限られた数の分子のみの分析の証拠によれば、双極性障害(BPD)および大うつ病性障害(MDD)の病態生理学に、また、こうした障害に対する薬物治療の機構に、遺伝子調節の変化および特有の遺伝子調節異常が関与し得ること示唆される。マイクロアレイ技術の最近の発達により、特定の遺伝子、系、およびシグナル伝達経路のmRNA発現プロフィールの包括的な見方が可能になる。
精神障害の遺伝的基準を理解するために、特に気分障害を患う対象の中枢神経系中に差次的に発現される遺伝子の発現パターンを調べるための研究が行われている。いくつかの研究では、知られている遺伝子および新規の遺伝子の差次的および特有の発現を、Affymetrix Gene Chips(登録商標)(高密度オリゴヌクレオチドプローブセットアレイを含有する)を用いて、BPおよびMDD患者の死後脳から精製された全RNAサンプルを調べることによって決定した。基本的な原理は、BPおよび/またはMDDにおいて、(健康な対照被検者と比較して)差次的に発現される遺伝子および経路を、上述のようなトランスクリプトームの全体的な発現プロファイリングを介して、同定することによるものであり、それによって、疾患を引き起こす、疾患に影響を与える、あるいは関連する遺伝子、あるいは診断および治療用途で使用するための疾患を治療するために使用される薬物と相互作用する遺伝子を、同定することができる。

一般に、対象タンパク質をコードする核酸は、cDNAまたはゲノムDNAをコードするように作製されるDNA配列ライブラリからクローン化される。特定の配列は、オリゴヌクレオチドプローブ(その配列は、PCRプライマーに対する基準を提供し、特定のプローブを単離するのに適した領域を定義する、図1、図5〜8、または表1〜4に記載される遺伝子の配列から得ることができる)とハイブリッド形成させることによって位置決めすることができる。あるいは、この配列を、発現ライブラリにクローン化させる場合、発現された組換え型タンパク質を、抗血清、あるいは、図1、図5〜8、または表1〜4に記載される遺伝子によってコードされるアミノ酸配列を含むポリペプチドに対して作製される精製された抗体を用いて免疫学的に検出することができる。
本発明の天然に存在するまたは組換え型ポリペプチドは、機能分析で使用するために精製することができる。天然に存在するポリペプチド(例えば、図1、図5〜8、または表1〜4に記載される遺伝子によってコードされるポリペプチド)は、例えば、マウスまたは脳などのヒト組織、あるいは任意の他の起源のオルソログから精製することができる。組換え型ポリペプチドは、任意の適切な発現系から精製することができる。
組換え型タンパク質が、一般的にプロモーター誘導の後、形質転換された細菌によって大量に発現される場合、発現は恒常的である可能性があるが、タンパク質は、不溶性の凝集体を形成する可能性がある。タンパク質封入体の精製に適した、いくつかのプロトコルが存在する。例えば、凝集タンパク質(以下、封入体と呼ぶ)の精製は一般的に、それだけには限らないが一般的に、約100〜150μg/mlのリゾチームおよび0.1% Nonidet P40(非イオン性界面活性剤)の緩衝液中での保温による細菌細胞の破壊による、封入体の抽出、分離、および/または精製を含む。細胞懸濁液は、Polytronグラインダー(Brinkman Instruments、Westbury,NY)を使用して、すりつぶすことができる。あるいは、細胞を、氷上で超音波処理することができる。細菌を溶解させる代替の方法は、AusubelらおよびSambrookら(どちらも上記文献)に記載されており、当業者には明白である。
1.溶解度分画
しばしば最初のステップとして、また、タンパク質混合物が複合的である場合、初期の塩分画によって、当該の組換え型タンパク質から、多くの望ましくない宿主細胞タンパク質(または細胞培地から得られるタンパク質)を分離することができる。好ましい塩は、硫酸アンモニウムである。硫酸アンモニウムは、タンパク質混合物中の水量を効率的に減少させることによって、タンパク質を沈殿させる。その後、タンパク質は、その溶解度に基づいて沈殿する。タンパク質は、より疎水性であるほど、低い硫酸アンモニウム濃度で、より沈殿しやすいと考えられる。典型的なプロトコルは、得られる硫酸アンモニウム濃度が20〜30%の間であるように、タンパク質溶液に飽和硫酸アンモニウムを加えることである。これによって、大抵の疎水性タンパク質が沈殿することとなろう。(当該のタンパク質が疎水性でないならば)沈殿物を捨て、硫酸アンモニウムを、当該のタンパク質を沈殿させることが知られている濃度で上清に加える。次いで、沈殿物を緩衝液に可溶化させ、必要に応じて、透析またはダイアフィルトレーションを介して、過剰な塩を取り除く。タンパク質の溶解度に依存する他の方法(冷エタノール沈殿など)は、当業者によく知られており、複雑なタンパク質混合物を分画するために使用することができる。
算出された分子量に基づいて、より大きなサイズ、またり小さなサイズのタンパク質を、異なる孔径の膜(例えば、AmiconまたはMillipore膜)を介する限外濾過を使用して単離することができる。第一段階として、タンパク質混合物を、当該のタンパク質の分子量よりも小さい分子量出口を有する孔径をもつ膜を介して限外濾過する。次いで、限外濾過の残留物を、当該のタンパク質の分子量よりも大きい分子出口をもつ膜に対して限外濾過する。組換え型タンパク質は、膜から濾液に通過する。次いで、この濾液を、以下で述べる通りのクロマトグラフィーにかけることができる。
