Abstract
Description
製薬産業分野では、近年、とりわけ、酵素、抗体及びサイトカイン(例えば、エリスロポエチン、インターフェロン、プラスミノーゲンアクチベーターなど)に基づいた製品が非常に成功を収めている。タンパク質治療剤に対する世界中からの要望は年々増している。治療モノクローナル抗体(mAbs、monoclonal antibodies)は、タンパク質治療薬の中でも重要な一群である。ポリクローナル抗体とは対照的に、モノクローナル抗体は、単一の抗体形成細胞に由来する免疫細胞(細胞クローン)により分泌されるので、モノクローナルと呼ばれる。モノクローナル抗体の特徴は、それらは各々、免疫原性物質の1つのエピトープのみに指向され、従って、1つの抗原性決定基に対してのみ指向され、それ故、疾病の処置において非常に特異的に使用され得るということである。タンパク質治療薬の例としては、Roche Diagnostics GmbH社のモノクローナル抗体のトラスツズマブ(市販名:ハーセプチン)、ダクリズマブ(市販名:ゼナパックス)及びリツキシマブ(市販名:マブセラ)があり、これらは、とりわけ乳ガン(トラスツズマブ)の処置に、臓器拒絶(ダクリズマブ)の処置に、そして非ホジキンリンパ腫(リツキシマブ)の処置に成功裏に使用されている。
本発明は、試料中の抗体及び抗体断片又は改変型の抗体を検出する方法を記載しており、以下の工程:
a)抗体及び/又はその開裂産物を含む試料を得る工程、
b)a)で得られた試料を、
i)IgG特異的システインプロテアーゼ、
ii)グリコシダーゼ、
iii)還元剤
と共にインキュベートする工程、
c)b)でインキュベートされた試料を、液体クロマトグラフィーと接続された質量分析計により分析することにより、a)で得られた溶液中に含まれるインタクトな抗体を検出、及び抗体の断片もしくは改変型を検出する工程
を含むことを特徴とする。
i)化膿性連鎖球菌に由来するIgG特異的システインプロテアーゼ、及び
ii)フラボバクテリウム・メニンゴセプチカムに由来するエンドグリコシダーゼであるN−グリコシダーゼF
を含む。
本研究は、治療モノクローナル抗体の分解産物及び改変体の分析に関し、これらは、例えば、抗体の作製中、抗体の保存中に、又は抗体の製剤化中のストレス条件により形成される。
抗体の酸化は、重鎖及び軽鎖のアミノ酸の共有結合的な改変に相当し、これは、反応性酸素種により誘発される。殆ど全てのアミノ酸が原則的には酸化され得るが、メチオニン(M)及びトリプトファン(W)が最も酸化に対して感受性が高い。これらのアミノ酸の酸化(図1参照)は、非常に多くの異なるタンパク質において起こり、その生物学的活性を低下または消失させたり、凝集を誘発したり、タンパク質分解を促進したりすることが多いために特に関心が高い(Houde, D., et al., J. Chromatogr. A 1123 (2006) 189-198)。メチオニンからメチオニンスルホキシドへと単純に酸化されることにより、酸化種においてΔm=+16Daの質量差が生じる。トリプトファンは通常2回酸化されて、その結果、Δm=+32Daの質量差が生じる。更なる反応において、2回酸化型のトリプトファンはキヌレニンへと再編成され、その結果、Δm=+4Daの質量差が生じる。
抗体分子中のアミノ酸の脱アミド化は、アスパラギン(N)及びグルタミン(Q)において起こり得る。しかしながら、通常、アスパラギンで起こる。更に、特定のアミノ酸配列及びアミノ酸組合せ、例えばアスパラギンとグリシン(NG)、アスパラギンとセリン(NS)、及びアスパラギンとトレオニン(NT)が特に受け易い。アスパラギンの脱アミド化が、保存中の生物学的分子の分解の主な原因である。折り畳まれたインタクトな抗体の脱アミド化は、最初、ストレスの高い条件下でゆっくりとしか起こらない。抗体の3次元構造が例えば、還元及び酵素的開裂後に破壊された場合に、脱アミド化が促進される。なぜなら、これらの場合、アミノ酸は、周囲の媒体とより反応し易くなるからである(図2)。