アデノウイルスは、中間程度のゲノムサイズ、操作の便宜性、高いタイター、広範囲なターゲット細胞および優れた感染性に起因して、遺伝子伝達ベクターとして多く利用されている。ゲノムの両末端は、100〜200bpのITR(inverted terminal repeat)を含み、これは、DNAの複製およびパッケージングに必須のシスエレメントである。ゲノムのE1領域(E1AおよびE1B)は、転写および宿主細胞遺伝子の転写を調節するタンパク質をコードする。E2領域(E2AおよびE2B)は、ウイルスDNAの複製に関与するタンパク質をコードする。現在開発されたアデノウイルスベクターのうち、E1領域が欠如した複製不能アデノウイルスが多く利用されている。一方、E3領域は、通常のアデノウイルスベクターから除去されて外来遺伝子が挿入されるサイトを提供する(Thimmappaya,B.et al.,Cell、31:543−551(1982);およびRiordan,J.R.et al.,Science、245:1066−1073(1989))。
レトロウイルスは、自分の遺伝子を宿主のゲノムに挿入させ、大量の外来遺伝物質を運搬することができ、感染させることができる細胞のスペクトルが広いので、遺伝子伝達ベクターとして多く利用されている。
アデノ随伴ウイルス(AAV)は、非分裂細胞を感染させることができ、多様な種類の細胞に感染できる能力を有しているので、本発明の遺伝子伝達体として適合している。AAVベクターの製造および用途に関する詳細な説明は、米国特許第5,139,941号および第4,797,368号に詳細に開示されている。
他のウイルスベクターも、本発明の遺伝子伝達体として利用することができる。ワクチニアウイルス(Puhlmann M.et al.,Human Gene Therapy 10:649−657(1999);Ridgeway,“ Mammalian expression vectors,”In:Vectors:A survey of molecular cloning vectors and their uses.Rodriguez and Denhardt,eds.Stoneham:Butterworth、467−492(1988);Baichwal and Sugden,“Vectors for gene transfer derived from animal DNA viruses:Transient and stable expression of transferred genes,” In:Kucherlapati R、ed.Gene transfer.New York:Plenum Press、117−148( 1986)およびCoupar et al.,Gene、68:1−10(1988))、レンチウイルス(Wang G.et al.,J.Clin.Invest.104(11):R55−62(1999))または単純ヘルペスウイルス(Chamber R.,et al.,Proc.Natl.Acad.Sci USA 92:1411−1415(1995))に由来するベクターも、運搬しようとする目的ヌクレオチド配列を細胞内に運搬できる運搬システムとして利用することができる。
リポソームは、水相に分散したリン脂質により自動的に形成される。外来DNA分子をリポソームで成功裏に細胞内に運搬した例は、NicolauおよびSene,Biochim.Biophys.Acta,721:185−190(1982)およびNicolau et al.,Methods Enzymol.,149:157−176(1987)に開示されている。一方、リポソームを利用した動物細胞の形質転換に最も多く利用される試薬としては、リポフェクタミン(Lipofectamine,Gibco BRL)がある。運搬しようとする目的ヌクレオチド配列を内包したリポソームは、エンドサイトーシス、細胞表面への吸着またはプラズマ細胞膜との融合などの機序を通じて細胞と相互作用して細胞内に運搬しようとする目的ヌクレオチド配列を運搬する。
マウスGM−CSF遺伝子は、Dr.Gerald C.O’Sullivan(Cork Cancer Research Centre,Mercy University Hospital and Leslie C.Quick Jnr.Laboratory,University College Cork,Cork,Ireland)から入手し、塩基配列は、Cancer Gene Therapy(2006)13,1061−10710に示された通りであり、配列番号1で表示される。
