Fmoc化学を利用したペプチド合成装置9050(Applied Biosystems社)を用いて、従来の固相合成により、配列番号16のペプチドおよびその種々の誘導体を合成した。ペプチドをRPクロマトグラフィーで精製し、分析的RP−HPLCおよびMALDI−MSによりその同一性(identity)および純度を確認した。
共受容体の指向性が異なるHIV−1、HIV−2、およびSIVの分子クローンは、リン酸カルシウム法(CalPhos(商標) Mammalian Transfection Kit、Clontech社)によってプロウイルスDNAを293T細胞に一過的にトランスフェクトすることにより作製した。一晩インキュベートした後、10%FCSを補充したDMEM培地でトランスフェクション混合物を置換し、トランスフェクションの48時間後にウイルスストックを回収した。その後、培養上清を3000rpmで5分間遠心して細胞片を取り除いた。得られたウイルスストックをp24(HIV)抗原またはp27(SIV)抗原のELISAにより定量化した。ウイルスストックは、すぐに用いるか、分注して−80℃で保存した。
HIV−1プロモーターの制御下にLacZレポーター遺伝子を含むTZM−blレポーター細胞を、96ウェルF底マイクロタイタープレート(Greiner Bio−One社)に播種した。翌日、細胞を、配列番号16のペプチドまたはその誘導体の種々の希釈物と1時間プレインキュベートし、その後、HIV−1、HIV−2、およびSIVを感染させた。1ステップTropix Gal−Screen Kitを製造業者の推奨に従って用いて、2日後に感染率を決定した。
Ficoll密度遠心を用いて、DRK−Blutspendedienst Baden−Wurttemberg−Hessenに由来するバフィーコートから末梢血液単核細胞を単離した。1ml当たり1×106個のPBMCを、1μg/mlフィトヘマグルチニン(PHA、Oxoid社、#3085280)および10ng/mlインターロイキン2(IL−2、Strathmann社、#9511192)を用いて3日間刺激した。その後、細胞をペレット化し、IL−2含有培地に再懸濁した。2×105個のPBMCを96ウェルF底マイクロタイタープレートに播種し、ペプチドを加え、10〜100pgのX4指向性ウイルスまたはR5指向性ウイルスのp24抗原を細胞に感染させた。子孫ウイルスを含む上清を感染後2〜3日毎に採取した。p24抗原のELISA(NIH AIDS試薬プログラム)によりウイルス産生を測定した。平均p24抗原値(ng/ml)は、3連(triplicate)の感染±標準偏差から得た。
細胞毒性効果を評価するために、TZM−bl細胞または予備刺激したPBMCを、ペプチド濃度を増加させてインキュベートした。CellTiter−Glo Luminescent Cell Viability Assay(PROMEGA社、G7571)を製造業者の推奨に従って用いて、細胞生存率を決定した。値は3連の測定から得た。細胞を含むPBS(ペプチドなし)中のATPレベル(100%)と比較して生存率を計算した。試薬添加の10分後にルミノメーターを用いてデータを記録した。
リガンド−受容体相互作用のリアルタイム蛍光モニタリング
抗ヒトCXCR4(クローン12G5、IgG2a)または抗ヒトCXCR7(クローン9C4)モノクローナル抗体(mAb)をR&D Systems社(ミネソタ州、ミネアポリス)から購入した。PE標識ヤギ抗マウスF(ab’)2 Ab(Dako社、デンマーク、グロストルプ)を用いて非結合の抗CXCR7 mAbおよび抗CXCR4 mAbの結合を明らかにし、CellQuestソフトウェアを用いてFACSCaliburフローサイトメーター(BD Biosciences社)で分析した。ヒトケモカインCXCL12およびCXCL12−TexasRedを、説明されているように(Valenzuela-Fernandez, et al. 2001)合成した。ヒトケモカインCXCL11はClinisciences SAS社(フランス)から購入した。
