Description
本発明は、一般に、癌の治療に関し、より具体的には、放射線または標準的な化学療法剤を用いない、固形腫瘍およびその転移を含む腫瘍の治療に関する。好ましい実施形態では、本発明は、癌ワクチンの使用による癌の予防および治療に関する。
現代の癌療法は、ほとんどが放射線、手術および化学療法剤の使用を伴う。しかし、これらの手段による結果は、一部の腫瘍では有益であるものの、多くの他の腫瘍ではわずかな効果しか得られないかまたは効果がない。さらに、これらのアプローチは、容認できない毒性を有する場合が多い。
本発明は、一般に、癌の予防および/または治療に関し、より具体的には、放射線または標準的化学療法剤を用いない、固形腫瘍およびその転移を含む腫瘍の治療に関する。一実施形態では、本発明は、a)腫瘍細胞を有する被験体を提供する工程と、b)前記被験体から前記腫瘍細胞の少なくとも一部を除去して除去細胞を作製する工程と、c)処理された腫瘍細胞を作製するため、タプシガルギンおよび/またはタプシガルギン関連化合物を含む刺激剤を用いて、前記除去細胞の少なくとも一部をエキソビボで処理する工程と、d)前記処理された腫瘍細胞を同じ被験体にインビボで導入して抗癌治療効果を生じさせる工程とを含む方法を含む。被験体にキャリーオーバーされる薬剤量を最少化するため、前記処理された腫瘍細胞を導入する前に、これらは(例えば緩衝液、培地などで)洗浄することができる。好ましい実施形態では、本発明は、a)腫瘍細胞を有する患者を提供する工程と、b)前記患者から前記腫瘍細胞の少なくとも一部を除去して除去細胞を作製する工程と、c)処理された腫瘍細胞を作製するため、タプシガルギンおよびタプシガルギン関連化合物から成る群から選択される薬剤に、前記除去細胞の少なくとも一部をエキソビボで曝露する工程と、d)前記処理された腫瘍細胞を溶解して細胞断片を作製する工程と、e)前記断片を同じ患者にインビボで導入して抗癌治療効果を生じさせる工程とを含む方法を含む。一実施形態では、工程(d)の前に、次の工程へキャリーオーバーされる薬剤量を最少化するため、処理された細胞を洗浄することが好ましい。本発明は、工程(c)の任意の特定の曝露/処理時間により限定されるものではない。1秒から72時間の間、より好ましくは30秒から1時間の間、さらにより好ましくは1分から30分の間の曝露時間を含む、様々な曝露時間が意図される。本発明は、工程(d)において溶解の性質により限定されるものではない。一実施形態では、処理された腫瘍細胞は、細胞を凍結/解凍して溶解される。別の実施形態では、溶解剤を使用する。溶解剤は、界面活性剤(例えばドデシル硫酸ナトリウム)、酵素(例えばQiagen Buffer B1 1ml、リゾチーム20μl、プロテアーゼ45μlおよびQiagen Buffer B2 0.35mlから成る酵素消化緩衝液)、または単純な溶液(例えばリン酸緩衝生理食塩水)もしくは例えば、低張溶解緩衝液(5mM リン酸ナトリウム pH7.4、プロテアーゼ阻害剤カクテル、5mM DTT)で、細胞を浸透圧で溶解するためのより複雑な溶液であり得る。別の実施形態では、細胞は超音波処理される。別の実施形態では、細胞抽出物が作製される(例えば膜抽出物、細胞質抽出物など)。別の実施形態では、これらの方法の組み合わせが使用される(例えば低張溶解、それに続くホモジナイザー、例えばポリトロンPT 3000、Kinematica社、ルツェルン、スイスでの処理)。本発明は、細胞断片または細胞成分の導入方法に限定されるものではなく、これらは静脈内(IV)、筋肉内(IM)、腹腔内(IP)、皮下(SC)、皮内(ID)などで投与することができる。特定の癌には特定の方法で断片/抽出物を投与することが好ましい場合がある。例えば、皮膚癌の場合、メラノーマなどの癌を治療するため皮膚を介して(例えば皮下、経皮などで)断片/抽出物を投与することが(一実施形態では)好ましい場合がある。前立腺癌の場合、断片/抽出物を腹腔内投与することが(一実施形態では)好ましい場合がある。一方、幾つかの実施形態では、断片/抽出物は、腫瘍から離れた部位、および腫瘍の組織型(例えば乳癌、肺癌など)に関係のない組織(例えば筋肉、皮膚など)に投与する。
