薬物や遺伝子などの活性物質を組織に的中させる能力は、臨床治療における目標として最も求められているものの一つである。「能動ターゲッティング」という用語で呼ばれる一つのアプローチは、特定の細胞に的中させるための、コロイド表面への特定のリガンドの結合に関する。この結果、リガンドはターゲット部位上の表面エピトープまたはレセプタに選択的に結合する[Moghimi SM, et al. Pharmacol Rev. 53(2): 283-318(2001)]。
本発明は、抗体などの標的物質をポリマベースナノ粒子(好ましくは治療用活性物質を具える粒子)に結合させる単純なアプローチの開発に基づくものであり、標的物質を粒子形成ポリマに結合させる事前試薬を必要としない。このことは、粒子のポリママトリックスに非共有結合している親油性部分と、続くステップで標的物質を結合することができるマレイミド化合物を具える第2の部分とを有する二官能性リンカの使用によって達成される。この新規なアプローチによれば、異なる各標的物質に、異なるナノ粒子成分を調整する必要がなくなり、必要に応じて様々な標的物質に単純にリンカを結合させることで様々な標的システムを作るのに使用することができる「ユニバーサル」なナノ粒子リンカ(細胞毒性薬などの活性物質を伴う)を形成することが可能である。
(i)ポリマナノ粒子と;

本発明は、一またはそれ以上の標的物質を、一又はそれ以上の薬物装填ナノ粒子へワンステップ結合させる新規なプロセスに基づく、薬物送達治療の改良を提供することを目的としている。特に、本発明は、選択した標的物質に順次結合することができるユニバーサルナノ粒子リンカ(選択的に薬物と組み合わせた)の調整が可能であり、従って、異なる各標的物質用に特別なナノ粒子を設計する必要がない。本発明による設計ナノ粒子によれば、二面メンブレイン低抗体密度を示す標的細胞をより良好に識別することができる。
ここで、Yは、ヘテロ原子、C1−C20アルキレンまたはアルケニレン、C5−C20シクロアルキレンまたはシクロアルケニレン、C6−C20アルキレン−シクロアルキキレン、をあらわし、ここで、前記アルキレンまたはアルケニレン中の炭素原子の一つがヘテロ原子で置換される;

例1−クロスリンカ(OMCCA)合成
オクタデシル-4-(マレイドメチル)シクロヘキサン-カルボン酸アミド(OMCCA)の合成のために、スルホスクシンイミジル-4-(N-マレイミドメチル)シクロヘキサン-1-カルボキシレート(SMCC Perce社 米国、イリノイ州)100mgと、ステアリルアミン(SA, Sigma Chemical社、米国、モンタナ州)80mgを、8mlのクロロフォルムと、トリエチルアミン(Reidel-de-Haen, Sigma-Aldrich Cemie GmbH社、シュテインハイム、ドイツ)41ulに溶解させ、反応液を50℃で、4時間インキュベートした。1%の塩化水素でこの溶液を3回洗浄し、減圧下でクロロフォルムを蒸発させた。生成物を一晩乾燥させ、重量を測った。収率は約90%であり、H−NMR(Mercury VX300, Varian, Inc.社、米国カリフォルニア州)、IR(Vector 22, Bruker Optics Inc. 社、米国、マサチューセッツ州)、及びLC−MS(Finnigan LCQDuo, ThermoQuest社、米国、ニューヨーク州)によってリンカの形成が、確認された。

(A)PEG−PLAの合成と特性
PEG−PLA(5:20)を、Bazile D. et al.[Bazile D, et al. J Pharm Sci, 84: 493-498(1995)]に記載されているような良く知られた手順に従って合成した。簡単に言えば、分子量5000のメトキシポリエチレングリコール(Sigma-Aldrich Chemie GmbH社、シュタインハイム、ドイツ)12gを、D, L-ラクチド(Purasob, Purac社、ゴリンヘム、オランダ)12gと、135℃の乾燥した条件で、2時間混合した。

ポリマ:ポリ乳酸(PLA)とポリ(エチレングリコール−co−乳酸)(mPEG−PLA)を、触媒(4)として第一スズ1−ヘキサノン酸エチルの存在下で開環重合法を用いて合成した。PLAを合成する場合、共触媒として、D,L−乳酸(30g)とベンジルアルコール(32mg)を250mlの乾燥トルエンに溶かし、一方、mPEG−PLAの合成の場合、メトキシポリエチレングリコール(mPEG、MW5000)1.5gを共触媒として使用し、すでに30gのD,L−乳酸を含む250mlの乾燥トルエンに加えた。還流混合物をディーンスターク装置で4時間攪拌して、水を共沸除去した。残りの水を除去した後、第一スズ1−ヘキサノン酸エチル(245mg)を加えた。次いで、混合物を135℃で4時間加熱した。粗ポリマを塩化メチレンに溶かし、4リットルの冷たいプロピルエーテル/石油エーテル混合物(3:2)の中に2回沈殿させた。特性を決定する前に、ポリマを真空乾燥させた。コポリマの合成は、次のスキーム2に記載されている。
