Abstract
Description
本願は、2007年6月13日に出願された米国仮特許出願第60/934,420号の利益を主張する。上記出願の全教示は、参照によって本明細書に援用される。
科学者らは、腫瘍細胞および病理組織構築物(例えばテラトカルシノーマ)が初期胚の細胞および組織に類似していることを認識している。正常であるが未分化の胚性幹細胞は、細胞数の急速な増加および器官原基への分化および続く器官形成を論理的に含む必要がある未定義のプロセスによって器官を生じることができた。悪性腫瘍は、初期胎児と類似した速度で増殖し、正常組織の組織学的外見と共に組織学的に未分化または組織化したニッチを含む。
幹細胞集団内の変異は、癌幹細胞を生じるために必要とされる。19世紀の病理学的観察に由来する現代の「腫瘍幹細胞」仮説は、腫瘍が正常幹細胞および胎児組織の両方と類似した性質を有する特有のサブセットの細胞を含むことを前提としている。これらの細胞は、非対称および非有糸分裂性回転の細胞分裂によって広範に増殖する。この集団の細胞は、おそらくは自己再生し、ほとんどの腫瘍の増殖および維持に重要である。従って、腫瘍幹細胞分裂の阻害は、現在のアプローチよりも改善された有効性を約束する新規治療介入を表すと広く考えられる。
本発明は、腫瘍幹細胞、例えば分裂し腫瘍をもたらす細胞が釣鐘形核を特徴とし、特有の形態の複製を起こすという発見に関するものである。釣鐘形核は、腫瘍組織を除いて、成体組織でごくまれに見られ、または非常に減少した数で見られ、ゲノムは単鎖DNA(ssDNA)として表わされる期間を経る。釣鐘形核は、結腸および膵臓ヒト腫瘍中の非有糸分裂プロセスによって対称および非対称の両方で分裂する(Gostjevaら、Cancer Genetics and Cytogenetics, 164:16-24 (2006); Gostjevaら、Stem Cell Reviews, 1 : 243-252 (2005))。これらの釣鐘形核は、釣鐘形核が管状構造の合胞体に包まれ、核全体の30%を含む5〜7週の胚性後腸および「未分化」ニッチが豊富な腫瘍組織の両方に多数で存在する。これらは、いくつかの幹細胞様性質、特に「標語」(shibboleth)の非対称分裂およびヒト結腸前新生物および新生物組織の増殖速度と一致した核分裂頻度を有する(Herrero-Jimenez, P.,ら、Mutat.Res., 400:553-78 (1998), Herrero-Jiminez, P., ら、Mut. Res. 447:73-116 (2000); Gostjevaら、Cancer Genetics and Cytogenetics, 164:16-24 (2006); Gostjevaら、Stem Cell Reviews, 1 : 243-252 (2005))。これらの以前に認識されていない核形態は、腫瘍生成および分化の両方の供給源であり、従って癌治療戦略の標的である。これら釣鐘形核はゲノムがssDNAとして実質的に表わされる段階を経るという観察によって、ターゲティングおよび破壊が可能になる。
本明細書に記載される予期しない観察は、腫瘍幹細胞複製が、複製中間体構造を伴い、釣鐘形核が2つの核に分裂し始めること、および核分裂中に腫瘍幹細胞の実質的な部分が最初にssDNAに分離することである。この複製中間体構造中に、日常業務の方法を用いて検出できる多量のssDNAが、核および細胞に存在する。この観察は、釣鐘形核の増殖の阻害/阻止で薬剤の有効性を評価し、本明細書に記載される有効な抗腫瘍剤のスクリーニングおよび同定方法の基礎を元にしている。さらに、腫瘍幹細胞特異的分子は、この複製中間体構造の細胞に存在することもでき、アッセイ法の基礎として機能することもできる。本明細書に記載される方法および米国特許出願第2006/0063144号およびPCT/US06/047136を用いて、(各教示は、その全体が参照によって本明細書に援用される)候補抗腫瘍剤への接触もしくは処理の前および後の両方で、固形腫瘍塊中のssDNAを含む釣鐘形核またはssDNAのみを視覚化でき、釣鐘形核(異型核)を有する細胞の相対密度を、候補/試験薬剤を用いた処理の前および後のいずれかと、または薬剤と接触/処理されていない他の腫瘍組織と比較できる。相対密度の減少、例えば細胞数もしくは腫瘍塊重量に対するssDNAを含む釣鐘形核またはssDNAのみの数の減少は、薬剤が腫瘍幹細胞の増殖を減少するまたは阻止することに有効であることを示す。
