Abstract
Description
(1)薬物送達用のキャリアを含む、投与計画に従って対象に投与するための組成物であって、
投与計画は、絶食させるか、または低血糖を誘発させた対象に該組成物を投与することおよび該対象において血糖値の上昇を誘発させることを含み、
該キャリアは、その外表面が第1の高分子と第2の高分子により修飾されており、第1の高分子は、数平均分子量が第1の所定分子量(例えば、4000Da)以上のポリエチレングリコールであり、第2の高分子は、数平均分子量が第2の所定分子量(例えば、3000Da)以下のポリエチレングリコールであり(ここで、第1の所定の数平均分子量は、第2の所定の数平均分子量よりも大きい)、
第1の高分子は、GLUT1リガンドにより修飾され、これにより該キャリアの外表面がGLUT1リガンドにより修飾されている、組成物。
(2)第1の高分子の分子数と第2の高分子の分子数との比が50:50〜30:70の範囲である、上記(1)に記載の組成物。
(3)第1の高分子の分子数と第2の高分子の分子数との比が45:55〜35:65の範囲である、上記(1)または(2)に記載の組成物。
(4)薬剤を脳に送達するための、上記(1)〜(3)のいずれかに記載の組成物。
(5)薬剤に血液脳関門を通過させるための、上記(1)〜(3)のいずれかに記載の組成物。
(6)薬剤を脳血管内皮細胞に送達するための、上記(1)〜(3)のいずれかに記載の組成物。
(7)血糖値の上昇がグルコース投与により誘発される、上記(1)〜(6)のいずれかに記載の組成物。
(8)組成物がグルコースの投与と同時にまたはその前に投与される、上記(7)に記載の組成物。
(9)組成物が、静脈内に輸液投与され、輸液投与が10分以上継続される、上記(1)〜(8)のいずれかに記載の組成物。
(10)キャリアが送達する薬剤を内包している、上記(1)〜(9)のいずれかに記載の組成物。
(11)GLUT1リガンドがグルコースである、上記(1)〜(10)のいずれかに記載の組成物。
(12)薬剤が、生理活性物質、抗体、核酸、生体適合性の蛍光色素、および造影剤から選択される少なくとも1つの薬剤である、上記(1)〜(11)のいずれかに記載の組成物。
(13)薬物送達用のキャリアを含む、組成物であって、
キャリアは、その外表面が第1の高分子と第2の高分子により修飾されており、第1の高分子は、数平均分子量4000Da以上のポリエチレングリコールであり、第2の高分子は、数平均分子量3000Da以下のポリエチレングリコールであり、

第2の所定の数平均分子量は、例えば、4000Da以上100kDa以下とすることができる。第2の所定の数平均分子量は、例えば、4000Da以上、5000 Da以上、6000 Da以上、7000 Da以上、8000 Da以上、9000 Da以上、または10 kDa以上、20 kDa以上、30 kDa以上、40 kDa以上、50 kDa以上、60 kDa以上、70 kDa以上、80 kDa以上、または90 kDa以上とすることができる。
但し、第1の所定の数平均分子量は、第2の所定の数平均分子量よりも大きいものとする。例えば、、第1の所定の数平均分子量は、第2の所定の数平均分子量よりも1 kD以上、2 kD以上、3 kD以上、4 kD以上、5 kD以上、6 kD以上、7 kD以上、8 kD以上、9 kD以上、または10 kD以上大きいものとすることができる。

本発明のある態様では、第2の高分子は、数平均分子量1500Daから3000Daのポリエチレングリコールである。この特定の態様では、第1の高分子は、数平均分子量5000Da以上、6000Da以上、7000Da以上、8000Da以上、または9000Da以上のポリエチレングリコールである。この特定の態様では、第1の高分子は、9000Da以上、20000Da以下、30kDa以下、40kDa以下、50kDa以下、60kDa以下、70kDa以下、80kDa以下、90kDa以下、または100kDa以下であり得る。
第1の高分子(共重合体の形態を含む)と第2の高分子(共重合体の形態を含む)との分子数比は、50:50〜20:80の範囲であり得、50:50〜30:70の範囲であり得、20:80〜40:60の範囲であり得、45:55〜35:65の範囲であり得る。分子数比は、第1の高分子と第2の高分子の混合比により調整することができる。
第2の高分子は、ブロック共重合体の一部であり得る。例えば、第2の高分子は、ポリカチオンまたはポリアニオンブロックとブロック共重合体を形成し得る。
