Abstract
Description
・罹患又は欠陥の大動脈、肺、僧帽弁、三尖弁及び末梢静脈弁を容易かつ選択的に代えるために使用することができるシームレスな人工組織弁の提供。
・血管へ治療後の内膜過形成及び従来のポリマー製及び金属製弁に関連する過酷な生物学的応答を実施的に減少又は序器するシームレスな人工組織弁の提供。
・宿主組織の増殖、部位特異的な構造的及び機能的特性を有する新組織及び組織構造のバイリモデリング及び再生を誘導するシームレスな人工組織弁の提供。
・宿主組織に薬剤を投与することができ、これにより、所望の生物学的及び/又は治療効果をもたらすシームレス人工組織弁の提供。
・所定の時間の間臓血管組織の近傍に弁を一時的に配置する係止機構を含むシームレス人工組織弁の提供。
・血管構造との最適な機械的適合性を示すシームレス人工組織弁の提供。
弁のいくつかの実施例に関連して用いられる「係止機構」及び「係止」という用語は、弁を血管組織に最も近くに一時的に置くために構成され、用いられる一時の構造を意味する。本明細書において詳細に述べられるように、本発明のいくつかの実施例においては、係止機構は所定の期間、受容者の心血管組織に最も近くに血管組織を一時的に置くように設計され、構成されている。そして、いくつかの態様においては、所定の期間は、好ましくは新たな組織の再生工程内である。

Claims (15)
- シームレス人工弁であって、
外側の管腔表面と、第一及び第二の端部と、中間の三重壁部と、少なくとも第一の弁尖とを有する連続的な管状部材を含んでなり、前記第一の弁尖は前記シームレス人工弁を通過する流体を選択的に制限し、
前記中間の三重壁部は、前記管状部材を覆うように前記管状部材の前記第一の端部を外にめくり返して、二重壁第一端部及び該第一端部から伸長する二重壁部を形成し、前記管状部材の前記二重壁部を覆うように前記管状部材の前記第一の端部を反転させることにより形成され、
前記第一の弁尖は第一の交連接続点で前記中間の三重壁部を縫合することにより形成される、シームレス人工弁。 - 前記中間の三重壁部が二つの交連接続点で縫合され、このことにより、前記第一の弁尖と第二の弁尖が形成される、請求項1に記載の人工弁。
- 前記中間の三重壁部が、三つの交連の接続点で縫合される、このことにより、前記第一の弁尖、第二及び第三の弁尖が形成される、請求項1に記載の人工弁。
- 前記管状部材が、哺乳動物組織源由来の細胞外マトリックス( ECM )材料を含む、請求項1に記載の人工弁。

- 前記ECM材料が追加の生物学的活性剤を含む、請求項4の人工弁。

- 前記生物学的活性剤が、血小板由来成長因子(PDGF)、表皮成長因子(EGF)、トランスフォーミング成長因子α(TGF−alpha)、トランスフォーミング成長因子β(TGF−beta)、線維芽細胞増殖因子2(FGF−2)、塩基性線維芽細胞増殖因子(bFGF)、血管内皮増殖因子(VEGF)、肝細胞増殖因子(HGF)、インスリン様成長因子(IGF)、神経成長因子(NGF)、血小板由来成長因子(PDGF)、腫瘍壊死因子α(TNA−alpha)、及び胎盤成長因子(PLGF)からなる群から選択される、請求項7に記載の人工弁。
- 前記ECM材料が追加の薬剤を含む、請求項4に記載の人工弁。
- 前記薬剤が抗炎症含む、請求項10に記載の人工弁。

- 前記スタチンが、アトルバスタチン、セリバスタチン、フルバスタチン、ロバスタチン、メバスタチン、ピタバスタチン、プラバスタチン、ロスバスタチン及びシンバスタチンからなる群から選択される、請求項12に記載の人工弁。
- 前記薬剤が抗不整脈薬を含む、請求項10に記載の人工弁 。