本明細書に提示されたデータの結果として、近接した環境の細胞への最小傷害の有り無しで、腫瘍幹細胞を標的とし、かつ破壊する薬剤を送達する方法が現在利用可能である。本発明の一態様において、ssDNA特異的薬剤または腫瘍幹細胞特異的分子を標的とする薬剤を投与することによって、個体の前新生物、新生物(癌)または他の病気の治療方法が利用可能である。用語「治療」は、本明細書で使用される場合、癌または病気と関連する症状の改善:癌の転移の低減、阻止または遅延;1つ以上の腫瘍の数、容積および/またはサイズの低減;および/または癌もしくは病気の症状の重篤度、持続期間または頻度の減少のことを示すことができる。「ssDNA特異的治療剤」は、本明細書で使用される場合、ssDNAの形成の阻止、ssDNAの複製の妨げまたはssDNAのコピー化の妨げによる破壊のための腫瘍幹細胞のssDNAを標的とする薬剤(例えば、化学療法剤)、またはそうでない場合は、癌を治療する、もしくは個体の(1つもしくは複数の)腫瘍もしくは病気の影響を減少するもしくは除去する薬剤のことをいう。本明細書に記載されるように、ssDNAは腫瘍幹細胞維持に重要であり、従ってssDNAの形成の阻止、ssDNAの複製の妨げまたはssDNAのコピーの妨げをもたらすssDNAの破壊が、腫瘍幹細胞複製を阻害するので、癌または病気が治療される。いくつかの態様において、記載される治療方法は、1つ以上の手術、ホルモン切除療法、放射療法または化学療法と共に使用される。本発明の化学療法、ホルモンおよび/または放射療法の薬剤および化合物が、同時に、別々にまたは連続して投与されてもよい。
本発明の方法において、薬剤は、それ自体が投与されてもよいか、または生理学的に適合性のある担体または賦形剤とともに薬剤を含む組成物(例えば、医薬組成物または生理学的組成物)で投与されてもよい。本発明の一態様において、インビボ(例えば、個体に対して)またはインビトロ(例えば、組織試料に対して)のいずれかで投与されてもよい。本発明の方法は、ヒト個体に使用されてもよいだけでなく、獣医学的使用(例えば、他の哺乳動物、例えば、家畜動物(例えば、ウマ、ウシ、ヒツジ、ヤギ、ブタ、イヌ、ネコ、トリ)および非家畜動物にも適用可能である。担体および組成物は滅菌されたものであり得る。製剤は、投与様式に適したものであるべきである。
以下に、本発明の例示的態様を説明する。
成人の組織および腫瘍被検物を、Massachusetts総合病院、病理科(Gostjeva,E. et al,Cancer Genet. Cytogenet.,164:16-24,2006)の共同研究者から外科的廃棄物として得た。匿名廃棄腫瘍および組織切片の使用は、Prof. W. G. Thillyの研究所を通じた実験用被検体としてのヒト使用のMIT委員会により承認された。
以下のプロトコルは、染色体および核の構造的および定量的観察について望ましい明確さの組織および腫瘍被検物の核の可視化を可能にする。重要な要素は、手術の30分以内に固定した新鮮腫瘍試料の使用および薄い切片作製の標準手順の回避である。釣鐘形核は、明らかに組織および腫瘍試料において自己分解の早期犠牲物であり、切除後45分程度で識別できなくなる。標準的な5ミクロン切片は、ほぼ全てが5ミクロンより大きい最小直径を有すると発見されたいくつかの核形態を通って単純にスライスしたものである。考案された特別な技術は、大きな進歩の印である。
本明細書において使用される定量的画像解析のためのソフトウェアは、初期のサテライトサーベイランスシステムから適合したバックグラウンド抑制のためのアプローチを利用する。この技術はドイツのKontron corporationによって獲得され、それ自体は以前にZeiss,Inc.によって獲得された。全ての画像は、パーソナルコンピュータに接続された電動式光学顕微鏡AxioscopeTM、カラーCCDカメラAxioCamTM(Zeiss、Germany)からなる特注KS-400 Image Analysis SystemTM、Version 3.0(Zeiss、Germany)を用いて得られた。フォイルゲン染色単独を使用した場合は可視光および560nm(緑色)フィルターを用いて、プラナーアポクロマート対物レンズ1.4/100倍率で顕微鏡から画像を送る。フォイルゲン-ギムザ染色が使用される場合は、フィルターは使用されない。フレーム取込み器および最適露光は、各スキャニングセッションの前に調整される。核の画像はピクセルサイズ0.0223 x 0.0223ミクロンで記録される。
