Abstract

レヴィー小体病(LBD)の特徴は、ドーパミン作動系の変性、運動性変調、認知障害、およびレヴィー小体(LB)の形成である(非特許文献1)。LDBとしては、パーキンソン病、びまん性レヴィー小体病(DLBD)、レヴィー小体異型アルツハイマー病(LBV)、パーキンソン病(PD)とアルツハイマー病(AD)との併発、および多系統萎縮症(MSA)とみなされる症候群が挙げられる。レヴィー小体を伴う痴呆(DLB)というのは、LBD間の専門用語の相違を調整するために作られた用語である。LBを有する疾患は、依然として、高齢者における運動性疾患および認識力低下に共通する原因となっている(非特許文献2)。この疾患の発生率は増加し続けており、深刻な公衆衛生問題を生じているが、今日まで、こうした疾患に対して認可された治療法はない(非特許文献3)。LBDの原因については意見の分かれるところであり、各種の神経毒および遺伝的感受性要因などのさまざまな要因が役割を果たしていると提唱されている。

「因子」という用語は、薬理活性を有するか、有する可能性のある化合物を記述するのに用いている。因子としては、既知の薬物である化合物、薬理活性は確認されているが、さらに治療上の評価が行われている化合物、および薬理活性についてスクリーニングされることになっているコレクションおよびライブラリを構成する物質である化合物が挙げられる。
I. 概要
本発明は、アルファ−シヌクレインをリン酸化し、および/またはその産生を阻害する1種以上のキナーゼを阻害することによりレヴィー小体病(LDB)を抑制することができるという洞察に、部分的に基づいている。アルファ−シヌクレインはレヴィー小体(LB)に存在する主要な蛋白質である。本発明の実施はメカニズムの理解に左右されるものではないが、アルファ−シヌクレインのコドン129におけるリン酸化は、アルファ−シヌクレインの細胞内沈着物の形成をもたらす一連の分子事象の1つであると考えられている。ser−129でリン酸化されているアルファ−シヌクレインは、びまん性レヴィー小体病(DLBD)、多系統萎縮症(MSA)および家族型パーキンソン病(PD)のレヴィー小体(LB)に極めて多く含まれている。LBにおけるリン酸化アルファ−シヌクレインの異常な蓄積は、リン酸化シヌクレインがLB形成を駆動する病原性種であること、およびそのリン酸化に関与し、またはアルファ−シヌクレイン自体の産生を調節するキナーゼ(類)が多発性シヌクレオパチー(multiple synucleinopathies)の治療における治療上の標的(群)であることを示している。このシリーズの他の事象としては、リン酸化に続く蛋白質分解的切断が挙げられそうである(2004年5月19日出願の国際公開公報第WO2005/013889号参照)。本願では、その阻害がアルファ−シヌクレインのリン酸化の減少および/またはトータル・アルファ−シヌクレイン濃度の低下を伴う数種のキナーゼを特定したことを報告する。本発明はこれらのキナーゼの活性および発現の調節物質を特定する方法ならびにレヴィー小体病を処置する方法を提供する。
表1A、1Bおよび1Cには、蛋白質であって、これを阻害することによりser−129の位置のリン酸化が調節される蛋白質を示した。表1Aにはセリンおよび/またはスレオニン残基および、場合によっては、チロシンをリン酸化することができるキナーゼを示した。表1Bにはセリン残基を(知られている限り)修飾することができないチロシンキナーゼを示した。表1Cにはキナーゼ活性を有しないことが知られている蛋白質を示した。表1Aの上部からのキナーゼはアルファ−シヌクレインのser−129を直接リン酸化することができる候補である。また、表1Bの上部からのキナーゼは、アルファ−シヌクレインを間接的にリン酸化する役割を介する治療上の有用な標的である。表1Cの上部の蛋白質も、同じ理由から治療上の有用な標的である。各表の第1、2および3欄には、遺伝子名、キナーゼ名およびキナーゼのジェンバンク・アセッション番号を示した。その次の欄には、キナーゼの阻害がリン酸化を低下させる(「下降(down)」)のか、増大させる(「上昇(up)」)のかを示した。その次の3欄には、リン酸化の測定レベルが3回の独立した実験の平均値から外れる標準偏差の数を示した。最後の2欄にはキナーゼファミリー(即ち、アミノ酸特異性)および群を示した。

(Uedaほか、Proc. Natl. Acad. Sci. USA (1993年) 90: 11282−6;ジェンバンク・アセッション番号:P37840)。この蛋白質は、認識される3つのドメイン、即ち、1から61番目のアミノ酸をカバーするKTKE繰返し配列ドメイン(repeat domain)、約60番目から95番目のアミノ酸に及ぶNAC(非アミロイド成分)ドメイン、および約98番目から140番目のアミノ酸に及ぶC末端酸性ドメインを有する。
