Abstract
Description
本発明は、ワクチンおよびアジュバントの送達のために用いうる製剤、組成物および方法に関する。より詳細には、本発明は、タンパク質ワクチンおよびアジュバントのより効率的かつ効果的な送達を可能にするミクロスフェア、ならびにミクロスフェアを調製するための方法に関する。
癌および慢性感染症の予防のため、さらには治療のための新たなワクチンの開発が進められている。最も効果的なワクチンは、Tヘルパー反応および抗体応答に加えてCTL応答も誘発する可能性が高いと考えられる。興味が持たれるワクチン送達様式の一つは、ミクロスフェア中への封入を介するものである。しかし、ほとんどのタンパク質は有機溶媒に不溶性のため、タンパク質を封入したミクロスフェアは通常、ダブルエマルション(double-emulsion)法によって作製される必要がある。ダブルエマルション法によって作製されたミクロスフェア製剤は、インビトロ放出試験によって示される放出動態が望ましくない(例えば、初期バーストが大きい、および/または経時的な追加放出が極めて緩徐である)ことが多い。
本発明は、生分解性高分子ミクロスフェア中に封入されたアジュバントおよび薬学的に許容される担体を含む組成物を提供する。1つの態様においては、タンパク質がミクロスフェア中にアジュバントとともに封入される。一般にタンパク質には、癌、自己免疫疾患または感染症と関連のある抗原が含まれる。本発明によれば、タンパク質およびアジュバントを、同じミクロスフェア中への共封入、または第1のセットのミクロスフェア中に封入されたタンパク質および第2のセットのミクロスフェア中に封入されたアジュバントとしての同時投与のいずれかを介する、高分子ミクロスフェア中への封入を介して送達することができる。好ましい態様において、タンパク質および/またはアジュバントは、疎水的イオン対形成(hydrophobic ion pairing;HIP)を介してミクロスフェア中に封入される。
本明細書において開示する本発明は、ミクロスフェア中に共封入されたタンパク質およびアジュバントを含むワクチン組成物に対して強くかつ広範囲にわたる免疫応答を得ることができるという驚くべき発見に基づく。癌、自己免疫疾患および感染症と関連のある抗原を用いて、強い免疫応答が得られている。封入されたタンパク質抗原および別個に封入されたアジュバントの同時投与は広範囲にわたる免疫応答を誘発し、同一セットのミクロスフェア中に共封入されたタンパク質およびアジュバントを用いるとさらに強い広範囲にわたる免疫応答を得ることができる。すなわち、本明細書中に提供する例において試験したアジュバントは疎水性であるにもかかわらず、ミクロスフェア中に封入されたアジュバントを用いて有効なアジュバント送達が達成されている。タンパク質および/またはアジュバントの封入は当技術分野で知られた種々の方法によって達成しうるが、封入は本明細書に記載の疎水的イオン対形成を介して行われることが好ましい。
別に指定する場合を除き、本明細書で用いられるすべての科学用語および技術用語は、当技術分野で一般に用いられているものと同じ意味を有する。本出願において用いる場合、以下の用語および語句は指定された意味を有する。
タンパク質および/またはアジュバントの封入は当技術分野で知られた種々の方法によって達成しうるが、封入は本明細書に記載の疎水的イオン対形成を介して行われることが好ましい。その他のミクロスフェア封入法は国際公開公報第02/03961号(PCT/US01/21780号、2001年7月9日に出願)および2002年1月7日に出願されたPCT/US02/00235号に記載されている。
本発明は、タンパク質および/またはアジュバントを送達するために有用な組成物を提供する。ミクロスフェア中に封入されるタンパク質には、癌または感染症を治療および予防するための組成物を提供する、癌、自己免疫疾患または感染症と関連のある抗原が含まれうる。1つの態様において、組成物は薬学的組成物である。組成物は、癌、自己免疫疾患または感染症と関連のある1つまたは複数の抗原を含む、タンパク質の治療的または予防的有効量を含みうる。有効量とは、例えば、T細胞を活性化することにより、免疫応答を誘発または増強するのに十分な量のことである。T細胞の活性化の指標の一つは、以下の実施例の項に記載するような、細胞毒性アッセイまたはインターフェロン-γ放出アッセイである。いくつかの態様において、組成物はワクチンである。
本発明はまた、タンパク質またはアジュバントを対象に送達するための、対象における抗原に対する免疫応答を誘発するための、および対象における状態を治療または予防するための方法も提供する。これらの方法は、対象に本発明の組成物を投与することを含む。投与は以下の記載の通りに行いうる。
治療(treatment)には予防および治療法(therapy)が含まれる。予防または治療は、単一時点または複数時点における単回の直接注射によって実施しうる。投与をほぼ同時に複数部位に行うこともできる。罹患生物または対象には、ヒト、ウシ、ウマ、イヌ、ネコ、ブタおよびヒツジなどの哺乳動物が含まれる。罹患生物または対象はヒトであることが好ましい。