7つの異なる核形態型(大楕円形、凝縮楕円形、卵形、豆-、葉巻-、ソーセージ-および釣鐘形)が、胎児性腸試料全体に見られた(図1A)。釣鐘形核は、凝縮染色体に類似した凝縮クロマチンによって開放が保持されているように思われた(図1B)。釣鐘形核は、約20〜50ミクロンの管状構造すなわち合胞体内で線形に「頭からつま先」の向きに組織化されていた(図2)。釣鐘形核の「頭からつま先」パターンは、観察した全ての胚性管状構造において保存されたが、管状構造は、平行な管状構造が局所で逆平行な向きの釣鐘形核を有するように前後に蛇行していた。
陰窩内のほぼ全ての核は、陰窩基部から管腔表面に観察することができた(図4A)。多くの陰窩は、個々の核の形状が識別できるような様式で広がっていた。卵形または楕円形の核を有する細胞が、基部のすぐ上から管腔内へ広がる上皮までの陰窩に並ぶ(図4C)。陰窩基部の最初の約25細胞において、約2〜3ミクロン厚および約10ミクロン直径の円盤状と特徴付けされる潜在的に異なる9つの核形態型が優勢であった(図4B)。細胞が充分に広がった全ての陰窩基部の1%未満において、孤立した釣鐘形核が明白な円盤状核間で識別された(図4Aおよび4B)。同様の低頻度の釣鐘形核が成人肝臓の調製物において観察された。新生物または前新生物の任意の病理学的徴候のない成人結腸では、千を超える充分に広がった陰窩の細胞毎の細胞スキャンにおいて他の核形態バリアントは観察されなかった。
腺腫は、各々約2000個の細胞を有する正常結腸陰窩と識別不可能な多くの陰窩を含んでいた。これらは、図5Aに示されるように分岐形態において頻繁に見られた。陰窩壁内の正常結腸陰窩内と同じ楕円形および卵形の核は、正常結腸よりも頻度が高いが、陰窩基部に1つまたは2つの釣鐘形核が存在した。約8000個の細胞を含む不定形の小葉構造もまた観察され、その細胞は、組織解離によってより容易に広がった。ほぼ全ての不定形構造において、構造本体の方向に釣鐘開口が向いた2つ以上の釣鐘形核が存在した(図5B)。また、多くの多様な細胞および群が陰窩と不定形構造間に散在した(図5C)。いくつかの規則的な構造は、約250個、約500個または約1000個の細胞を含む充分なサイズの正常陰窩に成長しているようであった。多くの細胞群は、各々が1個の釣鐘形核を有する正確に8、16、32、64および128個の細胞の「環」として見られた(図5D)。
腺腫と同様に腺癌は、陰窩、大きな不定形構造ならびに16、32、64および128個の細胞の陰窩内クラスターの混合物を含んだ。釣鐘形核は、なお陰窩の基底カップにおいて単一のもの、対または多数のものとして見られ、大きな不定形小葉構造の壁では複雑な渦巻き内に埋もれていた(図6)。腺癌における核形態型の組は、弾丸形形態型を含む腺腫で見られる組と同一のようである。
3D保存単一釣鐘形核および対称分裂釣鐘形核対の共焦点顕微鏡検査を行なうため、DeltaVision(登録商標)RT Restoration Imaging System at Imaging Center、Whitehead Instituteが使用される。該システムは、核画像の復元のためのリアルタイム2Dデコンヴォルーションおよび3D Z投影法を提供する。
FISHを用いて、染色体全体が、釣鐘形核の上部に「環」として出現する凝縮に関与していることを決定した。基本的に、「環」内の染色体の標識は、釣鐘形核が異なる形態の核(図10Bに示される)を生じたときの変化を解析するため、ならびに核形態ではなく他の手段によってこれらの核を認識するための蛍光マーカーの開発の手段として予測される。
本明細書に記載の技術により、ヒト細胞培養物において有糸分裂中の姉妹核の任意の2つの核間または後期終期間の2%という小さい差の検出が可能になる。いつDNAが、釣鐘形核を含む細胞または合胞体によって合成されるかを決定するため、これらの技術を使用した。これは、核分裂の過程にあると思われる核のスキャニングを伴った。一般に、二倍体ヒト細胞には、同じ染色スライド上のヒトリンパ球DNA含有量と比較して予測される量のDNAを含む胎児性釣鐘形核が見られる。また、ヒト前新生物病変および腫瘍の釣鐘形核中のDNAの量は、平均して二倍体DNA量より多く、平均あたりで広いばらつきを示すことに注目する。測定により、別の全く予想外の所見:釣鐘形核が関与する対称および非対称核分裂ではともに、DNA合成が核分裂プロセスより先ではなく同時であるということが明らかになった。核は、単一の核の量からの総DNA含有量の増加が明白に検出される前、「カップからカップへの」分離のプロセスに伴って良好なようである。DNAの総量は、明らかに分裂し始めている核内の単一腫瘍核の平均を近似する低い値から増加し、ちょうど分裂を終了したようである核の核内含有量の平均の約2倍に達する。
