定義
本発明を詳細に説明する前に、本明細書における発明を説明するのに使用する用語の意味及び範囲を例示および定義付けするために以下のように定義する:

パーフォリン2の発現、機能、または活性を調節するか、あるいは、パーフォリン2の発現、機能、または活性に関連する1つまたは複数の標的分子の発現、機能、または活性を調節する薬剤の治療的有効量を投与することを含む、感染性疾患生物による感染に罹患している患者を治療する方法をさらに提供する。
本明細書で使用する場合、「感染性生物」は、例えば、細菌、ウイルス、真菌、寄生生物、および原生動物が挙げられるがこれらに限定されない。

本実施例は、Mpeg1が、重合により膜を貫通する孔を形成し、パーフォリン2(P2)と命名された新規孔形成タンパク質をコードすることを示している。マクロファージにおいてP2は、メチシリン耐性黄色ブドウ球菌(methicillin resistant Staphylococcus aureus、MRSA)、マイコバクテリウム・アビウム(Mycobacterium avium、Ma)、マイコバクテリウム・スメグマチス(Mycobacterium smegmatis、Msm)、ネズミチフス菌(Salmonella typhimurium、St)、および大腸菌(E.coli)を含む病原性細菌に対し強力な細胞内殺傷活性を有する。また、P2は、活性酸素種(ROS)および一酸化窒素(NO)の殺菌活性も可能にした。
プラスミド構築物:マウスMpeg−1 cDNAの完全コード領域を、いくつかのESTクローンから構築し、pEGFP−N3プラスミド(Clontech)に挿入した。単量体RFPをGFPの代わりにクローニングしたものをいくつかの実験で使用した。
マクロファージは、Mpeg1 mRNAを構成的に転写する。Mpeg1 mRNAからは、典型的に細胞溶解性孔形成タンパク質で見られるMACPFドメインを有するタンパク質が予測される。MACPFドメインの主に見られるメンバーは、細胞外細菌を殺傷する膜攻撃複合体の孔形成補体成分C9、ならびにウイルス感染細胞や腫瘍細胞を殺傷するTおよびNKリンパ球の孔形成分子であるパーフォリン1である。孔の形成は、構造変化を介したMACPFドメインの重合によって達成され、これにより4個の両親媒性β鎖の膜挿入が起こり、穿孔され標的細胞の死へつながる。本実施例では、Mpeg1が、重合により集合して膜を貫通する孔を形成するパーフォリン2(P2)と命名された新規孔形成タンパク質をコードすることを示している。マクロファージにおいてP2は、マイコバクテリウム・アビウム(M.avium)、マイコバクテリウム・スメグマチス(M.smegmatis)、ネズミチフス菌(S.typhimurium)、および大腸菌(E.coli)、ならびにメチシリン耐性黄色ブドウ球菌(MRSA)の臨床分離株を含む細胞内の病原性細菌に対し強力な殺傷活性を有する。重要なことに、P2は、ROSおよびNOの抗菌活性にも寄与している。
srcキナーゼがP2の活性化の要因であり、オートファジーが細菌の殺傷に重要であるだろうと仮定した。さらに、酵母ツーハイブリッドシステムを用いて、P2の細胞質ドメインと相互作用する7個のタンパク質が同定されたことにより、これらがP2の活性化に関与している証拠となる。これらのタンパク質を表1に示す。
マクロファージ、樹状細胞、およびミクログリアは、病原性細胞内細菌を殺傷するパーフォリン2(P2)と命名された孔形成タンパク質を構成的に発現する。P2は、膜を損傷することにより細菌を殺傷し、活性酸素種(ROS)および一酸化窒素(NO)の殺菌効果を増強する。これにより物理的損傷を起こし細菌の感受性の高い層へアクセスさせることが証明される。上皮細胞、線維芽細胞、および他の非造血由来細胞は、細菌が侵入すると、オートファジー関連メカニズムおよび抗菌性化合物を利用することにより細胞内細菌感染を除去する。これらの実験以前は、非造血細胞はP2を発現して細菌を除去するために利用することができるのか否かが不明であった。これは、初代ヒト角化細胞が構成的にP2 mRNAを発現すること、そしてこれまでに分析した全ての体細胞は、1型および2型インターフェロンによりP2 mRNAを発現するように誘導できることを示す最初の報告である。また、P2特異的siRNAにより内因性P2をノックダウンすると、殺菌活性が阻害される。この細菌活性はP2−RFPを補充すると回復されるが、RFPを補充しても回復されない。
ヒト細胞:以下の細胞を使用した。臍帯静脈内皮細胞、MIA PaCa−2膵臓癌細胞(ATCC CRL−1420)、UM−UC−3膀胱癌細胞(ATCC CRL−1749)、UM−UC−9膀胱癌細胞23、HeLa(ATCC CCL−2)、HEK293T細胞(ATCC CRL−1573)、および初代角化細胞。HUVECをLonza EGM−2 bulletキットで増殖させた。初代ヒト角化細胞は、先に記載のように増殖させた(Wiens,M.et al.J Biol Chem 280,27949−27959,doi:10.1074/jbc.M504049200(2005))。他のすべての細胞は、ATCCの推奨に従って増殖させた。全ての細胞を、5%CO2を含む加湿雰囲気中37℃で培養した。
