を含む。好ましくは、異種タンパク質をコードするDNA配列には、その3’末端で、染色体の若しくは内在性毒性プラスミドの細菌エフェクタータンパク質からの送達シグナルの3’末端におけるDNA配列と相同な又はその断片と相同なDNA配列が隣接している。より好ましくは、相同タンパク質にその3’末端で隣接しているこのDNA配列は、染色体上の若しくは内在性毒性プラスミド上の細菌エフェクタータンパク質からの送達シグナルの3’末端において10kbp以内に位置するDNA配列と又はその断片と相同である。特に、相同タンパク質にその3’末端で隣接しているこのヌクレオチド配列は、上記DNA配列と相同であり、染色体上の又は内在性毒性プラスミド上の細菌エフェクタータンパク質からの送達シグナル又はその断片と同じオペロン内にある。この実施態様では、形質転換は、融合した第1及び第2のDNA配列が、組換え弱毒化グラム陰性菌株の内在性毒性プラスミド又は染色体、好ましくは内在性毒性プラスミドに対する相同組み換えにより挿入され、融合した第1及び第2のDNA配列が、内在性毒性プラスミドのプロモーターに、又は染色体の、例えば、染色体病原性アイランドのプロモーターに動作可能に連結されるように、通常は行われる。好ましくは、融合した第1及び第2のDNA配列は、内在性毒性プラスミドのプロモーターに動作可能に連結される。この実施態様では、第一のDNA配列は、第2のDNA配列が、内在性プロモーターに動作可能に連結されている染色体又は内在性毒性プラスミド送達シグナルの3’末端とインフレームで配置される結果となるような、染色体の又は内在性毒性プラスミドの相同部位での相同組換えをもたらす、細菌エフェクタータンパク質からの送達シグナル又はその断片、好ましくはその断片を含む。
エフェクターをコードしている遺伝子のコード領域において、コードされているエフェクタータンパク質の触媒活性を消失させるような変異を生じさせることもできる。エフェクタータンパク質の「触媒活性」は、エフェクタータンパク質の抗標的細胞機能、すなわち毒性を通常は意味する。そのような活性は、エフェクタータンパク質の触媒ドメインにおける触媒モチーフにより支配される。エフェクタータンパク質の触媒ドメイン及び/又は触媒モチーフを同定する手法は、当業者に周知である。例えば、[24、25]を参照されたい。
細菌エフェクタータンパク質からの送達シグナル又はその断片をコードする第1のDNA配列、
前記第1のDNA配列の3’末端とインフレームで融合している、異種タンパク質をコードする第2のDNA配列
を含むベクターで、形質転換される。
a)プロテアーゼが、細菌T3SSエフェクタータンパク質からの送達シグナルと異種タンパク質としてのプロテアーゼとの融合タンパク質を発現する本明細書に記載の組換え弱毒化グラム陰性菌株により真核細胞に移行される方法、又は
b)プロテアーゼが、真核細胞において構成的に若しくは一過性に発現される方法
である。
i)本明細書に記載の組換え弱毒化グラム陰性菌株を培養すること、
ii)悪性固形腫瘍の細胞をi)の組換え弱毒化グラム陰性菌株と接触させること(ここで、細菌エフェクタータンパク質からの送達シグナルと異種タンパク質とを含む融合タンパク質が、組換え弱毒化グラム陰性菌株により発現され、悪性固形腫瘍の細胞内に移行される)、及び任意選択的に、
iii)融合タンパク質を切断し、その結果、異種タンパク質を悪性固形腫瘍細胞内の細菌エフェクタータンパク質からの送達シグナルから切断すること
を含む。
A)材料及び方法
菌株及び増殖条件。この研究で使用した株を図14A−Nに列挙する。プラスミド精製及びクローニングに使用した大腸菌Top10、及び接合伝達に使用した大腸菌Sm10λpir、並びにpKNG101を伝播するために使用した大腸菌BW19610[28]を、LB寒天プレート上及びLB培地中、37℃で常套的に増殖させた。アンピシリンを200μg/ml(エルシニア属)又は100μg/ml(大腸菌)で使用して、発現ベクターについて選択した。ストレプトマイシンを100μg/mlの濃度で使用して、自殺ベクターについて選択した。Y.エンテロコリティカMRS40(O:9、生物型2)[17]、非アンピシリン耐性E40派生物[16]及びそれに由来する株をブレインハートインフュージョン(BHI:Difco)上で、室温で常套的に増殖させた。Y.エンテロコリティカ株に、ナリジクス酸(35μg/ml)を添加し、全てのY.エンテロコリティカasd株に、さらに、100μg/mlのメソ−2,6−ジアミノピメリン酸(mDAP、Sigma Aldrich)を補充した。サルモネラ菌SL1344をLB寒天プレート上及びLB培地中、37℃で常套的に増殖させた。アンピシリンを100μg/mlの濃度で使用して、サルモネラ菌における発現ベクターについて選択した。

