Abstract
Description
1)等温増幅と圧電検知を組み合わせて連動検出するため、装置の構造が簡略化されるとともに、検出効率が向上する。また、圧電センサの高特異性によって、核酸検出の感度がいっそう高まり、検出限界が0.1pg/mLにまで達する。
Claims (10)
- 圧電センサと、当該圧電センサの表面を通過する少なくとも1本のダクトを含み、検出対象の核酸が液状で当該ダクトに導かれ、前記圧電センサの表面を通過し、
前記ダクトには、圧電センサよりも上流側に2つの弁が間隔を置いて設けられており、検出対象の核酸は、圧電センサを通過する前に2つの弁の間のダクト内に密封されて等温増幅されることで、増幅された検出対象の核酸が圧電センサによって検知可能となる核酸検出板。 - 前記2つの弁は、それぞれ前記圧電センサに近い第1の弁と、前記圧電センサから離れた第2の弁であり、第2の弁の上流側には更に三方弁が設けられており、三方弁のうち一方の分岐口には試料セルが接続されており、当該試料セルは、検出対象の核酸と、核酸増幅のための助剤、及び増幅した検出対象の核酸が圧電センサにより検知されるのを助ける助剤を載置するために用いられ、三方弁の他方の分岐口は前記核酸検出板の外部に接続されており、ダクト内に洗浄液又は緩衝液を導入するために用いられることを特徴とする請求項1記載の核酸検出板。
- 前記ダクトには、圧電センサよりも下流側に廃液プールが接続されており、当該廃液プールは、圧電センサにより検知された検出対象の核酸を回収し、汚染を防止するために用いられることを特徴とする請求項2記載の核酸検出板。
- 請求項3記載の核酸検出板と、
当該核酸検出板を収容可能なインキュベータ、及び
前記圧電センサのデータに接続可能な信号プロセッサを含む核酸検出システム。 - 前記核酸検出システムは、液体がダクト内で流動するよう駆動するための注入ポンプを更に含むことを特徴とする請求項4記載の核酸検出システム。
- 前記第1の弁、第2の弁及び三方弁はいずれも電気制御弁であり、
前記核酸検出板の上方には昇降台が設けられており、当該昇降台における前記第1の弁、第2の弁及び三方弁と対向する位置には伸縮可能な第1のプローブがそれぞれ設けられており、選択的に電気制御弁の開閉を誘発することを特徴とする請求項4記載の核酸検出システム。 - 前記昇降台における前記圧電センサと対向する位置に伸縮可能な第2のプローブが設けられており、圧電センサと信号プロセッサのデータ接続を選択的に制御することを特徴とする請求項6記載の核酸検出システム。
- 前記インキュベータにおける前記試料セルの上方と正対する部分には、試料投入口が開設されていることを特徴とする請求項4記載の核酸検出システム。
- 前記試料投入口には密封材が充填されていることを特徴とする請求項8記載の核酸検出システム。
- 前記インキュベータの表面には、管配線のための配線孔が更に設けられ、当該配線孔内に密封材が充填されていることを特徴とする請求項4記載の核酸検出システム。
