Abstract
Description
(1)以下の(I)又は(II)のいずれかを有効成分とする、癌幹細胞における薬物耐性の低減剤。
(I)RAB3B遺伝子の発現を抑制する核酸分子;
(II)配列番号1に示されるRAB3Bタンパク質に結合する抗体若しくは低分子化合物;
(2)癌幹細胞が、肝癌幹細胞であることを特徴とする上記(1)記載の癌幹細胞における薬物耐性の低減剤。
(3)核酸分子がRAB3B遺伝子を標的とする二本鎖siRNA、microRNA、shRNA、アンチセンス核酸、及びリボザイムからなる群から選択される少なくとも1種の核酸分子であることを特徴とする上記(1)又は(2)記載の癌幹細胞における薬物耐性の低減剤。
(4)核酸分子がRAB3B遺伝子を標的とする二本鎖siRNAであることを特徴とする上記(3)記載の癌幹細胞における薬物耐性の低減剤。
(5)薬物が、抗腫瘍性抗生物質、微小管阻害剤、プラチナ製剤、ヒストン脱アセチル化酵素(HDAC)阻害剤、又はキナーゼ阻害剤であることを特徴とする上記(4)記載の癌幹細胞における薬物耐性の低減剤。
(6)薬物が、ドキソルビシン、ドセタキセル、カルボプラチン、ボリノスタット、又はスニチニブであることを特徴とする上記(5)記載の癌幹細胞における薬物耐性の低減剤。
(7)核酸分子がRAB3B遺伝子を標的とするmicroRNAであって、該microRNAがhas-miR-15a-5pであることを特徴とする上記(3)記載の癌幹細胞における薬物耐性の低減剤。
(8)薬物が、抗腫瘍性抗生物質、微小管阻害剤、プラチナ製剤、ヒストン脱アセチル化酵素(HDAC)阻害剤、キナーゼ阻害剤、又はトポイソメラーゼ阻害剤であることを特徴とする上記(7)記載の癌幹細胞における薬物耐性の低減剤。
(9)薬物が、ドキソルビシン、ドセタキセル、カルボプラチン、ボリノスタット、スニチニブ又はイリノテカンであることを特徴とする上記(8)記載の癌幹細胞における薬物耐性の低減剤。
(10)以下の(I)又は(II)のいずれかを有効成分とする、癌幹細胞における転移能の抑制剤。
(I)RAB3B遺伝子の発現を抑制する核酸分子;
(II)配列番号1に示されるRAB3Bタンパク質に結合する抗体若しくは低分子化合物;
(11)癌幹細胞が、肝癌幹細胞であることを特徴とする上記(10)記載の癌幹細胞における転移能の抑制剤。
(12)被検者から採取した生体試料中のRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を指標として、癌の転移性再発リスクを予測する方法。
(13)癌が肝癌であることを特徴とする上記(12)記載の癌の転移性再発リスクを予測する方法。
(14)生体試料が、原発肝腫瘍組織又は血液であることを特徴とする上記(12)又は(13)記載の方法。
(15)上記(12)〜(14)のいずれか記載の方法に用いるためのキットであって、以下の(I)又は(II)を含むキット。
(I)被検者から採取した生体試料中のRAB3B遺伝子のmRNAの発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を検出するためのプライマー対若しくはプローブ、又はそれらの標識物;
(II)被検者から採取した生体試料中のmRNAがコードするタンパク質の発現量を検出するための抗体、又はそれらの標識物;
(15)配列番号1に示されるRAB3Bタンパク質のアミノ酸配列のうち、連続する5-50個のアミノ酸からなるペプチドを有効成分とする癌ペプチドワクチン。
事前に摘除手術後5年以上再発無し肝癌患者と、摘除手術後1年又は2年以内転移性再発有り肝癌患者それぞれ2検体以上、好ましくは4検体以上、より好ましくは5検体以上、さらに好ましくは10検体以上から採取された生体試料中のRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を測定し、かかる発現量の中央値又は平均値等を算出し、前記中央値又は平均値等を基にカットオフ値を定める。次いで被検者から採取された生体試料中のRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を測定し、被検者から採取された生体試料中のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量と前記カットオフ値とを比較することで、被検者における肝癌の転移性再発リスクを予測することが可能である。