全ての動物実験は、認可されたものであり(license 1908;Kantonales Veterinaramt Basel−Stadt)、地域のガイドライン(Tierschutz−Verordnung;Basel−Stadt)及びスイス動物保護法(Tierschutz−Gesetz)に従って行った。6週齢C57Bl/6及びBALB/cマウスをJanvier Labsから取り寄せた。少なくとも1週間の馴化後、尾静脈への注射により、マウスをY.エンテロコリティカMRS40 ΔHOPEMT又はネズミチフス菌(S.typhimurium)ΔaroAに感染させた。この実験を通して、マウスの行動及び身体的外見についてのスコアを付け、体表温度並びに体重を測定した。マウスに静脈内投与した種菌を希釈平板法により検証した。注射後のそれぞれの日に、マウスをCO2吸入により屠殺した。直ちに心臓からの吸引により血液試料を単離した。肝臓、脾臓、肺及び腫瘍を単離し、それらの重量を判定した。臓器及び腫瘍をホモジナイズした。ナリジクス酸(35ug/ml)を含有するLB寒天プレート上への段階希釈物のスポッティングにより、各試料におけるCFUを判定した。

YopE融合タンパク質の3型分泌に基づくタンパク質送達系
Y.エンテロコリティカT3SSエフェクターYopE(配列番号1)のまさにN末端は、異種タンパク質を移行させるのに十分な分泌シグナルを有する[19]が、そのシャペロン(SycE)についてはシャペロン結合部位(CBS)を含まない[39]。本発明者らは、YopEのN末端138アミノ酸(配列番号2)を選択して、送達すべきタンパク質と融合させた。このYopEのN末端138アミノ酸は、他の異種T3S基質の移行について最良の結果をもたらすことが証明されていた[21]からであった。YopEのこれらのN末端138アミノ酸はCBSを含有するので、本発明者らは、さらに、SycEを共発現させることを決めた。精製Y.エンテロコリティカpYV40毒性プラスミドからクローニングしたSycE−YopE1−138断片は、YopEの及びそのシャペロンSycEの内在性プロモーターを含有する(図9)。したがって、SycEと任意のYopE1−138の融合タンパク質は、室温での増殖から37℃への急速な温度シフトにより誘導される。37℃での培養時間は、細菌内に存在する融合タンパク質量に影響を与えることになる。多重クローニング部位(MCS)をYopE1−138の3’末端に付加させ(図9B)、その後、Myc及び6×Hisタグ及び終止コドンを付加させた。
インビトロ分泌アッセイ(図1Aを参照されたい)では、周囲の液体へのタンパク質分泌を人工的に誘導する。TCAに基づくタンパク質沈殿の後、抗YopE抗体でのウェスタンブロット分析を使用して、分泌されたタンパク質量を決定した(図1B)。wt株は、完全長YopEを分泌したが、ΔHOPEMT asd株は、分泌しなかった。YopE1−138−Myc−His(さらなる名称YopE1−138−Myc;配列番号3)が存在すると、より小さなYopEバンドが目に見えるようになった(図1B)。したがって、YopE1−138断片は本明細書に記載の設定で十分に分泌される。真核細胞へのタンパク質移行の均一性を分析するために、本発明者らは、YopE1−138−Mycをコードする株にHeLa細胞を感染させ、IFによりMycタグを染色した(図2A及びB)。最初は細菌のみが染色されたが、感染後(p.i.)30分の時点では、細胞の輪郭が目に見え始め、これは感染時間を増加させると増強される(図2B)。この傾向は、HeLa細胞の内部のMycタグ染色強度によく反映される(図2A及びB)。YopE1−138−Mycは、核内を除いて[44]、細胞のどこででも検出することができる(図2A)。意外なことに、この手法により、全てではないが大多数の細胞に同等に到達した。Y.エンテロコリティカに多くの異なる細胞型が感染することが公知であるので[45]、本発明者らは、様々な細胞株へのYopE1−138−Myc送達を追跡した。感染したマウス線維芽細胞、ジャーカット細胞及びHUVECにおいて同じ同種抗Myc IF染色が観察された(図11)。さらに、感染多重度を高く又は低く調整することにより、なお大多数の細胞を標的としたまま、送達されるタンパク質量を調節することが可能になる(図2C)。細菌数を少なくすると、少数の細胞が大量の送達タンパク質を有することになるのではなく、大多数の細胞が少量の送達タンパク質を有することになる(図2C)。
YopE自体は細胞質に局在していたので(図2A)、YopE1−138断片が核融合タンパク質の局在化を妨げるかどうかを試験することは特に興味深い。したがって、本発明者らは、YopE1−138−EGFPのC末端(及びN末端、同様の結果)にSV40 NLSを付加させた(それぞれ配列番号39及び配列番号38)。感染させたHeLa細胞において、YopE1−138−EGFP(配列番号37)は、弱い細胞質染色をもたらしたが、YopE1−138−EGFP−NLSは、より強力な核内EGFPシグナルを生じさせた(図3)。これは、YopE1−138断片がNLSの使用と適合性であることを示す。mCherryは、植物病原体において既に使用されているが[46]、これは、T3SSをコードするヒト又は動物病原性細菌を介したGFP様タンパク質の送達成功を意味する。これにより、SycE及びYopE1−138依存性戦略は最適な多くのタンパク質の送達に非常に有望であることが検証される。