既知の配列の任意の目的遺伝子の発現を抑制するのに、亜鉛フィンガー蛋白質、リボザイム、siRNA、アンチセンスRNAなどの特性のよく分かった一般的クラスの多くの因子を用いることができる。亜鉛フィンガー蛋白質は、目的遺伝子の発現を活性化するのに用いることもできる。抑制の対象となる遺伝子は、好ましくはPLK2またはGRK6である。何故なら、これらの遺伝子によりコードされているキナーゼはアルファ−シヌクレインを直接リン酸化するからである。PLK2が特に好ましい。また、APEG1、CDC7L1、MET GRK1、GRK2、CRK6、IKBKB、CKIIおよびGRK7は、これらが直接キナーゼの間接的活性化因子である可能性が高いので、抑制の好ましい標的である。PRKG1、MAPK13およびGAKはレヴィー小体病における活性化の好ましい候補である。何故なら、これらはアルファ−シヌクレインのリン酸化の負の調節因子であるからである。シンフィリンは抑制の好ましい標的である。何故なら、これは、キナーゼではないが、(通常、PLK2などのキナーゼの存在下に)アルファ−シヌクレインのリン酸化の増大に関係しているからである。
アルファ−シヌクレインのリン酸化またはその産生を直接的もしくは間接的に調節するキナーゼは、実施例で示したように、特定することができる。一般に、潜在的阻害剤のライブラリはキナーゼ遺伝子のコレクションの既知の配列に基づいて設計される。このライブラリを構成する物質は、上述した種類の分子のいずれかとすることができる。次に、このライブラリの構成物質をアルファ−シヌクレイン発現細胞に導入する。この細胞およびアルファ−シヌクレインはいずれもヒトのものであることが好ましい。通常、このような細胞にはヒト・アルファ−シヌクレインをコードしているDNAおよび試験対象のライブラリ構成物質をコードしているDNAの両者がトランスフェクトされる。ライブラリ構成物質は個別に、または、まとめてスクリーニングすることができる。ライブラリ構成物質を導入し、このライブラリ構成物質が発現されてそのキナーゼを抑制するのに十分な期間培養した後、トータル・アルファ−シヌクレインおよびリン酸化アルファ−シヌクレインの濃度を測定し、キナーゼ発現を抑制するライブラリ構成物質を処置しなかったがその他の点では同様な対照細胞における対応する濃度と比較する。測定は、アルファ−シヌクレインのトータル濃度を測定するためのアルファ−シヌクレイン(好ましくは、ヒト・アルファ−シヌクレイン)に特異的な抗体およびリン酸化アルファ−シヌクレインの濃度を測定するためのリン酸化アルファ−シヌクレインに特異的な抗体を用いてイムノアッセイにより行った。代表的な抗体については2004年11月8日出願のPCT/US2004/037444に記載されている(引用により本明細書に組み込まれている)。通常の測定誤差の範囲から外れるという意味で有意である、処置および対照細胞間でのリン酸化アルファ−シヌクレイン濃度の低下は、この細胞に導入した阻害剤がキナーゼを阻害し、これによりアルファ−シヌクレインのリン酸化が直接的または間接的に影響を受けたことを示すものである。このキナーゼの同一性(identity)は、阻害剤を個々にスクリーニングすることにより、あるいは阻害剤をまとめてスクリーニングする場合には阻害剤をコードしている核酸の配列を決定することにより、阻害剤の同一性から決めることができる。同様に、アルファ−シヌクレインのトータル濃度に関して、そのような濃度の測定における通常の測定誤差の範囲を外れる、処置および対照細胞間での低下は、阻害剤がアルファ−シヌクレインの発現レベルに間接的に影響するキナーゼを阻害することを示すものである。
種々の化合物について、キナーゼの発現または活性を調節(通常、阻害)する能力をスクリーニングすることができる。PLK2またはGRK6は、アルファ−シヌクレインを直接リン酸化することができるキナーゼ候補であるため、阻害のための好ましいキナーゼである。PLK2は、GRK6またはここで試験した他のキナーゼよりもはるかに高レベルでアルファ−シヌクレインをリン酸化することが分かっているので、特に好ましいキナーゼである。APEG1、CDC7L1、MET GRK1、GRK2、GRK6、IKBKB、CKIIおよびGRK7も、この直接キナーゼの間接的な活性化因子である可能性が高いので、阻害のための好ましいキナーゼである。PRKG1、MAPK13およびGAKは、アルファ−シヌクレインのリン酸化の負の調節因子であるので、レヴィー小体病における活性化のための好ましいキナーゼである。あるいは、こうしたキナーゼ自体または同様な活性を有するその断片もしくは模倣剤はアルファ−シヌクレインのリン酸化の阻害剤としてそのまま用いることができる。シヌクレインは、アルファ−シヌクレインのリン酸化の阻害のための治療上の代替的な標的として用いることができる。例えば、アルファ−シヌクレインおよびPLK2発現を有するアッセイにシンフィリンを加えて、シンフィリンの阻害剤を特定することができる。
