アンジオテンシンIIIに特異的な受容体を探して、本発明者らは以前に、ラット脳cDNAライブラリーから7回膜貫通ドメインを有するGタンパク質共役受容体(GPCR)をコードする遺伝子を単離した(1)。この受容体のアミノ酸配列は、ラットアンジオテンシン受容体1型(AT1受容体)のアミノ酸配列と31%同一であり、O'Dowdらにより以前にクローニングされたヒトオーファン受容体APJのアミノ酸配列と90%同一であった(2)。ヒトAPJ受容体の内因性リガンドは、Tatemotoらにより発見され(3)、アペリンと名付けられた。アペリンは、アミノ酸77個のより大きな前駆体、プレプロアペリンから生成するアミノ酸36個のペプチド(アペリン36)である。哺乳類でのプレプロアペリンのアミノ酸配列アライメントにより、アペリン−17又はK17Fとして公知のC末端アミノ酸17個の厳密な保存が実証された。アペリンのいくつかの分子形態:インビボ型アペリン36、K17F及びピログルタミル形態のアペリン13(pE13F)が同定されている(4〜8)。

アペリンは迅速に代謝されるので(血液循環中のK17Fの半減期:40秒、個人的なデータ)、アペリン/アペリン受容体経路を活性化する代謝的に安定なアペリン類似体が、心不全を有する患者でのアペリンシグナル伝達を増加させる治療潜在性を決定するために必要とされる。この目的で、本発明者らは、アペリン13(pE13F)及びアペリン17(K17F:配列番号:1:KFRRQRPRLSHKGPMPF)の構造活性相関研究を行った。本発明者らは、代謝的に安定なアペリン類似体(P92化合物及びJFM V−0196B化合物)を得たが、それらのうち最も強力なのは化合物P92であった。この化合物は、[125I]pE13Fを用いた競合放射性リガンド結合アッセイ法により決定された0.2±0.06nMというKiをラットアペリン受容体に対して示した。AT1受容体に対するその選択性は、アペリン受容体と比較して100倍である。この化合物は、フォルスコリンにより誘導されるcAMP産生阻害に対して、及びアペリン受容体のインターナリゼーションに向けて全アゴニストとして挙動する。絶水マウスにおける漸増する用量のP92の脳室内(i.c.v.)注射は、K17Fのi.c.v.注射により誘導されるよりも高い効力で血漿AVPレベルに用量依存的減少を誘導した。そのうえ、P92は、ノルアドレナリンを用いて予備狭窄させた大動脈輪又はアンジオテンシンIIにより予備収縮させた糸球体細動脈の血管弛緩を、K17Fにより誘導される血管弛緩に類似して誘導した。麻酔された正常血圧ラットにP92を漸増する用量で静脈内経路により注射すると、動脈圧(BP)が用量依存的に減少する。分離された潅流ラット心臓に適用されたP92は、心収縮性を増加させる。さらに、本発明の化合物(P92化合物及びJFM V−0196B化合物)は、血漿中で半減期の安定性の増加を示す。

本発明の別の態様は、対象への投与のために調製された医薬組成物であって、上記のような本発明の代謝的に安定なアペリン類似体の1つ以上の治療有効量を含む医薬組成物を含む。代謝的に安定なアペリン類似体の治療有効量は、投与経路、対象である哺乳類のタイプ及び処置されようとする対象の身体的な特徴に依存する。考慮できる具体的な要因には、疾患の重症度及び時期、体重、食事及び同時の投薬が含まれる。開示された化合物の治療有効量を決定するためのこれらの要因の関係は、当業者により理解されている。
上に定義される代謝的に安定なアペリン類似体及び本発明の医薬組成物は、アペリン受容体により媒介される疾患、特に心血管疾患(心不全、腎不全、高血圧症、肺高血圧症)、多発性嚢胞腎疾患、低ナトリウム血症及びSIADHを処置するために使用される。
− 心血管疾患:心不全、腎疾患(例えば腎不全、腎炎など)、高血圧症、肺高血圧症、硬変、動脈硬化症、肺気腫、肺水腫;脳卒中、脳虚血、敗血症における心筋障害
− 神経原性糖尿病(例えば、糖尿病性腎症、糖尿病性神経障害、糖尿病性網膜症などの糖尿病性合併症)のような病理を含めた抗利尿ホルモン不適切分泌症候群(SIADH)、敗血症性ショック、口渇トラブル;