さらに他の実施形態では、患者は、顆粒球−マクロファージコロニー刺激因子(GM−CSF、一般名サルグラモスチム(sargramostim)、商品名ロイキン(Leukine))および顆粒球コロニー刺激因子(G−CSF、一般名フィルグラスチム、商品名ノイポジン)など、骨髄における白血球細胞の産生を刺激する1つまたは複数の薬剤と共に前、後または同時に治療され得る。好ましい発現構築物に関して、一実施形態では、本発明は、長期持続効果を生じさせるため断片/抽出物の前、後、または一緒にCD40L発現プラスミドを導入することを意図する。
腫瘍細胞が患者から除去される幾つかの実施形態では、a)原発腫瘍(またはその部分)を除去するための標準的な手術を使用することが意図される。b)処理された腫瘍細胞を作製するため、タプシガルギンおよび/またはタプシガルギン関連化合物を含む刺激剤を用いて、エキソビボで腫瘍細胞を処理し、c)前記処理された腫瘍細胞由来の腫瘍細胞断片/抽出物を調製し、d)前記断片/抽出物を同じ患者にインビボで導入した後、抗転移効果が生じることが意図される。このように、本発明の実施形態は、従来の外科的処置と併用することができる。例えば、一実施形態では、乳房の原発腫瘍は、従来の手術(すなわち部分または全乳房切除術)により除去することができ、断片/抽出物は、転移治療のため術後に患者に投与することができる。幾つかの実施形態では、患者が転移を有することがわかっているか否かにかかわらず(または転移は検出されたが転移性疾患の全範囲はわからない場合に)、治療が行われる。言い換えれば、本発明の保護療法は、従来の手術後、ならびに既知の転移性疾患の救急治療後に患者に適用することができる。
本明細書では、「被験体」はヒトまたは動物であり得る。患者は、病院であれ、通院患者としてであれ、診療所であれ、または医院であれ、医療を受けているヒトである。「癌のリスクのある被験体」および「癌のリスクのある患者」は、様々な理由で(年齢、遺伝的素因、放射線曝露、煙への曝露、粒子の吸入、変異誘発物質の消費、HIV感染などのためであれ何であれ)リスクのある可能性がある。例えば、ある人は、彼/彼女が癌の罹患歴を有する家族の一員であるという理由でリスクのある可能性がある。一方、ある人は、遺伝子スクリーニングアッセイの結果によりリスクのある可能性がある。後者に関して、突然変異の検出は、癌および/または癌になる傾向がある個人(すなわちリスクのある)の診断を含むがこれに限定されず、臨床診断においてますます重要な分野となっている。タンパク質トランケーションテスト(PTT)は、ナンセンス、および切断タンパク質産物の生成をもたらすフレームシフト突然変異を検出する方法である。ヒトmutLホモログおよびヒトnutSホモログ(いずれも結腸癌に関与)、ならびにBRAC1(家族性乳癌に関与)などの癌関連遺伝子は、他の方法と並んで、現在この方法で変異をスクリーニングすることができる。こうした切断変異(および他の種類の変異)を有することがわかっている者が、癌のリスクがあると認識される。
タプシガルギンは、セスキテルペンラクトン(以下の構造1参照)またはセスキテルペンラクトンテトラエステルと呼ばれることが多い。タプシガルギン誘導体は、様々な戦略で作製された。例えば、デアセチルタプシガルギン(deacetylthapsigargin)を得るためO−10のアセチル基の加水分解が行われた(以下の構造2参照)。
ヒドロキシル基の1つのアセチル化およびカルボニルラクトンのメチレン基への還元も行われた。本明細書では、「タプシガルギン関連化合物」には、タプシガルギン誘導体、類似体および類似の活性を有する化合物が含まれる。故に、天然の類似体(上記表2参照)および合成類似体(下記参照のこと)は、本発明の範囲内にあるものである。実際、一般にはセスキテルペノイドおよび具体的にはグアイアノリドは、本発明の範囲内にあるものである。幾つかのタプシガルギン類似体は、タプシガルギシンを還元して得られるラクトールのO−11およびO−12をアルキル化またはアシル化して合成された。α配置置換基の導入は、類似体のCa(2+)−ATPase阻害能力を減少させたのに対し、酵素はβ配置置換基に対しより寛容であった。これは、ラクトン環のα面(alpha−face)が、阻害剤が酵素に結合した場合、結合部位と密接に接触していることを示している。合成的に作製された幾つかの類似体は、強力な活性を有することを示した(以下の化合物77など)。他の類似体にはL12ADTが含まれる。
