Abstract
【解決手段】配列番号1記載の塩基配列からなるオリゴヌクレオチドを含むプライマーと、配列番号2記載の塩基配列からなるオリゴヌクレオチドを含むプライマーとからなるカワヒバリガイ検出用プライマーセットを用いてPCRを行い、被験試料の遺伝子増幅が生じたか否かにより被験試料にカワヒバリガイ幼生が存在しているか否かを判定するようにした。また、カワヒバリガイ幼生個体数が既知の複数の試料それぞれに対し、上記プライマーセットを用いてPCRを行うことによりカワヒバリガイ幼生個体数と遺伝子増幅量との関係を調べて予め作成された検量線を用い、被験試料に対しプライマーセットを用いてPCRを行うことにより得られる被験試料の遺伝子増幅量から被験試料中のカワヒバリガイ幼生個体数の定量を行うようにした。

Description
カワヒバリガイ、及びカワヒバリガイと比較的近縁であると考えられる二枚貝であるコウロエンカワヒバリガイ(Xenostrobus securis)、ムラサキイガイ(Mytilus galloprovincialis)、ホトトギスガイ(Musculista senhousia)、ミドリイガイ(Perna viridis)のCOI遺伝子の塩基配列のデータベース化を行い、このデータベースに基づいてカワヒバリガイに特異的な塩基配列を特定し、プライマーを設計した。データベース化に供した二枚貝は採集直後に99.5%エタノールに浸漬し4℃で保存した。
本発明のプライマーを用いた定量分析について検討した。
カワヒバリガイ幼生から総DNAを抽出するための条件と定量分析について検討を行った。
複数のプランクトンが混在しているプランクトンサンプルにカワヒバリガイ幼生が含まれている場合であっても、カワヒバリガイ幼生を特異的に検出できるか検討した。
4 DNAプローブ
Claims (6)
- 配列番号1記載の塩基配列からなるオリゴヌクレオチドを含むプライマーと、配列番号2記載の塩基配列からなるオリゴヌクレオチドを含むプライマーとからなるカワヒバリガイ検出用プライマーセット。
- 被験試料に対し、請求項1記載のプライマーセットを用いてPCRを行い、前記被験試料の遺伝子増幅が生じたか否かにより前記被験試料にカワヒバリガイ幼生が存在しているか否かを判定することを特徴とするカワヒバリガイ幼生の検出方法。
- カワヒバリガイ幼生個体数が既知の複数の試料それぞれに対し請求項1記載のプライマーセットを用いてPCRを行うことによりカワヒバリガイ幼生個体数と遺伝子増幅量との関係を調べて予め作成された検量線を用い、被験試料に対し前記プライマーセットを用いてPCRを行うことにより得られる前記被験試料の遺伝子増幅量から前記被験試料中のカワヒバリガイ幼生個体数の定量を行うことを特徴とするカワヒバリガイ幼生の定量方法。
- 前記PCRをリアルタイムPCRとし、前記遺伝子増幅量として前記リアルタイムPCRを行うことにより得られるCt値を利用することを特徴とする請求項3記載のカワヒバリガイ幼生の定量方法。
- 前記被験試料を分画処理してから前記PCRに供する請求項3または4に記載のカワヒバリガイ幼生個体数の定量方法。
- 配列番号1記載の塩基配列からなるオリゴヌクレオチドを含むプライマー及び配列番号2記載の塩基配列からなるオリゴヌクレオチドを含むプライマーのうちの少なくとも一方のプライマーをプローブとして含むカワヒバリガイ検出用キット。