トランスジェニック動物モデルは、上記のようにアルファ−シヌクレインをリン酸化し、レヴィー様小体を形成するキナーゼの能力を試験するのに有用である。また、トランスジェニック動物は、アルファ−シヌクレインのリン酸化または産生を調節する際の活性について因子をスクリーニングするのにも有用である。特に好ましい因子は、PLK2およびGRK6、またはAPEG1、CDC7L1、MET GRK1、GRK2、GRK6、IKBKB、CKIIおよびGRK7を含むキナーゼを阻害するか阻害すると考えられる因子である。また、キナーゼPRKG1、MAPK13およびGAKを活性化するか活性化すると考えられる因子も好ましい。さらに、ノックアウト動物(即ち、siRNA、亜鉛フィンガー蛋白質などによる挿入不活化またはトランス抑制によって内因性キナーゼが不活化されている動物)は動物に対するキナーゼ活性除去の効果を確認するのに有用である。例えば、PLK2−ノックアウトマウスを分析することによって、PLK2阻害剤が何らかの副作用を示すかどうかが分かる。類似のノックアウトマウスによれば、他のキナーゼについての同様の情報が明らかとなる。
アルファ−シヌクレインを直接的または間接的にリン酸化するキナーゼの発現または活性を調節する因子は種々のアッセイによって特定することができる。特に好ましい因子は、キナーゼPLK2もしくはGRK6、またはAPEG1、CDC7L1、MET GRK1、GRK2、GRK6、IKBKB、CKIIおよびGRK7を阻害する因子、あるいはPRKG1、MAPK13およびGAKを活性化させる因子である。発現を調節する因子は細胞を利用したアッセイで特定することができるが、このアッセイでは、アルファ−シヌクレイン、およびアルファ−シヌクレインを直接または間接的にリン酸化するかトータル・アルファ−シヌクレインの濃度を調節するキナーゼを発現する細胞中に試験対象の因子を導入する。必要に応じて、特にPLK2の場合、さらにシンフィリンを発現させることによりこのキナーゼの活性を増強することができる。この因子は、直接、またはこの因子をコードし発現することができる核酸の形で導入することができる。細胞は自然界でアルファ−シヌクレインおよびキナーゼを発現することができ、あるいはこれらの片方または両方を適当な核酸のトランスフェクトによって細胞内に導入することができる。この因子のキナーゼ発現に対する効果は、このキナーゼもしくはそのmRNAの濃度から直接的に、あるいは前述のように、リン酸化アルファ−シヌクレインまたはトータル・アルファ−シヌクレインの濃度を測定することにより間接的に測定することができる。キナーゼmRNAの濃度はハイブリダイゼーション型アッセイによって分析することができる。キナーゼの濃度はイムノアッセイによって分析することができる。必要に応じて、キナーゼは、検出を容易にするためにフラッグ(Flag)(商標)などのペプチド標識で標識する(Hoppほか、BioTechnology 6: 1204−1210 (1988年))。キナーゼの濃度を低下させ、アルファ−シヌクレインのリン酸化のレベルを減少させ、および/または因子を処置されていない同様な対照細胞との比較でトータル・アルファ−シヌクレインの濃度を低下させる因子は、レヴィー小体病の治療に有用な可能性のある薬理活性を有する。
レヴィー小体病(LBD)の特徴は、ドーパミン作動系の変性、運動性変調、認知障害、およびレヴィー小体(LB)の形成である。(McKeithほか、Clinical and pathological diagnosis of dementia with Lewy bodies (DLB): Report of the CDLB International Workshop, Neurology (1996年) 47:1113−24)。レヴィー小体は神経細胞に見出される球状の蛋白質沈着物である。これが脳内に存在すると、アセチルコリン、ドーパミンなどの化学的メッセンジャーの作用が遮断されて脳の正常な機能が妨げられる。