2007, Peptide Science 88: 279‐289、にてレビューされている)。細胞反応を誘発するCXCL12等の受容体アゴニストとは対照的に、受容体アンタゴニストは、その受容体と結合した際に、生物反応、すなわち、細胞遊走またはCa2+シグナル伝達、を誘発しないリガンドまたは薬剤である。受容体アンタゴニストは、HIV‐1感染の阻害(CCR5アンタゴニスト)またはアゴニストが媒介する細胞反応の低減が可能である有用な薬剤として既に臨床的に使用されている(例:アンジオテンシンアンタゴニスト、β‐アドレナリンアンタゴニスト、セロトニンアンタゴニスト、またはCCR5アンタゴニスト)。受容体アンタゴニストと受容体との相互作用により、アゴニストの機能が阻害される。ほとんどの薬剤アンタゴニストは、受容体上の構造的に決定される結合部位において内在性のリガンドまたは基質と競合することにより、その効力を発揮する。
Peptide Science 88: 279‐289、にてレビューされている)。さらに、関節リウマチが、CXCR4を発現するCD4+メモリーT細胞の炎症滑膜への蓄積によって引き起こされることが示されている。関節リウマチ患者では滑膜中のCXCL12濃度が非常に上昇しており、これがメモリーT細胞を誘引する。CXCR4拮抗性分子は、メモリーT細胞の滑膜への遊走を阻害する(Tsutsumi et al., 2007, Peptide Science 88: 279‐289、にてレビューされている)。
Z1‐LVRYTKKVPQVSTPTL‐Z2(ALB‐408)
を有するペプチド、ならびに、その生物学的活性誘導体、特にアミド化、アセチル化、硫酸化、リン酸化、および/もしくはグリコシル化誘導体、ならびに、多重合成によって得ることができるALB408‐423の生物活性を有するペプチド、に関し;
ここで、Zは、0〜10の数のアミノ酸残基を表す。
ヒト血液ろ過液からの抗ウイルス効果を有するALB408‐423の単離
ヒト血液ろ過液は、所望により水による希釈および酸性化を行ってもよい。pH値は、1.5〜3.5が好ましく、2.5〜3.0が特に好ましい。その後、この血液ろ過液を、例えばスルホン酸基で修飾された支持体材料(Fraktogel SP‐650(M), Merck,ダルムシュタット,ドイツ)などのカチオン交換樹脂に通す。カチオン交換樹脂に結合したペプチドは、比較的高濃度の塩溶液で溶離させる。溶出液のイオン強度は、0.5M〜1Mの酢酸アンモニウム溶液のそれとおよそ同じである。
800〜1000リットルの血液ろ過液を、HClにより2.7のpH値に調節し、水で希釈して導電率を5.5mS/cmとし、3L/分の流速で強カチオン交換体にチャージする。

B〜E. 単離の第四〜第七工程。阻害活性を、純粋物質が得られるまで精製した。
F. 精製されたALB408‐423の質量スペクトル(MALDI‐MS)および配列分析。
et al., J. Mol. Biol. 241: 651, 1994)を、100μlのDMEM(10%FCS、100U/mlペニシリンG、および100μg/ml硫酸ストレプトマイシン)中へ播種することによって実施した。P4‐R5細胞は、LTR‐lacZカセットによって安定的にトランスフェクトされ、HIV‐1による感染に成功すると、Tatに依存的にβ‐ガラクトシダーゼを発現し、これは、化学発光試験にて検出することができる。翌日、画分のアリコートを添加した。そのために、凍結乾燥された画分を80μlのDMEMへ再懸濁させ、その各々の25μlをP4‐R5細胞へピペットで分注し、37℃にて1時間インキュベートし、続いてHIV‐1 NL4_3(1ngのp24抗原)で感染させた。ウイルスストックは、リン酸カルシウム法(CalPhos(商標) Mammalian Transfection Kit,Clontech)によるプロウイルスDNAを用いた293T細胞の一過性の感染によって得た。ウイルスストックはトランスフェクションの48時間後に回収してろ過し、感染に用いた。感染後3日目に、感染したP4‐R5細胞のβ‐ガラクトシダーゼ活性を、製造元の推奨に従ってGalScreenアッセイ(Tropix)を用ることで検出した。簡単に述べると、上清を除去し、PBS/GalScreen+基質の1:1希釈物40μlを添加し、続いて室温にて30分間インキュベートした。次に、ライセート30μlを96ウェルのルミプレート(lumiplate)へ移した。続いて、発光を、ルミノメーター(Berthold,Orion)により、秒あたりの相対発光量(relative light units)として検出した。全測定値から、非感染コントロール細胞の平均β‐ガラクトシダーゼバックグラウンド活性を減じた。各感染についての感染%の値を、ペプチド非含有のコントロール(100%)に対して算出した。ALB408‐423の非存在下での測定における酵素活性を100%に設定し、その他のすべての値はこれに基づいたものとした。最終精製(第七工程)は、TZM‐bl細胞(X. Wei et al., Antimicrob. Agents Chemother.
