Abstract
Description

マウスの血統および寄生生物
雌BALB/cマウス(6〜8週齢)は、Harlan Interfauna Iberica S.A.(バルセロナ、スペイン)から購入した。
ホスホロチオエート修飾CpG−ODN(5’−TCAACGTTGA−3’および5’−GCTAGCGTTAGCGT−3’)(配列番号5、6)は、Isogen(オランダ)により合成された。
L.majorのL3およびL5タンパク質をコードするオープンリーディングフレーム(ORF)を、L.majorゲノムデータベース(www.genedb.org/genedb/leish)から、S.cerevisiaeのL3およびL5タンパク質配列をプローブとして使用して得た(56)。rLmL3およびrLmL5タンパク質の発現のために、これらのコーディング領域(CR)を、L.major(MHOM/IL/80(Friedlin))由来のDNAを鋳型として使用してPCR増幅させた。増幅させたDNAを、pBluescriptプラスミド(Stratagene、La Jolla CA)内にクローニングし、配列決定し、その後pQE30発現ベクター(QIAGEN、Hilden、ドイツ)内にクローニングした。
rLmL3およびrLmL5タンパク質を、pQE−LmL3またはpQE−LmL5のプラスミドを用いて形質転換させたEscherichia coliにおいて過剰発現させ、変性条件下でNi−ニトリロ三酢酸(Ni−NTA)アガロースカラム(Qiagen)において精製した。Ni−NTAアガロースに結合させた後、組換えタンパク質を、(57)に記載のように、親和性カラムにおいて再フォールディングさせた。組換えタンパク質を、ポリミキシン−アガロースカラム(Sigma、St.Louis、Mo.)に通した。残留エンドトキシン含有量(<12pg/μgの組換えタンパク質)を、Quantitative Chromogenic Limulus Amebocyte Assay QCL−1000(BioWhittaker、Walkersville、Md.)により測定した。
2つの独立したBALB/cマウス群(6匹/群)に、10μgのrLmL3または10μgのrLmL5を25μgの各CpG−ODNと混合して、右フットパッドに皮下(s.c.)接種した。対照として、2つの追加のマウス群に、25μgの各CpG−ODNを単独で、またはリン酸緩衝生理食塩水(PBS)を接種した。個々の群に、2週および4週後に、プライミングに使用した同じ用量をブーストした。寄生生物のチャレンジを、5×104の静止期前鞭毛型のL.major(WHOM/IR/−/173)を、左(未処置)フットパッドに最終接種後4週間にs.c.接種することによって実施した。フットパッドの腫脹を、メートル法のキャリパーを用いて測定し、左フットパッドの厚み−右フットパッドの厚みとして計算した。寄生生物の数を、耳、流入領域リンパ節(DLN)および脾臓において、(70)に記載のように限界希釈によって決定した。
L.majorのLRPの調製のために、109の前鞭毛型を収穫し、予備冷蔵の洗浄PBSで2回洗浄し、1mlのNP40溶解バッファー(10mMのTris HCl、pH8.0、150mMのNaCl、1.5mMのMgCl2および0.5%NP40)に再懸濁し、10回上下にピペッティングした。溶解後、試料を、3,000×gで2分間4℃において微量遠心分離し、核をペレット化した。上清を、13,000×gで15分間、4℃において2回微量遠心分離し、[57]に記載のようにサイトゾルの上清からリボソームを調製した。簡潔に言うと、サイトゾルを、90,000rpmで30分間、4℃においてBeckman TL100.3 ローターで高速遠心分離に供した。粗リボソームペレットを、バッファーA(20mMのTris−HCl、pH7.4、500mMのAcNH4、100mMのMgCl2、5mMのβ−メルカプトエタノール)に再懸濁し、バッファーA中不連続ショ糖勾配(20/40%)を介して、90,000rpmで4℃においてTL100.3ローターで遠心分離にかけた。洗浄されたリボソームのペレットをPBSに溶解し、リボソームのRNA分解が完了するまで超音波処理した。マウスリボソームタンパク質抽出液(MRP)を、5×107のRAW264.7マウスマクロファージ細胞から、同じ手順を使用して調製した。