レヴィー小体病としては、(特発性パーキンソン病(PD)を含む)パーキンソン病、レヴィー小体を有する痴呆(DLB)としても知られるびまん性レヴィー小体病(DLBD)、アルツハイマー病とパーキンソン病との併発、および多系統萎縮症(MSA)が挙げられる。DLBDは、アルツハイマーおよびパーキンソン病の症状を共有する。DLBDは、主にレヴィー小体の存在部位がパーキンソン病と異なる。DLBDではレヴィー小体は主として皮質に形成される。パーキンソン病ではこれは主として黒質に形成される。他のレヴィー小体病としては、自律神経失調症、レヴィー小体性燕下困難(Lewy body dysphagia)、偶発性LDB、遺伝性LDB(例えば、アルファ−シヌクレイン遺伝子の突然変異体PARK3およびPARK4)ならびに多系統萎縮症(例えば、オリーブ橋小脳萎縮症、線条体黒質変性症およびシャイ・ドレーガー症候群)が挙げられる。
本発明は、レヴィー小体病に罹患しているか、罹患するリスクを有する患者においてこの疾患を予防もしくは治療するいくつかの方法を提供する。治療剤としては、アルファ−シヌクレインのリン酸化を阻害し、および/またはアルファ−シヌクレインのトータル濃度を低下させる上記因子の全てが挙げられる。
実施例1:アルファ−シヌクレインのリン酸化を調節するキナーゼのスクリーニング
α−シヌクレインをセリン129においてリン酸化するキナーゼもしくはキナーゼ類を特定するために、定量化可能な量のリン酸化α−シヌクレインを含む細胞を用いてsiRNAキナーゼ・ライブラリ(アンビオン社)をスクリーニングした。CMVプロモータの制御下にあるヒトの野生型α−シヌクレインを安定的にトランスフェクトしたヒト胎児腎細胞株HEK293細胞(PEAK細胞)(PEAK−Syn細胞)に対して、597種のヒト・キナーゼを標的とするsiRNAを100nMトランスフェクトし、ELISAアッセイにより測定してトータルおよびリン酸化シヌクレイン濃度を定量した。リン酸化アルファ−シヌクレインの割合を変えるsiRNAに対して95種のキナーゼが特定された(表1乃至3参照)。このうち28種はセリン残基をリン酸化するキナーゼのクラスに属し、従って、α−シヌクレインをセリン129において直接リン酸化することができた。その他はチロシンキナーゼであった。チロシンキナーゼはα−シヌクレインをser−129において直接リン酸化しないが、アルファ−シヌクレインキナーゼの上流調節剤として作用することができる。これらのser/thrキナーゼのうちの2種、カゼインキナーゼ2およびカルシウム/カルモジュリン依存性蛋白質キナーゼIIはα−シヌクレインをインビトロでリン酸化することが報告されており(Proninほか、J. Biol. Chem. 275: 26515−26522 (2000年); Okochoaほか、J. Biol. Chem. 275: 390−397 (2000年); Nakajoほか、Eur. J. Biochem. 217: 1057−1063 (1993年))、カゼインキナーゼ2阻害剤は細胞内のリン酸化シヌクレイン濃度を増加させることが報告されている(Okochoaほか、2000年)。GRKファミリーの構成キナーゼのうちの数種は(GRKではないが)アルファ−シヌクレインをインビトロでリン酸化することが報告されている(Proninほか、2000年)。ハエにおけるGRK2の発現はリン酸化シヌクレイン濃度を増加させることが報告されている(Chen、Feany、Nature Neurosci. 8: 657−663 (2005年))。

アルファ−シヌクレインのリン酸化の増大または低下を示した実施例1のキナーゼについて、アルファ−シヌクレインに対する効果を確認するために再試験した。この確認のためのスクリーニングは、実施例1で特定した標的および対象とする数種の別のキナーゼに対して10nMのsiRNAを用いて行った。実施例1でより高い濃度のsiRNAを用いたのは、不完全に設計されたsiRNAに引き起こされるほんの僅かのノックダウンも観察することができるようにするためであった。この確認スクリーニングではるかに低いsiRNA濃度を用いることによって、候補のリストを極めて特異的な調節を示すものにのみ絞ることができた。アンビオン社によってその後無効であると報告された一部のsiRNAについても再スクリーニングした(下記のリプレースメントライブラリ・スクリーニング参照)。最後に、いく種かの新たに特定したキナーゼについてスクリーニングし、これらの結果をすでにプールされている結果に加えた。候補として特定したキナーゼを定量的RT−PCR(qRT−PCR)によって試験することにより、これらが実際にPEAK−Syn細胞内に存在すること確認した(実施例6参照)。この確認および再スクリーニングの実験方法および結果は以下の通りであった。
