Description
本発明は受動型固形腫瘍標的抗癌プロドラッグ及びその調製方法に関しており、抗癌剤の分野に属する。
正常組織の微小血管内皮は極小ギャップ及び無欠構造を有しているため、大型分子及び脂質粒子が血管壁を通過することは困難である。正常組織の毛管に比して固形腫瘍は、脈管が多い組織、不規則な形状及び構造、毛管の拡張及び微小血管壁の欠損、内皮細胞の無規律配列、劣った構造一体性、内皮細胞間の広くなったジャンクションギャップ並びにリンパ回復不調を特徴とし、大型分子物質及び脂質粒子に選択的に増強された透過性及び保持性を持たせる。この現象は固形腫瘍の増強された透過性及び保持性と称される(以降“EPR効果”)。走査電子顕微鏡は、人間の結腸腺癌における微小血管内皮細胞のジャンクションギャップは400nmまで至るが、正常組織の微小血管内皮細胞のジャンクションギャップは100nm未満であることを示す。固形腫瘍組織の病理学的構造の特徴は、大型分子抗癌剤を受動的に標的化するか固形腫瘍を選択させ、全身的投薬後に腫瘍組織に対して集中的に分布させる。この現象は受動型固形腫瘍標的特性としても知られている。しかし、小型分子抗癌薬は正常組織及び腫瘍組織の血管壁を自由に通過でき、正常組織及び腫瘍組織内の両方に一定割合で薬物を分布する。このことは抗癌効果の選択性の欠乏並びに強毒性及び副作用の重要な理由の1つである。従って、小型分子の抗癌薬には受動的な標的効果はない。グレイシュK他は、相対分子量が40000以上の大型分子物質は腎臓濾過並びに腎クリアランスを克服し、通常よりも長い血漿半減期を有しており、大型分子物質の全身循環時間の延長はEPR効果の実現に有効に作用し、投薬頻度も減少させることを報告している。
本発明により解決されるべき技術的課題は、ペクチン抗癌プロドラッグのEPR効果に基づく受動型腫瘍標的特性の提供、並びに固形腫瘍組織内進入後に薬剤を放出し、キャリアを小型分子に分解し、排出を助けることである。
a)Mw5000から45000(好適には10000から30000)の小型分子ペクチンを水中に溶解させ、塩酸ドキソルビシンを添加し、均質に混合した後にEDC・HClと反応させ、透析し、乾燥させてMw100000から1000000のペクチン-ドキソルビシン複合体を得る工程と、
b)そのペクチン-ドキソルビシン複合体を水に投入し、PVP及びグリセロールを添加し(あるいは一定量のレシチンまたはDMSOを添加し(添加量:ペクチン-ドキソルビシン複合体の2%未満))、均質に混合して懸濁液を調製し、その懸濁液を超高圧ナノホモジナイザーで処理し、粒径100nmから200nmの受動型固形腫瘍標的抗癌プロドラッグを得る工程と、
を含んでいる。
受動型固形腫瘍標的抗癌プロドラッグの調製方法は、大型分子ペクチンをMw5000から45000(好適には10000から30000)の小型分子ペクチンにカットオフし、続いて小型分子ペクチン-ドキソルビシンを(アミド結合により)合成し、ペクチン分子のカルボキシル基及びヒドロキシル基を縮合して大型分子を形成し、大型分子を超高圧ナノホモジナイザーで処理し、粒径100nmから200nm、分子量100000から1000000の受動型固形腫瘍標的抗癌プロドラッグを得る工程を含んでいる。
1.小型分子ペクチンの調製:ペクチンを蒸留水中に溶解させ、ペクチンをNaOHと反応させ、pHを濃塩酸で中和し、カットオフによってMw5000から45000(好適には10000から30000)の小型分子ペクチンを得る。
実験手法は次の通りである。まず分子量10000から30000の小型分子ペクチンを溶解させ、1-エチル-3-(3-ジメチルアミノプロピル)塩酸カルボジイミド(EDC・HCl)の存在下で塩酸ドキソルビシン溶液とpH5から7にて40℃から60℃で反応させ、透析して乾燥させ、42mg/Lの溶解性で融点220℃から245℃である赤茶色の水不溶性の固体(ペクチン-ドキソルビシン複合体)を得る。
