Description
第一の側面において、本発明は、眼新生血管形成の用量依存治療または予防における使用のための、式(I)の化合物、またはその薬学的に許容される塩、溶媒和物、水和物もしくはプロドラッグを提供する。
抗血管新生治療

本発明の化合物は、SRPK1阻害剤として、選択的にスプライシングされたVEGFXXXbアイソフォームが関与している他の障害の治療において治療剤としても使用され得る。例えば、その内容が参照により本明細書に組み込まれるWO2011/036429は、妊娠雌性哺乳類(pregnant female mammal)における低下したVEGFXXXbレベルは、雌性哺乳類が子癇前症を発症するリスクを高めることを示している。したがって、本発明の化合物は、妊娠雌性哺乳類におけるVEGFXXXbレベルを増加させ、子癇前症もしくはそれに関連する合併症を発症する雌性哺乳類のリスク、または母体の子癇前症に関連した胎児もしくは新生児の不全を発症する雌性哺乳類の胎児のリスクを低下させるために使用され得る。
本発明の化合物は、式(I)によって定義され、キナーゼSRPK1およびSRPK2のうち1つまたは両方の阻害剤であり、それゆえ本明細書に記載されるように治療に有用であることが示されている。

本明細書で使用される表現「治療または予防」および類似の用語は、一般的な医療および精神医学的プラクティスに従った任意の利用可能なテストに従って判断されるような、障害を取り除きもしくは回避し、またはその症状を緩和することを意図した、予防的、治癒的(curative)および緩和的(palliative)等の、全ての形態の健康管理(healthcare)を意味する。特定の結果を達成するために合理的に期待して意図するものの、常にそうするというわけではない介入(intervention)は、「治療または予防」という表現に含まれる。障害の進行を遅延させまたは停止させることに成功する介入は、表現「治療または予防」に含まれる。

初代ヒトRPE単離は、Bristol Eye bank (Bristol Eye Hospital (BEH))から死後24時間以内に得られたヒトドナー眼球に対して実施された。網膜は脈絡膜RPEシートと一緒にペトリ皿に移され、細かく刻まれ、0.3mg/mlコラゲナーゼが補充されたダルベッコ改変イーグル培地(DMEM):F12(1:1)+GlutaMax(Gibco)中で15分間37℃で消化された。消化された脈絡膜RPEシートは、10%ウシ胎児血清(FBS)、0.5%PenStep(Invitrogen)が補充された培地(DMEM:F12+GlutaMax)中に懸濁され、1500rpm(251g)で10分、ペレット細胞へと回転された。ペレットは25%FBS(Gibco)が補充された培地中に再懸濁され、細胞培養フラスコ(Greiner)内で増殖され、80%コンフルエンスで分割された。ARPE−19(ATCC)細胞は、DMEM:F12+10%FBS中で培養され、80%コンフルエンスで分割された。
MVRL09、MVRL10(SPHINX)、MVRL16(SPHINX6)、MVRL17(SPHINX7)、SPHINX8、SPHINX9、SPHINX10、SPHINX12、SPHINX13、SPHINX14およびSRPIN340を含む候補化合物は、Kinase−Glo assay(Promega; Koresawa and Okabe, 2004)によってスクリーニングされ、その結果が表1に示される。9.6mM MOPS pH7および0.2nM EDTA pH8を含む反応バッファーは、10μΜ SRSF1 RSペプチド(NH2−RSPSYGRSRSRSRSRSRSRSRSNSRSRSY−OH(配列番号:1))および0.1μgの精製SRPK1キナーゼに添加された。候補化合物は、10μΜ〜0.5nΜで系列希釈され、反応混合物に添加され、SRPK1キナーゼが省略された(omitted)および化合物が省略されたウェルもまた対照として加えられた。すべてのウェルは、1%DMSOを含んでいた。1マイクロモーラーのATPが添加され、ATPマイナスのウェルは、バックグラウンド対照として使用された。次いで、プレートは30℃で10分間インキュベートされた。等量のKinase−Glo(Promega, 25μl)が各ウェルに添加され、プレートは、ARVO5x(Perkin Elmer)を使用して発光について測定された。
