Description
i)上述の融合タンパク質を過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞内での融合タンパク質の局在化を経時的に測定するステップと
を含み、細胞内での融合タンパク質の局在化の変化が活性化の指標となる方法を提供する。
i)上述の融合ペプチドを過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞内での上記ペプチドの局在化を測定するステップと、
iii)上記細胞を上記作用因子で処理して、細胞内での上記ペプチドの局在化を測定するステップと

本明細書で用いる「タンパク質」、「ポリペプチド」及び「ペプチド」という用語は、鎖長にはかかわりなく、アミノ酸モノマー(残基)の天然又は合成ポリマーを表すものとして互換的に用いられ、ここでいうアミノ酸モノマーには、この場合、天然アミノ酸、天然アミノ酸の構造変異体、ペプチド結合に関与できる合成非天然類似体が包含される。
図1Aは、マイトジェン活性化プロテインキナーゼ14(MAPK14)のDNA配列を示す。
p38αMAPK14変異体2に対応する遺伝子は、Mammalian Gene Collection(クローン5181064; GenBank BC031574、配列番号1、図1)から入手した。配列番号2は、配列番号1でコードされたタンパク質の配列を示す。PCRプライマーは、MAPK14遺伝子全体が増幅され、その生成物をプラスミドベクターpCORON1002−EGFP−N1及びpCORON1002−EGFP−C1(GE Healthcare社(英国アマーシャム)、図2a及び2b)或いはアデノウイルスベクターpDC515−UBC−Emerald−N1及びpDC515−UBC−Emerald−C1(Microbix Biosystems Inc.、Toronto、カナダ、図3a及び3b)のN末端又はC末端融合タンパク質としてサブクローニングできるように設計した。MAPK14断片の5′末端でのNheI制限酵素部位の導入及び3′末端でのXhoI制限酵素部位の導入によって、ベクターのNheI及びSalI制限部位のサブクローニングが可能となった。pCORON1002ベクターは安定細胞株の生産ができるように細菌アンピシリン耐性遺伝子と哺乳類ネオマイシン耐性遺伝子を含んでおり、融合タンパク質はヒトユビキチンCプロモーターによって発現される。ヒトユビキチンCプロモーターを選択したのは、宿主細胞系の動揺を最小限に抑制して安定で頑強な細胞株及びアッセイを得るのに望まれる均質で一定レベルの融合タンパク質発現をもたらすためである。
ヒト軟骨肉腫細胞株SW1353(ATCC)を、FuGENE6トランスフェクション剤(Roche社(英国))を用いて、プラスミドベクターpCORON1002EGFP−C1−MAPK14及びpCORON1002EGFP−N1−MAPK14でトランスフェクションした。細胞は、10%(v/v)FBS、1%(v/v)ペニシリン−ストレプトマイシン、1%(v/v)グルタミン添加RPMI1640培地(Sigma−Aldrich社(英国))で維持した。組換え融合タンパク質を発現する安定クローンを、ジェネテシン(500μg/ml)での選択によって、4週間かけて選別した。継代数の少ない段階での一次クローンの特性決定には、均質性及び発現レベルを測定するとともに親細胞株との形態的関連性及び生物学的反応の生理学的関連性を比較するためフローサイトメトリー(FACSCalibur、BD Biosciences社製)を用いた。これらの特性をさらに継代を重ねた細胞についても分析し、細胞株の経時的安定性を評価した。適宜二次クローンを単離して同様に分析した。EGFP−MAPK14融合タンパク質の安定な発現を呈し、望ましい規準を満たす細胞株を、200μg/mlのジェネテシンを含有する増殖培地で維持した。選択したクローンでは、30代目まで融合タンパク質が安定に発現されることが実証された。
EGFP−MAPK14融合タンパク質集団は主に休止細胞の細胞質に存在するか、或いは休止細胞全体に均一に分布している。しかし、浸透圧ショック、タンパク質翻訳阻害又はサイトカイン処理のような外部ストレスによって細胞が刺激されると、標識MAPK14集団の一部は核に再局在化する(図12)。
EGFP−C1−MAPK14を安定に発現するSW1353生細胞を、0.4Mソルビトール又は300nMアニソマイシンで15分間処理し、その応答を、同じ構築物を発現する未処理の対照細胞と対比した。
IN Cell Analyzer 3000のデータから得た生細胞の経時的画像の分析(図14)から、35分間のアニソマイシン(300nM)処理に応答して、SW1353細胞においてEGFP−C1−MAPK14融合タンパク質の核から細胞質への顕著な再配置(N:C比)が認められる。この実験では、ヘキスト33342を使用したが、これによって対照細胞でのベースラインストレス応答が起きた。
