インスリン様成長因子−1(IGF−1)は、哺乳動物中枢神経系(CNS)における細胞の発達および生存において決定的な役割を果たすため、このタンパク質は、CNSに影響を及ぼす種々の状態のための潜在的に重要な治療薬と考えられてきた。動物モデルでは、ウイルスベクターおよびくも膜下腔内注射を含むいくつかの方法によるIGF−1の送達が、ALSの治療にとって有望であるとされている。しかし、臨床治験では、ヒト患者への成熟組換えIGF−1の皮下投与は、ALSの治療において有効性を実証しなかった。したがって、神経細胞喪失部位へ治療有効量のIGF−1を送達する改善された方法に対する必要性が存在する。

本開示は、例えば、神経栄養因子を含む成長因子をコードする外因性ポリヌクレオチドを含む神経幹細胞(例えば、胎児または胚に由来するヒト神経幹細胞)であって、成長因子が神経幹細胞によって安定に発現される、神経幹細胞を提供する。本発明者らは、驚くべきことに、神経幹細胞が神経細胞喪失部位に生着し、治療有効量で、例えば、成熟IGF−1などの神経栄養因子を含む成長因子を安定に発現することができることを発見した。神経栄養因子として、例えば、インスリン様成長因子1(IGF−1)(例えば、配列番号1に示される配列を有するIGF−1アイソフォーム)、グリア細胞株由来神経栄養因子(GDNF)、脳由来神経栄養因子(BDNF)、ニューロトロフィン−3(NT−3)または血管内皮細胞成長因子(VEGF)を挙げることができる。しかし、本開示において使用するための、神経幹細胞によって分泌され得る任意のタンパク質が企図される。このようなヒト神経幹細胞は、神経幹細胞株から得ることができ、それだけには限らないが、筋萎縮性側索硬化症(ALS)、脊髄損傷(SCI)、外傷性脳傷害(TBI)、アルツハイマー病(AD)、認知症、軽度認知障害、糖尿病、糖尿病関連CNS合併症、末梢神経障害、レチナール神経障害および多発性硬化症を含めた種々のCNS適応症を含む神経変性性疾患または障害の治療のために使用され得る。
神経栄養因子などの成長因子をコードする外因性ポリヌクレオチドを含む神経幹細胞(例えば、安定なヒト神経幹細胞)が提供される。神経栄養因子は、インスリン様成長因子1(IGF−1)、グリア細胞株由来神経栄養因子(GDNF)、脳由来神経栄養因子(BDNF)、ニューロトロフィン−3(NT−3)または血管内皮細胞成長因子(VEGF)であり得る。また、成長因子をコードする外因性ポリヌクレオチドを有する神経幹細胞を含む神経幹細胞株も提供される。神経幹細胞は、好ましくは、安定であり、60回超の細胞倍加後でさえ培養において分化しない。
本明細書において開示されるような神経幹細胞は、筋萎縮性側索硬化症(ALS)、脊髄損傷(SCI)、外傷性脳傷害(TBI)、アルツハイマー病(AD)、認知症、軽度認知障害、糖尿病、糖尿病関連CNS合併症、末梢神経障害、レチナール神経障害または多発性硬化症などの神経変性性疾患または障害を含めた疾患または障害を治療するための方法において使用され得る。このような方法は、例えば、注射によるものを含めて、対象に、治療有効量の、本明細書において開示される神経幹細胞を投与することを含み得る。一実施形態では、開示される神経幹細胞を用いて治療された対象は、神経幹細胞の投与の前に、その間および/またはその後に免疫抑制される。
(材料および方法)
(HK532調製)
