Abstract
Description
本出願は、2010年5月5日付で出願された米国仮特許出願第61/331,550号の出願日の恩典を主張するものであり、その開示は参照により本明細書に組み入れられる。
本出願は、EFS-Webを介して提出された配列表を含んでおり、その全体が参照により本明細書に組み入れられる。2011年5月2日に作出されたASCIIコピーは、Sequence Listing_Staphylococcus Aureus Leukocidins_ST25.txtと名付けられており、サイズが102キロバイトである。
黄色ブドウ球菌(Staphylococcus aureus)または「staph」は、通常、人および動物の皮膚にまたは鼻内にみられる。そのような細菌は、切創または他の創傷を通じて身体に侵入しない限り、一般的には無害である。典型的には、staph感染は健常人ではささいな皮膚の問題である。歴史的には、staph感染はメチシリンのような、広域抗生物質によって処置される。ところが、現在では、メチシリン、ならびにペニシリンおよびセファロスポリンのような他のβ-ラクタム系抗生物質に対して耐性を示すある種のstaph株が出現している。それらはメチシリン耐性黄色ブドウ球菌(多剤耐性黄色ブドウ球菌、または「MRSA」としても公知)といわれる。

以下の開示は、順繰りに、LukAポリペプチド、LukBポリペプチド、LukAおよびLukBポリヌクレオチド、抗LukA抗体および抗LukB抗体、LukAおよび/もしくはLukB、または抗LukA抗体および/もしくは抗LukB抗体を含有する治療用組成物、治療用組成物を使用する方法、LukAB媒介性細胞毒性の阻害剤を特定する方法、ならびに黄色ブドウ球菌感染の重症度を予測または評価する方法を対象にする。
黄色ブドウ球菌に生まれつき備わっているポリペプチドが、今回、本出願人らにより単離され、公知のS-サブユニットロイコシジン(例えば、LukS-PVL、LukEおよびHlgC)の活性プロファイルを示すと特定された。本明細書においてまとめてLukAと表されるこれらのポリペプチドは、ヒト食細胞を特異的に標的化し結合し(しかしヒト上皮細胞もしくはヒト内皮細胞、またはマウス細胞ではそうでない)、いったん食細胞膜に結合すると、LukAは黄色ブドウ球菌F-サブユニットロイコシジン(例えば、本明細書において開示されるLukF-PVL、LukDおよびHlgB、ならびにLukB)とオリゴマー化し、オリゴマー化によって膜貫通細孔を形成する(まとめてLukA活性といわれる)。図1に示されるアライメントには、大多数のLukA配列のアミノ酸配列(本明細書においてSEQ ID NO:1と表される)、およびその対応する13種の異なる黄色ブドウ球菌株由来のLukAポリペプチドのアミノ酸配列(本明細書においてSEQ ID NO:2〜14と表される)が含まれる。
黄色ブドウ球菌に生まれつき備わっているポリペプチドが、今回、本出願人らにより、公知のF-サブユニットロイコシジン(例えば、LukF-PVL、LukDおよびHlgB)の活性プロファイルを示すと特定された。本明細書においてまとめてLukBと表されるこれらのポリペプチドは、ヒト食細胞に結合される黄色ブドウ球菌S-サブユニットロイコシジン(例えば、本明細書において開示されるLukS-PVL、LukEおよびHlgC、ならびにLukA)と特異的にオリゴマー化し、オリゴマー化によって食細胞に膜貫通細孔を形成する(まとめてLukB活性といわれる)。図2に示されるアライメントには、大多数のLukB配列のアミノ酸配列(本明細書においてSEQ ID NO:15と表される)、およびその対応する12種の異なる黄色ブドウ球菌株由来のLukBポリペプチドのアミノ酸配列(本明細書においてSEQ ID NO:16〜27と表される)が含まれる。
LukAおよびLukBロイコシジンは、当技術分野において周知である組み換えDNA法によって合成することができる。例えば、黄色ブドウ球菌(Newman)のLukAポリペプチド(SEQ ID NO:2)をコードするヌクレオチド配列(SEQ ID NO:28と指定した)を以下に示す。このポリペプチドをコードする縮重(例えば、組み換え発現の目的のために選択の宿主においてコドン選択という点で有用でありうる)配列、および他のLukAポリペプチドをコードするポリヌクレオチドは、当技術分野において公知であり、または当業者によってデザインされることができる。