アスパラギンは、中間体産物の形態の環式アミド(スクシンイミド)を形成し得、これは自発的に加水分解されることにより、イソアスパルチルペプチドとアスパルチルペプチドの約3:1の比の混合物が生じる(Chelius, D., et al., Anal. Chem. 77 (2005) 6004-6011)。この反応は、塩基性pH値において優先的に起こる。脱アミド化種のΔm=+1Daの質量差は、アスパラギンからアスパルテート及びイソアスパルテートへの脱アミド化の結果として起こる。10kDaを超える分子量を有するペプチド及びタンパク質の場合、現在の質量分析計を使用してΔm=+1Daの質量差を直接検出することは不可能か又は非常に困難である。更に、荷電分布の変化、より正確には荷電の不均一性の変化が起こる。
非還元性チオエーテル架橋の形成は、モノクローナル抗体、特にIgG1サブクラスのモノクローナル抗体で頻繁に観察される現象である。これは、抗体の重鎖と軽鎖を共に連結しているか、又は、軽鎖及び重鎖を分子内的に安定化している、ジスルフィド架橋の中の硫黄原子の消失に基づく。この抗体の改変は、ストレスの高い条件下、この場合では特に高いpH値において好まれる。このような反応はβ脱離であると推定される(Cohen, S.L., et al., J. Am. Chem. Soc., 129 (2007) 6976-6977)。チオエーテル架橋の形成中に硫黄原子が失われるので、これにより、還元されていない改変された抗体種ではΔm=−32Daの質量差が生じる。これに対して、このジスルフィド架橋(S−S)は抗体の還元中に2つのSH基へと開裂される。この理由から、還元された抗体成分における生じる質量差は、Δm=−34Daである。
ストレスにより誘発される断片化反応は、通常、タンパク質のポリペプチド鎖中のペプチド結合の加水分解性開裂、例えば、抗体の重鎖及び軽鎖の加水分解性開裂に基づく。タンパク質分解及びこのようなペプチド結合の加水分解は、原則的には、全てのアミノ酸の間で起こり得、特に、立体的緊張、又は、加水分解を好む他のアミノ酸の側鎖が存在する場合に起こり得る。
モノクローナル抗体は、非常に大きなタンパク質であり、更に、その重鎖の糖構造に因り極めて不均一(微小不均一)である。分解産物及び改変体の形成について抗体を調べるために、分析前により小さな断片へと開裂することが便宜である。それ故、試料調製の一部としての抗体の開裂は、分析調査を実施する際の重要な方法である。殆どの場合、抗体はジスルフィド架橋の還元によりその重鎖及び軽鎖へと単純に分解される。しかしながら、更に、他の抗体開裂法も存在する。
IgG分子に存在する全てのジスルフィド架橋は、還元により開裂することができる。抗体の遊離の重鎖及び軽鎖が、還元中に得られる。トリス−(2−カルボキシエチル)−ホスフィン(TCEP)は、抗体の全てのジスルフィド架橋が短時間で完全に開裂され、還元が全pH範囲におよび起こるため、しばしば使用されている還元剤である(例えば、Hau, J.C. and Hau, C.Y., Anal. Biochem. 220 (1994) 5-10を参照されたい)。1つの態様において、pH範囲は1.5〜8.5である。ジチオトレイトール(DTT)もまた、ジスルフィド架橋の急速な開裂を特徴とする。しかしながら、酸性環境におけるDTT還元は、非常にわずかにしか進行しない。重鎖及び軽鎖の解離を完了させるために通常、変性工程が必要とされる。変性により更にジスルフィド基により近づき易くなる。変性は、例えば、グアニジン/HCl又はギ酸を用いて実施することができる。
