Abstract
Description
・保護される器官/組織の上に可撓性フィルムを形成し、
・保護される器官/組織に付着し、
・6ヶ月までの時間内に生物分解され得、
・生体適合性であり、
・細胞毒性および組織毒性を有さない分解生成物を生成し、
・用途に合わせて迅速に硬化し、
・硬化中に、組織に損傷を与える有意な発熱を示さず、
・微調整された粘度の故に適用し易く、組織のより深い層まで浸透できない、
癒着バリアを製造することである。
(A)一般式(I):
R1、R2は、ツェレビチノフ活性水素を含有しない同じまたは異なった有機基であり、
nは、少なくとも2の整数である。]
で示されるアミノ官能性アスパラギン酸エステル、
(B)(B1)脂肪族イソシアネートと
(B2)ポリエステルポリオールおよび/またはポリエステルポリエーテルポリオールおよび任意にポリエーテルポリオールを含有し、400g/mol以上の数平均分子量および2〜6の平均ヒドロキシル官能価を有するポリオール成分
との反応によって得ることのできる、1重量%未満、好ましくは0.1重量%未満、より好ましくは0.03重量%未満の残留モノマー含量を有するイソシアネート官能性プレポリマー、および/または
(C)任意に、23℃で10〜6000mPasの範囲のDIN 53019粘度を有する有機充填剤、および
(D)適切な場合には、成分(B)の定義のようなイソシアネート官能性プレポリマーと成分(A)のようなアスパラギン酸エステルおよび/または成分(C)のような有機充填剤との反応生成物
を含んでなるポリウレア系を提供する。
R1、R2は、ツェレビチノフ活性水素を含有せず、1〜20個、好ましくは1〜10個の炭素原子を含有する、同じまたは異なった、任意に分枝または環状であってよい有機基、より好ましくはメチルまたはエチルであり、
nは、2〜4の整数であり、
Xは、n官能性第一級アミンの第一級アミノ基を除去することによって得られ、2〜20個、好ましくは5〜10個の炭素原子を含有する、任意に分枝または環状であってよい、n価の有機基である。
反応温度が60℃を超えないように、窒素雰囲気下、2molのマレイン酸ジエチルを1molの2−メチル−1,5−ジアミノペンタンに徐々に滴加した。次いで、反応混合物中でマレイン酸ジエチルが検出できなくなるまで、反応混合物を60℃で加熱した。
原料
ポリエーテル1は、56mgKOH/gのヒドロキシル価を有し、約50重量%ずつのエチレンオキシド単位およびプロピレンオキシド単位を含有する、BMS AG製の、1,2−プロピレングリコール−出発、KOH−触媒のポリエーテルグリコールであって、エチレンオキシドが鎖末端に存在する。
ポリエーテル2は、190mgKOH/gのヒドロキシル価を有する、BMS AG製の、1,2−プロピレングリコール−出発、KOH−触媒のヒドロキシル−末端ポリエチレンオキシドである。
ポリエーテル3は、550mgKOH/gのヒドロキシル価を有する、BMS AG製の、1,1,1−トリメチロールプロパン−出発、KOH−触媒のヒドロキシル−末端ポリエチレンオキシドである。
表1に記載されているエステル基濃度は、生成物1kgについて使用されたカルボキシル基のモル数に相当する。
記載されている「エチレンオキシド基の割合」は、出発ポリエーテル1(50重量%)、ポリエーテル2(100%)、ポリエーテル3(100%)およびジエチレングリコール(100%)のエチレンオキシド基含量、またはポリエステル調製の出発物質におけるエチレンオキシド基の割合から計算される。
温度計、カラム、上部温度計を備えた還流分割器、下降冷却器および1リットル容受器を備えた6リットル容フラスコで、98g(1.07mol)のグリセロール、935g(0.47mol)のポリエーテル1、1615g(2.73mol)のポリエーテル2および467g(3.2mol)のアジピン酸を、反応水を留去しながら、大気圧および窒素雰囲気下、撹拌しながら200℃まで徐々に加熱した。5時間後、68mgの二塩化錫二水和物を添加し、同時に、減圧を適用して最終的に15mbarまで減圧した。更に20時間後、反応が終わった。分析結果を表1に示した。
実施例C−2a
236.95gのHDIおよび0.4gの塩化ベンゾイルを、500ml容の四ッ口フラスコに初期導入物として導入した。100℃で脱水した実施例B−2のポリエステル2の162.6gを、80℃で2時間の間に添加し、次いで、1時間撹拌した。続いて、140℃および0.1Torrで薄膜蒸留法を使用して過剰HDI(ヘキサメチレンジイソシアネート)を留去すると、5.67%のNCO含量を有するプレポリマー280gが残った。残留モノマー含量は0.03%未満の遊離HDIであった。
281.88gのHDIおよび0.4gの塩化ベンゾイルを、500ml容の四ッ口フラスコに初期導入物として導入した。100℃で脱水した、実施例B−2のポリエステル2の96.75gとモル質量200Daのポリエチレングリコール(PEG 200)の20.97gとの混合物を、80℃で2時間の間に添加し、次いで、1時間撹拌した。続いて、140℃および0.1Torrで薄膜蒸留法を使用して過剰HDIを留去すると、7.88%のNCO含量を有するプレポリマー311gが残った。残留モノマー含量は0.03%未満の遊離HDIであった。