YopE1−138断片は、細菌送達に大いに役立つものであるが、融合タンパク質の機能及び/又は局在化を妨げる可能性がある。したがって、タンパク質送達後にそれを除去することが最適であるだろう。この目的のために、本発明者らは、YopE1−138と融合パートナー(転写制御因子ET1−Myc(配列番号36及び41)[50]及びヒトINK4C(配列番号40及び配列番号43))との間に2つのTEV切断部位(ENLYFQS)[47−49]を導入した。提供する方法の優位性を保持するために、本発明者らは、別のY.エンテロコリティカ株においてTEVプロテアーゼ(S219Vバリアント;[51])をYopE1−138(配列番号42)とさらに融合した。HeLa細胞を一度に両方の株に感染させた。タンパク質の移行画分のみの分析を可能にするために、感染HeLa細胞を、感染の2時間後に(図4)、細菌を溶解しないことが公知であるジギトニンを用いて溶解した([52];コントロールについては図11を参照されたい)。ウェスタンブロット分析は、細胞が対応する株に感染した場合にのみ、YopE1−138−2×TEV切断部位−ET1−Myc又はYopE1−138−2×TEV切断部位−Flag−INK4C−Mycの存在を明示した(図4A及びC)。この細胞溶解物を精製TEVプロテアーゼで一晩消化すると、シフトしたバンドを観察することができた(図4A及びC)。このバンドは、TEV切断部位のN末端に残存物がある、ET1−Myc(図4C)又はFlag−INK4C(図4A)に対応し、この残存物は、1つのセリンのみの可能性が最も高い。TEVプロテアーゼを送達する株に細胞を同時感染させると、同じ切断ET1−Myc又はFlag−INK4C断片が目に見えるようになった。これは、T3SSを介して送達されたTEVプロテアーゼが機能性であること、及び単一細胞が両方の菌株に感染したことを示す(図4A及びC)。切断は完全なものではないが、移行したタンパク質の大多数が、感染の2時間後に既に切断され、精製TEVプロテアーゼで一晩消化しても、より高い切断率は得られなかった(図4B)。報告されているように、TEVプロテアーゼ依存性切断は、融合タンパク質次第で最適化を必要とする可能性がある[53、54]。したがって、移行後のYopE1−138付属物のTEVプロテアーゼ依存性除去は、アミノ酸組成をN末端アミノ酸1つだけしか変化させずに、殆どの天然異種タンパク質のT3SSタンパク質送達をもたらす初めてのものである。
サルモネラ菌からのSopEは、Cdc42と相互作用してアクチン細胞骨格リモデリングを促進する、よく特徴づけられているグアニンヌクレオチド交換因子(GEF)である[55]。HeLa細胞へのYopE1−138−Mycの移行の影響はなかったが、移行したYopE1−138−SopE(配列番号5及び135)は、アクチンネットワークの劇的な変化を誘導した(図5A)。別のGEFエフェクタータンパク質である、フレクスナー赤痢菌からのIpgB1(配列番号4)で、同様の結果が得られた。意外なことに、アクチン細胞骨格の最初の変化は、感染後2分という速さで観察された(図5A)。したがって、T3SS依存性タンパク質送達は、遠心分離により感染を開始した直後に起こると、結論づけることができる。厳密なT3SS依存性輸送を証明するために、真核細胞膜への移行孔を形成するT3SSタンパク質の1つを欠失させた(YopB、[56]を参照されたい)(図11)。
ヒトタンパク質がIII型分泌によって移行できることを明らかにするために、本発明者らは、ヒトアポトーシス誘導因子を、Y.エンテロコリティカによる送達のためにYopE1−138と、又はサルモネラ菌による送達のためにSteA1−20、SteA、SopE1−81若しくはSopE1−105と融合させた。次いで、本発明者らは、Bcl−2タンパク質ファミリーのアポトーシス促進性スメンバーであるヒトBH3相互作用ドメインデスアゴニスト(BID、配列番号24)の移行をモニターした。BIDは、カスパーゼ−8(CASP8)により誘導されるミトコンドリア損傷のメディエーターである。CASP8は、BIDを切断し、切断型BID(tBID、配列番号25)はミトコンドリアに移行し、そこでシトクロムC放出を誘発する。後述のトクロムC放出は、固有モードのカスパーゼ3(CASP3)活性化をもたらし、その間に、CASP3は17kDaサブユニットと12kDaサブユニットに切断される[61]。YopE1−138−Myc又はYopE1−138−BIDを発現するY.エンテロコリティカによる1時間の感染は、アポトーシスを誘導することができなかったが、ヒトtBIDの移行は、よく特徴づけられているアポトーシス誘導因子スタウロスポリンよりも大きな程度で細胞死を誘発した(図7A及びC)。