システインプロテアーゼであるパパインは、アルギニン(R)、リジン(K)、グルタミン酸(E)、ヒスチジン(H)、グリシン(G)及びチロシン(Y)の後で比較的非特異的にペプチド結合を開裂する。インキュベーション時間が十分に長ければ、パパインによる消化により全体の加水分解が行われる。しかしながら、抗体は、限定的なタンパク質分解によりそのヒンジ領域において比較的選択的に開裂され得る(Lottspeich, F., and Engels, J.W., "Bioanalytik Spektrum Akademischer Verlag" Munich 2nd Edition (2006) 201-214)。開裂は、2つの重鎖を共に接続しているジスルフィド架橋のN末端側で起こる。ジスルフィド架橋はこの過程において保持されているので、3つの断片(2つのFab断片、1つのFc断片)が消化後に得られる。2つのN末端断片は抗原結合断片(Fab、抗原結合断片(antigen-binding fragment))と称され、C末端断片は結晶性断片(Fc、結晶性断片(crystallizing fragment))と称される。各Fab断片は、完全な軽鎖と、重鎖のアミノ末端半分から構成される。Fc断片は、重鎖の2つのカルボキシ末端半分から構成され、これらは、ジスルフィド架橋により依然として共に連結されている。
IdeS(化膿性連鎖球菌の免疫グロブリンG分解酵素、immunoglobulin G-degrading enzyme of S. Pyogenes)は、病原性細菌である化膿性連鎖球菌から単離することができる細胞性システインプロテアーゼである。この酵素は、認識配列GPSVFLFPの直前で高い特異性でもってヒトIgGを開裂する。この配列は、IgGのヒンジ領域の、2つの重鎖(HC)を共に連結しているジスルフィド架橋のC末端側に位置している。開裂により2本の重鎖(2つのHC−Fc断片)のC末端とFab”断片が得られ、このFab”断片は、ジスルフィド架橋により連結された軽鎖及び重鎖のFab断片腕から得られる(図3)(von Pavel-Rammingen, U., et al., EMBO Journal 21 (2002) 1607-1615)。
N−グリコシダーゼF酵素は、いわゆるエンドグリコシダーゼであり、糖タンパク質の糖構造、例えば抗体の重鎖の糖構造を、ペプチド鎖と近くのN−アセチルグルコサミン残基の間で開裂する。
a)抗体及び/又はその開裂産物及び/又は抗体の改変型を含む試料を得る工程、
b)a)で得られた試料を、
i)IgG特異的システインプロテアーゼ、
ii)グリコシダーゼ、
iii)還元剤
と共にインキュベートする工程、
c)b)でインキュベートされた試料を、液体クロマトグラフィーと接続された質量分析計により分析することにより、a)で得られた溶液中に含まれるインタクトな抗体を検出及び抗体断片を検出する工程、
を含むことを特徴とする。
i)化膿性連鎖球菌由来のIgG特異的システインプロテアーゼ、及び
ii)フラボバクテリウム・メニンゴセプチカム由来のエンドグリコシダーゼであるN−グリコシダーゼF
を含むことを特徴とする。
1.抗体の還元のみ;
2.抗体を脱グリコシル化し、その後、還元;
3.抗体をIdeSで消化し、その後、還元;
4.抗体を脱グリコシル化し、その後、IdeSで消化し還元
材料及び方法
本研究で実験に使用された抗体は、IgG1型のヒト組換え抗体である。この抗体は、CHO細胞で発現され、アフィニティクロマトグラフィー及び様々なイオン交換クロマトグラフィー工程により精製された。
約22mgのIgG1抗体を、10mMトリス/HCl緩衝液(pH8.5)中で透析することにより再緩衝化した。再緩衝化された抗体溶液の一部を、40℃で30日間インキュベートした。他の部分は、対照として−80℃で凍結させた。
透析のために、1.5mlの抗体溶液(c=14.6mg/ml)を、スライドA−ライザー透析カセット(容量:0.5〜3ml、分子量排除サイズ:10kDa)に移し、透析緩衝液中に吊り下げた。透析中は緩衝液を連続的に撹拌し、約8℃に維持した。透析中、透析緩衝溶液を、数回、新しいのと取り換えることにより、緩衝溶液が可能な限り完全に交換されるようにした。表1は、緩衝溶液の交換の時間スキームを示す。
透析後、抗体溶液を、細菌レベルの低い条件下で1枚のMinisartシリンジフィルター(0.2μm、Sartorius)を通してろ過することにより滅菌した。抗体濃度の測定のためのサンプリングもまた、細菌レベルの低い条件下で行なった。