− 代謝疾患:肥満、食欲不振、過食症、大食症、高コレステロール血症、高トリグリセリド血症、高脂血症;
− 老人性認知症、脳血管性認知症、家系性変性疾患(genealogical denaturation degenerative disease)(例えば、アルツハイマー病、パーキンソン病、ピック病、ハンチントン病など)が原因の認知症、感染症(例えば、クロイツフェルトヤコブ病などの遅発性ウイルス感染)に起因する認知症、内分泌疾患、代謝疾患、又は中毒(例えば甲状腺機能低下症、ビタミンB12欠乏症、アルコール依存症、様々な薬物、金属、若しくは有機化合物により起こる中毒)に関連する認知症、腫瘍(例えば脳腫瘍)により起こる認知症、及び外傷性疾患(例えば慢性硬膜下血腫)が原因の認知症などの様々なタイプの認知症、抑うつ、過活動児童症候群(小頭症)、意識障害、不安障害、統合失調症、恐怖症;

I − 薬物及び放射性リガンド
1)K17F、pE13F及びそれらの誘導体をそれぞれPolyPeptide Laboratories(Strasbourg, France)及びGL Biochem(Shangai, China)が合成した。125I−pE13F(Bolton Hunterによりリシン8でヨウ素化されている)をPerkin Elmer(Wellesley, MA, USA)から購入した。
2)アペリン−13から誘導されたアルキルペプチド及びパーフルオロアルキルペプチドの合成を、Pr M. Hibert、Dr. D. Bonnet及びDr. JF Margatheにより指揮されるTherapeutic Innovationの研究所で行った。
Applied Biosystem ABI 433A合成装置(Appelar, France)を使用してFmoc−L−Phe−Wang樹脂(276mg、0.37mmol/g)上での標準自動化SPPSによりアペリン(62−77)配列を合成した。溶媒としてN,N−ジメチルホルムアミド(DMF)中でカップリング試薬として2−(1H−ベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルウロニウムヘキサフルオロリン酸(HBTU)、1−ヒドロキシベンゾトリアゾール(HOBt)、及びジイソプロピルエチルアミン(Huenig塩基)(DIPEA)を使用して10倍過剰のFmoc−L−アミノ酸誘導体のカップリングにより伸長を実施した。各カップリング段階の後に、ピペリジンによる処理に続く301nmのUVにより、Fmocの脱保護を行った。溶媒としてDMF中で、カップリング試薬としてHBTU、HOBt、及びDIPEAを使用して、5倍過剰のFmoc−L−Lys(Boc)−OH又はBoc−L−Lys(Fmoc)−OHのいずれかをカップリングさせることにより、Lys(61)を手作業で導入した。C18 Ascentis Express(2.7μm、4.6mm×75mm)を用いて、直線勾配(7.5分かけて溶媒A中に溶媒Bが5%から100%、流速1.6mL・min-1、検出220nm;溶媒A:水/0.1% TFA;溶媒B:アセトニトリル/0.1% TFA)を使用して分析用逆相高速液体クロマトグラフィー(RP−HPLC)分離を行った。Waters XBridge RP-C18カラム(5μm、19×100mm)を用いて、直線勾配(溶媒A中の溶媒B;溶媒A:水/0.1% TFA;溶媒B:アセトニトリル/0.1% TFA;流速20mL.min-1;220nmで検出)を使用して、半分取逆相高速液体クロマトグラフィー(RP−HPLC)分離を行った。精製された化合物は、下に示される保持時間(tR)で単一ピークで対称のピークとして溶出した(それにより純度≧95%が確認された)。Bruker MicroTof質量分析計を用いて、エレクトロスプレーイオン化(ESI)及び飛行時間型分析計(TOF)を使用して、高分解能質量スペクトル(HRMS)を取得した。
