PNAGは、多糖細胞間接着因子であり、場合によりN−アセチル基で置換されていてもよいβ(1→6)結合グルコサミンのポリマーから構成される。この多糖は、黄色ブドウ球菌及び表皮ブドウ球菌の両方に存在し、いずれの供与源からも単離することができる(Joyceら 2003, Carbohydrate Research 338; 903; Maira-Litranら 2002, Infect. Imun. 70; 4433)。例えば、PNAGは、黄色ブドウ球菌MN8m株から単離することができる(WO04/43407号)。

一実施形態では、本発明の免疫原性組成物は、米国特許第6294177号に記載の黄色ブドウ球菌336抗原を含む。
表皮ブドウ球菌のATCC−31432株、SE−360株及びSE−10株は、3つの異なる型の莢膜、それぞれI型、II型及びIII型を特徴とする(Ichiman及びYoshida 1981, J. Appl. Bacteriol. 51; 229)。表皮ブドウ球菌の各血清型から抽出される莢膜多糖は、I型、II型及びIII型多糖を形成する。多糖は、いくつかの方法(例えば、米国特許第4197290に記載の方法、又はIchimanら、1991, J. Appl. Bacteriol. 71; 176に記載の方法など)により抽出することができる。

このタンパク質は、ABC輸送体タンパク質であり、肺炎双球菌(S. pneumoniae)の接着因子PsaAのホモログ(相同体)である。このタンパク質は、高い免疫原性を有する32kDaのリポタンパク質であり、細菌細胞壁全体に分布している(Cockayneら Infect. Immun. 1998 66; 3767)。このタンパク質は、黄色ブドウ球菌及び表皮ブドウ球菌において32kDaのリポタンパク質として発現され、スタフィロコッカス・ホミニス(S. hominis)においては40kDaのホモログが存在する。表皮ブドウ球菌において、このタンパク質は、鉄制御オペロンの成分である。これは、ストレプトコッカス・パラサンギス(Streptococcus parasanguis)のFimAを含む接着因子、及び鉄輸送機能が立証された又は推測されているABC輸送体ファミリーのリポタンパク質の両方に対して顕著な相同性が見られる。

黄色ブドウ球菌のisd(iron-regulated surface determinant:鉄制御型表面決定因子)遺伝子は、ヘモグロビン結合及びヘム鉄の細胞質移行に関与するタンパク質をコードする。ヘム鉄は、必須栄養素として作用する。IsdA及びIsdBは、ブドウ球菌の細胞壁に局在してる。IsdAは、プロテイナーゼK消化感受性であることから細菌表面に露出していると考えられている。IsdBは、部分的に消化されたことから、細菌表面上に部分的に露出していることが示唆されている(Mazmanianら 2003 Science 299; 906)。
このタンパク質ファミリーのメンバーは、αトキシン、溶血素、エンテロトキシンB及びTSST−1のようなトキシン、並びにRAPのようなトキシンの生成を調節するタンパク質を含む。
αトキシンは、黄色ブドウ球菌の大部分の菌株によって産生される重要な病原性決定因子である。このタンパク質は、溶血作用をもった孔形成トキシンである。αトキシンに対する抗体は、動物モデルでαトキシンの有害な及び致死性の作用を中和することが示されている(Adlamら 1977 Infect. Immun. 17; 250)。ヒト血小板、内皮細胞、及び単核細胞は、αトキシンの作用に影響を受けやすい。
RAPは、それ自身はトキシンではないが、病原性因子の発現の調節因子である。RAPは、ブドウ球菌により産生され、分泌される。このタンパク質は、他のブドウ球菌のagr調節系を活性化し、かつ溶血素(ヘモリシン)、エンテロトキシンB及びTSST−1のような病原性因子の発現と、それに続く放出を活性化する。
蓄積関連タンパク質(Aap)
Aapは、140kDaのタンパク質であり、表面上での表皮ブドウ球菌株の蓄積に必須である(Hussainら Infect. Immun. 1997, 65; 519)。このタンパク質を発現している菌株は著しく多量の生物膜を産生したことから、Aapは生物膜形成に関与していると考えられている。Aapに対する抗体は、生物膜形成を抑制し、また表皮ブドウ球菌の蓄積を抑制することができる。