本発明の局面は、LukAに特異的に結合する抗LukA抗体、およびLukBに特異的に結合する抗LukB抗体、該抗体を含有する治療用組成物、ならびにその使用の方法を対象にする。本発明の目的で、「抗体」という用語は、モノクローナル抗体、ポリクローナル抗体、抗体断片、および抗体の遺伝子組み換え型、ならびにそれらの組み合わせを含む。より具体的には、「免疫グロブリン」という用語と互換的に用いられる「抗体」という用語は、全長(すなわち、天然に存在するまたは通常の免疫グロブリン遺伝子断片組み換え過程により形成された)免疫グロブリン分子(例えば、IgG抗体)、およびかさねて、天然に存在してもよい、または事実上合成による、その免疫学的に活性な断片(すなわち、全長免疫グロブリン分子の特異的な結合部分を含む)を含む。したがって「抗体断片」という用語はF(ab')2、F(ab)2、Fab'、Fab、Fv、scFvなどのような抗体の一部分を含む。構造にかかわらず、抗体断片は、全長抗体によって認識される同一抗原と結合し、本発明との関係においては、LukA、LukBまたはLukAB複合体と特異的に結合する。抗体断片を作出およびスクリーニングする方法は、当技術分野において周知である。
LukAおよびLukBは、限局性急性炎症状態を含む、急性炎症状態の処置において抗炎症剤として用いるための治療用組成物へ処方することができる。LukAおよびLukBはまた、能動ワクチンとして用いるための治療用組成物へ処方することもできる。抗LukA抗体および抗LukB抗体は、受動ワクチンとして用いるための治療用組成物へ処方することができる。受動および能動ワクチンを予防的に用いて、黄色ブドウ球菌感染の発症を阻止することができ、または治療的に用いて、黄色ブドウ球菌感染、具体的には、難治性であることが知られた、もしくは特定の対象の場合、他の従来の抗生物質治療による処置に対して難治性と証明された、MRSAのような黄色ブドウ球菌感染を処置することができる。
急性炎症状態
炎症は、病原体(例えば、細菌およびウイルス)、損傷細胞および刺激物などの有害な侵入性の刺激を除去するための、ならびに治癒を開始するための防御生物学的応答と一般的に理解されている。炎症は、より具体的には、損傷に対して血管新生された生きている組織の反応と理解されている。したがって、炎症は、物理的、化学的または生物学的因子により引き起こされる損傷または異常刺激に応じた罹患血管および隣接組織の細胞学的および化学的反応の、定型化した基本固定概念である。炎症は、通常、損傷部位での体液および血液細胞の蓄積をもたらし、通常、治癒過程である。炎症過程がなければ、創傷および感染は治癒せず、組織の進行性破壊が致命的となるであろう。急性炎症は、侵入性の刺激に対する身体の初期応答をいい、損傷組織または感染組織への血漿および白血球の動員を伴う。慢性炎症ともいわれる、長期の炎症は、炎症部位に存在する免疫細胞のタイプの進行性の変化を伴い、炎症過程からの組織の同時の破壊および治癒によって特徴付けられる。
本発明はまた、抗体組成物を、それを必要としている哺乳類対象に投与することにより黄色ブドウ球菌感染の発症を阻止するまたは黄色ブドウ球菌感染を処置する方法を提供する。本発明の目的上、標的の対象集団には、黄色ブドウ球菌に感染している、または黄色ブドウ球菌に感染するリスクがある、ヒトのような、哺乳類が含まれる。いくつかの態様において、処置される対象は、MRSAを含む黄色ブドウ球菌に感染しており、および/または抗生物質もしくは他の治療剤で既に処置されたが、処置に失敗している。
急性炎症状態の処置との関連で、LukAおよびLukBの量は、処置が症状の数の低減、少なくとも1つの症状の重症度の減少、または少なくとも1つの症状のさらなる進行の遅延、または場合により急性炎症状態の完全な緩和のいずれかの1つまたは複数を達成できるという意味で治療的に有効である。
投与の前に、本発明の治療用組成物は滅菌されてもよく、これは凍結乾燥および再構成の前にまたは後に、滅菌ろ過膜を通じたろ過によって容易に達成することができる。治療用組成物は、滅菌アクセスポートを有する容器、例えば、静脈内溶液バッグまたは皮下注射針によって貫通されうるストッパを有するバイアルの中に入れることができる。