今日までに分析されたヒトおよびマウスの初代細胞および細胞株は全て、インターフェロンの誘導によりP2 mRNAを急速に発現する(図14A〜14J、表2、および図18A〜18I)。刺激していない初代マウス胚性線維芽細胞(mEF)は、TAQMAN(商標)PCRによる39サイクル後でも、検出可能レベルのP2 mRNAを発現しない(図14A)。インターフェロン(IFNα、β、またはγ)は、添加によりそれぞれ単独でP2 mRNAを上方制御し、一緒に添加すると高レベルのP2 mRNAを誘導した。対照的に、LPS、IL−1α、およびTNFαはmEF中のP2 mRNAを誘導しない。個体差があるものの、検査したマウスおよびヒト由来の初代細胞および確立細胞株の全てにおいて、このパターンのP2誘導がみられた(図14A〜14F、図18A〜18I、および表2の完全なリスト)。IFNによるP2 mRNAの強力な上方制御にも関わらず、P2タンパク質は、プロテアソーム阻害剤MG132を添加しない限り、インターフェロンで活性化したmEFのウェスタンブロット分析によって検出されない(図14G)。このことは、急速なP2タンパク質のターンオーバーは、プロテアソームの分解を介して生じていることの証明となる。これらの発見の例外が、P2 mRNAおよびタンパク質を構成的に発現する初代ヒト角化細胞で起こる(図14H)。
反応性酸素および窒素の中間体ならびに透過性増加タンパク質は、重要な抗菌エフェクターであるが、追加的な殺菌エフェクターも存在すると考えられている。ここで、我々は、孔形成タンパク質パーフォリン2は、細胞内細菌の複製に対する防御に必須であることを示す。パーフォリン2は、身体全体にわたってまんべんなくグラム陽性および陰性菌そして抗酸菌を殺傷する。パーフォリン2欠損マウスは、通常のマウスでは殺菌される経口胃サルモネラの感染により死亡する。パーフォリン2は、MACPFキラードメインが、感染の際に細菌を含有する液胞に移行する膜小胞の内腔の方向を向いた膜貫通タンパク質である。パーフォリン2により殺傷された細菌は、細胞壁にクラスタ化された90Åの孔を有する。この孔より細菌の活性酸素および窒素の中間体に対する感受性がより高くなる。患者において非治癒性かつ慢性的な感染皮膚潰瘍では、病原性細菌がパーフォリン2の発現または活性化を妨害し、パーフォリン2のレベルを抑制している。パーフォリン2の作用経路を研究することは、生命を脅かす細菌感染に対する新しいアプローチの機会を提供することになるだろう。
生体内におけるパーフォリン2の生物学的な重要性を決定するために、我々は相同組換えによりパーフォリン2ノックアウト(P2-/-)マウスを作製した。マウスは、C57B16および129の混合株由来であり、これらは少数のMHC抗原に違いがあり、多様性が制限されたものである。P2-/-マウスは、病原体を含まない条件下では正常に発育し生殖する。ホモ接合型P2-/-、ヘテロ接合型P2+/-、および野生型P2+/+の同腹子に、ネズミチフス菌(サルモネラ菌)を経口で胃に接種し上述のように免疫性をテスト(チャレンジ)した。ストレプトマイシンで前処理してから24時間後のP2+/+マウスに105個のストレプトマイシン耐性サルモネラ菌を感染させると、感染後最初の5日間で軽度に体重減少(<10%)したが、その後9日目には完全に回復した(図25a)。対照的に、ホモ接合型P2-/-マウスは、出血性の下痢を発症し、それにより5日または6日目まで体重減少が進行したので、この時点でさらなる苦痛をなくし、分析するために安楽死させた。102個のサルモネラ菌によるチャレンジでさえも、P2-/-マウスでは、進行性の疾患が起こり死に致った。P2+/-同腹子は、P2+/+同腹子よりも重篤な疾患となったが回復した(図25a)。
P2-/-マウスでは、サルモネラ菌が急速に広まり、P2-/-マウスにおけるいずれの細胞も、細菌の浸潤を止めることができないことが示唆された。したがって、我々は、どの種類の細胞が構成的にP2を発現またはP2を発現するように誘導可能なのかを検討した。表2は、データをまとめたものである。PMN、マクロファージ、樹状細胞、およびミクログリアは、パーフォリン2 mRNAおよびタンパク質を構成的に発現し、これはさらに、IFN−γおよびLPSによって上方制御される(図示せず)。ヒト初代角化細胞および慢性創傷の縁から採取した組織試料由来の角化細胞(図27)も同様に構成的にP2を発現する。試験した他の全ての細胞および細胞株は、IFN−α、−β、または−γで処理した場合のみP2 mRNAを発現した。