Description
従って、本発明は互いに相互作用をするベイトとプレイを検出し得る新規な方法を提供する。
(a)(i)ベイト(bait)、第1の標識物質及び移動モジュールを含む第1の構成物;及び(ii)プレイ(prey)及び第2の標識物質を含む第2の構成物;を含む細胞を調製する工程;及び
(b)第1の構成物及び第2の構成物の細胞内分布を検出する工程
を含むベイト(bait)とプレイ(prey)の相互作用を検出する方法を提供する。

本発明は、細胞内で起こる生体物質間の相互作用を直接的に、さらに、実時間で分析できるようにするために、外部刺激または内在的な信号伝達機構によって起こる蛋白質の細胞内位置移動を利用したものである。つまり、外部刺激または内在的な信号伝達機構により移動する移動モジュールに相互作用の1つの対象となるベイトを融合し、これを追跡するための標識物質を含む第1の構成物を設計した。また、ベイトと相互作用するプレイ及びこれを追跡できるように、さらに他の標識物質を含む第2の構成物を設計し、前記第1の構成物及び第2の構成物が同時に存在するようにして、これらの相互作用を細胞内で直接的、さらに、実時間で分析できるようにした。

しかしながら、信号物質での処理は試験物質処理前に実施される必要はなく、当業者が適当に調節して行える。
図1は、本発明の基本構成物及び分析指標を示す。
1)第1の構成物は、移動モジュール、第1の標識物質及びベイトを含み、第1の構成物には1つ、または複数のベイトを含み得る。
2)第2の構成物は、第2の標識物質及びプレイを含み、第2の構成物には1つ、または複数のプレイを含み得る。
3)信号(Signal S)は、移動モジュールの移動を誘導する外部刺激(移動モジュールの位置変化を誘導する成長因子(growth factor)、血清因子(serum factor)、PMA(TPA)等)、内在的な刺激物質(DAG,diacyl-glycerol)、外部または内在的なATP及びカルシウムの濃度変化を含む。
4)相互作用分析指標は、ベイト及びプレイの内因的または外因的特性による第1の構成物と第2の構成物の細胞内の分布位置、分布位置の変化、各構成物等の細胞内移動特性、及び1つまたは複数の第1、第2の構成物による結合体同定を含む。
ただし、下記実施例は本発明を例示するのみであって、本発明の内容が下記実施例に限定されるものではない。
<1−1>動物細胞株及びその培養
CHO−k1(ATCC CCL-61, Cricetulus griseus, チャイニーズハムスター(hamster, Chinese))、HEK293(ATCC CRL-1573, Homo sapiens, ヒト(human))、HeLa(ATCC CCL-2, Homo sapiens, ヒト(human))、及びSH−SY5Y(ATCC CCL-2266, Homo sapiens, ヒト(human))細胞株を使用し、これらの動物細胞株の培養条件は、ATCC(American Type Culture Collection)社の細胞株培養方法(indtruction)に従った。ただし、CHO−k1細胞はFー12培養液を使用し、HEK293、HeLa、及びSH−SY5Y細胞はDMEM培養液を使用し、その他の培養条件は同様である。各細胞の培養方法は下記の通りである(当業者の目的により細部的な培養条件は異なり得る。)。25mM HEPES、10%ウシ胎仔血清(fetal bovine serum,FBS,v/v)、100units/mlペニシリン(penicillin)、及び100μg/mlストレプトマイシン(streptomycin)を含むpH7.4の培養液(F−12及びDMEM)内で各細胞株を、37℃、5%CO2が維持される培養器で培養した。
本発明の実施例で使用された細胞内遺伝子導入方法は、一般的に使用されるリポソームを用いた方法(liposome-based techniques)の1つであるExGene 500(Fermentas Life Science)を使用し、遺伝子の濃度等遺伝子導入のための全ての条件は製造社の指針により行った。より具体的に、継代培養されている細胞をカバースリップが入っている12−ウェルプレートに移して分株を行い、1日培養した後、0.9mlの新しい培養液に交換した。約1μg程度の形質転換試料を0.1mlの150mM NaCl溶液に添加し、十分に撹拌した後、3.3μlのExGene試薬を添加して15秒間ボルテックス(vortex)して混合した。この溶液を常温で10分間放置した後、細胞が成長しているカバースリップが入っている12−ウェルプレートの各ウェル(well)に添加して18時間培養することによって、細胞は形質転換された。
<2−1>第1の構成物の設計及び作製