いくつかの態様において、治療用組成物は、処置されている黄色ブドウ球菌感染または急性炎症状態の性質に依り、別の活性薬剤とともに併用療法の一部として投与される。そのようなさらなる活性薬剤には、抗感染症剤、抗生物質製剤、および抗菌剤が含まれる。本発明において有用でありうる代表的な抗感染症剤には、バンコマイシンおよびリソスタフィンが含まれる。本発明において有用でありうる代表的な抗生物質製剤および抗菌剤には、バンコマイシン、リソスタフィン、ペニシリンG、アンピシリン、オキサシリン、ナフシリン、クロキサシリン、ジクロキサシリン、セファロチン、セファゾリン、セファレキシン、セフラジン、セファマンドール、セフォキシチン、イミペネム、メロペネム、ゲンタマイシン、テイコプラニン、リンコマイシンおよびクリンダマイシンを含めて、ペニシリナーゼ耐性ペニシリン、セファロスポリンおよびカルバペネムが含まれる。これらの抗生物質の投与量は、当技術分野において周知である。例えば、MERCK MANUAL OF DIAGNOSIS AND THERAPY, Section 13, Ch. 157, 100th Ed. (Beers & Berkow, eds., 2004)を参照されたい。抗炎症剤、抗感染症剤、抗生物質および/または抗菌剤は、投与の前に混ぜ合わされてもよく、あるいは本発明の治療用組成物と同時に(同じ組成物の一部としてもしくは異なる組成物によって)または連続的に投与されてもよい。
本発明の治療用組成物は単回の用量で、または複数投薬プロトコルにしたがって投与することができる。例えば、1用量または2用量のような、比較的少用量の治療用組成物が投与される。数日間または数週間にわたる複数用量を一般的に伴う、従来の抗生物質療法を含む態様において、抗生物質は一定期間、例えば少なくとも5日間、10日間または場合により14日間もしくはそれ以上の日数の間、毎日1回、2回もしくは3回またはそれ以上の回数、服用されうるが、一方で抗体組成物は、通常、1回または2回だけ投与される。しかしながら、当業者は、治療用組成物および抗生物質のさまざまな投与量、投与のタイミングおよび相対量を選択および調節することができる。
抗LukA抗体および抗LukB抗体、およびその断片、ならびに他の潜在的な治療的部分(例えば、有機小分子)をさまざまな方法(アッセイ形式またはスクリーニングを含む)で用いて、LukAB媒介性細胞毒性を阻害するその能力を評価することができる。下記のように、これらの方法は、LukAB媒介性細胞毒性およびヒト食細胞の溶解をもたらす事象のカスケードのいくつかの局面を阻害する薬剤を特定するようにデザインされる。これらの方法はまた、天然の対応物に比べて低減した毒性を保有するLukAおよびLukBの改変型を特定するようにデザインされる。カスケードの一部であり標的化される事象には、例えば、食細胞膜へのLukAの結合、LukAへのLukBの結合(LukABオリゴマー化)、およびLukABオリゴマーによって形成された膜細孔の遮断が含まれる。アッセイ形式には一般に、ヒト食細胞(またはそのLukAB膜結合部分)、適当な培地、ならびに精製されたLukAまたは精製されたLukAおよびLukBが必要とされる。
傷害過程における第1段階であるものと考えられる、標的細胞へのLukAの結合を遮断または低減する阻害剤をスクリーニングするために、ヒト食細胞(例えばPMN-HL60細胞)を384ウェル平底組織培養処理プレート(Corning)の中に、10%の熱不活化ウシ胎仔血清(FBS)を補充したRPMI (Gibco)の終容量50μl中2.5×103個の細胞/ウェルでプレーティングすることができる。次に細胞を試験化合物/分子(およそ5μl/さまざまな濃度)で処理し、精製され、蛍光的に標識されたLukA (例えば、FITC、Cy3、Cy5、APC、PE)およそ0.01〜2μMの溶液5 ulで37℃、5% CO2にて1時間傷害することができる。試験される化合物/分子の効力を評価するため、例えば、ハイコンテントスクリーニングおよびハイコンテント分析用にデザインされた自動蛍光顕微鏡画像化システム(例えばCellomics ArrayScan HCS Reader (Thermo Scientific) (励起485 nm、発光535 nm))を用い、細胞へのLukAの結合の指標として細胞結合型の蛍光を測定することができる。
傷害過程における第2段階であるものと考えられる、LukA/LukB相互作用を遮断または低減する阻害剤をスクリーニングするために、ヒト食細胞(例えばPMN-HL60細胞)を384ウェル平底組織培養処理プレート(Corning)の中に、10%の熱不活化ウシ胎仔血清(FBS)を補充したRPMI (Gibco)の終容量50μl中2.5×103個の細胞/ウェルでプレーティングすることができる。次に細胞を試験化合物/分子で処理し、その後、精製されたLukAと、FITC、Cy3、Cy5、APC、およびPEのような蛍光分子で蛍光的に標識されている精製されたLukBとの混合物で傷害することができ、傷害過程を完了するように(例えば、37℃、5% CO2で1時間)静置させることができる。試験される化合物/分子の効力を評価するため、例えば、ハイコンテントスクリーニングおよびハイコンテント分析用にデザインされた自動蛍光顕微鏡画像化システム(例えばCellomics ArrayScan HCS Reader (Thermo Scientific) (励起485 nm、発光535 nm))を用い、LukA/LukB-FITC相互作用の指標として細胞結合型LukB-FITCの蛍光を測定することができる。