被検者の手術摘出標本を用いて、非癌部RAB3B mRNA発現量にする癌部RAB3B mRNA発現量の比を求め、かかる非癌部RAB3B mRNA発現量に対する癌部RAB3B mRNA発現量の比を用いることで、被検者における肝癌の転移性再発リスクを予測することが可能である。カットオフ値は、好ましくは発現比1.0、より好ましくは1.5、さらに好ましくはReceiver operating characteristic曲線におけるYouden indexによって定めた1.16である。また、事前にHCC患者2検体以上、好ましくは4検体以上、より好ましくは5検体以上、さらに好ましくは10検体以上から採取された生体試料中の癌部及び非癌部それぞれのRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を測定し、非癌部RAB3B mRNA発現量にする癌部RAB3B mRNA発現量の比を求め、かかる発現量の比の中央値又は平均値等を算出し、前記中央値又は平均値等を基にカットオフ値を定めても良い。
被検者から採取された生体試料中のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量、及び、摘除手術後5年以上再発無し肝癌者と、摘除手術後1年又は2年以内転移性再発有り肝癌患者それぞれ2検体以上、好ましくは4検体以上、より好ましくは5検体以上、さらに好ましくは10検体以上から採取された生体試料中のRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量とを組み合わせた判別式をロジスティック回帰分析等の統計解析により構築して予測する方法を挙げることができる。
(I)被検者から採取した生体試料中のRAB3B遺伝子のmRNAの発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を検出するためのプライマー対若しくはプローブ、又はそれらの標識物;
(II)被検者から採取した生体試料中のmRNAがコードするタンパク質の発現量を検出するための抗体、又はそれらの標識物;
を含んでいれば特に制限されず、上記キットには、必要と目的に応じた緩衝液、pH調製剤、反応容器、癌の転移性再発リスクを予測するための説明書等をさらに備えたものであってもよい。
例示に限定されるものではない。
(SK-HEP-1細胞株からの浮遊細胞塊の誘導)
本発明者らが以前開示した癌幹細胞誘導法(上記特許文献4)を用い、肝癌由来の肝癌細胞株SK-HEP-1より、浮遊細胞塊(sphere)形成能を有する癌幹細胞を誘導した。具体的には以下に示すとおりである。

200μg/mL ヒトEGF(シグマ−アルドリッチ社製) 10μL

NSF-1(50倍濃度)(カンブレックス社製) 2mL(最終濃度;2%[w/v])
60mg/mL N−アセチルシステイン(N-acetylcysteine)(シグマ−アルドリッチ社製) 100μL
SK-HEP-1細胞株を1.0×105個/mLとなるように10%ウシ胎児血清(FBS:fetal bovine serum)を含むDMEM培地(以下、単に「血清含有DMEM培地」という)に懸濁した後、培養シャーレに播種し、37℃、5% CO2条件下で3日間培養した。血清含有DMEM培地で培養したSK-HEP-1細胞株(以下、「SK-HEP-1」ともいう)は培養シャーレに接着する接着性の癌細胞株である。
高分化型肝細胞癌由来の細胞株であるHuH7細胞株を、上記NSC含有無血清培地、又は上記血清含有DMEM培地で培養した。具体的には、HuH7細胞株を1.0×105個/mLとなるように上記NSC含有無血清培地又は上記血清含有DMEM培地に懸濁した後、培養シャーレに播種し、37℃、5%CO2条件下でそれぞれ7日間又は3日間培養した。上記NSC含有無血清培地及び上記血清含有DMEM培地で培養したHuH7細胞株(それぞれ、以下「HuH7/NSC」、「HuH7/DMEM」ともいう)はいずれも培養シャーレに接着し、浮遊細胞塊は形成されなかった。したがって、NSC含有無血清培地を用いたとしても、低分化型肝癌由来のSK-HEP-1細胞株を培養した場合には浮遊細胞塊を形成するのに対し、低分化型肝癌由来のHuH7細胞株を培養した場合には浮遊細胞塊を形成しないことが明らかとなった。なお、本発明者らにより、上記非特許文献4により、NSC含有無血清培地を用いたとしても、低分化型肝癌由来のHLE細胞株を培養した場合には浮遊細胞塊を形成するのに対し、低分化型肝癌由来のHep3B細胞株を培養した場合には浮遊細胞塊を形成しないことも明らかにされている。
