Abstract
入れられたサンプルを、横方向指向性フロー装置の分析膜に向かわせる適用されたサンプルを配向する診断アッセイ装置を提供する。
【解決手段】
本発明のアッセイ装置は検査ストリップの一端にある材料の層により画成されるサンプル受け入れポートが画成されている。ポートはサンプルを含み特にこれを制御された方法で膜へ配向する。更なる特徴として略C字状の筐体の形状を含み、全差ストリップはC字状の開口部を横断し読み取り装置によるアクセスができるようにしてある。好適な方法では超常磁性粒子を用いて標的分析物を標識し電磁気的読み取り装置による検出及び測定を行なう。

Claims (14)
- サンプル中の標的分析物の定量的検出のための指向性フロー・アッセイ装置であって、前記装置は
第1の端部と第2の端部を有するアッセイ支持部材と、
前記支持部材に隣接して取り付けられ略平行な多孔質分析膜であって、前記分析膜は第1の端部と第2の端部を有しその前記第1の端部と第2の端部の中間に少なくとも一つの捕獲領域を有し、前記少なくとも一つの捕獲領域は前記分析膜の前記第1の端部から前記分析膜の前記第2の端部に向かって移動する標識された分析物を捕獲するように構成された多孔質分析膜と、
前記支持部材の一端に具備され分析しようとする前記サンプルを前記装置に導入するためのサンプル受け入れポートであって、
前記封止部材の上に配置されたチャネル層であって、開口部とチャネルとを含み、前記開口部は前記チャネルと液体連通を提供し、前記チャネルは前記多孔質分析膜と液性連通を提供することを特徴とするチャネル層と、
前記チャネル層の上に配置された親水性材料であって、前記チャネル層において前記開口部に対応する開口部を内部に有することを特徴とする親水性材料と、
前記親水性材料の上に配置されて前記ポートへの液体の導入ができるように内部に開口部を有するガスケット要素であって、前記アッセイ装置に液体封止を提供することを特徴とするガスケット要素とを含むサンプル受け入れポートと
を含むことを特徴とする装置。
- 前記支持部材に対向する側面において前記分析膜を被覆する保護膜をさらに含み、前記保護膜は光学的に透明ではないことを特徴とする請求項1に記載の装置。
- 前記保護膜は前記多孔質膜と一体的に形成されていることを特徴とする請求項2に記載の装置。
- 前記捕獲領域を通過した共役体の回収のために前記多孔質膜に設けた制御領域をさらに含んで前記検査ストリップが使用済みであることを示すことを特徴とする請求項1に記載の装置。
- 前記保護膜上に印刷された少なくとも1個の磁気構成領域をさらに含むことを特徴とする請求項4に記載の装置。
- 前記アッセイ支持部材と前記チャネル層の間に液体封止材料をさらに含むことを特徴とする請求項1に記載の装置。
- サンプル中に含まれる標的分析物の定量的検出のための分析装置であって、前記装置は、
検査ストリップであって、
第1の端部と第2の端部とを有するアッセイ支持部材と、
前記支持部材に隣接して略平行に取り付けられた多孔質分析膜であって、前記分析膜は第1の端部と第2の端部とを有しまた前記分析膜の前記第1と第2の端部の中間で前記分析膜に捕獲領域を備え、前記捕獲領域は前記分析膜の前記第1の端部から前記分析膜の前記第2の端部へ向かって移動する標識された分析物を捕獲するように構成される多孔質分析膜と、
分析しようとするサンプルを前記装置に導入するため前記支持部材の一端に設けたサンプル受け入れポートであって、
前記封止材料の上に配置されたチャネル層であって、開口部とチャネルとを含み、前記開口部は前記チャネルと液体連通を提供し、前記チャネルは前記多孔質分析膜と液体連通を提供することを特徴とするチャネル層と、
前記チャネル層の上に配置された親水性材料であって、前記チャネル層の前記開口部に対応する開口部を内部に有することを特徴とする親水性材料と、
前記親水性材料の上に配置されて前記ポートへの液体の導入を可能にするように内部に開口部を有するガスケット要素とを含むことを特徴とするサンプル受け入れポートと、
前記支持部材を支持するための底部筐体部分であって、略C字状の形状に構成されることを特徴とする底部筐体部分と、
前記底部筐体部分に対して構成的に略相補的な上部筐体部分であって、前記底部筐体上に嵌合し前記検査ストリップが前記C字状の開口部を横断するようにしてあり、前記ガスケット要素が前記サンプル受け入れポートと前記筐体の間の液体封止を提供することを特徴とする上部筐体部分と
を含むことを特徴とする装置。
- 前記支持部材に対向する側面上で前記分析膜を被覆する保護膜をさらに含み、前記保護膜が光学的に透明ではないことを特徴とする請求項7に記載の装置。
- 前記保護膜は前記多孔質膜と一体的に形成されることを特徴とする請求項8に記載の装置。
- 前記捕獲領域を通過した共役体の回収のために前記多孔質膜に設けた制御領域をさらに含んで前記検査ストリップが使用済みであることを示すことを特徴とする請求項7に記載の装置。
- 前記保護膜上に印刷された少なくとも1個の磁気構成領域をさらに含むことを特徴とする請求項10に記載の装置。
- 前記底部筐体部分は前記サンプル受け入れポートにより前記支持部材と分析膜に落し込まれるような形状と構成を有することを特徴とする請求項7に記載の装置。
- 前記アッセイ支持部材と前記チャネル層の間に液体封止材料をさらに含むことを特徴とする請求項7に記載の装置。
- 指向性フロー・イムノアッセイ、レセプタ・アッセイ、細胞アッセイ、又は分子アッセイを用いてサンプル中の標的分析物の定量的検出を行なうための方法であって、前記方法は、
前記サンプルと、超常磁性粒子であって前記サンプル中の標的分析物と結合するように処理された超常磁性粒子とを結合させ、前記結合により共役体を生成するステップと、
前記検査ストリップ上に位置するサンプル受け入れポートを用いて検査ストリップの多孔質膜の一端へ前記共役体を入れるステップであって、前記検査ストリップは捕獲領域を有することを特徴とするステップと、
前記共役体が毛細管現象により前記多孔質膜の中を移動するときに、前記多孔質膜の捕獲領域の中に前記共役体を捕獲するステップと、
磁気式アッセイ読み取り装置を用いて前記捕獲領域において標識された分析物の量を読み取るステップと
を含むことを特徴とする方法。