韓国特許出願第2015−0044507号と同じ遺伝子を使用し、韓国特許出願第2015−0044507号の製造例1と同一に製作した。
ヒトFlt3L−TRAIL遺伝子は、Flt3L(FMS−like tyrosine kinase 3 ligand)の1−181アミノ酸をコードする遺伝子とTRAIL(tumor necrosis factor−related apoptosis−inducing ligand)の95−281アミノ酸をコードする遺伝子がロイシンジッパー(leucine zipper)で融合したものであって、ジーンバンクU03858(Flt3L)とジーンバンクB032722またはU57059(TRAIL)で示したように、配列番号3で表示される。
Adlox−FETZ(Flt3L−TRAIL)はpFETZ(Flt3L−TRAIL)(Regression of human mammary adenocarcinomaby systemic administration of a recombinant gene encoding the hFlex−TRAIL fusion protein by Xiaofeng Wu et al Molecular Therapy vol.3 368−374,2001)をSal/BamHIで処理したFETZ(Flt3L−TRAIL融合遺伝子;Flt3Lは、可溶型(soluble form)のみで構成されており、TRAILは、細胞外ドメインである95−281部位のみを有するものであって、この二つは、イソロイシンジッパーで連結されている)をSalI/BamHIで切断したAdloxベクターに移したものである(図4a)。
韓国特許公開第2013−0012095号に開示されたように、配列番号4または5で表示されるTGF−β1の発現を抑制するshRNA(以下、shTGF−β1という)を製作した。
韓国特許公開第2013−0088792号に開示されたように、配列番号6または7で表示されるTGF−β2の発現を抑制するshRNA(以下、shTGF−β2という)を製作した。
韓国特許公開第2013−0088792号および第2013−0012095号を基盤として、ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−U6−shTGF−β(β1 orβ2)は、Bsp1191で切断し、pVAX1−3484−CMVp−△E1Bは、PmeIで線形化させて、大腸菌BJ5183に形質転換させて、相同組換えDNAを生成した。その結果、HindIIIパターンとPacI切断で組換えされたことを確認した(図5aおよび図5b)。
韓国特許公開第2013−0123244号に開示されたように、配列番号9で表示されるHSP27発現を抑制するshRNA(以下、shHSP27という)を製作した。
韓国特許公開第2013−0123244号を基盤として、ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−H1−shHSP27はBsp1191で切断し、pVAX1−3484−CMVp−△E1BはPmeIで線形化させて、大腸菌BJ5183に形質転換させて、相同組換えDNAを生成した。その結果、HindIIIパターンとPacI切断で組換えされたことを確認した(図6)。
配列番号8で表示されるHSP25発現を抑制するshRNA(shHSP25)を製作した。

前記参照例5において減少効果がある2番と3番のpSP72△E3−H1−shHSP25をdl324−IXと相同組換えをした。dl324−IX−shHSP25 2番への相同組換えの場合、pSP72−shHSP25−2シャトルベクターはDrdIで切断し、バックボーンDNAであるdl324−IXはSpeIで切断し、それぞれ線形化させた後、BJ5183に形質転換させて得たコロニーをHindIIIパターンとPacI切断で選別した(図8)。他方、dl324−IX−shHSP25 3番への相同組換えの場合は、pSP72−shHSP25−3シャトルベクターはXmnIで切断し、バックボーンDNAであるdl324−IXはSpeIで切断し、それぞれ線形化させた後、BJ5183に形質転換させて得たコロニーをHindIIIパターンとPacI切断で選別した(図9)。このようにして得たコンストラクトからHSP25 shRNAを発現するそれぞれのアデノウイルスから発現減少効果を確認した(図10)。したがって、今後のマウスshRNA HSP25の製作には、shHSP25−2配列[配列番号8]を使用した。
GM−CSFとFlt3L−TRAILを同時に発現する腫瘍選択的複製可能アデノウイルスを次のように製作した。
(1)ヒト