これらのポリカチオンブロックまたはポリアニオンブロックとブロック共重合体を形成した第1の高分子および第2の高分子はそれぞれ、ポリアニオンまたはポリカチオンとミセルを形成し得る。本発明では、このようにして得られたミセルは、第1の高分子と第2の高分子とで修飾されたミセルである。
キャリアは、その外表面が第1の高分子と第2の高分子により修飾されており、第1の高分子は、数平均分子量が第1の所定の数平均分子量以上のポリエチレングリコールであり、第2の高分子は、数平均分子量が第2の所定の数平均分子量以下のポリエチレングリコール(ここで、第1の所定の数平均分子量は、第2の数平均分子量よりも大きい)であり、
第1の高分子は、GLUT1リガンドにより修飾され、これにより該キャリアの外表面がGLUT1リガンドにより修飾されている、組成物が提供される。薬物送達用キャリアは、治療薬および/または診断薬を送達し得る。
本明細書では、n位の炭素原子を介して結合したグルコースを「Glc(n)」{但し、nは、1〜4および6のいずれかの整数である}と表記することがある。例えば、本明細書では、6位の炭素原子を介して結合したグルコースを「Glc(6)」と表記し、2位の炭素原子を介して結合したグルコースを「Glc(2)」と表記することがある。
meO-PEG-PAspは既報に従い合成した。α-メトキシ-ω-アミノ-ポリエチレングリコール(meO-PEG-NH2, PEGの分子量: 2, 5, 12 kDa)を開始剤としたα-アミノ酸-N-カルボン酸無水物の開環重合(NCA重合)によってメトキシ-PEG-ポリ(β-ベンジル-L-アスパラギン酸) (meO-PEG-PBLA)を合成した。具体的には、meO-PEG-NH2を秤量しジクロロメタン、ベンゼンに溶解後一晩凍結乾燥させた。100 mg/mLになるようジクロロメタンに溶解し、100 mg/mLになるようN, N-ジメチルホルムアミド(DMF)、ジクロロメタンの1:9混合溶媒に溶解したNCA-BLAと混合した。35 ℃で3日間攪拌した。これらの操作はすべてアルゴン雰囲気下で行った。再沈殿、減圧乾燥により生成物を白色固体として得た。プロトン核磁気共鳴分光法(1H-NMR)により生成物の構造及びポリアスパラギン酸の重合度(DP)を、ゲル濾過クロマトグラフィー(GPC)により分子量分布を評価した。生成物のPEGの分子量(単位:Da)とPBLAの重合度(DP)の組み合わせは12k-80, 5k-72, 2k-78であった。得られたmeO-PEG-PBLAの脱保護によってメトキシ-PEG-ポリ-アスパラギン酸(meO-PEG-PAsp)を合成した。meO-PEG-PBLAを秤量しベンジル基に対し5 eq.の0.25 N水酸化ナトリウム水溶液に溶解、室温で1時間攪拌し反応させた。純水に対し室温で1日透析を行い、凍結乾燥によって白色粉末として生成物を得た。1H-NMR、高速液体クロマトグラフィー(HPLC)により生成物を評価した。1H-NMRによりベンジル基由来のピークが消失していることが確認され、脱保護反応が完了したことが示された。HPLCにより得られた分子量分布は単峰性で、反応によって二量体の生成や主鎖の分解が進行していないことが確認された。
Gluc(6)-PEG-PAspは、WO2015/075942に記載されるグルコースが6位の酸素でPEG-PAspと連結した分子である。上記と同様にDIG(6)-PEG-NH2(PEGの分子量: 5, 12 kDa)を開始剤としたNCA重合、脱保護によりGluc(6)-PEG-PAspを合成した(WO2015/075942参照)。1H-NMRおよびHPLCにより生成物を評価した。得られたブロック共重合体のPEGの分子量、ポリアスパラギン酸のDPの組み合わせは5k-72, 12k-67であった。
1.1で合成したmeO-PEG-PBLA(PEGの分子量: 2, 5, 12 kDa)のアミノリシス反応により、既報に従いPEG-PAsp(AP)を合成した。具体的には、meO-PEG-PBLAを秤量しジクロロメタン、ベンゼンに溶解後一晩凍結乾燥させた。15 mg/mLになるようにN-メチル-2-ピロリドン(NMP)に溶解し35 ℃で1日攪拌し溶解させた。meO-PEG-PBLAのベンジル基に対し100等量の1,5-ジアミノペンタン(DAP)とその半量のNMPを混合し氷水浴中で滴下、12 ℃で1時間攪拌し反応させた。これらの操作は全てアルゴン雰囲気下で行った。氷浴中でDAPのアミノ基に対し1.5等量の5 N塩酸で中和した。4 ℃で10 mM塩酸に対し1日、純水に対し1日透析し、凍結乾燥により白色固体として生成物を得た。1H-NMR及びHPLCにより生成物を評価した。得られたブロック共重合体のPEGの分子量、DPの組み合わせは2k-85, 5k-65, 12k-80であった。