Fmoc−L−Lys(Boc)−Ap(62−77)−Wang樹脂(1)又はBoc−L−Lys(Fmoc)−FR(Pbf)R(Pbf)QR(Pbf)PR(Pbf)LS(tBu)H(Trt)K(Boc)GPMPF−Wang樹脂(2)(1当量)をDMF中で膨潤させ、過剰な溶媒を濾過により除去した。ピペリジンのDMF溶液(20%v/v-1mL)を添加し、この混合物を室温で15分間振盪した。溶液を排出し、操作を15分間繰り返した。溶液を排出し、樹脂をDMF及びCH2Cl2で洗浄した。別のバイアルの中、DIPEA(5当量)をヘキサデカン酸又は4,4,5,5,6,6,7,7,8,8,9,9,10,10,11,11−ヘプタデカフルオロウンデカン酸(2当量)、HBTU(2当量)、及びHOBt(2当量)のDMF溶液(1mL)に添加した。この混合物を室温で1分間撹拌し、樹脂に添加した。この混合物を室温で90分間撹拌した。溶液を排出し、この手順を90分間繰り返した。溶液を排出し、樹脂をDMF、CH2Cl2、及びジエチルエーテルで洗浄し、次に真空中で乾燥させた。乾燥した樹脂をTFA/フェノール/チオアニソール/1,2−エタンジチオール/Me2S/水/NH4I、81/5/5/2.5/2/3/1.5(試薬H、2mL)で処理し、この混合物を室温で3時間振盪した。溶液を収集し、ビーズをTFAで洗浄した。溶液を真空中で蒸発させ、粗生成物を半分取RP−HPLCにより精製した。予期される生成物が凍結乾燥により産出した。

CHO−K1(American Type Culture Collection; Rockville, MD, USA)細胞を、10%ウシ胎児血清、0.5mM グルタミン、100ユニット/mL ペニシリン及び100μg/mL ストレプトマイシン(全てInvitrogen, Carlsbad, CA, USAから)を補充したHamのF12培地中で維持した。Lipofectamine 2000(Invitrogen)を使用して、細胞に野生型アペリン受容体−EGFPをコードするプラスミドをトランスフェクトし、以前に記載されたように安定細胞系を樹立した(42)。
野生型ラットアペリン受容体−EGFPを安定発現しているCHOからの粗膜調製物を、以前に記載されたように調製した(39)。単独又は様々な濃度(10−14M〜10−4M)のアペリン若しくはその類似体の存在下の結合緩衝液(50mM Hepes、5mM MgCl2、1% BSA、pH7.4)中で、膜調製物(膜タンパク質総質量0.5〜300μg/アッセイ)を2.10−10M 125I−pE13F(PerkinElmer Life Sciences)と共に20℃で60分間インキュベートした。氷冷した結合緩衝液を添加することにより反応を停止させ、ガラスマイクロファイバーフィルター(Whatman GF/Cフィルター)を通して濾過した。放射能をWizard 1470 Wallacガンマカウンター(Perkin Elmer, Turku, Finland)で計数した。
ホモジニアス時間分解蛍光(HTRF)技法に基づくcAMP dynamic 2アッセイキット(Cisbio Bioassays, Codolet, France)を使用してcAMPを定量した。刺激緩衝液(HBSS、5mM Hepes、0.1% BSA安定化剤、1mM IBMX、pH7.4)中で刺激を行った。簡潔には、ラットアペリン受容体−EGFPを安定発現しているCHO細胞 2,000個/ウェルを384ウェルプレートに添加し、10−6M フォルスコリン(FSK)及び漸増する濃度(10−14〜10−4M)のアペリン又はその類似体を用いて室温で30分間刺激した。次に、細胞を溶解させ、製造業者の説明書に従ってcAMPレベルを決定した。
ラットアペリン受容体−EGFPを安定発現しているCHO細胞を用いて、以前に記載されたようにインターナリゼーションアッセイを行った(43)。簡潔には、細胞を10−6M アペリン又はその類似体で処理し、それらを37℃で20分間インキュベートすることにより、インターナリゼーションをトリガーした。次に、共焦点顕微鏡分析のために細胞をAquapolymount(Polysciences, Warrington, PA, USA)中に取り付けた(詳細については(42)を参照されたい)。