ヒトHK532 NSC株(NSI−HK532およびNSI−HK532.UbC−IGF−I)は、Neuralstem,Inc.(Rockville、MD)によって提供された。手短には、HK532は、人工妊娠中絶後の胎齢8週のヒト胎児から得られた皮質組織から調製した。材料は、Neuralstem,Inc.に献体され、国立衛生研究所(National Institutes of Health)(NIH)およびFDAのガイドラインに従ってインフォームドコンセントを行った。ガイドラインは、記載されるように外部の独立した審査委員会によって審査され、承認された(Joheら(1996年)Genes Dev.、10巻(24号):3129〜40頁)。不死化遺伝子およびネオマイシン耐性遺伝子を含有するレトロウイルスベクターを使用して、皮質NSCを条件的に不死化した。不死化遺伝子は、3’末端でヒトエストロゲン受容体のC末端リガンド結合ドメインをコードするcDNA断片と融合しているヒトc−myc cDNAを含んでいた。細胞をネオマイシン耐性について選択し、単細胞株(HK532)として増殖させた。次いで、細胞株を複製欠陥組換えレンチウイルスベクターを用いて形質導入して、ヒトユビキチンC(UbC)プロモーターによって駆動されるヒトIGF−Iの発現を誘導した。得られた細胞を単細胞株として増殖させ、さらなる選択は行わなかった(HK532.UbC−IGF−I)。同一UbCプロモーター下で緑色蛍光タンパク質(GFP)を発現する対照構築物を使用するHK532の形質導入は、およそ90〜95%のGFP陽性増殖性細胞をもたらした。
HK532およびHK532−IGF−I細胞両方の培養を、これまでに記載されたように実施した[42]。手短には、細胞を、10mM Hepesバッファー中の、100μg/mLのポリ−D−リシン(Millipore、Billerica、MA)を用いて24時間、続いて、PBS中の、25μg/mLのフィブロネクチンを用いて1時間コーティングされたフラスコ上で成長させた。あるいは、皮質ニューロン(CN)との共培養に先立って、ポリ−L−リシンでコーティングしたインサート上に細胞を播種した。細胞を、前駆細胞状態成長および維持のために、10ng/mLの線維芽細胞成長因子(FGF)を補給したN2B+培地(Neuralstem,Inc.、Rockville、MDによって供給される)で培養した。分化のために、細胞をFGFを含まないNSDM分化培地(4mMのL−グルタミン、20μMのL−アラニン、6μMのL−アスパラギン、67μMのL−プロリン、250nMのビタミンB12、25mg/Lのインスリン、100mg/Lのトランスフェリン、20nMのプロゲステロン、100μMのプトレシンおよび30nMの亜セレン酸ナトリウムを補給したDMEM)中で培養した。分化した細胞データは、分化後日数として示されている(すなわち、未分化(D0)、1日目(D1)、3日目(D3)など)。2日毎に培地を変更し、50%の培地を変更した。
HK532およびHK532−IGF−I細胞において、先に記載されたようにELISAおよびウエスタンブロッティングによってIGF−I発現およびシグナル伝達を調べた(Vincentら、Endocrine Society Abstracts(2003年)、P3−316 548頁;およびChiaら、Am J Epidemiol(2008年)、167巻(12号):1438〜45頁)。手短には、IGF−I産生を確認するために、未分化(D0)および分化した(D3およびD7)HK532およびHK532−IGF−I細胞からコンディショニング培地を集め、Centriconフィルター(3KDaカットオフ、Millipore、Billerica、MA)を使用して1mLに10倍濃縮し、ヒト特異的IGF−I ELISA(Assay Designs、Enzo Life Sciences Inc.、Farmingdale、NY)を製造業者の指示に従って実施した。IGF−Iシグナル伝達分析のために、HK532およびHK532−IGF−I細胞を、処理培地(インスリンが添加されていないNSDM分化培地)中で4時間培養し、その後、選択阻害剤を1時間添加し、その後、外因性IGF−I(20nM)を30分間添加した。