SsaAは、黄色ブドウ球菌及び表皮ブドウ球菌の双方で見出された30kDaの強力な免疫原性タンパク質である(Langら 2000 FEMS Immunol. Med. Microbiol. 29; 213)。心内膜炎罹患過程のその発現により、感染性疾患の発症に特異的な病原性の役割が示唆された。
ブドウ球菌感染は、いくつかの異なる段階を経て進行する。例えば、ブドウ球菌の生活環は、片利共生定着、隣接する組織若しくは血流に接近することによる感染の開始、血液中での嫌気的増殖、黄色ブドウ球菌病原性決定因子と宿主防御機構との間の相互作用、並びに心内膜炎、転移性膿瘍形成及び敗血症候群などの合併症の誘発を含む。細菌表面上の異なる分子が感染サイクルの異なるステップに関与する。ブドウ球菌感染の異なるプロセスに関与する特定の抗原の組み合わせに対する免疫応答を標的とすることにより、ブドウ球菌機能は多面的局面で影響を受ける。またこれは優れたワクチン効率をもたらし得る。

黄色ブドウ球菌の集団構造と関連する病原性因子の発生の解析により、黄色ブドウ球菌の自然集団における可変的な病原性遺伝子の存在が明らかとなった。
本発明の一実施形態において、免疫原性組成物は、ブドウ球菌の2以上の菌株に対して(例えば、黄色ブドウ球菌及び表皮ブドウ球菌の双方に由来する菌株に対して)有効な免疫応答を提供する。例えば、防御免疫応答は、黄色ブドウ球菌の5型及び8型血清型に対して発生する。

ライゲーション用の発現プラスミドpET24d又はpQE−30を調製するため、同様に適当な制限酵素で完全に消化した。調製したベクターに対してモル濃度で約5倍過剰の消化断片を用いてライゲーション反応を行った。標準的な約20μlのライゲーション反応(約16℃、約16時間)を、当該分野で周知の方法によりT4 DNAリガーゼ(約2.0ユニット/反応、Life Technologies社)を用いて行った。ライゲーション液のアリコート(約5μl)を用いて、M15(pREP4)又はBT21::DE3エレクトロコンピテント細胞を当該分野で周知の方法により形質転換した。約1.0mlのLB培地で37℃にて約2〜3時間増殖させた後、形質転換処理した細胞をアンピシリン(100μg/ml)及び/又はカナマイシン(30μg/ml)を含むLBアガープレートに塗布した。抗生物質は、選択に包含される。プレートを37℃で一晩(約16時間)インキュベートした。ApR/KanRの単一コロニーを滅菌した爪楊枝で拾い、新たに調製したLB ApR/KanRプレートに「パッチ」植菌し、同時に約1.0mlのLB Ap/Kanブロス培地に植菌するのに使用した。パッチプレート及びブロス培養の双方を標準インキュベータ(プレート)又は振とう式水浴のいずれかで37℃で一晩インキュベートした。全細胞に基づくPCR解析を用いて、DNA挿入物を含む形質転換体を確認した。ここで、約1.0mlの一晩培養したLB Ap/Kanブロス培養を1.5mlのポリプロピレンチューブに移し、細胞をBeckmann社の微量遠心機で遠心(室温にて約3分、約12,000×g)して回収した。細胞ペレットを約200μlの滅菌水に懸濁し、約10μlのアリコートを用いて、フォワード/リバース増幅プライマーの両方を含む最終容量約50μlのPCR反応液を調製した。最初に95℃の変性ステップを約3分間行い、細菌細胞の熱破砕及びプラスミドDNAの遊離を確実に行った。ABI社モデル9700サーマルサイクラーにより32サイクル、3ステップの熱増幅プロフィール(すなわち、95℃−45秒;55〜58℃−45秒;72℃−1分)を用いて、溶解した形質転換体のサンプルからBASB203断片を増幅した。熱増幅後、約20μlの反応液アリコートをアガロースゲル電気泳動で解析した(Tris−酢酸−EDTA(TAE)バッファ内で0.8%アガロースを用いて)。DNA断片をゲル電気泳動及びエチジウムブロマイド染色の後、UV照射により視認した。DNAの分子サイズ標準(1Kbラダー, Life Technologies社)を被検サンプルと同時並行で電気泳動し、それを用いてPCR産物のサイズを推定した。予想サイズのPCR産物を生成した形質転換体をタンパク質発現構築物を含む菌株として同定した。発現プラスミド含有株を、その後組換えタンパク質の発現誘導性について解析した。
