Abstract
Description
本出願は、2007年6月1日付け出願の米国出願番号60/941,524(その全開示を参照により本明細書に組み入れることとする)の利益を主張するものである。
本発明は、全般的には、予防用および治療用ワクチンの分野に関する。より詳しくは、本発明は、乳癌の治療および乳癌寛解患者における再発の予防のためのペプチドワクチンに関する。
特許、公開された出願、技術文献および学術論文を含む種々の刊行物が本明細書の全体において引用されている。これらの引用刊行物のそれぞれの全体をあらゆる目的で参照により本明細書に組み入れることとする。
本発明は、臨床的に寛解状態にある乳癌患者における乳癌の再発に対する免疫を誘導し維持する方法に関する。該方法は、製薬上有効な担体と配列番号2のアミノ酸配列を有するペプチドとを含む組成物の有効量を該患者に投与することを含む。該投与は、当技術分野における適当な任意の手段、例えば接種または注射、より具体的には皮内注射(これは1以上の別個の用量で行われうる)により達成されうる。そのような用量は、等濃度の該ペプチドおよび免疫アジュバントを含むことが可能であり、実質的に同時に投与されることが可能であり、皮膚の表面上の1つの接種部位にまたは互いに離れた部位に投与されることが可能である。該組成物は、防御免疫が確立されるまで、月1回で約3〜6回またはそれ以上投与されうる。いくつかの態様においては、該組成物は更に、アジュバント、例えば組換えヒト顆粒球マクロファージ-コロニー刺激因子(GM-CSF)を含む。

本明細書および特許請求の範囲の全体にわたって、本発明の方法および他の態様に関する種々の用語が用いられている。そのような用語には、特に示さない限り、当技術分野における通常の意味が与えられるべきである。他の具体的に定義されている用語は、本明細書に記載されている定義に合致するように解釈されるべきである。
節陽性(node-positive)(NP)および節陰性(NN)治験は、地域の施設内治験審査委員会(Institutional Review Boards)により承認され、治験新薬申請(BB-IND#9187)に基づいてWalter Reed Army Medical Center, Washington, DCおよびJoyce Murtha Breast Care Center, Windber, PAにおいて実施された。全ての患者は、組織学的に確認された乳癌(BCa)を有し、登録前に標準的な一連の手術、化学療法および放射線療法(必要に応じて)を完了していた。ホルモン療法を受けている患者は彼女らの特定の治療計画を継続した。適切なカウンセリングおよび同意の後、BCa患者は適切な治験(NPまたはNN)に登録され、ついでHLAタイプ決定された。なぜなら、E75は主として、一般集団の約40〜50%に見出されるHLA-A2に結合するからである。HLA-A2+ 患者にはワクチン接種し、HLA-A2- 患者は臨床再発について予め観察した。HLA-A3+ 患者は、該A2患者とのパラレル(並行)治験に登録され、登録の時点で能動投与スケジュールで治療された。ワクチン接種前に、患者を一連の想起(recall)抗原(ムンプス、破傷風およびカンジダ)で皮膚検査した。患者が>2の抗原と反応(>5mm)した場合には、該患者は免疫応答性とされた。
E75ペプチドは適正製造基準(good manufacturing practices)等級で、NeoMPS, Inc.(San Diego, CA)により商業的に製造された。ペプチド純度(>95%)は高速液体クロマトグラフィーおよび質量分析により確認され、アミノ酸含量はアミノ酸分析により決定された。無菌性および一般安全性試験は該製造業者により行われた。無菌生理食塩水中に凍結乾燥ペプチド(0.5ml中に100μg、500μgまたは1000μg)を再構成した。投与時に、該ペプチドを解凍し、0.5ml中のGM-CSF(Berlex, Seattle, WA)と混合し、その1.0mlの接種物を分割し、5cm離れた2つの部位に皮内投与した。全ての接種物は同じ四肢に投与された。
NP治験は2段階の安全性治験として設計され、最初の段階ではペプチドの用量を段階的に増加し、後の段階においてはスケジュールを変化させた。一連のワクチン接種の詳細は既に公開されている(Peoples GEら, (2005) J. Clin. Oncol. 23:7536-45)。簡潔に説明すると、3〜6人の患者(HLA-A2+またはHLA-A3+)をそれぞれ、100μg、500μgまたは1000μgのE75の4回または6回の月1回の注射(それぞれ100.6、500.4、500.6、1000.4および1000.6)を受けるよう割り当てた(表2)。NP患者における最適用量を決定し確認するために、最終的に群を拡張したため、後の用量群における患者数の方が多くなった。