抗体濃度は、Uvikon XL型(Goebel Company)のスペクトルフォトメーターで280nmでの吸光度の測定により決定した。使用された抗体の消衰係数は1.55mL・mg−1・cm−1であり、Pace, C.N., et al., (Protein Sci. 4 (1995) 2411-2423)の方法に従って計算された。
抗体溶液は、Power Ease 300電気泳動ステーション並びにInvitrogen社のゲル、緩衝液、及び他の試薬を使用してゲル電気泳動により分離した。トリス−グリシン4〜20%勾配ゲルを分離に使用した。使用前に2倍の蒸留水で1:10(v/v)に希釈したトリス−グリシンSDS泳動緩衝液(10×)を泳動緩衝液として使用した。
Tosoh Bioscience CompanyのTSKゲルG3000SWXLゲルろ過カラム(7.8×300mm;5μm、250Å)を使用することにより、サイズに従って抗体溶液の成分をクロマトグラフィーにより分離した。分離は、島津−HPLCシステムで行なった。試料成分は、200mMのリン酸カリウム、250mMの塩化カリウム緩衝液(pH7.0)を用いて均一濃度で、30分間かけて流速0.5ml・分−1で溶出した。TSKゲルG3000SWXLゲルろ過カラムの温度は分離中25℃で一定に維持した。オートサンプラーの温度は6℃であった。試料成分は、ダイオードアレイ検出器を使用して280nmの波長で検出された。
Agilent Companyの弱カチオン交換カラムSynChropak WCX (4.6 x 250 mm, 6 μm, 300A)が、イオンクロマトグラフィーによる抗体溶液の成分の分離に使用された。分離は、島津HPLCシステムで行なった。試料成分は、溶出液A(10mMリン酸ナトリウム緩衝液、pH7.0)及び溶出液B(10mMリン酸ナトリウム緩衝液、750mM塩化ナトリウム緩衝液、pH7.0)の2勾配システムを用いて、流速1ml・分−1、泳動時間55分間で溶出された。溶出プロファイルを表2に示す。SynChropak WCXカチオン交換カラムの温度は、分離中、25℃で一定に維持した。オートサンプラーの温度は6℃であった。試料成分は、ダイオードアレイ検出器を使用して280nmの波長で検出された。
熱ストレス
モノクローナル抗体の分解産物及び改変体の形成を加速させるために、インキュベーター(Venticell, MM-Medcenter)中で所定の熱ストレスにかけた。インキュベーションのために、透析された抗体溶液を、40℃で30日間インキュベートした。更に、250μlの抗体溶液を10μlずつのアリコートに分け、−80℃で保存した。これらは、ゼロ点の対照として使用した。
後の分析で、40℃の保存から得られた抗体溶液において観察された変化が、緩衝溶液中での変化に起因しないことを確かめるために、2つの15mlアリコートの10mMトリス/HCl緩衝液を、細菌レベルの低い条件下でろ過することにより滅菌し、また40℃で30日間インキュベーター(Venticell, MM-Medcenter)中でインキュベートした。その後の過程は、抗体溶液と同様に行なった。
40℃でインキュベートし、−80℃で保存された抗体のゲル電気泳動による分離の結果を図4に示す。40℃でのインキュベーションにより、抗体分子の凝集物並びに抗体分子の断片が形成される(レーン6及び10)。インキュベートされていない抗体試料は、非還元条件下でIgG1分子の典型的なバンドパターンを示す(レーン4及び5を参照)。主なバンドは、2本の重鎖(HC)及び2本の軽鎖(LC)からなるインタクトな抗体に相当する。更に、構造が完全に保存されていない抗体種のバンドも見られる。これらのバンドは、1本の軽鎖を欠失した2HC/LC種、抗体の半分(HC/LC)、遊離重鎖(HC)、及び遊離軽鎖(LC)である(レーン4及び5を参照)。