本発明の組成物は、癌細胞または感染細胞を標的とする抗原特異的免疫応答の発生を促進するために用いうる。本発明のある種の好ましい態様では、樹状細胞またはその前駆細胞を抗原提示細胞(APC)として用いる。樹状細胞は非常に作用の強いAPC(BanchereauおよびSteinman、Nature 392: 245-251、1998)であり、予防的または治療的な免疫を誘発させるための生理的アジュバントとして有効なことが示されている(TimmermanおよびLevy、Ann. Rev. Med. 50: 507-529、1999を参照のこと)。一般に、樹状細胞はその典型的な形態(原位置で星状であり、インビトロで顕著な細胞質突起(樹状突起)が認められる)、ならびに、標準的なアッセイを用いた評価でB細胞(CD19およびCD20)、T細胞(CD3)、単球(CD14)およびナチュラルキラー細胞(CD56)の分子マーカーが存在しないことに基づいて同定しうる。当然ながら、樹状細胞を、インビボまたはエクスビボの樹状細胞には一般に認められない特定の細胞表面受容体またはリガンドを発現するように操作することもでき、このような改変樹状細胞を本発明では考慮している。樹状細胞に代わるものとして、分泌小胞を含む樹状細胞(エキソソームと呼ばれる)をワクチンに用いることもできる(Zitvogelら、1998、Nature Med. 4: 594-600)。

本実施例では、タンパク質-ミクロスフェアおよびタンパク質-アジュバント-ミクロスフェア製剤の調製について述べる。
これらのミクロスフェア製剤を、アジュバントの非存在下で、疎水的イオン対形成(HIP)法を用いて調製した。3mgの凍結乾燥タンパク質を2.7mlの超純水中に溶解した。このタンパク質溶液に対して、0.3mlの100mM CaCl2溶液および55μlの0.1M HClを添加し、pHをpH 3〜5の範囲に低下させた。ボルテックス混合により、タンパク質を有機相である4.3mM AOT(ドキュセートナトリウム)ジクロロメタン溶液中に抽出した。タンパク質を含むこの有機相を遠心処理によって水相から分離した。水相を廃棄し、DCMの添加によって有機相の容積を10mlに増やした。続いて、PLGポリマー(300mgのRG502H;Boehringer Ingelheim GmbH(Ingelheim、ドイツ))を溶媒に溶解した。ミクロスフェアの形成は、有機相中にあるタンパク質およびポリマーを400mlの5%PVA(ポリビニルアルコール)水溶液に添加し、Silverson混合機を用いて9000rpmで約1分間処理することによって行った。ミクロスフェアを硬化させるために2時間〜3時間ゆっくりと攪拌した後に洗浄し、遠心処理によって回収した。凍結および凍結乾燥の前にマンニトールを添加剤として添加した。
これらのミクロスフェアには、タンパク質(Mtb8.4;本明細書ではDPVとも称する)およびアジュバント(MPL)の両方を含めた。共封入の間、タンパク質とMPLとの比は約1:1に固定した。この製剤は、タンパク質抽出後かつプロセス媒体(すなわち、400mlの5%PVA)への添加前に有機相に3mgのMPLを添加した点を除き、MJ071b/MJ087bと同じやり方で調製した。
これらのミクロスフェアには、タンパク質(Mtb8.4)およびアジュバント(MPL)の両方を含めた。それらを共封入する際のタンパク質とMPLとの比は固定し、1:1とした。この製剤は、MPLを添加した点を除き、MJ071b/MJ087bと同じ一般的なやり方で調製した。、MPL(3rng)は内水相を介して組み入れた。すなわち、まず3mgのMPLを1.0mlの水中に激しいボルテックス混合によって分散させた。PLGポリマーをDCMを含むタンパク相に添加した後に、水相中にあるこのMPLの1mlを添加し、30秒間のボルテックス混合によって乳化させた。この一次エマルションを前と同じように400mlの5%PVA中で乳化させた。
これらのミクロスフェアにはタンパク質(Mtb8.4)およびアジュバント(MPL)の両方を含めた。タンパク質とMPLとの比は固定し、0.37とした。この製剤は、3mgのMPLをPVAの代わりに400mlのプロセス媒体に添加した点を除き、MJ071b/MJ087bと同じやり方で調製し、エマルションを9000rpmで2分間〜1/2分間混合した後に、さらに300mlの超純水を添加することによって分散物をさらに希釈した。
本実施例には、150mM Tris、pH 8.1および0.01%Tween 20から構成される放出媒体へのDPVタンパク質のインビトロ放出を示す。DPVは9キロダルトンのタンパク質であり、pIは6.5である。図1は、DPVの放出を、5種類のミクロスフェア製剤に関して、時間の関数としてプロットしたものを示している。製剤には、JA-024、40%ミリスチン酸エチル(C14)(菱形印);AS-011(四角印);AS-012、15%コレステロール(三角印);AS-014、20%カプリン酸エチル(C10)(×印);AS-013、20%ステアリン酸エチル(C18)(星印);およびJA-002、RG-502(+印)を含めた。
本実施例には、HIP法を用いて調製したMtb8.4タンパク質ミクロスフェアが、強力なアジュバントとの混合物およびタンパク質のみよりも強いCTL応答を誘発したことを示す。マウス脾細胞の1回のインビトロ刺激によるクロム放出アッセイを用いた個々のマウスに関するCTL応答を図2A〜図2Cに示している。マイクロカプセル封入されたタンパク質(図2A)による免疫処置を受けたマウスでは、タンパク質+MPL/サポニン(図2B)およびタンパク質のみ(図2C)によって誘発された応答と比べて、最も強く最も一貫した免疫応答が誘発された。マウスは、0日目および21日目に5μgのタンパク質で皮下に免疫処置された。脾臓を35日目に採取した。未処置マウスでは特異的溶解は観察されなかった。Mtb8.4タンパク質-ミクロスフェアはタンパク質+MPL/サポニン群またはタンパク質のみの群(図2A〜図2C)のいずれよりも強く一貫したCTL応答を誘発した。