各ワクチン接種の前ならびに一連のワクチン接種の完了の1ヶ月後(ワクチン後(post-vaccine))および6ヶ月後(長期(long-term))にそれぞれ血液を採取した。50mlの血液を採取し、PBMCを単離した。PBMCを洗浄し、培地に再懸濁させ、リンパ球源として使用した。
患者をワクチン接種後1時間にわたって即時型過敏症に関して観察し、48〜72時間後に呼び戻して彼女らの注射部位を測定し、彼女らに毒性に関して質問した。毒性をNCI Common Terminology Criteria for Adverse Events, v3.0により等級付けし、0〜5の尺度で記録した。より低用量の群において重大な毒性が生じない場合に限り、1つの用量群から次の用量群へと進めた。これらの一連の試験中に生じた最大の局所および全身毒性に基づいて、患者特異的な結果を記録した。
患者の主治医の腫瘍専門医による指示どおりに、ケア癌スクリーニングの基準に従い、臨床的再発に関して全ての患者を観察した。生検により証明された場合、または腫瘍専門医主治医チームにより再発に対して治療された場合には、患者は再発とみなされた。
Kaplan-Meier法による生存分析を用いて群間で再発率を比較し、ログランク分析を用いて、再発を示した被験者の比率を比較した。適宜、ウィルコックソン、フィッシャー正確確率検定またはχ2を用いて、臨床-病理学的(clinico-pathologic)因子に関するP値を計算した。ワクチン接種前およびワクチン接種後の二量体レベルを比較するためのP値を、ウィルコックソン検定を用いて計算し、DTHに関しては、スチューデントt検定を用いて計算した。
患者からの新たに単離されたPBMCにおけるCD8+ E75特異的細胞の存在を、二量体アッセイを用いることにより直接的にアッセイした。簡潔に説明すると、HLA-A2:免疫グロブリン(Ig)二量体(PharMingen, San Diego, CA)をE75または対照ペプチド(E37, 葉酸結合性タンパク質(25-33)RIAWARTEL(配列番号5))と共にローディングした。これは、1μgの二量体を過剰(5μg)のペプチドおよび0.5μgのβ2-ミクログロブリン(Sigma , St. Louis, MO)と共に37℃で一晩インキュベートすることにより行った。ついでそれらを、使用するまで4℃で保存した。PBMCを洗浄し、PharMingen染色バッファー(Stain Buffer)(PharMingen)に再懸濁させ、5ml丸底ポリスチレンチューブ(Becton Dickinson, Mountain View, CA)内に5×105 細胞/100μl/チューブで加え、ローディングされた二量体および抗体で染色した。各患者において、それぞれの逐次ワクチン接種に応答したCD8+ E75特異的細胞のレベルを決定し、各患者ごとに全ての接種後測定値を平均し、彼女らの接種前レベルと比較した。
既に記載されているように(Peoples GEら, (2005) J. Clin. Oncol. 23:7536-45)して、一連のワクチン接種の完了の1ヶ月後、両方の治験において、0.5mlの正常生理食塩水中の100μgのE75(GM-CSFを含有しない)および体積対照としての0.5mlの正常生理食塩水を使用してDTH反応を評価した。感受性ボールペン法を用いることにより、48〜72時間の時点で二次元で該DTH反応を測定し、直交平均(orthogonal mean)として表し、対照と比較した。NN治験においては、ワクチン接種前にもDTHを実施した。
該ワクチンのin vivo有効性を測定するために、一連のワクチン接種の完了の1ヶ月後に、100μgのE75を生理食塩水体積対照と共に皮内注射することによりワクチン接種後DTHを測定した。全ワクチン接種患者のうち、74%が陽性ワクチン接種後DTHを示し、E75に対する平均硬化は14.0±1.4mmであり、一方、対照においては2.1±0.5mmであった(P<0.0001)(図4A)。
HLA-A3+ 患者に関するワクチン応答をE75特異的インターフェロンγ ELISPOTによっても評価した。ELISPOTにより、該A3患者はベースラインにおいて0〜30スポット/106 細胞の範囲を示し、これはワクチン接種後においては3〜448スポット/106 細胞の範囲に増加した(p=0.04)。最も重要なことに、臨床的再発は両方の群において同じであり(A3, 9.1%対A2, 8.2%)、これに対して、対照群においては14.8%であった。