サイズ排除クロマトグラフィー(SEC)により、ストレスにより誘発された断片及び凝集物の形成の概観が得られる。SECは天然条件下で行なわれるので、共有結合した凝集物並びに共有結合していない凝集物が分析で検出される。分解産物の形成と40℃でのインキュベーションの間の関連を確立するために、ストレスのかけられた抗体溶液に加えて、−80℃で保存されていたストレスのかけられていない試料も分析する。
イオン交換クロマトグラフィー(IEC)は、抗体分子の改変に起因する荷電の変化を検出するのに適している。
IdeSによる消化の最適化
酵素IdeSは、50mMトリス/HCl緩衝液(pH8.0)中に1mg/mlの濃度で存在していた。3つの異なるIdeS濃度を含む希釈シリーズを、消化溶液への酵素の添加のために調製した。希釈前溶液(V)中のIdeSの最終濃度は、0.1mg・ml−1(V1)、0.01mg・ml−1(V2)、及び0.001mg・ml−1(V3)であった。希釈は、50mMトリス/HCl緩衝液(pH8.0)を使用して行なった。
様々な試料調製法の比較
還元のみ:
43μgの抗体を、0.43Mのリン酸トリクロロエチル(H2O中0.5M)を用いてc=1mg・ml−1の濃度で還元した。この目的のために、38μlのTCEP溶液(H2O中0.5M)を、5μlのストレスのかけられていない抗体溶液(c=8.55mg・ml−1)に加え、37℃で30分間インキュベートした。その後、抗体溶液を、1:2(v/v)の比率でギ酸(1%、v/v)を用いて希釈した。ギ酸で希釈した後、抗体の濃度は、0.5mg・ml−1であった。ギ酸の比率は、0.5%(v/v)であった。得られた溶液を、13,400rpmで2分間、遠心分離にかけた(Minispin, Eppendorf)。上清をピペットを用いて注意深く取り出し、分析チューブに移した。4μl(2μg)の抗体をLC/MS実験のカラムにアプライした。
IdeSによる消化は、43μgの抗体を1mg・ml−1の濃度でインキュベートすることにより行なわれた。この目的のために、5μlのストレスのかけられていない抗体溶液(c=8.55mg・ml−1)を、34μlの50mMトリス/HCl緩衝液(pH8.0)で希釈し、4μlのIdeS溶液(50mMトリス/HCl緩衝液(pH8.0)中0.25mg・ml−1)を加え、37℃で2時間インキュベートした。その後、0.25MのTCEP(H2O中0.5M)を用いて0.5mg・ml−1の濃度で還元した。この目的のために、35μlの消化抗体溶液(c=1mg・ml−1)を、35μlのTCEP溶液(H2O中0.5M)に加え、37℃で30分間インキュベートした。その後、抗体溶液を、1:2(v/v)の比率でギ酸(1%、v/v)を用いて希釈した。ギ酸で希釈した後、抗体の濃度は、0.25mg・ml−1であった。ギ酸の比率は0.5%であった。得られた抗体溶液を、13,400rpmで2分間遠心分離にかけた(Minispin, Eppendorf)。上清をピペットを用いて注意深く取り出し、分析チューブに移した。8μl(2μg)の抗体を、LC/MS実験のカラムにアプライした。
LC−MS分析法
抗体を、抗体の分解産物を特徴づけるためのLC/MS分析のために、まずIdeSで消化し、その後、脱グリコシル化し、その後、還元した。
IdeSによる消化のために、4μlのIdeS溶液(50mMトリス/HCl緩衝液中c=0.25mg・ml−1、pH8.0)を各々、7.9μlのストレスのかけられた抗体(c=6.26mg・ml−1)及び5.8μlのストレスのかけられていない抗体(c=8.55mg・ml−1)に加えた。この溶液を、抗体濃度が1mg・ml−1となるまで50mMトリス/HCl緩衝液(pH8.0)で希釈し、37℃で2時間インキュベートした。酵素:抗体の比は1:50であった。
抗体の重鎖の糖構造を、N−グリコシダーゼF酵素を用いて開裂除去した。N−グリコシダーゼで結合された糖は、IdeSによる消化後に、50mMトリス/HCl緩衝液(pH8.0)中0.5mg・ml−1の抗体濃度でインキュベーションすることにより開裂した。開裂は、0.5μl(活性:10U・ml−1)のN−グリコシダーゼFの添加により開始された。インキュベーションは37℃で4時間行なわれた。
