Description
本出願は、2015年9月24日に出願された米国仮出願第62/222,983号に関し、35 U.S.C 119の下でその優先権の利益を主張する。当該仮出願の内容は、参照によりその全体が本明細書に組み込まれる。
本出願は、EFS−Webを介して提出され、かつ参照によりその全体が本明細書に組み込まれる配列表を含む。2016年8月11日に作成された、該ASCIIの複写は、P33079−WO_SL.TXTという名称であり、16.4kbの大きさである。
「てんかん」という用語は、本明細書で使用されるとき、個体が2回以上の非誘発性けいれんを有する臨床的現象を指す。てんかんは、例えば、全般発作けいれん及び焦点発作けいれん(症候性及び特発性)を含み、これには、小児欠神てんかん、若年性ミオクロニーてんかん、起床時の大発作を伴うてんかん、側頭葉てんかん、前頭葉てんかん、頭頂葉てんかん、後頭葉てんかん、ならびに大田原症候群、ウエスト症候群、ドラベ症候群、ミオクロニー脱力発作を伴うてんかん、及びレノックス・ガストー症候群を含むてんかん性脳症が含まれる。
一態様において、本発明は、FGF21受容体活性化因子がてんかんの動物モデルにおいて有効性を実証するという観察に部分的に基づく。したがって、FGF21受容体を活性化する薬剤を投与することによる、てんかんを有する個体の治療方法が提供される。
以下は、本発明の方法及び組成物の実施例である。上述の概要を得ると、様々な他の実施形態が実施され得ることが理解される。

角膜キンドリングモデルを使用して、抗FGFR1cアゴニスト抗体のけいれんに対する効果を試験した(このモデルについては、Rowley,N.M.and H.S.White,Comparative anticonvulsant efficacy in the corneal kindled mouse model of partial epilepsy:Correlation with other seizure and epilepsy models.Epilepsy Res,2010.92(2−3):p.163−9、Matagne,A.and H.Klitgaard,Validation of corneally kindled mice:a sensitive screening model for partial epilepsy in man.Epilepsy Res,1998.31(1):p.59−71に記載される)。成体オスCF1マウス(1群あたりn=8、18〜25g)を 5回の連続的な二次性全般発作の基準までキンドリングを形成した(kindled)(Racine,R.J.,Modification of seizure activity by electrical stimulation:II.Motor seizure.Electroenceph.Clin.Neurophysiol.,1972.32:p.281−294に記載されるステージ4または5)。1日2回、0.5%塩酸テトラカイン溶液を各眼に適用し、角膜電極(3mA、60Hz、3秒間)を通して視神経を刺激した。1日2回の角膜刺激を受けた後、CF1マウスは典型的に、およそ10〜14日目で初回のステージ5けいれんに達した。マウスが、発明者らが「キンドリングが完成した」とみなした5回の連続的なステージ5けいれんの基準に達するまで、1日2回の刺激を各マウスに対して継続した。群内のすべての他のマウスのキンドリングが完成されるまで、キンドリングが完成したマウスを次いで1日おきから2〜3日おきに刺激した。
Claims (34)
- てんかんの治療用の医薬品の製造におけるFGF21受容体活性化因子の使用。
- 前記FGF21受容体活性化因子は、FGF21、抗FGFR1c抗体、抗KLB抗体、及び二重特異性抗FGFR1c/KLB抗体からなる群から選択される、請求項1に記載の使用。
- 前記FGF21受容体活性化因子は、FGF21である、請求項2に記載の使用。
- FGF21は、異種分子に複合されている、請求項3に記載の使用。
- 前記異種分子は、PEGである、請求項4に記載の使用。
- 前記異種分子は、ポリペプチドである、請求項4に記載の使用。
- 前記ポリペプチドは、抗体Fcである、請求項6に記載の使用。
- 前記抗体はIgG1である、請求項7に記載の使用。
- 前記FGF21受容体活性化因子は、抗FGFR1c抗体である、請求項2に記載の使用。
- 前記抗FGFR1c抗体は、KLHAVPAAKTVKFKCP(配列番号3)及びFKPDHRIGGYKVRY(配列番号4)からなる群から選択されるペプチドに結合する、請求項9に記載の使用。
- 前記FGF21受容体活性化因子は、抗KLB抗体である、請求項2に記載の使用。
- 前記抗KLB抗体は、前記抗KLB抗体は、16H7及びh5h23からなる群から選択される、請求項11に記載の使用。

- 前記二重特異性抗FGFR1c/KLB抗体は、アミノ酸配列SSPTRLAVIPWGVRKLLRWVRRNYGDMDIYITAS(配列番号5)からなるKLBの断片内のKLBエピトープに結合する、請求項13に記載の使用。
- 前記二重特異性抗FGFR1c/KLB抗体は、YW182.5 YGDYからのアミノ酸配列を含む抗FGFR1cアーム及び抗8C5.K4.M4L.H3.KNVからのアミノ酸配列を含む抗KLBアームを含む、請求項14に記載の使用。
- 前記医薬品は、皮下投与される、請求項1に記載の使用。
- 前記医薬品は、レベチラセタム(「KEPPRA(商標)」)、レベチラセタム持続放出(XR)(「KEPPRA XR(商標)」)、ラモトリギン(「LAMICTAL(商標)」)、ラモトリギンXR(「LAMICTAL XR(商標)」)、オキシカルバゼピン(oxycarbazepine)(「TRILEPTAL(登録商標)」)、カルバマゼピン(「TEGRETOL(登録商標)」)、ラコサミド(「VIMPAT(登録商標)」)、バルプロ酸(「VPA」)、及びペランパネル(「FYCOMPA(登録商標)」)からなる群から選択される1つ以上の追加の治療薬との併用投与のためのものである、請求項1に記載の使用。
- 個体におけるてんかんの治療方法であって、前記個体に有効量のFGF21受容体活性化因子を投与することを含む、前記方法。