本発明で第1の構成物とは、細胞質において均一に発現されている蛋白質を細胞膜に移動させる移動モジュール、顕微鏡を利用した分析を可能にする蛍光蛋白質、及びベイトを結合させてできる融合配列で構成される。
第1の構成物に含まれた移動モジュールは、プロテインキナーゼより誘導されることから、本質的にリン酸化機能を有しており、このようなリン酸化機能は移動モジュールの役割ばかりでなく、内在的なリン酸化機能を介して構成物が過剰発現された細胞の信号伝達機構と、蛋白質相互作用を阻害または干渉させる可能性を有している。従って、本発明では移動モジュールが有するリン酸化機能を除去するために突然変異を誘導した。本発明に使用されたキナーゼは、リン酸化機能において極めて重要な部位として知られる311番目のアミノ酸(チロシン,tyrosine)をフェニルアラニン(phenylalanine)に(Y313F)、378番目のアミノ酸(リジン,lysine)をアルギニン(arginine)にそれぞれ置換するために、PCRによる突然変異誘導方法を使用して、最終的にTMD移動モジュールを作製した。
生体内に存在する蛋白質は固有の分布特性(標的(targeting))を有していて、ベイトとプレイは細胞内小器官に標的位置を有する。この場合、第1の構成物と第2の構成物の位置移動を感知し難いこともある。従って、ベイトとプレイの結合を実験で確認するためには、実験が可能な細胞質または核質で分布位置を制御する必要があり、このため、蛋白質を細胞質に分布位置を誘導するための核外搬出シグナル(nuclear exclusion signal,NES)と核質に分布位置を誘導するための核局在化シグナル(nuclear localization signal,NLS)を使用して、ベイトとプレイの細胞内分布位置を制御できるベクターを作製した。
第2の構成物は、第1の構成物のベイトと結合する内在的な特性を有するプレイの移動を分析するための標識物質を含む。第2の構成物は第1の構成物に使用された標識物質とは異なる蛍光、すなわち緑色蛍光蛋白質(EGFP,AzG)、赤色蛍光蛋白質(mRFP)、及び赤外線蛍光蛋白質(HcR)を使用して第1の構成物と同一の方法で作製した。第2の標識物質として、EGFPを使用するときは、pEGFP−C3ベクター(Clontech)を使用し、mRFPを使用するときは、pmRFP−C3ベクターを使用し、AzGを使用するときは、pAzG−C3ベクターを使用し、HcRを使用するときは、pHcR−C3ベクターを使用し、各原本C3ベクターはpEGFP−C3ベクターのEGFP遺伝子配列部位を、各AzG、HcR遺伝子で下記の通り置換することにより作製された。
<3−1>構成物の発現確認及び移動特性分析
第1の構成物及び第2の構成物ベクターが導入された細胞が成長しているカバースリップ(cover slip)を流体器(perfusion chamber)に固定し、共焦点レーザー蛍光顕微鏡(カルザイスLSM510)の載物台に装着し、外部刺激前と外部刺激(1μM PMA(Phorbolester)処理)後の構成物ベクターに対する映像を撮影した。
第1の構成物のうち、pTMD−mRFP−C3ベクターを利用した、外部刺激物質であるPMA(Phorbol ester)の濃度に伴う移動効率を測定するために、CHO−k1細胞に第1の構成物の発現ベクターを形質転換させ、過剰発現させた後、18時間培養して、1、5、10、20、40、50、80、100nM、1、5μMのPMAで処理した。5分間PMAを処理した後、3.8%ホルムアルデヒド(formaldehyde)で細胞を固定して、共焦点顕微鏡(Confocal microscope, Carl Zeiss LSM 510)を介して、赤色蛍光が発現された200個の細胞を無作為で選定し、濃度別赤色蛍光の細胞膜分布細胞の数を測定した。
<4−1>p53蛋白質とSV40T抗原の細胞内結合分析
潜在的発癌性遺伝子として知られているp53と、これに結合するものとして知られているSV40T抗原間の結合について、現在IPを利用した結合同定システムとして商用化されたものがある。ここに本発明の基本的な結合検出機能を検証するために、同一の蛋白質等を対象に分析を実施した。p53とSV40T抗原に対する細胞内結合を検証するために、p53蛋白質が融合された第1の構成物(TMD−mRFP−p53)及びSV40T抗原が融合された第2の構成物を次のように製造した。
多機能性疾病誘発遺伝子として知られている、リシル−tRNA シンセターゼ(Lysyl-tRNA synthetase,KRS)は、p38/AIMP2蛋白質及び少なくとも3個の異なるアミノアシル−tRNA シンセターゼ(Aminoacyl-tRNA synthetase,ARSs)と複合体をなしているものとして知られている(J. Cell Science. 2004, 117, 3725-3734参照)。従って、KRS蛋白質とp38蛋白質の細胞内結合の実時間分析の可能性、多様な動物細胞内での実験可能性、蛍光標識の多様性、及びベイトとプレイとの交換可能性を確認するために、KRSと結合するものとして知られているp38と、潜在的な結合蛋白質と予測されるGag及びLRを利用して細胞結合を分析した。
