Abstract
Description
(a)通常0.5マイクロメートル以上の深さと、通常約2マイクロメートルのデューティサイクル(duty cycle)からなるマイクロスケールのパターンを施すことによる、コンタクトガイダンスを介した強化された創傷治療。
(b)接着斑の関与無しのコンタクトガイダンス。これは、創傷に対する被覆材の接着の問題なく、創傷治療の改良を可能にする新たな生物学的概念である
創傷治療を補助するパッチは、混合比1:10のポリジメチルシロキサン(PDMS,Dow Corning社製、米国)を用いて製造される。混合したPDMSは、閉じ込められた空気を除去するため、真空チャンバにおいて10分間脱気され、2μmの周期、1μmの溝幅及び0.6μmの溝深さの平行な溝からなるマイクロパターンが施された環状オレフィン・コポリマー(COC)の型に、500μmの厚さで注ぐ。次に、PDMSは、簡単に二回脱気され、60°Cで4時間硬化させた。硬化させたPDMSを、ピンセットで型から分離し、メスで1cm2の正方形に切断した。比較目的のため、ブランクパッチを、同様に、平坦なCOC基板に注がれたPDMSを用いて作成した。続いて、全てのパッチを、いかなる非架橋材料をも溶解するエタノール中に一晩放置した。その後、表面の親水性を増加させるため、パッチを、酸素プラズマで処理した。30〜150秒の間隔の範囲をテストした後、10Wで120秒の処理時間が最も低い接触角(20.2±0.5°)を生じるものとして選択された。図9は、異なるプラズマ処理時の活性PDMSパッチ表面において水の静的接触角を測定し、未処理のPDMSパッチの接触角を、低電力(10W)のプラズマで30,60,90,120秒ごとに処理したパッチの接触角、及びゼラチンコーティングされたPDMSの接触角と比較したテストを示す。その結果、パッチの剛性を、一軸試験により測定したところ、それらのヤング率は、1.53±0.057MPaと算出された。個々の線維芽細胞が10〜100nNの範囲内においてけん引力を生成することができるので、創傷治療中の表面上の地形の変形は無視できるとする仮定は妥当である。
接着斑及びアクチンフィラメントの可視化を必要とする実験において、細胞を、ネオン(登録商標)トランスフェクションシステム(Invitrogen社製,米国)を用いてトランスフェクトした。このとき、LifeAct−EGFP(緑色蛍光タンパク質)及びビンキュリン−FP635(遠赤色蛍光タンパク質)構築物を使用した。
マウスモノクローナル[A17]抗フィブロネクチン抗体(ab26245)はAbcam社(米国)から購入し、二次ヤギ抗マウスIgG−FITC抗体はSigma Aldrich社(米国)から購入した。
ヒト皮膚包皮線維芽細胞(HDF)は、ヘルシンキ宣言の原則に従って得られた組織生物学研究ユニット(チューリッヒ大学小児病院外科、スイス)から提供された。ヒト若年包皮サンプルは、ディスパーゼ(0.5mg/ml,Roche社製,スイス)を含み、5μg/mlのゲンタマイシンを含有するハンクス緩衝塩溶液(Ca2+及びMg2+を除いたHBSS,Invitrogen社製)において4°Cで一晩消化させた。これにより、その後ピンセットを用いて、表皮と真皮の分離を可能とした。一次皮膚線維芽細胞の培養を行うため、真皮は、37°Cにて1時間、コラゲナーゼIII(1mg/ml,Worthington Biochem社製,米国)を含むHBSS及びディスパーゼ(0.5mg/ml,Roche社製)用いて単細胞懸濁液において解離させた。最後に、容積比10%のウシ胎児血清、2mMのL−グルタミン、100U/mlのペニシリン及び100μgのストレプトマイシン(全てSigma Aldrich社から)を補充したRPMI−1640培地にて培養し、37°C及び5%のCO2を維持した。すべての実験において、5未満の継代による細胞がイン・ビトロ実験に使用された。
完全に創傷が被覆された後に基底支持体上に細胞により沈着されたフィブロネクチン線維を撮像するため、パッチは、除去され、培養物はその後のフィブロネクチン染色のために脱細胞化された。このため、培養物は、PBSで洗浄され、細胞膜は、体積率0.5%のトリトンX−100(Sigma Aldrich社製)の溶液と20mMのNH4OHをPBSに加えることで溶解した。完全に細胞の破片を溶解するため、試料は、その後、4°CのPBS中に一晩放置した。翌日、PBSを、吸引して、沈着したフィブロネクチンが次のように染色された。試料を、最初に1時間ブロッキング緩衝液(PBS中5%のBSA)内で処理し、その後一晩(4°Cで)一次抗体とともにインキュベートし、ブロッキング緩衝液で3回洗浄した後、試料を二次抗体とともにRTで1時間インキュベートした。最後に、サンプルを、PBSで5回洗浄して、直ちに撮像した。