ポリ乳酸及びポリ(エチレングリコール−co−乳酸)の特性
NP’sの調整
PLAナノ粒子を、Fessi H et al.[Fessi H, et al. Int. J. Pharm. 55: R1-R4(1989)]に記載されているナノ粒子−ポリマ界面蒸着法によって準備した。簡単に述べると、ポリマPLA(Boehringer Ingelheim社から購入したポリ乳酸、30KDa)88mgとコポリマPEG−PLA38mg、5:20(分子量5000のポリエチレングリコールと、分子量20000のポリ乳酸)を、水溶性有機溶剤であるアセトン20mlに溶かした。この有機相に対して、薬剤ドセタキセル10mgを加えた。抗体を結合させるために、この有機相にリンカOMCCA20mgを加えた。結果物としての有機相を、次いで、界面活性剤(Macrogol 15 ヒドロキシステアレート)として、Solutol(登録商標)HS15(BASF社、ルードウイッヒシャーフェン、ドイツ)100mgを含有する水相50mlへ加えた。この分散液を900rpmで、1時間以上混合して、次いで、減圧下で20mlまで蒸発させた。NPsを、ビバスピン300KDaカットオフを用いて燐酸緩衝食塩水で5−6回洗浄した。球形ポリマ状の、ナノメートル(100−500nm)粒子が、この条件の下で自然発生的に形成された。
薬物カプセル化(取り込み)の効力は、Kontron Instruments(Watford社、英国)325ポンプと、227nmに調整したKontron Instruments 332検出器、及びKontron Instruments 360自動サンプラからなるHPLCシステムを用いて決定した。LichroCART (Merck Darmstadt社、ドイツ) C18 (250*4 mm, 5um)カラムによって分離した。移動相は、1ml/分の流量の水中の50%のアセトニトリルであった。ドセタキセルの滞留時間は10分であった。
(1)粒子サイズの分析
平均径と粒子サイズ分布の測定を、25℃でALV非観血式後方散乱高性能粒子寸法測定器(ALV-NIBS HPPS, ランゲン、ドイツ)を用いて、水(屈折率:1.332;粘度:0.894543)を希釈液として行った。波長632nmのレーザビームを用いた。感度レンジは0.5nm乃至5μmであった。
二重に蒸留したNPs/免疫NPsの界面動電位を、Malvernゼータ電位測定装置(Malvern社、英国)を用いて測定した。
免疫NPsの形態学的評価は、金標識ヤギ抗ヒトIgG(Jackson ImmunoResearch Laboratories米国、ペンシルバニア州)を用いて、透過型電子顕微鏡(TEM)で行った。
薬物取り込みの効果
ナノ粒子中及び免疫ナノ粒子中の細胞障害性薬物ドセタキセル(DCTX)のカプセル化効率を、HPLCによって測定したところ、それぞれ、100%と49%であることがわかった。DCTXを装填したNPsの理論上の薬物含有量である、7.4%、w/w(PLA:PEG−PLA:DCTXの初期重量比; 88:38:10)が、DCTX免疫ナノ粒子の薬物含有量である、3.3%、w/w(PLA:PEG−PLA:DCTX:OMCCAの初期重量比;88:38:10:20)より有意に高かった。DCTX含有物におけるこの顕著な差異は、ポリママトリックス中のリンカの存在に関連するかもしれない。ナノ粒子形成中に、このリンカは、おそらく、DCTXと競合して、その取り込み限度を7.4から3.3%に減らす。
様々なNPsの平均サイズ及び粒子サイズ分布を、ALV法を用いて測定した。ブランクNPs(活性成分を含まない)の平均径は、60nmであり、一方、ブランクの免疫NPs(活性成分を含まない)と、DCTX装填免疫NPsの径は、それぞれ、150nmと180nmであった。NPsのこの顕著な径の増加は、リンカの存在に関連している。リンカは、おそらくより大きいNPsを形成する水相へのアセトンの拡散を低減する。
ブランクNPsの界面動電位は−18mVであり、抗体結合NPsの界面動電位は−7mVに減った(図2)。この界面動電位の減少は、pH7.4のトラスツズマブの正電荷に関連する。なぜなら、この等電点は9だからである。
図3A−3Bに記載した結果から、各金黒スポットは、ナノ粒子表面に付着した一のトランスツズマブ分子を表すことがわかる。MAbは、リンカによってナノ粒子の表面に効率的に結合し、反応状態は、二次抗体へのMabの初期の親和性に影響しないことが推測された。
チオール基を生成するための抗体の修飾
