Abstract
Description
したがって本発明の第一の態様は、
(i)10〜16個のアミノ酸からなり;
(ii)
(a)グアニジニウム基を含む側鎖、または
(b)アミノ基を含む側鎖
のいずれかをもつ、少なくとも8個のカチオン性アミノ酸を有し、該カチオン性アミノ酸の7個以下が連続しており;
(iii)連続していない2個のトリプトファン残基、または全てが連続してはいない3個のトリプトファン残基を有するペプチド;または、上記ペプチドのペプチド疑似体を提供する。

アミノ基を含む側鎖を持つ好ましいアミノ酸は、リシン(3文字コードおよび1文字コードでLysまたはK)およびアミノカプロン酸やオルニチンなどのリシンの誘導体(側鎖は1以上のアミノ基を含んでいてもよい)である。アミノ基(‐NH2)は通常線状の(または分岐した)アルキル基(例えば、C2−C8アルキル)に結合しており、側鎖は通常他のヘテロ原子を含まない。
好ましくは本発明のペプチドは6個以下、さらに好ましくは5個以下の連続した、本明細書で定義したようなカチオン性アミノ酸を有する。
本発明のペプチドは好ましくは少なくとも8個のグアニジウム含有残基を含み、好ましくは全てのカチオン性アミノ酸がこの種のものである。あるいは、ペプチドは、そのカチオン性アミノ酸の全てが側鎖にアミノ基を含む種類のもの(例えばLys)であってもよいし、2種類のカチオン性アミノ酸の混合物、例えばアミノ基を含む側鎖を有する1〜3個のカチオン性アミノ酸であってもよい。
好適なペプチド疑似体としては、ボラン、または水素化アルミニウムリチウムなどのヒドリド試薬による還元剤処理により、アミド結合がメチレンアミンに還元された還元型のペプチドが挙げられる。そのような還元は、分子の全体的なカチオン性を増加させるという更なる利点を有する。
治療は、上述したように、腫瘍の成長を軽減すること、例えば腫瘍の大きさを、例えば少なくとも20%、好ましくは少なくとも30または50%、さらに好ましくは少なくとも70%縮小することを含む。治療により腫瘍が根絶することもあるが、腫瘍が根絶しなくとも治療が成功したと判断することもできる。主要な目標は平均余命を伸ばし、および/またはクオリティーオブライフを改善することである。リンパ腫と診断された後の予後は臨床医により評価することができ、生存率の改善は、1年、5年、または10年のどの時点でも、データまたは余命に対して判断すればよい。
活性成分は、必要に応じて他の活性剤と組み合わせて、錠剤、ピル,粉薬(例えば吸入用粉薬)、トローチ、分包剤、カプセル、エリキシエル〔elixir〕、懸濁液、乳濁液、クリーム、泡状剤、ゲル、溶液、シロップ剤、エアロゾル剤(固体または液状の媒質内)、ローション、軟膏、軟質および硬質のゼラチンカプセル、座薬、滅菌した注射用薬剤、滅菌した分包粉末剤などの従来のガレヌス製剤〔galenic preparations〕を製造するための、1以上の従来の担体、希釈剤および/または賦形剤と一体化してもよい。
非経口的に投与できる形態は、滅菌されており、生理学的に許容できない薬剤が入っていないものとすべきであり、また投与時の刺激または他の副作用を最小限にするためにモル浸透圧濃度が低いものとすべきであり、それゆえ溶液は、好ましくは等張であるか、わずかに高張であるもの、例えば高張塩水(生理食塩水)とすべきである。適当な担体としては、非経口的溶液の投与に慣習的に使われている、生食注射液、リンガー液、ブドウ糖注射液、ブドウ糖/生理食塩水注射液、乳酸リンゲル注射液、Remington’s Pharmaceutical Sciences, 15th ed., Easton: Mack Publishing Co., pp. 1405−1412および1461−1487 (1975)およびThe National Formulary XIV, 14th ed. Washington: American Pharmaceutical Association (1975)に記載されているような他の溶液などの、水性の担体が挙げられる。溶液は非経口溶液に通常用いられる保存料、抗菌剤、バッファーおよび抗酸化剤や、バイオポリマーに適合し、製品の製造、保存、または使用に支障をきたさない、賦形剤および他の添加剤を含んでもよい。