本実施例では、取り出した日に得た血清からの抗体応答(IgG1)を測定した実験について述べる。図3A〜図3Cに示されているように、本発明のHIP法を用いて調製したタンパク質ミクロスフェアは、5匹の被験マウスのすべてにおいて強い抗体応答を誘発した(図3A)。これらの応答はタンパク質のみによって誘発されたものよりも有意に強く(図3B)、MPL/サポニンアジュバント混合物によって誘発されたものと同程度であった(図3C)。各群5匹ずつのマウスに対して0日目および21日目に5μgのタンパク質を皮下に免疫処置した。血清を35日目に採取し、特異的抗体レベルをELISAによって測定した。個々のマウスによる結果を示している。
本実施例には、本発明のタンパク質ミクロスフェア製剤が免疫応答の誘発に有効であるというさらなる証拠を提供する。図4は、Mtb8.4タンパク質ミクロスフェアがCTL応答を誘発する能力と、DNA免疫処置およびタンパク質+MPL/サポニンアジュバント混合物によって誘発される能力と比較した、マウスでの実験の結果を示している。CTL応答を、脾臓をプールしたマウス群に関して(n=7)、マウス脾細胞の単回インビトロ刺激によるクロム放出アッセイを用いて測定した。マウスに対して0日目および21日目に15μgのタンパク質を皮下に免疫処置するか、または50μgのDNAを筋肉内に免疫処置した。脾臓を43日目に採取した。塗りつぶした記号はMtb8.4 DNAによる形質導入を受けた標的の溶解を表している。白抜き記号は対照EL4標的の溶解を表している。図4に示されているように、タンパク質-ミクロスフェアを投与した群では強いCTL応答が測定された(丸印)。この反応はタンパク質+MPL/サポニン群で測定された反応(三角印)よりも有意に強く、驚いたことに、DNAを投与した群での反応と同程度であった(四角印)。

本実施例には、MPLなどのアジュバントの存在下でのタンパク質ミクロスフェアの共封入の、Th1およびB細胞媒介性免疫に対する相乗効果を示す。
本実施例には、Mtb8.4-EL4標的細胞の特異的CTL媒介性溶解の、CD4+T細胞のIFNγ放出の、およびCD8+T細胞のIFNγ放出(それぞれ図12、図13および図14)の誘導に対する、MJ-082タンパク質-MPL共封入物の用量反応性を開示する。CTLアッセイおよびICCアッセイを本明細書の別の箇所に記載した通りに行ったところ、モノホスホリルリピドA(MPL(登録商標))などのアジュバントをMtb8.4-ミクロスフェア中に共封入した場合のCD4およびCD8 T細胞免疫応答に対する相乗効果がさらに裏づけられた。
本実施例には、本発明による最適なワクチン製剤のいくつかの様相を示す。第1に、タンパク質(Mtb8.4)と別のアジュバント(RC529)との共封入は、CD8+およびCD4+T細胞応答、CTL、IFN-γおよび抗体(IgG1およびIgG2a)免疫応答を誘発するのに有効である。このため、共封入されたタンパク質およびアジュバントに対する広範囲にわたる免疫応答は単一のアジュバントには限定されない。第2に、タンパク質およびアジュバントを別個に封入して混合しても、広範囲にわたる免疫応答は誘発されない。しかし、第3に、抗原およびアジュバントを同じ粒子中に封入すると、別個の粒子中に封入して混合した場合よりも強い応答が誘発される。第4に、ミクロスフェア中に封入されたアジュバント(MPLまたは529)は、標準的水性製剤(すなわち、リポソーム)中に製剤化されたアジュバントと少なくとも同程度に有効である。ミクロスフェア中への封入を介してアジュバントを送達するこの新規な手段は、従来のアジュバント送達に代わる魅力的な選択肢となる。
ミクロスフェアの調製:
添加した成分の違いを除き(Mtb8.4タンパク質の有無、MPL(登録商標) アジュバントの有無、RC529アジュバントの有無)、本実施例のための種々の製剤の調製には同じミクロスフェア調製法を用いた。これは上記の実施例1、実施例6および実施例7に記載したものと同じ方法である。簡潔に述べると、ドキュセートナトリウムのジクロロメタン溶液を用いて水相酸性溶液の抽出を行った。タンパク質を有する製剤に関しては、水溶液中にあるMtb8.4タンパク質(3mg)を有機相中に抽出した。続いてアジュバント(MPLまたは529)を有機相に必要に応じて添加し、その後に300mgのPLG(RG502H)を添加した。続いて有機相を400mlの5%PVA中にてSilversonホモジナイザーを用いて乳化させた。ヒュームフード内で数時間攪拌することによって溶媒を抽出した。続いてミクロスフェアを数回洗浄し、凍結乾燥した。凍結乾燥の前にマンニトールを添加した。
ミクロスフェアのタンパク質含量およびアジュバント含量を、HPLC分析またはアミノ酸分析およびバートレット無機リンアッセイを用いて決定した。サイズ分布の測定にはHoriba LA920を用いたほか、走査型電子顕微鏡による視覚的判定によっても行った。放出動態は、10mg〜20mgの製剤を100mM Tris緩衝液中に分散させ、上清から経時的に試料を採取した上で、逆相HPLCによってタンパク質濃度を定量することによって測定した。