MAb上のチオール基を、2-イミノチオレーン試薬 [トラウトの試薬、 Sigma-Aldrich Chemi GmbH社、シュタインハイム、ドイツ、Traut RR, et al. Biochemistry. 12(17): 3266-73(1973)]を用いて増やした。トラウトの試薬を、精製トランスツズマブと共に、30:1のモル比で、45分間インキュベートした。トラウトで修飾したMAbを、HiTrap脱塩コラム(Amersham Bioscience社、ウプサラ、スエーデン)で分離した。修飾MAbを含む画分を、280nmのUVで測定した。フリーチオール基を、5,5’-ジチオ-ビス(2-ニトロベンゾイック酸)(エルマンの試薬、Sigma-Aldrich Chemie GmbH社、シュタインハイム、ドイツ)を用いて、412nmの吸光度の変化をモニタすることによって測定した。トラウトの試薬と反応させると、mAbが反応スルフヒドリルを有する。これは、OMCCAなどのマレイミド基を支えるスルフヒドリル反応架橋試薬との結合プロトコルに用いルことができる。以下のスキーム(3)は、低減した抗体とリンカのマレイミド基との間の可能な結合反応を示す。
ポリマPLA88mg(ポリ乳酸、30KDm コポリマPEG−PLA38mg、5:20と、薬物ドセタキセル0又は10mgと、DCTXナノ粒子のブランクナノ粒子146mgまたは156mgの総量と同等の、クロスリンカOMCCA20mg)(PLA:PEG−PLA:DCTX:OMCCA; 88:38:0/10:20)を、0.1Nの水酸化ナトリウムでpH6.5に調整し、トラウト修飾トラスツズマブ(最終濃度1mg/ml)で、夜通し、4℃で、連続的に攪拌して、窒素雰囲気下でインキュベートした。非反応マレイミド基は、2-メルカプトエタノール(Pierce社、米国、イリノイ州)で30分間インキュベートする間にブロックされた。非結合抗体と2-メルカプトエタノールは、Sepharose CL−4Bコラム(Amersham Bioscience社、ウプサラ、スエーデン)で、ゲルフィルタレーションによって免疫ナノ粒子から分離した。BCAプロテインアッセィ(ビシンコニン酸プロテインアッセィ)(Pierce社、米国、イリノイ州)によって、 [Smith P.K., et al. Anal. Biochem. 150: 76-85(1985)] に記載されているように結合効率を評価した。
この測定の目的は、トラスツマブ分子がブランクナノ粒子、すなわち、表面に固定されたリンカを含まないナノ粒子に物理的に吸収されるかどうかを評価するためのものである。このため、トラスツマブ7.5mg/ml溶液100μl(1mg)を、ブランクの正及び負に荷電したナノ粒子水性分散液であって、トータルで125mgのナノ粒子を含む水性分散液1mlと室温で1時間以上混合した。この混合液(750μl)を、30mlのPBSで5回洗浄し、希釈した分散液を遠心分離(4000rpm、30分)を用いて、ビバスピン300KDaカットオフを通して濾過して、吸収されなかったMAb分子を除去した。
SH基の測定
修飾MAbのスルフヒドリル基の数を、エルマンの試薬を用いて、標準としてのシステアミンと比較して決定した。無傷トラスツズマブとトラウトで修飾トラスツズマブを、0.1M EDTA pH8を含有するPBS緩衝液で希釈し、エルマンの試薬でインキュベートした。未処理トラスツズマブ中の1.4に比べて、MAb当たりのトラウトで修飾したトラスツズマブSH基の測定値は31.5であった。
NPsに結合したMAbの量を、BCAプ7ロテインアッセィを用いて測定した。NPsを、50℃で、0.1Nの水酸化ナトリウムで分解して、アッセイ試薬でインキュベートした。免疫NPs(薬物DCTXを含まない)と、免疫DCTXに装填したNPsの結合効率は、それぞれ71%と77%であった。
正及び負の調剤のための分離を行う前後のトラスツズマブの量の比率は、それぞれ、4.2%と2.7%であった。
HER−2/neu過剰発現測定
HER−2/neu過剰発現を、乳癌細胞株:SK−BR−3中で、及び、前立腺癌細胞株:LNCaP中で評価した。SK−BR−3細胞を、カバースリップ上で半密集するまで成長させた。細胞は新しい4%のパラフォルムアルデヒドを用いて10分間固定化し、洗浄して、5%のBSAで自己蛍光性をブロックした。無処理の、あるいはトラウトで修飾した(ウエルあたり400μl中0.1mg/l、0.05mg/l)、一次MAbで、4℃で細胞を一晩インキュベートした。細胞を洗浄して、FITC結合ヤギ−抗ヒトIgG(Jackson ImmunoResearch Laboratories、米国、ペンシルバニア州)の1:50希釈液で、室温で1時間インキュベートした。固定化した後、二次抗体を洗浄して蛍光顕微鏡または共焦点顕微鏡(Zeiss社, Axiovert 135M、オーバーコッヘン、ドイツ)のいずれかを用いて細胞を観察した。
HER−2/neu過剰発現の測定
