Abstract
Description
−マイクロ波透過材料で作製され、中心軸を有する略円筒形の反応器であって、入口及び出口を有する、反応器と、
−反応器の出口に付随した多孔質フリットであり、反応器からのビーズの放出を防止し且つ溶媒の放出を可能にする多孔質フリットと、
−時計回り方向及び反時計回り方向交互の、中心軸を中心とする反応器の同心回転のための手段と
を含む、液体溶媒と混合された固相樹脂ビーズを使用するマイクロ波アシスト固相合成のための装置により達成される。
−前段で定められている通りに装置を設けるステップであって、反応器は官能性樹脂ビーズ、溶媒、及び反応物を含む、設けるステップと、
−交互の方向に中心軸を中心に反応器を回転させることにより反応器の内容物によって混合すると同時に、反応器をマイクロ波加熱するステップと
を含む、マイクロ波アシスト固相合成のための方法に関する。
−官能性樹脂ビーズを反応器内に配置し(例えばポリスチレンの各ビーズが、少なくとも1つの保護アミノ酸又はアミノ酸配列を支持している)、溶媒で満たし、ミキサ・モータ(4)を作動させることにより混合する。
−シリンジの出口(7)経由で、真空を用いて排出する。
−溶媒で脱保護し、ミキサ・モータ(4)を作動させることにより混合する。
−シリンジの出口(7)経由で、真空を用いて排出する。
−マイクロ波加熱中にアミノ酸をカップリングし、ミキサ・モータ(4)を作動させることにより混合する。
−シリンジの出口(7)経由で、真空を用いて排出する。
−溶媒で洗浄し、ミキサ・モータ(4)を作動させることにより混合する。
−シリンジの出口(7)経由で、真空を用いて排出する。
−必要に応じて、洗浄を繰り返す。
Claims (9)
- 液体溶媒と混合された固相樹脂ビーズを使用するマイクロ波アシスト固相合成のための装置であって、
マイクロ波透過材料で作製され、中心軸を有する略円筒形の反応器(1)であって、入口(5)及び出口(6)を有する前記反応器(1)と、
前記反応器(1)の前記出口(6)に付随した多孔質フリットであって、前記反応器からのビーズの放出を防止し且つ前記溶媒の放出を可能にする前記多孔質フリットと、
時計回り方向及び反時計回り方向交互の、前記中心軸を中心とする前記反応器の同心回転のための手段(4)と
を含む、装置。 - 前記フリットは、真空の適用により前記出口への溶媒の放出を可能にするようになされている、請求項1に記載の装置。
- 前記フリットは約1μmから約50μmまでの細孔径を有する、請求項1又は2に記載の装置。
- 前記フリットは約5μmから約30μmまでの細孔径を有する、請求項3に記載の装置。
- 前記フリットは疎水性材料で作製されている、請求項1から4までのいずれか一項に記載の装置。
- 前記フリットはPTFEで作製されている、請求項5に記載の装置。
- マイクロ波アシスト固相合成のための方法であって、
請求項1から6までのいずれか一項に記載の装置を設けるステップであって、前記反応器(1)は官能性樹脂ビーズ、溶媒、及び反応物を含む前記設けるステップと、
前記反応器(1)をマイクロ波加熱するステップと、
時計回り方向及び反時計回り方向交互に、前記中心軸を中心に前記反応器を回転させることにより前記反応器(1)の内容物を混合するステップと
を含む、方法。 - 前記固相合成は固相ペプチド合成である、請求項7に記載の方法。
- 前記反応器バイアル(1)は基本的に垂直に配置される、請求項1から6までのいずれか一項に記載の装置。