LukAB細孔の形成を遮断または阻害する阻害剤、細胞溶解をもたらすエフェクタ分子をスクリーニングするために、ヒト食細胞(例えばPMN-HL60細胞)を384ウェル透明底黒色組織培養処理プレート(Corning)の中に、10%の熱不活化ウシ胎仔血清(FBS)を補充したRPMI (Gibco)の終容量50μl中2.5×103個の細胞/ウェルでプレーティングすることができる。次に細胞を試験化合物/分子(さまざまな濃度を含有するおよそ5μl)で処理し、その後、精製されたLukAB (およそ0.001〜2μm)で37℃、5% CO2にて10分間傷害することができる。対照として、PMN-HL60細胞を培地で処理することができ(陰性対照)、および0.1% v/v Triton X100で処理することができる(陽性対照)。
本発明のさらなる局面は、感染した対象から得られた生体サンプルにおける、LukAおよび/もしくはLukB、またはその対応する遺伝子の存在または量の検出を伴う、黄色ブドウ球菌感染の重症度を予測または評価する方法を対象にする。したがって、ほんのわずかなまたは検出不能な量のLukAおよび/またはLukBを産生する対照(例えば、黄色ブドウ球菌株USA100およびUSA400)と比べて、LukAおよび/もしくはLukBの存在もしくは比較的多量の検出、またはその対応遺伝子(例えば、黄色ブドウ球菌株Newman、4645、ならびにMRSA株USA300およびUSA500によって示される)の検出は、重篤な感染を示す。LukAおよび/またはLukBの相対量の検出または存在に関して、図4Aのイラストを参照することができる。この方法の代表的な態様を以下に記述する。
LukABレベル(すなわち、LukAB産生)を判定するために、生体サンプル、例えば、体液(例えば、血液)または組織サンプルを感染対象から入手し、引き続いて黄色ブドウ球菌の増殖を可能とするのに適した培養条件への培養物の曝露、培養上清の入手、そこからの細菌タンパク質の分離、LukAおよび/またはLukBの特定、その後、LukAおよび/またはLukBの定量化を行う。より具体的には、1つの態様において、臨床分離株を適当な培地、例えば、1%カザミノ酸を補充したRoyal Park Memorial Institute培地1640 (RPMI; Invitrogen) (RPMI+CAS)中にて、適当な培養条件の下で、例えば、180 rpmで振盪させながら37℃で12〜18時間、選択および増殖することができる。次いで細菌を遠心分離によって沈殿させることができ、培養上清を回収することができる。次に培養上清(およそ30μl)をSDS-Laemmli緩衝液10μlと混合することができ、95℃で10分間煮沸することができる。次にタンパク質を、例えば、15% SDS-PAGEゲルを用いて分離することができ、その後、固体支持体、例えば、ニトロセルロース膜に転写することができる。この膜を次に、LukAまたはLukBに対して作製された抗体(例えば、ウサギポリクローナル抗体)とともにインキュベートすることができ、抗体-LukA/抗体-LukB複合体をフルオロフォアに結合された二次抗体(例えば、AlexaFluor-680に結合された抗ウサギ抗体; Invitrogen)で検出することにより、LukAまたはLukBの存在を可視化することができる。次いで膜を乾燥し、例えば、Odyssey赤外線撮像システム(LI-COR Biosciences)を用い走査して、LukAおよびLukBの量を判定することができる。