SK−BR−3(乳癌細胞)やLNCaP(前立腺癌細胞)などの様々な癌細胞株中のHER−2/neu過剰発現を測定するために、免疫染色とFACS分析を上述したとおりに行った。固定した細胞を、HER−2/neu過剰発現を検出するためにトラスツズマブでインキュベートした。トラスツズマブでインキュベートしなかったが、二次FITC結合ヤギ抗ヒトIgGでインキュベートした細胞をコントロールとして使用した。
免疫ナノ粒子細胞結合の蛍光及び共焦点レーザ走査顕微鏡(CLSM)分析
FITC標識ヤギ抗−ヒトIgGでインキュベートした後、固定化SK−BR−3細胞をトラスツズマブ結合ナノ粒子製剤でインキュベートした。細胞を蛍光顕微鏡(結果は示さず)と共焦点顕微鏡(図6A−6B)を用いて調べた。免疫NPsの細胞表面への結合は、クロスリンカの濃度に比例していた。初期重量比PLA:OMCCA;50:10(図6B)のこの製剤によって誘発された蛍光は、初期重量比PLA:OMCCA;50:6(図6A)の製剤の蛍光より有意に高かった。これは、次いで生じるより高い密度のMAb、すなわち、BCAアッセイによって測定された、BのMAb濃度が150ug/mlであるのに対して、Aの濃度は40ugmlに過ぎないことによる。
この研究の目的は、二つの異なるMAbが同じナノ粒子に結合できることを示すことである。このため、二つの異なるMAbsを用いた。トラスツズマブと、AMB8LKの抗H−フェリチンモノクロナール抗体(MAT、Evry社、フランスから購入)である。各MAbは、蛍光プローブが顕著に異なっていた。
トラスツズマブ(1ml中21mg)を重炭酸ナトリウム0.165M緩衝液、pH9.4で洗浄した。DMF溶液中1mg/mlのスルホローダミンB塩素酸100μlを、MAb溶液に徐々に加えて、攪拌した。反応液を4℃で1時間インキュベートした。標識MAbをフリースルホローダミンB塩素酸から分離するために、PD10カラムを用いてPBS−EDTA pH7.2(1.8g NaHPO3(60mM)、4.35g NaCl(150mM)、0.93g EDTA(5Mm))で洗浄した。

1ml中4.1mgのAMB8LKを重炭酸ナトリウム0.165M緩衝液pH9.4で洗浄した。DMF溶液中の10mg/mlのFITC50μlを、MAb溶液に徐々に加えて、攪拌した。反応液を4℃で1時間インキュベートした。標識MAbをフリーFITCから分離するために、PD10カラムを用いてPBS−EDTA pH7.2(1.8g NaHPO3(60mM)、4.35g NaCl(150mM)、0.93g EDTA(5Mm))で洗浄した。回収した標識MAbの最終体積は2400μlであった。溶液15μlを、1:67のPBS−EDTAで希釈して、サンプルを、波長280mn(タンパク質)と、492nm(FITC)で、UV分光光度計で読み取った。

30%PEG−PLA ナノ粒子3mlを標識トラスツズマブ4.1mgとAMB8LK1.4mgでインキュベートした。製剤を窒素雰囲気下、4℃で二晩インキュベートした。結合MAbからフリーMAbを分離するために、ナノ粒子を300Kビバスピンで3回洗浄した。
結合効率
分離前と後の製剤のUV吸収を、UV分光光量計で読み取った。各ナノ粒子製剤50μlを、アセトニトリル1mlで希釈した。この結果物の比率が結合効率を表す(表2)。スルホローダミンB塩素酸標識トラスツズマブは570nmでの吸光度であり、FITC標識AMB8LKは492nmでの吸光度である。
平均粒径を、ALV非観血式後方散乱高性能粒子寸法測定器を用いて測定したところ、平均粒径は313nmであった。
分離前後のナノ粒子(3つの異なるサンプル中の)のゼータ電位を、二重に蒸留した水中で、Malvernゼータ電位測定装置(Malvern社、英国)を用いて測定した。
ナノ粒子抗体結合の安定性の研究
結合粒子の安定性を、温度の上昇、攪拌などの促進試験、および、長期保存評価を用いて生体内で調べた。
免疫ナノ粒子からの生体内薬物放出プロファイルは、低圧で限外濾過技術を用いて以下のとおり行われる。薬剤添加粒子(1−6mgの薬剤を含む)0.4mlを、放出媒体(シンク状態を維持している)100mlを含むAmicon8200 攪拌器(Amicon, Danvers社、米国、マサチューセッツ州)に直接入れる。所定の時間インターバルで、放出媒体をYM−100限外濾過膜(0.5bar以下)を通して、窒素ガスを用いて、低圧で濾過する。透明なろ液1mlのアリコートを分析して、HPLCを用いて薬物含有率を調べる。膜の吸収と除去は、薬物の水性濃度を正確に測定するために計数するべきであり、したがって、有効性の確認は限外濾過技術を使用する前に行われる。