当該のタンパク質はまた、その大きさ、正味の表面電荷、疎水性、およびリガンドに対する親和性に基づいて、他のタンパク質から分離することができる。さらに、タンパク質に対して産生された抗体は、カラム基材および免疫精製されたタンパク質に結合させることができる。こうした方法はすべて、当分野でよく知られている。
当業者は、本発明のポリヌクレオチドの発現の検出が、多くの用途を有することを認識するであろう。例えば、本明細書に述べる通り、患者における本発明のポリペプチドまたはポリヌクレオチドのレベルの検出は、気分障害または精神病性障害、あるいは気分障害または精神病性障害の素因を診断するために有用である。さらに、遺伝子発現の検出は、本発明のポリペプチドまたはポリヌクレオチドの発現の調節因子を同定するために有用である。
核酸ハイブリダイゼーション技術を使用するポリヌクレオチド発現の検出に加えて、本発明のポリペプチドを検出するために、イムノアッセイを使用することもできる。イムノアッセイは、ポリペプチドを質的または量的に分析するために使用することができる。適用可能な技術の一般的な概要は、Harlow&Lane Antibodies:A Laboratory Manual(1988)において見出される。
当該のタンパク質または他の免疫原と特異的に反応するポリクローナルおよびモノクローナル抗体をもたらすための方法は、当業者に知られている(例えば、Coligan、上記文献;およびHarlowおよびLane、上記文献;Stitesら、上記文献、およびその中の引用文献;Goding、上記文献;ならびにKohlerおよびMilstein Nature,256:495−497(1975)を参照のこと)。こうした技術には、ファージまたは類似のベクターにおける、組換え抗体のライブラリからの抗体の選択による抗体調製が含まれる(Huseら、上記文献;およびWardら、上記文献を参照のこと)。例えば、イムノアッセイで使用するための抗血清を産生するために、当該のタンパク質またはその抗原性フラグメントは、本明細書に記述する通りに単離される。例えば、組換え型タンパク質は、形質転換細胞系で産生される。マウスまたはウサギの近交系は、フロイントアジュバントなどの標準のアジュバントおよび標準の免疫化プロトコルを使用して、タンパク質を用いて免疫される。あるいは、本明細書に開示された配列から得られ、かつ担体タンパク質に結合された合成ペプチドを、免疫原として使用することができる。
好ましい実施態様では、当該のタンパク質は、いくつかのよく知られた免疫学的な結合アッセイのいずれかを使用して検出および/または定量化される(例えば、米国特許第4,366,241号;第4,376,110号;第4,517,288号;および第4,837,168号を参照のこと)。一般的なイムノアッセイの総説については、Asai Methods in Cell Biology Volume 37:Antibodies in Cell Biology、Academic Press,Inc.NY(1993);Stites、上記文献も参照のこと。免疫学的な結合アッセイ(またはイムノアッセイ)は一般的に、分析物(この場合、本発明のポリペプチドまたはその抗原性の部分配列)と特異的に結合し、しばしばこれを固定するために、「捕獲(capture)剤」を利用する。捕獲剤は、分析物に特異的に結合する部分である。好ましい実施態様では、捕獲剤は、例えば、本発明のポリペプチドと特異的に結合する抗体である。抗体は、当業者によく知られたいくつかの手段のいずれかによって、上で述べた通りに産生することができる。
組織サンプルから当該のタンパク質を検出するためのイムノアッセイは、競合的または非競合的である可能性がある。非競合イムノアッセイは、捕獲された分析物(この場合タンパク質)の量が、直接的に測定される分析である。ある好ましい「サンドイッチ」分析では、例えば、捕獲剤(例えば図1、図5〜8、または表1〜4に列挙される遺伝子によってコードされるポリペプチドに特異的な抗体)を、それが固定される固体基板に直接的に結合させることができる。次いで、こうした固定された抗体は、試験サンプル中に存在するポリペプチドを捕獲する。このように固定されたポリペプチドは、次いで、標識をもつ第二抗体などの標識化薬剤によって結合される。あるいは、第二抗体は、標識を欠いている可能性があるが、これは、続いて、第二抗体が得られる種の抗体に特異的な、標識された第3の抗体に結合される可能性がある。第二抗体は、ビオチンなどの検出可能な部分で改変することができ、これに、酵素標識されたストレプトアビジンなどの第3の標識分子が、特異的に結合することができる。
競合アッセイでは、サンプル中に存在する(図1、図5〜8、または表1〜4に列挙される遺伝子によってコードされるポリペプチドなどの)分析物の量は、サンプル中に存在する分析物によって捕獲剤(例えば分析物に特異的な抗体)から置き換えられる(またはそれと競合する)加えられた(外因性の)分析物の量を測定することによって、間接的に測定される。ある競合アッセイでは、この場合では既知量の当該のタンパク質が、サンプルに加えられ、次いで、サンプルが捕獲薬剤と接触され、この場合には抗体が本発明のポリペプチドに特異的に結合する。抗体に結合される免疫原の量は、サンプル中に存在する免疫原の濃度に反比例する。特に好ましい実施形態では、抗体は、固体基板上に固定される。例えば、抗体に対する結合されるポリペプチドの量は、タンパク質/抗体複合体中に存在する対象タンパク質の量を測定することによって、あるいは、残留する非複合型タンパク質の量を測定することによって決定することができる。タンパク質の量は、標識されたタンパク質分子を提供することによって検出することができる。
