Description
本発明は、リボザイムを含む分子イメージングのための組成物及び該組成物を用いた分子イメージング方法に関する。より詳しくは、本発明は、イメージングレポーター遺伝子と結合し且つ疾病に関連する特定の遺伝子をターゲットとするトランス−スプライシングリボザイムを含む分子イメージングのための組成物、及び該組成物を用いた分子イメージング方法に関する。
リボザイム(ribozyme)は、酵素活性を有するRNA分子である。リボザイムは、RNAスプライシング(RNA splicing)、RNAプロセッシング(RNA processing)、RNAゲノムの複製及びリボーソームにおけるペプチド結合の形成といった、遺伝子の発現及び調節に、能動的に関わってきた。特に、テトラヒメナ好熱菌(Tetrahymena thermophila)から分離したグループIイントロンリボザイムの場合、イントロンに隣接する2つのエクソンを、一般的なシス−スプライシング反応を介して連結させることができ、またトランス−スプライシング反応を行うことによって、別々のRNA上に含まれる2つのエクソンを連結させることもできる。トランス−スプライシングは、2工程のトランスエステル化(transesterification)機構を通じた切断−ライゲーション反応(cleavage-ligation reaction)を介して進行する。リボザイムは、タ−ゲットRNAとリボザイムの内部ガイド配列(IGS)との間で塩基対を形成することにより、別個に存在する基質RNA(5’エクソン)を認識し結合する。一旦結合すれば、リボザイムはタ−ゲットRNAを切断して下流の切断生成物を放出し、エクソン配列を、その3末端で、基質RNAの5’エクソン切断生成物に、自己切断過程で切断された5’エクソンRNAの代りに、接合(splice)する。このようなスプライシングは、in vitroだけでなく、E.coli及び哺乳動物細胞でも起こる。トランス-スプライシング反応は、特定のターゲットRNAのターゲッティング及び切断、並びに所望のRNA配列との置換に、適用することができる。したがってトランス-スプライシングリボザイムは、新規の治療遺伝子としての可能性を有しており、このような新規治療遺伝子は、遺伝疾患に関連する欠陥のあるRNA転写物を正常RNAにより修繕したり、或いは特定のRNAをターゲッティングして、ターゲットRNAを発現する細胞内のみで所望のRNAが発現されるようにすることができる。
技術的な問題
そこで本願の発明者らは、トランス−スプライシングリボザイムを分子イメージングへ応用することについて、徹底的且つ集中的な研究を行った。研究の結果、特定の疾病関連遺伝子をターゲットとするトランス−スプライシングリボザイムを用いて分子イメージングを行えば、得られたイメージから、疾病の原因となる遺伝子のin vivoでの発現を、非侵襲的且つ正確に検出できることが見出され、本発明の分子イメージング方法が、従来法よりも早期における疾病の診断において卓越していることが示されたことから、本発明が導かれた。
本発明の別の目的は、特定の遺伝子をターゲットとするリボザイムベクターを用いることに基づいた、分子イメージングの方法を提供することにある。
一側面において、本発明は、特定の遺伝子をターゲットとするリボザイムベクターを含む、分子イメージングのための組成物に関する。
以下の実施例を通じて、本発明についてのさらなる理解が得られるが、これらの実施例は、本発明を説明するために記載されたものであって、本発明を限定するものとして解釈されるものではない。
1−1.癌遺伝子特異的なトランス−スプライシングリボザイムを発現する組換えアデノウイルスの構築
癌特異遺伝子mCKAP2をターゲットとするトランス−スプライシングリボザイムは、公知の技術を使用して構築した(Kim, A., et al., Oligonucleotides 17: 95-103, 2007)。β−ガラクトシダーゼ(lacZ)遺伝子又はルシフェラーゼ(luc)遺伝子を、リボザイムの3’に連結させ、得られたリボザイムをクローニングした。該レポーター遺伝子と融合されたリボザイムを、CMVプロモーターを含むシャトルベクターpAvCMV3.0のBglIIとNotIサイトの間にサブクローニングした。該シャトルベクターを、HEK293細胞内に、アデノウイルスをバックボーンとするベクターpJM17と共に、カルシウム−リン酸塩沈降法を用いてコトランスフェクトした。続いて、トランスフェクトされた細胞は、軟寒天下で約2週間増殖させ、ウイルスプラークを形成させた。