46: 1896, 2002)とまったく同一の条件下にて試験した。
本アッセイにて生物活性であった(図1A)pHプールIIからの画分6〜8の全量200mg(1087リットルの血液ろ過液等量に相当)を、セミ分取用逆相カラムを通して分離した。画分33+34が、本発明の物質を含有していた(図1B)。

質量測定
血液ろ過液から単離されたペプチド(実施例1の第七工程の画分31より)、および化学合成されたペプチド(実施例3)の質量測定を、MALDI質量分析器(Voyager DE‐Pro)で実施した。ペプチドの分子質量は、以下の質量数値(MW)に相当すると測定された。
ALB408‐423、ヒト血液ろ過液から単離(図1F): 1830.9Da
ALB408‐423、化学合成ペプチド: 1830.6Da
精製された天然のペプチドを、PROTEOMEFACTORY AG社,Dorotheenstr.94,10117 ベルリン(ドイツ)、から提供されたMS‐MSカップリング分析(ESI‐TRAP)により、Mascot検索エンジンによる確立されたESI MS‐MS質量のデータベース比較によって分析した結果、次の配列が最も確率の高かった。
LVRYTKKVPQVSTPTL(図1F)
SwissProtデータベースによるさらなるデータベース比較により、このペプチド配列が、ヒトタンパク質血清アルブミン(アクセス番号NP000468)のアミノ酸408‐423と100%の同一性を有することが示され、この配列は、アミノ酸:LVRYTKKVPQVSTPTL、を含有している。
実施例1の第七工程からの画分31の1.6mgを160μlのDMEMへ溶解した。続いて、ALB408‐423を含有する画分31の段階希釈物10μlを60μlのTZM‐bl細胞(60μl)へ添加し、1ngのp24抗原HIV‐1 NL4_3によって感染させ、全量を100μlとした。3日後、GalScreenアッセイにて感染率を測定した(実施例1参照)。画分31は、このX4向性HIV‐1 NL4_3による感染を用量依存的に阻害した。最大値の半分まで感染を阻害する用量(IC50)は、21.45μg/mlであった(図2)。
ALB408‐423の化学合成
ALB408‐423の化学合成は、公知のFmoc化学を用い、ペプチド合成機9050(Applied Biosystems)上での従来の固相合成によって実施した。得られたペプチドは、逆相クロマトグラフィで精製し、実施例2で述べた分析用RP‐HPLCおよびMALDI‐MS質量測定により、その同一性および純度を確定した。
X4‐向性HIV‐1変異体の感染を特異的に阻害する合成ALB408‐423
5000個のTZM‐bl細胞を、100μlのDMEM(10%FCS、100U/mlペニシリンG、および100μg/ml硫酸ストレプトマイシン)中へ播種した。ALB408‐423をPBSへ溶解した(10mg/ml)。1日後、PBSによるALB408‐423の段階希釈物20μlを細胞へ添加し、続いて、0.5ngのp24抗原HIV‐1で細胞を感染させ、全量を200μlとした。共受容体向性の異なる複数のHIV‐1分子クローンを用い、これらは記載に従って作出した(Papkalla et al., J. Virol. 76: 8455‐9, 2002)。
合成ALB408‐423はCXCR4向性レンチウイルスの感染を用量依存的に阻害する
X4向性HIV‐1 NL4‐3およびHIV‐2ROD10、またはCCR5向性HIV‐1 NL4‐3 92th014、HIV‐1‐7312、およびSIVmac239は、293T細胞の一過性のトランスフェクションによって作出し、これらを用いて、示した濃度のALB408‐423を含有するTZM‐bl細胞を感染させた。2日後、感染率を既述のようにして測定し算出した(実施例4)。結果から、ALB408‐423は、X4向性HIV‐1およびHIV‐2の感染を用量依存的に阻害したが(IC50は約10〜20μM)、一方このペプチドは、R5向性レンチウイルスの感染には影響を及ぼさなかったことが示され、これは、CXCR4向性HIV‐1およびHIV‐2の特異的な阻害を実証するものである(図4)。

ALB408‐423誘導体を用いた構造活性相関(SAR)研究