特に好ましい実施形態では、サンプル中の本発明のポリペプチドの存在を検出および定量化するために、ウェスタンブロット(免疫ブロット)分析が使用される。この技術は一般に、分子量に基づいて、ゲル電気泳動によってサンプルタンパク質を分離すること、適切な固体支持体(例えばニトロセルロースフィルター、ナイロンフィルターまたは誘導体化されたナイロンフィルターなど)に、分離されたタンパク質を移すこと、および当該のタンパク質と特異的に結合する抗体と共にサンプルを保温することを含む。例えば、抗体は、固体支持体上の当該のポリペプチドに特異的に結合する。こうした抗体は、直接的に標識することもできるし、当該のタンパク質に対する抗体に特異的に結合する標識された抗体(例えば標識されたヒツジ抗マウス抗体)を使用して、その後検出することもできる。
分析において使用される特定の標識または検出可能な基は、それが分析において使用される抗体の特定の結合を有意に妨げない限り、本発明の重要な態様ではない。検出可能な基は、検出可能な物理または化学特性を有している任意の材料であり得る。こうした検出可能な標識は、イムノアッセイの分野において十分に開発されており、一般に、こうした方法に有用なほとんどの標識を、本発明に適用可能である。したがって、標識は、分光学的、光化学的、生化学的、免疫化学的、電気的、光学的、または化学的手段によって検出可能な任意の組成物である。本発明において有用な標識には、磁気ビーズ(例えばDynabeads(商標))、蛍光色素(例えば、フルオレセインイソチオシアネート、テキサスレッド、ローダミンなど)、放射標識(例えば3H、125I、35S、14C、または32P)、酵素(例えば、ホースラディッシュペルオキシダーゼ、アルカリ性ホスファターゼ、およびELISAで一般に使用される他のもの)、およびコロイド金または着色ガラスもしくはプラスチック(例えばポリスチレン、ポリプロピレン、ラテックスなど)ビーズなどの比色標識が含まれる。
本発明のポリペプチドまたはポリヌクレオチドの調節因子、すなわちその活性のアゴニストまたはアンタゴニスト、あるいはポリペプチドまたはポリヌクレオチド発現の調節因子は、気分障害または精神病性障害を含めたいくつかのヒトの疾患を治療するために有用である。本発明のポリヌクレオチドまたはポリペプチドの発現を調節するアゴニスト、アンタゴニスト、または他の薬剤の投与を、気分障害または精神病性障害を患う患者を治療するために使用することができる。
細胞、特に哺乳類細胞、とりわけヒト細胞における本発明のポリペプチドおよびポリヌクレオチドの発現レベルまたは活性を調節する薬剤を同定するために、いくつかの異なるスクリーニング・プロトコルを利用することができる。一般的には、スクリーニング法は、例えば、本発明のポリペプチドに結合させること、あるいはポリペプチドに結合する抑制剤を調節すること、あるいはポリペプチドまたはポリヌクレオチドの発現を活性化することによって、ポリペプチド活性を調節する薬剤を特定するために、複数の薬剤をスクリーニングすることを含む。
こうして特定される少なくともいくつかの薬剤が、おそらくポリペプチド活性の調節因子であると考えられるので、本発明のポリペプチドへの結合が可能な薬剤に対するスクリーニングによって、予備的なスクリーニングが行われる可能性がある。結合アッセイは通常、本発明のポリペプチドを、1つ以上の試験薬剤と接触させることと、タンパク質と試験薬剤が結合複合体を形成するのに十分な時間を与えることを含む。形成されるいずれの結合複合体も、いくつかの確立された解析手法のいずれかを使用して検出することができる。タンパク質結合アッセイには、それだけには限らないが、共沈、非変性SDS−ポリアクリルアミドゲル上の共遊走およびウェスタンブロット上の共遊走を測定する方法が含まれる。Neurotransmitter Receptor Binding(Yamamura,H.I.ら編)内の、BennetおよびYamamura、(1985)「Neurotransmitter,Hormone or Drug Receptor Binding Methods」(pp.61−89)を参照のこと。こうした分析に利用されるタンパク質は、天然に発現される可能性もあるし、クローン化される可能性もあるし、合成される可能性もある。
特定のスクリーニング法は、本発明のポリペプチドまたはポリヌクレオチドの発現を上方または下方調節する化合物についてスクリーニングすることを含む。こうした方法は、概して、試験化合物を、本発明のポリペプチドまたはポリヌクレオチドを発現する1つ以上の細胞と接触させるような、細胞に基づく分析を行うことと、その後、発現の増減(転写産物、翻訳産物、または触媒産物のいずれか)を検出することが必要である。ある種の分析は、末梢細胞、または本発明の内因性のポリペプチドまたはポリヌクレオチドを発現する他の細胞を用いて実施される。
本発明のポリペプチドの触媒活性は、酵素による産物の産生を測定することによって、あるいは基質の消費を測定することによって決定することができる。活性は、触媒反応の速度か、該ポリペプチドが基質と結合する(Km)あるいは触媒産物(Kd)を放出する能力かのいずれかを指す。