患者
NPおよびNN治験は地域の施設内治験審査委員会(Institutional Review Boards)により承認され、Walter Reed Army Medical Center (WRAMC), Washington, DCおよびJoyce Murtha Breast Care Center, Windber, PAにおいて実施された。これらの臨床治験は、米国食品医薬品局(Food and Drug Administration)により承認された治験新薬申請(BB-IND#9187)に基づいて実施されている。全ての患者は、組織学的に確認された乳癌を有し、標準的な療法を完了しており、初期登録の時点で無疾患かつ免疫応答性であった。前記実施例に記載されているとおりに、6ヶ月の期間にわたる種々のスケジュールに基づき、HLA-A2+ 患者およびHLA-A3+ 患者に種々の用量のE75およびGM-CSFをワクチン接種した。患者がその一連の一次ワクチン接種の完了から少なくとも6ヶ月経過している場合には、E75(1mg)+ GM-CSF(0.250mg)の選択的ブースター用量を該患者に投与した。
E75ペプチドは適正製造基準(good manufacturing practices)等級においてNeoMPS(San Diego, CA)により商業的に製造された。ペプチド純度は高速液体クロマトグラフィーおよび質量分析により確認され、アミノ酸含量はアミノ酸分析により決定された。該ペプチドは95%超にまで精製された。無菌性および一般安全性試験は該製造業者により行われた。凍結乾燥ペプチドを無菌生理食塩水中で0.5ml中1000μgの濃度で再構成した。該ペプチドを0.5mL中のGM-CSF(Berlex, Seattle, WA)と混合し、その1.0mlの接種物を分割し、5cm離れた2つの部位に皮内投与した。ブースター接種は一連の一次接種と同じ四肢において行った。
患者をワクチン接種後1時間にわたって即時型過敏症に関して観察した。毒性をNCI Common Toxicity Criteria for Adverse Events, v3.0により等級付けし、0〜5の尺度で記録した。重大(等級2または3)な局所または全身毒性を既に示していた患者には0.125mgの低用量のGM-CSFを投与した。
ブースターワクチン接種の前ならびにブースター投与の3〜4週間後に血液を採取して末梢血単核細胞をVacutainer CPTチューブ内に単離し、それをリンパ球源として使用した。
患者からの新たに単離されたPBMCにおけるCD8+ E75特異的細胞の存在を、前記実施例3に記載されている二量体アッセイを用いることにより直接的にアッセイした。簡潔に説明すると、HLA-A2:免疫グロブリン(Ig)二量体(PharMingen, San Diego, CA)をE75または対照ペプチド(葉酸結合性タンパク質ペプチドE37 (25-33)RIAWARTEL(配列番号5))と共にローディングした。これは、1μgの二量体を過剰(5μg)のペプチドおよび0.5μgのβ2-ミクログロブリン(Sigma , St. Louis, MO)と共に37℃で一晩インキュベートすることにより行った。ついでそれらを、使用するまで4℃で保存した。PBMCを洗浄し、PharMingen染色バッファー(Stain Buffer)(PharMingen, San Diego, CA)に再懸濁させ、5ml丸底ポリスチレンチューブ(Becton Dickinson, Mountain View, CA)内に5×105 細胞/100μl/チューブで加え、ローディングされた二量体および抗体で染色した。各HLA-A2+ 患者において、該ワクチンブースターの前および後のCD8+ E75特異的細胞のレベルを決定した。
直ちに(ex vivo)またはペプチドとの7日間のインキュベーションの後にBD ELISPOTキットを使用して、IFNγ産生細胞を検出した。IL-7を含有する培地中(ex vivo)またはIL-7を添加したまたは添加しない培地中(7日)、新鮮なPBMCをウェル当たり5×105 細胞(ex vivo)または1×105 (7日)の濃度でELISPOTプレート内にプレーティングした。ペプチド(E37, FluM, E75, GP2, HER2/neu 1μgまたは5μg)の存在下または非存在下、細胞を16時間(ex vivo)または7日間刺激した。該7日ウェル内での追加的なインキュベーションはE75+HER2/neu 1μgまたは5μgの組合せを含むものであった。十分な細胞が入手可能であった場合には、各血液サンプルに関して合計16回のアッセイを行った。インキュベーションの終了時に、該プレートを製造業者の説明に従い発色させた。ビオチン化検出抗体を加え、該プレートを4℃で一晩インキュベートした。インキュベーション後、該プレートを洗浄し、アビジン-HRP溶液を1時間加え、AEC基質溶液を使用してスポットを発色させた。分析装置Immunospot Series 2およびImmunoSpotソフトウェアを使用して、スポットを計数した。