NもしくはC末端欠失、またはアミノ酸置換を含有する種々のALB408‐423誘導体(表1)を化学的に合成し、凍結乾燥したペプチドをPBS中へ溶解した。この抗ウイルス活性を、既述のようにしてX4向性HIV‐1 NL4‐3を用い、TZM‐bl細胞内で分析した(実施例4)。ALB408‐423のN末端欠失(409‐423、410‐423、411‐423、412‐423、413‐423、414‐423、415‐423)によって抗ウイルス活性が大きく減退または無効化され、このことは、ALB408‐423が介するX4向性HIV‐1の阻害に対して、N末端ロイシン(L408)が極めて重要であることを示している(図5および図6)。
ALB誘導体はいずれも細胞毒性を示さない

細胞を、最も強力な抗ウイルス活性を及ぼすペプチド(表1および図9)の増加する種々の濃度と共に、3日間インキュベートした。細胞生存率を、CellTiter‐Glo発光細胞生存率アッセイ(Luminescent Cell Viability Assay)(Promega,#G7571)を製造元の推奨に従って用いて測定した。この発光に基づくアッセイは、細胞内ATPの量に基づいて生存細胞数を測定する。データは、ルミノメーターを用い、試薬の添加後10分で記録した。PBSのみでインキュベートした細胞から得られた発光活性を100%に設定した。図9に示した結果より、試験したいずれのALB誘導体でも、300μM以下の濃度では細胞毒性効果が見られなかったことが明らかに示される。
ALB408‐423の抗ウイルス活性および最も強力な誘導体
種々のHIV‐1クローンに対する最も活性であるALBペプチドの効果を調べるために、293T細胞をプロウイルスプラスミドでトランスフェクトすることによって異なる共受容体を利用するウイルスを作出した(Papkalla et al., J. Virol. 76: 8455‐9, 2002)。ウイルスストックをまずTZM‐bl細胞に滴定した。次に、100μMのペプチドを含有するTZM‐bl細胞を、感染性を正規化した量のX4向性、二重向性(X4/R5)、またはR5向性HIV‐1で感染させた。既述のように(実施例4)感染率を測定し、野生型ALB408‐423、C末端切断型ALB408‐419およびALB L408I‐419変異体、ならびにALB‐T412Aが、分析したすべてのX4向性HIV‐1クローン(NL4‐3、P51‐Sc、P34‐s)の感染をほぼ完全に阻害したことが示された(図10)。これらのペプチドは、R5向性HIV‐1感染に対しては影響を及ぼさず、二重向性HIV‐1クローン92ht593.1によるTZM‐bl細胞の感染は中程度に阻害しただけであった。これらのデータは、抗ウイルス活性が高められたALB変異体(ALB408‐423と比較して)(表1)も、X4向性HIV‐1変異体の広範囲にわたる阻害剤であることを実証している。
ALB408‐423、ALB408‐419、およびALB L408I‐419はX4向性HIV‐1の感染およびPBMC内での複製を阻害する
該当する一次細胞におけるALB408‐423およびその誘導体の効果を分析するために、末梢血液単核細胞を、フィコール密度遠心分離を用いて、ドイツ赤十字献血サービス バーデンビュルテンベルクヘッセン(DRK‐Blutspendedienst Baden‐Wurttemberg‐Hessen)より得たバフィーコートから単離した。1mlあたり1×106個のPBMCを、1μg/mlのフィトヘマグルチニン(
PHA、Oxoid、#3085280)および10ng/mlのインターロイキン2(IL‐2、Strathmann、#9511192)にて3日間刺激した。その後、細胞をペレット化し、IL‐2を含有する培地へ再懸濁させた。1.5×105個のPBM
C(250μl)を96ウェルディッシュへ播種し、ペプチドを添加し、細胞をX4向性HIV‐1 NL4‐3のp24抗原50pg/mlで感染させた。子孫ウイルスを含有する上清を感染後の第1、3、および6日に採取した。ウイルス産生をp24抗原ELISA(SAIC‐Frederick,Inc[AIDS & Cancer virus program])で測定した。100μMのALB408‐423およびALB L408I‐419を含む細胞から第6日に得られた上清では、p24抗原は検出することができず、100μMのALB408‐419を含有する上清では、ほんの僅かのp24レベルしか検出することができなかった(図11)。20μMのペプチドの存在下では、ウイルスの複製が大きく減退した。これらのデータは、試験したALB408‐423およびその2種類の誘導体が、天然のHIV標的細胞内にて、X4向性HIV‐1の感染および複製を阻害することを示している。