P2を発現する全ての細胞は、細胞内細菌感染を制御することができるが、P2がsiRNAでノックダウンされると破壊される。ヒトPMNおよびマクロファージは、構成的にP2タンパク質を発現する(図26a)。HL60からレチノイン酸によって誘導されたPMNにおいてパーフォリン2をノックダウンすると、殺菌活性が有意に阻害され、MRSA、M.スメグマチス、およびネズミチフス菌の細胞内複製が可能になる(図26a)。同様に、P2をノックダウンすると、腸上皮細胞(直腸癌CMT93)(図26b)、ヒト内皮細胞(HUVEC)(図26c)、およびヒト子宮頸癌上皮細胞HeLa細胞(図26d)の細胞内細菌の殺傷が有意に阻害される。P2−GFPのトランスフェクションによりP2の発現を上昇させると、マイコバクテリウム・アビウムを除去するために重要な殺菌活性が増加する(図26e)。P2の3’非翻訳領域に特異的なsiRNAでノックダウンした内因性P2は、P2−GFPまたはP2−RFPを用いたトランスフェクションによって補完され、MEF(図26f)または貪食細胞(図示せず)において完全に殺菌活性が再構築される。我々のデータは、P2は、普遍的に発現するまたは誘導可能で、検査した少なくとも3種類の細菌を殺傷するかまたはそれらの細胞内複製を阻害するために必要であることを累積的に示している。
P2の強力な殺菌作用により、細胞内細菌はパーフォリン2を回避するための戦略を進化させなければならないことが示唆される。原則的に、細菌は、非貪食細胞においてパーフォリン2の転写をブロックすることができ、これによりパーフォリン2発現細胞において構成的に下方制御を引き起こし、またはパーフォリン2の活性化および重合を妨害する。天然MEFを実験室株大腸菌K12に感染させると、P2 mRNAの迅速な誘導およびその後の細胞内細菌の殺傷が起こる。大腸菌とは異なり、生きた野生型ネズミチフス菌(サルモネラ菌)は、MEFにおいてP2 mRNAの誘導を引き起こすことはない(図27a)。対照的に、熱殺菌されたかまたはPhoP突然変異体のネズミチフス菌は、大腸菌と同程度にMEFにおいてP2 mRNAを誘導する。これにより、ネズミチフス菌はP2の誘導を積極的に抑制することが示唆されている。同様に、偏性細胞内細菌であるクラミジア・トラコマティスによるHeLa細胞の感染も、P2の発現を誘導しない。外因性のIFNによるP2の誘導は、de novoクラミジアのタンパク質合成を必要とするメカニズムを介して積極的に抑制される(図27b)。EPECはエンドサイトーシスをブロックすることができるが、貪食細胞は、この阻害を克服できる(図27c)。エンドサイトーシスされたEPECは、それらがユビキチン様分子であるNEDD8を不活性化するデアミダーゼをコードするCifプラスミド(図27d)を有する場合にのみ、パーフォリン2から保護される。NEDD8は、NFκBの活性化など多くの基本的な細胞経路に関与するcullin−ring型ユビキチンE3リガーゼ(CRL)を活性化するのに必要である。NEDD化はNEDD8特異的E2リガーゼUbc12によって行われる。P2の細胞質ドメインは、酵母ツーハイブリッドシステムではUbc12と相互作用し、P2と共に免疫沈降される(補足図27)ことより、CRLのUbc12介在性NEDD化はパーフォリン2介在性の殺傷に必要であり、CIFがこのステップをブロックすることが示唆される。
現在のパラダイムでは、細胞内細菌は、活性酸素および窒素種により、そして貪食された細菌がリソソームと融合するによって殺傷されることが示唆される。ここでは、我々は、各エフェクター経路およびそれらの相乗効果による殺菌効率を分析する。我々は、個別に各エフェクターをブロックし、ネズミチフス菌(サルモネラ菌)およびM.スメグマチスの細胞内生存および複製または細胞内殺傷について測定した。これらの実験を、腸上皮細胞(CMT93)(図示せず)およびIFN−γで活性化したPEM(図27e)を用いて実施したところ、実質的に同一の結果であった。まず、我々は、IFN−γで活性化しLPSで刺激したPEMは、ROSとNOを生産し、この生産はノックダウンによるP2 siRNAによって影響されないことを確認した。そして、使用したROSおよびNOの阻害剤は、ROSまたはNOのブロックに特異的かつ活性であったことも確認した(図示せず)。3つのエフェクターが全てスクランブルsiRNAでトランスフェクションした細胞において活性である場合、PEMは感染後の最初の4時間で細胞内サルモネラ菌の約90%を殺傷できる(図27e、実線)。P2をノックダウンすると、サルモネラ菌に対する殺菌活性が完全に破壊され、それらの細胞内複製が可能になった(図27e、破線)。NACおよびNAMEを用いて追加的にROSおよびNOを阻害しても、さらなる効果を示さなかった。また、P2の存在下でNOまたはROSを阻害すると(スクランブルsiRNA、青と緑の実線)、有意に殺菌活性が減少するが、P2の存在下では、細菌の複製はブロックされたままであった。