細胞の撮像は、オルカR−2CCDカメラ(浜松ホトニクス社製,日本)を備えた倒立型のニコン−Ti広視野顕微鏡(ニコン社製,日本)を用いて行った。パッチ装着後、プレートはインキュベーションチャンバ内の顕微鏡下に置いた。インキュベーションチャンバは、温度、CO2濃度、及び湿度が、それぞれ、37°C、5%、及び95%に維持されていた。画像は、20倍、開口数0.45の長作動距離対物レンズ(Plan Fluor,ニコン社製)を用いて撮影した。9つの隣接する非重複フィールドが、それぞれサンプルとして平行に記録された。これは、1290×983μmの有効視野における創傷治療の並列タイムラプス撮像を可能とした。並列映像は、1時間の時間分解能と31時間以上の全継続時間で取得した。測定の各時間において、透過率及び蛍光画像は、それぞれ微分干渉コントラスト法(DIC)及びFITCフィルターセットを使用して取得した。実験中の焦点ドリフトは、顕微鏡のPFSオートフォーカスシステムを使用して回避した。
創傷治療の映像は、イメージJ(国立衛生研究所,米国)を使用して、次の手順で解析した。蛍光チャネルは、コントラストが増強され、白黒画像を提供するために閾値処理された。閾値処理された画像は、その後、ノイズ低減のため反転除去された。創傷境界は、イメージJの「トレーシング」ツールを使用して第一画像から自動的に抽出され、創傷治療動態を定量化するため保存された。タイムラプスのフレームごとに、元の創傷領域における細胞カバー率が測定され、このようにして各測定時における創傷カバー率(μm2)が提供された。
統計解析は、MATLAB(MathWorks社,米国)を用いて行った。パターン化されたパッチとブランクパッチの間における創傷治療、細胞移動、フィブロネクチン方向及び接着斑の数とサイズの違いは、マン・ホイットニー・ウィルコクソン順位和検定(α=0.05)を使用して調べた。細胞方向と細胞移動方向の比較は、カイ二乗独立性検定、α=0.05により行った。全ての定量的な測定は、平均値±標準誤差で表される。計数されたイベントの総数は、グラフの右上隅に表示される。明示的に表示されない場合、統計的検定の信頼区間は、p<0.05、p<0.01、及びp<0.001のように、それぞれ1つ、2つ及び3つのアスタリスクを用いてレポートされる。
線維芽細胞の創傷治療:
イン・ビトロでの細胞移動に対するPDMSパッチの効果をテストするため、新たに単離されたヒト皮膚線維芽細胞(HDF)を、ゼラチンコーティングされた基底支持体上でコンフルエントになるまで増殖させた。その後、ピペットの先端を用いて、創傷を単分子層に力学的に誘導し、図3に示すように、PDMSの活性ゼラチンコーティングされた表面を、培養物の頂端に適用した。図3は、実験のセットアップの説明図であり、(A)においてソフトリソグラフィによって生成されたPDMS活性表面又はパッチ、(B)においてゼラチンコーティング(緑色)を支持する親水性表面を得るためにプラズマ処理されたPDMSパッチ、(C)においてゼラチンコーティングされた基底支持体(ペトリ皿)上の培養細胞により得られた一次ヒト皮膚線維芽細胞のコンフルエント層(HDF)、(D)において力学的に傷付けられた単分子層、及び、(E)において創傷の上に適用されたパッチの活性表面を示す。
垂直格子(図4)の測定された効果が、ガイダンス機構に基づくものかどうかを評価するため、移動細胞の個別トラックを創傷治療映像から抽出した。垂直格子(図5A)又はブランクパッチ(図5B)のもとで得られた軌道の解析によって、地形が改良された表面に接触した細胞は、直線的な経路で長距離に亘って移動し、これにより、創傷領域に深く浸透することが明らかとなった。創傷治療の際における元の位置から最後の位置への平均細胞変位は、ブランクパッチのもとで移動する細胞と比べ、垂直格子のもとで移動する細胞は、21%高かった(図5C)。長い移動軌道は、速い移動(垂直パッチのもとでは平均移動速度が13%速かった)と改良された方向(垂直パッチのもとでは総変位に対する総移動距離の割合は、平均10%低かった)に起因する。創傷に対し60°〜90°の範囲内において、整列した軌道の割合が極めて増加する(13%)ことが示されているように、重要なことは、より良い軌道の方向によって、より早い創傷カバー率が得られたことである。