前述のスクリーニング法のいずれかによって最初に特定された薬剤は、明確な活性を検証するために、さらに試験することができる。こうした研究は、適切な動物モデルを用いて行われることが好ましい。こうした方法の基本形式は、最初のスクリーニング中に特定されたリード化合物を、人間に対するモデルとして働く動物に投与し、その後、本発明のポリヌクレオチドまたはポリペプチドの発現または活性が実際に上方調節されたかどうか決定することを含む。検証研究に利用される動物モデルは概して、任意の種類の哺乳類である。適切な動物の具体例としては、それだけには限らないが、霊長類、マウス、およびラットが挙げられる。本明細書に記述する通り、既知の治療薬の投与を使用するモデルが、有用であり得る。
精神障害の動物モデルはまた、調節因子のスクリーニングへの使用が見いだされている。一実施形態では、ショウジョウバエモデルなどの無脊椎動物モデルを、本明細書に開示されるヒト遺伝子のショウジョウバエ・オルソログの調節因子のスクリーニングのために使用することができる。別の実施形態では、例えば、適切な遺伝子ターゲティングベクターを用いる相同的組換えまたは遺伝子過剰発現の結果としての遺伝子ノックアウト技術を含めたトランスジェニック動物技術は、本発明のポリヌクレオチドまたはポリペプチドの発現を、存在させない、低下させる、あるいは増大させることとなる。同じ技術はまた、ノックアウト細胞を作製するためにも適用することができる。所望される場合、本発明のポリヌクレオチドまたはポリペプチドの組織特異的発現またはノックアウトが必要である可能性がある。こうした方法によって産生されるトランスジェニック動物は、精神病の動物モデルとしての使用が見い出されており、精神病の調節因子のスクリーニングに有用である。
本発明のポリペプチドまたはポリヌクレオチドの調節因子として試験される薬剤は、任意の小さな化学物質または生物学的実体(タンパク質、糖、核酸、または脂質など)であり得る。あるいは、調節因子は、ポリペプチドまたはポリヌクレオチドの遺伝子改変型(例えば、FGF2、NCAM、またはFGF系のペプチドインヒビターの組換え型または変化型)であり得る。一般的に、試験化合物は、化学小分子およびペプチドである。基本的には、いずれの化学物質も、本発明の分析において潜在的な調節因子またはリガンドとして使用される可能性があるが、多くの場合、水性または有機(とりわけDMSOベースの)溶媒に溶解可能である化合物が使用される。これらの分析は、一般には並行して行われる、分析ステップを自動化すること、また、分析に対する任意の好都合な起源由来の化合物を提供することによって(例えば、ロボットアッセイにおけるマイクロタイタープレート上のマイクロタイター形式で)、大きな化学的なライブラリをスクリーニングするために設計される。周知のことだが、Sigma(St.Louis,MO)、Aldrich(St.Louis,MO)、Sigma−Aldrich(St.Louis,MO)、Fluka Chemika−Biochemica Analytika(Buchs,Switzerland)などを含めて、化学物質の多くの供給会社が存在する。また、調節因子としては、本発明のmRNA(例えばアンチセンス分子、リボザイム、DNAzymeなど)のレベル、またはmRNAからの翻訳のレベルを低下させるために設計された薬剤が含まれる。
本発明のハイスループット分析では、1日で、数千にも及ぶ異なる調節因子またはリガンドをスクリーニングすることが可能である。特に、マイクロタイタープレートの各ウェルは、選択される潜在的調節因子に対して別々の分析を行うために使用することもできるし、濃度またはインキュベーション時間の影響が観察されることになっている場合、5〜10のウェル毎に、単一の調節因子が試験される可能性もある。このように、単一の標準のマイクロタイタープレートは、約100(例えば96)の調節因子を分析することができる。1536ウェルのプレートが使用される場合、単一のプレートは、約100から1500の異なる化合物を容易に分析することができる。1日につきいくつかの異なるプレートを分析することが可能であり、本発明の統合されたシステムを使用して、最高約6,000〜20,000までの異なる化合物に対する分析スクリーニングが可能である。最近では、試薬操作に対する微小流体アプローチが開発されている。
本発明のポリペプチドまたはポリヌクレオチドの活性を調節する化合物に対するさらに別の分析は、コンピュータ支援ドラッグデザインを含む。ここでは、コンピュータシステムは、そのアミノ酸またはヌクレオチド配列によってコードされる構造情報に基づいて、ポリペプチドまたはポリヌクレオチドの三次元構造を作り出すために使用される。入力シーケンスは、コンピュータプログラム中にあらかじめ確立されたアルゴリズムを用いて、直接的かつ能動的に相互作用し、分子の二次、三次、および、四次構造モデルがもたらされる。本発明のポリペプチドまたはポリヌクレオチドの潜在的受容体または結合パートナーに関して、類似の分析を実施することができる。次いで、例えば本発明のポリペプチドまたはポリヌクレオチドと結合する能力を有する構造の領域を特定するために、タンパク質またはヌクレオチド構造のモデルを調べる。次いで、こうした領域を、本発明のポリペプチドまたはポリヌクレオチドに結合するポリペプチドを特定するために使用する。
