Abstract

Description
本発明は、IL−13とそのレセプターおよびレセプター成分との相互作用の拮抗による、線維症の処置および阻害に関する。
インターロイキン−13(IL−13)を含む、サイトカインとして公知の種々の調節分子が、同定されている。IL−13の種々のタンパク質形態およびIL−13活性の種々の形態をコードするDNAは、McKenzieら、Proc.Natl.Acad.Sci.USA90:3735(1993);Mintyら、Nature362:248(1993);およびAversaら、WO94/04680に記載される。従って、用語「IL−13」は、これらの文献に記載される配列および/または生物学的活性を有するタンパク質を含み、このIL−13は、遺伝子組換え技術によって産生されるか;天然で因子を産生する細胞源から、または他の因子での誘導により精製されるか;化学技術によって合成されるか;あるいは上記の組み合わせである。
本発明に従って、インターロイキン−13レセプターのIL−13結合鎖をコードするポリヌクレオチドが開示され、これには、マウスレセプターおよびヒトレセプター由来のポリヌクレオチドが挙げられるが、これらに限定されない。特定の実施形態では、本発明は、以下からなる群から選択されるヌクレオチド配列を含む単離されたポリヌクレオチドを提供する:
(a)配列番号1のヌクレオチド256〜ヌクレオチド1404のヌクレオチド配列;
(b)配列番号3のヌクレオチド103〜ヌクレオチド1242のヌクレオチド配列;

(d)(a)または(b)において特定されるヌクレオチドに対して、ストリンジェントな条件下でハイブリダイズし得るヌクレオチド配列;
(e)(a)または(b)において特定される配列の種ホモログをコードするヌクレオチド配列;および
(f)(a)または(b)において特定されるヌクレオチド配列の対立遺伝子改変体。

本願の発明者らは、IL−13RのIL−13結合鎖(以後「IL−13bc」)をコードするポリヌクレオチド(マウスおよびヒトのIL−13bcをコードするポリヌクレオチドを含むが、限定はされない)を初めて同定および提供した。
(IL−13bc cDNAの単離)
(マウスIL−13レセプター鎖の単離)
5μgのポリA+RNAを、6〜8週齢のC3H/HeJマウスの胸腺から調製した。二本鎖のヘミメチル化cDNAを、StratageneのcDNA合成キットを製造業者の説明書に従って用いて調製した。手短に述べると、第1鎖を、オリゴdT−Xhoプライマーを用いてプライム(prime)し、そして第2鎖合成の後、EcoRIアダプターを添加し、そしてcDNAをXhoIで消化し、そして精製した。cDNAを、Zap Express(Stratagene)λベクターのXhoI−EcoRI部位に連結し、そしてGigapak II Goldパッケージング抽出物(Stratagene)を製造業者の説明書に従って用いてパッケージングした。1.5×106の得られた組換えファージのライブラリーを、製造業者の説明書に従って増幅した。このライブラリーを、配列KSRCTCCABK CRCTCCA(配列番号5)(K=G+T;S=C+G;R=A+G;B=C+G+T)の縮重17マーオリゴヌクレオチドプローブを用い、記載された(Current Protocols in Molecular Biology、Ausubelら編、John Wiley and Sons、1995、6.4.3節)通りの標準的なTMACハイブリダイゼーション条件を用いてスクリーニングした。クローンA25が同定された。なぜなら、クローンA25はこの17マープローブにハイブリダイズするが、公知の造血素レセプター由来のプローブにはハイブリダイズしないからである。このクローンを、製造業者の説明書に従ってZapExpressベクターからプラスミド形態で単離し、そしてDNA配列を決定した。DNA配列は、造血素レセプターファミリーの新規なメンバーをコードしていた。