ヒトCXCR7 cDNAをEGFP−cDNAと融合させて改変pIRES Hyg 3ベクター(Clontech社)にクローニングした。リン酸カルシウム−DNA共沈法によりEGFP−CXCR7を安定的に発現するHEK293T細胞を作製し、100nMのCXCL12の非存在下または存在下で9C4 mAbの結合について評価した。参照文献(Hachet-Haas et al., 2008)に記載されているように、モノクローナルマウス抗GFP(Roche Molecular Biochemicals社;1/100希釈)を1次抗体として、R−PE標識AffiniPure F(ab’)2断片ヤギ抗マウスIgG(Immunotech社;1/100)を2次抗体として用いたEGFPの細胞表面標識を用いて、受容体の内部移行を記録した。サイトメーター(FACScalibur、Becton−Dickinson社)を用い、フローサイトメトリー分析(1サンプルあたり10,000個の細胞)によりCXCR4またはCXCR7の染色を定量化した。CELLQuest(Becton−Dickinson社)ソフトウェアを用いて、CXCR4またはCXCR7の蛍光強度の平均を計算した。
市販の抗ヒトCXCR4 mAb(BD Pahrmingen社、クローン:12G5;または1D9)およびCCR5 mAb(BD Pahrmingen社、クローン:CD195)を用い、フローサイトメトリー分析(FACSCalibur;Becton−Dickinson社)により、CXCR4上の配列番号16のペプチドの結合部位を評価した。2×105個のJurkat T細胞を、無血清培地中でペプチドと共に4℃で30分間インキュベートした。インキュベーション後、細胞をFACSバッファー(PBS+1%FCS)で遠心により洗浄して、続いて、PE標識した抗ヒトCXCR4 mAbまたはCCR5 mAbのいずれかで4℃にて染色した。洗浄後、FACSバッファー中の4%パラホルムアルデヒドで、室温で5分間、細胞を固定し、その後、フローサイトメーターで分析した。CELLQUEST(Becton−Dickinson社)を用いてデータを処理した。配列番号16のペプチドの受容体優先性を分析するために、Cell Dissociation Solution(非酵素的、1×;Sigma社:C5914)を用いてGhost細胞(親、X4、Hi)を回収し、上述のようにFACS分析のために調製した。
組換えバキュロウイルスの産生:ヒトCXCR4、ラットGタンパク質αi2サブユニット、ならびにヒトGタンパク質β1サブユニットおよびウシGタンパク質γ3サブユニットをコードするバキュロウイルスの産生が記載されている(Moepps et al., 1997)。[35S]GTP[S]はPerkin−Elmer社(米国、ウォルサム)から入手した。CXCL12はPeproTech社(米国、ロッキーヒル)から入手した。

5μm孔フィルターを備えた直径6.5mmのチャンバー(Transwell、24ウェル細胞培養、Coster社、マサチューセッツ州、ボストン)を用いてJurkat細胞(ATCC)の遊走を分析した。2×105個のJurkat細胞を200μlのOptimizer T Cell Expansion SFMに懸濁し、この細胞懸濁液を上側のチャンバーに加えた。次に、600μlのT Cell Expansion SFM中の10nM CXCL12(R&D System社)および/または種々の濃度の配列番号16のペプチドもしくはその誘導体を、下側のチャンバーに加えた。細胞培養チャンバーを、細胞培養インキュベーター中で37℃にて150分間インキュベートした。インキュベーション後、チャンバーを取り出し、100μlの上清を採取し、下側の区画に遊走した細胞を、血球計を用いて直接計数するか、または、上述のようにCellTiter−Glo Luminescent Cell Viability Assayを用いて分析した。全ての値は、3連の実験±SDから得た、CXCL12のみで処理した細胞と比べた、遊走細胞の平均数を表す。
