Abstract
Description

本開示は、N−(4−{[6,7−ビス(メンチルオキシ)キノリン−4−イル]オキシ}フェニル)−N’−(4−フルオロフェニル)サイクロプロペイン−1,1−ジカルボキシアミド、c−Met阻害剤のメタボライトに関する。
WO2005/030140は、カボザンチニブの合成(実施例48)を説明し、また、キナーゼの信号伝達を阻止、規制及び/または調整するこの分子の治療活性を開示する(アッセイ、表4、エントリ289)。実施例48は、WO2005/030140の段落[0353]に記載されている。
現在も、カボザンチニブに類似する活性プロフィールを示す化合物を識別する必要はある。
a)R1またはR2の一方が、H、SO3Hまたはグルクロン酸部分であり、他方がMeである。
b)R3は、OHまたはOSO3Hである。
c)R4は、R4がO−である場合にNがN+であるという条件で、O−である。
d)R5は、OHまたはOSO3Hである。
e)R6は、OHまたはOSO3Hである。
a)R1またはR2はMeである。あるいは、R1またはR2の一方がH、SO3Hあるいはグルクロン酸部分であり、他方がMeである。

カボザンチニブを哺乳動物へ投与すること。
哺乳動物における、哺乳動物の組織または体液におけるカボザンチニブのメタボライトのレベルまたは濃度を検出あるいは測定すること。
この場合、メタボライトは、以下のものからなるグループから選択される。

この試験の目的は、ヒト血漿、尿及び糞便内のカボザンチニブのメタボライトを捉えて識別することであった。血漿、尿及び糞便サンプルは、健康な男性対象者における、[14C]カボザンチニブ(100μCi)を含むカボザンチニブ(L−リンゴ酸塩)の単一175mg経口投与後の、カボザンチニブの質量バランス試験から採集した。
試験デザインと、血漿、尿及び糞便の採取。
血漿サンプルは、投与前、及び投与後の0.5、1、2、3、4、5、8、14、24、72、168、336、504及び648時間に採集した。尿サンプルは、投与前、投与後の0から8時間、8から24時間、24時間間隔で480時間まで、及び、48時間を超える間隔で投与後504から1152時間までのものを採集した。糞便サンプルは、投与前、24時間間隔で投与後480時間まで、及び、48時間を超える間隔で投与後504から1152時間までのものを採集した。すべてのサンプルはQPSLLC(ニューアーク、DE)へ送り、−70℃で保管した。十分なレベルの放射能を持つサンプルに対するメタボライト・プロファイリング及び識別のために、放射性流通検出器(RFD)に結合したHPLC/タンデムMSを使用した。