M.スメグマチスは4時間以内のほうが殺菌エフェクターによる殺傷に対する耐性がより高い。それにもかかわらず、パーフォリン2のノックダウンによりM.スメグマチスの殺傷は有意に阻害され、追加的にROSまたはNOをブロックしてもそれ以上の効果がなかった。P2のノックダウンをせずに、ROSまたはNOをブロックしても、マイコバクテリアに対する殺菌活性への影響は制限され顕著ではなかった。対照的に、非病原性の実験用大腸菌K12は、P2がなくてもROSおよびNOによる細胞内殺傷に対して感受性があるが、P2の存在する場合よりも有意に効率が低かった。データでは、パーフォリン2が活性酸素および窒素種、そしてリゾチームと相乗作用することが示唆され、これは、以前に細菌が個々のエフェクターによる影響が異なると示されたことと一致する。
感染していない場合、パーフォリン2は、核周辺の小胞を区画する膜に埋め込まれて格納される(図28b)。したがって、パーフォリン2は、殺傷のために、細菌感染の場所に輸送されなければならない。パーフォリン2は、短く高度に保存された細胞質ドメインを有し、この細胞質ドメインが細胞質タンパク質と相互作用して、P2の移行および重合を引き起こす(図28a)。しかし、YからFに変異すると(図28aに赤い矢印で示す)P2の殺菌活性がブロックされる。よって、ここにP2の活性化に重要な機能があることが示唆される(図示せず)。
感染の数分以内に、パーフォリン2はそのエフェクタードメインが内腔の方を向いた状態で細菌を囲む膜上に蓄積するという発見により、パーフォリン2が、補体C9およびとパーフォリン1と同様の細胞傷害性のメカニズムで細菌の細胞壁に重合し、水充填孔を作成することによって、他の殺菌因子の透過を促進し、その細菌を死に至らしめることができることということが示唆される。我々は、今まで報告されていなかったパーフォリン2の孔形成能力を決定した。我々は、C9またはパーフォリン1モノマーと類似したパーフォリン2のモノマーが細胞傷害性ではないこと、そして、その孔形成にP2の細胞質ドメインと未知のシグナル伝達タンパク質との特異的相互作用を介するP2の重合が必要であることを推論した。パーフォリン2の細胞質ドメインは、保存されたRKYKKK配列(配列番号4)を含み(図28a、青い矢印)、膜の反対側にあるP2の重合を誘発するタンパク分解性切断部位として機能し得る膜貫通ドメインの隣にある。これは、P2−GFPでトランスフェクトしたHEK293細胞から、分画遠心法および低レベルのトリプシンでそれらを短時間処理して細胞質ドメインを切断することにより、パーフォリン2−GFPを有する膜を精製することによって検査した(図示せず)。トリプシン処理したパーフォリン2−GFP−膜の電子顕微鏡検査により、85〜95Åの平均内径を持つ典型的な貫通孔が示される(図29a、白矢印)。さらに、重合中にパーフォリン2の融合によって形成された半環状で8の字形の不規則な構造(黒矢印)は、環が閉じて終了するまで継続する重合プロセスと一致する。C9およびパーフォリン1と同様に、閉環は、膜タンパク質の立体障害によってブロックされ得る。トランスフェクトされていない細胞は、このような孔を有していなかった(図示せず)。これらのデータは、パーフォリン2が実際に、液胞の内部にある細菌の細胞壁に孔を形成することが潜在的に可能な孔形成膜貫通タンパク質であることを直接示している。パーフォリン2は、小胞の内腔の方向に向いているMACPFドメインを有する膜タンパク質であるので、孔の形成は、小胞内部のMACPFドメインに接触している膜に起こるのであろう。
プラスミド構築物
マウスmpeg1 cDNAの完全コード領域を、いくつかのESTクローンから構築し、pEGFP−N3プラスミド(Clontech)に挿入した。単量体RFP(R.Flavell,Yale)を、GFPの代わりにクローニングし、いくつかの実験で使用した。
RAW264.7、J774、HL−60、およびHEK−293細胞株を、ATCCから得た。BV2ミクログリア細胞株は、University of MiamiのDr.J.Bethea氏から譲渡した。全ての細胞を、ATCCの推奨に従って5%CO2を含む加湿雰囲気中、37℃で培養した。HL−60は、先の記載のように、レチノイン酸を用いてPMN表現型へ分化させた。マウス初代マクロファージは、先の記載のように、チオグリコール酸誘発性の腹腔または骨髄から得た。ヒトマクロファージおよびPMNを新鮮な健康なドナーのPBMCから単離した。ヒトマクロファージは、先の記載のように単球から分化させた。ヒトPMNは、先の記載のように単離した。マウス胚線維芽細胞(MEF)は、先の記載のように単離した。
ネズミチフス菌株LT2Z(University of MiamiのDr.G.Plano氏から譲渡)、K12大腸菌、およびメチシリン耐性黄色ブドウ球菌(University of MiamiのDr.L.Plano氏から譲渡)は、ルリアブロス(LB)で増殖した。