[実施例2]
(転移性肝内再発に特異的な遺伝子の同定)
実施例1で得られたSK-sphere、SK-HEP-1、HuH7/NSC、HuH7/DMEMそれぞれの肝細胞癌由来細胞、及び摘除手術後5年以上肝内再発無しHCC患者(HCC without IHR(≧5year))若しくは摘除手術後1年以内転移性肝内再発ありHCC患者(HCC with IHR(≦1year))からの摘除標本における癌部組織及び周辺非癌部組織のmRNA発現を解析することで、原発巣除去後の転移性肝内再発に特異的な遺伝子の同定を行った。
miRNeasy(QIAGEN社製)を用いて、実施例1で作製したSK-sphere、SK-HEP-1、HuH7/NSC、HuH7/DMEMそれぞれの癌由来細胞株、及び摘除手術後5年以上肝内再発無しHCC患者(HCC without IHR(≧5year))及び摘除手術後1年以内転移性肝内再発有りHCC患者(HCC with IHR(≦1year))からの摘除原発巣標本(それぞれn=5)における癌部組織からtotal RNAを抽出した。
TruSeq Stranded Total RNA with Ribo-Zero Gold LT Sample Prep kit(イルミナ社製)及びNextSeq500(イルミナ社製)を用いて、そのプロトコルに従ってribosomal RNA除去後にRNAシークエンスを行い、26,475遺伝子の網羅的なmRNA発現解析を行った。NextSeq 500より得られたfastqファイルからcutadaptによるデータトリミング、FastQCによるクオリティ確認を経てSTARによるhg38ヒトリファレンスゲノムへのマッピングを行った。26,475遺伝子それぞれにおけるリードカウント数について、R softwareにおけるTCCパッケージを用いて正規化及び群間比較を行った。群間比較に先立って正規化後に何れの分においても発現量が低い(read count = 10)遺伝子は除外した。発現変動遺伝子 (Differentially expressed Genes; DEGs) の検出にはGLM LRT法を用いた。解析にあたっては、(a)SK-sphereとSK-HEP-1、(b)SK-sphereとHuH7/NSC、(c)HCC with IHR(≦1year) vs HCC without IHR(≧5year)におけるmRNAの発現量を比較して、転移性肝内再発有り群及びSK-sphereに共通して発現亢進を示す遺伝子候補を選択した。選択基準は(Fold-change)2.0、q値〈0.05)とした。q値は、FDR多重検定(multiple-testing correction)により算出した。
さらに、上記それぞれの発現量解析の結果を統合してMA plot解析(SK-sphere vs SK-HEP-1及びShort-DFS(Disease-free survival)vs Long-DFS in Tumor)を行い、SK-sphere及び転移性肝内再発有り群に共通して発現亢進を示す遺伝子の探索を行った。肝癌におけるDFSは無再発生存期間を意味し、RFS (relapse-free survival) と同義である。上記遺伝子の探索手法の概略を図2に、MA plot解析の結果を図3及び4に示す。図3及び4中、横軸はSK-sphereとSK-HEP-1での総カウント数 (mRNAの発現量に相当)又はShort-DFSとLong-DFSでの総カウント数のLog2値、縦軸はSK-sphere/SK-HEP-1又はShort-DFS/Long-DFS発現比のLog2値を表す。
(mRNA発現レベルの比較及び生存曲線解析)
1.RNA−シークエンス
実施例3におけるmRNAの発現解析によるSK-sphere、SK-HEP-1、HuH7/NSC、HuH7/DMEMそれぞれの癌由来細胞株からのRAB3B mRNA発現をまとめたグラフを図5A(a)に示す。図5A(a)に示すように、SK-sphereはSK-HEP-1と比較して2.3倍もRAB3BのmRNAが発現していることが明らかとなった。一方、HuH7においては、培養条件にかかわらずRAB3BのmRNA発現レベルはSK-HEP-1におけるRAB3BのmRNA発現レベルよりも低かった。
定量PCRによりSK-sphere、SK-HEP-1、HuH7/NSC、HuH7/DMEMそれぞれの癌由来細胞株、及び摘除手術後5年以上肝内再発無しHCC患者(HCC without IHR(≧5year))若しくは摘除手術後1年以内転移性肝内再発有りHCC患者(HCC with IHR(≦1year))からの摘除標本(それぞれn=5)における癌部組織からのRAB3B mRNA発現量を解析した。定量PCR解析は、以下の方法で行った。
