(技術分野)
本願は、がんの治療において細胞障害性Tリンパ球(CTL)反応を誘発する能力がある抗原のデリバリーのためのビヒクルとしてのリポソームおよび連続的疎水相を含む組成物の使用に関する。
1つの実施態様において、本発明は、疎水性物質の連続的相を含む担体;リポソーム;および細胞障害性Tリンパ球(CTL)反応を誘発する能力がある少なくとも1つの抗原を含む組成物を提供する。また、組成物が、少なくとも1つのヘルパーTエピトープを含むのが好ましい。
本発明の実施態様を説明する図面は、例示のためにすぎない。
本願は、少なくとも1つのヘルパーTエピトープと共にCD8+ 細胞障害性Tリンパ球(CTL)反応を誘発する能力がある少なくとも1つの抗原および疎水性物質の連続的相を含む担体に懸濁したリポソームを含む組成物を提供する。さらに、本発明は、患者においてがんを治療するための方法における該組成物の使用を教示する。
治療される患者は、いずれかの脊椎動物であり、哺乳動物が好ましく、ヒトがより好ましい。
組成物の適当な抗原は、患者において細胞性(CTL)免疫反応を誘発する能力がある抗原である。
Tヘルパーエピトープは、Tヘルパー活性を有するアミノ酸(天然または非天然アミノ酸)の配列である。Tヘルパーエピトープは、免疫系の能力を確立し、最大化することにおいて重要な役割を演じ、細胞障害性Tリンパ球などの他の免疫細胞の活性化および方向付けに関与するTヘルパーリンパ球によって認識される。
組成物の担体は、疎水性物質、好ましくは液体疎水性物質の連続的相を含む。連続的相は、本質的に純粋な疎水性物質または疎水性物質の混合物である。さらに、担体は、疎水性物質が連続的相であるという条件で、疎水性物質中水型のエマルジョンまたは疎水性物質の混合物中水型のエマルジョンであってもよい。さらに、もう1つの実施態様において、担体は、アジュバントとして機能しても良い。
リポソームは、封入された水性ボリュームを含む完全に閉鎖した液体二重層膜である。リポソームは、単層小胞(単一の二重層膜を有する)または各二重層が水性層によって次の層から分離されてもされなくてもよい多膜二重層を特徴とする多重層小胞であってもよい。リポソームの一般的議論は、Gregoriadis G.Immunol.Today、11:89−97、1990;およびFrezard、F.、Braz.J.Med.Bio.Res.、32:181−189、1999に見出すことができる。
組成物は、当業界で公知のように、1つ以上の医薬的に許容しうるアジュバント、賦形剤などをさらに含んでもよい。たとえば、Remington's Pharmaceutical Sciences (Remington's Pharmaceutical Sciences、Mack Publishing Company、Easton、Pa.、USA 1985)およびThe United States Pharmacopoeia:The National Formulary(USP 24 NF19)、1999刊行を参照のこと。
1つの実施態様において、適当なアジュバントとして、CpG−含有オリゴデオキシヌクレオチド(CpG ODN)が挙げられる。たとえば、5'−TCCATGACGTTCCTGACGTT−3'など。当業者は、標的種および効力に基づいて、適切なCpGを選択することができる。CpGの代わりに、Pam3Cys−SKKK)(EMC Microcollections、Germany)またはその変異体、同族体および類縁体などのリポペプチドを用いてもよい。この点において、リポペプチドのPam2ファミリーが、リポペプチドのPam3ファミリーの代わりに有効であることが分かっている。