一晩培養した前培養液のアリコート(約1.0ml)を約25mlのLB Ap/Kan培地を入れた125mlのエルレンマイヤーフラスコに植菌し、37℃で振とう(約250rpm)しながら、培養液の濁度がO.D.600で約0.5、すなわち対数増殖期(通常約1.5〜2.0時間)に達するまで培養した。このとき、約半分の培養液(約1.25ml)を第二の125mlフラスコに移し、組換えタンパク質の発現をIPTG(滅菌水で調製した1.0Mストック、Sigma社)を1.0mMの終濃度で添加することにより誘導した。IPTG誘導及び非誘導の両培地をさらに約4時間、37℃で振とうしながらインキュベーションし続けた。誘導及び非誘導両培地のサンプル(約10ml)をインキュベーション後に取り出し、細胞を微量遠心機で室温にて約3分間遠心することによって回収した。個々の細胞ペレットを約50μlの滅菌水に懸濁し、2−メルカプトエタノールを含む等量の2×Laemelli SDS−PAGEサンプルバッファと混合した。沸騰水浴に約3分間置き、タンパク質を変性させた。等量(約15μl)の粗製IPTG誘導及び非誘導細胞溶解物の両方を2つの12%Tris/グリシンポリアクリルアミドゲル(1mm厚Mini−gels, Novex社)上に加えた。誘導及び非誘導溶解サンプルを予め染色した分子量マーカー(SeeBlue, Novex社)と共に慣用条件下で標準的なSDS/Tris/グリシンランニングバッファ(BioRad社)を用いて電気泳動した。電気泳動後、一方のゲルをクマシーブリリアントブルーR250(BioRad社)で染色し、その後脱色して新規のIPTG誘導タンパク質を視覚化した。第二のゲルには、PVDFメンブレン(孔サイズ0.45ミクロン、Novex社)上にBioRad社Mini−ProteanIIブロッティング装置とTowbin(トービン)メタノール(20%)転写バッファを用いて、4℃にて約2時間かけてエレクトロブロッティングを行った。メンブレンのブロッキング及び抗体インキュベーションを当該分野で周知の方法に従って行った。抗RGS(His)3モノクローナル抗体、続いてHRP結合ウサギ抗マウス二次抗体(QiaGen社)を用いて、組換えタンパク質の発現の確認及び同定を行った。抗His抗体反応パターンの視覚化は、ABT不溶性基質を使用して又はAmersham社のECL化学発光システムを用いたHyperfilmを使用して行った。
細菌株
大腸菌の組換え発現用菌株であり、プラスミド(pQE30)を含んだM15(pREP4)株、又はブドウ球菌タンパク質をコードするプラスミドpET24dを含んだBL21::DE3株を用いて、組換えタンパク質の精製用細胞集団を調製した。
組換えタンパク質の調製に用いた発酵培地は、100μg/mlのアンピシリン及び/又は30μg/mlカナマイシンを含む2×YT培地(Difco)からなる。消泡剤を培養槽の培地に0.25ml/L(Antifoam204、Sigma社)で添加した。組換えタンパク質の発現を誘導するために、IPTG(イソプロピルβ−D−チオガラクトピラノシド)を発酵槽に加えた(終濃度;1mM)。
非変性条件下
IPTGを終濃度1mMで加えて、培養液をさらに4時間培養した。その後、培養液を6,000rpmで10分間遠心し、ペレットをプロテアーゼインヒビターカクテルを含むリン酸バッファ(50mM K2HPO4/KH2PO4:pH7)に懸濁した。このサンプルを1500バールの圧力でフレンチプレスにより(2回のランで)溶解した。15,000rpmで30分間の遠心の後、上清をさらなる精製のために回収し、NaClを0.5Mになるまで加えた。その後、サンプルを50mM K2HPO4/KH2PO4(pH7)で調整したNi−NTA樹脂(XK16カラムPharmacia社、Ni−NTA樹脂Qiagen社)に加えた。サンプルを加えた後、カラムをバッファA(0.2M NaH2PO4(pH7)/0.3M NaCl/10%グリセロール)で洗浄した。結合したタンパク質を溶出するため段階勾配を用いた。この勾配では、バッファB(0.2M NaH2PO4(pH7)/0.3M NaCl/10%グリセロール/200mMイミダゾール)が異なる割合でバッファAに加えられている。バッファBの割合は、10%から100%へと徐々に増加している。精製後、タンパク質を含む溶出分画をプールし、濃縮し、0.002M KH2PO4/K2HPO4(pH7.0)/15M NaClに対して透析した。