確認スクリーニングの結果は下記に示す4つのカテゴリーにグループ分けした。
a)3つのsiRNAのいずれも10nMでリン酸化シヌクレイン濃度に何ら影響を及ぼさない、および/または
b)これらのsiRNAが100nMでの一次スクリーニングで認められたのと逆の表現型を生じる。

最初のスクリーニングで使用した一部のsiRNAは質が劣るとみなされたので、リプレースメントsiRNAを用いて両濃度でスクリーニングを行った。リプレースメントsiRNAのデータは、最初のスクリーニングからの特異的siRNAで得られた結果のデータと取替えるために用いた。統計データは各キナーゼの3種のsiRNAについて一覧にし、これを用いて、9種の別のキナーゼを一次スクリーニングとしてとらえ損ねた、最初のスクリーニングからの候補と認定した。これらを再試験したところ、このうちの2種のキナーゼが10nMのsiRNAで「部分的確認」であった。これらはBCKDKおよびFLJ25965(KSR2)であった。
上記のsiRNAスクリーニングからのキナーゼのどれがアルファ−シヌクレインを直接リン酸化するのかを明らかにするために、精製キナーゼをアルファ−シヌクレインとインキュベートしてインビトロのキナーゼ反応を行わせた。この結果から、PLK2ならびにGRK2、5、6および7 (GPRK2、5、6および7)は全てアルファ−シヌクレインを特異的にセリン129でリン酸化することができ、インビトロではセリン87をリン酸化しないことが示され、これらがアルファ−シヌクレインを直接リン酸化することができることが分かった。また、MET、CDC7L1およびIKBKBはアルファ−シヌクレインを直接リン酸化することができないことが示された(図1A乃至C)。
上記インビトロ・アッセイにおけるGRK6およびPLK2(GRKファミリーと系統的に関連のあるポロ様キナーゼ)による活性の有意なレベルは、上記RNAiスクリーニングにおけるリン酸化の低下因子としてのPLK2、GRK2およびGRK1の特定と相俟って、他のGRK類のさらに包括的な調査を促進した。図4AおよびBには、このインビトロ・アッセイで比較したGRK2、4、5、6および7ならびにPLK2の結果を示した。これらの結果は、デンシトメトリー比較においてS129リン酸化に対する特異性が明確であることを示すものである。この選択性は、CKI>GRK6>PLK2>GRK4>GRK5>GRK2と表すことができる。GRK7は容易に検出できるほどのレベルでリン酸化することができなかった。酸性配列フランキングアミノ酸129に特異性を示す全てのGRK類はser−87をリン酸化することはできなかった。こうした反応はELISA測定により定量化して確認した。これらの値は、GRK類に対してPLK2濃度の明らかな減少を示すイムノブロットデータとおおよそ一致した。AS基質の大部分は上記アッセイデザインに基づいて使い果たされた(リン酸化された)のであり、その測定値は反応における最大OD/基質を表すものと考えられる。
キナーゼは細胞内におけるよりもインビトロでより無規律となり得るので、アッセイを細胞株を用いて行うことによりアルファ−シヌクレインとの直接的相互作用を確認した。実施例3からのインビトロでアルファ−シヌクレインをリン酸化したキナーゼのcDNAをPEAK−Syn細胞株中にトランスフェクトしてどのキナーゼが細胞内でアルファ−シヌクレインのser−129をリン酸化することができるかを調べた。その結果から、GRK6および、さらに著しくはPLK2が細胞内でアルファ−シヌクレインのリン酸化を媒介することができることが分かった(図5)。
別の会社(ダーマコン(Dharmacon))製のPLK2 siRNAもアルファ−シヌクレインのリン酸化を阻害することを示すことによって、実施例4のデータはPLK2についてさらに裏付けられた。このことは、PLK2がアルファ−シヌクレインをセリン129で直接リン酸化する細胞キナーゼとしての有望な候補であることを示すデータ(表2および13)を補強するものであった。