7匹ずつのC57Bl/6からなる群に対して表8.1に記載した通りに免疫処置を行った。対照群のマウスには、未処置(naive)、Mtb8.4タンパク質のみ、Mtb8.4-DNAおよびMtb8.4タンパク質-ミクロスフェアを含めた。アジュバントMPLおよび529の各々に対する被験群は5つとし、試薬のさまざまな組み合わせをカバーした。Mtb8.4タンパク質の用量は5μgとした。MPLまたは529に関するアジュバント目標用量はマウス1匹当たり5μgとした。しかし、共封入ミクロスフェアによって投与されたアジュバントの実際の量を測定して設定した。すなわち、Mtb8.4-MPL共封入ミクロスフェアおよびMtb8.4-529共封入ミクロスフェアを投与されたマウスに関するアジュバント用量はそれぞれマウス1匹当たり4.0μgおよび6.5μgであった。したがって、MPLを投与したすべての群はマウス1匹当たり4.0μgを投与され、529を投与したすべての群はマウス1匹当たり6.5μgを投与された。
脾細胞を、CTLアッセイ、CD8+およびCD4+T細胞応答ならびにIFN-γに関するICCアッセイに用いた。Mtb8.4特異抗体の測定には血清を用いた。これらの方法は上記の実施例1〜7に用いたものと同じである。
ミクロスフェアの特徴分析:
ミクロスフェアのインビトロ物理化学分析の結果を表2に示している。タンパク質ミクロスフェア(a)、タンパク質-MPL共封入ミクロスフェア(d)およびタンパク質-529共封入ミクロスフェア(e)に関するMtb8.4タンパク質のコア充填率(core-loading)は同程度であり、0.62%〜0.87%の範囲であった。MPL-ミクロスフェア(b)およびタンパク質-MPL共封入ミクロスフェア(d)に関するMPLのコア充填率はそれぞれ0.64%および0.70%であり、一方、529ミクロスフェア(c)およびタンパク質-529共封入ミクロスフェア(e)に関する充填率はそれぞれ0.71%および0.80%であった。この一連のミクロスフェアはいずれも同程度のサイズであり、直径は約1μmであった。表2には、2時間時点で放出されたタンパク質の量がタンパク質ミクロスフェアについては3%であり、タンパク質-MPL共封入ミクロスフェアおよびタンパク質-529共封入ミクロスフェアについては17%および21%であったことも示されている。
1回のみの免疫処置の後に採取した脾細胞に対して行ったICCSアッセイによるCD8+およびCD4+T細胞応答の結果を図15A〜図15Dに示している。図15Aおよび図15Bは、MPL群および対照群に関するMtb8.4特異的CD8およびCD4 T細胞応答を表しており、一方図15C〜図15Dは529群および対照群に関する結果を表している。4つの図はすべて、MPLまたは529のいずれかを用いたタンパク質-アジュバント共封入ミクロスフェアにより、最も強いCD8 T細胞応答および最も強いCD4 T細胞応答が生じたことを示している。CD8およびCD4 T細胞応答両方について、タンパク質-MPL共封入ミクロスフェアの方がタンパク質-529共封入ミクロスフェアよりも強い応答を誘発した。Mtb8.4タンパク質のみ、タンパク質+MPL(登録商標)アジュバントおよび未処置のマウスのいずれによっても、CD8 T細胞応答もCD4 T細胞応答も本質的には生じなかった。図15A〜図15Dはまた、アジュバント-ミクロスフェアをタンパク質のみ(F群およびK群)またはタンパク質-ミクロスフェア(H群およびM群)と混合すると、CD8およびCD4 T細胞応答の両方が、-AF脂質製剤中に製剤化されたアジュバントによって生じたもの(E群、J群、G群およびL群)よりも増強されることも示している。すなわち、アジュバント-ミクロスフェアは、裸の(naked)タンパク質、ならびにMPLおよび529を用いたタンパク質-ミクロスフェアのいずれにも有効なアジュバント製剤である。
一次免疫処置から2週間後に採取した脾細胞からのIFN-γ放出を、図16A(MPL群および対照群)ならびに図16B(529群および対照群)に示している。図16A〜図16Bは、CD8およびCD4 T細胞応答と同じく、最も強いIFN-γ応答がタンパク質-MPL共封入ミクロスフェアおよびタンパク質-529共封入ミクロスフェアによって誘発され、2つの応答の中ではタンパク質-MPL共封入ミクロスフェア製剤の方が同じく強い応答を誘発することを示している。このアッセイにおいて、タンパク質ミクロスフェアを、アジュバントもしくはアジュバント-ミクロスフェアと混合して、または混合せずに投与された群(D、G、H、LおよびM群)はすべて同程度の応答を誘発し、これはタンパク質のみおよびタンパク質+アジュバント(B、E、F、JおよびK群)によって誘発されたものよりも明らかに強かったが、共封入群(IおよびN群)によって誘発されたものよりはかなり弱かった。
二次免疫処置から2週間後に採取した血清に関して測定したIgG1およびIgG2b抗体応答を、対照群およびMPL群については図17A〜図17Bに示し、対照群および529群については図17C〜図17Dに示した。IgG1については16000:1希釈、IgG2bについては4000:1希釈で測定したODを、応答として示した。図17Aおよび図17Cは、ミクロスフェア中に封入されたタンパク質を投与された群(G、H、I、L、MおよびN群)で最も強いIgG1応答が得られたことを示している。DNAおよびタンパク質のみによる免疫処置はIgG1応答をほとんどまたは全く誘発せず、タンパク質をMPL-AFおよび529-AFのいずれかと混合した場合は非常に弱いIgG1応答が得られた。図17Bは、タンパク質-ミクロスフェアをアジュバントと混合した場合(G、HおよびI群)に最も強いIgG2b応答が得られたことを示している。これらの応答は、DNA、タンパク質のみ、タンパク質とMPL-AFとの混合、およびタンパク質とMPL-ミクロスフェアとの混合によって誘発されたものよりもはるかに強かった。興味深いことに、脂質中に製剤化したアジュバントとミクロスフェア中に製剤化したアジュバントを比較した図17A〜図17D中のいずれの比較においても(例えば、IgG1およびIgG2bに関するEとF、GとH、JとK、およびLとMとの比較)、アジュバント-ミクロスフェア群はより高レベルの抗体を誘発した。このことは、アジュバント-ミクロスフェアがアジュバントの有効な製剤の一つであり、おそらくは-AF脂質製剤よりも優れていることを示している。
