Abstract
Description
注目の遺伝子を組み込んだクローンの選択は、選択圧に対する耐性を付与する選択マーカーを使用することで実施される。選択マーカーの大部分は、例えば、ネオマイシン、カナマイシン、ハイグロマイシン、ゲンタマイシン、クロラムフェニコール、ピューロマイシン、ゼオシン、又はブレオマイシンなどの抗生物質に対する耐性を付与する。
注目のタンパク質の高発現性クローンについてのスクリーニングは、多量のタンパク質の存在を直接的に明らかにする方法によって多くの場合実施される。通常、ELISA又は免疫組織化学的染色などの免疫学的方法が、細胞内又は細胞培養上清中の生成物を検出するために適用される。これらの方法は、退屈で、高価で、時間がかかっていて、そして、多くの場合高スループット・スクリーニング(HTS)に適していない。加えて、発現タンパク質に反応する抗体が入手可能でなければならない。
a)細胞を、(i)毒性化合物に対する耐性を付与するか又は代謝的優位性を付与するフラグメントと融合させたジヒドロ葉酸レダクターゼ(DHFR)の機能的なフラグメントを含んでなるres-DHFRキメラ・タンパク質と、(ii)注目のタンパク質をコードする発現ベクターによってトランスフェクションするステップ;
b)上述の毒性化合物に耐性であるか、又は上述の代謝的優位性を獲得した細胞を選択するステップ;及び
c)DHFRに結合する蛍光化合物を用いて、ステップ(ii)で選択された細胞の蛍光発光をアッセイするステップ、
を含んでなり、ここで、毒性化合物に対する耐性を付与するか、又は代謝的優位性を付与する上述のタンパク質はDHFRでない。
a)本発明の方法による細胞をスクリーニングするステップ;及び
b)上述の細胞から細胞株を樹立するステップ、
を含んでなる。
a)上述の注目のタンパク質の発現を可能にする条件下、先の方法に従って獲得した細胞株を培養するステップ;及び
b)上述の注目のタンパク質を回収するステップ、
を含んでなる。
配列番号1及び2は、それぞれ、本発明によるPuro-DHFRマーカーの核酸及びポリペプチド配列に相当する。
配列番号3及び4は、それぞれ、ストレプトマイセス・アルボニガー(Streptomyces alboniger)ピューロマイシンN-アセチルトランスフェラーゼ(pac)の核酸及びポリペプチド配列に相当する。
配列番号7〜10は、本発明に従ってPuro-DHFRマーカーを構築するときに使用したプライマーに相当する(実施例1)。
配列番号11〜16は、QPCRによって遺伝子コピー数を検出するときに使用したオリゴヌクレオチドに相当する(実施例3)。
本発明は、res-DHFRとも呼ばれる新規な四機能性キメラ・マーカーの構築と特徴づけから生じる。より詳しく述べると、本発明は、Puro-DHFRとも呼ばれるres-DHFRポリペプチドを開示するが、それは、DHFRと、ピューロマイシンに対する耐性を付与するタンパク質であるピューロマイシンN-アセチルトランスフェラーゼ(pac)との間の融合タンパク質に相当する。
‐ピューロマイシン毒性化合物と組み合わせて選択マーカーとして;
‐MTX毒性化合物と組み合わせて選択マーカーとして;
‐増幅可能な遺伝子として;そして
‐顕微鏡とFACSの両方によって検出され得る容易に計測可能な代用として、
使用され得る。
本発明によるポリペプチドは、毒性化合物に対する耐性又は代謝的優位性のどちらかを付与するフラグメントに融合したジヒドロ葉酸レダクターゼ(DHFR)の機能的なフラグメントを含んでなるキメラ・タンパク質であり、ここで、上記の毒性化合物に対する耐性又は代謝的優位性を付与するフラグメントは、DHFR又はその断片でない。そのようなキメラ・タンパク質は、本明細書中で、さらに、「本発明によるポリペプチド」又は「res-DHFR」とも呼ばれるだろう。
5,6,7,8-テトラヒドロ葉酸+NADP+=7,8-ジヒドロ葉酸+NADPH
を触媒するポリペプチドの断片を指す。
本発明の別の側面は、本発明によるres-DHFRポリペプチドをコードする核酸に関する。
別の側面は、注目のタンパク質を生産するための、本発明によるres-DHFR核酸を含んでなる細胞の使用に関する。好ましくは、前述の細胞は、Puro-DHFRベクターを含んでなる。
a)res-DHFRをコードする発現ベクターで細胞をトランスフェクションするステップ;
b)前述の毒性化合物に耐性である細胞を選択するステップ;及び
c)DHFRに結合する蛍光化合物を用いて、ステップ(b)で選択された細胞の蛍光発光をアッセイするステップ、
を含んでなる。