ALB408‐423ペプチドはCXCL12のCXCR4への結合を阻害する
ケモカインCXCL12のその受容体CXCR4への結合を阻害するALB408‐423の能力を試験するために、過去の記載(Valenzuela‐Fernandez, et al.; 2001, JBC 276:26550‐26558)に従って蛍光結合アッセイを生存細胞全体に対して実施した。CXCR4受容体は、EGFP蛍光タンパク質が受容体の細胞外アミノ末端部へ融合した融合タンパク質(EGFP‐CXCR4)として、ヒト胎児由来腎臓(HEK)細胞へ安定にトランスフェクトされる。ヒトケモカインCXCL12およびCXCL12‐テキサスレッドを記載に従って合成した(Amara et al., 1999, JBC 274:23916‐23925;
Valenzuela‐Fernandez, et al., 2001, JBC
276:26550‐26558)。リガンド‐受容体相互作用のリアルタイム蛍光モニタリングを以下のようにして実施した。融合受容体EGFP‐hCXCR4を発現するHEK293細胞を、5mMのEDTAを補ったpH7.4のリン酸緩衝生理食塩水中で採取し、遠心分離に掛け、プロテアーゼ阻害剤(40μg/mL ベスタチンおよびバシトラシン、20μg/mL ホスホラミドン、50μg/mL キモスタチン、および1μg/mL ロイペプチン)を補ったHEPES‐ウシ血清アルブミンバッファー(10mM HEPES、137.5mM NaCl、1.25mM MgCl2、1.25m
M CaCl2、6mM KCl、10mM グルコース、0.4mM NaH2PO4、
1% ウシ血清アルブミン(w/v)、pH7.4)中へ再懸濁させた。実験は、HEPES−BSAバッファー中に懸濁させた細胞(通常、106細胞/mL)に対して実施し
た。510nmで発光(470nmで励起)される蛍光の時系列での記録を、分光蛍光光度計(fluorolog 2,Spex)を用いて21℃にて行い、0.3秒ごとにサンプリングした。30秒の時点て100nMのCXCL12‐TRを1mLの細胞懸濁液へ添加することによって蛍光結合測定を開始した。競合実験では、EGFP‐CXCR4を発現する細胞を、種々の濃度の競合物質の非存在下または存在下にて、予め10分間インキュベートした。次に、CXCL12‐TR(100nM)を添加し、平衡に達するまで(300秒)蛍光を記録した。データの解析は、Kaleidagraph 3.08
ソフトウェア(Synergy Software,レディング,ペンシルベニア州,米国)を用いて行った。CXCL12のテキサスレッド(TR)基へのエネルギー移動に起因するEGFP蛍光発光の低下として、蛍光CXCL12との関連性が検出される。
ペプチドALB408‐423はCXCR4、CCR5、およびCXCR4を介するCa2+動員を誘発せず、CXCL12が誘発するカルシウム細胞応答を阻害する
CXCR4、CCR5、またはCXCR1が介する細胞応答を制御するALB408‐423の能力を、カルシウム指示薬をロードしたHEK293細胞上で調べた。indo‐1アセトキシメチルエステルをカルシウムプローブとして用い、記載に従って(Palanche et al., 2001, JBC 276:34853‐34861;
Vollmer et al., 1999, JBC 274:37915‐37922)細胞内Ca2+放出測定を実施した。細胞応答は、分光蛍光光度計を用いて励起を355nmに、発光を405nmおよび475nmに設定し、1mLキュベット内で攪拌しながら37℃にて記録した。ヒトケモカインCCL5およびCXCL8は、Becton