阻害剤は、Akt経路阻害剤LY294002(LY;20μM;Sigma−Aldrich、St.Louis、MO)、MAPK阻害剤U0126(U;20μM;Calbiochem、La Jolla、CA)またはIGF−IR阻害剤NVPAEW541(NVP;1μM;Sigma−Aldrich)を含んでいた。ウエスタンブロットのために、氷冷RIPAバッファー(20mM Tris、pH7.4、150mM NaCl、1mM EDTA、0.1%SDS、1mMデオキシコール酸Na、1%Triton X−100、0.1トリプシンユニット/L アプロチニン、10mg/mLロイペプチンおよび50mg/mL PMSF)中で総細胞タンパク質を抽出し、タンパク質濃度を調べ、試料をSDS−PAGEゲルで電気泳動し、ニトロセルロースに移した。一次抗体(特に断りのない限り、Cell Signaling Technology,Inc.(Danvers、MA)から得られた)は、ホスホ−IGF−IR(pIGF−IR)、IGF−IRβ(Tyr1135/1136)、ホスホ−Akt(Ser473)(pAkt)、Akt、ホスホ−ERK(pERK)、ERKおよびβ−アクチン(Chemicon、Temecula、CA)を含んでいた。一次抗体を4℃で一晩インキュベートした後、膜を、西洋ワサビペルオキシダーゼとコンジュゲートしている適当な二次抗体(Cell Signaling Technology、Danvers、MA)とともに22℃で1時間インキュベートし、化学発光基質(SuperSignal West Pico;Pierce、Fisher Scientific、Hampton、NH)を用いて発色させ、Kodak BioMax XARフィルム(Sigma−Aldrich)に曝露した。
未分化HK532およびHK532−IGF−I細胞を、4℃で一晩貯蔵した後(1×106細胞/mLまたは3×106細胞/バイアル)、遊走用インサートに添加するか、あるいは、代わりに、6ウェルプレート上で培養して、分化のD7でインサートに移した。IGF−I(10nMの最終濃度)を含むか、または含まないNSDMおよび10%FBSをインサートの下に添加した。24時間後、QCM 24ウェル比色定量細胞遊走アッセイ(Millipore)を使用してインサートを通って遊走した細胞を染色した。遊走は、530および590nmで標準LabSystems Fluoroskan Ascent FLマイクロプレートリーダーを使用して定量した。
細胞増殖および分化を、標準実験室免疫細胞化学(ICC)プロトコールを使用して評価した(Kimら、Journal of Biological Chemistry(1997年)、272巻:21268〜21273頁;Lunnら、Neurobiol Dis(2012年)、46巻(1号):59〜68頁)。手短には、HK532およびHK532−IGF−I細胞を、24ウェルプレート中、ポリ−L−リシンおよびフィブロネクチンコーティングしたガラスカバースリップ上で培養した。細胞を、10μM 5’−エチニル−2’−デオキシウリジン(EdU)とともに2時間インキュベートし、その後、Click−It EdUキット(Invitrogen)の製造業者のプロトコールに従って固定および処理することによって、細胞増殖を、D0、D3およびD7でこれまでに記載されたように測定した[45]。デジタルカメラを備えたOlympus BX−51顕微鏡を使用してとられた蛍光画像の定量化によってEdU組込みを測定した。すべての試料について増殖実験あたりおよそ2.5〜2.7×103個の細胞をカウントした(n=3)。