シンフィリンは、アルファ−シヌクレインを結合することが明らかにされているシヌクレイン関連蛋白質である。シンフィリンの存在がアルファ−シヌクレインのリン酸化を増強することができるかどうかを明らかにするために、アルファ−シヌクレインおよびPLK2を含むHEK細胞と含まないHEK細胞内にこれを過剰発現させた。標準的プロトコルに従ってトランスフェクトを行った後、アルファ−シヌクレインのELISAおよび分析を実施した。また、細胞を採取してウエスタンブロット分析を行った。トランスフェクトした細胞の溶解物について、1H7抗体を用いてトータルシヌクレイン、11A5抗体を用いてセリン129リン酸化シヌクレインを分析した。細胞内にトランスフェクトされたDNAの総量は96穴プレートの穴当たり0.16μgと一定のままであった。細胞内に導入されるDNAの種類は、DNAの完全な割り当て(quota)を形成するのに用いるべき空のベクターによってさまざまに異なる。種々の濃度のアルファ−シヌクレイン、PLK2およびシンフィリンcDNAを未処置のHEK細胞内に導入した。これら3種の全てをトランスフェクトした細胞はトータルシヌクレインのわずかな増加を示した。リン酸化シヌクレインについては、トランスフェクトしていない細胞中の濃度は定量限界未満であった。アルファ−シヌクレインを単独で導入すると、5.2%のリン酸化シヌクレインが得られ、これはシヌクレインとシンフィリンの同時トランスフェクトの場合(5.4%のリン酸化シヌクレイン)よりもわずかに少なかった。PLK2とシヌクレインを同時トランスフェクトすると、60%リン酸化シヌクレインと、HEK−syn安定細胞内にPLK2をトランスフェクトした場合に認められたのと同様な濃度が得られた。驚くべきことに、3種のDNA(PLK2、シヌクレインおよびシンフィリン)の全てを同時に過剰発現させると、HEK細胞中のリン酸化シヌクレイン濃度は83.3%となった。従って、PLK2、シヌクレイン過剰発現HEK細胞においてシンフィリンはシヌクレインのリン酸化を増大させた。
アルファ−シヌクレインの多くの既知家族性突然変異体のPLK2によるリン酸化を検討するために、インビトロトランスフェクト研究を行い、アルファ−シヌクレインおよびその突然変異体のリン酸化を分析した。これらの家族性突然変異体(FPD)はA30P、A53TおよびE46Kであった。
対象キナーゼがHEK−シヌクレインおよびSY5Y−シヌクレイン細胞で発現されるかどうかを明らかにするためにqRT−PCRを行った。表16では、全てのサンプルはGAPDH発現に対して標準化した。さらに、陰性キナーゼのうちの2種は各実験で対照として分析した。直接キナーゼである可能性のある24種の候補のうち、HEK293−シヌクレイン細胞ではPLK2を含む20種が検出された。従って、残る「完全確認」キナーゼが上記細胞内に検出された(図9)。試験した直接キナーゼである可能性のあるもののうち、4種、即ち、GPRK1、GPRK7、ERK8およびRIPK3は検出されなかった。GPRK6は辛うじて検出可能であった。
アルファ−シヌクレインを安定的にトランスフェクトした293細胞にPLKおよびGRKを過剰発現させた。ELISAおよびウエスタンブロットを行ってPLKおよびGRKキナーゼによるリン酸化シヌクレインの増加を確認し、リン酸化の増大を証明した。別の方法を用いてその増大を、293細胞に対して免疫染色にELISAで使用するのと同じビオチン化抗体(11A5)を用いることにより確認した。この方法では、PLK2および、程度は少ないがGRKをトランスフェクトした細胞中のリン酸化シヌクレインの増加も証明された。この増加は11A5で明るく染色される少数の細胞で検出され、全ての細胞での全般的な増加がみられるのではない。トータルシヌクレインの量(5C12抗体を用いて測定)は変化しないように思われた。これは293細胞内のリン酸化の有意な増大であった。従って、こうした結果が神経芽腫細胞で再現され得るかどうかを調べることは重要であった。
PLK2はG1相細胞周期調節蛋白質であるので、アルファ−シヌクレインのPLK2によるリン酸化が細胞周期調節に変化をもたらすかどうかを確認することは重要である。細胞がより集密的に増殖するにつれて細胞内のリン酸化シヌクレインの割合が減少することは、以前に(増殖が減速するに従って)観察された。このことは、アルファ−シヌクレインをリン酸化する1種以上の直接キナーゼが細胞周期の調節に関与していることと極めてよく一致している。従って、PLK2に対する1種以上の抗体を用いてウエスタンブロットにより標準化蛋白質濃度を分析することによって、PLK2濃度の減少がこの観察されたリン酸化シヌクレイン濃度の減少および細胞の集密化と相関しているかどうかを明らかにする
。