K17F、pE13F、P92及びP26がERK1/2のリン酸化を誘導する能力を比較するために、野生型アペリン受容体−EGFPを安定発現しているCHO細胞を、漸増する濃度のK17F、pE13F、P92及びP26(10−11〜10−5M)で10分間処理した。次に、Alphascreen technologyによりERK1/2のリン酸化をモニタリングした。
37℃のマウス血漿中でのK17F、pE13F、P26、P92、JFM V−0196A及びJFM V−0196Bの安定性を決定した。各化合物について、原溶液(水中に100μM)を血漿に希釈して最終インキュベーション濃度5μMにした。0.1%トリフルオロ酢酸を含有する氷冷アセトニトリル 1容を添加することにより、それぞれt0及び4時間で、37℃でのインキュベーションを停止した。試料を1分間ボルテックス撹拌し、次に4℃で遠心分離した後、上清をLC−MSに注入した。Kinetex RP-C18カラム(2.6μm、100Å、50×4.6mm)を用いて、直線勾配(溶媒A中に溶媒B、溶媒A:水/0.05% TFA;溶媒B:アセトニトリル;流速2mL.min-1、358nmで検出)を使用して分析を行った。t0に対する残留被験化合物のパーセンテージを、クロマトグラムのピーク面積をモニタリングすることにより測定した。
餌及び水を自由摂取させて12時間の明暗サイクル下で雄性Sprague Dawleyラット(130〜180g BW)、雄性成Wistarラット(300〜400g)、及び雄性Swissマウス(18〜20g)を維持し、これらの動物をCharles River Laboratories(L’Arbresle, France)から入手した。実験動物の管理と使用のための最新の施設内ガイドラインに従って全ての動物実験を実施した。
以前に記載されたように、細動脈の顕微解剖のために雄性ラットの左腎を調製した(44)。実体顕微鏡観察下で糸球体細動脈を単離した。輸入細動脈及び筋肉の輸出細動脈を糸球体と共に単離し、Helouら(45)により以前に記載されたようにその形態及び腎皮質内部での局在により同定した。
これらの実験のために、糸球体に付属した輸入細動脈及び筋輸出細動脈を顕微解剖した。対照、10−9M AngII及び10−9M AngII+5.10−7M P92について3つの実験条件で1分間隔でデジタルカメラ(カメラNikon DXM1200を装着した顕微鏡LEICA DMRB)を用いて同じ細動脈の連続写真を記録した。細動脈の直径をAdobe Photoshop CSで測定した。糸球体の約100μm上流の距離で直径を測定し、各細動脈について3つ組で行った。ステージのマイクロメーターを使用して較正を行った。統計解析のために各実験条件についての平均直径を使用した。輸入細動脈と筋肉の輸出細動脈との間の細動脈の直径の差を考慮して(45)、細動脈直径の変動を、対象に対するパーセンテージで表現した。
以前に記載されたように、ラット大動脈輪を用いて実験を行った(39)。ラットに麻酔し(ペントバルビタールナトリウム、腹腔内経路により60mg/kg)、胸部大動脈を慎重に切除し、以下(mmol/L):118.3 NaCl、4.7 KCl、2.5 CaCl2、1.2 MgSO4、1.2 KH2PO4、25 NaHCO3、0.016 EDTA及び11.1 グルコースを含有する冷生理食塩水溶液(PSS)中に入れた。大動脈から過剰の結合組織及び脂肪を取り除き、長さ約3〜4mmの輪に切断した。内皮の管腔表面の損傷を避けるよう特別な注意を払った。5% CO2、95% O2の混合物を連続的に通気した,37.4℃でpH7.4のPSS 20mLを満たしたジャケット型オルガンバス20mL中に大動脈輪を懸濁した。大動脈輪の一端を組織ホルダーに接続し、他方を等尺性力変換器(EMKA Technologies, Paris, France)に接続した。輪を休止張力2gで120分間平衡化した。平衡化期間中に、輪を30分ごとに洗浄した。次に、NA(3×10−8mol/L)で予備収縮した輪における完全性又は内皮の不在をチェックするために、アセチルコリン(Ach、10−4mol/L)に対する最初の弛緩を実行した。ベースライン張力までPSSですすいだ後、輪を90分間平衡化した。