術後無再発生存期間解析として、上記RAB3B mRNA発現に基づいてHCC患者20例の群分けを行い、Kaplan-Meier生存曲線及びLog-Rank検定によるP値を示した。結果を図5Cに示す。群分けのカットオフ値として、非癌部におけるRAB3B mRNA発現量に対する癌部RAB3B mRNA発現量の比が1.0, 1.16, 1.5 , 2.0としたものをそれぞれ図5C(a)-(d)に示す。カットオフ値1.16はReceiver operating characteristic曲線におけるYouden indexを用いて得た。また、Cox回帰解析からのハザード比 (HR) 及びP値を各グラフの左下に記した。
(siRNAによるノックダウン解析)
SK-HEP-1細胞に対して、Lipofectamine RNAiMAX Transfection Reagent (Thermo Fisher Scientific社製) を用いてreverse transfection法により以下のsiRNAsをトランスフェクションした。siRNAとして、配列番号2(Sense 5’ to 3’: GCUUCAUUCUGAUGUAUGAtt)に示されるセンス配列と配列番号3(Antisense 5’ to 3’: UCAUACAUCAGAAUGAAGCcc)に示されるアンチセンス配列とから形成されるsiRAB3B#1、又は、配列番号4(Sense 5’ to 3’: GCUAUGCUGAUGACACGUUtt))に示されるセンス配列と配列番号5(Antisense 5’ to 3’: AACGUGUCAUCAGCAUAGCgg)に示されるアンチセンス配列とから形成されるsiRAB3B#2の塩基配列、及びコントロールsiRNA (siNegaCont: Silencer(登録商標) Select Negative Control No.1 siRNA; Thermo Fisher Scientific社製)を用いた。Reverse transfection法には以下の条件を適用した。1x105 cells/ 5nM each siRNA/1 μL Lipofectamine RNAiMAX Transfection Reagent/ 1mL Opti-MEM I Reduced Serum培地 (Thermo Fisher Scientific社製)。結果を図6(a)、(b)、図7、図8に示す。
(抗癌剤に対する感受性解析−1)
上記実施例5の結果を踏まえて、SK-HEP-1細胞株をNSC含有無血清培地又は血清含有DMEM培地で培養し、RAB3B遺伝子の発現をsiRNAによりノックダウンした場合の抗癌剤感受性を解析した。抗癌剤としては、ドキソルビシン(Dxorubicin)、ドセタキセル(Docetael)、カルボプラチン(Carboplatine)、ボリノスタット(ヒストン脱アセチル化酵素(HDAC)阻害剤(Suberoylanilide hydroxamic acid:SAHA))、スニチニブ(Sunitinib)、イリノテカン(Irinotecan)を用いた。抗癌剤感受性はMTS(3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホフェニル)−2H−テトラゾリウム)アッセイ(上記非特許文献1)と同様の方法により評価した。簡潔に述べると、実施例5に示す条件にてReverse transfectionを行い、96-well plateに50μL (5,000 cells)/well播種し、翌日にDMEM培地又はNSC培地と交換した。培地交換後1日又は2日後に抗癌剤含有培地を添加し、さらに24時間後にMTS試薬を添加し、その2時間後に吸光度を測定した。細胞の培養は37℃、5%CO2条件下で行った。試薬製造者の指示に従い、MTS染料の吸収を測定することにより、細胞の増殖を評価した。吸収は、Wallac 1420 ARVOsx Multilabel Counter (Perkin-Elmer社製)を用いて行った。実験は2回の独立系で、夫々、4穴複製からの値を用いた。
[実施例7]