実施例1:本発明の受動型固形腫瘍標的抗癌薬P(A)nの調製
1.小型分子ペクチンの調製
この調製は、13.3gのペクチンを計量し、1Lの蒸留水を加え、混合して溶解させ、pHを5MのNaOHで13に調整し、65℃で10時間反応させ、反応を停止させ、さらに、反応溶液を濃塩酸で中和し、カットオフ分子量30000のミリポアで濾過し、濾過物をカットオフ分子量10000のミリポアで濾過し、カットオフ分子量10000の不透過性固形物を回収し、減圧濃縮して乾燥状態とし、真空下で乾燥させて分子量10000から30000の小型分子を得るものである。
配合及び架橋の工程は、分子量10000から30000の小型分子ペクチン-ドキソルビシンを計量して反応フラスコに投入し、100mlの水を加え、混合して溶解させ、0.5gの塩酸ドキソルビシンを計量し、超音波溶解のために50mlの蒸留水を加え、その塩酸ドキソルビシン溶液を反応フラスコに投入し、さらに50mlの蒸留水で反応フラスコに付着したドキソルビシンを洗浄し、続けて1gのEDC・HClを計量して反応フラスコに加え、50℃に温度を上昇させて6.5時間反応させ、反応後にカットオフ分子量(Mw)3500の透析バッグを投入して1日透析し、蒸留水を3時間毎に交換し、続いて溶媒を蒸発させて乾燥状態とし、真空下で12時間乾燥させて赤茶色の固形物、すなわち42mg/Lの溶解性と、融点220℃から245℃である1.1gの水不溶性大型分子ペクチン-ドキソルビシン複合体を得るものである。
標準曲線の設定:標準塩酸ドキソルビシン溶液を10.00、20.00、30.00、40.00及び50.00μg/mLの濃度で正確に調製し、479.5nm(紫外線可視吸光分光機を使用したスペクトル走査により決定)での吸光を確認。
エステル結合及びアミド結合に対するP(A)n内のカルボン酸塩の影響を回避させるべく、P(A)nはプロドラッグサンプルを調製するために脱塩処理された。ドキソルビシン、ペクチン、本発明サンプル及びドキソルビシンとペクチンの混合物は二次水に溶解された。紫外線スペクトル及び赤外線スペクトルは図1と図2でそれぞれ示すように現れた。
0.468gの大型分子不溶性ペクチン-ドキソルビシン複合体(薬剤含有率21.4%)に1gのPVP、3mlのグリセロール及び50mlの水が加えられ、粉砕処理され、懸濁液が調製され、超高圧ナノホモジナイザー(河北廊坊通用机器制造有限公司製造T−200D型)で処理された。懸濁液はまず120mpaにて処理され、2回目は180mpaで処理され、3回目は190mpaで処理され、計3回の超高圧ナノホモジナイザー処理された。超高圧ナノホモジナイザーで処理後の粒径分布は図3に示されている。
実施例2:本発明の受動型固形腫瘍標的抗癌薬P(A)nの調製
1.小型分子ペクチンの調製
この調製は、次のステップを含む。13.3gのペクチンを計量し、1Lの蒸留水を加え、混合して溶解させ、pHを5MのNaOHで13に調整し、65℃で10時間反応させ、反応を停止し、反応溶液を濃塩酸で中和し、10000のカットオフ分子量のミリポアで濾過し、濾過物をカットオフ分子量7000の透析バッグで透析し、透析物を回収し、減圧し、濃縮して乾燥状態とし、真空下で乾燥させて、7000から10000の分子量の小型分子ペクチンが得られる。