この平衡期間の終わりに、NA(3×10−6mol/L)を用いた予備収縮後に、K17F(10−12〜10−4M)、pE13F(10−12〜10−4M)、P26(10−12〜10−4M)又はP92(10−12〜10−4M)に対する累積濃度反応曲線を描いた。各濃度の薬物を、先行する濃度で最大効果となったときに添加した。将来の分析(Datanalyst v2.1, EMKA Technologies, Paris)のために、濃度反応曲線をIOX v2.4(EMKA Technologies, Paris)によりPCに継続的に記録した。
Wistarラットを、100mg/kg腹腔内(i.p.)イナクチン[5−エチル−2−(1c−メチルプロピル)−2−チオバルビツール酸](RBI, IL, USA)で麻酔した。アペリン断片(K17F又はP92)をKrebs緩衝液(mM: NaCl 118.5、KCL 4.75、CaCl2 1.4、NaHCO3 24、MgSO4 1.19、KH2PO4 1.21、グルコース 11)0.2ml中に溶解させた。結果として生じた溶液を、右大腿静脈に挿入されたカテーテルを介してラットに投与し、直後にKrebs緩衝液0.2ml単独に切り替えて静脈カテーテルを洗い流した。以前に記載されたように(48)、平均動脈圧(MBP)のモニタリングのために、以前に記載されたように、右大腿静脈に追加的なカテーテルを挿入した(47)。Maclabシステム(Phymep, Paris, France)に連結したCOBE CDX III圧変換器(Phymep, Paris, France)に動脈カテーテルを接続した。血圧シグナルによりHR測定をトリガーした。実験全体にわたりBPを連続的に記録した。動脈カテーテルが圧変換器に接続された15分後に、各ラットは、アペリン断片のi.v.注射を受けた。
水を自由摂取させた覚醒マウス又は以前に記載されたように24時間絶水した覚醒マウス(4)に、K17F(1μg)及びP92(0.01μg〜1μg)をi.c.v.経路により投与した。注射の1分後に動物を屠殺し、0.3M EDTA(pH7.4)50μlを含有する冷チューブ内に体幹血(trunk blood)(0.5〜1ml)を収集した。特異的バソプレッシン−[Arg8]RIAキット(Peninsula Laboratories International Inc, San Carlo, USA)を使用することにより、以前に記載されたように(4)、血漿0.2mlからAVP濃度を決定した。
ペントバルビタールナトリウム(50mg/kg、腹腔内)で動物を麻酔した。ヘパリン(200IU/kg)を大腿静脈内に投与した。心臓を手早く切除し、以下(mM):NaCl 118.5、KCl 4.75、MgSO4 1.19、KH2PO41.2、NaHCO3 24、CaCl2 1.4及びグルコース 11を含有する氷冷Krebs-Henseleit溶液中で停止させた。次に、潅流装置に心臓を取り付け、蠕動ポンプ(Minipuls 3, model 172)を用いて一定流速6ml/minで上行大動脈を介して逆行性潅流を樹立した。pH7.4を保つために95% O2/5% CO2を通気している37℃のKrebs-Henseleit重炭酸緩衝液で心臓を潅流した。右房に挿入したサーモプローブにより温度を連続的にモニタリングした。心臓は洞調律で自発的に拍動する。国内食品ラップ製液体充填等容性バルーンを、左心耳を経由して左室内に導入し、膨張させて、8〜10mmHgの前負荷を与えた。データ取得システム(Chart V5, Powerlab 16/30, ADInstruments, UK)を経由して左室圧を連続的にコンピューターに記録した。左室圧の最大上昇速度(dP/dtmax)及び心拍数を左室圧から得た。20分の平衡時間の後に、輸液ポンプ(Harvard Apparatus Pump 11)を用いて潅流液に速度100μl/minで30分間添加した薬物により心臓を処置した。