M.アビウム・イントラセルラーレ(University of MiamiのDr.T.Cleary氏から譲渡)、およびM.スメグマチス(ATCC)は、ミドルブロック7H9ブロスで増殖した。
リポ多糖(LPS)は、Sigmaから購入し1ng/mlの最終濃度で使用した。組換えヒトおよびマウスIFN−γはpreprotechから購入し、100U/mlの最終濃度で使用した。N−アセチルシステインおよびL−NAMEいずれもSigmaから購入した。
ウサギ抗Mpeg1、ヒト抗Mpeg1、および抗GFPポリクローナル抗体をAbcamから入手し、ウェスタンブロット分析に使用した。ウサギ抗パーフォリン2(細胞質ドメイン)抗血清は、21st Centuryより作製したものを得た。
RNAを、RNeasy(Qiagen)の説明書に従い細胞から抽出した。1μgのRNAを、QuantiTect逆転写キット(Qiagen)を用いて同社のプロトコルに従いcDNAに変換した。qRT−PCRは、マウスmpeg1、GAPDH、およびβ−アクチンについて、TAQMAN(登録商標)遺伝子発現アッセイ(Applied Biosystems)を用いて行った。ヒト組織ついては、ヒトmpeg1、GAPDH、およびβ−アクチンプローブを利用した。全てのアッセイは、Applied Biosystems 7300 PCRプラットフォーム上で実施した。
細胞内殺菌活性を応用した。簡単に言うと、細菌を、ルリアブロス(LB)中で16〜18時間(ネズミチフス菌、黄色ブドウ球菌、および大腸菌)またはミドルブロック7H9ブロス中で24時間(マイコバクテリア)振盪しながら37℃で増殖してその後感染させた。ネズミチフス菌、大腸菌、および黄色ブドウ球菌については、培養物をその後LBで1:33に希釈し、そしてさらに3時間増殖させて対数期に到達させ、ネズミチフス菌については侵襲的な表現型を誘導した。真核細胞は、各細胞について最適化されたLonzaのプロトコルに従ってトランスフェクトし、トランスフェクト後12ウェルプレートにプレーティングした。RAで分化させたHL−60細胞には刺激を与えなかった。RAW264.7細胞は、LPS(1ng/ml)およびIFN−γ(100U/ml)で14時間刺激し、マクロファージ系統に分化させた。他の全ての細胞は種特異的なIFN−γ(100U/ml)で刺激した。細胞を37℃、5%CO2のインキュベーターで、30分間(ネズミチフス菌)または1時間(黄色ブドウ球菌、大腸菌、およびM.スメグマチス)、10〜60の感染多重度(MOI)で感染させた。感染後、細胞を氷冷PBSで2回洗浄し、50μg/mlゲンタマイシンを含有する新鮮な培地を添加した。2時間後、培地のゲンタマイシンの濃度を5μg/mlに低下させた。指示した時点で、細胞をPBSで洗浄し、1%Igepalを含むddH2Oで溶解、希釈し、LB寒天プレート(ネズミチフス菌、黄色ブドウ球菌、大腸菌)またはミドルブロック7H11プレート(マイコバクテリア)に三連でプレーティングし、十分なコロニーが増殖してからCFUを決定した。
ゲンタマイシン保護アッセイを変更して、ゲンタマイシン非含有細胞内細菌殺傷アッセイを作成した。上記の変更には、感染において90〜100%のコンフルエンスを達成するように真核細胞をプレーティングすること、および感染多重度(MOI)を5〜15に減少することが含まれた。侵入時間は変更せず、ネズミチフス菌については30分、黄色ブドウ球菌、大腸菌、およびM.スメグマチスについては1時間とし、感染は、37℃、5%CO2のインキュベーターで行った。細胞外細菌を確実に最大除去するために、洗浄工程を変更し、細胞を氷冷PBSで2回洗浄し、付着した細胞外細菌を除去しやすくするようトリプシン処理し、そして氷冷PBSでさらに3回洗浄した。4時間ごとに培地を除去し、細胞をPBSで2回洗浄し、その後、新鮮な培地を再度追加した。指示した時点で、細胞をPBSで3回洗浄し、1%Igepalを含むddH2Oで溶解、希釈し、LB寒天プレート(ネズミチフス菌、黄色ブドウ球菌、大腸菌)またはミドルブロック7H11プレート(マイコバクテリア)に三連でプレーティングし、十分なコロニーが増殖してからCFUを決定した。