本実施例には、タンパク質およびアジュバントの共封入の代替的方法によっても、強く広範囲にわたる免疫応答を誘発しうるミクロスフェアを作製しうることを示す。詳細には、ダブルエマルション法によってミクロスフェアを作製し、シングルエマルション法を用いて調製した共封入ミクロスフェアと比較した。さらに、本実施例には、有効なミクロスフェアの作製にHIPを利用する、代替的でしかもより製造しやすい手法も示す。この方法では、水性タンパク質溶液をダブルエマルション工程における内水相として、溶媒中のHIP剤とともに用いる。タンパク質はおそらく、一次エマルション段階の間に溶媒中に抽出されると考えられる。
Mtb8.4-MPL共封入ミクロスフェアを、いくつかの特徴について共通する3種類の異なる技法を用いて調製した。すなわち、用いた製剤にはすべて、3mgのMtb8.4タンパク質、3mgのMPL(登録商標)アジュバント、300mgのRG502Hポリマー、プロセス媒体としての400mlの5%PVAを用い、ミクロスフェアの調製にはSilverson混合機を混合速度9000rpmで用いた。ロット番号MJ102の第1のミクロスフェア製剤は、実施例1、実施例6および実施例7に記載された通りに調製した。簡潔に述べると、Mtb8.4タンパク質を水溶液からドキュセートナトリウム-ジクロロメタン(DCM)溶液中に疎水的イオン対形成を介して抽出した。続いて水溶液を廃棄した。ポリマーおよびMPL(登録商標)アジュバントをDCMに添加してミクロスフェアを調製した。ロット番号MJ107の第2の製剤には、HIPおよび/またはドキュセートナトリウムを用いずにタンパク質を有機相中に可溶化するために、改変した溶媒系を用いた。Mtb8.4は単独ではDCM中には不溶性である。このため、Mtb8.4を酸性pHの水溶液から凍結乾燥し、ジメチルスルホキシド(DMSO)中に溶解して、それに同容積のDCMを添加した。続いてポリマーおよびMPL(登録商標)アジュバントを溶媒相に溶解し、ミクロスフェアを調製した。ロット番号MJ108の第3の製剤には、ダブルエマルション法の変法を用いた。内水相は、MJ102の抽出時と同じく、Mtb8.4タンパク質の酸性pHにある1.0ml水溶液とした。有機相はMJ102の抽出時と同じく約8mgのドキュセートナトリウムを含み、さらにポリマーおよびMPL(登録商標)アジュバントを含むDCMとした。一次エマルションは、溶媒相にあるタンパク質溶液をボルテックスを用いて激しく混合することによって形成させた。タンパク質はおそらく、MJ102の場合と同じく、疎水的イオン対形成を介して有機相に抽出されたと考えられる。しかし、この技法は、MJ102工程の面倒な面である水相の分離および廃棄の必要がない点で、より簡単である。続いて、ミクロスフェアを上記の一次エマルションを用いて調製する。
ミクロスフェアのタンパク質含量およびアジュバント含量を、HPLC分析またはアミノ酸分析およびバートレット無機リンアッセイを用いて決定した。サイズ分布の測定にはHoriba LA920を用いたほか、走査型電子顕微鏡による視覚的判定によっても行った。放出動態は、10mg〜20mgの製剤を100mM Tris緩衝液中に分散させ、上清から経時的に試料を採取した上で、逆相HPLCによってタンパク質濃度を定量することによって測定した。
7匹ずつのC57Bl/6からなる群に対して、マウス1匹当たり5μgの封入Mtb8.4タンパク質により免疫処置を行った。MPL(登録商標)アジュバントに関する目標用量はマウス1匹当たり5μgとした。MJ102、MJ107およびMJ108に関する実際の用量はマウス1匹当たりそれぞれ4.1μg、4.3μgおよび6.4μgであった(表3)。対照群のマウスには、未処置、Mtb8.4タンパク質+MPL-AF、タンパク質+-AF媒体(脂質水溶液)およびMtb8.4-DNAを含めた。全タンパク質製剤は、尾の基部への皮内経路によって投与し、一方、DNAは筋肉内に投与した。各群のマウス3匹または4匹を一次免疫処置から2週間後に取り出して脾臓および血清を採取し、評価のためにプールした。残りの1群当たり3匹または4匹のマウスには一次免疫処置から3週間後に追加免疫処置を行い、その2週間後に取り出した。
脾細胞を、CTLアッセイ、CD8+およびCD4+T細胞応答ならびにIFN-γに関するICCSアッセイに用いた。Mtb8.4特異抗体の測定には血清を用いた。これらの方法は上記の実施例1〜実施例7に用いたものと同じである。
ミクロスフェアの特徴分析:
タンパク質含量およびMPL(登録商標)含量(「コア充填率」、CLと略記)ならびに封入効率を表3に挙げた。MJ102、MJ107およびMJ108に関してタンパク質の封入効率は同程度かつ妥当であり、64%〜76%の範囲であった。MPL(登録商標)アジュバントの封入効率も同程度かつ妥当な値であり、62%〜79%の範囲であった。アジュバントと抗原との比もいずれも均一に近く、0.81〜1.28の範囲であった。