1つの実施態様において、本明細書に記載の組成物は、抗原(遊離抗体および/またはリンパ球上の抗原結合受容体と特異的に相互作用する物質として定義される)または抗原/アジュバント複合体をリポソーム内に封入して、リポソーム−封入抗原を形成し、疎水性物質の連続的相を含む担体をリポソーム−封入抗原と混合することによって製剤することができる。抗原/アジュバント複合体を第1段階で用いないならば、適当なアジュバントをリポソーム−封入抗原に、リポソーム−封入抗原および担体の混合物に、またはリポソーム−封入抗原と混合する前に担体に加えてもよい。プロセスの順番は、使用するアジュバントのタイプに応じて変わる。次いで、得られるリポソーム−封入抗原を担体と混合する(用語「リポソーム−封入抗原」は、状況に応じて、抗原単独のリポソーム封入または抗原/アジュバント複合体の封入を意味することに留意すべきである)。これは、アジュバントと抗原との間の密接な接触を促進し、少なくとも部分的に、良好な免疫反応の原因となりうる。いくつかのアジュバントの使用を促進するために、抗原を最初にリポソーム内に封入し、次いで、疎水性物質の連続的相を含む担体中で、得られるリポソーム−封入抗原をアジュバントと混合してもよい。
以下の非限定的実施例によって、本発明をさらに説明する。
細胞反応
(a)活性化
CTL反応の特異性および迅速性を試験するために、ユニバーサルTヘルパーエピトープであるPADRE(AKXVAAWTLKAAA−OH(配列番号:10);50 μg/投与)に融合し、PBS/FIA(リン酸緩衝食塩水フロイント不完全アジュバント)エマルジョン(100 μl/投与)に懸濁したリポソーム(0.2 g レシチンおよび0.02 g コレステロール/投与)内に封入した、CpG ODN アジュバント、ヒトパピローマウイルス(HPV)16、すなわち、R9F(E7(H2−Db)ペプチド RAHYNIVTF、アミノ酸49−57;配列番号:1)のCTLエピトープを含む組成物で、マウスを1回処置した。処置後14日目に、脾細胞(エフェクター細胞)をR9Fまたは無関係ペプチド(KIMCNSSCM;配列番号:13)と共に6時間共培養した。脾細胞のエクスビボ細胞内IFN−γ染色は、脾細胞を無関係ペプチドに曝露する場合(脾細胞の0.12 %;またはペプチド無し;図1)よりも、脾細胞をR9Fペプチドに曝露する場合に、IFN−γ陽性CD8+ T細胞(CTLs)の割合が13倍多いこと(脾細胞の1.6 %)を実証した。図1にて実証されるように、上述のR9Fペプチドを含む組成物でのマウスの処置は、無関係ペプチドを含む組成物で処置したマウスと比べて、HPVエピトープによる刺激に対する特異的反応を示すCTLの有意な増殖を引き起こした。
特定の本発明の具体例での単回処置によって誘発された記憶応答の持続期間を、以下の組成物で処置後130日目のマウスから得られた脾細胞によるEL−4細胞の溶解によって実証した(図3):(i)PBS/FIA油中水型エマルジョンに包含されたリポソーム内に封入した、融合ペプチド(PADREと融合したR9Fペプチド)およびCpG ODN(菱形);(ii)PBS/FIA油中水型エマルジョン中の封入しない融合ペプチドおよびCpG ODN(三角および十字)または(iii)PBS/FIA油中水型エマルジョン中の、CpG ODNアジュバント無しでリポソーム内に封入した融合ペプチド(黒丸)。リポソーム−封入融合ペプチド、リポソーム−封入CpG ODN、非封入−融合ペプチドおよびCpG ODN、ならびに非封入−融合ペプチド単独は、コントロールとして働いた。
子宮頸がんの根絶