本発明者らのこれまでに公開されたプロトコールに従って一次CNを単離した(Lunnら、Stem Cells Dev(2010年)、19巻(12号):1983〜93頁)。手短には、E15 Sprague−Dawleyラット胚から得たCNを集め、膜を除去し、組織を2〜3mm片に刻んだ。0.5%トリプシン/EDTA中、37℃で10分間組織をインキュベートし、続いて、血清でコーティングされたガラスピペットを用いて1分間トリチュレートすることによって、細胞を解離した。得られた細胞懸濁液を、24ウェルプレート中で、ポリ−L−リシンコーティングされたガラスカバースリップ上に塗布し、2.5mg/mlアルブミン、2.5μg/mlカタラーゼ、2.5μg/ml SOD、0.01mg/mlトランスフェリン、15μg/mlガラクトース、6.3ng/mlプロゲステロン、16μg/mlプトレシン、4ng/mlセレン、3ng/ml β−エストラジオール、4ng/mlヒドロコルチゾン、1×ペニシリン/ストレプトマイシン/ネオマイシン(Gibco BRL)および1×B−27添加剤(Gibco BRL)を補給したNeurobasal培地(Gibco BRL、Invitrogen)を含んでいた成長培地中でインキュベートした。
in vivo移植後にHK532−IGF−I細胞が生存し、海馬領域中に組み込まれることを実証するために、Jackson Laboratory(Bar Harbor、ME)から、6週齢のB6C3−Tg(APPswe/PSEN1ΔE9)85Dbo/J(APP/PS1;n=5)および野生型B6C3F1/J(WT;n=8)マウスを得た。11週齢で、マウスに皮下タクロリムスペレット(FK−506;Neuralstem,Inc.によって供給される)を与え、12週齢で細胞移植手術を実施した。手短には、イソフルオランを用いてマウスに麻酔し、標準定位固定フレーム(Stoelting Company、Wood Dale、IL)に入れた。皮膚を切開し、予想される注射の領域で大きな開頭術を実施した。十字縫合から測定される以下の座標(それぞれ、後側/側面/腹側):−0.82/±0.75/2.5、−1.46/±2.3/2.9、−1.94/±2.8/2.9に従って表される3部位での海馬采脳弓への両側注入(合計6回の注射)によってHK532−IGF−I細胞懸濁液を投与した。各注射は、30,000細胞/μLの細胞濃度で1μLの容量からなっていた(60秒かけて投与され、ニードルを引き抜く前に60秒遅れさせる)。次いで、吸収性縫合糸を使用して皮膚を閉じた。手術後、マウスに腹膜内麻薬性鎮痛薬を2日間与え、研究を通じてタクロリムスペレットを継続した。細胞移植後2および10週で、分析のためにマウスを屠殺した。手短には、動物に麻酔し、氷冷生理食塩水を用いて灌流し、脳を解剖し、半球間境界に沿って切開した。脳を4%PFA中で一晩、後固定し、免疫組織化学分析(IHC)のために30%スクロース中で凍結保護した。
すべてのデータは、平均±標準偏差(SD)、n=3として、または3回の独立実験の代表画像として示されている。GraphPad Prism(GraphPad Software,Inc.、La Jolla、CA)を使用して統計分析を実施した。ペアワイズ比較のために対応t検定を使用した。p<0.05の値を統計上有意と考えた(*p<0.05)。
(IGF−I産生およびシグナル伝達)
HK532細胞は、これまでに記載されていない新規皮質NSCである。細胞治療薬としてのそれらの可能性ある効き目を増強するために、およびそれらの神経保護能に対するIGF−I産生の影響を調べるために、全長ヒトIGF−Iをコードするレンチウイルスベクターを使用して、HK532−IGF−I細胞株を生成した。コンディショニング培地のELISA分析は、親HK532細胞は、極めて少ない〜検出不能な基礎レベルのIGF−Iしか産生しないのに対し、HK532−IGF−I細胞は、D0からD7の間に3〜5ng/mLのIGF−Iを産生する、およそ50倍の増大(図1A)を実証した。したがって、HK532−IGF−I細胞は、適切なレベルのIGF−Iを産生し、これは初期分化を通じて維持され、頑強で安定なIGF−I発現が確認される。