医薬用途においては、 ペプチドまたは疑似体は通常5〜500mg/l、好ましくは20〜100mg/l、例えば40〜60mg/lの濃度で簡単に投与することができる。
材料および方法
使用した化学物質および試薬
Annexin V−FITC(Biovision)、ヨウ化プロピジウム(Sigma−Aldrich)、トリブチルすずクロリド(Sigma−Aldrich)、RPMI−1640(Sigma−Aldrich)、テトラメチルローダミンメチルエステル過塩素酸塩(Invitrogen)、ハンクス平衡塩類溶液(Sigma Aldrich)、Caspase Fluorometric Substrate Set II Plus kit (Biovision)。
全てのペプチドは標準的なFmoc合成プロトコールおよびアミノ酸誘導体を用いて、ペプチド合成装置Tribute(Protein Technologies社)により合成した。Tbt β−(2,5,7−トリ−tert−ブチルインドール−3−イル)アラニン)のFmoc−誘導体はHaug et al. (2008) J. Med. Chem. 51(14) pp. 430614に記載された方法で調製した。
C18(20×250mm、5μm、100Å)Inersil ODS−3カラム(GL Sciences社)を用いたウォーターズ社の600E セミ分取HPLCシステムで原料を精製した。ペプチドの純度はC18(4.6×250mm、5μm、100Å)Sunfireカラムを用いた分析用RP−HPLC(ウォーターズ社 2695システム)で測定した。1.0ml/分の流量で0〜50%Bのグラジエントで30分、214nmにセットしたピークを検出することで解析を行った。全てのHPLCの溶媒(水およびアセトニトリル)は0.1%TFAを含んでいた。試験に用いた全てのペプチドは95%以上の純度を有していた。
用いられた全ての細胞株はマイコプラズマ試験を行い、5%CO2、37℃で培養した(Hera Cell 150 incubator、Thermo Electron Corp.)。用いられた種々の細胞株(ATCC番号および継代数を含む)は以下の表に記載されている。
2人の健康なドナーから採取した血液サンプルにヘパリン処理を行い、洗浄した。RBCをPBSで再懸濁し、ヘマトクリット値が10%になるまで希釈した。RBCを直ちにペプチドで1時間37℃で処理した。処理後に細胞をスピンダウンし、上清100μLを96ウェルのプレートに移した。405nmにおける吸光度を測定した。
A20およびMRC−5に対する最初のスクリーニングは手作業で行ない、リンパ腫パネルスクリーンは自動化プラットフォーム(Biomek NX、 Beckman Coulter社)を用いてMarbio(Tromsoe)において行った。
このアッセイはMTTアッセイに準じて、およびToerfoss et al. in J. Pept. Sci 2012, 18(3):p.1706)を基にして行った。簡潔にいうと、A20細胞は最終濃度3×105細胞/mlで用い、化合物6aおよび11(IC50濃度)を含有したRPMI−1640培地に播種した。アポトーシス誘導のコントロールとして、0.5μM TBTCで処理した細胞を用いた。ネガティブコントロールとしては未処理細胞を用いた。1時間のインキュベーション後、細胞を回収し、遠心分離し、0.14M NaClおよび2.5mM CaCl2を含有する10mM HEPES/NaOHバッファーに1×106細胞/mLとなるように再懸濁した。
このアッセイはAusbacher et al.(2012)Biochem−Biophys. Acta, 1818(11) p. 291725を基にして行なった。簡潔にいうと、A20細胞(3×105細胞/ml)を12ウェルのプレートに播種し、1〜4時間、決定されたIC50値の6aおよび11でインキュベートした。ポジティブコントロールとしてTBTC(0.1μm)で処理した細胞を用い、ネガティブコントロールとして未処理細胞を用いた。各実験終了20分前に最終濃度が100nMとなるようにTMREを添加した。次に、細胞を回収し、最終濃度が1×106細胞/mlとなるようにHBSSで再懸濁して、FACSCaliburフローサイトメーターのFL−2チャネルで解析した。