予測通り、tBIDの移行は、CASP3 p17サブユニットの産生を、もっと言えば、スタウロスポリンを用いた場合より多い量での産生をもたらす(図7A)。移行したタンパク質量と内在性Bidを比較することができるように、HeLa細胞をジギトニンで溶解し、抗Bid抗体を使用するウェスタンブロット法により分析した(図7B)。T3SSにより送達されたYopE1−138−tBIDは、HeLa細胞内でほぼ内在性Bidレベルに達したが、送達されたYopE1−138−BIDのほうが多い量で存在した(2.5倍)(図7B)。HeLa細胞のディーププロテオーム及びトランスクリプトームマッピングにより、単一細胞当たりBIDコピー数105の4.4倍が推定された[62]。したがって、T3SS依存性ヒトタンパク質送達は、1細胞当り105−106タンパク質に達すると結論づけることができる。これらの数は、大腸菌T3SSにより移行されるナノボディの細胞当たりのコピー数[63]と一致する。感染多重度が及び感染期間が10倍、抗生物質添加の時点及び感染前の37℃での培養時間が3.2倍のレベルになると仮定すると、送達されるタンパク質コピー/細胞を、数1000コピー/細胞から数106コピー/細胞まで調整することができる。総じて、これらの結果は、移行したtBIDが機能性であり、有意味なレベルで送達されることを示した。これにより、細胞生物学の中心的側面である、アポトーシスの調節におけるタンパク質の役割を研究するための移行ツールが検証された。
リン酸化は、生物学的プロセスを活性化するか又は不活化することができる広範な翻訳後修飾であり、したがって、シグナル伝達事象を研究するための好適な標的である[65]。それにもかかわらず、現在利用できる、アポトーシスに関するリン酸化の系レベルの分析はない。HeLa細胞に送達されたヒトtBidの影響を分析するために、本発明者らは、LC−MS/MSによるラベルフリーリン酸化プロテオミクス手法を使用した。3つの独立した実験において、細胞を未処理のまま放置するか、又はΔHOPEMT asd+YopE1−138−Myc若しくはΔHOPEMT asd+YopE1−138−tBidに30分間感染させた。細胞を溶解し、その後、酵素的消化、リン酸化ペプチド濃縮、並びに個々のリン酸化ペプチドの定量及び同定を行った。本発明者らは、ΔHOPEMT asd+YopE1−138−Mycに感染させた細胞とΔHOPEMT asd+YopE1−138−tBidに感染させた細胞を比較し、それにより、363のtBid依存性リン酸化事象を同定することができた。本発明者らがtBidリン酸化プロテオームと定義した、243の異なるタンパク質に対応する、tBid送達時に、286のリン酸化ペプチドは、リン酸化の増加を示したが、77は、然程リン酸化されなかった。STRINGデータベースを使用して、tBidリン酸化プロテオームのタンパク質間相互作用ネットワークを生成した[66](図8A)。さらに、ミトコンドリアでのアポトーシスに関係することが公知の27のタンパク質をネットワークに追加して、中心クラスターを構築した。興味深いことに、tBidリン酸化プロテオームからのほんの少数のタンパク質しかこの中心クラスターにつながらず、これは、多くのタンパク質が、これまでアポトーシスタンパク質に直接関連づけられなかったリン酸化の変化を受けることを示す。tBidリン酸化プロテオームによりカバーされる生物学的機能を特徴づけるために、本発明者らは、Database for Annotation,Visualization,and Integrated Discovery(DAVID、http://david.abcc.ncifcrf.gov/)[67、68]の機能アノテーションツールを使用して遺伝子オントロジー分析を行った。同定された生物学的機能は、多様な細胞プロセスがtBidによる影響を受けることを示す。クロマチン再構成及び転写制御に関与する多くのタンパク質は、リン酸化の変化を受ける(すなわち、CBX3、CBX5、TRIM28、HDAC1)。例えば、HDAC1は、転写制御に関与するヒストンデアセチラーゼである。HDAC1が、アポトーシスにも関与するタンパク質であるNF−kBの転写活性を調節することができることは、証明されている。本発明者らは、アポトーシスの調節に重要な役割を果たすことが以前に証明されている、RNAプロセシングに関与するタンパク質のクラスターをさらに同定した[69]。例えば、HNRPKは、DNA損傷に対するp53/TP53応答を媒介し、アポトーシスの誘導に不可欠なものである[70]。さらに、タンパク質翻訳に関与するタンパク質のリン酸化も影響を受ける。いくつかの真核生物開始因子(すなわち、EIF4E2、EIF4B、EIF3A、EIF4G2)がリン酸化の変化を受け、これは、アポトーシス細胞においてタンパク質合成全体が減少するという観察と一致している。