たとえば、ガルダシルTMおよびセルバリクスTMなどのヒトパピローマウイルス(HPV)誘発性子宮頸がんおよび外陰がんのための予防ワクチンならびに子宮頸がんおよび外陰がんの治療上の処置の開発は、依然として優先度が高い。この実施例においては、ヒトパピローマウイルス(HPV)16のCTLエピトープ、すなわち、R9F(E7(H2−Db)ペプチド RAHYNIVTF、アミノ酸49−57;配列番号:1)を含む治療組成物を用いてCTLを誘発した。これらのCTLは、その分化および増殖ならびに機能性記憶CTLへの成熟のために、CD4+ T細胞の補助を必要とする。CD4+ T細胞の補助を介する潜在的CTL反応を達成するために、R9FペプチドをユニバーサルTヘルパーエピトープ、PADRE(配列番号:10)に融合して、融合ペプチドを得た。融合ペプチドを、CpG ODNモチーフまたはリポペプチド(Pam3Cys−SKKKK)を含む合成オリゴヌクレオチドと共にリポソーム内に封入した。治療用組成物は、単回処置において治療用製剤をデリバリーするためにPBS/FIA油中水型エマルジョンを用いた。C3腫瘍細胞を発現しているHPV 16を用いて、C57BL/6マウス(マウス10匹/グループ)をチャレンジさせ、次いで、チャレンジ後14日目に上記治療組成物またはコントロール組成物でマウスを処置して治療的処置の効能を実証した。30日目(すなわち、処置後16日目)までに、C3腫瘍にチャレンジさせ、次いで処置を与えたグループ内の10匹のマウスすべてにおいて、触診可能な腫瘍の完全な根絶が実証された(図4;白四角)。対照的に、融合ペプチド以外の上記治療的処置の成分をすべて含む組成物で処置したコントロールマウス10匹のすべてにおける腫瘍は、サイズが増大し続けた(黒四角)。
予防
C3 腫瘍細胞によるインビチャレンジに対する本発明組成物の保護能力をさらに実証するために、雌性C57BL/6マウスの尾のつけね部に、油中水型エマルジョンに懸濁したリポソーム内にCpG ODNと共に封入した、PADRE(配列番号:10)と融合したR9Fペプチド(RAHYNIVTF;配列番号:1)(融合ペプチドと称する)を含む組成物を皮下注射した。組成物が、CpG ODNの代わりに別のアジュバントを含む組成物と同じくらい保護的であるかどうかを決定するために、CpG ODNが別のCpGアジュバント、すなわち、Pam3c(Pam3Cys−sKKKK)と置き換えられている上述の組成物をマウスに投与した。コントロールグループには、PBS、PBS中のCpG ODN、PBS中の融合ペプチド、CpG ODNと共にPBS中に懸濁した融合ペプチド、またはアジュバント無しでリポソームに封入した融合ペプチドを注射した。
確立した触診可能なC3腫瘍の治療を評価するために、マウスの左脇腹において、0.5 x 106個のC3細胞を皮下に植え込んだ。腫瘍植え込み後、4、5、6または9日目のいずれかに、リポソームに封入し、PBS/FIA油中水型エマルジョンに懸濁した、CpG ODNおよびPADRE(配列番号:10)と融合したR9Fペプチド(RAHYNIVTF;配列番号:1)(融合ペプチド)を含む組成物またはプラセボ(PBSエマルジョン中の融合ペプチドおよびCpG ODN)でマウス(n=10)を処置した。単回免疫感作は、腫瘍植え込み後4、5または6日目に免疫感作した処置グループ10匹のマウスすべて、および腫瘍植え込み後9日目に免疫感作したグループの30匹のマウスすべてにおいて40日目まで腫瘍を根絶した。腫瘍植え込み後5日目に処置したグループにおいて、1匹のマウスのみが、40日目まで腫瘍を維持した(第1表)。対照的に、腫瘍植え込み後4または6日目にプラセボ組成物で処置したグループにおいて、9/10のマウスに腫瘍が発生した。チャレンジ後5日間プラセボを投与したマウスのグループにおいて、10/10のマウスに腫瘍が発生し、腫瘍植え込み後9日目にプラセボワクチンで処置したグループにおいて、27/30のマウスに腫瘍が発生した。