本発明は、本発明のポリペプチドまたはポリヌクレオチド、本発明のポリペプチドまたはポリヌクレオチドのための特定の抗体などを使用する、本明細書に記述する分析を実践するための組成物、キット、および統合されたシステムを提供する。
本発明のポリヌクレオチドまたはポリペプチドの調節因子(例えば、FGF2、NCAMなどのアンタゴニストまたはアゴニスト、FGF系のペプチドインヒビター、または精神障害を患う対象において過剰発現する遺伝子のsiRNAおよび/またはアンチセンスインヒビターを、in vivoでそれらの分子の活性を調節するために、哺乳類の対象に直接的に投与することができる。投与は、調節因子化合物を、治療される組織と最終的に接触させるために通常使用され、当業者によく知られている経路のいずれかによるものである。特定の組成物を投与するために、2つ以上の経路を使用することもできるが、ある特定の経路が、別の経路よりも即時的で、より有効な反応をしばしば提供する可能性がある。
様々なヒトの疾患は、遺伝子が転写され、遺伝子産物が細胞中に産生されるように、遺伝子をヒト細胞に安定に導入することを含む治療アプローチによって治療することができる。この手法による治療が適用できる疾患には、単一または複数の遺伝子に欠損があるものを含めた遺伝性疾患が含まれる。遺伝子治療はまた、後天性疾患および他の状態の治療にも有用である。遺伝性疾患ならびに後天性疾患の治療に対する遺伝子治療の適用に関する議論については、Miller、Nature 357:455−460(1992);およびMulligan、Science 260:926−932(1993)を参照のこと。
細胞または生物への送達のために、本発明のポリヌクレオチドを、ベクターに組み込むことができる。こうした目的のために使用されるベクターの例には、標的細胞中の核酸の発現を導くことが可能な発現プラスミドが含まれる。他の例では、ベクターは、ウイルスベクター系であり、ここでは、核酸は、標的細胞を形質移入することが可能であるウイルスゲノムに組み込まれる。好ましい実施態様では、ポリヌクレオチドは、所望の標的宿主細胞の遺伝子の発現を導くことが可能な発現および制御配列に作動可能に連結することができる。このように、適切な状態下で、標的細胞における核酸の発現を達成することが可能である。
核酸の発現に有用なウイルスベクター系には、例えば、天然に存在するまたは組換え型のウイルスベクター系が含まれる。特定の用途に応じて、適切なウイルス性のベクターには、自律複製可能な(replication competent)、自律複製不能な(replication deficient)、および条件つきで複製可能な(conditionally replicating)ウイルスベクターが含まれる。例えば、ウイルスベクターは、ヒトまたはウシアデノウイルス、ワクシニアウイルス、ヘルペスウイルス、アデノ随伴ウイルス、マウスのマイニュートウイルス(MVM)、HIV、シンドビスウイルス、および(それだけには限らないがラウス肉腫ウイルスを含めて)レトロウイルス、およびMoMLVのゲノムから得ることができる。一般的に、当該の遺伝子は、一般的に、ウイルスDNAに付随する遺伝子構築物のパッケージング、それに続く感受性宿主細胞の感染、および当該の遺伝子の発現を可能にするために、こうしたベクターに挿入される。
同様に、本発明の核酸を含む遺伝子構築物をパッケージングするために使用されるウイルスのエンベロープは、特定の細胞への受容体依存性エンドサイトーシスを可能にするために、受容体に対して特異的な受容体リガンドまたは抗体の添加によって改変することができる(例えば、WO 93/20221、WO 93/14188、およびWO 94/06923を参照のこと)。本発明のある実施形態では、本発明のDNA構築物は、エンドサイトーシスを容易にするために、アデノウイルス粒子などのウイルスタンパク質に連結される(Curielら、Proc.Natl.Acad.Sci.U.S.A.88:8850−8854(1991))。他の実施形態では、本発明の分子結合体として、微小管インヒビター(WO/9406922)、インフルエンザウイルス血球凝集素を模倣する合成ペプチド(Plankら、J.Biol.Chem.269:12918−12924(1994))、およびSV40 T抗原などの核局在化シグナル(WO93/19768)を挙げることができる。
医薬品目的で使用される場合、遺伝子治療のために使用されるベクターは、適切な緩衝液中に調製される。これは、リン酸緩衝生理食塩水またはリン酸ナトリウム/硫酸ナトリウム、トリス緩衝液、グリシン緩衝液、滅菌水、およびGoodらBiochemistry 5:467(1966)によって記載されているものなどの当業者に知られている他の緩衝液などの、任意の薬学的に許容される緩衝液であり得る。
本発明の処方物は、当業者に知られている任意の送達方法を使用して、任意の組織または器官に送達することができる。本発明のある実施形態では、本発明の核酸は、粘膜、局所用、および/または口腔内処方物、特に、粘膜付着性ゲルおよび局所用ゲル処方物で調製される。経皮送達のための例示的な浸透増強組成物、ポリマーマトリックス、および粘膜付着性ゲル調製物は、米国特許第5,346,701に開示されている。
本発明の遺伝子治療処方物は一般的に、細胞に対して投与される。細胞は、上皮性膜などの組織の一部として、あるいは組織培養におけるものなどの、単離された細胞として提供することができる。細胞は、in vivoで、ex vivoで、またはin vitroで提供することができる。