局所反応(LR)を応答のin vivo機能評価として測定した。LRをワクチン接種の48〜72時間後に測定し、感受性ボールペン法を用いて2方向において測定し、直交(orthogonal)平均±SEとして記録した。LRをその患者自身の以前のLRと比較して、ブースターに対する応答を評価した。
HLA:IgG二量体値を中央値として表し、ウィルコックソン検定を用いてP値を計算した。比例比較のために、フィッシャー正確確率検定を用いた。局所反応の比較は、適宜、対応のある又は対応の無いスチューデントt検定で行った。
節陽性および節陰性BCa患者において、HER2/neu E75ペプチドワクチンを使用して臨床治験を行った。これらの患者は、あらゆるレベルのHER2/neu発現のものから構成されていた。HER2/neu状態の決定は、主に、2種類の試験、すなわち、免疫組織化学法(IHC)および蛍光in situハイブリダイゼーション(FISH)により行った。IHCはHER2/neuタンパク質の過剰発現を検出し、0〜3+(0 = 陰性、1+ = 低発現、2+ = 中等度、および3+ = 過剰発現)の半定量尺度で記録されている。一方、FISHはHER2/neu遺伝子の増幅(過剰コピー)を検出し、17番染色体に対するHER2/neuの比率として表され、FISHが>2.0コピーであれば過剰発現と解釈される。IHCおよびFISHの一致は約90%である。
HER2/neu発現のレベルに基づいて、本発明者らの第II相E75ワクチン治験に登録された163人のBCa患者において、サブセット(部分集団)分析を行った。患者は、低発現体(LE = IHC 1+〜2+およびFISH>0であるが<2.0)と過剰発現体(OE = IHC 3+および/またはFISH>2.0)を比較して、さらにIHC状態(0, 1+, 2+, 3+)により、評価した。標準的な臨床病理学的因子、該ワクチンに対する免疫応答(in vivo DTH反応およびin vitro HLA-A2:IgG二量体アッセイ)および臨床応答(絶対的再発および致死率)の分析を行った。
E75 NPおよびNN治験が施設内治験審査委員会(Institutional Review Boards)により承認され、治験新薬申請(BB-IND#9187)に基づいてWalter Reed Army Medical Center, Washington, DCおよびJoyce Murtha Breast Care Center, Windber, PAにおいて実施された。全ての患者は、組織学的に確認された乳癌(BCa)を有し、登録前に標準的な一連の手術、化学療法および放射線療法(必要に応じて)を完了していた。ホルモン療法を受けている患者は彼女らの特定の治療計画を継続した。適切なカウンセリングおよび同意の後、BCa患者は適切な治験(NPまたはNN)に登録され、ついでHLAタイプ決定された。なぜなら、E75は主として、一般集団の約40〜50%に見出されるHLA-A2に結合するからである。HLA-A2+ 患者にはワクチン接種し、HLA-A2- 患者は臨床再発について予め観察した。ついでHLA-A3+ 患者にワクチン接種した。ワクチン接種前に、患者を一連の想起(recall)抗原で皮膚検査した(マントゥー試験)。患者が>2の抗原と反応(>5mm)した場合には、該患者は免疫応答性とみなされた。
E75ペプチドは適正製造基準(good manufacturing practices)等級においてNeoMPS, Inc.(San Diego, CA)により商業的に製造された。ペプチド純度(>95%)は高速液体クロマトグラフィーおよび質量分析により確認された。無菌性および一般安全性試験は該製造業者により行われた。0.5mlの無菌生理食塩水中に100mcg、500mcgまたは1000mcgの凍結乾燥ペプチドを再構成した。該ペプチドを0.5ml中のGM-CSF(Berlex, Seattle, WA)と混合した。その1.0mlの接種物を分割し、同じ四肢における5cm離れた2つの部位に皮内投与した。