Biocoat Matrigel細胞浸潤チャンバー(BD BioCoat(商標) Matrigel(商標) Invasion Chamber)を製造業者の推奨に従って用いて、癌細胞の細胞浸潤をアッセイした。5×104個のDU145細胞(ATCC)を、0.1%BSA(KPL社)を含む300μlの無血清RPMI(Gibco社)に懸濁した後、上側チャンバーに加えた。100nMのCXCL12、および種々の濃度の配列番号16のペプチドを含むか、または含まない、700μlの無血清培地を下側チャンバーの各々に加えた。チャンバーを5%CO2/95%空気の湿潤雰囲気中、37℃で24時間インキュベートした。こすり洗うことにより、膜の上側表面から非浸潤細胞を除去した。上述のようにCellTiter−Glo(登録商標) Luminescent Cell Viability Assayキットを用いて、膜の底の方に浸潤した細胞を定量化した。
培地、ALB誘導体、またはAMD3100(全て545μM)のいずれかとプレインキュベートしたJurkat細胞を、37℃でCXCL12(100μg/ml)を用いて記載されている時間刺激し、5%ホルムアルデヒド(Carl Roth社;ドイツ、カールスルーエ)で固定し、0.1%サポニン(Carl Roth社)で透過処理した。F−アクチンをAlexaFluor568標識ファロイジン(Molecular Probes社;オレゴン州、ユージーン)で染色した後、相対染色強度のフローサイトメトリー分析を行った。
C57Bl/6Jマウス(Janvier社;フランス、ル ジュネスト サン ティスル)を、餌および水を不断給餌して、フランクフルトのゲーテ大学メディカルセンターの通常飼育の動物施設で飼育した。10mg/mLの配列番号16のペプチドを含む200μLの水または生理食塩水をマウスに腹腔内注射した。注射後、記載の時間に、注意深く皮膚を消毒した後、頬袋から血液を採取した。低張溶解後、サイトカインが豊富な市販の半固体培地(3434、Stem Cell Technologies社;ブリティッシュコロンビア州、バンクーバー)中、標準的な条件下で、2連(duplicate)で白血球をインキュベートした。説明されているように(Bonig et al., 2006)、7日目にCFU−Cをスコア化した。CFU−Cは、インキュベートした血液容積を基準に標準化した。全ての動物実験は、研究機関内の動物実験委員会(IACUC)の許可を得て、実験動物ケア評価認証協会(AAALAC)のガイドラインに従って行われた。
C57BL/8動物に、配列番号16のペプチド(食塩水中2mg、腹腔内)または対照食塩水を注射した。末梢血液を注射の1時間後に個別に回収し、計数、混合し、4×105個のC57BL/6 CD45.1 BM細胞と共にC57BL/6 Cd45.1レシピエントに競合的に移植した(660μlの血液に相当)。
食塩水中の2mgの水酸化アルミニウム(Sigma−Aldrich社、23918−6)に吸着させた50μgのオボアルブミン(OVA、Sigma−Aldrich社、A5503)を、0日目、1日目、および2日目に腹腔内(i.p.)注射することによって、マウスを感作した。麻酔(50mg/kgケタミンおよび3.3mg/kgキシラジン、腹腔内)下で、5日目、6日目、および7日目に、食塩水25μl中の10μgのOVA(12.5μl/鼻孔)、または対照マウスでは食塩水のみの鼻腔内(i.n.)注入により、マウスを曝露した。各OVA曝露の2時間前に、配列番号16のペプチドまたはALB409−423を含む食塩水を腹腔内投与(16μmol/kg)した。以前に報告されているように(Rebber et al., 2012)、最後のOVA曝露の24時間後に、気管支肺胞洗浄(BAL)および細胞分画の測定を行った。