そして、前述の注目のタンパク質の最高の発現を示す細胞の約1%〜約20%を選択できる。例えば、前述の注目のタンパク質の最高の発現を示す細胞の約1%、1.5%、2%、3%、4%、5%〜約15%、18%、又は20%を選択できる。好ましくは、前述の注目のタンパク質の最高の発現示す細胞が選択される。注目のタンパク質の発現に基づくこの選択は、ステップ(d)(すなわち、蛍光に基づく最後の選択)の最後の反復後に好ましくは実施される。
a)先の方法に従って細胞をスクリーニングするステップ;及び
b)上述の細胞から細胞株を樹立するステップ、

1.1. Puro-DHFR核酸の構築
本明細書中で以下に記載した構築物を、図1に描く。すべての構築物が、pGL3基礎プラスミド骨格(Promega)に基づいている。
安定したトランスフェクタントを選択するためのプロトコールを、図2に図式化する。
CHO-S-細胞を、チャイニーズハムスターから得、そして、無血清培地(Invitrogen/Gibco、La Jolla, CA)に順応させた。CHO-S DX11-F10は、無血清培地中の懸濁状態での培養に適合させた、DHFR欠損CHO DUKXB11細胞株に由来する細胞株である(Urlaub et al. 1980)。両細胞を、ProCho5(Lonza Biologies)中でごく普通に培養する。DXB11-F10細胞の培養用培地には、反対のものが示されない限り、ヒポキサンチン(100μM)とチミジン(16μM)を補った(HTサプリメント;Invitrogen/Gibco)。
選択のために、ピューロマイシンを10μg/mlにて使用し、そして、葉酸類似体であるメトトレキサート(Calbiochem)を50〜100nMの濃度にて使用した。
安定にトランスフェクションされた細胞プールを、125ml振とうフラスコ内に2.5×105細胞/mlにて播種し、そして、バッチ培養により最長7日間培養した。細胞培地を、様々な時点で回収し、そして、SEAP測定値の日差変動を避けるために、サンプルを分析まで−20℃に保った。相対SEAP活性を、動力学的酵素アッセイにより測定した。hepes緩衝生理食塩溶液(HBSS)中への培地の段階希釈物10μlを、96ウェル・プレートに加え、次に、ホスファターゼ基質溶液(Pierce)100μlを各ウェルに加え、そして、405nMのODの測定値を、規定の時間間隔で読み取った。OD対時間のプロットの線形ウィンドウだけを、分析に考慮した。
製造業者の取扱説明書に従いGenElute Mammalian Genomic DNA Miniprepキット(Sigma)を使用して、ゲノムDNAを単離し、そして、分光光度測定により定量化した。遺伝子コピー数の測定のために、10ngのゲノムDNAを、多重アッセイにおける標準的なサイクル条件を使用して7500 Real-Time PCR施設(Applied Biosystems)を用いて定量的PCRによって分析した。レポーター構築物を検出するために、ピューロマイシン特異的TaqManプローブを使用し、そして、ハムスター・グリセルアルデヒド・リン酸デヒドロゲナーゼ(GAPDH)遺伝子を検出する第2のTaqManプローブを、内在性コントロールとして使用した。検量線を、ピューロマイシン遺伝子のコピー数をサザンブロットによって測定した細胞株からのゲノムDNAを使用して作成した。
全RNAを〜5×106細胞から‐DNase処理ステップを含めた‐NucleoSpin RNA IIキット(Macherey-Nagel)を使用して単離し、そして、RNA濃度を、260nmにて分光光度測定を用いて測定した。
2.5〜5×105個の細胞を、10mMのフルオレセイン標識メトトレキサート(F-MTX、Molecular Probes/Invitrogen)を含む0.5mlの培地中、29℃にて一晩インキューベートした。標識細胞を、無血清培地で洗浄し、そして、は、FITCフイルター・セットを使用した蛍光顕微鏡(DP50デジタルカメラを備えたOlympus CKX41顕微鏡)を使用することで記録した。
2.1. Puro-DHFRは、トランスフェクションされた細胞にピューロマイシンに対する耐性を付与する(すなわち、Puro-DHFRには、ピューロマイシン・アセチルトランスフェラーゼ活性がある)
DHFR欠乏細胞は、MTXに対する感受性を欠き、且つ、成長のために培地中にHT(ヒポキサンチン及びチミジン)の存在を必要とする。
実施例1.2に記載のとおり、内因的にDHFRを発現するCHO-S-細胞に、Puro-DHFRをコードするp325ベクターでトランスフェクションした。その細胞を、ピューロマイシン(10μg/ml)の存在下で選択した。耐性個体群の限界希釈によって、クローンを得た。興味深いことに、本明細書中に示された実験が、内因性DHFRバックグラウンド発現にもかかわらず、Puro-DHFR選択及び増幅がCHO-S-細胞において実現可能であることを実証した。