本発明はまた、気分障害(大うつ病または双極性障害など)、精神病性障害(精神分裂症など)、またはこうした障害の症状の少なくともいくつかの素因を診断する方法を提供する。診断は、患者における本発明のポリペプチドまたはポリヌクレオチドのレベルを決定すること、次いで、そのレベルをベースラインまたは範囲と比較することを含む。一般的に、ベースライン値は、気分障害または精神病性障害を患っていない健康な人における、あるいは薬物投与または他剤に対する影響下での、本発明のポリペプチドまたはポリヌクレオチドを表すものである。本発明のポリペプチドまたはポリヌクレオチドのレベルの、ベースライン範囲からの(上または下への)変動は、患者が、気分障害または精神病性障害を患う、あるいは気分障害または精神病性障害の少なくともある種の様相を呈するリスクがあることを示唆する。ある実施形態では、本発明のポリペプチドまたはポリヌクレオチドのレベルは、患者から血液、尿、または組織サンプルを取り出し、本明細書に論じられるものなどの多くの検出方法を使用して、サンプル中の本発明のポリペプチドまたはポリヌクレオチドの量を測定することによって測定される。
遺伝子発現の欠如が、障害に関連する場合には、遺伝子のゲノム構造は、罹病性関連の欠失または挿入突然変異を検出するために、PCRなどの既知の方法を用いて評価することができる。逆に、特定の遺伝子に対応するmRNAまたはタンパク質の存在は、個人が、遺伝子発現の欠如を伴う遺伝子突然変異またはそれに付随する障害を有さないことを示唆するだろう。したがって、mRNAまたはタンパク質の有無を検出することによって、または遺伝子のゲノム構造を調べることによって診断を行うことができる。
以前の研究では、遺伝子発現を分析するために、マイクロアレイおよびPCRを使用して、気分障害および自殺の傾向に関連する遺伝子を調査していた(Sibilleら、2004;Yanagi Mら、J Hum Genet.,50(4):210−6(2005))。しかし、どちらの調査も、自殺に最も典型的である可能性がある遺伝子を検出する、気分障害と比較した場合の自殺、および対照と比較した場合の自殺の厳密な分析を使用していなかった。この実施例は、自殺をしたBPDおよびMDD患者由来の死後脳の扁桃体、前側帯、および小脳におけるマイクロアレイ遺伝子発現プロフィールを、白質、乏突起膠細胞、ミエリン、および他の経路を調節する分子のmRNA発現レベルに焦点を合わせて記述する。ここで同定される遺伝子は、自殺の挙動を検出および治療するためのバイオマーカーとして使用することができる。
この実施例は、ヒト以外の霊長類(健康なアカゲザル)における特定の遺伝子が、気分安定薬、リチウム(Li)、すなわちBPの治療のための最適な薬物を用いる治療に応答して、差次的に発現されることを実証する。アカゲザルの前部帯状皮質(AnCg)、背外側前頭前野(DLPFC)、海馬(HC)、および扁桃体(AMY)に対して、本明細書に記載されている遺伝子発現検出方法を使用して、遺伝子発現プロファイリングを実施し、上述のヒトの剖検結果と比較した。表2Aは、文献中にあらかじめ特定されていたリチウム応答性遺伝子と、本発明の調査によって確認されたリチウム応答性遺伝子を示す。表2Bは、霊長類におけるリチウム応答性遺伝子として新たに特定された遺伝子(これはまた、双極性障害を患うヒトの対象において異常調節される)を示す。
FGF受容体2(FGFR2)転写産物は、うつ状態の対象のいくつかの脳領域において低下することが、一貫して発見されている(例えば、2004年8月5日に米国特許公開番号20040152111−A1として公開された、米国2003年11月3日に出願の特許出願第10/701,263号を参照のこと)。ヒトのFGFR2遺伝子は、19のエキソンを含有し、13ものスプライスバリアントを産生する。こうしたバリアントは、3つの主要な機能的クラスに分類される:第1は、可溶性の受容体であると考えられる膜貫通ドメインおよびチロシンキナーゼドメインを欠くバリアント;第2は、エキソン9によってコードされるIg IIIc型ドメインを含有するバリアント;第3は、エキソン8によってコードされるIg IIIb型ドメインを含有するバリアント。Ig III型ドメインは、リガンド特異性を与えるので、これら後者の2バリアントは、IIIcまたはIIIbドメインのその使用に基づく異なる薬理学的プロファイルを有する。この実施例は、ヒトの皮質(背外側前頭前野および前側帯)および海馬から得られる全RNA中に存在するエキソンのPCRに基づく測定を記載する。
A.FGF2のマイクロインジェクション
この一連の実験は、対象動物における抑うつを評価するための、強制水泳試験(FST)および高架式十字迷路(EPM)を使用する、FGF2のマイクロインジェクション後の有意な影響を示す。FSTでは、FGF2注射された動物(n=12)は、対照(n=13)よりも多くの水泳(t[23]=2.20、p<0.05)および少ない不動(t[23]=2.88、p<0.0l)を示した。これは、FGF2動物では、より抑うつ様でない挙動を示すこととなる。しかし、EPMでは、FGF2注射された動物は、クローズドアームで過ごす時間が有意に多く(t[13]=3.18、p<0.01)、オープンアームで過ごす時間は有意に少なかった(t[13]=2.46、p<0.05)。これらの結果(図2)は、FGF2の急性注射の後、不安様挙動が増大されることを示す。