興味深いことに、細胞骨格リモデリングに関与する多くのタンパク質(例えば、PXN、MAP1B9)のリン酸化がtBid送達時に変更される。これは、細胞の形態がtBid送達時に劇的に変化するという観察と一致している(図8B)。細胞収縮、及び接触の喪失は、ZO2及びパキシリンのような接着関連タンパク質のリン酸化が観察されるという事実に反映される。同様に、核の収縮には、ラミンA/C及びラミンB1のような層状タンパク質のリン酸化が伴う。総じて、tBID送達は、ミトコンドリア完全性の破壊によっても示される迅速なアポトーシス応答を誘導する(図8B)。本発明者らは、tBid誘導アポトーシスが、多様な細胞プロセスにおける何百ものリン酸化事象に影響を与えることを明らかにした。同定された多くのタンパク質がアポトーシスに関係しているが、アポトーシス誘導時にリン酸化されることが分かっているのはほんの少数であった。したがって、リン酸化プロテオミクス手法は、アポトーシスに関するさらなる研究のための有用な方策となる。
反復した同一又は可変タンパク質ドメインからなる異種融合タンパク質がIII型分泌により移行することができることを証明するために、本発明者らは、Y.エンテロコリティカによる送達のためのマウスアポトーシス誘導因子をYopE1−138と融合させた。コントロールとして、本発明者らは、Y.エンテロコリティカによる送達のために、マウスtBID(Y.エンテロコリティカに対してコドン最適化されたもの;配列番号194)又はマウスtBID若しくはマウスBAX(どちらの場合もY.エンテロコリティカに対してコドン最適化されたもの;配列番号200及び201)のBH3ドメインを、YopE1−138と融合させた。異種融合タンパク質は、結果としてYopE1−138−(tBID−BH3)2(配列番号202)になるようにそれ自体と融合しているtBIDのマウスBH3ドメインの1つの事例に存する。第2の事例では、異種融合タンパク質は、結果としてYopE1−138−(tBID−BH3)−(BAX−BH3)(配列番号203)になる、BAXのマウスBH3ドメインと融合しているtBIDのマウスBH3ドメインからなる。マウスtBID及びマウスBAXの事例では、コドンをY.エンテロコリティカに対して最適化した。
Y.エンテロコリティカの場合、毒性は、「エルシニア属外部タンパク質」(Yop)と呼ばれる6つの内在性エフェクタータンパク質、詳細には、YopH、O、P、E、M、T(MRS40 pIML421[yopHΔ1−352、yopOΔ65−558、yopP23、yopE21、yopM23、yopT135])の欠失により低減される[40]。これらのYopは、約70kbpサイズのプラスミドである、「エルシニア属毒性プラスミド」(pYV)上にコードされており、プラスミド上には、完全3型分泌系(T3SS)はもちろん他の毒性プレーヤーもコードされている(図24)。YopH、O、P、E、M及びTは、免疫系を調節し弱めるために細菌3型分泌系により宿主細胞に送達される6つのエフェクタータンパク質である。各Yopは、宿主細胞において特異的生化学的活性を有する。YopTは、Rho GTPaseのC末端システインを切り離し、かくて、GTPaseを膜に固定化するイソプレニル基を除去する。誤った局在化に起因するRhoのこの活性化により、マクロファージ及び好中球のような免疫細胞による食作用が回避される[71]。同じ経路で、YopEは、Rho GTPaseに対してGTPase活性化タンパク質(GAP)として作用して、Rho GTPaseを不活性化する。この結果、免疫細胞による食作用が減少し、IL−1ベータ放出が阻害されることになる[71]。さらに、YopOは、グアニジンヌクレオチド解離阻害因子(GDI)として作用して、Rho GTPaseを不活性化する。YopOは、アクチン細胞骨格に対してまだ定義されていない方法で作用する、セリン/スレオニンキナーゼドメインを有する[71]。YopHは、接着斑キナーゼ(Fak)のような接着斑タンパク質、パキシリン等に対して作用し、かくてマクロファージ及び好中球による食作用を強力に防止する、チロシンホスファターゼである[71]。YopPは、偽結核菌(Y.pseudotuberculosis)又はペスト菌(Y.pestis)ではYopJと称するものであり、免疫細胞におけるMAPK/NFkB経路を不活性化して、これらの細菌の存在による刺激を受けた免疫細胞からのTNFa及びIL−8放出を防止することが判明した。さらに、YopPは、免疫細胞においてアポトーシスを誘導することが判明しており、これは、MAPK経路における効果に関係がある可能性があり、これは活性化状態で細胞をアポトーシスから保護する[71]。YopMの役割は、まだ完全に明らかになってはいないが、YopMは、リボソームS6キナーゼ1(RSK1)及びプロテインキナーゼC様2(PRK2)に関連して見出された。YopMは、RSK1のリン酸化を刺激し、かくて、例えば細胞周期進行のような、下流の標的に作用するように思われる[71]。これらのYopのうちの1つ又はいくつかを欠失させることにより、免疫系に対する細菌の防御機構に劇的な影響を与えられる[72]。それぞれのyopの変異を、それぞれの領域に関してPCRにより確認し、またインビトロ分泌アッセイにより確認した(図25)。SDS−PAGE及びクマシーブルー染色によるインビトロでの分泌の分析により、完全長YopH、O、M及びYopEの非存在が確認された。