結合実験及びcAMP実験からのデータをGraphPad Prism(GraphPad Software, La Jolla, CA, USA)で解析した。Studentの独立t検定又はDunnetの事後検定を伴う一元配置分散分析(ANOVA)を用いて統計的比較を行った。Studentの独立t検定又は加重平均に対する一元配置分散分析(ANOVA)に続くFisherの検定を使用して、カルシウム測定についての統計的な差を評価した。
1 − CHO細胞に安定発現されたラットアペリン受容体−EGFPに対するK17F、pE13F及びアペリン類似体の親和性
pE13Fをインビボ酵素分解から保護するために、本発明者らは、pE13Fの各アミノ酸をそのD−異性体又は合成アミノ酸により置き換える。本発明者らは、最初に、pE13Fのどのアミノ酸がアペリン受容体への改変ペプチドの結合に影響せずに置き換えることができるかを決定する。アペリン受容体についてのD−スキャニング実験からのKi値は、pE13Fについて0.6±0.1nMであり、pE13F(D−Arg2)、pE13F(D−Arg4)、pE13F(D−Leu5)、pE13F(D−Ser6)、pE13F(D−His7)、pE13F(D−Lys8)、pE13F(D−Ala9)及びpE13(D−Phe13)についてそれぞれ37.5±11.3nM、40.2±9.0nM、3.4±0,4nM、23.3±4.7nM、4.1±2.0nM、12.2±4.6nM、4.3±1.8nM、8.6±2.7nMであった。本発明者らは、また、Ac−R12Fについて1.4±0.7nM、pE13F(Aib7)について2.6±2.3nM、pE13F(Nle11)について0.8±0.2nM及びpE13(4Br−Phe13)について0.06±0.02nMという、アペリン受容体に対するKi値を得た。pE13FにおけるpGluの欠失並びにN−アセチルArg2、D−Leu5、Aib7、D−Ala9、Nle11及び4Br−Phe13の付加の組み合わせは、化合物P26を提供し、この化合物は、Ki値2.1±0.4nMを示した(表2)。
ラットアペリン受容体−EGFPを安定発現しているCHO細胞を、漸増する濃度のK17F、pE13F及びそれらの類似体(10−14〜10−6M)の存在下で10−6M フォルスコリンと共にインキュベーションすることで、pE13F及びK17FについてそれぞれIC50値1.0±0.2nM及び0.3±0.2nM、並びにpE13F(D−Arg2)、pE13F(D−Arg4)、pE13F(D−Leu5)、pE13F(D−Ser6)、pE13F(D−His7)、pE13F(D−Lys8)、pE13F(D−Ala9)及びpE13(D−Phe13)についてそれぞれIC50値927±312nM、419±58nM、25±9.6nM、350±180nM、317±143nM、76±10nM、43±12nM、109±27nMを有して、フォルスコリン誘導cAMP産生の濃度依存性阻害を生じた。本発明者らは、また、Ac−R12Fについて2.4±1.4nM、pE13F(Aib7)について1.2±0.6nM、pE13F(Nle11)について1.0±0.5nM及びpE13(4Br−Phe13)について0.1±0.05nMのIC50値を得た。K17F類似体P92、P95、P96の方は、K17Fに類似した阻害効力を示した(表1)。最終的に、類似体P26及びP92についてのIC50値は、それぞれ4.4±0.9nM及び0.2±0.06nMであった(表2)。
本発明者らは、また、pE13F又はK17F及びそれらの類似体がラットアペリン受容体を安定発現しているCHO細胞におけるラットアペリン受容体−EGFPのインターナリゼーションを誘導する能力を検討した。組換えアペリン受容体をそのC末端部分でEGFPによりタグ付けしたので、本発明者らは、形質膜区画から小型の細胞質蛍光小胞への蛍光の再分布を追従することにより、アペリン受容体のインターナリゼーションを視覚化することができた。アペリン受容体−EGFPを安定トランスフェクトされたCHO細胞を、それぞれ1.7及び0.26nMのEC50値を有する漸増する濃度のpE13F又はK17Fと共に20分間インキュベートすることで、形質膜での蛍光消失及び多数の細胞内蛍光小胞の出現により示されるように、アペリン受容体の進行性で顕著なエンドサイトーシスが生じた(表3、図1)。