配合及び架橋は、次のステップを含む。1gの分子量7000から10000の小型分子ペクチン−ドキソルビシンを計量して反応フラスコに投入し、100mlの水を加え、混合して溶解させ、0.5gの塩酸ドキソルビシンを計量し、50mlの蒸留水を加えて超音波溶解させ、その塩酸ドキソルビシン溶液を反応フラスコに投入し、さらに50mlの蒸留水で反応フラスコに付着したドキソルビシンを洗浄し、1gのEDC・HClを計量して反応フラスコに投入し、50℃に温度を上昇させ、6.5時間反応させ、反応後に透析のために1日、カットオフ分子量(Mw)3500の透析バッグを配合し、3時間毎に蒸留水を交換し、続いて溶媒を蒸発させて乾燥状態とし、12時間真空下で乾燥させて赤茶色の固形を得るものである。すなわち、24.2%の薬剤含有率である1.2gの大型分子不溶性ペクチン-ドキソルビシン複合体が得られる。
実施例3:本発明の受動型固形腫瘍標的抗癌薬P(A)nの調製
1.小型分子ペクチンの調製
この調製は、次のステップを含む。13.3gのペクチンを計量し、1Lの蒸留水を加え、混合して溶解させ、pHを5MのNaOHで13に調整し、65℃で10時間反応させ、反応を停止し、反応溶液を濃塩酸で中和し、50000のカットオフ分子量のミリポアで濾過し、濾過物をカットオフ分子量20000の透析バッグで48時間透析し、3時間毎に蒸留水を交換し、減圧し、濃縮して乾燥状態とし、真空下で乾燥され、20000から50000の分子量の小型分子ペクチンが得られる。
配合及び架橋は、次のステップを含む。1gの分子量20000から50000の小型分子ペクチンを計量して反応フラスコに投入し、100mlの水を加え、混合して溶解させ、0.5gの塩酸ドキソルビシンを計量し、50mlの蒸留水を加えて超音波溶解させ、その塩酸ドキソルビシン溶液を反応フラスコに投入し、さらに50mlの蒸留水で反応フラスコに付着したドキソルビシンを洗浄し、1gのEDC・HClを計量して反応フラスコに投入し、50℃に温度を上昇させ、6.5時間反応させ、反応後に透析のために1日、カットオフ分子量(Mw)3500の透析バッグを配合し、3時間毎に蒸留水を交換し、続いて溶媒を蒸発させて乾燥状態とし、12時間真空下で乾燥させて赤茶色の固形を得るものである。すなわち、25.2%の薬剤含有率である1.2gの大型分子不溶性ペクチン-ドキソルビシン抱合体が得られる。
試験例1:本発明の受動型固形腫瘍標的抗癌プロドラッグのリソソーム加水分解試験
リソソーム源:文献「細胞生物学の実験方法及び技術」に従って純化リソソームが抽出された。
移動相:メタノール:アセトニトリル:リン酸バッファー塩=7:4:6
検出波長:480nm
流量0.8mL/分
超濾過後に残るフィルターケーキが回収され、95%エタノールにより液体の赤色が消えるまで繰り返し洗浄され、溶媒は蒸発されて乾燥状態にされ、蒸留水が加えられて沈殿物が溶解され、分子量がゲル透過クロマトグラフィにより確認された。
クロマトグラフカラム:ウルトラヒドロゲル(登録商標)リニアカラム(7.8x300mm、水)
カラム温度:40℃
フローセル温度:35℃
移動相:0.005MのKNO3
流量:0.5mL/分
サンプル濃度:5mg/mL(0.05%のアジ化ナトリウムを溶解に使用)
サンプルサイズ:20μL
試験例2:MTT法による腫瘍細胞ラインの成長活性に対する受動型固形腫瘍標的抗癌プロドラッグの阻害活性の観察
塩酸ドキソルビシンは浙江海正薬業股▲分▼有限公司から購入され、生理塩水内に溶解させることで2mg/mlのドキソルビシンを含むように調製された。
試験例3:肺転移腫瘍に対する受動型固形腫瘍標的抗癌プロドラッグの実験研究
塩酸ドキソルビシンは浙江海正薬業股▲分▼有限公司から購入された。
細胞培養:細胞はRPMIの1640が100mL/Lウシ胎児血清の培養液及びペニシリン-ストレプトマイシン溶液(100U/mLペニシリン、100mg/Lストレプトマイシン)内で日常的に培養され、培養のために37℃の50mL/LのCO2培養器に入れられ、対数増殖相の細胞が3または4安定継代培養後に回収され、2.5g/Lのトリプシンにより消化され、血清を含まない培養液の懸濁により回収され、細胞密度はその後の利用のために無血清1640媒液によって調整された。