カスパーゼ2、−3/7、−6、−8、−9、および−10の関与について調べるために、Ausbacher et al.(前述)に従ってカスパーゼFluorometric Substrate Set II Plus kit(Biovision Research Products, Mountain View, CA, USA)を用いて活性スクリーニングを行った。簡潔にいうと、A20細胞(3×105細胞/ml)を1×IC50の6a、1×IC50の11または0.5μMのTBTCで1時間処理した。次に細胞を溶解し、ライセートをDL−ジチオトレイトール溶液と共に反応バッファーに添加した。7−アミノ−4−トリフルオロメチルクマリンで標識したカスパーゼの基質を添加し、37℃で2時間インキュベートし、蛍光プレートリーダー(SpectraMAX Gemini EM、Molecular devices、Sunnyvale, CA, USA)で解析した。カスパーゼ活性は処理したサンプルおよび未処理のコントロールサンプルの蛍光強度を比較することにより決定した。
Ramos細胞(2×105細胞/ml)を、ペプチドを含んだ血清無添加RPMI−1640培地で懸濁し、培養フラスコ(NUNC Easy flask 25cm2、Thermo Fischer Scientific社, Langenselbold, Germany)に移した。既に決定したIC50値によりペプチド濃度を選択した。全ての細胞をカルノフスキー〔Karnovsky〕固定液(ホルムアルデヒド−グルタルアルデヒドのカコジルサン緩衝溶液(pH7.2))により、4℃、オーバーナイトで前固定した。固定液をカルノフスキー緩衝溶液に置き換え、1%の四酸化オスミウムで後固定を行った。濃度を段階的に変えたエタノールで脱水したあと、サンプルをアセトニトリルおよびepon resin(AGAR 100、DDSA、MNAおよびDMP−30)を1:1で混合したものにオーバーナイトで浸透させた。次の日純粋な樹脂を加え、24時間重合させた。超薄切片を準備してホルムバール、炭素を固定した銅製のグリッド上に置いた。サンプルを酢酸ウラニル(5%)およびレイノルズクエン酸鉛で染色して対比した。JEOL−1010透過型電子顕微鏡(JEOL,Akaishima,Japan)でサンプルを解析した。画像の取得にはオリンパスMorada TEM サイドマウントCCDカメラ(Olympus soft imaging solutions GmbH, Muenster, Germany)を用いた。
SEMの研究ではTEMを用いた実験と同じ手法でインキュベーションを行った。全ての細胞をMcDowell固定液で4℃、オーバーナイトで固定した。後固定に1%四酸化オスミニウムを用い、濃度を段階的に変えたエタノールで脱水を行った。化学的脱水にはヘキサメチルジシラザンを使用した。切片をアルミニウムのスタブに載せ、試験の前に、90秒間スパッタリングにより被覆した。サンプルをJEOL社JSM−6300走査電子顕微鏡(JEOL,Akaishima,Japan)で解析し、画像の取得をEDAX Phoenix EDAM III data acquisition module(EDAX Inc.,Mahwah,NJ,USA)により行った。
短いペプチドに対する最初の工程として、1b中の、抗微生物ペプチド(AMP)(14アミノ酸)のモデルを、トリプトファンおよびアルギニン残基(1〜6個のアミノ酸)を交互に含む一連の短いAMPで置き換えた(図1および下記表1参照)。
最初のスクリーニング工程で、ペプチドアナログを正常ヒト繊維芽細胞で試験し(SI−1)、その結果を腫瘍周辺の健康な細胞に対する細胞毒性を評価するために用いた。