対照的に、ラットアペリン−受容体−EGFPを安定発現しているCHO細胞を1μM pE13F(D−Arg2)、pE13F(D−Arg4)及びpE13F(D−Lys8)と共にインキュベートすることは、アペリン受容体のインターナリゼーションを誘導しなかった。実際に、CHO細胞は、細胞内蛍光小胞なしに形質膜レベルで強いアペリン受容体−EGFP蛍光を示した。対照的に、類似体AcR12F、pE13F(Aib7)、pE13F(D−Lys8)、pE13F(D−Ala9)、pE13F(Nle11)、pE13(4Br−Phe13)は、アペリン受容体のインターナリゼーションの強力な誘導物質である。同様に、P26、P92及びJFM V−0196Bは、それぞれ2.11±1.14、0.38±0.11及び0.41±0.16nMのEC50値でアペリンRのインターナリゼーションを誘導することができた(表2、7、図1、9)。pE13F及びK17Fと比較した類似体P26及びP92により誘導されるインターナリゼーションの定量により、効率の順序:K17F=P92>pE13F=P26が示された。
K17F、pE13F、P92及びP26に対するERK1/2リン酸化の用量反応曲線は、K17F及びペプチド92についてそれぞれ等価のEC50値4.08±1.17nM及び3.42±2.41nMを有して類似の最大効果を示したが、一方で、pE13F及びペプチド26についてのEC50値は、K17Fよりも3及び18倍高い(それぞれEC50=12.0±2.79 10−9M及び71.3±19.1 10−9M)(表2)。様々な化合物を用いたERK1/2リン酸化の用量反応曲線は、K17F、P92及びJFM V−0196Bについてそれぞれ4.08±1.17nM、3.42±2.41nM及び0.89±0.61nMのEC50値を有して、類似の最大効果を示した(表7)。対照的に、pE13F及びP26は、それぞれ対応するEC50値12.01±2.79nM及び72.10±19.60nMを有して、ERK1/2リン酸化のより弱いプロモーターであった(表7)。したがって、以下の化合物についての効率の順位は、JFM V−0196B>P92=K17F>pE13F≫P26であった。
アペリン受容体に対するアルキルK17F類似体の親和性は、パーフルオロアルキルペプチドの親和性よりも低かった(表3)。
マウス血漿中の安定性。37℃のマウス血漿中のK17F、pE13F、P26、P92、JFM V−0196A及びJFM V−0196Bの安定性を決定した(表4)。K17F及びpE13Fの血漿中半減期の値は、それぞれ4.6及び7.2分である一方、K17F及びpE13Fのそれぞれの誘導体:P92及びP26は、半減期の値が24及び86分と血漿安定性の増加を示す。そのうえ、K17Fのアルキル又はパーフルオロアルキル誘導体は、4時間インキュベーション後であっても分解が観察されなかったので、より高い血漿安定性を示す。
ノルアドレナリン(NA)により予備収縮したラット大動脈輪に対して
3×10−6M NAを用いて予備収縮されたラット大動脈輪において、K17F、pE13F、P92及びP26は、濃度依存性弛緩を誘導した(図2)。P26の効力(pD2)(6.27±0.42)は、K17F(8.30±0.44)、pE13F(8.00±0.59)及びP92(7.68±0.57)の効力よりも有意に低かった(P<0.05)。対照的に、pE13Fにより誘導される最大効果(62±6%)は、K17F(87±8%、P<0.01)、P26(93±3%、P<0.001)及びP92(100±1%、P<0.001)により誘導される最大弛緩効果よりも有意に低かった(図2A)。対照的に、最大濃度100μMで、pE13F(62±6%)及びJFM V−0196B(60±3%)により誘導される血管弛緩効果は、同濃度のK17Fにより誘導される効力よりも有意に低く(93±3%、P<0.01)、K17Fと比較してこれらの2つの化合物が低い効力であることを示した(図10A)。