マウス細胞については、3つのパーフォリン2特異的に化学合成された19ヌクレオチドのsiRNA二重鎖を、Sigmaから入手した。2つのsiRNAは、パーフォリン2の3’UTRに相補的で、もう1つはコード領域に相補的であった。配列を次に示す:CCACCUCACUUUCUAUCAA(配列番号1)、GAGUAUUCUAGGAAACUUU(配列番号2)、およびCAAUCAAGCUCUUGUGCAC(配列番号3)。反応の対照としてスクランブルsiRNAも作製した。ヒト細胞については、3つのヒトパーフォリン2特異的サイレンサー選択siRNAをAmbion(Invitrogen)から購入した。そのサイレンサー選択番号は、s61053、s47810、s61054である。Ambion(Invitrogen)のサイレンサー選択陰性対照#2も使用した。すべての細胞へのsiRNAのトランスフェクションは、Amaxa Nucleofector System(Lonza)を用いて製造元の指示に従い行った。全てのトランスフェクションは、指定する場合、1〜4×106個の細胞、最終濃度が300nMのsiRNA(パーフォリン2特異的siRNAをプールした)、および2μgのプラスミドDNAを用いて行った。トランスフェクション後直ちに細胞を抗生物質を含まない培地にプレーティングした。
原核生物膜:膜は、N2キャビテーションおよび分画遠心によって安定的にトランスフェクトしたパーフォリン2−GFP HEK−293細胞から単離した。膜は、小量の中性トリス緩衝食塩水中に再懸濁し、37℃で100μg/mlトリプシンで1時間処理してから洗浄し、そして5%の中性リンタングステン酸Naで30秒間ネガティブ染色した。画像は、Phillips CM10透過型電子顕微鏡で52,000倍の初期倍率で撮影した。
パーフォリン2−GFPを局在化するために、RAW264.7細胞をパーフォリン2−GFPで一過性トランスフェクトし、ガラス底のディッシュ内でNo.1.5カバーガラス(MatTek Corp)を用いLPS(1ng/ml)およびIFN−γ(100U/ml)で一晩刺激した。細胞をPBSで1回洗浄し、オルガネラを標識した。小胞体(ER)の標識では、ER−Tracker(商標)Blue−White DPX(Invitrogen)を1μΜの作用濃度で37℃で30分間使用した。他の全ての染色では、トランスフェクトした細胞を室温で15分間、3%パラホルムアルデヒド(PFA)で固定し、0.5%サポニンで透過し、10%正常ヤギ血清でブロックし、そして一次抗体および二次抗体と共にインキュベートした。抗CD107a(LAMP−1)(BD Pharmingen)、抗CD11b(BD Pharmingen)、抗golgin97(Invitrogen)、抗EEA1(Calbiochem)、抗GM130(BD biosciences),およびHoechst 33258(Invitrogen)を使用して細胞オルガネラを同定した。二次抗体は、全てヤギで上昇した。標本を、PBS中に保持し、電動ステージとLeica DFC495カメラ付のLeica SP5 SP5倒立共焦点顕微鏡にて室温で撮影した。画像を、Leicaアプリケーションスイートの改良型蛍光ソフトウェアを用い、デコンボリューション処理を応用して分析した。
マクロファージ、ミクログリア、mEF、HeLa、および他の新鮮な外植細胞あるいは細胞株におけるP2 siRNAによるP2のノックダウンにより、病原性ネズミチフス菌、メチシリン耐性黄色ブドウ球菌(MRSA)、マイコバクテリウム・スメグマチス(図30A)、およびマイコバクテリウム・アビウム(図示せず)等の細胞内細菌の殺傷および除去をブロックすることが、ゲンタマイシン保護アッセイおよび抗生物質の非存在下での測定(図示せず)によりわかった。図30の直腸上皮細胞(A)またはマウス胚線維芽細胞(MEF)(B)では、感染の24時間前に、P2の3’UTRに特異的な2つのsiRNAのプール、またはP2 RNAおよびタンパク質をノックダウンするスクランブルsiRNAを用いて、一過性トランスフェクトした(図31C)。P2の補完について(B)、細胞を、P2 siRNAおよびノックダウン耐性P2−RFP(内因性P2の3’UTRを発現していない)、または対照RFPと共にトランスフェクトした。P2−RFPは、モノマーDsRed(RFP)を有しP2がC末端にタグされた融合タンパク質である。トランスフェクトしたP2−RFPは、細菌殺傷の点で完全に活性である(図30B、C)。16時間目に、IFN−γを加えP2 mRNAを誘導した(P2 mRNAは、P2 siRNAが存在する場合に抑制される)。時刻0において、細胞を、1個の細胞あたり30〜50個の細菌という感染多重度(MOI)で病原性MRSA、ネズミチフス菌、またはマイコバクテリウム・スメグマチスを30〜60分間感染させた。細胞内感染の後、細胞外の細菌をPBSで洗い流し、そして細菌の細胞外の複製を防止するために細胞をゲンタマイシンに再プレーティングした。宿主細胞は、感染直後およびそれ以降のいくつかの時点で、穏やかな界面活性剤(NP40)で溶解し、生存している細菌を反復コロニー形成アッセイによってカウントした。穏やかな界面活性剤は、宿主細胞の膜を溶解するが、細菌の細胞壁は溶解しない。