Dickinson Biosciences(サンホゼ,カリフォルニア州)より購入した。CXCR4、CCR5、またはCXCR1を発現する細胞を用いたCa2+動員アッセイにより、ケモカインアゴニスト、CXCL12(CXCR4)、CCL5(CCR5)、およびCXCL8(CXCR1)それぞれがCa2+動員を誘発し(図13)、一方、ALB408‐423単独ではいずれのカルシウム応答も誘発せず、従って、CXCR4、CCR5、およびCXCR1アゴニスト特性を示さないことが実証される(図13)。
ALB408‐423はCXCL12が誘発するCXCR4の内在化を阻害する
適切なケモカインで刺激すると、数多くのGタンパク質共役受容体が、クラスリン被覆ピットにより内在化する。CXCR4反応のアンタゴニストとして、ALB408‐423は、ケモカインが誘発するCXCR4受容体の内在化を変化させることもできる。CXCL12が媒介するCXCR4の内在化に対するALB408‐423のアンタゴニスト特性を分析するために、EGFP‐CXCR4受容体を発現する細胞を分割し、ラットI型コラーゲンをコーティングした12mmカバーガラス上で24ウェルプレートにて2日間増殖させた。次に、細胞を0〜30分の範囲の時間、100nM CXCL12、または50μM ALB408‐423、または100nM CXCL12プラス50μM ALB408‐423、を含有する、プロテアーゼ阻害剤を補ったHEPES‐BSAバッファー中にて37℃でインキュベートした。細胞を氷上へ配置することで内在化を停止させ、氷冷HEPES‐BSAバッファーで直ちに洗浄した。次に、細胞をPBS中の4%パラホルムアルデヒド中にて4℃で15分間固定し、続いて50mMのNH4Cl中に
て15分間インキュベートした。カバーガラスを抗退色剤(anti‐fading agent)である
(Calbiochem)を用いて顕微鏡のスライドグラス上へ載せ、24時間室温で維持した後、−20℃で保存した。次に、細胞を倒立顕微鏡(Leica)およびHCX PL APO Ibd.BL 63× 1.40 OIL UV 対物レンズ(n°506192)を用いたレーザー走査型共焦点イメージングシステム(Leica AOBS
SP2 MP)で分析した。電子ズームは3に設定し、ピンホールは1エアリー(Airy)であり、得られたピクセルサイズは0.154μmであった。EGFPはアルゴンレーザーの488nmレーザー線で励起し、495〜550nmのいわゆるmCFPチャネル内の1つの光電子増倍管(PMT)によって検出および増幅した(PMT1 610高電圧‐HV‐、オフセット0)。良好なシグナル対ノイズ比を得るために、連続して取得した4つのイメージから平均化した。
CXCL‐12が媒介するジャーカットT細胞の遊走を阻害するALB408‐423
CXCL12‐CXCR4シグナル伝達は、HIV/AIDS、癌、白血病、および関節炎等のいくつかの疾患において極めて重要な役割を担っている。CXCL12を発現する器官、組織、または細胞は、CXCR4を発現する腫瘍細胞を引き寄せることができ、転移を起こさせる。ALB408‐423がCXCL12の媒介による腫瘍細胞遊走を阻害することが可能かどうかを調べるために、モデルシステムとしてCXCR4を発現するジャーカットT細胞を用いて遊走アッセイを実施した(Princen et al.,
2004, J. Virol. 78: 12996−13006)。ジャーカットT細胞を0.4×106(200μl)にて10%FBSを含有する媒体へ懸濁させ、次
に、この細胞懸濁液(200μl)を5μmポアフィルターデバイス(Transwell,24‐ウェル細胞培養,Costar)の上部コンパートメントに添加した。次に、CXCL12含有(100ng/ml)または非含有の培養液600μlを下部コンパートメントに添加し、上部コンパートメントから細胞を誘引させた。CXCL12の誘発によるジャーカットT細胞遊走に対する阻害効果を研究するために、下部コンパートメントのCXCL12を種々の濃度のALB408‐423と混合した。この細胞培養プレートを、細胞培養インキュベータ内にて37℃で2時間インキュベートした。インキュベーションの後、プレートを取り出し、下部コンパートメントに遊走した細胞100μlを、計数チャンバーを用いて直接計数するか、または増殖アッセイ(CellTiter‐Glo(登録商標)試薬,Promega)を用いて製造元の推奨に従って分析した。増殖アッセイは、細胞数と直接比例する細胞内ATPレベルを測定する(データ示さず)。図16に示すデータは、ALB408‐423が、CXCL12が媒介するジャーカットT細胞遊走を用量依存的に阻害することを実証している。高濃度(360μg/ml)では、ALB408‐423はCXCL12が誘発する細胞遊走ほぼ完全に阻害し、いずれのペプチドも存在しない状態(CXCL12もALB408‐423もなし)で観察された比率と同等であった。これらのデータは、CXCR4アンタゴニストALB408‐423が、CXCL12によって媒介される腫瘍細胞の誘引を阻害することができることを示している。