SK−BR−3及びLNCaP細胞は、24個のウエルプレートで半密集状態まで成長させる。細胞を、クマリン−6標識ナノ粒子(ブランク粒子、DCTX装填NPs及びDCTX装填免疫NPs)で、37℃で、様々な時間インターバルをとってインキュベートした。励起波長485nm、及び発光波長520nmのFluoStar- Galaxy (BMG Labtechnologies社)で、プレートを蛍光測定する。各プレートにつき4回読み取りを行い、平均値を計算する。同じサンプルでインキュベートしていないウエルは、全蛍光に対する基準になる。
放射能標識ポリマ[3H]-ポリ(乳酸)でできた様々な粒子製剤(ブランク粒子、DCTX装填NPs及びDCTX装填免疫NPs)を、健康なオスのBALB/cマウス(20−26g)の尾静脈に、5ml/kg注入する。注入の後、以下の時間インターバルで、動物をエーテルで麻酔した:5、10、30分、1、2、8、24、48、72時間、1、2週。次いで、血液を心臓から回収して、動物を犠牲にする。心臓、肺、肝臓、脾臓、すい臓、腎臓、乳腺、結腸、腸及び脳を切除して、食塩水ですすぐ。血液を遠心分離にかけて血漿を得る。各時間インターバルにつき、5匹の動物を使用した。様々な器官サンプルをプラスチックバイタルに保存して、分析を行うまで冷凍する(−80℃)。器官と血漿の放射活性を、液体シンチレーションカウンタを用いて測定し、体内分布を評価した。HPLCまたはLC−MSによって、ドセタキセルを測定する。
PC−3.38ヒト前立腺癌細胞株を半密集に培養して、トリプシン処理を行い、PBSで洗浄した。オスのSCID/ベージュのマウス、8週齢に、ケタミン100mg/mlとキシラジン20mg/ml、85:15を筋肉注射して麻酔をかける。正中線下側腹部切開を行い、前立腺を露出させ、上述したように、腫瘍細胞(0.05mlのPBS中に5×105細胞)を前立腺に注入する[Honigmana A. et al. Mol Ther. 2001 Sep; 4(3):239-49]。
(A) 生体内研究用NP’sの作成
ポリマPLA(MW100,000)とmPEG−PLA(MW100,000)(2:1)を0.2%w/vツウイーン80を含むアセトン50ml(Sigma社、モンタナ、セントルイス)に溶かして、濃度0.6%w/vとした。薬剤装填用に、薬剤の0.08%w/vのパクリタキセル−パラミテート(pcpl)をポリマ混合物に加えて、有機相へ溶解させた。リンカOMCCA[オクタデシル-4-(マレイミドメチル)シクロヘキサン-カルボン酸]を、濃度0.04%w/vで有機相に組み入れた。この有機相を、0.25%w/vのSolutol(登録商標)HS15(BASF社、ルードウィッヒシャーフェン、ドイツ)を含む水相100mlに加えた。懸濁液を900rpmで1時間以上攪拌し、蒸発させて10mlに濃縮した。OMCCAを含む製剤をpH8.5に調整して、チオール化モノクロナール抗体(MAb)と共に窒素雰囲気下で、4℃で一晩インキュベートした。すべての製剤を0.1%のツウイーン80溶液(ビバスピン300、000MWCO、Vivascience, ストーンハウス、英国)100mlと共にダイアフィルタにかけ、1.2umのフィルタ(FP 30/1.2 CA、Schleicher & Schuell社、ダーセル、ドイツ)で濾過した。
トラスツズマブに結合したpcpl装填NPsを上述のとおり準備した。2mlの媒体(RPIM1640、Biological Industries社、 Beit Aemek, Israel)中のヒト前立腺癌細胞過剰発現HER−2(PC−3.38細胞300、000)を、12ウエルプレートのカバースライドに乗せて、37℃で、5%の二酸化炭素雰囲気中で24時間インキュベートして半密集にした。細胞を、4%のパラフォルムアルデヒド溶液(Fluka社、シュタインハイム、スイス)で固定化して、1%のBSA溶液(Sigma社、セントルイス、モンタナ州)で、室温でインキュベートした。BSA溶液を捨てた後、希釈した製剤(1:100)をこの細胞で4℃で2時間インキュベートした。細胞を冷たいPBS溶液(Biological industries社、 Beit Aemek, Israel)で3回洗浄し、次いで、FITC標識ヤギ抗ヒトIgG(Jackson ImmunoResearch Laboratories, 米国、ペンシルバニア州)でインキュベートした。冷たいPBS溶液で細胞を再度洗浄し、ガラススライドに固定して、Olympus 1X70共焦点レーザ走査顕微鏡(Olympus Co., Ltd. 東京、日本)で検査した。
モノクロナール抗体トラスツズマブに結合したpcpl免疫NPsは、図7A−7Dに示すように、PC−3.38細胞に過剰発現したHER2レセプタに対して親和性を示しており、結合プロセスが、トラスツズマブの元々の親和性結合に影響しないことを確認するものである。