重要なのはレヴィー小体病および/またはPD病状の細胞モデルの特定であった。このため、イン・ビボでアルファ−シヌクレイン沈着のモデルを確立するために、ヒト培養皮質細胞においてアルファ−シヌクレインのレンチウイルス媒介発現を行った。従って、ドナーおよび野生型および変異型アルファ−シヌクレインを過剰発現するHCC細胞で実験を行い、細胞を断片化して細胞内の野生型アルファ−シヌクレインおよび変異型アルファ−シヌクレインの場所を特定した。以下の結果から、HCC細胞はLB病に合致する仕方でアルファ−シヌクレインの凝集を示したことが分かる。さらに、PLK2を発現させると、その凝集だけでなくアルファ−シヌクレインのリン酸化も増大した。
推定アルファ−シヌクレインキナーゼの確認のためにアルファ−シヌクレインノックアウト(アルファ−シヌクレインKO)マウス脳を利用することは、以下の点でsiRNAスクリーニングよりも有利である: 1) 脳材料を用いることにより、HEK細胞株では得られないと思われる適切で恐らくより高いレベルの脳特異的キナーゼ活性が得られること、2)脳には、上記細胞には存在しないと思われる補因子(脂質、蛋白質など)が存在すること、および3)リン酸化ASとして検出される可能性のある如何なる内因性アルファ−シヌクレインも存在しないこと。測定可能なキナーゼ活性が粗材料中に存在するかどうかを明らかにするために、組換えアルファ−シヌクレイン(rAS)含有(可溶性および界面活性剤可溶性)抽出物25乃至50μgの封入体を250μMのATPを用いて評価した。図8AはrASの対応する使用量を表す各反応におけるトータルアルファ−シヌクレインを示す。図8Bでは、ser−129リン酸化アルファ−シヌクレインの濃度を調べた。TBS(蔗糖可溶性)およびTX抽出物の両者中のrASがリン酸化され、TBS材料からのシグナルのレベルはTX材料からのそれのほぼ2倍であった(但し、反応は蛋白質に対して標準化しなかった)。リン酸化レベルはCKIの添加によって増大したが、大豆由来リン脂質の添加では有意な影響を受けなかった。図9のser−87リン酸化では同一のブロットをプローブした。このpAbは100ngのrASと交差反応性を示すので、バックグラウンドを超えるレベルはS87部位での真のリン酸化を表す。TBSおよびTX反応のいずれにおいても、この部位での有意なリン酸化は認められなかったが、CKIスパイクは測定可能なレベルのリン酸化に達した。これらの実験から、測定可能な紛れもないキナーゼ活性(類)がKOマウス脳の可溶性および膜画分に存在し、S87に比しS129に対して特異的であることが示唆される。アルファ−シヌクレインには他のセリンまたはスレオニン部位におけるリン酸化の可能性が存在するが、そのような修飾を検出するための抗体はまだ利用可能ではない。このように、測定可能なレベルのser−129特異的キナーゼ活性/活性類がアルファ−シヌクレインKOマウス脳抽出物に存在するので、このような活性を有するものを脳からのキナーゼ精製の原料とすることができるかも知れない。
Claims (34)
- レヴィー小体病(LBD)を治療する活性について因子をスクリーニングする方法であって、
(a)表1A、B;C、表2、表12または表13に示したキナーゼの活性または発現を調節する因子を同定する工程、および
(b)該因子がLBDの動物モデルにおいて該疾患を治療するのに有用な活性を示すか否かを決定する工程
を含む、方法。 - 前記調節が阻害である、請求項1に記載の方法。
- 工程(a)は、前記因子が前記キナーゼを阻害するか否かを特定することを含む、請求項1に記載の方法。
- 工程(a)が、前記キナーゼおよび/またはアルファ−シヌクレインを発現する核酸で形質転換した細胞において行われる、請求項3の方法。
- 前記細胞が神経細胞である、請求項4に記載の方法。
- 工程(a)がインビトロで行われる、請求項3に記載の方法。
- 工程(b)がLB疾患のトランスジェニック動物モデルにおいて行われる、請求項1に記載の方法。
- 前記細胞がヒト・アルファ−シヌクレインを発現する導入遺伝子を含む、請求項5に記載の方法。
- 前記アルファ−シヌクレインがアルファ−シヌクレインの家族性突然変異体である、請求項4に記載の方法。
- 前記家族性突然変異体がA30P、A53TおよびE46Kからなる群より選択される、請求項9に記載の方法。
- 前記キナーゼがAPEG1、PLK2、CDC7L1、PRKG1、MAPK13、GAK、RHOK、ADRBK1、ADRBK2、GRK2L、GRK5、GRK6、GRK7、CKIIA2、CKIIA1、IKBKBおよびMETからなる群より選択される、請求項1に記載の方法。
- 前記キナーゼがAPEG1、PLK2、CDC7L1、RHOK、ADRBK1、ADRBK2、GRK2L、GRK5、GRK6、GRK7、CKIIA2、CKIIA1、IKBKBおよびMETからなる群より選択される、請求項2に記載の方法。