マウスレセプターのヒトホモログの部分的フラグメントを、マウス配列に由来するオリゴヌクレオチドを用いたPCRによって単離した。cDNAを、Clontechから得たヒト精巣ポリA+RNAから調製した。274塩基対のDNAフラグメントを、30サイクルのインキュベーション(94℃×1分間、42℃にて1分間および72℃にて1分間)について1.5mM MgCl2を含む1×Taq緩衝液中で、以下のオリゴヌクレオチドを用い、AmpliTaqポリメラーゼ(Promega)を用いたPCRによってこのcDNAから増幅した:ATAGTTAAACCATTGCCACC(配列番号6)およびCTCCATTCGCTCCAAATTCC(配列番号7)。このフラグメントのDNA配列を決定し、そして2つのオリゴヌクレオチドを、以下の配列を用いてこのフラグメントの内部部分から調製した:AGTCTATCTTACTTTTACTCG(配列番号8)およびCATCTGAGCAATAAATATTCAC(配列番号9)。これらのオリゴヌクレオチドをプローブとして用いて、CLONTECH(カタログ番号HL1161)から購入したヒト精巣cDNAライブラリーをスクリーニングした。フィルターを、標準的な5×SSCハイブリダイゼーション条件を用いて52℃にてハイブリダイズし、そして2×SSC中で52℃にて洗浄した。400,000クローンのスクリーニングにおいて両方のオリゴヌクレオチドにハイブリダイズする22個のクローンを単離した。これらのcDNAクローンのうちの4つのDNA配列を決定し、そして全てが同じ新規な造血素レセプターをコードしていた。全長ヒトレセプター鎖の推定されるDNA配列を配列番号3として示す。
(可溶性IL−13bcタンパク質の発現および活性のアッセイ)
(可溶性IL−13bc−Igの産生および精製)
マウスIL−13bcの細胞外ドメインのアミノ酸1〜331をコードするDNAを、gly−ser−glyをコードするスペーサー配列にPCRによって融合し、そしてCOS−1発現ベクターpED.FcのヒトIgG1のヒンジCH2 CH3領域をコードする配列とインフレームで連結した。IL−13bc−IgをDEAE−デキストラントランスフェクトCOS−1細胞から産生し、そしてプロテインAセファロースクロマトグラフィー(Pharmacia)によって精製した。
IL−13またはIL−4に応じたB9細胞(Aardenら、Eur.J.Immunol.1987.17:1411−1416)の増殖刺激を、DNA中への3H−チミジン取り込みによって測定した。細胞(5×103/ウェル)を、1μg/mlのIL−13bc−Igの存在下または非存在下で種々の濃度の増殖因子を含む培地を有する96ウェルプレートに播種した。3日間のインキュベーション後、1uCi/ウェルの3H−チミジンを添加し、そして細胞をさらに4時間インキュベートした。取り込まれた放射能を、LKB 1205 Plateリーダーを用いて決定した。
(表面プラズモン共鳴(Biacore分析)によって測定した、IL−13に対する可溶性IL−13bcの直接結合)
Biacoreのバイオセンサーを用いて、精製されたIL−13bc−Ig(Pharmacia、Johnssonら、1991)に対するIL−13の特異的結合を直接測定した。約10,000〜17,000の共鳴単位(RU)の精製されたIL−13bc−Ig、ヒトIgG1または無関係のレセプターを各々、製造業者によって推奨されるようにセンサーチップ上の異なるフローセルに共有結合によって固定した。(RUは、センサーチップ表面に結合したタンパク質の質量を反映するもの(refelction)である。)精製されたIL−13を、過剰の精製IL−13bc−Igの存在下または非存在下で10分間にわたって5μl/分でフローセルと交差して注入した。結合を、サンプル注入の前と後とのRUの差として定量した。481.9RUの特異的IL−13結合は、固定化IL−13bc−Igについてのみ観察され、一方、IL−13+IL−13bc−Igの結合体化は、固定化IL−13bc−Igに対する結合がないことをもたらした(4RU)。IL−13結合は、固定化IgGまたはIL−11R−Igのいずれについても観察されなかった(それぞれ、5.4RUおよび3.7RU)。