IPTGを終濃度1mMで添加し、培養液をさらに4時間培養した。その後、培養液を6,000rpmで10分間遠心し、ペレットをプロテアーゼインヒビターカクテルを含むリン酸バッファ(50mM K2HPO4/KH2PO4:pH7)に懸濁した。このサンプルを1500バールの圧力を用いてフレンチプレスにより(2回のランで)溶解した。15,000rpmで30分間遠心した後、ペレットを1M尿素を含むリン酸バッファで洗浄した。このサンプルを15,000rpmで30分間遠心し、ペレットを8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris−Hcl(pH8)に再懸濁し、室温に一晩置いた。サンプルを15,000rpmで20分遠心して、上清をさらなる精製のために回収した。それからサンプルを8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris−Hcl(pH8)で調整したNi−NTA樹脂(XK16カラムPharmacia社、Ni−NTA樹脂Qiagen社)に加えた。フロースルーの通過後、カラムをバッファA(8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris(pH8.0))、バッファC(8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris(pH6.3))、バッファD(8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris(pH5.9))及びバッファE(8M尿素/0.1M NaH2PO4/0.5M NaCl/0.01M Tris(pH4.5))で連続的に洗浄した。組換えタンパク質はバッファDとEでの洗浄の間にカラムから溶出した。変性組換えタンパク質は、尿素を欠く溶液に溶解することができる。このため、8M尿素に含まれる変性タンパク質を4M尿素/0.1MNa2PO4/0.01M Tris−HCl(pH7.1)、2M尿素/0.1M NaH2PO4/0.01M Tris−HCl(pH7.1)/0.5Mアルギニン、及び0.002M KH2PO4/K2HPO4(pH7.1)/0.15M NaCl/0.5Mアルギニンに対して連続的に透析した。
CDAPを用いた天然PS8の活性化及びカップリング化学

活性化及びカップリングを室温で連続的な攪拌下において行った。10mgの天然多糖を0.2M NaClに溶解し、最終PS濃度で2.5mg/mlの溶液を得た。その溶液を活性化ステップ前にpH6.0+/−0.2に調製した。
CDAPを用いたサイジングPS8の活性化及びカップリング化学
PSは、理論上10%の含水率で加重されている。2gの天然の含湿PSを初期濃度10mg/mlでWFIに一晩溶解した。サイジング前に、天然PS溶液を5μmのカットオフフィルターで浄化した。
活性化及びカップリングは、室温にて連続的な攪拌下において行った。

活性化及びカップリングを室温で連続的な攪拌下において行った。
破傷風トキソイドを0.2M NaClに溶解した5〜10mgの誘導体化多糖に加え、0.5N HClの添加によりpHをpH5.0又はpH6.0に調整した。EDACを0.1M Trisバッファ(pH7.5)に溶解し、その後EDACを10分間にわたって(1/5容量を2分毎)添加した。
活性化及びカップリングを室温で連続的撹拌下において行った。