6aおよび11が細胞毒性作用を及ぼすメカニズムについて検討した。十分に確立されたフローサイトメトリーのアッセイを用いることで、ミトコンドリア膜電位の変化(ΔΨm)を、細胞膜の組成および完全性とともに調べた。
材料および方法
ペプチド
試験に使う2つのペプチド6a(WRWRWGGRRRRRRR−アミド、配列番号:24)およびペプチド11(WRRRRRWRRRRW−アミド、配列番号:25)は標準的なFmoc法プロトコールに従って独自に合成した。全てのペプチドは分取HPLCを用いて精製し(>95%)、凍結乾燥させて、トリフルオロ酢酸塩として用いた。
自然に発生するBALB/cマウス由来のマウスB細胞リンパ腫であるA20細胞を、1.5g/L 炭酸水素ナトリウム、4.5g/L グルコース、10mM HEPES、1.0mM ピルビン酸ナトリウムおよび50μM 2−メルカプトエタノールを含むRPMI−1640培地中で増殖させた。細胞培養は5%のCO2、37℃の加湿条件に保った。
6〜8週齢の雌BALB/cマウスはCharles River, Germanyから購入した。全てのマウスは地方および欧州倫理委員会のガイドラインに沿って、特定の病原体が存在しないゲージで飼育した。
A20腫瘍細胞を採取し、RPMI−1640内で洗浄し、BALB/cマウスの腹部左側に接種(皮内)した(1マウスあたり5×106細胞/50μl RPMI−1640)。腫瘍の大きさが20mm2に達したら、マウスを3つのグループに分けて処理した;グループ1−溶媒(0.9%NaCl/滅菌H2O)で処理した9匹のマウス、グループ2a−腫瘍をペプチド6aで処理した8匹のマウス、グループ3a−腫瘍をペプチド11で処理した9匹のマウス。病巣内注射は1日1回、3日連続して行った。
6aおよび11の処理後にA20リンパ腫が完全に退縮した動物に、再びA20リンパ腫細胞を移植した。動物が回復してからて4週間後に、腹部右側(1回目の腫瘍移植部位の反対側)に細胞(5×106)を接種(皮内)した。
生存曲線はログランク検定により比較した。各群は一元配置ANOVA検定およびTukeyの多重比較検定を用いて比較した。P<0.05で有意差があるとした。
1匹を除いてA20細胞を接種された全ての動物に腫瘍が発生し、6〜7日以内に腫瘍はペプチド処理を開始するのに十分に大きく(約20mm2)なった。
試験を行ったいずれのペプチドも、全身的な毒性を引き起こすことなしに、処理したマウスの大部分で完全な腫瘍の退縮を示し、一次的な抗腫瘍効果を発揮した。しかしながら,再移植の研究では明らかな違いが見られた。いずれのペプチドも二次的な効果を誘導したが、真の防御免疫を引き起こしたのはペプチド11のみであった。
実施例1で記載したように、WRWRW−GG−KKKKKKK(配列番号:26)の配列であるペプチドをA20細胞で試験した。IC50値は13.2μMであり、活性はあるが、そのポリアルギニンの等価物であるペプチド6aのように効果的なものではない。
材料および方法
テストシステム:雄のSprague−Dawley(Charles River, Portage, MI)ラット(320〜400g)にはCorDynamics社により、DSI社(St. Paul, MN)のテレメートリートランスミッターが取り付けられた。
EWL−6は約−20℃に設定された冷凍庫内において、遮光下で保存された。EWL−6が含まれる製剤を1mL/kg/回の投与量となるように静脈注射用製剤用の0.9%塩化ナトリウム注射溶液(U.S.P)中で調製した。注射前に投与量を0.2μMのポリプロピレンフィルターを通して濾過した。
血行動態評価:脈圧(図8)および平均動脈圧(図9)は初日の投与から−1〜8時間にわたってテレメートリーにより記録し、初日の投与から−1〜8時間の1分平均で報告された。
EWL−6の投与後、コントロールと比較して、脈圧(図8)または平均動脈圧(図9)において顕著な差は見られず、これは本発明のペプチドには循環器に対する毒性がないことを示している。
Claims (21)
- (i)10〜16個のアミノ酸からなり;
(ii)
(a)グアニジニウム基を含む側鎖、または