(IGF−I発現は、HK532増殖または遊走を変更しない)
EdU組込みを使用して、HK532およびHK532−IGF−I細胞増殖に対するIGF−Iの効果を評価した。未処理D0 HK532およびHK532−IGF−Iのおよそ36%および33%が、それぞれEdU陽性であった(図2A〜E)。D3で、HK532およびHK532−IGF−Iの6%および9%が、それぞれEdU陽性であり、D7までに、いずれかの細胞株の3%未満が、EdU陽性であった。したがって、D0、D3またはD7で増殖プロファイルの相違は観察されず、両株がD7で最小の増殖を示した。これらのデータは、IGF−Iが、分化の最初の段階の間に増殖を促進または維持しないことを実証する。
(IGF−Iを発現するHK532は、神経分化能を保持する)
次いで、HK532前駆細胞状態の維持および軸索伸長に対するIGF−Iの効果を調べた。D0 HK532およびHK532−IGF−Iのおよそ92%および90%が、それぞれNestin陽性であり、これは、IGF−I発現が前駆細胞状態の維持に影響を及ぼさなかったことを示す。ニューロン分化の初期指標として確立された神経指数アプローチを使用して神経突起成長に対するIGF−Iの効果も評価した。HK532およびHK532−IGF−I両細胞について、細胞が分化するにつれ、D0およびD7の間に神経指数は増大し、試験された任意の時点で細胞株間で相違は観察されなかった(図3F)。これらのデータは、IGF−Iが、初期HK532分化に影響を及ぼさないことを実証する。
(HK532−IGF−Iでは、GABA作動性であって、グルタミン酸作動性ではない表現型が増大される)
最終分化に対するIGF−Iの効果を調べるために、D0、D3およびD7でのグルタミン酸作動性(VGLUT)およびGABA作動性(GAD65)表現型を示す細胞の割合。GAD65陽性細胞を数量化し、それぞれ、総細胞の74%および67%で、親HK532細胞と比較してHK532−IGF−Iにおいて大幅に増大した(図4A、B、E)。HK532(61%)およびHK532−IGF−I(67%)培養物中のVGLUT陽性細胞のパーセンテージは、大幅に異ならなかった(図4C、D、F)。これらのデータは、IGF−Iの存在が、細胞分化に起因するGABA作動性ニューロン数を増大させるが、グルタミン酸作動性ニューロン数に対しては有意な効果を有さないこと実証する。
(HK532−IGF−Iは、in vitroで、Aβ毒性に対して耐性であり、一次CNを保護する)
Aβ(1−42)は、AD関連毒性のよく使用されるin vitroモデルである(Bruceら(1996年)、PNAS 93巻(6号):2312〜6頁)。Aβに曝露された場合に、一次CNおよび両NSC株において大幅なアポトーシスおよびCC3活性化が観察された(図5A)。HK532およびHK532−IGF−Iにおけるアポトーシスレベルは、一次CNにおいて観察されたものよりも大幅に低かった(p<0.05;図5A)。修飾された前駆細胞の保護能を調べるために、HK532およびHK532−IGF−Iとともに間接的に共培養された一次CNにおいてAβ毒性も評価した(図5B〜E)。CC3活性化によってやはり示される一次CNにおけるアポトーシスは、HK532とともに共培養した場合には40%未満に、HK532−IGF−Iとともに共培養した場合には30%未満に大幅に低減した(p<0.05;図5F)。これらのデータは、HK532−IGF−I細胞株が、神経保護性であり、Aβ誘導性一次CN死を防止可能であることを示す。
(ADマウスモデルにおいてHK532−IGF−Iは移植を生き抜き、in vivoで組み込まれる)