PC−3.38細胞(300,000細胞)を成長させて、12ウエルプレート上に半密集させた。NPsと免疫NPsをクマリン−6で標識化した。次いで、細胞を、1:1000に希釈した標識NPsと、トラスツズマブ免疫NPsで、1mlの培地に、37℃で、5%のCO2雰囲気中で、3時間以上インキュベートした。PBSで3回洗浄した後、細胞を4%のPFAで固定し、グラススライドに装填して、CLSM(LSM410、Zeiss社、オーバークロッケン、ドイツ)で観察した。
蛍光NPsと免疫NPsを上述のとおり準備した。これらの製剤の物理的特性を表4に示す。
2mlの媒体(RPIM1640及びDMEM、それぞれ、Biological Industries社、 Beit Aemek, イスラエル)中のヒト前立腺癌細胞(300,000、PC−3.38、過剰発現H−フェリチン)及びヒトすい臓癌細胞(300,000、CAPAN−1、ヒトすい臓癌、過剰発現HER-2)を、12ウエルプレートのカバースライドに乗せて、37℃で、5%の二酸化炭素雰囲気中で24時間インキュベートして半密集にした。細胞を、4%のパラフォルムアルデヒド溶液(Fluka社、シュタインハイム、スイス)で固定化して、1%のBSA溶液(Sigma社、セントルイス、モンタナ州)で、室温でインキュベートした。BSA溶液を捨てた後、希釈した蛍光製剤(1:2000)をこの細胞と共に4℃で2時間インキュベートした。細胞を冷たいPBS溶液(Biological industries社、Beit Aemek, Israel)で3回洗浄し、ガラススライドに固定して、Olympus 1X70共焦点レーザ操作顕微鏡(Olympus Co., Ltd. 東京、日本)で検査した。
図9A乃至9Dに示すデータは、AMB8LKに結合した免疫NPs(図9B)またはトラスツズマブに結合した免疫NPs(図9C)、または1:1の比率のトラスツズマブとAMB8LKに結合した免疫NPs(図9D)を示し、AMB8LK免疫NPsは、特に、CAPAN-1中に過剰発現することが知られているH−フェリチンを認識し、トラスツズマブ免疫NPsは、HER-2受容体が過剰発現しないので、CAPAN-1を認識しなかった。しかし、組み合わせた免疫NPsをCAPAN−1細胞でインキュベートすると、NPsが細胞を認識して、AMB8LKの親和性が、トラスツズマブの存在と同じナノ粒子への結合によって影響を受けないことを明らかに示している。
培地中の細胞による放射能標識化製剤からの薬物の取り込みを、[3H]−パクリタキセル−パルミテート([3H]-pcpl)を含む溶液で、37℃で、3時間以上インキュベートした後、行った。2mlの媒質(RPMI1640)中のPC−3.38細胞(500,000)を12ウエルプレートに入れて、37℃で、24時間、5%の二酸化炭素雰囲気でインキュベートした。各ウエルにおいて、使用した初期放射線活性の総量が45μCiの [3H]-pcpl溶液、 [3H]-pcpl装填NPs、及びトラスツズマブに結合した[3H]-pcpl装填NPsであり、これは22μgのpcplと等価であった。37℃で、3時間、5%の二酸化炭素雰囲気でインキュベートした後、製剤を捨てて、細胞をPBSで3回洗浄した。細胞をトリプシン処理して、水酸化ナトリウム溶液で処理した。放射線活性を、Ultima-Goldシンチレーション混合物(Packard Instruments社、 米国、マサチューセッツ州、ボストン)中で、ベータカウンタ(Kontron Instruments社、ミラノ、イタリア)でモニタした。
図11に示すように、総放射線活性から取り込み率計算した。pcpl免疫NPsの取り込み率は、pcplNPs及びpcpl溶液の取り込み率より、顕著に高かった。これらの発見は、MAbsを用いた所望の組織への薬物装填コロイドキャリアの特定の標的化を確証するものである。
クレモフォールEL:エタノール溶液中の[3H]-pcplと、[3H]-pcpl装填NPsと、トラスツズマブに結合した[3H]-pcpl装填NPsの生体分散及び動的薬理学的プロファイルを、オスのBalb/Cマウス、8週齢で調べた。4匹のマウスを各群に振り分けた。この群では、pcpl7.5mg/kgの総投与量と同等である[3H]-pcplの0.225μCi放射線活性投与が、1回の大量瞬時投与で尾静脈に注入された。頚椎脱臼によって動物を犠牲にして、対象の組織(すなわち、心臓、肝臓、脾臓、腎臓、血液及び血漿)を同定し、単純な外科的技術を用いて取り除いた。滅菌食塩水(0.9%塩化ナトリウム)1mlで洗浄した後、組織の重量を測定し、水溶性組織可溶化剤(Packard社、 グロニンゲン、オランダ)1mlでインキュベートし、組織を30%の過酸化水素溶液(Fluka社、シュタインハイム、スイス)で脱色した。放射線活性を、Ultima-Goldシンチレーション混合物(Packard Instruments社、 米国、マサチューセッツ州、ボストン)中で、ベータカウンタ(Kontron Instroments社、ミラノ、イタリア)でモニタした。対数直線グラフに、時間に対する[3H]-pcplの血中濃度をプロットし(図6)、薬剤の動的薬理学パラメータを、WinNonlin(登録商標)Professionalソフトウエア、バージョン4.0.1を用いて無隔壁分析によって更に調べ、結果を表5に示した。組織中の薬剤のパーセンテージを薬剤初期投与量からグラム組織に標準化するときの、対象組織中の[3H]-pcplの生体分布を、選択された時間インターバルで示す(図12A乃至12F)。