TNF−alphaにより、調節されるNFkB及びIkB蛋白質の相互作用は、細胞内で多様な細胞信号伝達経路に関与するものと知られている。多様な経路において、特に注目されているのは、生体内で発生する各種の危害性信号に対応する炎症誘発と密接な関連性を有していることである。従って、NFkB複合体のうち、多機能性炎症関連遺伝子であるRelA蛋白質と阻害蛋白質であるIkBの結合可否を検証することと同時に外部刺激後、細胞固定技法でなく生細胞を利用した直接分析可能性を確認するために、実時間分析技法(time-laps)を利用して実験を実施した。
NFkB複合体は、基本的にNFkB(RelA/p50)と陰性調節因子であるIkB蛋白質が複合体を成すものとして知られている。本実験ではこの複合体を構成するRelA、p50、及びIkBの3種の蛋白質の結合を同定できるかを検証した。
酵母ツーハイブリッド法(Y2H,yeast two-hybrid)は、現在、生細胞内の蛋白質結合をスクリーニングする最も一般的な方法として使用されているばかりでなく、今まで知られた新薬標的スクリーニング方法のうち、多くの部分を占めている代表的な方法である。しかしながら、酵母ツーハイブリッド法は、偽陽性(false-positive)が高く、酵母を使用するという短所が有り、新薬標的及び新薬候補検証に弱点を有していることが周知の事実である。従って、本実験例では酵母ツーハイブリッド法により、スクリーニングされた候補物質を検証し得る方法としての本発明の適用性を分析した。
潜在的発癌性遺伝子として知られるp53蛋白質は、少なくともmdm2蛋白質の結合により、発癌が促進されるものとして知られており、これを阻害し得る物質であるヌトリン3が強力な抗癌剤として使用できることが多くの研究を介して確認された。ビアコア社の表面プラズモン共鳴(surface plasmon resonance technology,SPR)技術を使用してイン・ビトロ(in vitro)で実施した結合阻害実験によれば、p53とmdm2の結合は、最小限1μM以上のヌトリン3を使用した時、90%以上の結合阻害効果を表し、50%の阻害が起こる濃度(IC50;median inhibitory concentration)は、90nMであると測定された(Vassilev, L. T. et al., Science 303, 844-848)。

- 下記の工程(a)及び(b)を含むベイト(bait)とプレイ(prey)の相互作用を検出する方法:
(a)(i)ベイト、第1の標識物質及び移動モジュール(translocation module)を含む第1の構成物;及び(ii)プレイ及び第2の標識物質を含む第2の構成物を含む細胞を調製する工程;及び
(b)第1の構成物及び第2の構成物の細胞内分布を検出する工程。 - 下記の工程(a)、(b)及び(c)を含むベイト(bait)とプレイ(prey)の相互作用を検出する請求項1記載の方法:
(a)(i)ベイト、第1の標識物質及び移動モジュールを含む第1の構成物;及び(ii)プレイ及び第2の標識物質を含む第2の構成物を含む細胞を調製する工程;

- 前記細胞が、(i)プロモータ及びこれと作動可能に連結されたベイト、EGFP(Enhanced Green Fluorescent Protein)またはmRFP(Monomeric Red Fluorescent Protein)である第1の標識物質及び配列番号1、配列番号3、配列番号5及び配列番号7からなる群より選ばれるアミノ酸配列を有する移動モジュールをコードするヌクレオチドを含むベクター及び(ii)プロモータ及びこれと作動可能に連結されたプレイ及びEGFP(Enhanced Green Fluorescent Protein)、mRFP(Monomeric Red Fluorescent Protein)、AzG(Azami Green)及びHcR(HcRed, Heteractis crispa red Fluorescent Protein)からなる群より選ばれる第2の標識物質をコードするヌクレオチドを含むベクターで同時形質転換されたものである請求項16記載の細胞。
- 前記細胞が、CHO−k1細胞、HEK293細胞、HeLa細胞、SH−SY5Y細胞、Swiss 3T3細胞、3T3−L1細胞、NIH/3T3細胞、L−929細胞、Rat2細胞、RBL−2H3細胞、及びMDCK細胞からなる群より選ばれる請求項15項乃至18のいずれか1項記載の細胞。
- 請求項15乃至18項のいずれか1項記載の細胞を含むベイトとプレイの相互作用検出用キット。