(標識IL−13BC−Ig融合タンパク質に対する、COS細胞中で発現されたIL−13の結合:IL−13bc−Fcを用いたIL−13のCOSインサイチュ検出)
IL−13、IL−4、IL−11についての発現ベクターまたは空のベクターを、DEAE−デキストラン方法によって二連のプレート中のCOS−1細胞中にトランスフェクトした。トランスフェクションの2日後、細胞をリン酸緩衝化生理食塩水(PBS)中で2回洗浄し、そして4℃にてメタノールで10分間、培養ディッシュ中で固定した。固定後、細胞をPBSで2回洗浄し、次いで結合緩衝液(PBS、1%(w/v)ウシ血清アルブミン、0.1%(w/v)アジ化ナトリウム)で1回リンスし、そして1.0ug/mlでIL−13bc−Fcを有するか関連する抗サイトカイン抗血清を有する結合緩衝液中で4℃で2時間インキュベートした。細胞を、PBSで2回洗浄し、そして結合緩衝液中に500倍希釈したアルカリホスファターゼ標識ウサギF(ab)2’抗ヒトIgG(Fc融合体検出のため)またはウサギF(ab)2’抗ラットIgG(抗サイトカイン検出のため)中で振盪しながら4℃でインキュベートした。細胞をさらに、PBS中で2回洗浄した。アルカリホスファターゼ活性を、ニトロブルーテトラゾリウム(nitro blue tetrazolium)および5−ブロモ−4−クロロ−3−インドリル−ホスフェートを用いて可視化した。
(IL−13bcタンパク質の生物学的活性を決定するための他の系)
他の系を用いて、特定のIL−13bcタンパク質が、本明細書中で定義される通りのIL−13bcの「生物学的活性」を示すか否かをを決定し得る。以下は、このような系の例である。
IL−13bcタンパク質がIL−13またはそのフラグメントを結合する能力は、このような結合を検出し得る任意の適切なアッセイによって決定され得る。いくつかの適切な例が以下に続く。
酵母の遺伝的選択方法である「相互作用捕捉(interaction trap)」[Gyurisら、Cell 75:791−803,1993]を用いて、IL−13bcタンパク質が、本明細書中で定義したとおりのIL−13bcの生物学的活性を有するか否かを決定し得る。この系では、LexAop−Leu2およびLexAop−LacZの両方からのレポーター遺伝子の発現は、ベイト(bait)タンパク質(例えば、この場合、ヒトIL−13bcと相互作用する種)と、プレイ(prey)(例えば、この場合、ヒトIL−13bcタンパク質)との間の相互作用に依存する。従って、Leu2発現またはLacZ発現のレベルによって相互作用の強さを測定し得る。最も単純な方法は、LacZコードタンパク質であるβ−ガラクトシダーゼの活性を測定することである。この活性は、X−Gal含有培地またはフィルター上の青色の程度によって判断され得る。β−ガラクトシダーゼ活性の定量的測定について、標準的なアッセイは、「Methods in Yeast Genetics」、Cold Spring Harbor、New York、1990(Rose,M.D.、Winston,F.およびHieter,P.による)に見出され得る。
(可溶性IL−13Rを用いた線維症の阻害)
線維性組織の発達は、損傷後の治癒の正常なプロセスの一部である。それにもかかわらず、いくつかの状況では、罹患組織の正常な機能を妨害する、過剰のコラーゲンの破壊的蓄積が存在する。実際、コラーゲン合成およびおよび組織瘢痕形成(tissue scaring)は、いくつかの自己免疫、アレルギー性疾患および感染性疾患を含めた多数の慢性および消耗性の病気の主な病理学的症状発現である1-7。瘢痕組織形成のプロセスに関する機構のたくさんの情報が存在する8、9が、線維性プロセスの開始における炎症細胞およびサイトカインの役割の理解にはなお大きな隔たりが存在する。