microRNAは配列依存的に複数の遺伝子発現を制御するnon-coding RNAである。近年、microRNAは細胞内だけでなく、エクソソーム小胞に内包され細胞外に分泌されることも知られるようになった。すなわち、microRNAは細胞内における一つの遺伝子発現制御だけでなく、他の細胞における遺伝子発現をも制御しうる。まず、SK-HEP-1とSK-sphereにおける培地中のRNA濃度を調べたところ、SK-sphereはSK-HEP-1と比較してRNA濃度が上昇していた(図10(a))。そのため、CSLCにおいてはmicroRNAによって癌微小環境や周辺細胞の制御が考えられた。
(抗癌剤に対する感受性解析−2)
SK-HEP-1細胞株をNSC含有無血清培地又は血清含有DMEM培地で、has-miR-15a-5のmimic RNA又はコントロールRNAを加えて培養し、実施例6と同様の方法で抗癌剤感受性を解析した。抗癌剤としては、ドキソルビシン(Dxorubicin)、ドセタキセル(Docetael)、カルボプラチン(Carboplatine)、ヒストン脱アセチル化酵素(HDAC)阻害剤(Suberoylanilide hydroxamic acid:SAHA)、スニチニブ(Sunitinib)、イリノテカン(Irinotecan)を用いた。結果を図11A-Fに示す。
(CRISPR/Cas9によるノックアウト解析)
CRISPR/Cas9を用いたゲノム編集技術を用いて、SK-HEP-1株を親株としてmono-allelic RAB3B knock-out株を樹立した。ゲノム編集にはCas9 mRNA及び配列番号9に示す(-)鎖配列(GTTTCACCCGCTTCTCGTGA)及び続くCGGをそれぞれ標的配列及びPAM配列とし、配列番号10に示す配列 (GUUUCACCCGCUUCUCGUGA guuuuagagcuagaaauagcaaguuaaaauaaggcuaguccguuaucaacuugaaaaaguggcaccgagucggugcuuuu) をguide RNAとして合成し、guide RNAをlipofectamine RNAi Max(Thermo Fisher Scientific社製)によりSK-HEP-1細胞株に移入した後にsingle cell cloningを行った。得られたcloneからgenome DNAを抽出し、サンガーシーケンスにより変異導入を確認した。上記guide RNAとCas9 mRNAを用いてゲノム編集を行うことで、図12に示されるように、SK-HEP-1細胞株におけるgenome DNA中のPAM配列に隣接する標的配列に二本鎖切断が起こり、結果として配列番号1に示されるRAB3Bのアミノ酸配列が、配列番号11に示されるアミノ酸配列へと変異したクローンが得られた。
免疫不全マウスNOD-Rag1null IL2r null double mutant mice (NRG mice)へ細胞を脾臓注入することによって、肝腫瘍形成頻度を調べた。HBSSに懸濁した1000個のSK-HEP-1細胞株又はRAB3B-KO8細胞株をマウス脾臓に注射し、脾臓を摘出したうえで閉腹し、8週後に肝腫瘍の有無を確認した。培地は血清含有DMEM培地(normal)又はsphere誘導のNSC培地(NSC)を用いた。結果を図16に示す。図中、yesは肝腫瘍有りのマウスの数、noは肝腫瘍無しのマウスの数である。また、上段がSK-HEP-1細胞株を注射した場合、下段がRAB3B-KO8細胞株を注射した場合である。図16に示すように、SK-HEP-1細胞株ではsphere誘導のNSC培地にて7日間処置すると肝腫瘍形成頻度が向上していたが、RAB3B-KO8細胞株ではsphere誘導のNSC培地にて7日間処置しても肝腫瘍形成頻度の向上は認められなかった。すなわち、RAB3B-KO8細胞株は癌幹細胞における転移性が抑制されていることが明らかとなった。
(抗癌剤に対する感受性解析−3)
実施例6同様に、RAB3B-KO8細胞株を用いてMTSアッセイにより抗癌剤に対する感受性解析を行った。SK-HEP-1細胞株又はRAB3B-KO8細胞株を、各抗癌剤に24時間暴露後の生存率を調べた結果を図17に示す。培地は血清含有DMEM培地(normal)又はsphere誘導のNSC培地(NSC)を用いた。図17に示すように、NSC培地においてSK-HEP-1細胞株と比較してRAB3B-KO8細胞株での抗癌剤耐性の有意な減弱が認められた。したがって、RAB3B遺伝子の発現を抑制することで、癌幹細胞における抗癌剤耐性を低減することが可能であることが確認された。
(細胞周期における影響)