LukABをコードする遺伝子の存在について判定するために、生体サンプルを感染対象から入手し、引き続いて黄色ブドウ球菌の増殖を可能とするのに適した培養条件への培養物の曝露、培養黄色ブドウ球菌からの核酸の抽出、その後、LukAおよび/またはLukB特異的なプライマーを用い、PCRまたは他の適当な増幅プロトコルを用いた少なくとも1ラウンドの核酸増幅の実施、ならびにLukAおよび/またはLukBの検出を行う。したがって、1つの代表的な態様において、最初のサンプル調製の後、臨床分離株を固形培地、例えば、37℃にて1.5%の寒天で固化されたトリプティックソイブロス(TSB)中で/上で増殖させることができる。次いで黄色ブドウ球菌コロニーを選択し、例えば、37℃で10分間TSM緩衝液(100 mM TRIS pH 7、500 mMスクロース、10 mM MgCl2)中2 mg/mlのリソスタフィン(AMBI PRODUCTS LLC)で酵素的に消化することができる。次いでサンプルを遠心分離し、上清を捨て、ペレットを滅菌水100μlで再懸濁し、引き続き100℃で5分間の煮沸、および遠心分離を行うことができる。上清は、出発材料ならびにLukAおよび/またはLukB特異的なプライマーを用いたPCRなどの増幅反応用のDNA鋳型をもたらす。

LukAおよびLukB遺伝子産物をNde1およびSal1 (New England BioLabs)で消化し、pMAL-c4Xベクター(New England BioLabs)へ核酸連結した。この構築体を大腸菌株DH5αへ形質転換し、配列決定によってプラスミド挿入断片を確認した。形質転換体を、培養物がおよそ0.5のA600に達するまで、100 ug/mlのアンピシリンおよび0.2%のグルコースを有するTerrific Broth中にて37℃で増殖させた。6-his-タグ付MBP-LukAまたは6-his-タグ付MBP-LukBの発現を180 rpmで振盪させながら、16℃にて終夜、0.3 mMのイソプロピルβ-D-1-チオガラクトピラノシド(IPTG)で誘導した。

遺伝子産物をXho1およびBamH1 (New England BioLabs)で消化し、pET14bベクター(Novagen)へ核酸連結し、遺伝子の5'領域にヒスチジンタグのコード配列を融合した。この発現プラスミドを大腸菌株DH5αへ形質転換し、プラスミド挿入断片を配列決定によって確認した。プラスミドを精製し、発現大腸菌株T7 lysY/lq (New England BioLabs)へ形質転換した。
LukAおよびLukB遺伝子を、以下のプライマーを用いアニーリング温度55℃の標準的PCRの設定の下でVentポリメラーゼ(New England BioLabs)により黄色ブドウ球菌DNAから増幅した: LukAの場合には

LukAおよびLukB遺伝子産物をNde1およびSal1 (New England BioLabs)で消化し、pET41bベクター(Novagen)へ核酸連結した。この構築体を最初に、DH5α細胞へ形質転換し、その後、大腸菌発現株ER2566 (New England BioLabs)へ形質転換した。形質転換体を37℃で2.5時間、カナマイシン25 ug/mlを有するTerrific Broth中で増殖させ、LukAおよびLukBの発現を180 rpmで振盪させながら、37℃にて2時間、0.3 mMのIPTGで誘導した。細胞をペレット化し、1×TBS (500 mM Tris、1.5 M NaCl、pH 7.5)中に再懸濁し、1分間氷上で超音波処理(10秒のパルス)した。超音波処理した細菌を50,000 rpmで30分間、超遠心分離した。
フロイント完全アジュバント(FCA)中で乳化された変性組み換えLukA (250μg)をNew Zealand Whiteウサギに注射した。動物を、フロイント不完全アジュバント(FCA)中で乳化された組み換えLukA (125μg)で21日目におよび49日目に追加免疫した。
使用した細胞株
LukABがヒト食細胞をいかにして標的化し、死滅化させるかを調べるためのモデルとして、HL-60細胞(ATCC CCL-240、ヒト前骨髄球細胞株)を用いた。HL-60細胞を、10%の熱不活化ウシ胎仔血清(FBS)を補充したRPMI培地(Gibco)中、5% CO2にて37℃で増殖させた。HL-60を好中球様細胞(PMN-HL60)へ分化させるために、培養物に1.5% (v/v)のジメチルスルホキシド(DMSO)を補充し、これを4日間増殖させた。
細胞毒性アッセイ法