12pMのIL−1βで処理した生細胞では、細胞質から核へのEGFP−MAPK14シグナルの明瞭な転位置が認められ(図15A及び15B)、20〜40分で核におけるシグナルの蓄積が最大となった(図16)。炎症反応で予想されるように、40〜90分の間にシグナルのゆっくりとした平衡化が起こった(図16)。
SEGFP−C1−MAPK14を安定に発現するW1353細胞に、IL−1βを0.017〜333pMの最終濃度で加えた。単一クローン由来の安定細胞集団の8代目及び15代目の細胞の用量反応曲線から、この細胞株の応答が、経時的に安定していることが分かる(図17)。EC50値はそれぞれ2.5及びpMであった(N=8、S:Nはそれぞれ6.5及び4)。
安定な発現を示すSW1353細胞におけるGFP−MAPK14融合タンパク質の転位置に関するアッセイの再現性について検討した。アッセイあ3枚の別個の96ウェルプレート上で実施した。各プレートのウェルに、対照培地で処理した細胞(n=48)とIL−1β(12pM)で処理した細胞を交互に入れた。全体のシグナル/ノイズ比は3.61:1(n=144)であった。
処理細胞及び未処理細胞での核/細胞質比の平均の標準偏差に寄与するばらつきを分離できれば、p38MAPKシグナル伝達を活性化/阻害する化合物のスクリーニングのための改良アッセイの作成に役立つはずである。
EGFP−MAPK14アッセイの経時的及び生物学的応答を、p38(MAPK14)活性化の広く認められたアッセイであるリン酸化型(Thr180/Tyr182)−p38(MAPK14)に対する免疫蛍光アッセイとの同時分析によって評価した。EGFP−MAPK14融合タンパク質を安定に発現するSW1353細胞を、IL−1βで処理し、固定化し、上述の通りヘキスト染色した。次いで、細胞を洗浄し(PBS中1%ヤギ血清、0.1%Tween)、室温で15分間透過処理し(洗浄緩衝液中0.5%Triton X)、再度洗浄した。50μlのウサギ抗リン酸化型p38抗体(1:200)(Zymed Laboratories社(米国サンフランシスコ))を添加して室温で1時間インキュベートしてから、2回洗浄し、50μlのヤギ抗ウサギAlexa 647抗体(Molecular Probes社製)を1:200で加え、室温で1時間インキュベートした。2回洗浄した後、IN Cell Analyzer 1000装置でPBS中の細胞の画像を取得して、ヘキスト、EGFP−MAPK14及びAlexa 647を検出した。
EGFP−MAPK14融合タンパク質を安定に発現する細胞を、(100μl維持培地に懸濁して)Packard社製ブラック96ウェルViewPlatesにウェル当たり0.8×104の細胞数で播種した。次いで、培地中の阻害剤2μlを加えて細胞を30分間予めインキュベートしておいてから、培地中で予め調製しておいた試験活性化因子及び対照25μlを加えてさらに30分間インキュベートした。細胞を上述の通り、固定化、画像化し、分析した。

- 請求項13乃至請求項18のいずれか1項記載のヌクレオチド配列で形質転換された宿主細胞。
- 請求項13乃至請求項18のいずれか1項記載のヌクレオチド配列で安定に形質転換された請求項23記載の宿主細胞。
- 植物、昆虫、線虫、鳥類、魚類及び哺乳類細胞から選択される、請求項23又は請求項24記載の宿主細胞。
- 前記哺乳類細胞がヒト細胞である、請求項25記載の宿主細胞。
- 前記ヒト細胞がヒト軟骨肉腫細胞株SW1353である、請求項26記載の宿主細胞。
- 請求項1乃至請求項12のいずれか1項記載の融合タンパク質を発現することのできる、請求項23乃至請求項27のいずれか1項記載の宿主細胞。
- 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化を検出する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合タンパク質を過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞内での融合タンパク質の局在化を経時的に測定するステップと
を含み、細胞内での融合タンパク質の局在化の変化が活性化の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合ペプチドを過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞内での上記ペプチドの局在化を測定するステップと、
iii)上記細胞を上記作用因子で処理して、細胞内での上記ペプチドの局在化を測定するステップと
