Abstract
【解決手段】圧搾した脱脂オイルシード10を、中性、弱塩基性または弱酸性のpHの水で抽出し濾過する。濾過材によって、固体細胞ミートの小断片は濾液中に通過することができる。濾液を塩基で処理することによりpH9を超えるpHに高め、固体を分離する。タンパク質を、例えば熱誘発沈殿または等電点沈殿および/または限外濾過によって液体の他の成分から分離し濃縮する。残った液体は、糖質が豊富である。この方法によって、人や動物の食品成分または化粧品製造に適した1種類以上のタンパク質生成物、発酵または飼料成分の使用に適した高糖質生成物、および繊維性タンパク質の動物飼料成分が得られる。

Description
第I期では、溶媒除去した脱脂オイルシードをpH3〜9、好ましくはpH7からpH9までのpHで水中にてスラリー化する。該脱脂オイルシードを水と完全に混合する。混合温度は重要ではなく、周囲温度以上であるのが好ましい。50℃〜95℃の温度まで、混合物を若干加熱するのが特に好ましい。それは、この加熱が脱脂オイルシードの一部成分の溶解度を高めるからである。簡便法として、加熱は、水を予め加熱した後に脱脂オイルシードに添加することで実施できる。該混合物は、確実に完全混合するように攪拌する。攪拌時間は、重要ではないが、例えば5分間から2時間までの時間が適している。この後、スラリーを濾過し、大半の固体材料を液体から分離する。簡便法として、例えばベルトプレスでプレスした後、希望すれば、(パルプ除去した)分離液体をさらに濾過して固体をさらに除去することによって、これを実施できる。濾過によって、液体抽出物と固体残渣(「プレスケーキ」として知られている)が得られる。希望すれば、プレスとパルプ除去を多段階的に実施し、液体‐固体分離を高めることができる。
第I期処理から得た抽出物(小粒子の細胞ミートを含有するのが好ましい)をアルカリ物質で処理し(本明細書では、第II期処理と呼ぶ)、pHをpH9を超えるpHに調整する。好ましいpHは10.5〜11.5である。
第III期は、第II期固体の処理のための任意の方法である。
第IV期は、第II期液体用の任意の処理である。第II期処理からの液体分画は、タンパク質が豊富であるので、処理すればそのタンパク質を回収することができる。数種類の任意の方法を使用できる。ある適切な方法は限外濾過であって、低分子量化合物をフィルターから通過させるが、タンパク質は保持することができる。他の好ましい方法はタンパク質沈殿である。タンパク質沈殿の好ましい方法は、熱誘発凝固、すなわち液相を70℃〜120℃、好ましくは90℃〜110℃の温度までタンパク質のアミノ酸構成要素が破壊しない程度の短い時間加熱 (例えば、95℃、約5分間) するか、あるいは等電点沈殿、すなわち希薄な酸を緩徐に添加して液体のpHを該液体中の主要タンパクの等電pHに接近させるかであり、これらは溶液からタンパク質を除去するために当分野では公知である。タンパク質沈殿の場合、沈殿しなかった可溶性タンパク質を更に除去するためには、沈殿後に限外濾過段階を利用するのがやはり好ましい。
カナダサスカチュワンのオイルシード圧搾業者からヘキサン含有オイル抽出済みカノーラ白色フレーク25kgを得た。ヘキサンが溶媒検出器で検出できなくなるまで、原料中ヘキサンを室温で蒸去して溶媒除去白色フレークを得た。溶媒除去白色フレークをローラーで粉砕し、大きな塊を潰し、均質な抽出用出発原料を作成した。次に、予め50℃まで加熱しておいた水75kgと該原料を混合し、CaOの10%スラリー1.7Lを該混合物に添加した。材料を均一な堅さになるまでリボンミキサー中で混合した。該混合物のpHを測定し、8.0であることを確認した。その後、該材料をリボンミキサー中で10分間混合した。
ベルトプレス3回通過からのパルプ除去抽出物(第I期液体)を100L蒸気釜に入れ、CaOの10%スラリー1.7Lを該抽出物に加えた。抽出期中、抽出物の温度は室温まで下降した。CaO添加後の室温での抽出物pHは11.0であった。釜への蒸気流は、該抽出物温度が50℃に上昇するまで回転した。釜中を30分間定速攪拌しながら、該抽出物を50℃に維持した。
実施例1で作成した第II期固体を、第III期処理を行うことを希望する日まで、保存、凍結した。しかし、希望すれば、第III期処理は、第II期直後に実施できる。
実施例2の抽出物(第II期液体)の遠心分離から得た上清を合わせ、100L蒸気釜に入れた。釜に通す蒸気は、抽出物の温度が95℃に達するように供給した。95℃の温度を5分間維持した後、冷水を蒸気釜のジャケットに通した。冷水を 20分間流した。この加熱およびその後の冷却処置の間に、抽出物の最上部にタンパク沈殿またはカードが生じた。次に、蒸気釜の内容物を、登録商標ナイテックス(グレートウェスタンマニュファクチャリング・カンパニー社)として販売されているナイロンメッシュの口径200ミクロンの篩に注いだ。液体が篩を通過している間に、篩にカードを集積し、容器に集めた。

- 脱脂オイルシードから得られる生成物であって、
少なくとも50乾燥重量%タンパク質と、1乾燥重量%未満フィチン酸塩とを含み、タンパク分散指数が5%未満であることを特徴とする組成物。