定型的なPCRが、VEGF165およびVEGF165b mRNAを検出するために使用された。5−10%のcDNAが、以下を含む反応混合物に添加された:2×PCR Master Mix (Promega)、エキソン7b(5’−GGC AGC TTG AGT TAA ACG AAC−3’(配列番号:2))およびエキソン8bの3’UTR(5’−ATG GAT CCG TAT CAG TCT TTC CTG G−3’(配列番号:3))に相補的なプライマー(各1μΜ)、およびDNase/RNaseフリー水。全てのサンプルは、陰性対照(逆転写酵素なしの、水およびcDNA(−RT))、および陽性対照(プラスミド発現ベクター(pcDNA)中のVEGF165およびVEGF165b pcDNA)と並行して測定された。反応混合物は95℃で60秒間変性し、55℃で60秒間アニーリングし、72℃で60秒間伸長させて、30〜35回、温度サイクルされた(PCR Express, Thermo Electron Coorporation, Basingstoke)。PCR産物は、0.5μg/ml臭化エチジウム(BioRad)を含む2.5%アガロースゲル上で分離され、紫外線トランスイルミネーター(BioRad)下で可視化された。
タンパク質サンプル(30〜50μg)は、1×ドデシル硫酸ナトリウム(SDS)ローディングバッファー(100mM Tris−HCl、4%SDS、20%グリセロール、0.2%(w/v)ブロモフェノールブルー、および最終濃度5%2−メルカプトエタノール、 pH6.8)と混合された。タンパク質を変性させるために、サンプルは100℃で5分間煮沸された。
1×106 PC−3 RFP(赤色蛍光タンパク質)細胞が、ヌードマウスの前立腺に外科的に投入された。腫瘍体積は、IVIS Lumina imaging systemを用いて毎週2回測定された(総フラックス:光子/秒として表される)。腫瘍が2×l09平均光子/秒に達すると、マウスは、20μg SPHINXまたはビヒクルのいずれかのIP注射で週に三回処理された。31日後、マウスは間引きされ、さらなる分析のために腫瘍が抽出された。N=9、p<0.01、2要因ANOVA。
インビボでのSRPIN340の薬物動態を決定するために、質量分析ベースの戦略が採用された。初めに、SRPIN340および分子誘導体MVRL09は、100μg/mlから0μg/mlまで水で系列希釈され(最初のストックはDMSOに溶解された)、分析された。クロマトグラムは、SRPIN340(349.1Da)およびMVRL09(351Da)に予想された分子量で明瞭なピークを作り出した。ピーク下面積が統合され、線形応答を確認するための濃度に対するプロットが観察された。SRPIN340は、20ngの硝子体内注射後の眼組織で、および5μgの単回局所適用後の眼組織で調べられた。単回投与後、マウスは1、4、8、24および48時間の時点で安楽死させられた。サンプル、SRPIN340(検体)および対照処理されたものは、ホモジナイズされ、等量(100μl)アセトニトリルまたはアセトンでサンプルからタンパク質が沈殿させられた。
MVRL09の内部標準が、調製中のサンプルのロスについて勘定するためにサンプルに添加された。50%サンプルおよび50%アセトニトリルおよび100μg/ml MVRL09の溶液は、タンパク質をペレット化するために4℃で15分間遠心分離され、分析のために上清が採取された。溶液は8時間37℃で蒸発させられ、注入およびMALDI−TOF質量分析法分析器(Absciex)の準備のために30μlアセトニトリルに再懸濁された。各サンプルは、5分間測定され、クロマトグラムが作成された。標準曲線は、濃度を決定するために、両方のヒト血漿水マトリックス中で作成された。半減期は、対数プロットを使用して、1時間から48時間までのサンプル濃度に対して1つの位相指数関数的減衰をフィッティングすることにより、Prismを用いて計算された。
特記しない場合、データは平均±SEMとして示される。すべてのデータ、グラフおよび統計的分析は、Microsoft Excel(Microsoft Office Software)、GraphPad Prism(GraphPad Software Inc.)、およびImage Jにより算出された。全ての結果は、p<0.05(*)、P<0.01(**)およびp<0.001(***)で統計的に有意であるとみなされた。
新規SRPK1阻害剤の同定
新規SRPK1および高度に関連するSRPK2阻害剤を同定するために、ある範囲の阻害剤がインビトロキナーゼアッセイ(Promega; Koresawa and Okabe, 2004)でスクリーニングされた。以前に同定されたSRPK阻害剤SRPIN340(Fukuhara et al., 2006)は、新規候補化合物MVRL10(5−メチル−N−[2−(モルホリン−4−イル)−5−(トリフルオロメチル)フェニル]フラン−2−カルボキサミド)およびMVRL09(N−[2−(モルホリン−4−イル)−5−(トリフルオロメチル)フェニル]ピリジン−3−カルボキサミド)の同定のための陽性対照として使用された。構造的誘導体SRPIN349は、陰性対照として使用された(図1)。SRPIN349はSRPIN340と同様の構成を有しているが、トリフルオロメチル部分の代わりに水素原子を有する。