(住血吸虫症病因における、IL−4、IL−13またはIL−4/IL−13二重欠損の匹敵する効果:sIL−13Rα2−Fc処置は、S.mansoni感染マウスにおける肝臓線維症を有意に減少させる)
住血吸虫症の病因におけるIL−4およびIL−13の調節の役割を比較するために、本発明者らは、C57BL/6WTマウスおよびIL−4欠損マウスを25のS.mansoni cercariaeに経皮的に感染させた。別個の群の動物を、「材料および方法」に記載されるように、sIL−13Rα2−Fcまたはコントロール−Fcのいずれかで処置した。処置を産卵開始時である5週目に始め、そして全ての動物を感染8週後に屠殺し、そしていくつかの寄生生物学的および免疫学的パラメーターについて調べた。表IIIに示すように、4群のマウスは全て、類似した寄生生物負荷を宿しており、そして1対の寄生生物あたりに産生される組織卵は群の間でそれほど異ならなかった。ピーク組織応答の時点である45感染8週間後に、WTマウスは、IL−13遮断の結果として、肉芽腫の大きさにおける有意な変化を示さなかった(図2A)。興味深いことに、コントロール−Fc処置IL−4欠損マウスはまた、減少した肉芽腫性応答を示すことができず、そして実際に、肉芽腫はこれらのマウスにおいて有意に大きかった。これらの観察とは著しく対照的に、IL−4欠損マウスは、IL−13が阻害された場合著しく減少した肉芽腫性応答を提示した(図2A、一番右)。実際、IL−4二重欠損/sIL−13Rα2−Fc処置マウスは、コントロールまたはsIL−13Rα2−Fc処置WT動物のいずれもと比較した場合、肉芽腫容量における平均40%〜50%の減少を提示し、そしてコントロール−Fc処置IL−4欠損マウスと比較した場合、75%を超える減少を提示した。
IL−4はCD4+Th2細胞発達を駆動する主なサイトカインであることが周知である21、22が、Th2型応答の生成および維持におけるIL−13の役割は、議論の余地があり、そして研究されている宿主の遺伝および感染性疾患モデルの両方によって影響を受け得る30、34、38。それゆえ、肝臓病理におけるsIL−13Rα2−Fc誘導性変化がTh1/Th2サイトカインバランスにおける変化によって生じたか否かを決定するために、本発明者らは、腸間膜リンパ節および脾臓を、感染させたマウスから単離し、単細胞懸濁物を調製し、そしてこの培養物を寄生生物抗原でインビトロで再刺激した。さらなる細胞培養物を、抗CD4 mAbの存在下で寄生生物抗原に曝露して、サイトカイン産生がCD4+T細胞応答に依存性であるか否かを決定した。培養上清を、IL−4、IL−13、IL−5、IL−10およびIFN−γについてELISAによって分析した。予測され得るように15、WTマウスから調製された腸間膜培養物(図5)および脾臓培養物(データは示さず)は、非常に偏ったTh2型サイトカイン応答を提示した。これらは、SEA刺激に応じて高レベルのIL−4、IL−5、IL−10およびIL−13を産生し、そしてIFN−γをほとんどまたは全く産生しなかった。対照的に、IL−4欠損マウスは、より混ざったTh1/Th2型プロフィールを示した。実際、有意なSEA特異的IFN−γ応答がIL−4欠損マウスにおいて検出された。これは、以前の研究23、24と一致する。IL−13、IL−10、そしてより低い程度でIL−5もまた、これらの動物において検出され、一方、これらのサイトカインのレベルは、WTマウスと比較した場合、顕著に減少した。重要なことに、低いがしかし有意なIL−4依存性IL−13応答の維持が、IL−4の非存在下で維持される著しい肉芽腫性応答を説明するようである(図2)。驚くべきことに、肝臓線維症に対するその顕著な阻害効果にもかかわらず、sIL−13Rα2−Fcは、WTマウスまたはIL−4欠損マウスのいずれでも、Th1型サイトカイン応答にもTh2型サイトカイン応答にも有意な影響を有さなかった。全ての場合、サイトカイン産生がCD4+T細胞応答に非常に依存していたことにもまた留意すべきである。なぜなら、抗CD4 mAb処置SEA刺激培養物のいずれにおいてもサイトカイン発現がほとんどまたは全く検出されなかったからである。