IFN−γ、IL−10およびIL−4の放出を、組換えタンパク質を用いて刺激された脾細胞の細胞培養液の上清において、市販のELISAキット(Diaclone、Besancon、フランス)を使用して測定した。簡潔に言うと、3×106脾臓細胞を、48−ウェルプレートに、48時間37℃において、rLmL3(6μg ml−1)またはrLmL5(6μg ml−1)または媒体単独の存在下で播種した。
イヌ血清試料を、エストレマドゥーラ(Extremadura)地方(スペイン)のLeishmania infantumに自然に感染した、20匹の臨床的に症候性のイヌから採取した。感染した動物を、Department of Parasitology of the Veterinary School、Extremadura University、Caceres、スペインにおいて臨床的および分析的に評価した。間接的免疫蛍光法によって試験した場合、すべての血清は陽性であり、寄生生物の無鞭毛型の存在が、膝窩および前肩甲骨のリンパ節における直接観察によって確認された。対照血清は、Department of Parasitology(Extremadura University)において飼育された8匹の健康な動物から得た。
統計分析を、スチューデントt検定によって実施した。P<0.05の場合、差は有意であると考えた。
LmL3およびLmL5タンパク質の同定、クローニングおよび発現
L.majorのL3およびL5のコーディング領域を同定するために、本発明者らは、プローブとしてS.cerevisiaeのL3およびL5のアミノ酸配列(それぞれ、YOR063wおよびYPL131w)を使用して、BLASTP検索を実施した[58]。推定LmL3およびLmL5タンパク質として注釈を付けた、2つの異なるエントリ(LmjF34.2880およびLmjF35.1890)を、BLASTスコアの有意値を用いてレスキューした。データベースの配列データに基づき、PCRプライマーを設計し、クローニングを目的とするさまざまな制限酵素のためのカット部位を含む、LmL3およびLmL5のCRを増幅した。増幅されたDNAを、pBluescriptにサブクローニングし、配列決定した。L.majorの推定L3タンパク質は、分子量47.5kDaおよび予測等電点11.67を有する、419個のアミノ酸を保有する。L.majorの推定L5タンパク質は、分子量36.6kDaおよび予測等電点10.69を有する、328個のアミノ酸を保有する。L.majorのL3およびL5の推定アミノ酸配列と、それらのS.cerevisiaeカウンターパートとの比較により、高度の相同性:L3タンパク質に関して57%の同一性、73.5%の類似性、L5タンパク質に関して;51.2%の同一性、66.3%の類似性が明らかにされた(アライメントを参照されたい)。ここに提示されたアライメントは、LeishmaniaのL3およびL5タンパク質が、高度の類似性を有するいくつかのドメインを含有することを示す。両方の寄生生物タンパク質は酵母タンパク質より長く、LmL3に関してはカルボキシ末端およびLmL5に関しては両方の末端に余分なアミノ酸残基を提示することが観察されることもまた、注目に値する。感染期間中にこれらのタンパク質に対して誘発された液性応答および細胞性応答は、宿主カウンターパート由来の相同タンパク質との交差反応性を有さない寄生生物に対して特異的であるので、保存タンパク質ファミリーに属するLeishmaniaタンパク質の稀な一次構造は、免疫学的に関連していると思われる(39、36、58)。