NP治験は2段階の安全性治験として設計され、最初の段階ではペプチドの用量を段階的に増加し、後の段階においてはスケジュールを変化させた。一連のワクチン接種の詳細は既に公開されている。簡潔に説明すると、3〜6人の患者をそれぞれ、100mcg、500mcgまたは1000mcgのE75ペプチドの4回または6回の月1回の注射(それぞれ100:6、500:4、500:6、1000:4および1000:6)を受けるよう割り当てた。NP患者における最適用量を決定し確認するために、最終的に群を拡張したため、後の用量群における患者数の方が多くなった。
患者をワクチン接種後1時間にわたって即時型過敏症に関して観察し、48〜72時間後に呼び戻して彼女らの注射部位を測定し、彼女らに毒性に関して質問した。毒性をNCI Common Terminology Criteria for Adverse Events, v3.0により等級付けした(0〜5の尺度で記録した)。より低用量の群において重大な毒性が生じない場合に限り、1つの用量群から次の用量群へと進めた。これらの一連の試験中に生じた最大の局所および全身毒性に基づいて、患者特異的な結果を記録した。
各ワクチン接種の前ならびに一連のワクチン接種の完了の1ヶ月後(ワクチン後(post-vaccine))および6ヶ月後(長期(long-term))に血液を採取した。50mlの血液を採取し、PBMCを単離した。PBMCを洗浄し、培養培地に再懸濁させ、リンパ球源として使用した(既に記載されているとおりである)。
患者からの新たに単離されたPBMCにおけるCD8+ E75特異的細胞の存在を、既に記載されているとおりに二量体アッセイを用いることにより直接的にアッセイした。簡潔に説明すると、HLA-A2:免疫グロブリン(Ig)二量体(PharMingen, San Diego, CA)をE75または対照ペプチド(E37, 葉酸結合性タンパク質(25-33)RIAWARTEL(配列番号5))と共にローディングした。これは、1mcgの二量体を過剰(5mcg)のペプチドおよび0.5mcgのβ2-ミクログロブリン(Sigma , St. Louis, MO)と共に37℃で一晩インキュベートすることにより行った。ついでそれらを、使用するまで4℃で保存した。PBMCを洗浄し、PharMingen染色バッファー(Stain Buffer)(PharMingen)に再懸濁させ、5ml丸底ポリスチレンチューブ(Becton Dickinson, Mountain View, CA)内に5×105 細胞/100μl/チューブで加え、ローディングされた二量体および抗体で染色した。各患者において、それぞれの逐次ワクチン接種に応答したCD8+ E75特異的細胞のレベルを決定し、平均接種後レベルを彼女らの接種前レベルと比較した。
既に記載されているようにして、一連のワクチン接種の完了の1ヶ月後、両方の治験において、0.5mlの正常生理食塩水(GM-CSFを含有しない)中の100mcgのE75ペプチドおよび体積対照としての0.5mlの正常生理食塩水を使用してDTH反応を評価した。感受性ボールペン法を用いることにより、48〜72時間の時点で二次元で該DTH反応を測定し、直交平均(orthogonal mean)として記録し、対照と比較した。NN治験においては、ワクチン接種前にも同様にDTHを実施した。
患者の主治医の腫瘍専門医による指示どおりに、ケア癌スクリーニングの基準に従い、臨床的再発に関して全ての患者を観察した。生検により証明された場合、または腫瘍専門医主治医チームにより再発に対して治療された場合には、患者は再発とみなされた。
Kaplan-Meier法による生存分析を用いて群間で再発率を比較し、ログランク分析を用いて、再発を示した被験者の比率を比較した。適宜、ウィルコックソン、フィッシャー正確確率検定またはχ2を用いて、臨床病理学的因子に関するP値を計算した。ワクチン接種前およびワクチン接種後の二量体レベルを比較するためのP値およびDTHを、対応のある又は対応の無い両側スチューデントt検定を用いて計算した。
LE(対照 = 44、ワクチン = 56)対 OE患者(対照 = 22、ワクチン = 29)およびIHC状態対照およびワクチン群(それぞれ、0 = 5対7, 1+ = 15対25, 2+ = 24対26, 3+ = 13対19)を評価した。LE対OEおよび全てのIHC状態ワクチン接種群は共に免疫学的に応答したが、LE患者、より詳しくは、IHC 1+患者は、増加した長期in vitro免疫応答を示した(それぞれ、p=0.04およびp=0.08)。また、それらの対照群と比較して、LE患者は死亡率の減少傾向を示し、IHC 1+患者は死亡率の減少を示した(それぞれ、p=0.08およびp=0.04)。
