Abstract
Description
URA3遺伝子、アンピシリン耐性遺伝子、酵母2μ複製開始点、グリセルアルデヒド−3−リン酸デヒドロゲナーゼ(GPD)プロモーター及びホスホグリセリン酸キナーゼの酵母転写終結シグナルを含む、国際公開第2008080989号の実施例1に記載の酵母エピソームベクターpTT−10301を、3’末端でHis−タグを有し、天然のhTF配列(配列番号5)の潜在的なNグリコシル化部位の内の1つを不活性化するAsn124Ala変異を有する成熟hTFタンパク質(配列番号1のアミノ酸33−295位)をコードする配列番号6のcDNAの、GPDプロモーターの制御下でのクローニングに使用した。
この試験において参照として使用するFSは、Tissucol(登録商標)として市販されているFSである。1mLのTissucol(登録商標)は、500Uのヒトトロンビン、115mgのフィブリノーゲン、アプロチニン3000U及び塩化カルシウム40μmolを含有する。
実施するアッセイは、The Journal of Trauma Injury, Infection, and Critical Care, 2000, vol. 49 (2), p. 246-250でKemal Karakayaらにより開示されているものである。
実施例2で使用した各参照FS及び組み合わせを、2.5mLのヘパリン(Clexane(登録商標))処理血漿を含有する4cm長のレーンの一方の端部上に投与した。
国際公開97/29792号に開示された組成物の止血特性及び閉塞特性を測定するために、いくつかのトロンビンフリーのシーラント組成物を調製し、メーカーの推奨(ROTEM(登録商標)の取扱説明書)に従ってTEMにより分析した。凝塊全体の強度に関連するパラメーターMCF(最大凝塊硬度)及びパラメーターG(弾性モジュラス強度(elasticmodulus strength))を記録した。
1:FII、FV、FVII、FX、FXIII、FI(起源:5例の異なる健康なドナーから得た、貯蔵した血漿)+脂質修飾TF 200ng/mL(起源:TT−173)+塩化カルシウム 5mM。
2:FII、FV、FVII、FX、FXIII、FI(起源:5例の異なる健康なドナーから得た、貯蔵した血漿)+脂質修飾TF 200ng/mL(起源:TT−173)+FI 3mg/mL(起源SIGMA.商品番号F4129)+塩化カルシウム 5mM
3:FII、FV、FVII、FX、FXIII、FI(起源:5例の異なる健康なドナーから得た、プールした血漿)+脂質修飾TF 200ng/mL(AmericanDiagnostica 商品番号4500L)+塩化カルシウム 5mM

5:FII:100μg/mL、FV:10μg/mL、FVII:0.5μg/mL、FX:8μg/mL、FXIII:10μg/mL、FI:6mg/mL(起源:FI(商品番号F4129 SIGMA)、FII:(商品番号HCP−0010 CellSystems)、FXIII:(商品番号HCXIII−0160 CellSystems)、FX(商品番号HCX−0050 CellSystems)、FV(商品番号HCV−0100 CellSystems)、FVII(商品番号HCVII0030 CellSystems))+脂質修飾TF 200ng/mL(起源:TT−173)+塩化カルシウム 5mM。これらの成分の混合物を、その使用10分前に新たに調製した。

以下により産生された凝塊
(a)市販されていないFS(実施例5での組成物を参照)30μL
(b)3成分の組み合わせFS+TT173(実施例6での組成物を参照)30μL、又は
(c)同様の3成分の組み合わせではあるが、TFの起源としてTT−173の代わりに脂質修飾rTF(200ng/mL。起源AmericanDiagnostica 商品番号4500L)を含有する3成分の組み合わせ30μL
を、300μLのヘパリン添加血漿(0.6U/mL)に添加した。次いで、凝塊時間(CT=凝塊の投与から最初のフィブリンネットの出現までの時間)パラメーター、凝塊形成時間(CFT=最初のフィブリンネットの形成から全凝塊の形成までの時間)パラメーター及び最大凝塊形成(MCF=凝塊硬度)パラメーターを、メーカーの推奨(ROTEM(登録商標)の取扱説明書)に従ってTEMにより測定した。