細胞内においてRASA2がP2の細胞質ドメインと相互作用することを酵母ツーハイブリッドシステムで検証するために、我々は、RAWマクロファージにおける2つのタンパク質の局在を決定した。マクロファージは、構成的にP2を発現する。共局在を決定するために、我々は、P2の細胞質ドメインのC末端にリンカーを介して融合したP2−GFP融合タンパク質を作成した。P2−GFPは、補完アッセイにおいて細胞内細菌の殺傷に機能する(図30B、図31)。P2−GFPは、RAW細胞の核周辺膜小胞に局在しており(図33、上)、RASA2(図33、中央)と類似する。画像をマージすると、P2−GFPおよびRASA2が多数の核周囲小胞に共局在しているように見える(図33、下)。我々は、RASA2がvps34リン酸化の際に細菌を含有する液胞に移行していることを示す。
P2の誘導、補充、移行、活性化、または重合に必要な任意の分子をブロックすることもまた、細胞内細菌の殺傷を阻止すると考えられる。RASA2がP2の移行、活性化、および/または殺傷に必要な場合は、そのノックダウンにより、細胞内細菌の殺傷が阻害されるはずである。このことについて、細胞内M.スメグマチスを殺傷するBV2ミクログリア細胞を用いた最初の実験で検査した(図35a)。RASA2のノックダウンは、P2のノックダウンと同様に細胞内殺傷を完全にブロックし、細胞内複製を可能にしたので、RASA2は、細胞内細菌の殺傷に重要であることが示唆される。
我々は、抗RASA2モノクローナル抗体でBV2−およびmEF−溶解物を免疫沈降し、SDS−PAGEによって分離、そして抗P2抗体を用いた免疫ブロッティングをする予定である。また、これらの研究は、それぞれP2−GFPのトランスフェクト、および抗GFPを用いた免疫沈降、ならびに抗GFPおよび抗RASA2を用いた免疫ブロッティング(図35b)の後に実施する。GFPトランスフェクション(P2を融合していない)は、対照として機能する(図示せず)。実験は、IFN前誘導あり及びなし、そして細菌感染あり及びなしで実施する。また、我々は、P2−GFP免疫沈降物でタンパク質を染色することによってさらなるタンパク質バンドを探し、質量分析によってそれらを特徴づける予定である。GFP免疫沈降物は対照として機能する。我々はまた、P2の細胞質ドメインおよびRASA2の突然変異体の変異解析を実施する予定である。
RASA2は、N末端セグメント内に2つのC2ドメイン(C2AおよびC2B)を有しており、そこからC末端方向へRasGapドメインおよびPH/Btkドメインが続く。C2ドメインは、細胞膜へタンパク質を標的にすることに関与するタンパク質構造ドメインであるが、RASA2におけるそれらの機能についてはあまり研究されていない。PH−Btkドメインは、PIPの結合に必要である。RASA2は、可溶性のIP(3,4,5)P3、PI(1,3,4,5,6)P5、IP6に結合するが、IP(1,4,5)P3には結合しない。また、RASA2は、PtdIns(3,4,5)P3にも結合するので、その結合にはイノシトール3リン酸化が必要であることが示唆される。RASA2が内在性膜上でvps34により構成的に合成されるPtdInsP(3)Pに結合するか否かについては、知られていない。
図35におけるP2の移行は、ネズミチフス菌および実験用株大腸菌K12の感染によるものである。我々はまた、サルモネラ菌、マイコバクテリア、および他の細菌を用いて、野生型および変異型P2、ならびに野生型および変異型RASA2の移行をそれぞれ決定する予定である。マイコバクテリアは、PI3Pの生成と液胞の成熟を妨害する。サルモネラ3型分泌エフェクターSopBは、液胞の成熟を妨げ、サルモネラ菌を含む液胞、SCV内の細胞内生存を増強するというPI3Pに対する複数の効果を有する。サルモネラ菌を含む液胞に対しPI3Pを変化させると、P2およびRASA2の移行の両方に対し、大腸菌に対する場合よりも影響を及ぼす可能性がある。
マウスP2の細胞質配列において保存されているアミノ酸は(RKYKKK配列番号4)(ハイライト)であり、それに続いて、保存されたY、そして酸性EIEEQE(配列番号5)があり、その後保存されたSが続き、C末端の近くに追加的なSがある(図28A)。いくつかのキナーゼファミリーを、潜在的なリン酸化部位についてのアルゴリズムを用いて同定した。我々は、P2の細胞質ドメインは、細菌感染時に、細菌を含有する液胞へのP2の移行および細菌を死に至らしめる細胞壁でのP2の重合をもたらす信号を受けることを示す。我々は、(a)KYKK(配列番号7)をQYQQ(配列番号8)に;(b)保存されたYをFに、そして(c)2個の保存されたSをA(ブロック)またはD(構成的に活性)に変異させる予定である(図28a参照)。我々はすでにフローサイトメトリーによる決定においてP2−RFPにおけるYからFへの変異が安定して発現されるが、siRNAによる内因性P2のノックダウンを補完するために使用する場合、細菌殺傷活性が破壊されることを確立した。
オートファジー(ゼノファジー)は感染によって活性化される。オートファジーの細胞内開始部位は、オートファゴソームへと成長するファゴフォアーの核形成部位を決定するvps34複合体によるイノシトールの3位をPI(3,4,5)P3やPI3Pへリン酸化することによって決定される。