誘導体に関して、本発明は、8−O−デブタノイルタプシガルギン、8−O−(4−アミノシンナモイル)−8−O−デブタノイルタプシガルギン(ACTAと呼ばれる)を含むがこれらに限定されない多様性を意図する。タプシガルギンC8誘導体(ZTG)は、4−アジド[カルボキシル−14C]安息香酸でデブタノイルタプシガルギンをアシル化することにより合成された。
以下は、本発明により意図される特定の実例および実施形態を示し、限定されるものではない。
基本的実験手順
この例は、以下の実施例II〜VIIで使用される基本的な材料および方法を提供する。これらの特定の実験は、特定の化合物および方法を利用しているが、他の類似の化合物および方法も、類似のデータをもたらすことができる。
ヒト子宮頸上皮細胞株HeLaおよびマウスB16−F10メラノーマは、アメリカンTCCから購入し、10%ウシ胎仔血清(Sigma−Aldrich社、セントルイス、MO)を補充したダルベッコ改変イーグル培地[DMEM(Cellgro(登録商標)、ハーンドン VA)]で維持した。以下に記載された全ての細胞ストレス/死関連処理は、初回スクリーニングにおいてHeLa細胞を用いて行った。ヒト前単球白血病THP−1(ATCC、マナッサス VA)および前マクロファージMonoMac6細胞を、この試験で利用された細胞を提示するレポーター抗原として利用した。Ziegler−Heitbrockら、「Establishment of a human cell line(Mono Mac 6)with characteristics of mature monocytes」Int.J.Cancer 41:456〜461頁(1988年)。両方の系は、10%ウシ胎仔血清を補充したRPMI−1640培地(Cellgro(登録商標))で維持した。特に注記のない限り、細胞は全て5% CO2を補充した加湿インキュベーターで37℃にて維持した。
以下のリストには、この試験で評価される小分子化合物が含まれる:タプシガルギン、タプシガルギシン(MP Biomedicals社、アーバイン CA)、1,2−ビス(o−アミノフェノキシ)エタン−N,N,N’,N’−四酢酸テトラ(アセトキシメチル)エステル[BAPTA−AM、(EMD Biosciences社、サンディエゴ、CA)]、エチレングリコール−ビス(β−アミノエチル)−N,N,N’,N’−テトラアセトキシメチルエステル[EGTA−AM(EMD Biosciences社)]、1,2−ビス(2−アミノフェノキシ)エタン−N,N,N’,N’−四酢酸(EGTA)、テトランドリン、KCl、アミロイドペプチドAβ1−42、1−オレオイル−2−アセチル−sn−グリセロール(OAG)、カルシマイシンA23187、ジフェニルボリン酸2−アミノエチル(2−APB)、ヒスタミン、シクロピアゾン酸、2,5−ジ−(t−ブチル)−1,4−ヒドロキノン[BHQ(EMD Biosiences社)]、GdCl3、塩酸ニカルジピン、1−(o−クロロ−α,αジフェニルベンジル)イミダゾール(クロトリマゾール)、アパミン、カリブドトキシン、ラジシコール、ブレフェルジンA、ツニカマイシン、タモキシフェン、イオノマイシン、スタウロスポリン、シス−ジアミン白金(II)ジクロリド(シスプラチン)、パクリタキセル、メトトレキサート、シクロヘキシミド、およびラパマイシン。特に注記のない限り、化合物は全てSigma−Aldrich社から購入した。化合物を、公開された文献に基づく一連の濃度でHeLa細胞に添加する前にメーカーの推奨により溶解し、1分間から2日間インキュベートした。炎症pam3cys−ser−(lys)4,ヒドロクロリド(Pam3Cys)、細菌フラジェリン、およびリポ多糖の既知のメディエーターは、EMD Biosiences社から購入した。