多数のクローンを分離した。組換えウイルスは、HEK293細胞内にて増幅させた後、公知の方法を用いて、すなわちCsCl勾配超遠心法を2回行うことにより、精製した(Kobayashi, K., et al . , J. Biol. Chem. 271:6852-6860, 1996)。mCKAP2を特異的にターゲットとするトランス−スプライシングリボザイム及びlacZ遺伝子を含む、精製組換えアデノウイルスを、「Ad−mCR−lacZ」と命名した。mCKAP2特異的なトランス−スプライシングリボザイム及びluc遺伝子を含む、別の精製組換えアデノウイルスを、「Ad−mCR−luc」と命名した。Ad−mCR−lucの模式図を図2に示した。ネガティブコントロールとしては、アデノウイルスをバックボーンとするベクター(Ad−Mock)並びにlacZ遺伝子を保有するアデノウイルスベクター(Ad−lacZ)及びluc遺伝子を保有するアデノウイルスベクター(Ad−luc)を使用した。分離された組換えアデノウイルスベクターの力価は、プラーク形成単位(pfu)として、TCID50法を用いて決定した。
マウス肝癌細胞株Hepa1−6、マウス大腸癌細胞株CT−26、マウス線維芽細胞株NIH3T3、ヒト肝癌細胞株HepG2細胞、マウス肝癌細胞株BNL1ME A.7R.1、及びマウス黒色腫細胞株B16F10を使用した。これらの細胞株は、10%ウシ胎仔血清(Invitrogen社)及び100U/mLのペニシリン/ストレプトマイシンを添加したダルベッコ変法イーグル培地(DMEM;Gibco社)にて、37℃、5%のCO2下で、インキュベータ中で培養した。
6週〜8週齢の雄C57BL6マウスをOrient社(ソウル、韓国)から購入した。該動物は、韓国食品医薬品安全庁の動物実験室で無菌条件下に維持され、使用する前に少なくとも1週間、実験条件に順適応させた。該動物は、午前6時に灯をオンにし、午後6時に灯をオフにする規則的な12時間の光/暗周期の下で飼育した。
肝癌の動物モデルはHepa1−6細胞をマウスの脾臓に注入し、確立したが、その方法は公知の方法を若干改変して利用した(Giavazzi, R., et al., J Natl Cancer Inst 77:1303-8, 1986)。詳細には、Hepa1−6細胞を培養皿でコンフルエントに培養し、マウスに注射する1日前に継代培養した。Hepa1−6細胞を0.025%トリプシン−EDTAを用いて回収し、PBSで3回洗浄した。細胞はトリパンブルーで染色し、生きている細胞の数を数えた。生きている細胞が95%より多い場合に、該培養細胞をマウスに注射した。Hepa1−6細胞は、50μl当たり2×106個細胞の密度で懸濁した。マウスの左脇を縦に小さく切開して脾臓を露出させ、50μl量中の2×106個の細胞を、29グレードの注射針を用いて脾臓に徐々に注入した。注射針を除去後、脾臓を腹腔に戻し、腹部の切開を絹縫合して閉じた。Hepa1−6細胞を注射の後、8〜10日内に多数の肝癌小結節が生成されることを全体的な観察によって容易に探し出すことができた。
2−1.マウスの組織又は細胞株におけるmCKAP2の発現の評価
マウスの組織又は細胞株においてmCKAP2タンパク質が発現されているか否かを判定するため、ウエスタンブロット解析を行った。マウスの組織又は各臓器から、Trizol試薬(Invitrogen社)を用いてトータルRNAを単離後、RNaseフリー水及びバッファ中にて、RQ1 RNAaseフリーDNaseI(1U/μgRNA;Promega社)で、37℃で30分間処理した。cDNAの合成に関しては、DNaseI処理したトータルRNA、4Uの逆転写酵素(Omniscript−RTase、Qiagen社)、dNTPミックス、10×RTバッファ、250ngのランダムプライマー(Invitrogen社)、及び40UのRNase阻害剤により反応混合液を作製した。逆転写反応を、25℃で10分、37℃で1時間の条件で行った後、95℃に加熱して酵素を非活性化させた。合成されたcDNAを鋳型として用い、下の表1にまとめられたmCKAP2特異的プライマー、及び下記の反応条件下の反応混合液を用いて、mCKAP2を増幅した。mCKAP2遺伝子の発現は、アガロースゲル電気永動を通じて検出した。

その結果、マウス癌細胞株CT−26及びHepa1−6でmCKAPの発現を確認することができた(図4及び5)。