保護に関与する免疫学的パラメータを決定するために、SLA、LRP、MRPおよび対応する組換えタンパク質(L3およびL5)により駆動されるサイトカインの産生(IFN−γ、IL−4およびIL−10)を、ワクチン接種されたマウス群および対照マウス群においてチャレンジ後8週に分析した。L3およびL5をワクチン接種されたマウス由来の脾臓細胞は、チャレンジ後8週に、対照マウス由来の脾臓細胞より多くのSLA、LRPおよび組換え抗原特異的IFN−γを産生した(L3に関して図5aおよびL5に関して図6a)。MRPを用いた脾臓細胞培養液の刺激は、IFN−γの産生をもたらさなかったので、このサイトカインの産生は、Leishmaniaのリボソームタンパク質により特異的に誘導されることが見出された(L3に関して図5aおよびL5に関して図6a)。さらに、対照マウス(CpGおよび食塩水)と比較した場合、低レベルのSLAおよびLRP特異的IL−10(L3に関して図5bおよびL5に関して図6b)およびIL−4(L3に関して図5cおよびL5に関して図6c)が、保護されたマウスから得られた脾臓細胞の上清において見出された。さらに、MRP特異的L3およびL5依存性のIL−10およびIL−4の産生は、保護されたマウスにおいて非常に低かった。したがって、感染後、保護の表現型は、寄生生物により誘導されるIL−4およびIL−10の応答を調節できる、L3およびL5のTh1応答の誘導と関連したことを結論付けることができる。
L3およびL5のホモログのアライメント
本発明者らは、さまざまなリーシュマニア(Leishmania)種間のL3およびL5リボソームタンパク質の保存の程度を分析した。この目的のために、L.infantumおよびL.mexicana(配列番号48および50)由来のL3タンパク質ならびにL.infantum(クローンJPCM5[MCAN/ES/98/LLM−877])、L.braziliensis(MHOM/BR/75/M2904)およびL.mexicana(MHOM/GT/2001/U1103)(配列番号52、56および54)由来のL5タンパク質のアミノ酸配列を、ゲノムデータベース(www.genedb.org)からin silico分析によってレスキューし、L.majorオルソログ(L3に関して配列番号1およびL5に関して配列番号3)からのアミノ酸配列と比較した(次ページにアライメントを示す、パートAおよびB)。L.majorのL5タンパク質中のN末端の伸張の存在を除き、高度の保存が、さまざまな種の間で観察された。表1A−Bはそれぞれ、L3およびL5オルソログの間の同一性および類似性の百分率を示す。
本研究に用いた組換えLmL3およびLmL5が、寄生生物リボソーム中に位置するタンパク質に対応することを実証するために、本発明者らは、組換えタンパク質に特異的な抗体を、組換えタンパク質およびLRP抽出液を含有するウェスタンブロットに用いた。図8Aは、組換えタンパク質を用いて免疫化されたマウスから得られた抗LmL3抗体が、LRP抽出液中の予想分子量(47.5kDa)を有する単一バンドを認識したことを示す。同様の結果が、L5タンパク質に関しても観察された(図8B)。本発明者らが、先に記載のようにrLmL5−Shepharose4Bカラムにおける親和性クロマトグラフィーによって、L.infantumに自然に感染したイヌ血清から精製された抗LmL5抗体を用いた場合、36.6kDaの単一バンドがLRP抽出液中に観察された(71)。陽性対照として、両方の事例において、対応する組換えタンパク質は、特異的抗体によって認識された。
本発明者らは、L.braziliensisの感染により引き起こされた皮膚リーシュマニア症の発症における、Th1誘導性アジュバント(CpG−ODN)と組み合わせたL.majorの組換えタンパク質rLmL3およびrLmL5の免疫化の効果を分析した。この目的のために、5匹のマウスの群を、50μgのCpG−ODN(25μgのCpG−1[5’−TCAACGTTGA−3’]+25μgのCpG−2[5’−GCTAGCGTTAGCGT−3’)](配列番号5および6))と組み合わせて、10μgのrLmL3または10μgのrLmL5を用いて独立に免疫化した。