Description
ホスト細胞は、フラジェリンポリペプチド生産のための本明細書に記載される発現又はクローニングベクターでトランスフェクション又はトランスフォーメーションされ、プロモーターの誘導、トランスフォーマントの選択又は所望の配列をコードする遺伝子の増幅のために、必要に応じて改変した従来の栄養培地において培養される。
細菌の調製:Streptococcus pneumoniae血清型1(臨床分離株E1586)を、National Reference Laboratory - Ministry of Health、ウルグアイ(39)から入手した。ワーキングストックを以下のように調製した。Todd Hewitt Yeast Broth(THYB)(Sigma-Aldrich、セントルイス−ミズーリ州、米国)を、血液寒天培地プレートで増殖させたS. pneumoniaeの新鮮コロニーで播種し、培養物が0.7〜0.9単位のOD600nmに達するまで37℃でインキュベーションした。培養物を、THYB+グリセロール12%(v/v)中、−80℃で最大3ヶ月保存した。マウス感染のために、ワーキングストックを解凍し、滅菌生理食塩水溶液(生理食塩水)で洗浄し、適切な濃度に希釈した。連続希釈物を血液寒天培地プレートに蒔くことによって、ストック中の細菌数を確認した。
フラジェリンの鼻腔内送達は、S. pneumoniaeを用いた致死的チャレンジからマウスを保護する:最初に、本発明者らは、鼻腔内(i.n.)投与でBALB/cマウスを100%死亡させるS. pneumoniaeの最小用量を決定した。S. pneumoniae血清型1の臨床分離株を漸増用量で動物に感染させ、生存を毎日評価した。本発明者らは、4x105CFUを、72〜120時間以内に動物を殺す最小致死用量(MLD)と定義した。
S. pneumoniaeによる気道の早期コロニー形成を防ぐためのTLR及びMyD88要件によって示されるように、先天性免疫は、肺炎球菌感染症をコントロールするのに不可欠である(Albiger, B., Sandgren A., Katsuragi H., Meyer-Hoffert U., Beiter K., Wartha F., Hornef M., Normark S. and Normark B. Immun. 73:298-307.)。気道の肺炎球菌感染症に対する免疫反応は、好中球の大量かつ活発な肺へのリクルートによって特徴付けられ、PMNによる肺炎球菌の食作用殺傷は、主な防御機構であると考えられる。それにもかかわらず、S. pneumoniaeは、食細胞の補体沈着及び呼吸バーストを阻害するか、又は遅らせることによって、ホストの先天的防御から逃れることができる。従って、血清型特異的抗体が産生され、補体沈着阻害を回避して、オプソニン食作用を増強するまで、好中球の流入は、感染症を排除するのに効果がないことが多い。本研究において、本発明者らは、S. pneumoniaeによって通常は捕捉されないTLRのアゴニスト(すなわち、TLR5アゴニストフラジェリン)の外因性投与が、先天性免疫を強化して、気道の急性肺炎球菌感染症をコントロールできるか否かを評価した。本発明者らは、フラジェリンの局所投与が、局所的及び全身的な細菌クリアランスを増強することによって、致死用量のS. pneumoniae血清型1でチャレンジしたマウスの生存を促進することを見出した。フラジェリン処置は、BALB/c、C57BL/6及びNMRIマウスにおいて、感染を確立する前、感染を確立する間及び感染を確立した後に実施した場合に、効果的であった。

- 気道感染症を処置するための方法における使用のための、TLR5アゴニスト。
- 有機小分子;抗体、アプタマー又はポリペプチドからなる群より選択される、請求項1記載のTLR5アゴニスト。
- フラジェリンポリペプチドである、請求項1又は2記載のTLR5アゴニスト。
- 前記フラジェリンポリペプチドが、エシェリキア(Escherichia)、例えばイ・コリ(E. coli)、エンテロバクター(Enterobacter)、エルビニア(Erwinia)、クレブシエラ(Klebsiella)、プロテウス(Proteus)、サルモネラ(Salmonella)、例えばサルモネラ エンテリカ セロバ チフィムリウム(Salmonella enterica serovar Typhimurium)、セラシア(Serratia)、例えばセラチア マルセッセンス(Serratia marcescans)、シゲラ(Shigella)、バシリ(Bacilli)、例えばB.スブチリス(B. subtilis)及びB.リチェニホルミス(B. licheniformis)、シュードモナス(Pseudomonas)、例えばP.エルギノーサ(P. aeruginosa)、及びストレプトミセス(Streptomyces)からなる群より選択される細菌から単離される、請求項3記載のTLR5アゴニスト。
- 前記フラジェリンポリペプチドの前記アミノ酸配列が、配列番号1、配列番号2、配列番号3又は配列番号1、配列番号2若しくは配列番号3と少なくとも約90%、少なくとも約95%、少なくとも約97%、少なくとも約98%若しくは少なくとも約99%同一であるアミノ酸配列からなる群より選択される、請求項3又は4記載のTLR5アゴニスト。
- 前記フラジェリンポリペプチドがa)配列番号3の1位に位置するアミノ酸残基から開始し、配列番号3の99〜173位に位置するアミノ酸残基のいずれか1つからなる群より選択されるアミノ酸残基で終了するアミノ酸配列と少なくとも90%のアミノ酸同一性を有するN末端ペプチド;及びb)配列番号3の401〜406位に位置するアミノ酸残基のいずれか1つからなる群より選択されるアミノ酸残基で開始し、配列番号3の494位に位置するアミノ酸残基で終了するアミノ酸配列と少なくとも90%のアミノ酸同一性を有するC末端ペプチドを含む請求項3記載のTLR5アゴニストであって、前記N末端ペプチドは、前記C末端ペプチドに直接的に連結されているか、又は前記N末端ペプチド及び前記C末端ペプチドは、スペーサー鎖を介して互いに間接的に連結されている、TLR5アゴニスト。