トランス−スプライスされた分子(TSM)を検出するため、RT−PCRを行った。CKAP2保有のHepa1−6細胞、及びCKAP2を消失したHepG2細胞は、トランス−スプライシングリボザイムを保有するアデノウイルスAd−mCR−lacZ、Ad−lacZ及びAd−Mockで、30moiにて感染させた。トータルRNAを感染細胞から単離し、上記記載と同様の方法を用いてcDNAを合成した。TMSの生成を評価するため、合成されたcDNAを鋳型として用いて、表1にまとめられたTSM特異的プライマー及び上記の反応条件下(アニーリングは60℃で行い、変性及び伸長は上記と同じ温度にて行った)の反応混合液を用いて、TSMを増幅した。フォワード及びリバースプライマーは、mCKAP2又はトランス−スプライシング分子を増幅するように設計した。より具体的には、TSM特異的プライマーは、トランス−スプライシング接合のmCKAP2上流領域及び該トランス−スプライシング接合の下流領域とアニールするように設計した。
リボザイムがmCKAP2をターゲットとしているか、及びリボザイムが発現しているかを判定するために、リボザイム遺伝子の3’末端にlacZ遺伝子が融合されている組換えアデノウイルスAd−mCR−lacZを、細胞に感染させた。β−ガラクトシダーゼの発現を、X−gal染色及びβ−ガラクトシダーゼアッセイを通じて、定量的に分析した。X−gal染色に関しては、Hepa1−6細胞及びHepG2細胞を2×105細胞/穴の密度で6穴培養プレートに播種し、37℃で培養した。翌日、Ad−mCR−lacZアデノウイルスを、0、10、30又は50moiで、細胞に感染させた。CMVプロモーターの調節下にlacZ遺伝子を保有するAd−lacZアデノウイルスを、ポジティブコントロールとして使用した。30moiのAd−lacZで細胞を感染させた。48時間後、β−ガラクトシダーゼ染色キット(Invitrogen Corporation、CA、アメリカ合衆国)を用いて、細胞を染色した。
3−1.ルシフェラーゼアッセイ
ルシフェラーゼの発現を評価するため、1×104個数のHepa1−6細胞を、黒色の96−ウェル培養プレートに播種し、培養した。翌日、mCKAP2をターゲットとし且つルシフェラーゼを発現する組換えアデノウイルスAd−mCR−luc、及びCMVプロモーターの調節下にルシフェラーゼ遺伝子を保有するAd−lucアデノウイルスを、0、1、5、10、15、30及び50moiで、細胞に感染させた。5時間後、溶解バッファ(Promega社)を用いて細胞を溶解した。ルシフェラーゼ活性は、Blight−Glo(商標)ルシフェラーゼアッセイシステム(Promega社)を用いて決定した。発光強度は、ルミノメーターを用いて、10秒ごとの相対発光量(RLU)で測定し、測定値をRLU/s/μgタンパク質で表した。
細胞株またはマウス組織は、20mM Tris−HCl(pH7.5)、5mM EDTA、10mM Na4P2O7、10mM NaF、2mM Na3VO4、1%NP−40、PMSF及びプロテアーゼ阻害剤を含むタンパク質抽出溶液(Sigma社)を用いて溶解した。タンパク質は、公知の方法を用いてニトロセルロース膜に移した。より詳細には、精製されたタンパク質(25μg又は50μg)をSDS−PAGEによって分子の大きさの順に分離した後、ブロット機器を用いてNC膜(Millipore社)に移した。ブロットは、ブロッキングバッファ(0.05% Tween20を含むTBS中の5%スキムミルク)にて30分間ブロックした。続いてブロットを1次抗体のウサギポリクローナル抗mCKAP2抗体又は抗β−アクチン抗体(Sigma社)中で室温で1時間、あるいは摂氏4℃で一晩インキュベートした。ブロットは、Tweenトリス−バッファ生理食塩水(T−TBS)で2回洗い、結合していない1次抗体を取り除いた。結合した1次抗体は、西洋わさびペルオキシダーゼが接合した抗−ウサギ抗体(Amersham社)で検出した。
Xenogen IVIS2000冷CCDカメラ(Xenogen社、Hopkinton、MA)を用いて、in vivo分子イメージングを行った。Ad−mCR−luc組換えアデノウイルス、ポジティブコントロールとしてのAd−lucアデノウイルス、及びネガティブコントロールとしてのAd−mockウイルスを、50μLのダルベッコPBS(Life Technologies社)中、1011個のウイルス粒子(vp)のウイルス量になるように調製した後、尾静脈注射によって、マウスに全身投与した。翌日、ルシフェラーゼの基質のD−ルシフェリンを、200μLのPBS中、マウスの体重1kg当たり150mgの用量で、マウスの腹腔内に投与した。in vivo分子イメージングのため、イソフルラン混合酸素(isofluran-mixed oxygen)によりマウスを麻酔した。10分後にリファレンスイメージを撮り、そしてin vivoイメージを1分〜5分間撮影した。Living Image software(Xenogene社)を用いて全身のイメージングを行い、その結果を秒当たり、cm2当たり、ステラジアン(steridian)当たりのフォトン(p/s/cm2/sr)にて表した。