対照として、5匹のマウスの群を、50μgのCpG−ODN単独またはPBS(賦形剤として用いたバッファー)を用いて免疫化した。マウスに、耳の真皮(左耳)に接種した。各群に、2週および4週後に、プライミングに使用した同じ用量をブーストした。寄生生物のチャレンジを、右(未処置)耳に105の静止前鞭毛型のL.braziliensis(MHOM/BR/01/BA788)を、2対のLutzomya intermediaスナバエの唾液腺と組み合わせて注射することにより実施した。感染のこのモデルにおいて、寄生生物のチャレンジ後、BALB/cマウスは、感染した耳において炎症性病変を発症し、着実に進行し、およそ5週に最大に達する。その後、病変サイズは退行し、完全な耳の瘢痕化が、感染後およそ9週に観察された(72)。本発明者らは、4匹のマウス群における皮膚病変の発症を、寄生生物チャレンジ5週まで、メートル法のキャリパーを用いて病変の厚みを測定して分析した。図7Aに示すように、rLmL3+CpG−ODNまたはrLmL5+CpG−ODNをワクチン接種されたマウスの耳病変は、PBSまたはCpG−ODNの対照群と比較して有意に低かった。さらに、寄生生物負荷量を、感染した耳において感染後5週に分析した。寄生生物の数を、(73)に記載のように限界希釈アッセイにより決定した。対照群と比較した場合、寄生生物負荷量の減少が、ワクチン接種されたマウスにおいて観察された。差は、両方の対照群に対してrLmL5+CpG−ODN群において有意であったことが見出された(P<0.05、スチューデントt検定)。さらに、5匹のマウスのうちの4匹(rLmL5+CpG−ODN群)および5匹のマウスのうちの1匹(rLmL3+CpG−ODN群の耳において寄生生物は検出できなかった(図7B)。したがって、組換えタンパク質として発現され、CpG−ODNと組み合わされたL.majorのL3およびL5リボソームタンパク質をワクチン接種されたマウスは、L.braziliensisによる異種チャレンジに対して保護されることが結論付けられた。
5つのマウス群(n=4/群)を、配列番号5および6(50μg)で先に同定されたCpG ODNの存在下で、10μgのS6(配列番号34)、L2(配列番号22)、L7(配列番号24)、L8(配列番号26)およびL16(配列番号28)を用いて独立に免疫化した。対照として、マウスの群を、アジュバントを用いて免疫化し、他の群を、賦形剤(PBS−食塩水)を用いて免疫化した。3用量を2週の間隔で投与した。すべての免疫化は、右のフットパッドに実施した。最終用量の1ヵ月後、マウスを、105の静止期前鞭毛型のL.majorを左フットパッドに皮下注射し、感染させた。真皮の病変の発症を、フットパッドの腫脹を、チャレンジ後8週まで測定することによって評価した(図9A)。S6+CpG ODNをワクチン接種されたマウス由来のフットパッドの腫脹は、対照および他の4種のタンパク質を用いて免疫化されたマウスにおいて観察されたフットパッドの腫脹に対して有意に低かったが、すべての群からのマウスが炎症性病変を発症した。さらに、マウスの流入領域リンパ節(DLN)および脾臓における寄生生物負荷量を分析した。S6+CpG ODNを用いて免疫化された動物は、それぞれ、食塩水およびCpG ODNの群と比較して、脾臓における寄生生物の数が2−logの減少を示した(図9B)。しかし、S6+CpG ODNの群のDLN中に見出される寄生生物負荷量は、対照において観察された寄生生物負荷量と同様であった。このアッセイから、CpG ODNアジュバントの存在下のS6組換えタンパク質を用いたワクチン接種は、BALB/cマウスにおいてL.major感染によるCLに対する部分的保護をもたらす免疫状態の誘導であると結論付けることができる:感染部位における対照より低い炎症性病変および脾臓における対照より低い寄生生物負荷量の存在。他方では、他の4種の抗原(L5、L7、L8およびL16)を用いたワクチン接種では、対照と比較してCLの進行に有意な変化はもたらされなかった。