メラノーマの治療的処置
チロシナーゼは、メラノーマにおいて過剰発現されたタンパク質である。チロシナーゼタンパク質からのペプチドは、メラノーマの治療のための一般的に不十分な抗原である。本明細書に記載するように、マウスMHC、H2K2およびヒトHLA−A2.1に結合するチロシナーゼ関連タンパク質(TRP−2)(アミノ酸181−188;VYDFFVWL;配列番号:6)由来のペプチドであるV8Lを治療的処置において用いて、IFN−γ産生細胞の産生を刺激した。TRP−2特異的IFN−γ産生細胞の数の刺激は、メラノーマに対する特異的治療効果が、予想されうることを示す。
乳がんの治療的処置
p53遺伝子産物は、悪性腫瘍、特に乳がんの治療のための理想的で広範に発現される標的である。ヒトのがんの大部分が、腫瘍発生における早期イベントとしてp53突然変異を示す。p53の過剰発現は、より悪性度の高いがん、リンパ節転移、標準的治療計画および最終的がん関連死亡率の独立予測因子である。
1つ以上の標的に対する治療的がん処置
いくつかのがんは、1つ以上の腫瘍関連タンパク質を同時に発現する。このようながんは、治療的処置のための1つ以上の標的を提供する。たとえば、メラノーマ細胞は、p53およびTRPの両方を過剰発現し、p53およびTRP標的の両方を発現している細胞は、治療に対して、より攻撃されやすいので、p53およびTRPの両方を同時に狙う治療が、より有効で特異的でありうるという可能性をもたらす。
ヒトHLA A2主要組織適合性複合体(MHC)遺伝子を有し、したがって、ヒトMHCを発現し、ヒト子宮頸がんを良好に模倣するHLA A2トランスジェニックマウスを用いた。HLA A2 MHCに適合させるために、これまでの実施例で用いたものとは相異するCTLエピトープを利用した。HLA A2マウスを以下の組成物の1つで処置した:
(1)4つのE6/7 ヒトパピローマウイルス(HPV)由来ペプチド(MP)の混合物、各ペプチドの配列は、以下の通り:
Y10T(E7:アミノ酸11−20;YMLDLQPETT;配列番号:2);
L9V(E7:アミノ酸82−90;LLMGTLGIV;配列番号:3);
T81(E7:アミノ酸86−93;TLGIVCPI;配列番号:4);および
T10V(E6:アミノ酸29−38;TIHDIILECV;配列番号:5);
(2)「aay」リンカーと結合して、長ペプチドとなった上記の4つのペプチド(AB2;配列番号:14)、その配列は以下の通り:
TIHDIILECVaayYMLDLQPETTaayLLMGTLGIVaayTLGIVCPI;
または
(3)上述の4つのペプチドから選ばれる単一のペプチド、すなわち、L9V(E7:アミノ酸82−90;LLMGTLGIV;配列番号:3)。
実施例7において、リンカー「aay」(−アラニン−アラニン−チロシン−)を用い、4つのHPV 16 E6/E7ペプチドを一緒に結合して、長ペプチドを形成した。このリンカーは、天然では疎水性であり、ペプチド製造を困難にし、長ペプチドを可溶化するためのジメチルスルホキシドの使用を必要とする、融合長ペプチドの疎水性を増大させる。
メラノーマに対する治療的処置