図12A乃至12F及び表5に示すデータは、pcpl NPsと免疫NPsの滞留時間が、pcpl溶液より、血液中においてより長いことを示している。両ナノ粒子送達システム共、循環における薬物放出の引き延ばしに成功しており、ナノ粒子の隠れた特性によって、pcplの本質的な動的薬理学プロファイルが隠れていた。実際、NP表面に位置するPEG部分によって誘発された立体障害が、NPsのオプソニン化を防げて、循環時間を引き延ばす。pcplNPsと免疫NPsのターミナル半減期がそれぞれ14.6時間と20時間であり;pcpl溶液によって誘発される半減期8.3時間より有意に長いことは興味深い。更に、免疫NPsは、NPsより高い半減期値を示した。これは、おそらく、NP表面にトラスツズマブが結合した結果である。巨大分子である抗体は、おそらく、いくらかの追加の立体障害を与え、表5に示すデータからわかるように、正常にペグ化したNPsに比較して滞留時間を長くする。pcplNPsと免疫NPsは共に、pcpl溶液の無限大でのAUC値に比較して、CmaxとAUC値を顕著に上げた(表5)。しかし、pcplNPsとpcpl免疫NPs間のCmaxとAUC値には差がなかった。
Claims (31)
- 送達システムにおいて:
(i)ポリマベースのナノ粒子と;
(ii)前記ナノ粒子に非共有結合的に固定された第1の部分であって、前記第1の部分の少なくとも一部が、前記ナノ粒子に埋め込んだ親油性セグメントを具える第1の部分と、前記ナノ粒子の外側表面に露出させたマレイミド化合物を具える第2の部分と、を具えるリンカと;
を具えることを特徴とする送達システム。 - 請求項1に記載の送達システムにおいて、前記リンカが両親媒性分子であることを特徴とする送達システム。
- 請求項1または2に記載の送達システムにおいて、前記親油性部分が炭化水素または少なくとも8個の炭素を具える脂質を具えることを特徴とする送達システム。
- 請求項1乃至3のいずれか1項に記載の送達システムにおいて、前記リンカが以下の一般式(I):
を有し、
ここで、Yはヘテロ原子、C1−C20アルキレンまたはアルケニレン、C5−C20シクロアルキレンまたはシクロアルケニレン、C6−C20アルキレン−シクロアルキキレンを表し、前記アルキレンまたはアルケニレンの炭素原子の一つがヘテロ原子と交換可能であり;
Xは、-C(O)-R1, -C(O)-NH-R1, -C(O)-O-C(O)-R1, C(O)NH-R2-R1, または -C(O)-NH-R2-C(O)-NH-R1から選択されたカルボニル含有部分を表し、ここで、R1は,炭化水素または少なくとも8つの炭素を具える脂質、及びR2は,疎水性ポリマを表す、
ことを特徴とする送達システム。 - 請求項4に記載の送達システムにおいて、前記R1が、モノまたはジアクリルグリコール、リン脂質、スフィンゴ脂質、スフィンゴリン脂質、または脂肪酸であることを特徴とする送達システム。
- 請求項4または5に記載の送達システムにおいて、前記Yがアルキレン−シクロヘキサンであることを特徴とする送達システム。
- 請求項6に記載の送達システムにおいて、前記Yが、式 -CH2-C6H10- を有するアルキレン−シクロアルキキレン;Xが、式 -C(O)-NH-R1を有する部分を含むカルボニルであり、R1が脂肪酸であることを特徴とする送達システム。
- 請求項1乃至7のいずれか1項に記載の送達システムにおいて、前記リンカが、オクタデシル-4-(マレイミドメチル)シクロヘキサン-カルボン酸アミド (OMCCA); N-1 ステアリル-マレイミド (SM); スクシニミジルオレート; 1,2-ジエステアロイル-sn-グリセロ-3-ホスホエタノールアミン-N-[マレイミド(ポリエチレングリコール) 2000]; 及びこれらの混合物から選択されることを特徴とする送達システム。
- 請求項6または7に記載の送達システムにおいて、前記リンカが、オクタデシル-4-(マレイミドメチル)シクロヘキサン-カルボン酸アミド (OMCCA)であることを特徴とする送達システム。
- 請求項1乃至9のいずれか1項に記載の送達システムが活性物質を具えることを特徴とする送達システム。
- 請求項10に記載の送達システムにおいて、前記活性物質が前記粒子中に埋め込まれている、含侵している、あるいはカプセル化されている、または、前記粒子の表面に吸着されていることを特徴とする送達システム。
