ガラクトース媒介(GAM(−))rhTSHのPEG化
カタラーゼ/ガラクトースオキシダーゼ処理:1mgあたり2Uのカタラーゼ(Sigma 442U/μl)および1mgあたり4μgのガラクトースオキシダーゼ(Worthington GAO、1.2mg/ml)をTSHに添加した。次に、混合物は、37℃にて16時間インキュベートした。インキュベーション後、混合物を緩衝液交換し、PEG化を行うために、少なくとも5.5mg/mlの最終濃度に、100mM酢酸ナトリウム(pH5.6)中に濃縮した。
rhTSHのN末端PEG化

PBS(リン酸緩衝生理食塩水)緩衝液(pH7.2)中で、様々な(PEG:タンパク質)比(例えば、(0.5:1)、(1:1)、(2:1)、(4:1)、(8:1))で、最終濃度が0.8mg/mlであるrhTSHに適切なサイズのNHS(N−ヒドロキシスクシンイミド)PEG(dH2O中の50mg/ml)を添加した。インキュベーションは、25℃にて1.5時間行った。
部位特異的コンジュゲーションのためのシステイン突然変異体の生成
TSH単一突然変異体を設計および調製し、部位特異的PEG化のためにシステインを導入した。これらの突然変異体は、タンパク質の折り畳み、受容体結合における効果を最小限にするように、また効果的にコンジュゲートされるそれらの可能性について設計された。
PEG化されたrhTSHのmonoS AKTA精製
monoSカラム(GE Healthcare)上で試料を精製し、10mM酢酸ナトリウム(pH5)および10mMの酢酸ナトリウム、1M塩化ナトリウム(pH5)による勾配を用いて、流速4ml/分にて溶出させた。勾配は、0%移動相B(10mM酢酸ナトリウム、1M塩化ナトリウム(pH5))で開始し、次に、25カラム体積にわたって50%移動相Bに増大させ、続いて、100%移動相Bにしてカラムを洗浄した。
SDS−PAGE分析および染色
予め注いだ勾配ゲル(4〜12%Bis Tris、Invitrogen)に4〜5μgのTSHを装填した。MOPS(3−(N−モルホリノ)プロパンスルホン酸)泳動緩衝液(Invitrogen)を調製した。電気泳動装置を、氷を入れた氷のバケツに設置した。ゲルは、約50分間、200Vで泳動され、次に、蒸留水で3回、それぞれ5分間濯がれた。50mlの5%塩化バリウムをゲルに添加し、次に、10分間振とうした。塩化バリウムは、蒸留水で5分間、ゲルを濯ぐことによって除去された。次に、蒸留水を除去し、バンドが視認できるまで、最初に、1×ヨウ化カリウム/ヨウ素溶液でPEGについてゲルを染色した。次に、ゲルを蒸留水で脱色し、すぐにスキャンした。タンパク質染色について、クマシー脱色(10%酢酸、20%メタノール)を用いて、PEG染色の全ての痕跡を除き、次に、ゲルをクマシーブルー染色で染色した。精製された糖質PEG化されたrhTSHコンジュゲートのSDS−PAGE分析結果を図10C(PEG染色)および図10D(クマシー染色)に示す。
PEG化されたrhTSHのSEC−HPLC分析
試料をSuperdex 200 10/300GLカラム(GE Healthcare)上で泳動し、50mMリン酸ナトリウム、150mM塩化ナトリウム緩衝液(pH7)中で、流速0.4ml/分にて溶出させた。40kDのSAM反応物と精製されたPEG化されたrhTSHコンジュゲートの典型的なSEC−HPLCプロファイルをそれぞれ図11Aおよび図11Bに示す。
PEG化されたrhTSHのペプチドマッピングとN末端配列決定
20μgの試料それぞれは、窒素で重層された、6M塩酸グアニジンと38mMジチオスレイトールを含有する0.1M Tris(pH8.5)中に希釈され、25℃にて一晩インキュベートされた。インキュベーション後、ヨードアセトアミドを50mMで添加した。次に、窒素を試料に重層し、25℃にて2時間インキュベートした。アルキル化反応は、1/1(v/v)の0.25%トリフルオロ酢酸を添加することによってクエンチされ、0.1M Tris(pH8.5)中に、3,500MWCO Slide−A−Lyzerミニ透析ユニット(Thermo Scientific)を用いて試料を透析した。試料は、1:25(酵素:試料)の比で37℃にて一晩消化された。消化反応物は、1/1(v/v)の0.25%トリフルオロ酢酸を用いてクエンチされた。