実施例4、5および6は、TRP−2およびp53特異的IFN−γ産生脾細胞の産生を増加する本発明組成物の能力による、メラノーマ関連タンパク質に対する細胞性免疫反応の刺激の確立を実証する。B16−F10細胞(10 x 103個の細胞/マウス)を無菌C57BL/6マウスの左脇腹の皮下に植え込んだ。マウスは、植え込みの時点で6−8週齢であり、水および食事を自由摂取させ、フィルタートップ条件下で飼育した。メラノーマ細胞の植え込みの5日後、TRP−2由来の2つのペプチドの内の1つ(V8LまたはS9L(配列番号:6または7);25 μg/マウス)、p53由来の1つの修飾ペプチド(mK9M(配列番号:8);25 μg/マウス)またはこれらのペプチドの混合物(25 μgの各ペプチド/マウス)を含む組成物の皮下注射による単回処置をマウスに施した。また、すべての組成物は、PADRE(25 μg/マウス)およびCpG ODN(50 μg/マウス)の両方を含み、PBS/ISA51油中水型エマルジョンに懸濁したリポソーム内にデリバリーした。コントロールマウスには、PBS単独の単回投与を施した。すべての注射は、尾部の付け根に投与した。次の式:最長測定値x(最短測定値)(Pilon−Thomasら、J.of Immunother.、29(4)、2006)を用いて、4−5日毎に腫瘍サイズを決定した。
B16メラノーマ腫瘍モデル
先の実施例において、2つの独立したHPV子宮頸がんモデル(C3およびTC1/A2)において確立した腫瘍における本発明組成物の有効性を実証した。HPV関連(bearing)腫瘍は、腫瘍細胞の表面上に存在するHPVのCTLエピトープを標的化することによって根絶された。この方策は、ウイルス誘発性がんを治療する場合に特に有効である。しかし、過剰発現した「自己」抗原を提示する腫瘍は、免疫系では見ることができないので、治療するのがより困難である。自己抗原は、耐性機構によってしっかりと護られている。効果的な治療的がん処置は、過剰発現した腫瘍関連自己抗原に対する免疫反応を誘発する能力をもたなければならない。メラノーマ(B16腫瘍モデルなど)は、クラスIMHCの発現および自己抗原の提示をダウンレギュレートすると考えられる。メラノーマの治療のための治療組成物は、腫瘍の表面上の自己エピトープを標的化する能力がある低親和性T細胞クローン型を活性化しなければならない。
C3腫瘍にマウス(C57BL/6)をチャレンジさせると、チャレンジ後8日目に腫瘍は、触診可能なサイズに発達した。チャレンジ後8日目に、マウスを2つのコントロールグループ(10匹のマウス/グループ)および油中水型エマルジョンに懸濁したリポソーム内に封入したCpG ODNおよびPADRE(配列番号:10)に融合した融合ペプチド(R9Fペプチド(配列番号:1)を含む組成物の単回投与で処置した処置グループに分割した。
T−ヘルパーエピトープとして破傷風トキソイドペプチド F21Eを用いるメラノーマの治療
破傷風トキソイド由来のT−ヘルパーエピトープと組み合わせたメラノーマ関連抗原、TRP−2を、PBS/ISA51油中水型エマルジョン中のCpG ODNを含む組成物に一緒に封入した。種々のT−ヘルパーエピトープが使用可能であることを実証するために、破傷風トキソイドペプチドを、先の実施例で用いたT−ヘルパーエピトープであるPADREと置き換えた。多数のTRP−2ペプチド特異的IFN−γ産生細胞の刺激が、メラノーマに対する特異的な治療効果が予想されうることを示す。
細胞系
10 % 加熱不活性化ウシ胎児血清(シグマ、セントルイス、ミズーリ)、2 mM L−グルタミン(ギブコ、バーリントン、オンタリオ)、50 mM 2−メルカプトエタノール(ギブコ、バーリントン、オンタリオ)、100 U/ml ペニシリンおよび100 μg/ml ストレプトマイシン(ギブコ、バーリントン、オンタリオ)を補足したイスコーブダルベッコ変法培地(IMDM;シグマ、セントルイス、ミズーリ)中で、C3細胞系を維持した。細胞を37℃/5 % CO2にてインキュベートした。
アメリカン・タイプ・カルチャー・コレクション(ATCC)、マナッサス、VAからB16F1(B16)メラノーマ細胞系を入手した。