単回免疫処置から2週間後に測定したCTL応答を図18に示している。Mtb8.4タンパク質+MPL(登録商標)アジュバントを投与されたマウスは明らかな特異的CTL応答を示さなかった。同様に、未処置マウスおよびタンパク質+-AF媒体(脂質)を投与されたマウスも特異的CTL応答を生じなかった。しかし、有効性が以前に判明しているMtb8.4タンパク質-MPL共封入製剤(実施例1、実施例6、実施例7および実施例8を参照)であるMJ102は、この場合もCTLを誘発することに成功した。同様に、ダブルエマルション法を用いて調製した共封入ミクロスフェアMJ108も同程度の強さのCTL応答を誘発した。2時間時点でほぼ80%が放出された共封入製剤であるMJ107は、この実験におけるこの時点では強いCTL応答を誘発しなかった。
一次免疫処置または一次および二次免疫処置のいずれかを受けたマウスについて測定した脾細胞培養物からのIFN-γ放出を図19に示している。図19は、タンパク質+MPL(登録商標)アジュバント、タンパク質+-AF媒体を投与されたマウスおよび未処置マウスではIFN-γが本質的に全く誘発されなかったことを示している。DNAを投与されたマウスは背景値を辛うじて上回る程度のIFN-γ応答を誘発した。これに対して、MJ102およびMJ108ミクロスフェアはいずれも、1回または2回の免疫処置を受けたマウスにおいて強く同程度のIFN-γ応答を誘発した。CTL応答についても、MJ107ミクロスフェアは初期放出動態の低い2つのミクロスフェア製剤よりもIFN-γ誘発が弱かったが、これは背景値を上回るレベルであり、DNA群のものと同程度であった。
図20および図21はそれぞれ、マウスにおいて誘発されたMtb8.4特異的IgG2bおよびIgG1抗体応答を示している。図20は、3種類のミクロスフェア製剤が最も強いIgG2b応答を誘発したことを示している。タンパク質+MPL(登録商標)アジュバントおよびMtb 8.4-DNAはミクロスフェアよりもIgG2b誘発レベルが低かった。CTLおよびIFN-γ応答の場合と同じく、MJ102およびMJ108はMJ107よりも強い応答を誘発した。同様に、図21は3種類のミクロスフェア製剤が最も強いIgG1特異抗体応答を誘発したことを示している。タンパク質+MPL(登録商標)アジュバントおよびMtb8.4-DNAはミクロスフェアよりもIgG1誘発レベルが低かった。他の免疫応答測定値とは異なり、3種類のミクロスフェア製剤に対するIgG1応答はすべて同程度であった。
本実施例には、好ましい結核抗原である72fの送達の最適化に関係するいくつかの点を示す。72fは3つのポリペプチド:RA12、TbH9およびRA35の融合体である。本実施例は以下のことを示す:(1)ミクロスフェア中に封入された組換え72f(r72f)タンパク質は、強いCD4+およびCD8+T細胞応答、CTLおよび抗体応答を誘発しうる;(2)r72fとアジュバント(MPL)との共封入物はアジュバントを含まないr72f-ミクロスフェアよりも強い免疫応答を誘発する;(3)ダブルエマルションおよびシングルエマルション製剤はいずれも最も強い免疫応答を誘発する;および(4)最適なミクロスフェアによって誘発されるCD4+T細胞応答(Elispot)はタンパク質+AS-2(MPLおよびサポニン)、タンパク質+MPLおよびタンパク質のみによって誘発されるものよりも著しく強い。
72fタンパク質ミクロスフェアをシングルエマルション法およびダブルエマルション法を用いて調製した(表5)。表1に記載したすべての製剤は、300mgのPLGポリマー、3mgの72fタンパク質、10mlの有機溶媒(表5)を有し、ミクロスフェアの作製にはSilverson混合機を9000rpmで用いた。製剤の違いには、シングルエマルション(SE)工程であるかダブルエマルション(DE)工程であるか、ポリマーの末端基がカルボン酸であるかエステル基であるか、ポリマー末端基の頻度(すなわち、分子量:約10kDaに対して約40kDa)、DCMであるかDCM:DMSO(35:65)であるか、MPL(登録商標)アジュバントの添加の有無、プロセス媒体の容積(280mlであるか400ml)および安定剤(CMC 1.4%であるかPVA 5%)が含まれる。MPL(登録商標)アジュバントをAM123では共封入した点を除き、AM123はAM117と同じ様式で調製されたことに注意されたい。同様に、AM124ではMPL(登録商標)アジュバントを共封入した点を除いて、AM124はAM120と同じ様式で調製された。
ミクロスフェアのタンパク質含量およびアジュバント含量を、アミノ酸分析およびBartlett無機リンアッセイによって決定した。サイズ分布の測定にはHoriba LA920を用いたほか、走査型電子顕微鏡による視覚的判定によっても行った。
マウス1匹当たり10μgの封入72fタンパク質を3週間間隔で尾の基部に接種することで(すなわち、ID)、3匹ずつのC57BL/6マウスの群を2回免疫処置した。免疫応答の発生に関する評価のために、2回目の免疫処置から2週間後にマウスから脾臓および血清を採取した。MPL(登録商標)アジュバントの目標用量はマウス1匹当たり10μgとした。タンパク質およびMPL(登録商標)アジュバントを共封入した2種類のミクロスフェア製剤であるAM123およびAM124の実際の用量はそれぞれ5μgおよび12μgであった。対照群には、未処置マウス、タンパク質のみ(10μg)、水中油型エマルション中のタンパク質+AS-2(MPL(登録商標)およびサポニン)、ならびにタンパク質+MPL(登録商標)-SEアジュバントを含めた。