以前に認識されていない一連の核の形態が、釣鐘形核を生じるヒト胎児性調製物において同定された。これらの形態は、第5週に検出され、最初は、釣鐘形核を含む管状合胞体であった。これらの例を図13A〜Dに示す。これは、初期胚形成の有糸分裂の球形核から、発生上の「幹」細胞系統の正味成長および分化を表す後の無糸分裂の釣鐘形核までの形態変化を示す重要な所見である。
合胞体外部釣鐘形核は、実際にはヒトDNAを含む。ほとんどの動原体は、胎児性試料において釣鐘形核の開口部の凝縮DNA領域と関連している。興味深いことに、標準的なFISHプロトコルでは、合胞体内の釣鐘形または他の形状の核は染色されず、合胞体の収縮性要素含有鞘がFISH試薬の侵入をブロックし得ることを示す。図15は、ヒト12週の胎児性結腸組織由来の球形(図15A)、「葉巻」形状(図15B)および釣鐘形(図15C)の核の動原体(緑色)を示す。
形態が図13A〜Dに示すものとほぼ同一である、特に、釣鐘形核、前合胞体形態および合胞体形態を含む種々の形態の全ての核が、12.5日目に最初に検出され、次いで、14.5〜16.5日目に検出される前合胞体形態を有する胎児性マウス組織に見られ、胎児は、胎児性マウスの器官形成期間にほぼ平行であった。マウスにおけるこれらの所見は、ヒト観察を考慮すると驚くべきことではないが、ヒトにおいて倫理的または可能でない非ヒト種における器官形成の研究の可能性の範囲を大きく開くものである。
染色体および染色体要素、種々の収縮性分子(例えば、アクチン)ならびに「幹細胞マーカー」と一般的に称されるものを含む他の同定可能なマーカーの位置の規定のために、FISHを含む一連の組織化学的手順を適用するために原腸の大量の合胞体および釣鐘形核が使用される。本明細書に記載の技術は、ZEISS-P.A.L.M.顕微解剖装置を用いた合胞体および個々の核を収集する作業に適用される。成功の基準は、細胞内mRNA、最も一般的なタンパク質およびグリコサミノグリカンのスキャニングに充分な数である10,000以上の数の核相当物における合胞体形態または核形態型に関して均一な一連の試料の回収である。
複製中間体構造を同定するためのさらなるデータを図16〜19に示す。図18Bは特に重要である。これは、釣鐘形核内のssDNAゲノムの存在を決定的に示す。ssDNAに結合すると特異的に単独に赤色蛍光を発するアクリジンオレンジ染色が使用される。
モノクローナル抗体Mab F7-26をssDNA中間体の検出に使用した(Bender MedSystems GmbH,Vienna,Austria)。使用前に、抗体のストック溶液を(PBSおよび5%ウシ胎児血清で)10倍希釈した。ssDNA抗体を適用する前に、10mM HCl中0.005%ペプシン、50mM MgCl2および50mM MgCl2中1%ホルムアルデヒド(Fomina et al.,2000に詳細に記載)を用いて一連の洗浄工程を行なった。各洗浄工程後、スライドをPBSで3回洗浄した(それぞれ5分間)。最後に、スライドを0.05%Tweenで洗浄し、次いで、Tween中ブロッキングタンパク質3%BSA、および再度0.05%Tween中で洗浄した。その後、スライドを5%FBS中Mab F7-26で室温で60分間処理した後、抗マウスIg-G-FITC、Alexa 488を30分間適用した。スライドをDAPIで染色し、一連のエタノール70%、90%および100%で脱水し、空気乾燥し、「Citiflour」媒体中にマウントした。
Claims (23)
- A)被験体または宿主動物の腫瘍と候補薬剤とを接触させる工程であって、該腫瘍が釣鐘形核を含む腫瘍幹細胞を含み、該釣鐘形核のいくつかまたは全てがssDNAと結合した複製中間体構造である工程;
B)薬剤が腫瘍と相互作用するために適切な条件下で、候補薬剤と接触する腫瘍を維持する工程;
C)腫瘍試料を切除して、工程B)の接触した後の腫瘍試料中のssDNAを含む釣鐘形核の存在、非存在もしくは数またはssDNAのみの存在、非存在もしくは量を決定する工程;および
D)候補薬剤と接触した後の腫瘍試料中のssDNAを含む釣鐘形核の存在、非存在もしくは数またはssDNAのみの存在、非存在もしくは量と、対照試料中のssDNAを含む釣鐘形核の存在、非存在もしくは数またはssDNAのみの存在、非存在もしくは量とを比較する工程を含み、