CTLエピトープを含むHPV 16 E7(H−2Db)ペプチド RAHYNIVTF49−57(R9F)は、ダルトン・ケミカル・ラボラトリーズ・インコーポレイテッド(トロント、オンタリオ)によって、CD4+ヘルパーエピトープを含むPADREに融合された。この融合ペプチドを50 μg/投与にて用いた。指示した場合には、R9Fを、抗原(25 μg/投与)として、または細胞障害性アッセイにおいて用いた。ペプチド KYMCNSSCM(配列番号:13)(ダルトン)を無関係コントロールペプチドとして用いた。
ELISPOT決定に用いた無関係ペプチドのアミノ酸配列は、KIMCNSSCM(ダルトン・ケミカル・ラボラトリーズ・インコーポレイテッド)であった。
CpG ODN(下線の5'−TCCATGACGTTCCTGACGTT−3'CpGモチーフをもつ合成ODN 1826、50 μg/投与)(配列番号;12)を、クーレイ・ファーマシューティカル(ウェルズリー、マサチューセッツ)から入手した。リポペプチド(Pam3Cys−SKKKK、(100 μg/投与)は、EMC・マイクロコレクションズ、ドイツから入手した。
リポソームを以下の通り製造した:10:1(0.2 gのレシチンおよび0.02 gのコレステロール/投与)の比率のレシチンおよびコレステロールをクロロホルム/メタノール(1:1;v/v)に溶解し、PTFE 0.2 μmフィルターを用いて溶液を濾過滅菌した。ロータリーエバポレーターを用いてクロロホルムおよびメタノールを減圧除去し、得られる薄い脂質層から痕跡量の溶媒をさらに減圧除去した。リポソーム封入のために、融合ペプチドとCpGを滅菌PBSに溶解し、得られる溶液を混合しながら薄い脂質層に加えてリポソームを形成した。得られるリポソームの懸濁液を、FIAにリポソーム/PBS懸濁液を加えることによって、FIA(シグマ、セントルイス、ミズーリ)中で乳化して、油中水型エマルジョン(PBS:FIA;1:1,v/v;100 μl/投与)を形成した。いくつかの実施例では、油性担体として、FIAの代わりにモンタニドISA51(セピック、フランス)を用いた。
腫瘍植え込みに用いたC3細胞を95 %集密で増殖させ、0.05 %トリプシンで集菌した。マウスにおいて腫瘍を確立するために、マウスの左脇腹に0.5 x 106個のC3細胞を皮下注射した。次の式:最長測定値x(最短測定値)2÷2を用いて、4−5日毎に腫瘍サイズを決定した。
マウス(HLA A2)を左脇腹に皮下植え込みしたTC1/A2 腫瘍細胞(1 X 105個の細胞/マウス)にチャレンジさせた。5日毎に腫瘍サイズを測定し、個々のマウスにおける腫瘍サイズと腫瘍を有するマウスのパーセントで記録する。
B16−F10細胞(10 x 103個の細胞/マウス)を、チャレンジの時点で6−8週齢の無菌C57BL/6マウスの左脇腹に皮下植え込みした。2−5日毎に腫瘍サイズを測定し、結果を、腫瘍を有するマウスのパーセントで記録した。
インターフェロン(IFN)−γについてのCTLアッセイ、ELISPOTおよび細胞内染色は、無関係ペプチドは、バックグラウンドを越えるCTL活性またはIFN−γ産生を引き出さなかったので、治療的反応が選択されたE7ペプチドに対して特異的であることを明らかにした。これらの実験は、治療的処置を与えられたマウスからの脾細胞中の活性化した治療特異的細胞障害性T細胞が、腫瘍サイズの縮小と相関することを示す。手順の詳細を以下に記載する。