pIRES(Clontech)のMCS(multi cloning site)AにはGM−CSF遺伝子を挿入し、MCS BにはFlt3L−TRAIL遺伝子を挿入した。
ヒトGM−CSF遺伝子をpIRESでMCS A部位にサブクローニングするために、PCR用プライマーを製作した。センスストランドは、5’末端にXhoI部位を有する5’−CCG CTCGAG ATGTGGCTGC AGAGCCTGCT G−3’とアンチセンスストランドとして5’末端にMluIを有する5’−CCGACGCGTTCACTCCTGGACTGGCTCCCA−3’を製作した。PCR鋳型としては、pORF−GMCSFを使用して初期変性:95℃、2min後、変性:95℃、1min、アニーリング:55℃、1min、伸長:72℃、1minを30回繰り返した。そして、最後に、最終伸長:72℃、5minを行った。
マウスGM−CSF遺伝子をpIRESでMCS A部位にサブクローニングするために、PCR用プライマーを製作した。センスストランドは、5’末端にXhoI部位を有する5’−CCG CTCGAG ATGTACAGGATGCAACTCCTGTCT−3’とアンチセンスストランドとして5’末端にMluIを有する5’−CCGACGCGT TCATTTTTGGCCTGGTTTTTTGCA−3’を製作した。PCR鋳型としては、pCA14−mGM−CSFを使用して初期変性:95℃、2min後、変性:95℃、1min、アニーリング:55℃、1min、伸長:72℃、1minを30回繰り返した。そして、最後に、最終伸長:−72℃、5minを行った。
腫瘍溶解性(Oncolytic)シャトルベクターへのクローニングは、次のように行った。
ウイルス生成のために、ウイルスバックボーンDNAであるdl324−BstBIは、Bsp1191で切断し、pVAX1−3484−CMVp−△E1B−GMCSF−IRES−Flt3L−TRAILはPmeIで線形化させて、大腸菌BJ5183に形質転換させて、相同組換えDNAを生成した。その結果、HindIIIパターンおよびPacI切断で全部成功裏に組換えされたことを確認した(図13)。

前記pSP72△E3−U6−shTGFβ2をHindIIIブランティング処理した後、KpnI処理して、SV40 poly A部位を除去した後、前記pSP72△E3−H1−shHSP27でSphIブランティング後、KpnI処理して、H1プロモーター−shHSP27−SV40 poly Aを取り出して、前記HindIII(blunt)/KpnI処理したpSP72△E3−U6−shTGFβ2と連結させて、pSP72△E3−U6−shTGFβ2−H1−shHSP27シャトルベクターを製作した。
1)pSP72△E3−U6−マウスshTGFβ1の製作
ヒトshTGFβ1の代わりに、マウスshTGFβ1を使用することを除いて、前記製造例7の1)と同一に製作した。
ヒトshTGFβ2の代わりに、マウスshTGFβ2を使用することを除いて、前記製造例7の1)と同一に製作した。
ヒトshHSP27の代わりに、マウスHSP25を使用することを除いて、前記製造例7の1)と同一に製作した。
pSP72△E3−U6−マウスshTGFβ1をHindIIIで切断し、pSP72ΔE3−H1−shHSP25をSphIで切断した後、ブランティングさせた。そして、それぞれをKpnIで切断した後、ライゲーションさせて、pSP72△E3−H1−mshHSP25−U6−mshTGFβ1を得た(図24a)。
腫瘍選択的殺傷アデノウイルスの製作は、次の通りである。
マウス形態のshRNAを発現する腫瘍選択的アデノウイルスの製作のためのdl324−3484−CMVp−△E1B−H1−shHSP25−U6−mshTGFβ1の相同組換え過程は、次の通りである。
ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−U6−shTGF−β1は、製造例7で提示したpSP72△E3−U6−shTGFβ1とdl324−BstBIとの相同組換えを通じて得た。dl324−BstBI−U6−shTGF−β1は、Bsp1191で切断し、製造例6で得たpVAX1−3484−CMVp−△E1B−GMCSF−IRES−Flt3L−TRAILをPmeIで線形化して、大腸菌BJ5183に形質転換させて相同組換えを誘導した。その結果、HindIIIパターンとPacI切断で組換えされたことを確認した(図26)。
ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−U6−mshTGF−β1は、製造例8で提示したpSP72△E3−U6−mshTGFβ1とdl324−BstBIとの相同組換えを通じて得た。dl324−BstBI−U6−mshTGFβ1と製造例6で得たpVAX1−3484−CMVp−△E1B−mGMCSF−IRES−Flt3L−TRAIL相同組換えのために、シャトルベクターであるpVAX1−3484−CMVp−△E1B−mGMCSF−IRES−Flt3L−TRAIL(図12b)をPmeIで切断した後、バックボーンであるdl324−BstBI−U6−mshTGFβ1をBsp1191で切断した後、相同組換えした(図27)。
ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−H1−shHSP27は、製造例7で提示したpSP72△E3−H1−shHSP27とdl324−BstBIとの相同組換えを通じて得た。得られたdl324−BstBI−H1−shHSP27はBsp1191で切断し、製造例6で得たpVAX1−3484−CMVp−△E1B−GMCSF−IRES−Flt3L−TRAILはPmeIで線形化して、大腸菌BJ5183に形質転換させて、相同組換えを誘導した。その結果、HindIIIパターンとPacI切断でレーン1、2、3がいずれも成功裏に組換えされたことを確認した(図28)。
ウイルス生成のために、ウイルスバックボーンであるdl324−BstBI−H1−shHSP25は、製造例8で提示したpSP72△E3−H1−shHSP25とdl324−BstBIとの相同組換えを通じて得た。得られたdl324−BstBI−H1−shHSP25は、Bsp1191で切断し、製造例6で得たpVAX1−3484−CMVp−△E1B−mGMCSF−IRES−Flt3L−TRAIL相同組換えのために、シャトルベクターであるpVAX1−3484−CMVp−△E1B−mGMCSF−IRES−Flt3L−TRAILをPmeIで切断し、バックボーンであるdl324−BstBI−mshHSP25をBsp1191で切断して、相同組換えを実施した(図29)。
pVAX1−3484−CMVp−△E1B−GMCSFシャトルベクターは、pCA14−GMCSFをBglII処理後、ブランティングさせて、SalIで切断したインサートをEcoRIで切断した後、ブラントさせた後、さらにSalIで切断したpVAX1−3484−CMVp−△E1Bにサブクローニングさせて製作した(韓国特許出願第2015−0044507号の製造例1参照)。
dl324−3484−CMVp−△E1B−mGMCSF−shHSP25−mshTGFβ1への相同組換えは、E1シャトルベクターであるpVAX1−3484−CMVp−△E1B−mGMCSF(製造例1に基づく韓国特許出願第2015−0044507号を参照)は、PmeIで切断して線形化させ、製造例8で得たpSP72−ΔE3−H1−shHSP25−U6−mshTGFβ1とdl324−BstBIと相同組換えして得たバックボーンDNAであるdl324−BstBI−H1−shHSP25−U6−mshTGFβ1は、Bsp1191で切断して線形化させた後、BJ5183に形質転換させて得たコロニーをHindIIIパターンとPacI切断で選別した(図32)。
製造例2で得たシャトルベクター(pVAX1−3484−CMVp−△E1B−Flt3L−TRAIL)と製造例9で得たバックボーンであるdl324−BstB1−H1−shHSP27−U6−shTGFβ1またはdl324−BstB1−U6−shTGFβ2−H1−shHSP27を相同組換えさせた(図33および図34)。
ウイルス生成のために、ウイルスバックボーンである製造例8で得たpSP72−ΔE3−H1−shHSP25−U6−mshTGFβ1(製造例8)とdl324−BstBIを相同組換えして得たバックボーンDNAであるdl324−BstBI−H1−shHSP25−U6−mshTGFβ1(製造例9)は、Bsp1191で切断し、製造例2で得たpVAX1−3484−CMVp−△E1B−Flt3L−TRAILはPmeIで線形化させて、大腸菌BJ5183に形質転換させて、相同組換えDNAを生成した。その結果、HindIIIパターンとPacI切断で組換えされたことを確認した(図35)。
製造例9で得たdl324−BstBI−ΔE3−U6−TGF−β2−H1−HSP27またはdl324−BstBI−ΔE3−H1−HSP27−U6−TGF−β1と製造例6で得たpVAX1−3484−CMVp−△E1B−GMCSF−IRES−Flt3L−TRAILとの相同組換えを通じてdl324−3484−CMVp−△E1B−GMCSF−IRES−Flt3L−TRAIL−U6−shTGFβ2−H1−shHSP27(YSC−01)(図36、図38)。またはdl324−3484−CMV−△E1B−GMCSF−IRES−Flt3L−TRAIL−H1−shHSP27−U6−shTGFβ1(YSC−02)を製作した(図37、図39)。