CXCR4へのALB408‐423の結合はN末端アミノ酸の完全性に依存する
CXCR4への結合を媒介し、従ってX4向性HIV‐1の感染およびCXCL12の結合を阻害するALB408‐423の領域を同定するために、本発明者らは、いくつかのALB408‐423誘導体(表1参照)の、CXCL12が誘発するCa2+動員およびCXCL12‐Trの結合に対する影響を分析した。実験の詳細については実施例xおよびyを参照されたい。図17のAおよびBに示すように、N末端ロイシンが欠失したALB409‐423は、CXCL12が媒介するCa2+応答もCXCL12‐TrのCXCR4受容体への結合も阻害しなかった。興味深いことに、ALB409‐423は、HIV‐1阻害アッセイにおいても不活性であり、ALB409‐423がCXCR4へ結合できないことが、抗ウイルス活性の喪失の原因でもあることが示唆される。対照的に、C末端切断型のALB誘導体はいずれも、依然としてCXCR4受容体との相互作用が可能であり(図17A)、CXCL12が媒介するCa2+応答を阻害し(図17B)、野生型ペプチドに近い解離定数(30μM)を示すが、ただし、最も短い408‐413は、受容体に対する親和性が下がり(>200μM)、X4向性HIV‐1感染の阻害にもほとんど効果がない(図6および表1)。まとめると、これらのデータは、いくつかのC末端切断型ALB誘導体が、CXCR4と結合することができ、それによってCXCL12の結合およびシグナル伝達、またはX4向性HIV‐1感染を阻止するCXCR4アンタゴニストであることを示している。
Claims (12)
- 配列表の配列番号8〜14、16〜18、20〜22、25〜30および32からなる群より選択されるいずれか1つのアミノ酸配列からなるペプチドを含有する、CXCR4アンタゴニスト剤。
- 前記ペプチドが、配列番号8、12、21および29からなる群より選択されるいずれか1つのアミノ酸配列からなるペプチドである、請求項1に記載のCXCR4アンタゴニスト剤。
- 配列表の配列番号8〜14、16〜18、20〜22、25〜30および32からなる群より選択されるいずれか1つのアミノ酸配列からなるペプチドが、アミド化、アセチル化、硫酸化、リン酸化、および/もしくはグリコシル化されているものである、請求項1に記載の、CXCR4アンタゴニスト剤。
- 配列表の配列番号8〜14、16〜18、20〜22、25〜30および32からなる群より選択されるいずれか1つのアミノ酸配列からなるペプチドが、CXCL12のCXCR4への結合を阻害するものである、請求項1または請求項3に記載の、CXCR4アンタゴニスト剤。
- 配列表の配列番号8〜14、16〜18、20〜22、25〜30および32からなる群より選択されるいずれか1つのアミノ酸配列からなるペプチドが、gp120のCXCR4への結合を阻害するものである、請求項1、請求項3または請求項4のいずれか1項に記載の、CXCR4アンタゴニスト剤。
- 請求項1〜請求項5のいずれか1項に記載のCXCR4アンタゴニスト剤を活性成分として含有する、CXCR4向性ウィルスの感染阻害剤。
- 請求項1〜請求項5のいずれか1項に記載のCXCR4アンタゴニスト剤を活性成分として含有する、抗HIV剤。
- 配列番号12または配列番号21のアミノ酸配列からなるペプチド。
- 請求項8に記載のペプチドをコードするポリヌクレオチド。
- DNAまたはRNAから構成されることを特徴とする、請求項9に記載のポリヌクレオチド。
- 請求項9または請求項10に記載のポリヌクレオチドを含有するベクター。
- 請求項11に記載のベクターを含有する、遺伝子操作された宿主細胞。