267.82gのHDIおよび0.4gの塩化ベンゾイルを、500ml容の四ッ口フラスコに初期導入物として導入した。100℃で脱水した、実施例B−2のポリエステル2の91.92gとPEG 400の39.85gとの混合物を、80℃で2時間の間に添加し、次いで、1時間撹拌した。続いて、140℃および0.1Torrで薄膜蒸留法を使用して過剰HDIを留去すると、7.57%のNCO含量を有するプレポリマー302gが残った。残留モノマー含量は0.03%未満の遊離HDIであった。
実施例D
10gのプレポリマーBを、ビーカー内で、当量のアミノ官能性アルパラギン酸エステル(アスパルテートA)と十分に混合した。その直後、反応混合物を、腎臓、肝臓および筋肉組織に薄く適用した。硬化時間および硬化温度、並びに組織への付着性を測定した。
実施例1
アルパルテートを混合することなく、プレポリマーA−1を組織に適用すると、30分以内に硬化しなかった。
HDIの代わりにTDIを用いて、実施例Bに記載したように、プレポリマーA−1を調製した。得られたプレポリマーを、様々な量の水と混合し、組織に適用した。いずれも15分以内に硬化しなかった。
465gのHDIおよび2.35gの塩化ベンゾイルを、1リットル容の四ッ口フラスコに初期導入物として導入した。TMP(三官能性)から開始した、63%のエチレンオキシド含量と37%のプロピレンオキシド含量(各々、全アルキレンオキシド含量に基づく)とを有するポリエーテルの931.8gを、80℃で2時間の間に添加し、次いで、1時間撹拌した。続いて、130℃および0.1Torrで薄膜蒸留法を使用して過剰HDIを留去すると、2.53%のNCO含量を有するプレポリマー980g(71%)が残った。残留モノマー含量は0.03%未満のHDIであった。

Claims (13)
- (A)一般式(I):
R1、R2は、ツェレビチノフ活性水素を含有しない同じまたは異なった有機基であり、
nは、少なくとも2の整数である。]
で示されるアミノ官能性アスパラギン酸エステル、
(B)(B1)脂肪族イソシアネートと
(B2)ポリエステルポリオールおよび/またはポリエステルポリエーテルポリオールおよび任意にポリエーテルポリオールを含有し、400g/mol以上の数平均分子量および2〜6の平均ヒドロキシル官能価を有するポリオール成分
との反応によって得ることのできる、1重量%未満の残留モノマー含量を有するイソシアネート官能性プレポリマー、または
(C)任意に、23℃で10〜6000mPasの範囲のDIN 53019粘度を有する有機充填剤、および
(D)適切な場合には、成分(B)の定義のようなイソシアネート官能性プレポリマーと成分(A)のようなアスパラギン酸エステルおよび/または成分(C)のような有機充填剤との反応生成物
を含んでなるポリウレア系。 - 成分(B1)に使用するポリエーテルエステルポリオールが、30〜140mgKOH/gのヒドロキシル価、0.05〜10mgKOH/gの酸価、および23℃で200〜8000mPasのDIN 53019剪断粘度を有することを特徴とする、請求項1に記載のポリウレア系。
- 成分(B2)に、100〜2000g/molの数平均分子量を有し、そのエーテル基の全てまたは一部がエチレンオキシドに由来する、ポリエーテルポリオールを使用することを特徴とする、請求項1または2に記載のポリウレア系。
- ポリエーテルポリオールの割合が、成分(B2)全体に基づいて50重量%以下であることを特徴とする、請求項3に記載のポリウレア系。
- 成分(B2)が、0.5〜5.5のエステル基濃度(単位:mol/kg)を有することを特徴とする、請求項1〜4のいずれかに記載のポリウレア系。
- 成分(C)の有機充填剤が、ヒドロキシ官能性化合物、好ましくはポリエチレングリコールを含むことを特徴とする、請求項1〜5のいずれかに記載のポリウレア系。
- 成分(A)、成分(B)、適切な場合には成分(C)および適切な場合には成分(D)を混合することによる、請求項1〜6のいずれかに記載のウレア系の製造方法であって、遊離アミノ基またはブロックトアミノ基と遊離NCO基の比が1:1であり、充填剤成分(C)とアスパラギン酸エステル成分(A)の重量比が0:1〜20:1の範囲である、方法。
- 請求項7に記載の方法によって得ることのできるポリウレア系。
- 請求項1〜6または8のいずれかに記載のポリウレア系を使用する、細胞組織のシーリング方法、結合方法または被覆方法。
- 細胞組織のシーリング方法、結合方法または被覆方法を実施するためのポリウレア系の使用。
- 請求項1〜6または8のいずれかに記載のポリウレア系を用いて得ることのできる、フィルムおよび複合部材。
- 請求項1〜6または8のいずれかに記載のポリウレア系を用いて得ることのできる、術後癒着バリア。
- 請求項1〜6または8のいずれかに記載のポリウレア系を含む2チャンバーディスペンシングシステムであって、一方のチャンバーが成分(A)のプレポリマーを含み、もう一方のチャンバーが硬化剤成分(B)および適切な場合には成分(C)および適切な場合には成分(D)を含む、2チャンバーディスペンシングシステム。