オートファジーは、オートファジー膜の段階的な組み立ておよびその後のリソソームとの融合により媒介される。我々は、感染性オートファジーにおいて、P2が細菌を含有する液胞および/または初期のオートファゴソームに補充され、ここで、膜貫通ドメインを介してP2が膜に繋留したまま細菌を死に致らしめていることを示す(図28aを参照)。P2により殺傷された細菌は、細胞壁に孔があるので、パーフォリン2の重合により穿孔されたことが強く示唆される(図37)。MRSAは、感染後に4時間界面活性剤を用いて溶解することによりmEFから得られ、電子顕微鏡により分析したところ、典型的に90Åの細胞壁の孔が見られ(図36左パネル)、これは真核生物膜のP2孔の大きさと類似していた(図36の右パネル)。P2による孔と推定されるものも、mEFで殺傷したマイコバクテリアの細胞壁に存在する(データは示さず)。P2孔は、ROS、NO、およびリゾチームを細菌の細胞壁を通して透過させやすくして殺傷を完了する可能性がある。P2/オートファジーのリンクについての我々の示唆によれば、P2の補充前の段階におけるオートファジーをブロックすると細菌の殺傷がブロックされるであろう。P2が補充され重合した後にオートファジーの段階をブロックしても、殺傷が損なわれることはないだろう。Atg14Lはオートファジーにおけるvps34 PI3−キナーゼ複合体の標的成分である。P2のノックダウンと同様に、siRNAによりAtg14Lをノックダウンさせると、完全に活性なP2を存在させても、BV2におけるマイコバクテリアの殺傷がブロックされる(図37)。
図32は、感染の5分以内に細菌を含有する液胞へP2が移行する例を示す。この実験では、内因性P2をsiRNAでノックダウンし、同時に細胞をP2−RFPでトランスフェクトした。16時間後、細胞をGFP発現大腸菌K12に感染させ、感染後5分間パラホルムアルデヒドで固定し、共焦点顕微鏡によって撮影した。図32aは、P2およびLC3が、感染の5分以内に細菌を含有する液胞上に共局在していることを示す。
ATG9L1は、IFNによってP2を発現するよう誘導していないmEFにおけるサルモネラ菌の細胞内複製を遅延させるのに必要である。我々は、P2が存在しない場合、細菌がmEFにおいて複製するだろうこと、そして図34bにおいてサルモネラ菌がP2誘導を抑制していることを示した。したがって、我々は、P2発現を誘導したまたはしない野生型およびATGL1ノックダウン細胞を用いた画像化によって、ATG9L1の役割を研究する予定である。我々はまた、P2−GFPまたは/およびASA2を用いた共免疫沈降についても観察する予定である。ATG9L1は60〜90nm以下の脂質小胞に埋め込まれた6個の膜貫通性タンパク質であるので、P2と相互作用し、同様の移行経路を用いている可能性のある候補である。
Claims (17)
- パーフォリン2(P2)の機能、活性、または発現を調節する方法であって、P2の発現、機能、または活性に関連する1つまたは複数の分子の機能、活性、または発現を調節する少なくとも1つの薬剤の有効量を患者に投与し;P2の機能または発現を調節する;ことを含む方法。
- P2の機能、活性、または発現に関連する1つまたは複数の分子は、src、ユビキチン結合酵素E2M(Ubc12)、GAPDH、P21RAS/gap1m(RASA2)、ガレクチン3、ユビキチンC(UCHL1)、プロテアソーム、vps34、ATG5、ATG7、ATG9L1、ATG14L、ATG16L、LC3、Rab5、またはそれらの断片を含む、請求項1に記載の方法。
- 前記分子は、P2の転写または翻訳を阻害する、請求項1に記載の方法。
- 薬剤は、低分子、タンパク質、ペプチド、ポリペプチド、修飾ペプチド、修飾オリゴヌクレオチド、オリゴヌクレオチド、ポリヌクレオチド、合成分子、天然分子、有機または無機分子、またはそれらの組み合わせを含む、請求項1に記載の方法。

- パーフォリン2タンパク質をコードする遺伝子の破壊を含むトランスジェニックマウス。
- 前記破壊は、パーフォリン2タンパク質をコードする前記遺伝子のヘテロ接合性またはホモ接合性破壊を含む、請求項13に記載のトランスジェニックマウス。
- 前記破壊は、ホモ接合性破壊を含み、前記ホモ接合性破壊は、前記トランスジェニックマウスにおける前記遺伝子を不活性化し、機能的パーフォリン2タンパク質の発現を阻害する、請求項14に記載のトランスジェニックマウス。
- 前記トランスジェニックマウスは、野生型マウスと比較して、細胞内病原体による感染に対する感受性の増加を示す、請求項13〜15のいずれか1項に記載のトランスジェニックマウス。
- 請求項13〜15のいずれか1項に記載のトランスジェニックマウス由来の器官、組織、細胞、または細胞株。