黄色ブドウ球菌ロイコシジンAB (LukAB)による傷害後の哺乳類細胞の生存性を評価するため、PMN-HL60細胞を96ウェル平底組織培養処理プレート(Corning)の中に、10%の熱不活化ウシ胎仔血清(FBS)を補充したRPMI (Gibco)の終容量100 ul中1×105個の細胞/ウェルでプレーティングした。細胞を三つ組で、20%〜0.16% v/vに及ぶ黄色ブドウ球菌株Newman由来の培養ろ液の連続2倍希釈液により37℃、5% CO2で2時間、傷害した。野生型菌株由来の細胞外タンパク質およびLukABを欠く菌株(変異体菌株)由来の細胞外タンパク質を用いて、実験を行った。100%生存性の対照には、20% v/vの組織培地(RPMI+10%の熱不活化ウシ胎仔血清)、および20% v/vの黄色ブドウ球菌増殖培地(RPMI+Casアミノ酸)を含めた。0.1% v/v Triton X100を100%細胞死の対照として用いた。傷害の後、CellTiter (Promega) 10μlを各ウェルに加え、細胞を37℃、5% CO2でさらに3時間インキュベートした。CellTiterでは、代謝的に活性な細胞(色の変化)、つまり死細胞で失われる特性をモニタリングする。Perkin Elmer Envision 2103 Multilabel Readerを用い492 nmで比色反応を測定した。以下の方程式を用いて%生存細胞を計算した: %生存性 = 100×[(Ab492 サンプル - Ab492 Triton X)/(Ab492 組織培地)。
LukAB媒介性細胞毒性を測定するための代替的なアッセイ法は、宿主細胞膜の完全性を評価することである。この目的を達成するために、SYTOXグリーン(Invitrogen)透過性アッセイ法を利用した。健常細胞はSYTOXグリーンに対して非透過性であるが、細胞膜の完全性がいったん損なわれると、色素に対して透過性になる。細胞内部で、SYTOXグリーンはDNAに結合し、強力な蛍光を示す。
LukABはヒト初代食細胞を標的化および死滅化する
黄色ブドウ球菌株Newman由来のろ過培養上清での初代ヒト末梢単核球(PBMC) (図3a)、単球、マクロファージ、樹状細胞(図3b)および多形核細胞(PMN) (図3c)の傷害は、細胞毒性アッセイ法(Cell Toxicity Assay)によって調べた場合に強力な細胞死をもたらした(図3a〜c)。溶血素(hla)、γ-ヘモリジン(hlg)、ロイコシジンE/D (LukED)またはロイコシジンA/B (LukAB)を欠く黄色ブドウ球菌株Newman由来のろ過培養上清でのこれらの細胞の傷害から、hla-、hlg-、およびLukED-陰性菌株由来の細胞外タンパク質がNewman野生型(WT)菌株と同じ細胞毒性である(細胞毒性アッセイ法によって調べた場合)ことが明らかにされ、Newmanによって産生される既述のロイコトキシンのどれも、これらの細胞の細胞毒素媒介性の死滅化に寄与しないことが示唆された(図3a〜c)。対照的に、LukABを欠く菌株Newman (ΔlukAB)由来の細胞外タンパク質で細胞を傷害した場合、ごくわずかな細胞死しか観察されなかった。LukABを欠く菌株Newmanによる細胞毒性活性の欠如は、プラスミドにてトランスにlukAB遺伝子を与えること(ΔlukAB/pLukAB)によりレスキューされた(図3c)。重要なことには、この表現型は、精製された組み換えLukAおよびLukBでPMNを傷害することによって判定した場合、LukABに完全に依存している。個々のサブユニットはPMNに対して検出可能な細胞毒性を示さなかった(図3d)。対照的に、両方のサブユニットの組み合わせでは、用量依存的にこれらの細胞に対する強力な細胞毒性が引き起こされた(図3d)。全体として、これらの結果は、感染性因子から宿主を防護するのに必要とされる重要な免疫細胞である初代ヒト食細胞を標的化および死滅化する黄色ブドウ球菌の能力にLukABが関与していることを実証している。
黄色ブドウ球菌株Newman由来のろ過培養上清での好中球様細胞株(PMN-HL60)およびマクロファージ様細胞株(THP1+PMA)の傷害は、細胞毒性アッセイ法(Cell Toxicity Assay)によって調べた場合に強力な細胞死をもたらした(図3)。黄色ブドウ球菌WTならびに同質遺伝子的な細胞毒素変異体菌株(hla、hlgABC、LukED、およびLukAB)由来のろ過培養上清でのこれらの細胞の傷害から、細胞毒性アッセイ法によって判定した場合、これらの細胞を死滅化する黄色ブドウ球菌の能力にLukABが関与していることが明らかにされた(図4aおよび4b)。LukABを欠く菌株Newmanによる細胞毒性活性の欠如は、lukABを発現するプラスミドでLukABを欠く菌株Newmanを形質転換すること(ΔlukAB/pLukAB)によりレスキューされた(図4c)。この菌株由来の細胞外タンパク質はPMN-HL60細胞とTHP-1+PMA細胞の両方に対し極めて細胞毒性であり、LukABが、ヒト細胞を標的化および死滅化する強力なブドウ球菌毒素であるという強力な証拠が示された。