この一連の実験は、NCAM投与の後の強制水泳試験における動物の気分に対する有意な影響を示す(アミノ酸配列=EVYVVAENQQGKSKA;図3を参照のこと)。ここでは、NCAM注射された動物(n=13)は、対照(n=13)よりも少ない不動(t[24]=2.13、p<0.05)を示した。また、これは、より抑うつ様でない挙動の指標である。これはまた、FGF2データと同じ傾向であり、FGF2およびNCAMがFGF受容体と相互作用するという事実と整合性がある。同様に、NCAM注射された動物は、クローズドアームで過ごす時間が多く(有意に長い時間ではないが)、オープンアームで過ごす時間が短く、不安様挙動の増大と整合性があった。NCAM注射された動物はまた、センターの4分の1で過ごす時間が有意に短かった(t[18]=2.40、p<0.05)。
この一連の実験は、強制水泳試験および高架式十字迷路試験を使用する、FGF系ペプチドインヒビター(アミノ酸配列=HFKDPKRLY)を用いる注射の後の、動物の気分に対する有意な影響を示す。結果を図4に示す。FSTでは、インヒビターを注射された動物(n=7)は、対照(n=7)よりも、有意に少ないクライミング(t[12]=2.06、p<0.05)、有意に少ない水泳(t[12]=1.92、p<0.05)、および有意に多い不動(t[12]=3.58、p<0.005)を示した。これらの結果は、FGF系の抑制が、抑うつ様挙動の増大をもたらす可能性があることを示す。これらの結果は、以前のデータセットの結果を確認し、進歩させるものであり、大うつ病を伴う個人の死後組織の研究と整合性があった。インヒビターを注射された動物はまた、EPMのセンターの4分の1で過ごす時間が有意に長かった(T[7,7]=35.0、p=0.03)。同じ動物は、オープンアームで過ごす時間が短く、不安様挙動の増大の指標となるクローズドアームで過ごす時間は等しかったが、過ごす時間の長さに有意な差はなかった)。しかし、これらの知見は、上述のマイクロインジェクション研究から引き出される結論と整合性がある。
方法。スプラーグ‐ドーリーラット(Sprague Dawley rat)に、出生の翌日、賦形剤またはFGF2(20ng/g、皮下)を注射し、成体としてのモリス水迷路試験を行った後、屠殺した。本発明者らは、歯状回のレーザー捕獲顕微手術、それに続くマイクロアレイ分析を用いる、候補遺伝子探索法(candidate approach)と遺伝子発見アプローチの両方を使用して、遺伝子発現の変化を評価した。
不安障害は、大うつ病(MD)を含めた他の神経精神病学的障害との同時罹患率が高い。この実施例は、長期にわたるFGF2投与が、ラットにおける抗不安作用を有することを示す。ラットは、様々な運動試験および行動試験におけるその挙動に基づいて「高い不安」(LR)または低い不安」(HR)群に配置した。どちらの群の動物にも、3週間、48時間ごとにFGF2(5ng/g)または賦形剤を、腹腔内注射によって投与した。このFGF2注射の1日後、すべての動物を、高架式十字迷路(EPM)および明暗(LD)不安試験で試験した。
成体のCNSでは、線維芽細胞成長因子受容体2(FGFR2)と線維芽細胞成長因子受容体3(FGFR3)は、差次的に分布している。これらの受容体のmRNAは、Ig様ドメインIIIのカルボキシル末端をコードするエキソンにおいて、選択的スプライシングを受ける。この相互排他的なmRNAスプライシングによって、有意に異なるリガンド結合プロフィールをもつ、FGFR2およびFGFR3という2つのアイソフォームが産生される:片方のアイソフォームは、エキソンIIIb(FGFR2b/FGFR3b)を発現し、もう片方のアイソフォームは、エキソンIIIc(FGFR2c/FGFR3c)を発現する。エキソン選択は、発生中に、厳密に組織依存性であり、エキソンIIIbは上皮性系統に発現され、エキソンIIIcは、間葉性系統に発現される。しかし、細胞周期依存性のIIIbからIIIcへのスイッチは、FGF1およびFGF2の外因的な添加によって、in vitroで誘導される。この実施例は、長期間にわたるストレスが、FGFR2のエキソンIIIc:IIIbスプライスバリアント発現割合の低下を誘導することを示す。
青斑核(LC)および背側縫線(DR)は、それぞれ、脳のノルアドレナリン作動性およびセロトニン作動性神経支配の主な源である。これらの神経伝達物質の調節異常は、精神障害に関係している。この実施例は、その脳のLCおよびDR領域中の遺伝子発現プロフィールを比較するために、MDD(N=12)、BPD(N=6)を患う患者、および健康な対象(N=9)の死後脳を使用する。すべての対象は、脳pH>6.6および苦痛ファクターゼロという包含基準を満たしていた。レーザー捕獲顕微解剖されたLCおよびDRサンプルから得られる全RNAサンプルを抽出し、増幅し、Affymetrix高密度オリゴヌクレオチド・マイクロアレイを用いてプローブで探索した。遺伝子発現データは、確固たるマルチチップ平均アルゴリズム(p≦0.1)のANOVAによって、また、MAS5.0「現在の(present)」コール(call)アルゴリズム(3種の健康状態のうちの1つにおける50%存在の最小値)によって分析した。これらの基準を満たしている遺伝子を、Ingenuity Pathway Analysis(IPA)を使用して分析した。健康な個人と比較すると、774および636遺伝子は、LCでは発現変化を示し、627および656遺伝子は、それぞれ、MDDおよびBPD患者のDRで発現変化を示す。