製造例10で得たdl324−BstBI−H1−mshHSP25−U6−mshTGFβ1と製造例6で得たpVAX1−3484−CMVp−△E1B−mGMCSF−Flt3L−TRAILを相同組換えするために、シャトルベクターであるpVAX1−3484−CMVp−△E1B−mGMCSF−Flt3L−TRAILをPmeIで切断し、バックボーンDNAであるdl324−BstBI−H1−shHSP25−U6−mshTGFβ1をBsp1191で切断した後、組換えされたことを確認した(図40および図41)。
ヒトMDAMB−231−Her2細胞株1×107cell/100μlの細胞を腹壁に注入し、腫瘍サイズが60mm3に達したとき、二日間隔で3回(1×109pfu/50μl)、アデノウイルス(対照群:control virus、oncolytic NC、製造例3の1種oncolytic shTGF−β1、製造例4の1種のoncolytic shHSP27)、製造例7の2種(shTGF−β1+shHSP27)腫瘍内注射(intratumoral injection)で感染させた。各実験群当たり6匹ずつを使用した。
膵臓癌細胞株MiaPaCa−2、HPAC、BxPC3、AsPc−1、Capan−1、肝癌細胞株SK−Hep1、Huh7、肺癌細胞株A549細胞株、前立腺癌細胞株DU−145、黒色腫細胞株A375、神経膠腫U251N、そして乳癌細胞株MDAMB231にHer2を過発現させた細胞株を6ウェルプレートに2×x105cellで敷設し、欠損アデノウイルス/NC(陰性対照群)、製造例3のアデノウィルス/shTGF−β1、アデノウィルス/shTGF−β2をそれぞれ100moiで感染させ、二日後、細胞を剥き取って、6ウェルに1×105cell/wellで分注した後、二日に一回ずつ5%FBS含まれた培地に交替した。10日後、培養液を除去し、4%パラホルムアルデヒドで固定させた後、クリスタルバイオレットで染色して観察した。
MDAMB231−Her2にshTGF−β1の他にもshHSP27が必要であるかを調べるために、欠損アデノウイルス/NC(陰性対照群)、製造例3の1種、アデノウイルス/shTGF−β1、製造例7の2種(アデノウイルス/shTGFβ1−shHSP27)をそれぞれ100moiで感染させた後、ウェスタンブロットを行った。
一つの特異な事項は、TGF−βとHSP27を同時に低下させる場合、TRAILの受容体であるDR4、DR5が増加し、その代わりに、TRAIL抵抗性に寄与するCDK9は減少するなど、TRAILによる細胞毒性増加の可能性を増加させる。図50は、そのような可能性を裏付けている。実際にTGF−β/HSP27低下およびTRAIL発現させる場合、生存に関係する信号がさらに減少することを確認した(図51)。
YSC−01とYSC−02のウイルスDNAからウイルス中に外来遺伝子4個の遺伝子挿入が正常に起こったことをウイルスDNAの塩基配列分析を通じて確認した(図52)。
MDA−MB231−Her2、MiaPaCa−2、A549、Huh7、DU145等のp53変異型とA375、HPACなどのp53正常型癌細胞株、そして正常な膵臓細胞(Pancreatic normal cell)とChangなどの正常細胞株を含むいくつかの細胞株において生存率確認のためのクローン形成法を行った。
MiaPaCa−2細胞株8×106cell/100μlの細胞を腹壁に注入し、腫瘍サイズが60mm3に達したとき、二日間隔で3回(1×109pfu/50μl)、アデノウイルス(陰性対照群:Ad−3484−NC)、製造例6の2種(Ad−3484−GM−CSF−Flt3L−TRAIL)、製造例19の4種(Ad−3484−GM−CSF−Flt3L−TRAIL−shTGFβ2−shHSP27(YSC−01)、Ad−3484−GM−CSF−Flt3L−TRAIL−shHSP27−shTGFβ1(YSC−02))を腫瘍内注射(intratumoral injection)で感染させた。各実験群当たり6匹ずつを使用した。
マウスタイプのYSC−02による抗腫瘍効果を様々な比較対象グループと比較するために、免疫可能マウス モデルをまず製作した。すなわち、免疫能があるマウス(Balb c)にアデノウイルスの感染および複製が可能なマウス肝癌細胞株(BNL−CAR−E1B55K−HSP25)を製作した。