ラット糸球体細動脈の血管反応性にこれらの化合物が及ぼす効果を評価するために、1)基礎状態で、2)AngIIを添加後に、並びに3)AngII及びP92の存在下で直径を測定した。結果を図2Bに示すが、1nM AngIIがベースライン状態で測定された値と比較して細動脈直径を有意に減少させたことを示した(それぞれ100.0±4.1対87.6±1.9%、n=5、P<0.05)。1nM AngIIにより予備狭窄した細動脈への500nM P92の添加は、87.6±1.9から98.1±2.5%に細動脈直径を増加させた(n=5、p<0.05)。これらの結果は、P92が、K17KとしてAngIIにより予め予備狭窄した糸球体細動脈の血管拡張を誘導可能であったことを示した。
A)動脈圧に及ぼすP92の効果
イナクチン(用量100mg/kg)で麻酔された正常血圧Wistarラット(300g)において基礎MBPは100.3±1.2mmHgであった。K17F(15nmol/ラット=50nmol/kg)の静脈内注射は、平均動脈圧(MABP)を7.8mmHg減少させた。おそらく血流中でのペプチドの急速な分解を反映して、降圧応答は、注射の1.2分後に最大であり、単に一過性であった。ベースラインへの復帰は、注射の4.2分後に観察された。用量15nmol/ラットで、P92は、K17FよりもBPの減少にずっと有効であり(15nmol K17Fの投与を受けているWistarラットと比較して、1分で−20.8mmHg、P<0.05)、注射の10.8分後にベースラインに復帰した(15nmol K17Fの投与を受けているWistarラットと比較してP<0.05)(図3A)。K17Fの静脈内注射(100nmol/ラット=333nmol/kg=0.3μg/kg)は、MABPを30.5mmHg減少させ、プラトー値−12mmHgへの復帰が、約22分で観察された。同用量のP92について、本発明者らは、約35分で観察された−約20mmHgというプラトー値への復帰を伴うMABPにおける55.4mmHgの減少を観察した(図3B)。
第2系列の実験では、本発明者らは、麻酔されたWistar正常血圧ラットにおけるBPに及ぼす化合物JFM V−0196Bの効果を測定した(図5)。漸増する用量(5〜15nmol/ラット)でi.v.注射されたJFM V−0196Bは、用量依存的にBPを減少させ、用量15nmolについて最大減少50mmHgであり、それに対し、K17Fについて7.8mmHgであった。プラトー値−25mmHgへの復帰が、約33分で10又は15nmol P92について観察された。対照的に、15nmol K17Fの投与を受けているWistarラットでは、10.8分後にベースラインへの復帰が観察された。
マウスの24時間水欠乏は、2セットの実験において血漿AVPレベルを有意に増加させる(425±46pg/ml、n=18に対して対照マウス175±17pg/ml、n=20;P<0.001、図6A及びB並びに644±60pg/ml、n=14に対して対照マウス232±54pg/ml、n=8;P<0.001、図6A及びB)。以前に記載されたように{Iturrioz, 2010 #32}、絶水マウスにおける用量1μg(468pmol)でのK17Fのi.c.v.注射は、食塩水を注射された絶水マウスと比較して(425±46pg/ml)、血漿AVPレベル(190±27pg/ml、n=7)を有意に減少させた(P<0.001)(図6)。
単離されたラット心からの左室圧の記録から、K17F(0.01〜300nM)は発生する圧を用量依存的に増加させたことが明らかとなった。100から300nMの間に含まれる用量について最大効果が観察された。他方で、用量200及び400nMのP92は、K17Fに類似した振幅で心収縮性を増加させる(図7)。試験した2つの化合物について変時効果は観察されなかった。
本発明の他の化合物について実施例1に記載されたように、これらの化合物を合成する。フルオロ−P92及びリポ−P92の化学構造を、それぞれ図12及び図13に開示する。
本出願にわたり、様々な参考文献が、本発明が属する技術の現状を記載している。これにより、これらの参考文献の開示は、参照により本開示に組み入れられる。