評価する細胞死誘導トランス遺伝子には、ヒトカスパーゼ8、カスパーゼ9、カスパーゼ3、ヒトインスリン増殖因子−1受容体細胞内ドメイン(IGF−1R IC)、およびニューロキニン−1受容体(NK1R)が含まれ、全てpcDNA3.1ベクター(Invitrogen社)にクローニングした。マウスCD40L(またはCD154)発現プラスミドは、NCBIに公開されているマウスCD40Lのヌクレオチド配列(図12)を使用し、XbaI制限部位が隣接する配列をコードするオリゴヌクレオチド(図13)を重複させてCD40L相補的DNAを作製した後、Pfu DNAポリメラーゼ(Stratagene社、サンディエゴ CA)を使用してポリメラーゼ連鎖反応を行った。得られたDNAを、XbaI(New England Biolabs社、イプスウィッチ MA)で消化し、同様に消化したpVAX1発現ベクター(Invitorogen社、カールスバッド CA)に連結した(図14)。pVAX−CD40L構築物はDNA配列決定により確認した。
トランスフェクションは全て、メーカーの指示によりHeLa細胞およびリポフェクタミン2000(登録商標)トランスフェクション試薬(Invitorogen社)を用いて行った。NK1R−トランスフェクト細胞を、1μM サブスタンスPペプチド(Sigma−Aldrich社)と共にインキュベートしてトランスフェクションの翌日に死を誘導した。
血清飢餓は、1〜24時間の間、血清を含まないDMEMでHeLa細胞をインキュベートした後に行った。熱ショックは、1〜6時間の間、42〜45℃で細胞をインキュベートして誘導した。
細胞ストレス/死の後、得られた細胞/細胞残屑を回収し、過剰量のリン酸緩衝生理食塩水で3回洗浄し、凍結/解凍(−80℃から25℃)を3回素早く繰り返し、UV分光光度法(A280)により定量した。ストレス/死細胞内の炎症誘発性シグナルの存在を判定するため、40μgのストレス/死細胞を50,000個のTHP−1に添加し、18時間インキュベートした。腫瘍壊死因子−α(TNF−α)、インターロイキン8(IL−8)、およびIL−12を含む炎症誘発性サイトカインの存在について、上清を市販のELISAシステムにより評価した。
B16−F10細胞を、タプシガルギン(2.5μM)の添加前に、約80%コンフルエントになるまで150mm組織培養プレートで培養した。細胞を、回収前に48時間インキュベートし、過剰量のリン酸緩衝生理食塩水で3回洗浄し、凍結−解凍サイクルを行い、上述の通りに定量した。未処理細胞を同様に回収し、両方の抽出物を、インビボでのさらなる使用の前に上述のTHP−1システムにより炎症活性についてインビトロで評価した。
記載された以下のマウス試験は全て、Buck Instituteの所内動物実験委員会(Institutional Animal Care and Use Committee)(IACUC)により開始前に承認された。6〜8週齢のメスC57Bl/6およびnu/nuマウスは、Charles River Laboratories社から購入した(ウィルミントン MA)。免疫感作は、皮下、腹腔内、および皮内経路を含む様々な免疫感作経路により、500μgのB16−F10抽出物(タプシガルギン処理の有無にかかわらず)を2週間間隔で3回注射して行った。生腫瘍の曝露は、最後の追加免疫から2週間後、対側部位に100,000個の生B16−F10細胞を皮下注射して行った。
タプシガルギン感受性内因性危険シグナル(danger signal)の同定
薬剤を誘導する様々の異なる、可能性のある細胞ストレスおよび細胞死をスクリーニングした。植物由来タプシガルギン毒素は、患者由来子宮頸癌細胞株(HeLa)においてTNF−α分泌を誘導し、故に上述の様々な抗癌実施形態で使用するため同定された。反対に、他の可能性のある細胞ストレッサー(すなわち、例えば、タキソール、ツニカマイシン、ブレフェルジンA、シクロヘキシミド、およびカルシマイシン)をテストしたが、免疫賦活活性は誘導しなかった(図1参照のこと)。
サイトカインのタプシガルギン誘導分泌
この例は、癌細胞のタプシガルギン処理が、サイトカインを放出するように活性化される抗原提示細胞(単球、マクロファージ、樹状細胞など)に対し、この細胞を免疫賦活性にすることを例証する。発明の機構を理解する必要はないが、IL−12は、細胞性免疫応答(癌および細胞内病原体による感染の制御に必要となり得る免疫応答の種類)の発生に役割を果たしていると考えられる。