類似のパターンのサイトカイン発現が、肉芽腫形成部位でインビボで観察されるか否かを決定するために、本発明者らは、感染8週間後に種々の群のマウスから肝臓mRNAを単離し、そして定量的RT−PCRを行った。図5に示すように、感染したWTマウスは、強力なTh2型サイトカインmRNAプロフィールを示した。これは、IL−4、IL−13、IL−5およびIL−10のmRNAにおける顕著な増加を示す。WTマウスはまた、IFN−γ mRNAの発現における中程度の増加を示した。これは、以前の観察19と一致した。これらの知見とは対照的に、IL−13およびIL−5のmRNAレベルは、IL−4欠損マウスにおいてずっと低く、一方、IL−10およびTNF−αのmRNAは有意に増加し、そしてIFN−γ mRNA発現は変化しなかった。さらに、腸間膜リンパ節培養物および脾細胞培養物から得られたインビトロでの結果と同様に、IL−13遮断は、WTマウスまたはIL−4欠損マウスのいずれにもサイトカインmRNA発現に対して有意な影響を有さなかった。しかし、sIL−13Rα2−Fcで処置したIL−4欠損マウスにおけるIL−10 mRNAレベルには中程度の増加が存在したが、これは、線維症における減少を説明しないようである。なぜなら、IL−4欠損マウスと比較して非常に多様なレベルのIL−10がsIL−13Rα2−Fc処置WTマウスにおいて検出され、依然として線維症における類似の減少が観察されたからである。TGF−β1およびTGF−β2bのmRNA発現をまた、肉芽腫性組織において調べたが、感染したIL−4欠損マウスでもsIL−13Rα2−Fcで処置した動物でも有意差は観察されなかった(データは示さず)。
上記のインビトロおよびインビボでのサイトカイン研究は、sIL−13Rα2−Fcの抗線維症効果が、Th1型またはTh2型のサイトカイン発現における変化によって説明されないようであることを示唆した。それゆえ、その後の実験では、本発明者らは、コラーゲンI(Col I)およびコラーゲンIII(Col III)のmRNA発現パターンを調査して、sIL−13Rα2−Fc誘導性の線維症減少が、これらの2つの重要なコラーゲン産生遺伝子19における発現における直接変化を伴うか否かを決定した。図6に示すように、IL−13遮断は、Col IおよびCol IIIのmRNA発現をWTマウスおよびIL−4欠損マウスの両方で有意に減少させた。IL−4欠損マウスにおいてCol IまたはCol IIIのmRNAの感染誘導レベルにおける変化は存在せず、そしてsIL−13Rα2−Fc処置WTマウスと比較した場合、同様に処置したIL−4欠損マウスにおいてさらなる減少は存在しなかった。
IL−13遮断が、感染させたWTマウスおよびIL−4欠損マウスの肝臓におけるCol IおよびCol IIIのmRNA発現をインビボで有意に減少させることを示したので、本発明者らは、IL−13が線維芽細胞におけるコラーゲン合成を直接刺激するか否かを決定しようとした。この問題に答えるために、本発明者らは、マウス3T3線維芽細胞におけるI型コラーゲンの誘導を、ウェスタンブロッティングによって調べた。図7に示すように、IL−13は、刺激の48時間後にコラーゲン合成を誘導した。最小のI型コラーゲンが、未刺激の細胞において検出された(図7、レーン1)かまたはサイトカイン活性化培養物において、より早期の時点で検出された(データは示さず)。IL−4はまた、コラーゲンI合成を誘導し(レーン2)、そして高レベルの分泌コラーゲンが、サイトカインで刺激した両方の培養物において得られた上清において容易に検出可能であった(データは示さず)。反応の特異性を、精製したI型コラーゲンを用いて確認し(レーン5)、そして細菌のコラゲナーゼ処置は、抗体がコラーゲンについて特異的であることを示した(データは示さず)。