3つ全ての阻害剤は、SRPK1キナーゼを阻害するいくらかの能力を示し、MVRL09は10μΜより大きいIC50で最も低い効果である。SRPIN340およびMVRL10の両方が、それぞれ0.96μΜおよび0.85μΜのIC50値でSRPK1活性を強力に阻害した(図1B)。このアッセイで得られた結果は、SRPIN340について公表されたIC50と一致している(0.89μΜ; Fukuhara et al., 2006)。新規候補化合物MVRL16(SPHINX6)、MVRL17(SPHINX7)がまた、同じインビトロキナーゼアッセイを用いて同定され、それぞれ173nMおよび54nMのIC50値を示している(図7)。ピペラジン構造が欠落しているMVRL18(SPHINX8)、SPHINX9およびSPHINX10は、インビトロアッセイでは活性を示さなかった。
我々は、血管新生促進VEGFアイソフォームVEGF165の発現を低下させ、インビボにおいてCNVを阻害するため、SRPK1の小分子化合物阻害剤を使用した。我々は、阻害剤が、インビトロで、SRSF1 RSドメインのセリン残基のSRPK1リン酸化を防止することを同定した。SRPK1のSRSF1との相互作用は、文献(Aubol et al., 2003; Velazquez−Dones et al., 2005; Ngo et al., 2005)に詳しく記載されている。SRSF1は、VEGFを含む多数の遺伝子の選択的スプライシングを制御するプロトオンコジーンSRタンパク質である(Sanford et al., 2005)。ここで、我々は、ARPE−19細胞へのSRPK1阻害剤の反復投与がSRPK1発現非依存的にSRSF1の発現を低下させたことを示し、これはSRSF1のリン酸化がその細胞質安定性のために必要となる可能性があることを示唆している。さらに、SRPK1阻害剤SRPIN340およびMVRL09は、RNAレベルでVEGF165発現を低下させ、タンパク質レベルでの総VEGF発現を有意に低下させた。MVRL10はVEGFタンパク質レベルに影響を与えなかったが、促進型−および抗−血管新生VEGFアイソフォームの発現比率についてのさらなる調査が、MVRL10は抗血管新生VEGFアイソフォームの発現を優位にする比率に有意に切り替えたことを示した。100nMでのSPHINX7は、RNAレベルでVEGF165の量を低減すること(図9A)、タンパク質レベルでVEGF165bを増加させること(図9B)、および10μΜでSRSF1リン酸化を阻害すること(図10Aおよび10B)が示されている。マウスレーザー誘発CNVモデルにおいてインビボで試験した場合、3つ全ての阻害剤が新生血管病変の面積を低下させ、SPHINX7、SRPIN340およびMVRL10(SPHINX)は有意を達成した。

Claims (17)
- 式(I)の化合物、またはその薬学的に許容される塩、溶媒和物、もしくは水和物を含む眼新生血管形成の用量依存もしくは局所治療または予防剤;
ここで:
R1は1つ以上の置換基を有していてもよいC1−6アルキル基、ハロゲン原子、ニトロ基、シアノ基、アジド基、ヒドロキシ基、1つ以上の置換基を有していてもよいC1−6アルコキシ基、1つ以上の置換基を有していてもよいC1−6アルキルチオ基、1つ以上の置換基を有していてもよいC1−6アルキルスルホニル基、カルボキシル基、ホルミル基、1つ以上の置換基を有していてもよいC1−6アルコキシカルボニル基、アシル基、アシルアミノ基、またはスルファモイル基を表し;
R2は水素原子、または1つ以上の置換基を有していてもよいC1−6アルキル基を表し;
R3は1つ以上の置換基を有していてもよいC6−10アリール基、1つ以上の置換基を有していてもよい含窒素複素環、1つ以上の置換基を有していてもよい含酸素複素環、または1つ以上の置換基を有していてもよい縮合芳香族複素環を表し;
R4は水素原子またはハロゲン原子を表し;
Qは−C(O)−、−C(S)−、−C(S)NHC(O)−、−C(O)NHC(O)−、またはC(O)NHC(S)−を表し;ならびに
Wは1つ以上の置換基を有していてもよい1−ピペラジニル基を表す。 - 用量依存および局所治療または予防剤である、請求項1に記載の眼新生血管形成の治療または予防剤。
- 前記眼新生血管形成が、脈絡膜新生血管形成である、請求項1または2に記載の眼新生血管形成の治療または予防剤。
- 前記眼新生血管形成が、網膜新生血管形成である、請求項1または2に記載の眼新生血管形成の治療または予防剤。
- 式(I)の化合物、またはその薬学的に許容される塩、溶媒和物、もしくは水和物;
ここで:
