Abstract
【解決手段】 魚鱗由来のコラーゲンで被覆された、表面が線維化状を有する細胞培養基材である。このようなコラーゲンは、従来の哺乳類由来のコラーゲンで被覆した基材よりも、長期にわたって優れた細胞接着性、伸展性を示す。とりわけ哺乳類由来の細胞培養に優れた基材を提供するものである。

Description
更に、本発明は、魚鱗由来のコラーゲンが、変性温度が30℃以上のテラピア鱗由来のコラーゲンである上記細胞培養基材に関する。
更に、本発明は、細胞が哺乳類の細胞である上記細胞培養基材に関する。
コラーゲンの変成温度は、International Journal of Biological Macromolecules, 32, 199 (2003)に記載の方法に従って、CD分光計(Jasco model 725 spectrometer) を使用してコラーゲン水溶液の温度を段階的に上昇させることによって求めた。即ち、コラーゲン水溶液をpH 3の希塩酸で希釈し、0.03wt%となるように調整し、光路長1mmの石英セルに入れた。セルの温度を1℃/minで上昇させ、221nmにおける旋光度を0.2℃ごとに測定した。各温度における旋光度を温度に対してプロットすると、旋光度の値がコラーゲン螺旋の値からランダムコイルの値へと急激に変化する変成曲線が得られる。それらの旋光度値の中間値を与える温度、すなわち、螺旋率(Helicity;%)が50%になるときの温度を変成温度とした。この測定は3 回行い、平均値を使用した。
テラピアの鱗を水で十分洗浄し、さらに10%塩化ナトリウム溶液で十分洗浄し、鰭などの夾雑物を除去した後、室温にて乾燥した。含水率は18.5%であった。
このテラピア鱗1kgをpH2の塩酸溶液9kgに分散し、1Mの塩酸溶液を添加しながらpHを2に保った状態で、25℃、2時間穏やかに攪拌し、鱗に含まれる無機成分を溶かしだした。これをザルにあげて、十分水洗した後、総重量が4kgとなるようにpH2の塩酸溶液を添加した。
上記テラピア鱗由来のコラーゲン溶液を、pH3の塩酸溶液を用いて0.03wt%に希釈した。これをアイスプレート上で24ウェル(BD社製)に薄く広げて、余分なコラーゲン溶液を除去した。次いでクリーンベンチ内で25℃、5分間乾燥した。対照として、同様の操作にて豚由来のコラーゲン(新田ゼラチン製、Cellmatrix Type I-C)を使用しコーティングしたウェルを作成した。
Growth Medium(5%FBS(SIGMA製)/alphaMEM(GIBCO製)/1%Antibiotec)にL929細胞を懸濁して、前記テラピア鱗由来のコラーゲンを用いて処理した24ウェルに播種密度3.4x103cells/cm2になるように播種した。37℃, 5%CO2において、 2、4dayインキュベートした。適宜、Growth Mediumを交換した。対照として、未処理のウェル、上記豚由来のコラーゲンで処理したウェルについても同様の試験を行った。
Growth MediumにL929細胞を懸濁して、上記テラピア鱗由来のコラーゲン溶液を用いて処理したチャンバースライド上に播種密度5.0x103cells/cm2になるように播種した。これをセミコンフルエントまで増殖した後、リン酸バッファーを用いて培地を洗い流し、4%ホルムアルデヒドで固定した。さらにリン酸バッファーにより洗浄した後、0.5%TritinXで5分間透過処理を行い、再度リン酸バッファーで洗浄した。これを湿箱に移して、ローダミン-ハロイジン溶液を添加し、暗所に30分間置いた。これをPBSで洗浄し、DAPI in vector shield を添加し、カバーガラスをかけ、蛍光顕微鏡で観察した。対照として、未処理のチャンバースライド、豚由来コラーゲン、タイ鱗由来コラーゲンで処理したものについても同様の試験を行い蛍光顕微鏡で観察を行った。

- 魚鱗由来のコラーゲンで被覆された、表面が線維化状を有する細胞培養基材。
- 魚鱗由来のコラーゲンが、変性温度が30℃以上のテラピア鱗由来のコラーゲンである請求項1記載の細胞培養基材。
- 細胞が哺乳類の細胞である請求項1または2記載の細胞培養基材。