LukBに対して作製されたポリクローナル抗体による免疫ブロット分析から、院内感染および市中感染と関連するMRSA菌株を含む一連のブドウ球菌株(USA300、400、および500; 図5a)によってLukBが産生されることが明らかにされた。重要なことには、LukBレベルはこれらの菌株の細胞毒性表現型と関連している(図5aおよび5b)。高いレベルのLukBを産生する菌株(例えば、Newman、4645、USA 500、およびUSA 300)は、少量のまたは検出不能なLukBを産生する菌株(例えばUSA100およびUSA400)よりもPMN-HL60細胞に対して細胞毒性であった(図5b)。MRSA菌株におけるLukABの役割について詳しく調べるために、臨床分離株USAタイプ300 LAクローンにおいてLukAB同質遺伝子変異体を作出した(図5c)。菌株Newmanで認められるように、LukABを欠く菌株USA300由来の細胞外タンパク質は親菌株由来の細胞外タンパク質よりも細胞毒性が著しく低かった(図5d)。これらのデータは、LukABがMRSA菌株によって産生される重要な細胞毒素であることを実証している。
黄色ブドウ球菌由来の細胞外タンパク質でのPMN-HL60の傷害は、LukABに依る表現型である細胞円形化および核膨潤をもたらした(図6a)。この細胞円形化および膨潤の表現型は、膜損傷アッセイ法(Membrane Damage Assay)によって判定した場合に膜透過性の増大と関連していた(図6bおよび6c)。重要なことには、LukAB陰性菌株由来の細胞外タンパク質は膜透過性、つまりプラスミドからLukABを産生することによってレスキューされた表現型、に及ぼす効果をほとんどまたは全く示さず(図6b)、組み換えLukABでは用量依存的に膜損傷が引き起こされるが、しかし個々の毒素サブユニットでは引き起こされない(図6c)。さらに、メチシリン感受性黄色ブドウ球菌(MSSA)株およびメチシリン耐性黄色ブドウ球菌(MRSA) USA300株の両方での初代ヒトPMNの感染は、LukAB依存的な膜損傷をもたらした(図6d)。これらの結果は、LukABがエクスビボ感染中に宿主細胞の原形質膜を損なうことを実証している。
黄色ブドウ球菌WT、ΔlukAB、およびlukAB発現プラスミドを持つΔlukAB (ΔlukAB/pLukAB)でのPMN-HL60細胞の感染から、膜損傷アッセイ法(Membrane Damage Assay)によって判定した場合に、ブドウ球菌と食細胞との間の相互作用中に食細胞の膜を破壊する黄色ブドウ球菌の能力にはLukABが必要とされること(図7a)が明らかにされた。重要なことには、LukABを過剰産生する黄色ブドウ球菌(ΔlukAB /plukAB)は、WT菌株よりも多くの膜損傷を示したこと(図7a)から、LukABが宿主細胞膜を強力に損なうことが実証された。ヒト全血(図7b)および精製された初代ヒト好中球(PMN; 図7c)の感染から、lukAB陰性のブドウ球菌がWT菌株と比べてさらに効率的に死滅化されることが明らかにされた(図7bおよび7c)。重要なことには、ΔlukAB陰性のブドウ球菌が示す弱毒化された表現型はlukAB発現プラスミドでレスキューされた(図7bおよび7c)。黄色ブドウ球菌WT、ΔlukAB、およびlukAB発現プラスミドを含有するΔlukAB変異体菌株の培養ろ液での初代ヒトPMNの傷害から、LukABが初代ヒトPMNを標的化および死滅化することが明らかにされた(図7d)。これらのデータから、LukABが、膜破壊を通じてPMNを標的化および死滅化する強力なブドウ球菌細胞毒素であり、したがってPMN媒介性の死滅化から黄色ブドウ球菌を防護することが強く示唆される。
LukABプロモーターがルシフェラーゼレポーターに融合されているルシフェラーゼレポーター構築体(pLukAB-Xen1)を含んだ黄色ブドウ球菌を眼窩後方から感染させたマウスは、腎膿瘍においてLukABプロモーター活性を示したが、その一方でプロモーターを持たないレポーター(pXen1)は活性を示さなかった(図8a)。これらのデータは、LukABが感染の腎膿瘍モデルにおいてインビボで発現されることを実証している。さらに、黄色ブドウ球菌WTを眼窩後方から感染させたマウスでは、腎臓での広範囲に及ぶコロニー形成が示されたが、LukABを欠く黄色ブドウ球菌株では示されなかった。LukAB陰性の菌株において観察されたコロニー形成の欠陥は、LukABをトランスに与えることによってWTレベルまで回復した(図8b)。全体として、これらのデータは、LukABが、細菌の病原性に寄与する重要なブドウ球菌細胞毒素であることを示している。