R1は1つ以上の置換基を有していてもよいC1−6アルキル基、臭素原子、塩素原子、ニトロ基、シアノ基、アジド基、ヒドロキシ基、1つ以上の置換基を有していてもよいC1−6アルコキシ基、1つ以上の置換基を有していてもよいC1−6アルキルチオ基、1つ以上の置換基を有していてもよいC1−6アルキルスルホニル基、カルボキシル基、ホルミル基、1つ以上の置換基を有していてもよいC1−6アルコキシカルボニル基、アシル基、アシルアミノ基、またはスルファモイル基を表し;
R2は水素原子、または1つ以上の置換基を有していてもよいC1−6アルキル基を表し;
R3は1つ以上の置換基を有していてもよい含酸素複素環、または1つ以上の置換基を有していてもよい2−もしくは3−もしくは4−ピリジル基を表し;
R4は水素原子またはハロゲン原子を表し;
Qは−C(O)−、−C(S)−、−C(S)NHC(O)−、−C(O)NHC(O)−、またはC(O)NHC(S)−を表し;ならびに
Wは1つ以上の置換基を有していてもよい1−ピペラジニル基を表す。 - 式(I)の化合物、またはその薬学的に許容される塩、溶媒和物、もしくは水和物を含む哺乳動物対象の抗血管新生治療剤;
ここで:
R1は1つ以上の置換基を有していてもよいC1−6アルキル基、ハロゲン原子、ニトロ基、シアノ基、アジド基、ヒドロキシ基、1つ以上の置換基を有していてもよいC1−6アルコキシ基、1つ以上の置換基を有していてもよいC1−6アルキルチオ基、1つ以上の置換基を有していてもよいC1−6アルキルスルホニル基、カルボキシル基、ホルミル基、1つ以上の置換基を有していてもよいC1−6アルコキシカルボニル基、アシル基、アシルアミノ基、またはスルファモイル基を表し;
R2は水素原子、または1つ以上の置換基を有していてもよいC1−6アルキル基を表し;
R3は1つ以上の置換基を有していてもよい含酸素複素環、または1つ以上の置換基を有していてもよい2−もしくは3−もしくは4−ピリジル基を表し;
R4は水素原子またはハロゲン原子を表し;
Qは−C(O)−、−C(S)−、−C(S)NHC(O)−、−C(O)NHC(O)−、またはC(O)NHC(S)−を表し;ならびに
Wは1つ以上の置換基を有していてもよい1−ピペラジニル基を表す。 - 請求項6で規定される化合物を含む、微小血管透過性亢進障害(microvascular hyperpermeability disorders)のための、またはVEGFXXXアイソフォームの血管新生促進透過性促進特性(pro−angiogenic pro−permeability properties)の制御のための、または増加した透過性の無い(without increased permeability)上皮細胞生存の支援における、上皮濾過膜の開窓(fenestrations)の性質の低下のための治療または予防剤。
- 請求項6で規定される化合物を含む、神経障害(neuropathic)および神経変性障害(neurodegenerative disorder)のための、またはインビボもしくはインビトロでの神経保護(neuroprotective)もしくは神経再生(neuroregenerative)のための治療または予防剤。
- 請求項6で規定される化合物を含む、VEGFR2媒介性非炎症性疼痛(VEGFR2−mediated non−inflammatory pain)の治療または予防剤。
- 請求項6で規定される化合物を含む、子癇前症もしくはそれに関連する合併症を発症する雌性哺乳類(female mammal)のリスク、または母体の子癇前症に関連した胎児もしくは新生児の不全を発症する雌性哺乳類の胎児のリスクの低下における使用のための治療または予防剤。
- 請求項6に記載の式(I)の化合物、任意に1つ以上の他の活性成分および薬学的に許容される担体を含む医薬組成物。
- 請求項5に記載の式(I)の化合物、任意に1つ以上の他の活性成分および薬学的に許
容される担体を含む、眼内注射に適した形態の医薬組成物。 - 請求項5に記載の式(I)の化合物、任意に1つ以上の他の活性成分および薬学的に許容される担体を含む、眼への局所投与に適した形態の医薬組成物。
- R1がトリフルオロメチル基を表す、請求項5に記載の化合物、または請求項1〜4もしくは6〜10のいずれか1項に記載の治療もしくは予防剤、または請求項11〜13のいずれか1項に記載の医薬組成物。
- R3が、含酸素複素環または2−もしくは3−もしくは4−ピリジル基を表し、それらのそれぞれが1つ以上の置換基を有していてもよい、請求項14に記載の化合物、治療もしくは予防剤、または医薬組成物。
- R3が、フェニル基または2−もしくは3−もしくは4−ピリジル基により置換された含酸素複素環を表す、請求項15に記載の化合物、治療もしくは予防剤、または医薬組成物。
- Wが、1−ピペラジニル基または4−メチル−1−ピペラジニル基または4−(ジメチルアミノ)エチル)−1−ピペラジニル基または4−(ジメチルアミノ)プロピル)−1−ピペラジニル基を表す、請求項14〜16のいずれか1項に記載の化合物、治療もしくは予防剤、または医薬組成物。