Claims (19)
- トランス−スプライシングリボザイムベクターを含む、分子イメージング用組成物。
- 前記リボザイムが、イメージングレポーター遺伝子と結合されている、請求項1に記載の組成物。
- 前記イメージングレポーター遺伝子が、蛍光タンパク質又は比色酵素をコードする遺伝子である、請求項2に記載の組成物。
- 前記リボザイムが、ルシフェラーゼ(luc)遺伝子と結合されている、請求項2に記載の組成物。
- 細胞骨格関連タンパク質2(CKAP2)をターゲットとするリボザイムを含む、請求項1に記載の組成物。
- 前記リボザイムが、トランス−スプライシンググループIリボザイムの改変型であり、且つ、
1)ターゲットのマウス細胞骨格関連タンパク質2(mCKAP2)のRNAのウリジン残基(45U)の下流領域に相補的なアンチセンス配列であり、83ヌクレオチド(mCKAP2のRNAの+59部位から+141部位のヌクレオチドに相補的)又は300ヌクレオチド(mCKAP2のRNAの+59部位から+358部位のヌクレオチドに相補的)からなるアンチセンス配列;
2)該アンチセンス配列の3’末端に連結され且つ内部ガイド配列(IGS)、5’−GAGCGT−3’を含むP1ヘリックス領域;
3)該P1ヘリックス領域の3’末端に連結されたP10ヘリックス領域;及び
4)該P10ヘリックス領域の3’末端に連結され且つβ−ガラクトシダーゼ(lacZ)又はルシフェラーゼ(luc)をコードする配列:
を含む、請求項1に記載の組成物。 - 前記リボザイムベクターが、アデノウイルスベクターである、請求項1に記載の組成物。
- 前記リボザイムベクターが、図2に示す遺伝地図を有する、請求項1に記載の組成物。
- 1)イメージングレポーター遺伝子を、特定の遺伝子をターゲットとするリボザイムに連結させ、得られるリボザイムをクローニングする工程;
2)クローニングされたリボザイムをベクターへ挿入し、該リボザイムを発現させる工程;及び
3)該リボザイムの発現が同定されたベクターを分離し、該リボザイムを確認する工程:
を含む、分子イメージングのためのベクターの製造方法。 - トランス−スプライシングリボザイムベクターを用いた、分子イメージング方法。
- 前記リボザイムが、イメージングレポーター遺伝子と連結されている、請求項10に記載の分子イメージング方法。
- 前記イメージングレポーター遺伝子が、蛍光タンパク質又は比色酵素をコードする遺伝子である、請求項11に記載の分子イメージング方法。
- 前記蛍光レポーター遺伝子が、蛍光タンパク質又は比色酵素をコードする遺伝子である、請求項12に記載の分子イメージング方法。
- 前記リボザイムが、ルシフェラーゼ(luc)遺伝子と連結されている、請求項11に記載の分子イメージング方法。
- 細胞骨格関連タンパク質2(CKAP2)をターゲッティングするリボザイムを含む、請求項10に記載の分子イメージング方法。
- 前記リボザイムが、トランス−スプライシンググループIリボザイムの改変型であり、且つ、
1)ターゲットのmCKAP2のRNAのウリジン残基(45U)の下流領域に相補的なアンチセンス配列であり、83ヌクレオチド(mCKAP2のRNAの+59部位から+141部位のヌクレオチドに相補的)又は300ヌクレオチド(mCKAP2のRNAの+59部位から+358部位のヌクレオチドに相補的)からなるアンチセンス配列;

を含む、請求項10に記載の分子イメージング方法。 - 前記リボザイムベクターが、アデノウイルスベクターである、請求項10に記載の分子イメージング方法。
- 前記リボザイムベクターが、図2に示す遺伝地図を有する、請求項10に記載の分子イメージング方法。
- 1)イメージングレポーター遺伝子を、トランス−スプライシングリボザイムに連結させ、得られるリボザイムをクローニングすること;
2)クローニングされたリボザイムをベクターへ挿入し、該リボザイムを発現させること;及び
3)該リボザイムの発現が同定されたベクターを分離し、該リボザイムを検出すること:
を含む、分子イメージング方法。