を含み、上記作用因子で処理していない対照細胞と比較したときの細胞内での上記ペプチドの局在化の差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して上記作用因子の有する効果の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合タンパク質を過剰発現するように形質転換した第一及び第二の細胞を培養するステップと、
ii)第一の細胞を上記作用因子で処理して、第一の細胞内での上記タンパク質の局在化を測定するステップと、
iii)上記作用因子で処理していない第二の細胞内での上記タンパク質の局在化を測定するステップと
を含み、第一の細胞と第二の細胞内での上記タンパク質の局在化の差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して上記作用因子の有する効果の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合タンパク質を過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞を上記作用因子で処理して、細胞内での上記タンパク質の局在化を測定するステップと、
iii)上記作用因子の存在下での上記タンパク質の局在化を、上記作用因子の非存在下でのタンパク質の局在化についての既知の値と比較するステップと
を含み、上記作用因子の存在下でのタンパク質の局在化と上記作用因子の非存在下での既知の値との差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化に際して上記作用因子の有する効果の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合ペプチドを過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞を上記作用因子で処理するステップと、
iii)上記細胞をp38マイトジェン活性化プロテインキナーゼ(MAPK)の既知の活性化因子で処理して、細胞内での上記ペプチドの局在化を測定するステップと
を含み、上記作用因子で処理せずに上記既知の活性化因子で処理した対照細胞と比較したときの細胞内での上記ペプチドの局在化の差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して上記作用因子の有する効果の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合タンパク質を過剰発現するように形質転換した第一及び第二の細胞を培養するステップと、
ii) 第一の細胞を上記作用因子で処理するステップと、
iii)第一の細胞と第二の細胞を、p38マイトジェン活性化プロテインキナーゼ(MAPK)の既知の活性化因子で処理して、第一の細胞と第二の細胞内での上記タンパク質の局在化を測定するステップと
を含み、第一の細胞と第二の細胞内での上記タンパク質の局在化の差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して上記作用因子の有する効果の指標となる、方法。 - 生細胞中でのp38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して作用因子の有する効果を測定する方法であって、
i)請求項1乃至請求項12のいずれか1項記載の融合タンパク質を過剰発現するように形質転換した細胞を培養するステップと、
ii)上記細胞を上記作用因子で処理するステップと、
iii)上記細胞をp38マイトジェン活性化プロテインキナーゼ(MAPK)の既知の活性化因子で処理して、細胞内での上記ペプチドの局在化を測定するステップと、
iv)上記作用因子及び活性化因子の存在下でのタンパク質の局在化を、活性化因子は存在するが上記作用因子の非存在下でのタンパク質の局在化についての既知の値と比較するステップと
を含み、上記作用因子の存在下でのタンパク質の局在化と上記作用因子の非存在下での既知の値との差が、p38マイトジェン活性化プロテインキナーゼ(MAPK)の活性化の調節に際して上記作用因子の有する効果の指標となる、方法。 - 前記既知の値がデータベースに保存されている、請求項23又は請求項35記載の方法。
- 前記融合タンパク質の局在化をその発光、蛍光及び放射能特性によって測定する、請求項29乃至請求項36のいずれか1項記載の方法。
- 前記作用因子が、p38マイトジェン活性化プロテインキナーゼの活性化を誘導する、請求項30乃至請求項37のいずれか1項記載の方法。
- 前記作用因子が、p38マイトジェン活性化プロテインキナーゼの活性化を阻害する、請求項30乃至請求項37のいずれか1項記載の方法。
- 前記作用因子が化学物質又は物理的実体である、請求項30乃至請求項39のいずれか1項記載の方法。
- 前記化学物質が薬剤候補物質である、請求項40記載の方法。
- 前記薬剤候補物質が炎症誘発性又は抗炎症性化合物である、請求項41記載の方法。