IdeSにより消化し、60μlのTCEP(リン酸トリクロロエチル;H2O中0.5M)の添加により脱グリコシル化した後に、還元が行なわれた。その後、この混合物を1:2(v/v)の比でギ酸(1%、v/v)で希釈し、これにより、還元溶液中の抗体の濃度が0.16mg・ml−1となった。TCEPの濃度は0.1Mであり、溶液中のギ酸の比率は0.5%(w/v)であった。ギ酸及びTCEPの両方の存在下で37℃で30分間インキュベーションを行なった。その後、得られた抗体溶液を、13400rpmで2分間遠心分離にかけた(Minispin, Eppendorf)。上清をピペットにより注意深く取り出し、分析チューブに移した。13μl(2μg)の抗体を、LC/MS実験のカラムにアプライした(図8)。
IdeSにより消化し、60μlのTCEP(リン酸トリクロロエチル;H2O中0.5M)の添加により脱グリコシル化した後に、還元が行なわれた。その後、この混合物を1:2(v/v)の比でギ酸(1%、v/v)で希釈し、これにより、還元溶液中の抗体の濃度が0.16mg・ml−1となった。TCEPの濃度は0.1Mであり、溶液中のギ酸の比率は0.5%(w/v)であった。ギ酸及びTCEPの両方の存在下で70℃で10分間インキュベーションを行なった。その後、得られた抗体溶液を、13400rpmで2分間遠心分離にかけた(Minispin, Eppendorf)。上清をピペットにより注意深く取り出し、分析チューブに移した。13μl(2μg)の抗体を、LC/MS実験のカラムにアプライした(図9)。
調製物の逆相(RP)分離が、ミクロ脱気装置、キャピラリーポンプ、ミクロオートサンプラー(温度制御ユニット、カラムオーブンを含む)、及び多波長検出器を備えた、Agilent 1100 Cap-LCシステムで行なわれた(Agilent, Waldbronn)。粒子サイズ3.5μm、孔サイズ300Å、寸法1.0×250mmを有するJupiter C18カラム(Phenomenex, Aschaffenburg)が標準的な分離に使用され、これにより参照基準点が示される。クロマトグラフィー分離は、75℃、流速40μl・分−1で行なわれた。各々の調製物において2μgの抗体がカラムにアプライされた。試料成分は、溶出液A(H2O中0.5%ギ酸(v/v))から溶出液B(70%の2−プロパノール、20%アセトニトリル、9.5%H2O、及び0.5%ギ酸(v/v))へと混合された二相勾配を用いて溶出された。表7は、溶出に使用された勾配のプロファイルを示す。
液体クロマトグラフィーにより分離された溶出液のオンラインESI−TOF質量分析を、電子スプレーイオン源を備えたmicromass LCT質量分析計(Waters, Eschborn)で行なった。データは、80℃のコーン温度及び100℃の脱溶媒和温度でポジティブモード(ES+)で600〜2000amu(原子質量単位、atomic mass units)の質量範囲で記録された。キャピラリー電圧は3000Vであり、コーン電圧は30Vであり、RFレンズ電圧は400Vであり、抽出コーン電圧は1Vであった。ESI−TOF質量分析における他のパラメーター設定は表8に要約されている。
Pursuitジフェニルカラムのマススペクトルと、Jupiter C18カラムのマススペクトルの比較
Jupiter C18カラムと同様に、Pursuitジフェニルカラムでは、ピーク1のプレショルダーにおける酸化HC−Fc種(23778Da)の部分的な分離しかできなかった(図8参照)。また、抗体の軽鎖のピログルタメート種でも分離の向上は全く達成されなかった。Jupiter C18カラム並びにPursuitジフェニルカラムではピーク3aにおいて同等な分解能で溶出された。HC−Fab断片(アミノ酸(AA)1〜220)もまた2つのカラムで同時に溶出され、インタクトなHC−Fab種は溶出プロファイルの最後のピークで溶出された。従って、この断片を、Pursuitジフェニルカラムを使用した分離で別個のピークとして分離することは不可能であった。