CD4+Th2型サイトカインパターンは、S.mansoniで感染されたマウスにおける免疫応答を支配する12、13。しかし、IL−4欠損マウスを用いた以前のIL−4除去の研究および実験は、住血吸虫症の病因におけるこのサイトカインの不可欠な役割を示すことができなかった15、23、24。実際は、線維症における部分的な減少がいくつかの研究で観察されたが15、卵細胞(egg)誘導肉芽腫形成は、IL−4が完全に無い中で進行し得た23、24。これらの知見と対照的に、肝線維症の肉芽腫形成および発達は、Stat6欠損マウスにおいて厳しく損傷しており16、これは、いくつかのTh2関連サイトカインの産生における主要な欠損を示した46。IL−4およびIL−13は両方ともStat6を介してシグナル伝達し、従って、感染されたIL−4欠損マウスとStar6欠損マウスとの間に観察される明らかな病理の差違は、IL−13によって説明され得る。それにもかかわらず、疾患進行におけるIL−4およびIL−13の異なる寄与は、Stat6欠損マウスまたはIL−4欠損マウスの単独研究からは認められ得なかった。本研究において、本発明者らは、感染されたWTおよびIL−4欠損マウスにIL−13の強力なインヒビターを使用し、IL−13およびIL−4が重複することならびに住血吸血病の病因における独特の役割を示すことを証明する。
(動物、寄生虫およびAg調製物)
6〜8週齢の雌のC57BL/6マウスおよびIL−4欠損マウス(C57BL/6背景、10代戻し交配)を、Taconic Farms,Inc.(Germantown,NY)から得た。全てのマウスは、研究動物の取り扱いの認定に関するNIH米国協会が承認した動物施設で、滅菌したフィルタートップのケージ中で飼育し、そして滅菌水で維持した。Schistosoma mansoniのPuerto Rican種のケルカリア(NMRI)は、感染させたBiomphalaria glabrata巻貝から得た(Biomedical Research Institute,Rockville,MD)。可溶性卵細胞抗原(SEA)は、以前に記載されるように15、ホモジナイズした卵細胞から精製した。
可溶性IL−13レセプターα2−Fc融合タンパク質(sIL−13Rα2−Fc)は、以前に記載されるように調製され35、そしてGenetics Institute,Cambridge MA.によって提供された。内毒素混入は、Cape Cod Associates LALアッセイ(Limulus Amebocyte Lysate,Woods Hole,MA)によって測定された場合<2EU/mgであった。B9増殖アッセイにおいて3ng/mlのマウスIL−13を中和する能力によって決定された場合、インビトロID50は、約10ng/mlであった。sIL−13Rα2−Fcに対するコントロールとして使用されたヒトIgG(コントロール−Fc)は、sIL−13Rα2−Fcに関して記載されるように35、組換えプロテインA−セファロースクロマトグラフィーによってアフィニティー精製された。以前に記載されたように、コントロール−Fcは、感染されたマウスにおける病理学またはサイトカイン発現に検出可能な効果を有さなかった30。
マウスは、20と25との間のケルカリアを含む水の中で、尾の皮膚に40分間経皮チャレンジをすることによって感染させた。動物は、卵細胞産生が発症した後(第5週)1日おきに、0.5ml PBS中、ヒトコントロールFcまたはsIL−13Rα2−Fcのいずれかを用い腹腔することによって処置した。インビボ使用のための最適な濃度(200μg/マウス/日)は、動力学的アッセイおよび感作/静脈内卵細胞注射されたマウスにおける用量応答実験に基づいて選択した30。血清は、屠殺した日にマウスから収集した。全ての動物は、第8週にフェノバルビタールナトリウムの腹腔内投与(18mg/マウス、Sigma、St.Louis,MO)によって屠殺し、そしてクエン酸生理食塩水を用いて灌流し、寄生虫負荷を評価した15。処置されたいずれの群においても死亡は観察されなかった。