前臨床試験の実現可能性を確立するために、HK532−IGF−I細胞を、APP/PS1二重トランスジェニックマウス、ADのよく使用されるモデルに移植した(Caoら、J Biol Chem(2007年)、282巻(50号):36275〜82頁)。このパイロット研究は、海馬の海馬采脳弓における細胞の正確で妥当な解剖学的配置を確認するように働き、移植された細胞の生存を経時的に評価した。注射されたすべての動物において標的化の正確性は達成された。移植されたヒト細胞は、HuNuおよびDCXについて2週間で(データは示さず)および移植後10週間でIHCによって検出された(図5E、F)。グラフトされた細胞は、両ADの海馬領域(図5E)およびWT動物(図6F)において明白であった。ニューロン前駆体を標識する微小管結合リン酸化タンパク質であるDCXによるHuNu標識細胞の同時染色は、神経発生を示し、移植された皮質前駆細胞は、初期ニューロン分化相にあったことを示唆する。
(HK532−IGF−Iの投与は、ADマウスモデルにおいてin vivoでAβプラーク形成を低減する)
Aβ病理に対するin vivo HK532−IGF−I移植の全体的な効果を評価するために、5種のAβアイソフォーム(Aβ−37、38、39、40および42)に対するポリクローナル抗体を用いてマウスあたり複数の海馬および皮質切片で免疫染色を実施した。総免疫反応性領域の尺度および強度に基づいて、切片の蛍光画像を定量化した。予測されたように、結果は、媒体が注射されたAPP/PS1マウスにおける明確なAβプラーク形成および非tg動物には、Aβがないことを示す(図6A〜B、D〜E)。さらに、HK532−IGF−Iを用いて処理されたAPP/PS1マウスでは、媒体が注射されたAPP/PS1マウスと比較してAβレベルの大幅に有意な低減があった(P<0.0001;対応のないt検定)(図6B〜C、E〜G)。このデータは、HK532−IGF−Iが、Aβ誘導性損傷から神経組織を保護するように機能するだけではなく、Aβ沈着物を排除する、および/またはAβが蓄積しにくくすることによってAβの沈着を減弱したことを示す。
(HK532−IGF−Iの投与は、ADマウスモデルにおいてin vivoでコリン作動性活性を増大する)
本発明者らのADモデルにおけるコリン作動性ニューロンの存在を評価し、これらのニューロンに対するHK532−IGF−I移植の効果を調べるために、各マウスに由来する線条体切片をChATに対する抗体を用いて免疫染色して、コリン作動性ニューロンを発現するChATの強いレベルを同定した(図7A〜D)。本発明者らは、各切片について線条体の全体を画像化し、各々のChAT陽性細胞の数をカウントした。細胞カウントは、APP/PS1マウスにおいて、WTと比較して線条体コリン作動性ニューロンの大幅な喪失を示した(P=0.0115;対応のないt検定)(図7E)。さらに、NSC処理APP/PS1マウスにおいて、媒体が注射されたADマウスと比較してコリン作動性ニューロンの数の大幅な増大があった(P=0.0366;対応のないt検定)(図7F)。これらの結果は、APP/PS1マウスにおけるHK532−IGF−I移植による線条体におけるコリン作動性機能のレスキューを示す。
(HK532−IGF−Iの投与は、ADマウスモデルにおいてin vivoで前シナプス活性を増大する)
HK532−IGF−Iが、APP/PS1マウスにおいてシナプスの密度を増大するか否かを調べるために、すべての動物から得た海馬切片を、前シナプスマーカー、シナプトフィジンを用いて免疫染色した。HK532−IGF−Iが移植されたAPP/PS1マウスの海馬で、媒体が注射されたトランスジェニックマウスと比較して、蛍光強度において識別可能な増大が見られた(図8A〜F)。この増大された強度は、注射されていない、および偽処理された非tgマウスの両方において見られたレベルと同等であった。これらのデータは、HK532−IGF−I移植が、ADにおいてシナプスを修飾することによって記憶および認知をレスキューすることを示す。
(脊髄におけるHK532.UbC−IGF1の生存および組込み)