創傷領域内の線維芽細胞の移動により新たに沈着されたフィブロネクチン細胞の構造は、生体における創傷の解消や瘢痕に強く影響を及ぼす。垂直格子により誘導したガイダンスの効果が、HDFの移動によるECMの沈着に影響を与えた(図4−6)かどうかをテストするため、創傷領域内に沈着した繊維状のフィブロネクチンの広範な構造を、完全な治療後に可視化した(図7)。フィブロネクチン線維は、均一に分布して、格子の方向に優先的に整列したバスケットウィーブ組織を見せる垂直格子のもとで創傷内に浸透した線維芽細胞によって基底支持体(図3)上に沈着(又は再構築)する。著しく対照的に、ブランクパッチのもとで移動した細胞により沈着したマトリックスは、あまり組織化されておらず、単分子層の非創傷領域で見られるもの(図7C)と同様の様々なフィブロネクチン密度の領域を示した(図7B)。広範なマトリックス配列のフーリエ解析により、ブランクパッチのもとで沈着した線維の整列が著しく悪化(59.8°±6.1°)している間に、線維芽細胞が、創傷に対しフィブロネクチン細胞が整列した垂直格子のもとで移動することが確認された(図7D)。次に、創傷治療において沈着するフィブロネクチンの均一性を定量化した。垂直パッチのもとでの創傷治療におけるピクセル強度の極めて低い標準偏差により、マトリックスが、ブランクパッチの場合又はコントロールされた非創傷領域の場合よりも、均一であることが明らかになった(図7E)。
垂直格子(図4〜6)の頂端適用により誘導されるガイダンスの効果は、地形的特徴と接着斑の間の相互作用を必要とすることを検証した。線維芽細胞により生成する接着斑のサイズと位置は、ビンキュリン−FP635の一時的な発現により明らかになった(図8A)。実験条件下で、ビンキュリンの過剰発現は、HDFの移動と極性に影響を与えなかった。点状蛍光信号からわかるように(図8A)、線維芽細胞により基底支持体を有する境界において接着斑が生じた。重要なことは、両方の実験条件下で、最小数の細胞のみしか、光学的に分析可能な接着斑が頂端パッチを有する境界において生じなかったことである(図8B)。実際、基底支持体を有するHDFにより生じた接着斑の平均数は、垂直格子のもとで移動する細胞の場合101±12、ブランクパッチのもとで移動する細胞の場合56±12であり、両ケースにおいて、頂端パッチを用いて生じた接着斑の平均数は、2倍少なかった。この結果は、基底支持体を伴う場合の生物学的相互作用が、頂端パッチを伴う場合よりも極めて強かったことを示している。この仮説を確認するため、パッチは、完全な創傷治療の後に除去した。図11は、完全な創傷治療後のパッチ除去を示し、(A)においてパッチ適用前の傷ついた単分子層が示され、(B)においてパッチ適用後の傷付いた単分子層が示されている。(C)においてパッチ除去前の治療された単分子層が示され、(D)においてパッチ除去後の治療された単分子層が示されている。(E)はパッチ除去前の治療領域のDIC画像であり、(F)はパッチ除去後の治療領域のDIC画像である。すべてのケースにおいて、パッチの除去は、治療された単分子層(図11E及び図11F)を損傷することなく又は細胞をはぎ取ることなく達成することができる。図10は、完全な創傷治療後のパッチの除去を示す。(A)においてパッチ適用前の傷付いた単分子層が示され、(B)においてパッチ適用後の傷付いた単分子層が示されている。(C)においてパッチ除去前の治療された単分子層が示され、(D)においてパッチ除去後の治療された単分子層が示されている。(E)はパッチ除去前の治療領域のDIC画像であり、(F)はパッチ除去後の治療領域のDIC画像である。

Claims (14)
- 損傷細胞層の改良された治療のための活性表面要素(2)であって、
少なくとも1つの地形的に構造化された表面を、パターン周期(p)でパターン長(l)に沿って延びる隆起(5)及び溝(6)を交互に備えたパターンを有する基板上に備え、
前記パターン周期(p)は10μmより小さく、
前記パターン長(l)は1mmより大きく、及び/又は、前記パターン長(l)は前記構造の前記周期(p)の20倍分より大きい
ことを特徴とする表面要素。 - 前記隆起(5)の幅及び/又は前記溝(6)が、1〜9μの範囲内にあり、好ましくは、前記隆起(5)の幅が1〜5μの範囲内にあり、前記溝(6)の幅が1〜5μの範囲内にあり、好ましくは、両方の幅が基本的に等しい