肉芽腫の測定に関して、肝臓の約半分をBouin−Hollande固定液を用い、そして以前に記載されたように15進めて固定した。肝肉芽腫の大きさは、ライトギムザ染色(Histopath of America,Clinton,MD)によって染色された組織学的切片で測定された。単一の生存可能な卵細胞を含む肉芽腫の各直径は、接眼マイクロメータを用いて測定し、そして肉芽腫の各体積は、球形を想定して算出した。最も長い直径およびこれに対して垂直の直径の平均を使用した。好酸球、肥満細胞および他の細胞型のパーセンテージは、同じ切片中で評価した。実質ネクローシスは、0〜4のスコアで得点付けし、0は最も小さい広がりのネクローシスであり、4は最も大きい広がりのネクローシスであった。肥満細胞のこの頻度はまた、類似の尺度に対して割り当てられ、これは0〜4の範囲を用いた。肝臓および腸における住血吸虫卵細胞の数ならびに肝臓のコラーゲン含量(ヒドロキシプロリンとして測定される)は、以前記載されたように測定した15。線維症はまた、ピクロシリウスレッド(picrosirius red)を用いて染色された切片を用いて組織的に得点付けした。ピクロシリウス試薬は、コラーゲンを特異的に染色し、切片が偏光下で観察される場合、コラーゲンが沈着する明るい領域が照射される。各切片内の全ての肉芽腫は、尺度1〜4に基づいてピクロシリウス(レッド)「密度」に対して得点付けされ、「関連する領域」の第2の測定はまた、同じ尺度を用いて測定された。全線維症スコアは、各肉芽腫に対する密度と領域を乗算することによって決定された(すなわち、16のスコアが最大である)。マウス1匹あたり平均30個の肉芽腫が、全ての分析に含まれた。一貫性を制御するために、同じ個体が全ての組織学的特徴を得点付けされ、そして実験的な設計は全く認識していない。
各動物由来の肝臓の2つの部分を合わせて、1mlのRNA−STAT 60(Tel−Test)中に置き、ドライアイス上で凍結しそして使用するまで−70℃で保存した。組織ポリトロン(Omni International Inc.,Waterbury,CT)を用いて組織をホモジナイズし、そして全RNAを製造者らの推奨に従って抽出した。RNAをDEPC処理水に再懸濁し、分光光度的に定量化した。
RT−PCR手順を使用して、IL−4、IL−5、IL−10、IL−13、IFN−γ、コラーゲンI、コラーゲンIII、TGFβl、TGFβ2およびHPRT(ヒポキサンチン−グアニンホスホリボシルトランスフェラーゼ)のmRNAの相対量を測定した。cDNAは、記載されるように14、1μgのRNAの逆転写後に得た。全ての遺伝子のプライマーおよびプローブは、以前に公開された14、19、54。各サイトカインに対して使用したPCRサイクルは、以下のようであった:IL−4(33)、IL−5(31)、IFN−γ(29)、コラーゲンI(26)、コラーゲンIII(22)、TGFβl(34)、TGFβ2(34)およびHPRT(23)。
増幅されたDNAを、以前に記載されるように14、電気泳動、サザンブロットおよび非放射性サイトカイン特異的プローブを用いたハイブリダイゼーションによって分析した。PCR産物は、ECL検出系(Amersham)を用いて検出した。化学発光シグナルは、平らな土台のスキャナー(Microtek model 600 ZS,Torrance,CA)を用いて量を測定した。PCR産物の量は、個体サンプルにおける、HPRT特異的シグナル密度に対するサイトカイン特異的シグナル密度の比を比較することによって決定した。引き続いて、個体サンプルに関する任意の濃度測定単位に100という因数を掛けた。