脊髄における生存および組込みを実証するために、HK532.UbC−IGF1を、SOD1G93Aラット、筋萎縮性側索硬化症(ALS)の確立された動物モデルの頸髄中に移植した。各動物に、頸髄の前角(C4〜C6脊髄レベル)を標的化して合計1.8×105個の細胞を注射した。一時的なミコフェノール酸モフェチル(30mg/kg IP、グラフト後7日間)によって、および連続タクロリムス送達によって動物を免疫抑制した。動物は56日生存しており、その後、標準灌流−固定した。凍結免疫組織化学分析は、前角におけるヒト細胞移植片の存在および灰白質および白質中の広い分布を示す(図9a〜c)。

Claims (25)
- インスリン様成長因子1(IGF−1)をコードする外因性ポリヌクレオチドを含む安定なヒト神経幹細胞。
- IGF−1をコードする外因性ポリヌクレオチドを含まない神経幹細胞と比較して、有意に増大した数のGAD65陽性GABA作動性ニューロンに分化可能である、請求項1に記載のヒト神経幹細胞。
- 成長因子をコードする外因性ポリヌクレオチドを含むヒト神経幹細胞。
- 前記成長因子が、インスリン様成長因子1(IGF−1)、グリア細胞株由来神経栄養因子(GDNF)、脳由来神経栄養因子(BDNF)、ニューロトロフィン−3(NT−3)および血管内皮細胞成長因子(VEGF)からなる群から選択される神経栄養因子である、請求項3に記載のヒト神経幹細胞。
- IGF−1がIGF−1アイソフォームである、請求項4に記載のヒト神経幹細胞。
- 前記IGF−1アイソフォームがIGF−1アイソフォーム4である、請求項5に記載のヒト神経幹細胞。
- 前記IGF−1アイソフォーム4が、配列番号1に示されるヌクレオチド配列を含む、請求項4に記載のヒト神経幹細胞。
- 皮質、海馬、視床、中脳、小脳、後脳、脊髄および後根神経節からなる群から選択される組織に由来する、請求項3に記載のヒト神経幹細胞。
- 胎児または胚から得られる、請求項3に記載のヒト神経幹細胞。
- 胎齢が約5〜約20週である胎児から得られる、請求項9に記載のヒト神経幹細胞。
- 前記成長因子をコードする前記外因性ポリヌクレオチドが、ユビキチンC(UbC)プロモーター、ヒトホスホグリセリン酸キナーゼ1プロモーター、ヒトシナプシンプロモーターまたは合成CAGプロモーターと作動可能に連結している、請求項3に記載のヒト神経幹細胞。
- インスリン様成長因子1(IGF−1)をコードする外因性ポリヌクレオチドを含むヒト神経幹細胞であって、IGF−1が配列番号1に示されるヌクレオチド配列を含み、前記IGF−1ヌクレオチド配列が安定に発現される、ヒト神経幹細胞。
- 神経変性性疾患または障害の治療のための方法であって、対象に、治療有効量の、成長因子をコードする外因性ポリヌクレオチドを含む1または複数のヒト神経幹細胞を投与することを含み、前記成長因子が安定に発現される、方法。
- 前記成長因子が、インスリン様成長因子1(IGF−1)、グリア細胞株由来神経栄養因子(GDNF)、脳由来神経栄養因子(BDNF)、ニューロトロフィン−3(NT−3)および血管内皮細胞成長因子(VEGF)からなる群から選択される神経栄養因子である、請求項13に記載の方法。
- IGF−1がIGF−1アイソフォームである、請求項14に記載の方法。
- 前記IGF−1アイソフォームがIGF−1アイソフォーム4である、請求項15に記載の方法。
- 前記IGF−1アイソフォーム4が、配列番号1に示されるヌクレオチド配列を含む、請求項16に記載の方法。
- 前記神経変性性疾患または障害が、筋萎縮性側索硬化症(ALS)、脊髄損傷(SCI)、外傷性脳傷害(TBI)、アルツハイマー病(AD)、認知症、軽度認知障害、糖尿病、糖尿病関連CNS合併症、末梢神経障害、レチナール神経障害または多発性硬化症である、請求項13に記載の方法。