トリプシン消化された試料は、自動化されたインジェクターと画分コレクター、バイナリー溶媒ポンプ、サーモスタット付きカラムコンパートメント、および可変波長検出器を備えたAgilent 1200 HPLCを用いて分画された。試料は、50℃に保持されたPoroshell 300SB−C8カラム(2.1×75mm、5μm粒子、Agilent Technologies、CA)に装填され、98%溶媒A(水中の0.1%トリフルオロ酢酸)および2%溶媒B(アセトニトリル中の0.08%トリフルオロ酢酸)中で予め平衡化された。トリプシンペプチドは、流速0.4ml/分にて、25分間かけて、2%から60%の溶媒Bの直線勾配を用いて溶出させた。PEG化された断片は、PEG化されていない断片よりも大きな%Bで溶出し、回収され、遠心濃縮器(Thermo Scientific、MA)中で乾燥させた。自動化されたN末端配列分析は、Prociseプロテインシーケンサー(Applied Biosystems、CA)を用いて行われた。200pmolの試料と対照は、予めプログラムされたパルス液体法を用いて、自動化された18回のEdman分解に供された。図12は、40kDのSAMコンジュゲートのペプチドマップとN末端配列決定の結果を示す。たった3つのトリプシン糖ペプチド(AT9、AT6、BT3)が検出され、糖質PEG化の部位特異性を示した。下線のNは、N連結型グリコシル化部位に対応する。
それぞれのサブユニットにおけるPEG化の相対量の決定
サブユニット特異的なPEG化は、2つの連続した逆相HPLC稼働を用いて、PEG化されたサブユニットからPEG化されていないαおよびβサブユニットを単離した後、それらの相対量を測定することによって計算された。PEG化されたαおよびβサブユニットを分離するクロマトグラフィー条件を同定することができなかったため、この推定法を用いた。遠心限外ろ過によって試料(100μg)を20μlに濃縮し、次に、6M塩酸グアニジン、10mMリン酸ナトリウム、100mM塩化ナトリウム(pH7.0)中で変性させた。25℃にて一晩インキュベートした後、75%溶媒A(10mMリン酸ナトリウム、pH6.5)および25%溶媒B(40%の10mMリン酸ナトリウム、pH6.5、60%アセトニトリル)を用いて予め平衡化されたPoroshell 300SB−C8カラム(2.1×75mm、5μm粒子、Agilent Technologies、CA)に試料を装填した。5分間の25〜50%B、続く20分間の50〜75%Bを用いて、0.4ml/分、25℃にてカラムを溶出した。PEG化されていないTSHサブユニットに対応するピーク画分(約9.5〜12.5分)は、その全体で回収され(図13B)、遠心限外ろ過によって50μl未満に濃縮された。水を用いて試料を50μlに調整し、次に、4.7μlの2Mジチオスレイトールおよび150μlの6M塩酸グアニジン、0.1M Tris(pH8.5)の添加によって還元し、窒素で重層し、25℃にて一晩インキュベートした。次に、遊離したチオールは、9.3μlの2Mヨードアセトアミドを添加し、窒素を重層し、2時間25℃にてインキュベートすることによってアルキル化された。アルキル化反応は、150μlの0.25%トリフルオロ酢酸を添加することによってクエンチされた。還元およびアルキル化されたPEG化されていないTSHサブユニットは、第2の逆相HPLC稼働によってプロファイリングされた。HPLCカラムのセットアップは、水中の0.1%トリフルオロ酢酸からなる溶媒Aとアセトニトリル中の0.08%トリフルオロ酢酸からなる溶媒Bおよびカラムが50℃に保持されることを除いて、上記に指示されたものと同一であった。カラムは、0.3ml/分で15分間、2〜75%Bの直線勾配により溶出された。PEG化されていないα対βサブユニットの相対的割合は、試料あたり、三重の注入からの得られたA214nmのクロマトグラム(図13C)の積分により決定された。次に、PEG化されたα対βサブユニットの相対的割合は、これらの値の逆数とした。この分析は、SDS−PAGE(図13A)によって得られた結果と一致して、40kDのモノPEG化されたGAM−、GAM+およびSAMコンジュゲートにおいて修飾されたαサブユニットの画分がそれぞれ77%、66%および58%であることを示した。
インビトロ受容体結合アッセイ