タプシガルギン処理したB16抽出物によるインビボ免疫感作
この例は、タプシガルギン処理細胞による免疫感作が、癌の進行に対する防御をもたらすことを示す。
B16+TG注射により誘導された免疫防御記憶(immunoprtective memory)
この例は、B16+TG(すなわちこの後、細胞断片溶液を作製するため洗浄され、定量化され、ならびに凍結および解凍により溶解された、TG処理したB16細胞)による免疫感作は、注射したメラノーマに対する長期防御をもたらす一実施形態を提供する。
これらのデータは、TG処理したB16抽出物で免疫感作したマウスは、B16メラノーマに対する長期持続免疫を発生できることを示す。この実験では、この免疫程度は、投与経路にある程度依存するように思われる。例えば、腹腔内または皮下的に与えられた免疫感作は、0日目または116日目ではB16メラノーマ細胞注射後の腫瘍増殖を完全には抑制しなかった。(図7参照)。しかし、条件を最適化していなかった。
B16−TG抽出物により誘導される単球分化
この例は、タプシガルギン処理細胞が、抗原提示細胞の他のサブタイプへの単球分化を誘発することができるかどうかについて例示的データを示す。過去の研究は、特定の薬剤が、抗原提示細胞上で作用する結果、成熟樹状細胞への分化をもたらすことを示している。Hertzら、「Microbial lipopeptides stimulate dendritic cell maturation via Toll−like receptor 2」J.Immunol.166:2444〜2450頁(2001年)。発明の機構を理解する必要はないが、樹状細胞は、生体における任意の抗原について適応免疫応答を活性化するか免疫寛容を活性化するかの決定に関与する主な細胞型であると考えられる。
TGリポソーム製剤により誘導された腫瘍減少
この例は、タプシガルギン含有リポソームが、用量依存的な様式で腫瘍の進行を減少させることを示すデータを示す。
毒素誘導NF−κBプロモーター系
この例は、毒素感受性ハイスループットレポーター(すなわちマーカーまたは標識)系の一実施形態を示す。
(TLR2を介してシグナル伝達する)TLRリガンドPam−3−Cysおよび(TLR5を介してシグナル伝達する)細菌フラジェリン((TLR4を介してシグナル伝達する)細菌リポ多糖ではない)のTHP−1由来クローンA9認識を、TGインキュベーション中に96ウェルホワイトプレート自動化アッセイでNF−κB活性をモニターして判定した。(図10A参照)
様々な濃度のタプシガルギンも、予め播種したHeLa細胞の存在下または非存在下で使用した。これらの結果は、クローンA9が、未処理HeLa細胞またはタプシガルギン単独よりも相乗的な高レベルでタプシガルギン処理したHeLa細胞を認識したことを示した。(図10B参照)。
第2の安定なNF−κBレポーター構築物形質転換体を、MonoMac−6細胞を用いて作製した。クローンA9とは対照的に、MonoMac−6由来クローンC5は、細菌リポ多糖の認識を改善したことを示した。クローンC5もまた、Pam3Cys、フラジェリン、およびタプシガルギン処理したHeLa細胞を認識した。(図11参照)。
タプシガルギン誘導脱毛
この例は、タプシガルギン(TG)が、脱毛を誘導する局所用溶液で使用される一実施形態を示す。様々な濃度(対照0、ならびにテスト試料40uM、1.2mMおよび40mM)でTGを含有する擦り込み溶液(rub−on solution)(75%エタノール、22%シクロヘキサン、および3%ジメチルスルホキシド)を調製した。様々な溶液を、異なるマウスに局所適用した。高濃度では、インサイチュで誘導炎症を素早く見ることができた(データ不図示)。高濃度での3日目の炎症は、この濃度が不適切であることが明らかになるほど多くの炎症を誘導した。低濃度では、擦り込み部位で一過性の脱毛症/抜け毛を誘導した(データ不図示)。脱毛は、数週間、および数カ月間も続き、4.5カ月(18週)でも依然として脱毛を見ることができた。何匹かの動物では、TG治療した動物(1回治療)でひとたび完全に脱毛すると、治療部位は、18週時点で若干毛が再生しはじめていた。故に、この低用量(uM)のTGは、脱毛を誘導するため、または化粧品および他の医学的適用のための他の脱毛製品で使用するのに(例えばローションで)使用することができる。