急性毒性を評価するために、健常な免疫正常マウス(C57BL/6)に関する用量漸増研究を行った。この実験では、Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、T Δasd(Δasd起因して複製欠損があり、ΔyopH、O、P、E、M、Tに起因してさらに弱毒化されたもの)を、ネズミチフス菌ΔaroA(他者により使用されたような、ΔaroAに起因して増殖が減弱されたもの[73−75])と比較した。感染後の急性毒性を、細菌の静脈内注射後のマウスの体重増加及び体重減少に基づいて評価した。さらに、様々な臓器における細菌数を、臓器のホモジナイズ、段階希釈及びプレーティングにより決定した。急性毒性を評価し、細菌増殖が主な関心事でなかった場合、デスフェロキサミンでの前治療を適用しなかった。4つの異なる細菌負荷量を各株について試験した(動物1匹当り105、106、107、108cfu)。Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、T Δasdは、2種のより低い用量(105、106cfu)では体重減少をもたらさなかった(図26)。細菌107個の用量で、体重は、最初の24時間以内に約6%低下し、その後、安定化し、継続的に上昇した。最高用量を用いると、感染の48時間後に体重の7.5%低下が見られた(図26)。Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、T Δasdは、感染を確立することができず、デスフェロキサミンでの前治療の非存在下ではよりいっそう確立することができないので、細菌数は急速に低下すると予想し、実際、急速な低下が認められた(図27−30)。それにもかかわらず、例えば内毒素ショックに起因する、強力な急性毒性は、適用したY.エンテロコリティカの最高用量についてでさえ観察されなかった。これは、他者によるマウス研究[73−75]において何度も使用されてきた、増殖及び感染可能なネズミチフス菌ΔaroAとは対照的であり、臨床治験(NCT01099631)においてもaroA及びaroD二重突然変異型とは対照である。身体的外見及び行動のスコアづけに促されて、本発明者らは、最高用量のネズミチフス菌ΔaroAに感染させた全てのマウスを感染後1日目、及び107cfu用量については4日目に屠殺した。漸進的に減少する体重減少が3つの最高用量について認められた(図26)一方で、最低用量は、持続性の軽度体重減少しか誘導しないことが判明した(図26)。試験した最低用量ででさえ、感染の8日後まで肝臓、脾臓及び肺に存在するネズミチフス菌ΔaroAが認められた(図27−30)。これは、他者により確認されたネズミチフス菌ΔaroAの非最適体内分布[76]を反映している可能性がある。
腫瘍特異的ビヒクルとしての、T3SSエフェクターのような重要な毒性決定因子の変異を有するグラム陰性菌を検証するために、十分に確証されているB16F10メラノーマモデル([86]、ATCC番号CRL−6475)を使用するマウス同種移植腫瘍研究を行った。皮下腫瘍がある特定のサイズ(約100−200mm3)に達したら、マウスを2×105cfuのY.エンテロコリティカ亜種パレアークティカMRS40又はY.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、Tに静脈内感染させた。細菌増殖を可能にするために、感染の24時間前にマウスをデフェロキサミンで前治療した。wt Y.エンテロコリティカ亜種パレアークティカMRS40株に感染したマウスは、身体的外見及び行動について高いスコアが付き(図31−32)、感染の最初の48時間にわたって有意な体重減少を示し(図33)、これに促されて、本発明者らは、この群のマウス全てを既に感染後2日目に屠殺した。対照的に、Y.エンテロコリティカ亜種パレアークティカMRS40ΔyopH、O、P、E、M、T株に感染したマウスは、感染後4日目でも、有意な体重減少を示さず、身体的外見及び行動について正常なスコアが付与された(図31−33)。wt株(Y.エンテロコリティカ亜種パレアークティカMRS40)に感染したマウスでは、評価した全ての臓器において、及びさらには血液中で、生きている細菌が検出された(図35)。悪性固形腫瘍内に存在するwt細菌が認められたが、同程度に多い又はそれ以上に多い数が他の臓器において認められ、脾臓内が最も多かった(図35)。際だって対照的に、Y.エンテロコリティカ亜種パレアークティカMRS40ΔyopH、O、P、E、M、Tに感染したマウスでは、生きている細菌は、感染後1日目に主として悪性固形腫瘍内で認められ、少ない細菌数が脾臓、肝臓及び肺で観察された。特に、感染後4日目に、悪性固形腫瘍内の細菌数は、数桁増加した(腫瘍組織1g当り108cfu超に達した)が、評価した全ての他の臓器では、細菌数が検出限界未満に低下した(図34)。したがって、Y.エンテロコリティカ亜種パレアークティカMRS40ΔyopH、O、P、E、M、Tは、感染後4日目に、脾臓又は肝臓と比較して(検出限界に対して割当量を計算して)悪性固形腫瘍部位で約(最小限)100万倍の割当量で蓄積した。