SK-HEP-1細胞株又はRAB3B-KO8細胞株を70%エタノールにより固定した後に、RNase処理及びPIによるDNA標識を行い、フローサイトメーターを用いた、各細胞周期における細胞集団の割合を求めた。結果を図18に示す。図18に示すように、SK-HEP-1株ではNSC培地でのsphereを誘導によってG0/G1期停止が見られたのに対して、RAB3B-KO8細胞株ではnormal培地での培養によるS期の増加とNSC培地でのsphereを誘導によるG2/M期停止が観察された。したがって、癌幹細胞においてRAB3B遺伝子の発現を抑制することで、G0/G1期停止が解除されることが確認された。
(ABCG2タンパク質の発現)
RAB3Bを標的とする薬物耐性の原理を検討する中で、多剤耐性を担う薬剤排出ポンプとして働くABCG2タンパクの発現に着目し、かかるABCG2タンパクの発現をフローサイトメトリーにより解析した結果を図19に示す。図19に示すように、RAB3B-KO8細胞株ではABCG2陽性細胞割合が減少していた。したがって、癌幹細胞においてRAB3B遺伝子の発現を抑制することで、癌細胞における薬剤排出機能が低減し、その結果薬物耐性が低減していることが明らかとなった。
Claims (16)
- 以下の(I)又は(II)のいずれかを有効成分とする、癌幹細胞における薬物耐性の低減剤。
(I)RAB3B遺伝子の発現を抑制する核酸分子;
(II)配列番号1に示されるRAB3Bタンパク質に結合する抗体若しくは低分子化合物; - 癌幹細胞が、肝癌幹細胞であることを特徴とする請求項1記載の癌幹細胞における薬物耐性の低減剤。
- 核酸分子がRAB3B遺伝子を標的とする二本鎖siRNA、microRNA、shRNA、アンチセンス核酸、及びリボザイムからなる群から選択される少なくとも1種の核酸分子であることを特徴とする請求項1又は2記載の癌幹細胞における薬物耐性の低減剤。
- 核酸分子がRAB3B遺伝子を標的とする二本鎖siRNAであることを特徴とする請求項3記載の癌幹細胞における薬物耐性の低減剤。
- 薬物が、抗腫瘍性抗生物質、微小管阻害剤、プラチナ製剤、ヒストン脱アセチル化酵素(HDAC)阻害剤、又はキナーゼ阻害剤であることを特徴とする請求項4記載の癌幹細胞における薬物耐性の低減剤。
- 薬物が、ドキソルビシン、ドセタキセル、カルボプラチン、ボリノスタット、又はスニチニブであることを特徴とする請求項5記載の癌幹細胞における薬物耐性の低減剤。
- 核酸分子がRAB3B遺伝子を標的とするmicroRNAであって、該microRNAがhas-miR-15a-5pであることを特徴とする請求項3記載の癌幹細胞における薬物耐性の低減剤。
- 薬物が、抗腫瘍性抗生物質、微小管阻害剤、プラチナ製剤、ヒストン脱アセチル化酵素(HDAC)阻害剤、キナーゼ阻害剤、又はトポイソメラーゼ阻害剤であることを特徴とする請求項7記載の癌幹細胞における薬物耐性の低減剤。
- 薬物が、ドキソルビシン、ドセタキセル、カルボプラチン、ボリノスタット、スニチニブ又はイリノテカンであることを特徴とする請求項8記載の癌幹細胞における薬物耐性の低減剤。
- 以下の(I)又は(II)のいずれかを有効成分とする、癌幹細胞における転移能の抑制剤。
(I)RAB3B遺伝子の発現を抑制する核酸分子;
(II)配列番号1に示されるRAB3Bタンパク質に結合する抗体若しくは低分子化合物; - 癌幹細胞が、肝癌幹細胞であることを特徴とする請求項10記載の癌幹細胞における転移能の抑制剤。
- 被検者から採取した生体試料中のRAB3B遺伝子のmRNAの発現量、前記mRNAがコードするタンパク質の発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を指標として、癌の転移性再発リスクを予測する方法。
- 癌が肝癌であることを特徴とする請求項12記載の癌の転移性再発リスクを予測する方法。
- 生体試料が、原発肝腫瘍組織又は血液であることを特徴とする請求項12又は13記載の方法。
- 請求項12〜14のいずれか記載の方法に用いるためのキットであって、以下の(I)又は(II)を含むキット。
(I)被検者から採取した生体試料中のRAB3B遺伝子のmRNAの発現量、又はRAB3B遺伝子を標的とするmicroRNAの発現量を検出するためのプライマー対若しくはプローブ、又はそれらの標識物;
(II)被検者から採取した生体試料中のmRNAがコードするタンパク質の発現量を検出するための抗体、又はそれらの標識物; - 配列番号1に示されるRAB3Bタンパク質のアミノ酸配列のうち、連続する5-50個のアミノ酸からなるペプチドを有効成分とする癌ペプチドワクチン。