- 前記キナーゼがPRKG1、MAPK13およびGAKからなる群から選ばれ、前記調節が活性化である、請求項1に記載の方法。
- 前記キナーゼがPLK2、IKBKBおよびGRK6からなる群より選択される、請求項13に記載の方法。
- 前記キナーゼがPLK2である、請求項14に記載の方法。
- 前記キナーゼがIKBKBである、請求項14に記載の方法。
- 前記同定工程がシンフィリンの存在下に前記因子をPLK2蛋白質と接触させることを含む、請求項15に記載の方法。
- 工程(b)が前記トランスジェニック動物を前記因子と接触させ、該因子が、該因子を処置しなかった対照トランスジェニック動物と比べてアルファ−シヌクレインの沈着物の形成を阻害するか否かを決定することを含む、請求項7に記載の方法。
- 前記動物モデルがA30P、A53TおよびE46Kからなる群より選択される突然変異を有するアルファ−シヌクレイン導入遺伝子を含むトランスジェニック動物である、請求項1に記載の方法。
- LB疾患の治療もしくは予防を行う方法であって、
該疾患に罹患しているか罹患するリスクのある患者に対して、表1A、BもしくはC、表2、表12または表13に示したキナーゼの活性または発現を調節するのに有効な因子の有効な療法を施行する工程を含み、該因子が
該キナーゼに対する抗体、
該キナーゼの発現を調節する亜鉛フィンガー蛋白質、
該キナーゼの核酸配列に相補的な配列を有するアンチセンスRNA、siRNA、リボザイムもしくはRNA
である、方法。 - 前記調節が阻害である、請求項20に記載の方法。
- 前記キナーゼがAPEG1、PLK2、CDC7L1、RHOK、ADRBK1、ADRBK2、GRK2L、GRK5、GRK6、GRK7、CKIIA2、CKIIA1、IKBKBおよびMETからなる群より選択される、請求項20に記載の方法。
- 前記キナーゼがPLK2、IKBKBおよびGRK6からなる群より選択される、請求項22に記載の方法。
- 前記キナーゼがPLK2である、請求項23に記載の方法。
- 前記キナーゼがIKBKBである、請求項23に記載の方法。
- さらにシンフィリンの存在下にPLK2を調節する因子を特定することを含む、請求項20に記載の方法。
- 前記キナーゼがPRKG1、MAPK13およびGAKからなる群から選ばれ、前記調節が活性化である、請求項20に記載の方法。
- アルファ−シヌクレインをリン酸化するキナーゼを同定する方法であって、
アルファ−シヌクレインを発現する細胞に対してキナーゼをコードしている遺伝子に相補的な配列を有する核酸または該遺伝子に特異的に結合する亜鉛フィンガー蛋白質をトランスフェクトする工程であって、これによって該トランスフクトされた核酸または亜鉛フィンガー蛋白質が該キナーゼの発現を阻害する、工程、および
該細胞のリン酸化アルファ−シヌクレインの量を、siRNAまたは該siRNAをコードしている核酸をトランスフェクトしていない対照細胞と比較して測定する工程であって、リン酸化アルファ−シヌクレインの減少が該キナーゼがアルファ−シヌクレインをリン酸化する指標を提供する、工程
を包含する、方法。 - さらに、前記核酸をトランスフェクトしていない対照細胞と比較して、前記細胞により産生されるアルファ−シヌクレインの量を測定することを含む、請求項28に記載の方法。
- 前記核酸がsiRNAまたは該siRNAをコードしているDNA分子である、請求項28に記載の方法。
- 前記アルファ−シヌクレインがアルファ−シヌクレインの家族性突然変異体である、請求項28に記載の方法。
- 前記家族性突然変異体がA30P、A53TおよびE46Kからなる群より選択される、請求項31に記載の方法。
- レヴィー小体病(LBD)を治療する活性について因子をスクリーニングする方法であって、
(a)シンフィリンの活性または発現を調節する因子を同定する工程、および
(b)該因子がLBDの動物モデルにおいて該疾患を治療するのに有用な活性を示すか否かを決定する工程
を包含する、方法。 - Ser−129リン酸化アルファ−シヌクレインを産生するための方法であって、
細菌細胞内にアルファ−シヌクレインをコードしているプラスミドおよびPLK2をコードしているプラスミドを提供する工程、
該細胞を培養し、それにより該プラスミドが同時発現してアルファ−シヌクレインおよびPLK2を産生する工程であって、それにより該PLK2が細菌細胞内でアルファ−スイ(alpha−suy)をリン酸化する、工程、ならびに
該細胞からリン酸化アルファ−シヌクレインを単離する工程
を包含する、方法。