急性およびメモリーCTL反応を調べるために、他に特記しない限り、標的マウスからの脾細胞を、免疫感作後それぞれ7、14または130日目に分析した。記載した場合、1ラウンドのIVSにおいて細胞障害性アッセイを行った。簡単に述べると、インビトロ刺激の3日前に、CO2窒息によって、未処置のC57BL/6マウスを屠殺し、脾臓を採取し、分離した。脾細胞を洗浄し、RPMI−10(ここで、RPMIは、10 % 加熱不活性化ウシ胎児血清(シグマ、セントルイス、ミズーリ)、50 mM 2−メルカプトエタノール(ギブコ)、100 U/mlペニシリンおよび100 μg/mlストレプトマイシン(ギブコ)を補足される)中でカウントした。脾細胞(106個の細胞/ml)を、リポ多糖(25 μg/ml)および硫酸デキストラン(7 μg/ml)とともに培養し、リンパ芽球を処置した。
EL−4細胞を、5 μCi/ml [メチル−3H] チミジン(アマシャム・ファルマシア、エルランゲン、ドイツ)で標識した。細胞を37℃ /5 % CO2にて24時間インキュベートし、次いでR9Fまたは無関係ペプチド(10 μg/ml)とともに1時間搭載した。次いで、標識した標的細胞の懸濁液を採取し、RPMI−10中で2回洗い、2 x 103個の細胞/ウエルの密度で96ウエルのU底プレートに蒔いた。2 x 105個のエフェクター細胞/ウエルの濃度で開始する連続希釈によって、エフェクター細胞を加えた。プレートを37℃ /5 % CO2にて4時間インキュベートした。ガラス線維フィルター上に細胞を吸引し、Packard TopCountシンチレーションカウンターを用いてトリチウムを計数した。以下の式:DNAフラグメント化%=(S−E)/E x 100 (ここで、Sは、処置の不在下において保持されたDNA(カウント)であり、Eは、エフェクター細胞の存在下において保持されたDNA(カウント)である)を用いてDNAフラグメント化のパーセントを計算した。
C57BL/6マウスから採取した脾細胞中の処置活性化抗原特異的CTLを、BD ELISPOT(BD バイオサイエンス、サン・ディエゴ、カリフォルニア)を用いて検出した。簡単に述べると、処置後7日目に96ウエルのニトロセルロースプレートを、補足抗体である精製抗マウスIFN−γ抗体でコーティングし、4℃にて一夜インキュベートした。抗体を捨て、プレートを2時間ブロックし、次いで、ブロッキング溶液を除去した。最終体積100μl中、1x百万細胞/ウエルの初期濃度にて脾細胞を各ウエルに加え、次いで、次の列のウエルに連続希釈した。以下の刺激およびコントロールを、培地100μlに加えて、所望の最終濃度を得た。C3細胞(5 x 105個の細胞/ml)、R9Fペプチド(10μg/ml)または無関係ペプチド(10μg/ml)をウエルに加えるかまたはペプチドを加えなかった。PMA(5ng/ml、シグマ)、イオノマイシン(500ng/ml、シグマ)は、ポジティブコントロールとなり、培地単独は、ネガティブコントロールとなった。プレートを37℃/5% CO2にて一夜インキュベートした後、検出抗体であるビオチニル化抗マウスIFN−γ抗体を室温にて2時間加えた。インキュベーション期間の後、検出抗体を捨て、酵素複合体(ストレプトアビジン−HRP)を1時間加え、最後に、プレートをAEC基質溶液で20分間染色した。プレートを洗浄し、拡大レンズを用いてスポットを視覚化するために一夜風乾した。
先に述べたように、腫瘍なしマウスの脾臓から脾細胞を回収し、RPMI−10(500 x g、5分)で2回洗浄し、RMPI−10に懸濁した(10 x 106個の細胞/ml)。96ウエルの平底プレートのウエルに脾細胞(1 x 106個の細胞/ウエル)を加え、R9Fまたは無関係ペプチドとともに、各ペプチドについて複製カラムにおいて最終濃度3 μg/mlでインキュベートした。IFNγ産生CD8+ CTLの保護機能を実証するためにEL−4細胞を用いる実験において、細胞障害性測定の前に、R9Fまたは無関係ペプチドのいずれかを搭載したEL−4細胞(1 x 105個の細胞/ウエル)を37℃/5% CO2にて6時間インキュベートした。