腸間膜リンパ節(MLN)細胞および脾臓を、マウスから抽出し、そして単一細胞懸濁液を調製した。赤血球を、ACK溶解緩衝液(Biofluids,Inc.,Rockville,MD)を用いた浸透処理によって溶解した。細胞を、37℃、5% CO2で、10% FCS、2mMグルタミン、100U/mlペニシリン、100μg/mlストレプトマイシン、25mM HEPES、1mM ピルビン酸ナトリウム、0.1mM非必須アミノ酸、および50μM 2−MEを補充したRPMI 1640培地に置いた。細胞を、24ウェルプレート(3×106/ml,lml)に置き、SEA(20μg/ml)で刺激し、そして上清をIL−4、IL−5、IL−10、IL−13およびIFN−γのレベルを測定するために72時間後に収集した。さらなるSEA刺激された培養物はまた、50μg/mlの抗−CD4 mAb(GK1.5)を用いて処理した。抗−CD4 mAb単独で処置した培養物は、コントロール培地の培養物で観察されたものと比較した場合、サイトカイン発現に変化を見せなかった(データには示さない)。IL−5、IL−10およびIFN−γは、特定のサンドイッチELISAを用いて測定した15。IL−13レベルは、マウスIL− 13ELISAキット(R&D Systems,Minneapolis,MN)を用いて測定した。サイトカインレベルは、組換えサイトカインを用いて作成されたカーブから計算した。IL−4は、IL−4感受性細胞株CT.4Sを用いて測定した。これらの細胞の増殖は、(3H)TdR取り込みによって量を測定し、そしてサイトカインの量は、既知の量の組換えIL−4を用いた比較によって測定した。
3T3線維芽細胞を、37℃、5%CO2で、10% FCS、2mM グルタメート、100U/mlペニシリン、100μg/ml ストレプトマイシン、25mM HEPES、1mM ピルビン酸ナトリウム、0.1mM 非必須アミノ酸、および50μM 2−MEを補充したRPMI1640培地で培養した。コンフルエントの細胞を、24ウェルプレート(500,000細胞/ml)にプレートし、そしてIL−4(1000U/ml)またはrIL−13(R&D Systems,Minneapolis,MN)(20ng/ml)を用いて6、24および48時間刺激した。培養上清を収集し、分泌されたコラーゲン1を分析した。細胞を、リン酸緩衝化生理食塩水で一度洗浄し、そしてSDS−PAGEサンプル緩衝液に溶解した。細胞溶解液および培養上清を、還元状態を使用する6%トリス−グリシンゲル(Novel Experimental Technology,San Diego,CA)中の電気泳動分離に供し、そしてニトロセルロース膜(Schleicher & Schuell,Keene,NH)に移した。ブロットは、ウサギIgG抗マウスI型コラーゲン(Biodesign International,Kennenbunk,ME)を用いてプローブ化し、そしてペルオキシダーゼ標識抗ウサギIgG(Amersham Pharmacia Biotech,Inc.,Piscataway,NJ)を2次Abとして使用した。バンドは、ウエスタンブロット化学発光試薬を用いて可視化した(NEN Life Science Products,Boston,MA)。コラーゲンバンドの同一性を確認するために、細胞溶解液を、PBS(1mM CaCl2および1%FCSを補充する)中の0.5 mg/mlのコラゲナーゼ(Boehringer Mannheim,Indianapolis,IN)を用いて、37℃で1時間処理した。精製されたラットコラーゲンI調製物をまた、コントロールとして使用した。
住血吸虫および卵細胞の数、サイトカインmRNAの変化、およ分泌されたサイトカインタンパク質の値を、スチューデントの2テールの(two−tailed)t−検定を用いて比較した。肝線維症は、共分散の分析によって比較し、これは、卵細胞1個あたりの、共分散(covariate)としての全肝臓卵細胞のログおよびヒドロキシプロリンのログを使用した。p<0.05が有効であると考えられた。