Jupiter C18カラムのマスプロファイルと、Pursuitジフェニルカラムのマスプロファイルの比較により更に、Pursuitジフェニルカラムのピーク3bの質量は、Jupiterカラムのピーク4と相関していることが示された。両方が33630Daの質量を示し、これは、IdeS酵素に割り当てられ得る。更に、Pursuit DPカラムのピーク5は、Jupiter C18カラムのピーク5と相関している。両方の場合において、チオエーテルを含むHC−Fab−LC複合体の質量(48918Da)、不完全に還元されたHC−Fab断片の質量(25377Da)、及びN−グリコシダーゼFの質量(34783Da)が存在している。
Claims (22)
- 試料中の抗体及び抗体断片を検出するための方法であって、以下の工程:
a)抗体及び/又は抗体断片を含む試料を得る工程、
b)a)で得られた試料を、
i)IgG特異的システインプロテアーゼ、
ii)グリコシダーゼ、及び
iii)還元剤
と共にインキュベートする工程、
c)b)でインキュベートされた試料を、液体クロマトグラフィーと接続された質量分析計により分析することにより、a)で得られた試料中に含まれるインタクトな抗体を検出及び/又は抗体断片を検出する工程、
を含むことを特徴とする、該方法。 - IgG特異的システインプロテアーゼは、IdeSであることを特徴とする、請求項1記載の方法。
- IgG特異的システインプロテアーゼは、配列番号1のアミノ酸配列を有することを特徴とする、請求項1記載の方法。
- IgG特異的システインプロテアーゼとのインキュベーションは、pH5.5〜8.5のpH範囲で行なわれることを特徴とする、請求項1〜3のいずれか1項記載の方法。
- pH範囲は、pH6.5〜8.5であることを特徴とする、請求項4記載の方法。
- pH範囲は、7.0〜8.0であることを特徴とする、請求項5記載の方法。
- IgG特異的システインプロテアーゼと、試料中に含まれる抗体及び/又は抗体断片のモル比は、1:25から1:2500であることを特徴とする、請求項1〜6のいずれか1項記載の方法。
- モル比は、1:25から1:100であることを特徴とする、請求項7記載の方法。

- 液体クロマトグラフィーは、逆相液体クロマトグラフィーであることを特徴とする、請求項1〜15のいずれか1項記載の方法。
- 逆相クロマトグラフィーは、C8基又はC18基を有するクロマトグラフィー材料を使用することを特徴とする、請求項16記載の方法。
- 液体クロマトグラフィーは、疎水性相互作用クロマトグラフィーであることを特徴とする、請求項1〜14のいずれか1項記載の方法。
- 疎水性相互作用クロマトグラフィーは、ジフェニル基を有するクロマトグラフィー材料を使用することを特徴とする、請求項18記載の方法。
- 試料中の抗体又は抗体断片を検出するための化膿性連鎖球菌(Streptococcus pyogenes)由来のIgG特異的システインプロテアーゼIdeSの使用であって、該試料を、IgG特異的システインプロテアーゼと共にインキュベートし、そして、フラボバクテリウム・メニンゴセプチカム由来のN−グリコシダーゼFと共にインキュベートした後、得られた断片を、液体クロマトグラフィーに接続された質量分析計により分析することを特徴とする、該使用。
- 抗体又は抗体断片を検出するためのキットであって、該キットは、
i)化膿性連鎖球菌由来のIgG特異的システインプロテアーゼIdeS、及び
ii)フラボバクテリウム・メニンゴセプチカム由来のグリコシダーゼであるN−グリコシダーゼF
を含むことを特徴とする、該キット。 - 試料中の改変型の抗体を検出する方法であって、以下の工程:
a)抗体及び/又はその開裂産物を含む試料を得る工程、
b)a)で得られた試料を、
i)IgG特異的システインプロテアーゼ、
ii)グリコシダーゼ、
iii)還元剤
と共にインキュベートする工程、
c)b)でインキュベートされた試料を、疎水性相互作用クロマトグラフィーにより分析し、これにより、試料中の改変型の抗体を検出する工程
を含むことを特徴とする、該方法。