100mgのTTを0.2M NaCl溶液中の50mgの誘導体化多糖に添加した。pHを0.3N HClの添加によりpH5.0±0.02に調整した。EDACを0.1M Trisバッファ(pH7.5)に溶解し、それから10分間にわたって(毎分1/10量)添加した。使用した条件(表8参照)に応じて、30〜180分後に1M Tris−HCl(pH7.5)の添加によって反応を停止した。SephacrylS400HRで精製する前に、複合体を5μmのMinisartフィルターを用いて清澄化した。その後、複合体をSephacryl S400HR(XK50/100)に注入した。150mM NaClを溶出バッファとして用いて、流速は60ml/時に固定した。溶出プールをレゾルシノール及びμBCAプロフィール(それぞれ多糖及びタンパク質用量を測定する)に基づいて選択した。その後、複合体を0.22μmの滅菌メンブレン(Millipack20)により10ml/分でろ過した。
脱−O−アセチル化
0.1N NaOHを16mlのサイジングしたPS(10mg/ml)に添加し、最終PS濃度9mg/ml及び最終NaOH濃度0.1Nとなるようにした。37℃で1又は2時間の処理後、PSは非処理PSと比較してそれぞれ35%及び12%(Hestrin用量)のO−アセチル化レベルを有していた。

本明細書の以下に記載する手法を用いて、下記の複合体を生成した:

1gのPNAGを20mg/mlの濃度で5N HClに溶解し、1時間インキュベートした。続いて5N NaOHを用いて中和した。この溶液を5μm膜メンブレンを用いて清澄化し、Sephacryl S400HRにおいて精製した。目的の画分、すなわち「中間の分子サイズ」(Infection and Immunity, 70: 4433-4440 (2002)を参照)に相当するものをプールし、濃縮したあと、脱N−アセチル化処理を行った。
S−GMBS(N−(γ−マレイミドブチリルオキシ)スルホスクシンイミド、Pierce)を0.2M NaCl中のdPNAGに添加し(S−GMBS/PS比(w/w):1/1)、pH7.0(1M NaOHを用いてpH調節)にて室温で2時間にわたりインキュベートした。ToyopearlHW−40FにおいてPBS、10mM EDTA、50mM NaCl(pH7.2)を溶出バッファーとして60ml/時の一定の流速で用いて精製することにより過剰のGMBSと副産物を除去した。溶出プールを光学密度(UV=206nm)の関数で選択し、続いてVivaspin管3,000MWCO又はAmiconUltra 10,000MWCOにおいて濃縮した。
GMBS活性化dPNAG及びDTT還元(reduced)TT−SPDPを混合し、室温で攪拌した。使用した条件に応じて、20〜120分後にシステイン(リン酸ナトリウムバッファーpH8.0中4mg/ml)を30分かけて添加することにより反応を停止させた。複合体を5μmフィルターを用いて清澄化し、精製のためにSephacryl S300HR樹脂(XK16/100)に注入した。200mM NaClを用いて30ml/時の一定の流速で溶出を行った。溶出画分をヘキソサミンにより及びタンパク質用量により分析した。目的の画分をプールし、0.22μm Sterivexにおいてろ過した。最終的な複合体は、多糖(ヘキソサミン用量)及びタンパク質組成(ローリー用量)について試験した。
DMSO(ジメチルスルホキシド,Merck)に溶解した5倍モル過剰のSPDP(N−スクシンイミジル−3−(2−ピリジルジチオ)プロピオネート,MW:312.4,Pierce)を、100mMリン酸ナトリウム(pH7.2)中5mg/mlのdPNAG100mgに添加し、室温で1時間インキュベートした。反応混合物をAmiconUltra 10,000MWCO(3000rpmで28分間の遠心)で±6mlにまで濃縮した後、Sephacryl S100HR(XK16/40)で精製を行った。リン酸バッファー(pH7.4)を用いて60ml/時の一定の流速で溶出を行った。目的の画分(206nmで読み取り)をプールし、AmiconUltra 10,000MWCO(3000rpmで30分間の遠心)において1.1mlになるまで濃縮した。
DMSO(ジメチルスルホキシド,Merck)に溶解した15倍モル過剰のSPDP(Pierce)を、100mMリン酸ナトリウム(pH7.2)中のTT(50/ml)1gに添加し、室温で80分間インキュベートした。続いて、生成物をSephacryl S100HR(XK16/40)に注入し、100mM酢酸ナトリウム(pH5.6),100mM NaCl,1mM EDTAを60ml/時の一定の流速で用いて溶出した。溶出プールを光学密度(UV=280nm)の関数で選択し、続いてAmiconUltra 10,000MWCO(3000rpmで75分間の遠心)において19.6mlになるまで濃縮した。