腫瘍特異的ビヒクルとしての、T3SSエフェクターのような重要な毒性決定因子の変異を有するグラム陰性菌を検証するために、十分に確証されている4T1乳がんモデル(ATCC番号CRL−2539)を使用するマウス同種移植腫瘍研究を行った。皮下腫瘍がある特定のサイズ(約100−200mm3)に達したら、マウスを2×105cfuのY.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、Tに静脈内感染させた。細菌増殖を可能にするために、感染の24時間前にマウスをデフェロキサミンで前治療した。弱毒化Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、T株に感染したマウスは、感染後8日目でも、有意な体重減少を示さず、身体的外見及び行動について正常なスコアが付与された。Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、Tに感染したこれらのマウスでは、生きている細菌は、感染後8日目に悪性固形腫瘍内で排他的に認められた(図36)。したがって、Y.エンテロコリティカ亜種パレアークティカMRS40 ΔyopH、O、P、E、M、Tは、感染後8日目に、脾臓又は肝臓と比較して(検出限界に対して割当量を計算して)悪性固形腫瘍部位で約(最小限)数10,000倍の割当量で蓄積した。
上述の実験において、アポトーシス促進性タンパク質(例えば、t−BID(配列番号25)又はBIM(配列番号21))のT3SSに基づく送達は、がん性細胞を含むマウス細胞及びヒト細胞両方において、細胞死を効率的に誘導したこと、並びに細菌のコドン使用頻度に対して最適化したマウスtBID(配列番号138)を使用したときこの効果を増大させることができたことが分かる。この細胞殺滅増加は、使用される最適なコドンに起因するタンパク質産生量の増加及びT3SSによる後続の送達量の増加を反映する可能性が非常に高い。
表IV: 様々なアポトーシス促進性タンパク質で形質転換された株
遺伝子修飾Y.エンテロコリティカによる腫瘍定着実験を同系マウス同種移植モデル(4T1乳がんモデル)において繰り返し、細菌の定着を2週間にわたって追跡した。このとき、マウスを1×106コロニー形成単位(CFU)のY.エンテロコリティカΔyopH、O、P、E、M、Tに感染させた。感染後まだ間もない頃にB16F10モデルと同様の結果を得たが、本発明者らは、腫瘍定着が、感染後8日目に、そして14日目まで一貫して認められることをさらに明らかにすることができた(図39)。さらに、この定着は非常に特異的であり、評価した他の全ての臓器においてほんの少数の細菌しか検出されなかった(図40)。これらの研究結果は、Y.エンテロコリティカΔyopH、O、P、E、M、Tが、腫瘍の持続的定着を確立することによって、免疫系によるクリアランスを防止することができることを示す。
インビボでの腫瘍細胞に送達されるYopE1−138−(tBID BH3)2(配列番号202)の影響を評価するために、本発明者らは、4T1乳がん細胞を皮下同種移植した野生型Balb/Cマウスにおいて研究を行った。本発明者は、エルシニア属毒性プラスミドpYV上のYopEの天然部位に天然YopEプロモーターのもとでYopE1−138−(tBID BH3)2(配列番号202)をコードする、Y.エンテロコリティカΔHOPEMT株を評価することを目標とした。腫瘍が150−250mm3のサイズに達したら、PBS又は1×107のY.エンテロコリティカΔHOPEMT pYV−YopE1−138−(tBID BH3)をマウスに静脈内注射した。細菌の静脈内注射の日を0日目と定義した。その後数日(細菌の静脈内注射後0日目から9日目)にわたって腫瘍体積をノギスで測定した。腫瘍サイズの一切の初期均質性を補償するために、腫瘍体積を0日目の腫瘍体積に対して正規化した。Y.エンテロコリティカΔHOPEMT pYV−YopE1−138−(tBID BH3)2での治療は、腫瘍体積進行に対する影響を示し、細菌投与後8、9及び10日目に統計的に有意な腫瘍低減があった(図41)。重要なこととして、単独でのY.エンテロコリティカΔHOPEMTは、4T1マウスがんモデルにおいて腫瘍進行に影響を与えないことが判明した(図42)。これらの実験結果は、そのような細菌及びそれらのT3SSを腫瘍進行の妨害に用いることができることに光を当てるものである。