図5は、Puro-DHFRをコードするp325ベクターでトランスフェクションされたCHO DXB11-F10細胞の蛍光メトトレキサートでの標識を示している。蛍光発光を、実施例1.6に記載のとおり検出した。非トランスフェクションDXB11-F10細胞は、トランスフェクションしたものより蛍光発光がかなり弱かった。加えて、MTX(50nM)の存在下のより高い選択圧が、puro-DHFR選択マーカーと、より強い細胞標識の発現増強につながる。
よって、Puro-DHFRは、DHFR+(CHO-S)及びDHFR-(CHO-DXB11-F10)細胞の両方で検出できる。
3.1. Puro-DHFRは、注目のタンパク質を高レベルで発現するクローンの単離を可能にする
DXB11-F10細胞に、Puro-DHFRをコードするp325ベクターでトランスフェクションした。このベクターは、レポーター遺伝子(注目のタンパク質)としてSEAPをさらに含んでなる。安定なプールを、MTXの存在下、又は不在在下で選択し、そして、SEAPの発現を、実施例1.3に記載のとおり計測した。図4に示されているように、MTXの存在下で選択したプールは、ピューロマイシンを用いて、又はHTだけの不存在下で選択したプールに比べて、約2倍高いSEAP発現を示した。
Puro-DHFRを、Puro-DHFRと注目のタンパク質の両方を発現するベクターを持つ候補クローンを樹立し、そして、スクリーニングするための選択及び代用マーカーとして使用する。トランスフェクション、選択、そして、増幅の後に、一次スクリーンを、高度な注目の遺伝子の発現も示すクローンを選択する確率が高い、蛍光発光(すなわち、高いPuro-DHFR発現)についてFACSによっておこなう。次に、二次スクリーンを、注目のタンパク質の発現について、できる限り、ELISAによって直接的に実施する。
Claims (35)
- 注目のタンパク質の発現についての細胞のスクリーニング方法であって:
a)(i)毒性化合物に対する耐性を付与するか、又は代謝的優位性を付与する断片に融合させたジヒドロ葉酸レダクターゼ(DHFR)の機能的な断片を含んでなるキメラ・タンパク質;及び(ii)注目のタンパク質、をコードする発現ベクターで細胞をトランスフェクションするステップ;
b)上記毒性化合物に耐性であるか、又は上記代謝的優位性を獲得した細胞を選択するステップ;及び
c)DHFRに結合する蛍光化合物を用いて、ステップ(b)で選択された細胞の蛍光発光をアッセイするステップ、
を含んでなり、ここで、上記の毒性化合物に対する耐性を付与する断片はDHFR又はその断片ではない、方法。 - ステップ(c)を実施する前に、前記注目の組み換えタンパク質を増幅するステップをさらに含んでなる、請求項1に記載の方法。
- 前記増幅ステップを、メトトレキサート(MTX)の存在下で細胞を培養することによって実施する、請求項2に記載の方法。
- 前記のDHFRに結合する蛍光化合物が、蛍光メトトレキサート(f-MTX)又は蛍光トリメトプリム(f-TMP)である、請求項1〜3のいずれか1項に記載の方法。

- マウスDHFRの断片に融合させたピューロマイシンN-アセチルトランスフェラーゼの断片を含んでなるPuro-DHFRポリペプチドであって、以下の:
(i)ピューロマイシンN-アセチルトランスフェラーゼ活性;及び
(ii)ジヒドロ葉酸レダクターゼ活性、
を示す前記Puro-DHFRポリペプチド。 - 前記ポリペプチドが、配列番号2の配列を含んでなる、請求項24に記載のPuro-DHFRポリペプチド。
- 請求項24又は25に記載のPuro-DHFRポリペプチドをコードする核酸。

- 請求項26又は27に記載の核酸を含んでなるベクター。
- 注目のタンパク質をコードする核酸をさらに含んでなる発現ベクターである、請求項28に記載のベクター。
- 少なくとも2つのプロモーターを含んでなり、その一方が前記Puro-DHFRポリペプチドの発現を駆動し、そして、もう一方が前記注目のタンパク質の発現を駆動する、請求項29に記載の発現ベクター。
- 前記Puro-DHFRポリペプチドをコードする核酸が、前記注目のタンパク質をコードする核酸と同じプロモーターによって駆動され、ここで、上記核酸の間に位置する、内部リボソーム侵入部位(IRES)又は2A配列を含んでなる、請求項29に記載の発現ベクター。
- 請求項26又は27に記載の核酸を含んでなる細胞。
- 請求項28〜32のいずれか1項に記載のベクターを含んでなる、請求項32に記載の細胞。
- ヒト細胞、CHO細胞、マウス細胞、及びハイブリドーマから成る群から選択される、請求項33に記載の細胞。