- 請求項1乃至11のいずれか1項に記載の送達システムにおいて、前記ポリマが、ポリヒドロキシブチル酸、ポリヒドロキシバレル酸、ポリカプロラクトーン、ポリエステルアミド、ポリチアノアクリレート、ポリ(アミノ酸)、ポリカーボネート、ポリアンヒドライド、ポリアルキルシアノアクリレート、及びこれらの混合物から選択された生体分解性ポリエステルであることを特徴とする送達システム。
- 請求項12に記載の送達システムにおいて、前記ポリエステルが、ポリ乳酸(PLA)、ポリグリコライド、ポリ乳酸−ポリグリコライド、ポリ(乳酸−co−グリコライド)、またはポリエチレングリコール−co−乳酸(PEG−PLA)であることを特徴とする送達システム。
- 請求項4に記載の送達システムにおいて、前記疎水性ポリマが、ポリエチレングリコール(PEG)、ポリシアル酸、ポリ乳酸(ポリラクチドとも言う)、ポリグリコール酸(ポリグリコライドとも言う)、アポリ乳酸−ポリグリコール酸、ポリビニルアルコール、ポリビニルピロリドン、ポリメトキサゾリン、ポリエチルオキサゾリン、ポリヒドロキシエチルオキサゾリン、ポリヒドロキシプロピルオキサゾリン、ポリアスパルトアミド、ポリヒドロキシプロピル メタクリルアミド、ポリメタクリルアミド、ポリジメチルアクリルアミド、ポリビニルメチルエーテル、ポリヒドロキシエチルアクリレート、ヒドロキシメチルセルロースまたはヒドロキシエチルセルロースなどの誘導体セルロース、から選択されることを特徴とする送達システム。
- 請求項14に記載の送達システムにおいて、前記疎水性ポリマが、平均分子量が2000乃至5000Daの間のPEGであることを特徴とする送達システム。
- 請求項10乃至15のいずれか1項に記載の送達システムにおいて、前記活性物質が、薬物、造影剤、あるいはこれらの混合物であることを特徴とする送達システム。
- 請求項1乃至16のいずれか1項に記載の送達システムが、一またはそれ以上の標的物質であって、各々が前記マレイミド化合物に共有結合している物質を具えることを特徴とする送達システム。
- 請求項17に記載の送達システムにおいて、前記標的作用物質が、アミノ酸ベース、核酸ベース、またはサッカライドベースのポリマ及び、これらの組み合わせから選択されたポリマであることを特徴とする送達システム。
- 請求項18に記載の送達システムにおいて、前記標的ポリマが、リガンド、抗体、抗原、グリコプロテインから選択されることを特徴とする送達システム。
- 請求項17乃至19のいずれか1項に記載の送達システムにおいて、前記標的物質が低分子量リガンドであることを特徴とする送達システム。
- 請求項19に記載の送達システムにおいて、前記抗体がモノ−またはポリ−クロナール抗体(MAb)であることを特徴とする送達システム。
- 請求項21に記載の送達システムにおいて、前記MAbが天然、あるいは遺伝子操作抗体であることを特徴とする送達システム。
- 請求項18または22に記載の送達システムが、少なくとも二つの抗体、または抗体フラグメントであって、それぞれが異なる結合特性を有する抗体または抗体フラグメントを具えることを特徴とする送達システム。
- 請求項21乃至23のいずれか1項に記載の送達システムにおいて、前記遺伝子操作抗体がトラスツズマブ、AMB8LK、またはこれらの組み合わせであることを特徴とする送達システム。
- 薬学的に許容可能なキャリアと組み合わせた請求項1乃至24のいずれか1項に記載の送達システムを具えることを特徴とする組成物。
- 疾病または疾患を治療または防止する方法において、当該方法が:必要のある対象に、請求項1乃至24のいずれか1項に記載のある量の送達システム、または、請求項26に記載のある量の組成物を提供するステップであって、前記送達システムが、前記疾病または疾患を治療または防止に有効な量の薬剤を具えるステップ、を具えることを特徴とする方法。
- 対象の身体内の標的細胞または標的組織を撮像する方法において、当該方法が:
(a)前記対象に請求項1乃至24のいずれか1項に記載の送達システムと共に造影剤を搬送するステップであって、前記ナノ粒子が一またはそれ以上の標的物質に結合されて、前記標的細胞または標的組織に前記送出システムを効率よく的中させるステップと;
(b)前記身体内の造影剤を撮像するステップと;
を具えることを特徴とする方法。 - 請求項25に記載の方法において、前記造影剤がクマリン−6を具えることを特徴とする方法。
- 請求項25に記載の方法において、前記送達システムが前記粒子内に埋め込んだ薬物を具えることを特徴とする方法。
- 請求項29に記載の方法において、前記薬物が細胞障害薬剤であることを特徴とする方法。
- 請求項30に記載の方法において、前記細胞障害薬剤が、ドセタキセルと、パルミチン酸パクリタキセルから選択した抗癌剤であることを特徴とする方法。