Claims (10)
- ペクチンとドキソルビシンとを結合させて調製される受動型固形腫瘍標的抗癌プロドラッグであって、Mwが5000から45000の小型分子ペクチンをドキソルビシンと反応させてMwが100000から1000000のペクチン-ドキソルビシン複合体を取得し、該複合体を懸濁して懸濁液を準備し、該懸濁液を超高圧ナノホモジナイザーで処理し、粒径が100nmから200nmで、融点が220℃から245℃の受動型固形腫瘍標的抗癌プロドラッグを取得して調製されるものであり、前記ペクチンと前記ドキソルビシンはアミド結合により結合しており、前記ペクチンはペクチン分子のカルボキシル基とヒドロキシル基を縮合することで形成されるエステル結合によって結合されていることを特徴とする受動型固形腫瘍標的抗癌プロドラッグ。
- 前記受動型固形腫瘍標的抗癌プロドラッグの粒径は130nmから180nmであることを特徴とする、請求項1記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 前記受動型固形腫瘍標的抗癌プロドラッグの水溶度は42mg/Lであることを特徴とする、請求項1又は2記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 前記ペクチン-ドキソルビシン複合体は、pH5からpH7、40℃から60℃で、EDC・HClの存在下で前記小型分子ペクチンをドキソルビシンと反応させることで取得されることを特徴とする、請求項1記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 前記小型分子ペクチンのMwは10000から30000であることを特徴とする、請求項4記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 前記ペクチン-ドキソルビシン複合体は水中に投入され、PVP及びグリセロールが加えられ、均質に混合されて懸濁液が作製され、前記懸濁液は超高圧ナノホモジナイザーで処理されて取得されることを特徴とする、請求項1から5のいずれかに記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 請求項6記載の受動型固形腫瘍標的抗癌プロドラッグを調製する方法であって、前記懸濁液を超高圧ナノホモジナイザーにて、まず120mpaで処理し、続いて180mpaで処理し、さらに190mpaで処理する3回の処理を特徴とする方法。
- 前記PVPの量は前記ペクチン-ドキソルビシン複合体の1倍から6倍であり、前記グリセロールの量は前記ペクチン-ドキソルビシン複合体の0.1%から0.8%であることを特徴とする、請求項7記載の受動型固形腫瘍標的抗癌プロドラッグ。
- 受動型固形腫瘍標的抗癌プロドラッグを調製する方法であって、
(a)Mwが5000から45000の小型分子ペクチンを水中に溶解させ、塩酸ドキソルビシンを加え、均質に混合した後にEDC・HClと3時間から8時間反応させ、透析して乾燥させ、Mwが100000から1000000のペクチン-ドキソルビシン複合体を取得するステップと、
(b)前記ペクチン-ドキソルビシン複合体を水中に投入し、PVP及びグリセロールを加え、均質に混合して懸濁液を作製し、前記懸濁液を超高圧ナノホモジナイザーで処理し、粒径が100nmから200nmである受動型固形腫瘍標的抗癌プロドラッグを取得するステップと、
を含んでいることを特徴とする方法。 - 前記小型分子ペクチンは、ペクチンを水中に溶解させ、前記ペクチンをpH13のNaOH溶液と反応させ、濃塩酸でそのpHを中和させ、分子量をカットオフすることで取得されることを特徴とする、請求項9記載の受動型固形腫瘍標的抗癌プロドラッグを調製する方法。