脾細胞をElispotアッセイおよびCTLアッセイに用いた。Elispotアッセイに関しては、細胞をTbH9タンパク質、RA12タンパク質、RA35タンパク質、およびRA12に対するCD8 10アミノ酸Dbエピトープによって刺激した。CTLアッセイに関しては、Ra12タンパク質による形質導入を行ったEL4細胞で細胞を刺激した。血清中のTbH9タンパク質、RA12タンパク質およびRA35タンパク質に対する抗体の測定には抗原特異的ELISAを用いた。
ミクロスフェアの特徴分析:
タンパク質含量およびMPL(登録商標)含量(「コア充填率」または「CL」)を表6に挙げている。タンパク質のコア充填率は同程度であり、0.63%〜1.35%の範囲であった。AM123およびAM124に関するMPL(登録商標)アジュバントのコア充填率は0.68%および0.93%であり、このため、ある一定のタンパク質用量(すなわち、10μg)では、AM124の方がAM123よりもほぼ2倍のアジュバントが投与される。表6は、直径約1μmにおいてミクロスフェアのサイズも同様であったことを示している。
Elispotアッセイの結果は、マウス3匹からのプール脾細胞に対する三重反復試験に基づく。表記した数はウェル当たりのIFN-γ Elispots数を表している。アッセイの陰性対照として、各群の各マウスからの細胞を培地のみによって刺激した。培地で刺激した各ウェルにはわずかな(<15)陽性スポットしかなかった。これに対して、TbH9組換えタンパク質(10μg/ml)で刺激した細胞は陽性応答を示したが、その程度は群間に違いがあった。すなわち、未処置マウス(「培地」行)およびタンパク質のみを投与されたマウスでは、TbH9特異的陽性応答が誘発されなかった。AS-2とともに製剤化したタンパク質、ならびにタンパク質+MPL(登録商標)-SEアジュバントはいずれも陽性のTbH9特異的応答を誘発し、ウェル当たりのスポット数はそれぞれ52個および154個であった。すべてのタンパク質ミクロスフェア群でも陽性のTbH9特異的応答が誘発され、AM116、AM117、AM118およびAM119に関するウェル当たりの平均スポット数はそれぞれ43個、41個、77個および69個であった。しかし、タンパク質ミクロスフェア製剤AM120(MPL非含有)ならびにAM123およびAM124(MPL共封入)は非常に強いTbH9応答を誘発したために、ウェル全体にスポットの集密化がみられた。AM117(MPL非含有)とAM123(MPL共封入)との比較により、MPL共封入によって生じた免疫応答の顕著な増強が明らかに示された。AM120(MPL非含有)応答はすでに最大限であるため、AM124(MPL共封入)との間に差は認められなかった。未処置マウスと比較してRa35特異的応答もRA12特異的応答も観察されなかった。Ra12 CD8+T細胞エピトープ(ペプチド番号86〜95)に対する最も強い応答も、ミクロスフェアを用いた場合に認められた。この読み取りに関しては、8種類のうち6種(AM116、AM117、AM119、AM120およびAM123)では応答がほぼ均一であったが、AM118およびAM124では応答が幾分弱かった。すなわち、タンパク質ミクロスフェアはTbH9に対する最も強いCD4+T細胞応答を生じるとともに、Ra12エピトープに対してもCD8+T細胞応答を生じさせた。これらの応答はタンパク質のみの場合よりもはるかに強く、MPLおよびAS-2アジュバント系によって誘発されたものよりも強かった。
2回目の免疫処置から2週間後にRa12に対する二次CTL応答を測定した。72fタンパク質+MPL(登録商標)アジュバントを投与されたマウスは明らかな特異的CTL応答を示さなかった。未処置マウスおよびタンパク質のみを投与されたマウスでは特異的溶解は観察されなかった。タンパク質+AS-2はマウス3匹中3匹でCTL応答を生じさせ、これは背景値に対して中程度〜高いレベルであった。AM118を唯一の例外として、すべてのタンパク質-ミクロスフェア製剤がCTL応答を誘発した。最も強く最も定常的なCTL応答は、AM123、AM116、AM117およびAM119の場合であった。AM117(MPL非含有)とAM123(MPL共封入)との比較では、この場合もMPLの共封入によって免疫応答がかなり増強することが示された。すなわち、タンパク質ミクロスフェアは、タンパク質のみおよびタンパク質+MPL(登録商標)アジュバントよりも明らかに強いCTL応答を生じさせた。
本実施例には、組換えher-2/neu細胞内ドメイン(rICD)タンパク質ミクロスフェアが、有効な腫瘍防御をもたらしうることを示す。複数種の(n=4)ICDタンパク質ミクロスフェア製剤を調製した。これらはすべて同程度のサイズであり、同一ロットのタンパク質を用いて作製され、PLGポリマーの一種をミクロスフェアマトリックスとして含んでいた。しかし、ミクロスフェアの作製には異なるポリマー、添加物および技法が用いられ、その結果、インビトロまたはインビボでの測定によれば異なる特性を有するミクロスフェアが得られた。詳細には、これらのアプローチは、製造工程時および最終ミクロスフェア産物の双方において、タンパク質とそれ自体、ポリマー、溶媒との相互作用の程度が変化するように、さらには溶質、ポリマー、安定剤および水の相互同士の相互作用が変化するように設計した。これらの製剤にはいずれもアジュバントを含めなかった。これらの4種類の製剤を腫瘍防御モデルにおいて評価した。