Claims (24)
- a)腫瘍細胞を有する患者を提供する工程と、b)前記患者から前記腫瘍細胞の少なくとも一部を除去して除去細胞を作製する工程と、b)処理された腫瘍細胞を作製するため、タプシガルギンおよびタプシガルギン関連化合物から成る群から選択される薬剤に、前記除去細胞の少なくとも一部をエキソビボで曝露する工程と、c)前記処理された腫瘍細胞を溶解して細胞断片を作製する工程と、d)前記断片を前記患者にインビボで導入して抗癌治療効果を生じさせる工程とを含む、方法。
- 工程(c)が、前記処理された腫瘍細胞由来の腫瘍細胞抽出物を調製する工程をさらに含み、工程(d)が、同じ患者にインビボで前記抽出物を導入して抗癌治療効果を生じさせる工程を含む、請求項1に記載の方法。
- 前記抽出物が無細胞である、請求項2に記載の方法。
- 工程(b)の前記処理が、1分と30分との間の時間にわたる、請求項1に記載の方法。
- CD40L発現構築物を前記患者に導入する工程をさらに含む、請求項1に記載の方法。
- 前記腫瘍細胞が、前立腺癌細胞、乳癌細胞、子宮頸癌細胞、子宮癌細胞、膵臓癌細胞、結腸癌細胞、肺癌細胞、および骨癌細胞から成る群から選択される、請求項1に記載の方法。
- 前記腫瘍細胞が皮膚癌細胞である、請求項1に記載の方法。
- 前記腫瘍細胞がリンパ腫細胞である、請求項1に記載の方法。
- 前記抗癌治療効果が、腫瘍サイズの減少を含む、請求項1に記載の方法。
- 前記薬剤がタプシガルギンを含む、請求項1に記載の方法。
- 前記薬剤がタプシガルギシン(thapsigargicin)を含む、請求項1に記載の方法。
- a)癌を発症するリスクのある患者および癌細胞株を提供する工程と、b)処理された癌細胞を作製するため、タプシガルギンおよびタプシガルギン関連化合物から成る群から選択される薬剤で、前記癌細胞株由来の細胞をエキソビボで処理する工程と、c)細胞断片を作製するため、前記処理された癌細胞を溶解する工程と、d)前記患者にインビボで前記断片を導入して抗癌予防効果を生じさせる工程とを含む、方法。
- 工程(c)が、前記処理された腫瘍細胞由来の腫瘍細胞抽出物を調製する工程をさらに含み、工程(d)が、同じ患者にインビボで前記抽出物を導入して抗癌治療効果を生じさせる工程を含む、請求項12に記載の方法。
- 前記抽出物が無細胞である、請求項13に記載の方法。
- 工程(b)の前記処理が、1分と30分との間の時間にわたる、請求項12に記載の方法。
- 前記癌細胞株が、前立腺癌細胞、乳癌細胞、子宮頸癌細胞、子宮癌細胞、膵臓癌細胞、結腸癌細胞、肺癌細胞、および骨癌細胞から成る群から選択される細胞に由来する、請求項12に記載の方法。
- 前記癌細胞株が皮膚癌細胞を含む、請求項12に記載の方法。
- 前記癌細胞株がリンパ腫細胞を含む、請求項12に記載の方法。
- 前記薬剤がタプシガルギンを含む、請求項12に記載の方法。
- 前記薬剤がタプシガルギシンを含む、請求項12に記載の方法。
- a)癌細胞、および前単球細胞株を提供する工程であって、前記前単球細胞株は、マーカータンパク質の発現を促進するNF−κBプロモーターをコードするDNA構築物をトランスフェクトされている、工程と、(b)処理された癌細胞株を作製するため、タプシガルギンおよびタプシガルギン関連化合物から成る群から選択される薬剤で前記癌細胞をインビトロで処理する工程と、c)前記処理された癌細胞由来の細胞断片を調製する工程と、d)インビトロで前記前単球細胞株に前記断片を導入する工程と、e)発現した前記マーカータンパク質の量を測定する工程とを含む、方法。
- 前記マーカーがホタルルシフェラーゼを含む、請求項21に記載の方法。
- 前記前単球細胞株が、ML1、HL60およびU−937から成る群から選択される、請求項21に記載の方法。
- 前記前単球細胞株が、ヒト前単球細胞株THP−1およびヒト前単球細胞株MonoMac−6から選択される、請求項21に記載の方法。