DTTをTT−SPDP又はTT−LC−SPDPにDTT/TT比(mg/mg)が0.7/1となるよう添加した。室温で2時間後、ピリジン−2−チオンが放出されて、その特徴的な吸収が343nmとなった。ゲルろ過(PD−10,Amersham)によりチオール化タンパク質を過剰のDTTから精製した。AmiconUltra 10,000MWCOにおいて濃縮した後、タンパク質含量をローリー用量により評価した。
カップリングは、連続的に攪拌しながら、初期TT/PS比(w/w)2/1で、室温にて実施した。
11.6mgのDTT(1,4−ジチオトレイトール,Boerhinger Mannheim,MW:154.24)を16.5mgのdPNAG−SPDPに添加した。室温で2時間後、ピリジン−2−チオンが放出されて、その特徴的な吸収が343nmとなった。ゲルろ過(Toyopearl HW40F)によりチオール化PSを過剰のDTTから精製し、続いてAmiconUltra 10,000MWCOにおいて860μlになるまで濃縮した。
カップリングは、連続的に攪拌しながら、初期TT/PS比(w/w)1.7/1で、室温にて実施した。
アジュバント組成物
アジュバントAをアジュバントとして添加した又はアジュバントを添加していない、以下の組成を有する複合体を接種した。

雌CD−1マウス(8〜10週齢)は、Charles River Laboratories社(Kingston, Mass)から購入した。致死率検証のために、9〜11匹のCD−1マウスからなる5つの群の腹腔内(i.p.)にCSAプレート上で増殖させた黄色ブドウ球菌を段階希釈して投与した。接種量は、約1010から108CFU/マウスの範囲とした。死亡率は、3日間毎日評価した。50%致死用量(LD50S)は、用量応答関係のプロビットモデルを用いて評価した。通常のLD50の帰無仮説は、尤度比検定で検証した。亜致死性菌血症は、8〜20匹のマウスからなる群に約2×106CFU/マウスを静脈内(i.v.)経路で、又は約2×107CFU/マウスをi.p.経路で投与することによって生じさせた。接種後、個々の動物群について、特定の時間に尾から血液を採取し、菌血症レベルを5%ヒツジ血液を含んだトリプシン大豆アガープレート(Becton Dickinson Microbiology Systems社)で二重試験にて行った量的平板計数により評価した。統計的有意性は、ウエルチの改変型対応のないスチューデントt検定で判定した。
30匹のマウスの群をアジュバント添加していない又はアジュバントAと組み合わせた糖用量3μgの黄色ブドウ球菌PS8−TT複合体を用いて、0日目、14日目、28日目、及び42日目に皮下接種した。0日目に、0.001〜0.013μgを含む最初の糖用量をマウスに接種した。さらに食塩水に溶解した0.3μgの用量で免疫付与を3回行った。55日目に、血清をそのマウスから回収し、各血清サンプルをELISAによって検証し、PS8に対する免疫応答を評価した。10匹のマウスの群をコントロール群に用いて、それらには食塩水又はアジュバントAを含む食塩水のいずれかを接種した。
リン酸緩衝食塩水(PBS)で希釈したメチル化HSA(2.5μg/ml)と混合した精製PNAG(2.5μg/ml)を高結合性マイクロタイタープレート(Nunc Maxisorp社)上に4℃で一晩かけて被覆した。
ヒト多形核白血球(PMN)による黄色ブドウ球菌のin vitroオプソニン貪食性殺傷をXuら 1992 Infect. Immun. 60; 1358に記載のように行った。ヒトPNMは、ヘパリン添加した血液から3%デキストランT−250における沈降により調製した。オプソニン反応混合物(1ml)は、熱不活化ウシ胎児血清を10%添加したRPMI1640培地中に約106のPMN、約108CFUの黄色ブドウ球菌、及び0.1mlの検査血清又はIgG調製物を含んでいる。過免疫付与したウサギ血清をポジティブコントロールとして用い、また0.1mlの免疫付与していないウサギ血清をIgGサンプルの完全な供与源として用いた。反応混合物を37℃でインキュベートし、細菌サンプルを0分、60分、及び120分の時点で水に移し、続いて希釈し、トリプシン大豆アガープレート上に広げ、一晩インキュベート後の細菌数をカウントするため37℃でインキュベートした。