(b)アミノ基を含む側鎖
のいずれかをもつ、少なくとも8個のカチオン性アミノ酸を有し、該カチオン性アミノ酸の7個以下が連続しており、
(iii)連続していない2個のトリプトファン残基、または全てが連続してはいない3個のトリプトファン残基を有するペプチドであって、必要に応じて塩、エステルもしくはアミドの形であるペプチド;
または、上記ペプチドのペプチド疑似体。 - 前記カチオン性アミノ酸が、アルギニン、ホモアルギニン、リシン、アミノカプロン酸、およびオルニチンからなる一覧から選択された、請求項1のペプチドまたはペプチド疑似体。
- 前記カチオン性アミノ酸が、グアニジニウム基を含む側鎖を有する、請求項1に記載のペプチドまたはペプチド疑似体。
- カチオン性アミノ酸またはトリプトファンのいずれでもない1〜6個の他のアミノ酸をさらに含む、請求項1〜3のいずれか1項のペプチドまたはペプチド疑似体。
- 前記他のアミノ酸が、トリプトファン残基および請求項1〜3のいずれか1項で定義されているカチオン性アミノ酸の双方に隣接している、請求項4のペプチドまたはペプチド疑似体。
- 前記他のアミノ酸が、水素原子が4個以下の側鎖を有し、非電荷であり、かつ非極性である、請求項4または5のペプチドまたはペプチド疑似体。
- 前記他のアミノ酸がグリシンおよび6−アミノヘキサン酸から選択された、請求項4〜6のいずれか1項のペプチドまたはペプチド疑似体。
- 3個のトリプトファン残基を有する、請求項1〜7のいずれか1項のペプチドまたはペプチド疑似体。
- 請求項1〜3のいずれか1項で定義されたカチオン性アミノ酸を9または10個有する、請求項1〜8のいずれか1項のペプチドまたはペプチド疑似体。
- N末端および/またはC末端にトリプトファン残基を有する、請求項1〜9のいずれか1項のペプチドまたはペプチド疑似体。
- 請求項1〜3のいずれか1項で定義されたカチオン性アミノ酸を少なくとも5個連続して有する、請求項1〜10のいずれか1項のペプチドまたはペプチド疑似体。
- 以下の配列の1種を有する、請求項1〜7のいずれか1項のペプチドまたはペプチド疑似体:
WXRRRRRXWXRRRRXW;
WXRRRRWRRRRXW;
RRRRXWXRRRRW;
KKKKXWXKKKKW;
KRKRXWXKRKRW;または
WRWRWXXRRRRRRR。
[配列式中、Wはトリプトファンをあらわし、Rはアルギニンをあらわし、Xはカチオン性アミノ酸またはトリプトファン残基ではないアミノ酸をあらわす]。 - 以下の配列の1種を有する、請求項1のペプチドまたはペプチド疑似体:
WRWRWGGRRRRRRR、WRRRRRWRRRRW、またはWGRRRRRGWGRRRRGW。 - 請求項1〜13のいずれか1項で定義されたペプチドまたはペプチド疑似体と、薬学的に許容可能な希釈剤、担体、または賦形剤とを含む医薬組成物。
- 治療に使用するための、請求項1〜13のいずれか1項で定義されたペプチドもしくはペプチド疑似体、または請求項14で定義された医薬組成物。
- 腫瘍またはがん細胞の治療に使用するための、請求項1〜13のいずれか1項で定義されたペプチドもしくはペプチド疑似体、または請求項14で定義された医薬組成物。
- 前記腫瘍またはがん細胞がリンパ腫である、請求項16に記載された使用のためのペプチド、ペプチド疑似体、または医薬組成物。
- 前記リンパ腫がカスパーゼに依存しないやり方で治療される、請求項17に記載された使用のためのペプチド、ペプチド疑似体、または医薬組成物。
- 腫瘍またはがん細胞の治療方法であって、請求項1〜13のいずれか1項で定義されたペプチドもしくはペプチド疑似体、または請求項14で定義された医薬組成物を、必要とする患者に薬学的に有効な量投与することを含む方法。
- 腫瘍またはがん細胞の治療のための薬剤の製造における、請求項1〜13のいずれか1項で定義されたペプチドもしくはペプチド疑似体、または請求項14で定義された医薬組成物の、使用。
- がん細胞を溶解するための、請求項1〜13のいずれか1項で定義されたペプチドまたはペプチド疑似体のin vitroにおける使用。