NMRスペクトルを得るために、配列番号16のペプチドの1mM溶液を、10mMのリン酸ナトリウムを含むH2O/D2O=10:1中に調製し、HClで最終pHを7.0に調整した。Bruker社製の分光計を用いて800MHz、600MHz、および500MHzでTOCSYおよびNOESY 1H−NMRスペクトルを記録した。質が良好であったため、800MHzの装置で得られたスペクトルを用いた。スペクトルは、外部TSPを基準とし、ウォーターサプレッション(water suppression)のためのwatergate 3−9−19パルスシーケンスを組み込んだStates−TPPI法(Jeener et al., 1979)を用いて記録した。一般に、256個の均等間隔の展開時間t1の値を取得し、平均して2048ポイントの16の過渡現象(transient)であった。時間領域データマトリックスは全て、両次元において4Kまでゼロフィルして、3.41Hz/ptのデジタル分解能が得られた。全ての実験で、フーリエ変換の前に、種々のパラメーターのLorentz−Gaussウィンドウをt1次元およびt2次元の両方に適用した。混合時間(0.30s)でNOESYスペクトル(Griesinger et al., 1988)を取得し、0.060sのDIPSI2混合パルス(Bartels et al., 1995)を用いてTOCSY実験(Braunschweiler et al., 1983; Rucker et al., 1989)を記録した。NOESY実験およびTOCSY実験はいずれも、298Kで行った。
NMRスペクトル中の1H化学シフトの分散により、TOCSYおよびNOESY分光測定を組み合わせて、標準的な方法を用いて、全てのNH−αおよびCH−αの共鳴、ならびに大多数の側鎖プロトン(97.7%)を簡単且つ明瞭に帰属することができた。CHi−NHi+1およびNHi−NHi+1のNOEを追跡することにより、連鎖的な骨格結合性が確認された。
ヒト血清に1mMの配列番号16のペプチドまたは改良された誘導体を添加して37℃でインキュベートした。サンプルを2時間毎に採取し、直ちに−20℃で保存した。インキュベートしたペプチドの血清中における抗ウイルス活性を評価するために、10μlのサンプルを5×104個/100μlのTZM−bl細胞に加えた。続いて、細胞に90μlのHIV−1 NL4−3を感染させ、ペプチドおよび血清の混合物の20倍希釈物を得た。1ステップTropix Gal−Screen Kitを用いて、感染の2日後に感染力を測定した。プロテアーゼ阻害剤が配列番号16のペプチドの分解に与える影響を評価するために、1MMの配列番号16のペプチドを加える前に、血清に最初にプロテアーゼ阻害剤カクテル(1×Complete mini(Roche社)および1mM PMSF(Roche社))を添加した。
別記(参照文献)されているように、配列番号16のペプチドであるペプチドを用いたニワトリの免疫処置により配列番号16のペプチドに対するポリクローナル抗血清を作製し(Davids Biotechnologie社、レーゲンスブルク)、マウスの免疫処置によりモノクローナル抗体を作製した(ViroPharmaceuticals社、ハノーバー)。ポリクローナル抗体およびモノクローナル抗体の反応性および特異性を特徴付けるために、100μlの段階希釈された配列番号16のペプチド、その誘導体(20μM)、またはHSA(Sigma社)でELISAプレート(Corning costar)を4℃で一晩コーティングした。翌日、200μlのELISA洗浄バッファー(KPL社)でプレートを2回洗浄し、150μlのPBS中の1%ウシ血清アルブミン(BSA)で処理して非コーティング表面をブロックした。さらに洗浄した後、100μlの段階希釈されたモノポリクローナル抗体またはポリクローナル抗体を加えて1時間インキュベートした。その後、プレートを洗浄し、100μlのホースラディッシュペルオキシダーゼ(HRP)標識二次抗体(抗ニワトリまたは抗マウス)をさらに1時間加えた。その後、プレートを5回洗浄し、100μlのSureBlue TMB 1−Component Microwell Peroxidase Substrate(KPL社)を加えた。100μlの1N HClを各ウェルに加えて発色を停止させ、マイクロタイタープレートリーダー(Molecular Devices社、VMax Kinetic Microplate Reader)を用いて、650nmを基準として、450nmで光学密度を記録した。