組み換えLukAB (rLukAB)でのPMN-HL60細胞の傷害から、LukAおよびLukB特異的抗体での免疫ブロットによって判定した場合、rLukAおよびrLukBの両方が標的細胞に結合することが明らかにされた(図9a)。蛍光活性化細胞選別(FACS)およびHis特異抗体を用いてPMN-HL60への6His-タグ付rLukABの結合も確認された(図9b)。rLukABオリゴマーがまた、組み換え毒素による傷害後にPMN-HL60膜に対する免疫ブロットにより検出され、LukABが標的細胞膜上に高次構造を形成することが示唆された(図9c)。重要なことには、rLukABでのPMN-HL60の傷害から、LukABが、ポリエチレングリコール分子(PEG)を用いて遮断できる標的細胞膜上の臭化エチジウム透過性の細孔を形成すること(図9d)、およびLukAB細孔の遮断が細胞の生存性を増大すること(図9e)が実証された。さらに、サポニンによりPMN-HL60膜において形成された細孔がPEGによって遮断されなかったことや、結果的に、これらの細胞が細孔媒介性の死から防護されなかったことから、PEGはLukAB細孔を特異的に遮断する。これらのデータは、LukAB細孔を小分子によって遮断できること、およびLukAB細孔の遮断によってLukAB媒介性の死滅化から細胞が防護されることを実証している。
異なる2羽のウサギにおいて作出されたα-LukAポリクローナル抗体の各種量とともにプレインキュベートされた黄色ブドウ球菌培養ろ液でのPMN-HL60の傷害は、用量依存的に培養ろ液の毒性の低下を引き起こした(図10)。この中和効果は、培養ろ液を免疫前血清とともにプレインキュベートした場合、認められなかった。重要なことには、異なる2羽のウサギにおいて作出された抗体は、非常に似通った挙動をし、これらの抗体の中和能は、後期採血由来の抗体の中和効果を初期採血由来の抗体の中和効果と比較することによって認められたように、成熟度とともに増した(図10)。これらのデータは、黄色ブドウ球菌由来の培養ろ液で認められた細胞毒性を、α-LukAポリクローナル抗体で中和できることを示している。
LukAは、これがN末端およびC末端の両方に伸長部を有するという点で他のブドウ球菌ロイコトキシンS-サブユニットとは異なる。この伸長部は、N末端の33アミノ酸およびC末端の10アミノ酸からなる(図11a)。N末端伸長部を欠く精製された組み換えLukA (rΔ33NLukA)と精製されたrLukBとの組み合わせでのPMN-HL60の傷害は、精製されたrLukA+rLukBの細胞毒性に匹敵した細胞に対する強力な細胞毒性をもたらした(図11b)。しかしながら、C末端伸長部を欠く精製された組み換えLukA (rLukAΔ10C)とrLukBとの組み合わせでのPMN-HL60の傷害は、細胞毒性効果をもたらさなかった(図11b)。これらのデータから、N末端伸長部はLukAの細胞毒性効果にとって重要ではないが、C末端伸長部は毒性に必要であることが実証される。重要なことには、LukABの効果を中和するα-LukAポリクローナル抗体(図10)は、α-Hisポリクローナル抗体と全く同じように6×Hisタグ付の非細胞毒性rLukAΔ10C変異体を依然として認識する(図11c)。これらのデータは、中和抗体であるα-LukAポリクローナル抗体をインビボで作出するためにrLukAΔ10Cを活用できることを示唆している。それゆえ、rLukAΔ10Cを能動ワクチン組成物において用いることができる。
Claims (1)
- a) SEQ ID NO:1〜14のいずれかのアミノ酸配列、もしくはSEQ ID NO:1〜14のいずれかと少なくとも70%の配列類似性を有する配列を有するLukAポリペプチド(「LukA」)の治療的有効量、b) SEQ ID NO:15〜27のいずれかのアミノ酸配列、もしくはSEQ ID NO:15〜27のいずれかと少なくとも70%の配列類似性を有する配列を有するLukBポリペプチド(「LukB」)の治療的有効量、c) LukAに特異的に結合する抗LukA抗体の治療的有効量、または、d) LukBに特異的に結合する抗体の治療的有効量のうちの少なくとも1つを含む、黄色ブドウ球菌(Staphyloccocus aureus)感染の発症を阻止するまたは黄色ブドウ球菌感染を処置するのに有用な活性薬剤。