Claims (49)
- 対象における気分障害の診断を容易にするための方法であって、
(i)図5、図6、図7、図8、表1、表2B、表3、および表4に列挙される遺伝子からなる群から選択される遺伝子の発現レベルを測定するステップと、
(ii)前記遺伝子が対照に対して異常調節されるか否かを決定する(ここでは、前記遺伝子の調節異常は、前記対象が気分障害を患う可能性の増大を示唆する)ステップと、
(iii)前記増大された可能性に関する任意の発見を記録または報告するステップ
を含む方法。 - 前記気分障害が双極性障害であり、前記遺伝子が、図5、図7、図8、表2B、および表4に列挙される遺伝子からなる群から選択される請求項1に記載の方法。
- 前記気分障害が大うつ病障害であり、前記遺伝子が、図6、図8、表1、および表3に列挙される遺伝子からなる群から選択される請求項1に記載の方法。
- 前記遺伝子調節異常が、前記対象の脳組織内に生ずる請求項2または3に記載の方法。
- 前記脳組織が、青斑核、背側縫線、前部帯状皮質、背外側前頭前野、海馬、および扁桃体からなる群から選択される請求項4に記載の方法。
- 前記遺伝子発現が、前記遺伝子から転写されるメッセンジャーRNAを検出することによって分析される請求項2または3に記載の方法。
- 前記遺伝子発現が、前記遺伝子のタンパク質産物を選択的に検出することによって分析される請求項2または3に記載の方法。
- 前記遺伝子から転写されるメッセンジャーRNAを検出することが、
(i)前記mRNAを、前記メッセンジャーRNAと選択的に結びつく試薬と接触させることと、
(ii)前記mRNAと選択的に結びつく前記試薬のレベルを検出すること
を含む請求項6に記載の方法。 - 前記遺伝子の発現レベルが、気分障害を患わない人間に関するレベルよりも高い請求項1に記載の方法。
- 前記遺伝子の発現レベルが、気分障害を患わない人間に関するレベルよりも低い請求項1に記載の方法。
- 前記遺伝子の発現レベルが、マイクロアレイ分析を使用して検出され、かつ前記遺伝子が、マイクロアレイ上の少なくとも2つの遺伝子のうちの1つである請求項1に記載の方法。
- 前記遺伝子が表1から選択され、前記気分障害が自殺性である請求項1に記載の方法。
- 前記対象が、自殺行動の可能性の増大を伴う気分障害とあらかじめ診断されている請求項12に記載の方法。
- 前記対象が、大うつ病、双極性障害、および精神分裂症からなる群から選択される気分障害とあらかじめ診断されている請求項12に記載の方法。
- 前記対象による自殺企図の可能性を低下させる前記対象のための治療を指示することをさらに含む請求項12に記載の方法。
- 前記対象が、双極性障害および大うつ病性障害の症状を有し、前記遺伝子が、双極性の対象において、大うつ病障害の対象とは違うように発現され、それによって、前記対象における双極性障害または大うつ病性障害の診断が容易になる請求項1に記載の方法。
- 前記遺伝子が、薬物乱用者でないMDD対象に対して、薬物を乱用するMDD対象において異常調節され、かつ前記遺伝子が、表1Cに列挙される異常調節された遺伝子から選択され、それによって、薬物乱用と結びつけられるMDDに対するMDDの診断が容易になる請求項1に記載の方法。
- 気分障害の治療または予防のための化合物を特定する方法であって、
(i)図5、図6、図7、図8、表1、表2B、表3、および表4に列挙される異常調節された遺伝子の群から選択される異常調節された遺伝子に相当するポリペプチドと化合物を接触させるステップと、
(ii)前記ポリペプチドに対する前記化合物の機能効果を決定し、それによって、気分障害の治療または予防のための化合物を特定するステップ
を含む方法。 - 前記接触ステップが、in vitroで実施される請求項18に記載の方法。
- 前記ポリペプチドが細胞中に発現され、前記細胞が前記化合物と接触される請求項18に記載の方法。
- 前記気分障害が、双極性障害、大うつ病障害、自殺性、および薬物乱用合併症からなる群から選択される請求項18に記載の方法。

を含む方法。 - グルタミン酸作動性不均衡を患う対象を治療するための方法であって、グリア輸送体、グルタミンシンテターゼ、AMPA、カイニン酸、GRM1、およびGRM7からなる群から選択される分子を標的にする十分な量の化合物を対象に投与することを含む方法。

(i)表3中に列挙された遺伝子から選択される異常調節された遺伝子の発現レベルを測定することと、
(ii)前記測定を、前記対象におけるBPまたはMDDの増大されたリスクと相関させることを含む方法。 - BPまたはMDDを発症する前記リスクを記録または報告することをさらに含む請求項38に記載の方法。

- げっ歯類において記憶力を向上させるための方法であって、前記動物の出生の48時間以内に前記動物にFGF2を投与することを含む方法。

- 前記FGF2が20ng/g(体重)で投与される請求項45に記載の方法。
- 請求項45、46、または47に記載の方法に従って治療されたげっ歯類。
- 成体のげっ歯類であり、未治療のげっ歯類に対して記憶力が向上され、前記げっ歯類におけるNCAMの発現は、未治療のげっ歯類に対して低下し、GAP−43、Rgs4、trkB、CCK、SST、およびVgfからなる群から選択される少なくとも1つの遺伝子の発現が、未治療のげっ歯類に対して増大された請求項48に記載のげっ歯類。