4種類の異なるICDタンパク質ミクロスフェア製剤を調製し、特徴分析して、腫瘍防御モデルにおける評価を行った。これらの製剤には共通した特徴がいくつかあった。すなわち、3種類の製剤はいずれも、2.5mgのICDタンパク質、300mgのPLGポリマー、プロセス媒体としての5%PVAを含み、ミクロスフェアの調製にはSilverson混合機を用いた。これらのミクロスフェアの製剤パラメーターの違いを表7にまとめている。これらには、シングルエマルション法であるかダブルエマルション法であるか、ポリマーの末端基の種類(カルボン酸「H」であるかエステル末端基であるか)、ポリマー末端基の頻度(すなわち、ポリマー分子量が約10kおよび約40K)、用いた溶媒系(DCMおよびDMC:DMSO(1:3.3))、プロセス媒体の容積、用いた混合速度(7000rpmおよび9000rpm)、および添加物(ステアリン酸エチル)の添加が含まれる。
ミクロスフェアのタンパク質含量をアミノ酸分析によって決定した。サイズ分布の測定にはHoriba LA920を用いたほか、走査型電子顕微鏡による視覚的判定によっても行った。
マウス1匹当たり25μgの封入ICDタンパク質を3週間間隔で接種することで、8匹ずつのC57Bl/6マウスの群を2回免疫処置した。対照群には、(この系に適していることが以前に判明しているアジュバントである)モンタニド中に製剤化された25μgのICD、ICD-DNAを投与するか未処置マウスとした。タンパク質製剤は皮内経路によって投与し、DNAは筋肉内に投与した。
2回目の免疫処置から2週間後にマウスにEL4-Her-2/neu腫瘍細胞を免疫処置した。腫瘍サイズを6週間にわたって定期的に測定した。
ミクロスフェアの特徴分析:
AM049、AM050、AM051およびAM051 ICDミクロスフェアに関するタンパク質含量(「コア充填率」、CLと略記)およびミクロスフェアの直径を表8に挙げた。表8は、これらのミクロスフェア製剤のコア充填率が同程度で0.96%〜1.24%の範囲であり、サイズも同程度であることを示している。
図23〜図29は腫瘍防御実験の結果を示している。図22は未処置マウスにおける腫瘍の進行を示している。マウス8匹中6匹では腫瘍の移植から間もなく腫瘍増殖がみられ、マウス8匹中7匹は実験終了時には顕著な腫瘍増殖を示した。図23および図24は、モンタニド中にあるICDタンパク質およびICD-DNAのそれぞれを投与された陽性対照群における腫瘍増殖を示している。図23は、この腫瘍モデルで有効なことが以前に判明しているアジュバントであるモンタニドアジュバント中に製剤化されたICDが、未処置マウスと比較して防御の増強を示したことを示している。第6週までには、モンタニド群のマウス8匹中3匹には顕著な腫瘍増殖がみられた。以前の実験で最も一貫性がみられた陽性対照であるICD-DNAは顕著な腫瘍防御をもたらした(図24)。DNA免疫処置を受けたマウスでは実験がほぼ終了するまで測定可能な腫瘍増殖はみられず、終了時になってマウス1匹の腫瘍が増殖していることが明らかになった。

(図3)図3A〜図3Cは、本発明のHIP法を用いて調製したMtb8.4タンパク質ミクロスフェアが抗体応答を誘発し(IgG1;図3A)、これがMPL/サポニンアジュバント混合物によって誘発された応答と同程度に強く(図3B)かつタンパク質のみによって誘発された応答よりも有意に強かった(図3C)ことを示したグラフである。個々のマウスによる結果を個別の線として示している。
(図4)脾臓がプールされたマウス群に関して(n=7)、マウス脾細胞の単回インビトロ刺激後のクロム放出アッセイを用いて測定したCTL応答を示している。塗りつぶした記号はMtb8.4 DNAによる形質導入を受けた標的の溶解を表している。白抜き記号は対照EL4標的の溶解を表している。タンパク質-ミクロスフェアを投与した群では強いCTL応答が測定された(丸印)。この反応はタンパク質+MPL/サポニン群で測定された応答(三角印)よりも有意に強く、かつ驚いたことに、DNAを投与した群での応答と同程度であった(四角印)。
(図5)細胞内サイトカイン(ICC)アッセイによって評価した、rMtb8.4により活性化された脾細胞培養物からのIFNγ放出を示している。個々のマウスの脾臓におけるCD8+IFNγ+細胞の比率を単回のSCまたはID免疫処置から14日後に測定した。MJ-071bは、MPLを含まないrMtb8.4が封入されたミクロスフェアを示している。
(図6)酵素結合免疫吸着アッセイ(ELISA)によって評価した、脾細胞培養物からのIFNγ放出を示している。脾細胞培養物(マウス3匹からプールした脾臓)を組換えMtb8.4によって72時間活性化した。MJ-071bは、MPLを含まないrMtb8.4が封入されたミクロスフェアを示している。

Claims (54)
- 生分解性高分子ミクロスフェア中に封入されたアジュバントおよび薬学的に許容される担体を含む、薬学的組成物。
- アジュバントとともにミクロスフェア中に共封入されたタンパク質をさらに含む、請求項1記載の薬学的組成物。
- アジュバントが、MPL、AS-2、サポニン、リン酸アルミニウム、リン酸カルシウム、アミノアルキルグルコサミニドリン酸、RC-529、RC-557、R544、イソツセラソール(isotucerasol)、細胞壁骨格、および/またはCpG含有オリゴヌクレオチドを含む、請求項1記載の薬学的組成物。
- タンパク質が、癌、自己免疫疾患または感染症と関連のある抗原を含む、請求項2記載の薬学的組成物。