- 5’から3’方向に、
プロモーター、
前記プロモーターに動作可能に連結されている、細菌エフェクタータンパク質からの送達シグナルをコードする第1のDNA配列、
前記第1のDNA配列の3’末端とインフレームで融合している、異種タンパク質をコードする第2のDNA配列
を含むベクターで形質転換された、対象の悪性固形腫瘍を治療する方法において使用するための組換え弱毒化グラム陰性菌株であって、組換え弱毒化グラム陰性菌株が悪性固形腫瘍内に蓄積し、方法が前記組換え弱毒化グラム陰性菌株の対象への投与を含み、組換え弱毒化グラム陰性菌株が、対象を治療するために十分である量で投与される、組換え弱毒化グラム陰性菌株。 - エルシニア属(Yersinia)、大腸菌属(Escherichia)、サルモネラ菌(Salmonella)及びシュードモナス属(Pseudomonas)からなる群から選択される、請求項1に記載の使用のための組換え弱毒化グラム陰性菌株。
- 真核細胞に対して毒性である少なくとも1の細菌エフェクタータンパク質の産生が不十分である、請求項1又は2に記載の使用のための組換え弱毒化グラム陰性菌株。
- 真核細胞に対して毒性である全ての細菌エフェクタータンパク質の産生が不十分である、請求項1又は2に記載の使用のための組換え弱毒化グラム陰性菌株。
- 真核細胞に対して毒性である細菌エフェクタータンパク質をコードする、全てのエフェクターをコードする遺伝子が、結果として得られるエルシニア属が、真核細胞に対して毒性である何れの機能性細菌エフェクタータンパク質ももはや産生しないように変異している変異型エルシニア属株である、請求項1に記載の使用のための組換え弱毒化グラム陰性菌株。
- 変異型エルシニア属株が、Y.エンテロコリティカ(Y.enterocolitica)であり、細菌エフェクタータンパク質からの送達シグナルが、細菌T3SSエフェクタータンパク質からの送達シグナルであり、細菌T3SSエフェクタータンパク質からの送達シグナルがY.エンテロコリティカYopEエフェクタータンパク質のN末端138アミノ酸を含む、請求項5に記載の使用のための組換え弱毒化グラム陰性菌株。
- 異種タンパク質が、アポトーシス又はアポトーシス調節に関与するタンパク質、細胞周期制御因子、アンキリンリピートタンパク質、細胞シグナル伝達タンパク質、レポータータンパク質、転写因子、プロテアーゼ、低分子量GTPase、GPCR関連タンパク質、ナノボディ融合構築物及びナノボディ、細菌T3SSエフェクター、細菌T4SSエフェクター並びにウイルスタンパク質からなる群から選択される、請求項1から6の何れか一項に記載の使用のための組換え弱毒化グラム陰性菌株。
- 真核細胞に対して毒性である少なくとも1の細菌エフェクタータンパク質の産生が不十分であるか、又は分泌系機構の一部である少なくとも1の細菌タンパク質の産生が不十分である、対象の悪性固形腫瘍を治療する方法における使用のための組換え弱毒化グラム陰性菌株であって、組換え弱毒化グラム陰性菌株が悪性固形腫瘍内に蓄積し、方法が前記組換え弱毒化グラム陰性菌株を対象へ投与することを含み、組換え弱毒化グラム陰性菌株が、対象を治療するために十分である量で投与される、組換え弱毒化グラム陰性菌株。
- エルシニア属、大腸菌属、サルモネラ菌及びシュードモナス属からなる群から選択され、真核細胞に対して毒性である全ての細菌T3SSエフェクタータンパク質の産生が不十分である、請求項8に記載の使用のための組換え弱毒化グラム陰性菌株。
- 真核細胞に対して毒性である全ての細菌エフェクタータンパク質の産生が不十分であり、プロドラッグ変換酵素を発現する、請求項8に記載の使用のための組換え弱毒化グラム陰性菌株。
- 組換え弱毒化グラム陰性菌株の細菌約105−約109個が対象に投与される、請求項1から10の何れか一項に記載の使用のための組換え弱毒化グラム陰性菌株。
- 対象にシデロホアと共投与される、請求項1から11の何れか一項に記載の使用のための組換え弱毒化グラム陰性菌株。
- 約20−60日の期間内の2−20日毎の単一投与からなる投薬レジメンに従って対象に投与される、請求項1から12の何れか一項に記載の使用のための組換え弱毒化グラム陰性菌株。
- 悪性固形腫瘍が存在しない臓器内の組換え弱毒化グラム陰性菌株の細菌数が、対象への組換え弱毒化グラム陰性菌株の最終投与の少なくとも4日後に検出限界未満である、請求項1から13の何れか一項に記載の使用のための組換え弱毒化グラム陰性菌株。
- 対象の悪性固形腫瘍を治療する方法における使用のための、請求項1から14の何れか一項に記載の組換え弱毒化グラム陰性菌株と薬学的に許容される担体とを含む薬学的組成物であって、組換え弱毒化グラム陰性菌株が悪性腫瘍内に蓄積し、方法が前記薬学的組成物を対象へ投与することを含み、薬学的組成物が、対象を治療するために十分である量で投与される、薬学的組成物。