精製された、PEG化されたコンジュゲートは、RSR Limited(Kronus、Star、ID)のTSH受容体自己抗体の第2世代ELISAキットを用いたインビトロでのブタTSH受容体結合アッセイによって分析された。キットが提供するTSH受容体に対するビオチン化ヒトモノクローナル自己抗体を用いる代わり、本発明者らは、TSH受容体への競合結合阻害に使用するために、Thyrogen(登録商標)(rhTSH)をビオチン化した。固定化されたブタTSH受容体へのビオチン化rhTSHの結合は、rhTSH対照またはPEG化されたrhTSHコンジュゲートのいずれかによって阻害され、IC50値を測定した。

rhTSHおよびPEG化されたrhTSH(20kDのSAM、20kDのGAM(−)、20kDのGAM(+)、40kDのGAM(+))の薬物動態について、単回筋内(IM)注射後の雄性および雌性ラットにおいて評価した。
単回IM注射後の雄性と雌性Sprague DawleyラットにおけるPEG化されたrhTSHの薬物動態分析
単回筋内(IM)注射後の雄性と雌性ラットにおいて、PEG化されたrhTSH(10kDのマルチSAM、10kDのSAM、40kDのSAM、40kDのGAM(−))の薬物動態を評価した。
薬物動態アッセイ:rhTSHまたはPEG化されたrhTSHのELISAによる血清試料中のタンパク質濃度の測定
高結合の96ウェルELISAプレートは、0.1M炭酸水素ナトリウム緩衝液(pH9.2)中に希釈され、ウェルあたり100μLで添加された1.33μg/mLのマウス抗hCG捕捉抗体で被覆された。標準のrhTSHまたはPEG化されたrhTSH曲線は、精製されたタンパク質から調製され、1×プレート洗浄液中の1.0%w/vのBSAからなる試料希釈緩衝液(SDB)において希釈された。標準は、アッセイごとにrhTSHまたはPEG化されたrhTSH種の限定された線形範囲に依存して、2:3の連続希釈スキームを用いて、25〜1.463ng/mL、8.334〜0.488ng/mLまたは5.556〜0.325ng/mLから希釈された。1:10希釈を下回らないように、試料をSDBに希釈した。正常なラット血清中に調製された500ng/mLと25ng/mLのrhTSH対照は、SDB中にそれぞれ1:100と1:10に希釈される。SDB中に調製された0.5ng/mLのrhTSH対照は、希釈せずに添加された。被覆されたプレートを洗浄し、100μLの試料、標準および対照をプレートに添加し、1時間37℃にてインキュベートした。
PEG化されたrhTSH薬力学のマウスT4バイオアッセイ評価
PEG化されたrhTSHの薬力学は、T3ペレット移植の3日後、単回IP注射後の雄性マウスにおいて評価された。このマウスPDモデルにおいて、内因性マウスT4は、投薬の3日前に、徐放T3ペレットの移植によって研究期間中に抑制された(図18A〜図18Dにおけるビヒクル群を参照されたい)。従って、rhTSH対照またはPEG化されたrhTSHコンジュゲートによって放出されたT4量だけを測定した。マウスの限定された血液体積のため、投薬の6、24、48および72時間後の4つの時間点で回収した。
PEG化されたrhTSH薬力学の用量応答曲線の研究評価(40kDのSAM)
PEG化されたrhTSH(40kDのSAM)の薬力学は、T3ペレット移植の3日後、単回腹腔内(IP)注射後の雄性マウスにおいて、3つの異なる投薬レベルで評価された。

Claims (52)
- 少なくとも1つのポリアルキレングリコールポリマーは甲状腺刺激ホルモン(TSH)の糖質部位に結合している、TSHを含む組成物。
- TSHは哺乳動物から単離されている、請求項1に記載の組成物。
- 哺乳動物はヒトである、請求項2に記載の組成物。
- TSHは組換え哺乳動物TSHである、請求項1に記載の組成物。
- TSHは組換えヒトTSH(rhTSH)である、請求項4に記載の組成物。
- 糖質部位はシアル酸である、請求項5に記載の組成物。
- シアル酸基は、rhTSHαサブユニットのASN52、rhTSHαサブユニットのASN78、およびrhTSHβサブユニットのASN23またはこれらの組合せからなる群から選択されるTSH上の部位に位置している、請求項6に記載の組成物。
- 糖質部位はガラクトースである、請求項5に記載の組成物。
- ガラクトースは、rhTSHαサブユニットのASN52、rhTSHαサブユニットのASN78、rhTSHβサブユニットのASN23またはこれらの組合せに位置している、請求項8に記載の組成物。