L3、L5およびS4リボソームタンパク質により誘導される保護をより詳細に分析するために、新たな免疫化−感染実験を実施した。マウスの12の群(n=4/群)が分析に含まれた。すべての場合において、マウスを、皮下に3回(2週間空けて)、右のフットパッドに免疫化した。次の群をアッセイした:

Claims (33)
- 対象における寄生虫症を治療する、または予防する、または遅らせるための、L5および/またはL3供給源の使用。
- L3供給源が、配列番号1のアミノ酸配列と少なくとも60%の配列の同一性または類似性を有するアミノ酸配列を含むポリペプチドであり、および/または配列番号2と少なくとも60%の同一性を有するヌクレオチド配列によりコードされるポリペプチドであり、且つ、L5供給源が、配列番号3のアミノ酸配列と少なくとも60%の配列の同一性または類似性を有するアミノ酸配列を含むポリペプチドであり、および/または配列番号4と少なくとも60%の同一性を有するヌクレオチド配列によりコードされるポリペプチドである請求項1に記載の使用。
- S4および/またはS6供給源がL3および/またはL5供給源と併用される請求項1または2に記載の使用。
- 対象における寄生虫症を治療する、または予防する、または遅らせるためのS6供給源の使用。
- アジュバントが存在する請求項1から4の何れか1項に記載の使用。
- 前記L3および/またはL5および/またはS4および/またはS6供給源はワクチンである請求項1から5の何れか1項に記載の使用。
- 前記L3および/またはL5および/またはS4および/またはS6供給源は、タンパク質、タンパク質断片、ペプチド、核酸である請求項1から6の何れか1項に記載の使用。
- 前記アジュバントがTh1促進アジュバントである請求項5から7の何れか1項に記載の使用。
- 前記Th1促進アジュバントがCpG ODNである請求項8に記載の使用。
- L3および/またはL5供給源を含む組成物。
- S4および/またはS6供給源が前記組成物に含まれている請求項10に記載の組成物。
- S6供給源を含むか、またはS6供給源から成る組成物。
- アジュバントをさらに含む請求項10から12の何れか1項に記載の組成物。
- 前記アジュバントがTh1促進アジュバントである請求項13に記載の組成物。
- 前記Th1促進アジュバントがCpG ODNである請求項14に記載の組成物。

- 対象における寄生虫症をインビトロで診断するためのL3および/またはL5供給源の使用。
- S4および/またはS6供給源がさらに使用される請求項19に記載の使用。
- 対象における寄生虫症をインビトロで診断するためのS6供給源の使用。
- 前記L3および/またはL5および/またはS4および/またはS6供給源がリーシュマニア(Leishmania)種から得られる請求項1から9の何れか1項または請求項19から21の何れか1項に記載の使用。
- 前記リーシュマニア種は、Leishmania major、Leishmania infantum、Leishmania braziliensisまたはLeishmania mexicanaである請求項22に記載の使用。
- 前記寄生虫症はリーシュマニア症またはマラリアである請求項1から9の何れか1項または請求項19から23の何れか1項に記載の使用。
- 前記寄生虫症は、リーシュマニアまたはマラリア原虫(Plasmodium)種によって引き起こされる請求項1から9の何れか1項または請求項19から24の何れか1項に記載の使用。
- 前記寄生虫症は、前記L3および/またはL5および/またはS4および/またはS6供給源の由来となる種とは異なる種によって引き起こされる請求項1から9の何れか1項または請求項19から25の何れか1項に記載の使用。
- L3および/またはL5の供給源を使用して対象における寄生虫症を診断するための方法であって、前記対象から得た試料中に、前記供給源を認識する抗体が存在するかどうかを決定することを含む方法。
- S4および/またはS6供給源がさらに使用される請求項27に記載の方法。