186人を該E75ワクチン研究に登録した。9人は離脱(4人の対照患者および5人のワクチン接種患者 - 毒性により離脱した者はいなかった)して177人が該治験を完了した。NP治験における全ての対照(C)およびワクチン接種(V)患者(C=46, V=45, 合計=91人の患者)にIHC、FISHまたは両方の試験を行った。NN治験(C=35, V=51, 合計=86人の患者)においては、12人の患者がHER2/neu IHC 0腫瘍を示した(C=5, V=7)。また、NN治験においては、14人の患者(C=7, V=7)の腫瘍はIHCもFISHも受けなかった。これらの14人の患者はサブセット分析から除外されている。したがって、163人の患者が分析に利用可能であった。
発現ごとの患者:
LE(IHC 1+〜2+またはFISH>0および<2.0)をOE(IHC 3+またはFISH>2.0)と比較するサブセット(部分集団)分析を行った。対照群の66人の患者にIHCまたはFISHを行った(LE=44, OE=22)。E75ワクチン群の合計85人の患者にIHCまたはFISHを行った(LE=56, OE=29)。同等数のCおよびV患者がLE群(それぞれ、67%対66%)およびOE群(それぞれ、33%対34%)に含まれていた。
該E75ワクチンはLE患者およびOE患者の両方においてin vitro免疫応答を惹起することが可能であった。ワクチン接種前から最大値までのE75特異的CD8+ T細胞の有意な増加が両方の群において認められた(LE p<0.001, OE p<0.001)。LE患者は、OE患者に比べて統計的に高い最大免疫応答を示した(p=0.04)(図11A)。
再発および死亡率により評価された臨床応答を図11Cおよび11Dに示す。C患者と比較した場合、全てのV患者(LE=10.7%対OE=13.8%)が再発率の減少を示したようであったが(LEおよびOE=18.2%)、これらの数は統計的に有意ではなかった。より重要なことに、V患者においては死亡率の減少の傾向が認められ、これはLE患者において最も印象的に認められた(C=6.8%対V=0.0%; p=0.08)。
IHC状態ごとの患者:
C群は、IHC(0=5, 1+=15, 2+=24, 3+=13)に付された57人の患者の病理学的標本を有していた。E75 V群は、IHC(0=7, 1+=25, 2+=26, 3+=19)に付された77人の患者の病理学的標本を有していた。同等のパーセンテージのCおよびV患者が各IHC群に含まれていた(0 C=8.8%対V=9.1%; 1+ C=26.3%対V=32.5%, 2+ C=42.1%対V=33.8%, 3+ C=22.8%対V=24.7%)。
該E75ワクチンは全てのIHCカテゴリーにおいてin vitro免疫応答を惹起することが可能であった。ワクチン接種前から最大値へのE75特異的CD8+ T細胞への有意な増加により認められるとおり(0 p=0.007, 1+ p<0.001, 2+ p=0.004, 3+ p=0.002)、全てのIHC群(0, 1+, 2+, 3+)が該ワクチンに応答した。IHC 1+患者のみが、ワクチン接種前から長期へのE75特異的CD8+ T細胞における有意な増加の傾向を示した(p=0.08)(図12A)。
再発および致死率により評価された臨床応答を図12Cおよび12Dに示す。全てのIHCカテゴリー(再発患者がいなかったIHC 0を除く)において、その数は統計的有意性に達していないもののC患者とV患者との比較において再発率の減少が認められた。より重要なことに、IHC 1+ 患者の致死率における有意な減少が認められた(C=20%およびV=0%の致死率)(p=0.04)。

Claims (34)
- 被験体における乳癌再発に対する防御免疫または治療免疫を誘導する方法であって、製薬上有効な担体と配列番号2のアミノ酸配列を有するペプチドとを含む組成物の有効量を該被験体に投与することを含んでなる方法。
- 該組成物を注射または接種により投与する、請求項1記載の方法。
- 該注射が皮内注射である、請求項2記載の方法。
- 該組成物を1以上の分割用量として注射する、請求項2記載の方法。
- 該用量が該ペプチドの最適化量を含有する、請求項4記載の方法。
- 身体上の複数の注射部位が互いに約5cm離れて位置する、請求項4記載の方法。
- 防御免疫が確立されるまで該組成物を6ヶ月にわたって6回投与する、請求項1記載の方法。
- 製薬上有効な担体と配列番号2を有するペプチドとを含むワクチンブースター組成物の有効量を含むブースターを該被験体に投与することを更に含む、請求項1記載の方法。
- 該ブースター組成物を注射により投与する、請求項8記載の方法。