動物に精製したブドウ球菌タンパク質を免疫し、過免疫血清を発生させた。マウスに3回(0日、14日、及び28日目)、スペコール(Specol)をアジュバントとして添加した10μgの各タンパク質を免疫した。ウサギに3回(0日、21日、及び42日目)、スペコールをアジュバントとして添加した20μgの各タンパク質を免疫した。免疫血清を回収し、抗タンパク質及び抗全死菌ELISAで評価した。
精製したタンパク質を、リン酸緩衝生理食塩水(PBS)に溶解した1μg/ml溶液として高結合性マイクロタイタープレート(Nunc Maxisorp社)上に4℃で一晩かけて被覆した。プレートをPBS−1%BSAで室温にて30分間撹拌しながらブロッキングした。試験サンプルを1/1000に希釈し、室温で撹拌しながら1時間インキュベートした。洗浄後、結合したマウス及びウサギ抗体をPBS−0.05%tweenで1:5000に希釈したJackson ImmunoLaboratories Inc.社のペルオキシダーゼ結合AffiniPureヤギ抗マウスIgG(H+L)(カタログ番号115−035−003)又はAffiniPureヤギ抗ウサギIgG(H+L)(カタログ番号11−035−003)を用いて検出した。検出抗体を30分間室温で撹拌しながらインキュベートした。発色は、10mlの0.1Mクエン酸バッファ(pH4.5)あたり4mgのOPD(Sigma社)+5μlのH2O2の溶液を用いて15分間室温で暗所にて行った。反応は、50μlのHClで停止させ、650nmに対して490nmで光学密度を読み取った。
5型及び8型黄色ブドウ球菌又は表皮ブドウ球菌Hay株に由来する全死菌(熱又はホルムアルデヒドで不活化したもの)をリン酸緩衝生理食塩水(PBS)に溶解した20μg/ml溶液として、高結合性マイクロタイタープレート(Nunc Maxisorp社)上に4℃で蒸発させながら一晩かけて被覆した。プレートをPBS−1%BSAで室温にて30分間撹拌しながらブロッキングした。プロテインAを、PBS−0.05%tweenで希釈した10μg/mlのアフィニティー精製ニワトリ抗プロテインA(ICLカタログ番号CPA−65A−2)の添加によって中和した。続いて、室温で1時間インキュベートした。その後、試験サンプルをPBS−0.05%の入ったマイクロプレート上で1/10の開始希釈から2倍に希釈し、1時間室温で撹拌しながらインキュベートした。洗浄後、結合したマウス又はウサギ抗体を、PBS−0.05%tweenで1:5000に希釈したJackson ImmunoLaboratories Inc.社のペルオキシダーゼ結合AffiniPureヤギ抗マウスIgG(H+L)(カタログ番号115−035−003)又はAffiniPureヤギ抗ウサギIgG(H+L)(カタログ番号11−035−003)を用いて検出した。検出抗体を30分間室温で撹拌しながらインキュベートした。発色は、10mlの0.1Mクエン酸バッファ(pH4.5)あたり4mgのOPD(Sigma社)+5μlのH2O2を用いて15分間室温で暗所にて行った。反応は50μlのHClで停止させ、650nmに対して490nmで光学密度を読み取った。
3匹のコットンラットからなる15の群に8種のブドウ球菌抗原の組み合わせを接種し、またコントロールとして機能する5匹のコットンラットを抗原なしで処置した。これら16の群については以下の通りである:

鼻腔内保菌アッセイの結果を表19に示す。コントロール群は、平均3.51335のlogCFU/鼻腔を有しており、鼻腔内保菌の減少は、ブドウ球菌タンパク質を接種したコットンラットの全群で見ることができた。群4、9、及び13は、CFU/鼻腔において2ログを超える減少を示しており、鼻腔内保菌での最大減少を示した。群12及び16もまた、CFU/鼻腔で約2ログの減少を示した。