既報に従いポリイオンコンプレックス(PIC)ミセルを調製した(WO2015/075942参照)。
糖の定量法であるフェノール硫酸法を用いて実施例2の手順で調製した高分子ミセル表層のグルコース分子の定量を行った。高分子ミセルを10 mM PBに溶解した溶液100 μLに、試験管中で5 w/v%フェノール溶液を100 μL添加、さらに濃硫酸500 μLを添加し、10秒程度激しく振蕩攪拌した。30分間室温で静置し、反応後の溶液を96 well black plateに200 μL/wellとり、490 nmにおける吸光をマルチプレートリーダーで測定した。高分子ミセルの調製に用いたグルコース修飾ブロック共重合体で検量線を作成し、高分子ミセル中のグルコース分子量を算出した。算出した高分子ミセル中のグルコースリガンド率を表2に示す。ブロック共重合体混合比とほぼ等しい比率でグルコース分子が高分子ミセルに修飾されていることが示された。
本実施例では、薬物として、DNA、RNAまたは抗体のミセル化を試みた。
本実施例では、薬物を封入しないミセルで脳への蓄積量を評価した。
培養細胞を用いた評価では、MDA-MB-231細胞を2000 cells/wellになるように96 well black plateに播種し、10 v/v%仔ウシ血清(FBS)、RPMI 200 μL/well中で24時間インキュベーションした。インキュベーション後培地を除去し、培地50 μL/well、1 mg/mL高分子ミセルPBS溶液15 μL/wellになるように混合した溶液に置換する。さらに1時間インキュベーションし、PBSで洗浄後100 μL/wellのPBSを添加しマルチプレートリーダーを用い高分子ミセル中のCy5に由来する蛍光の強度を測定した。結果を図4および5に示す。図4に示されるように、Gluc-PEG-PAsp(12k-67)を有する高分子ミセルのうち、分子量12 kDaのPEGのみからなるものはグルコース分子を有さない高分子ミセル(Null/m)と比較しMDA-MB-231への取り込み量に有意な差は確認されなかったが、分子量2 kDaのPEGからなるブロック共重合体を混合し調製した高分子ミセルはPEG(12 k) : PEG(2 k) = 50 : 50もしくは40 : 60のものでNull/mと比較しMDA-MB-231への取り込み量が有意に増加した。PEG(12 k): PEG(2 k) = 30:70の高分子ミセルについてはグルコースリガンドを導入した高分子ミセルとNull/mとで取り込み量に有意な差は確認されなかった。導入できるグルコースリガンド分子の比率の上限が30 %に制限されていたためと考えられる。また、図5に示されるように、Gluc-PEG-PAsp (5k-80)を有する高分子ミセルのうち、分子量5 kDaのPEGのみからなるものはNull/mと比較して取り込み量に有意な差は確認されなかったが、50 %の比率で分子量2 kDaのPEGを混合し調製した高分子ミセルはNull/mと比較し有意に取り込み量が増加した。
6.1に示した手順において、高分子ミセル添加前にGLUT1阻害剤(フロレチン, サイトカラシンB)をそれぞれ10 μL/wellを添加した培地で1時間インキュベーションし、GLUT1阻害に伴う高分子ミセル取り込み量の低下を評価した。結果を図6に示す。GLUT1の阻害剤としてフロレチンまたはサイトカラシンBいずれを添加した場合でも鎖長の異なるPEGを有するグルコース修飾高分子ミセル(Gluc/m)の取り込み量は有意に減少した一方でNull/mの取り込み量は阻害剤の添加による有意な差は確認されなかった。鎖長の異なるPEGを有するGluc/mのMDA-MB-231への取り込みがGLUT1とグルコース分子の相互作用によるものであることが示唆された。
鎖長の異なるPEGを混合することによるリガンド認識能の向上を、動物実験により検討した。
本実施例では、薬物として、DNA、RNAまたは抗体のミセル化を試みた。
また、図8Bに示されるように、pDNA封入ミセルでは、脳への送達効率の高い2k−50のみを用いてミセルを形成させようとするとその形成効率は低かった。これに対して、12k−50と2k−50とを4:6のモル比で含む混合物を用いて作製したミセルは、ミセルの形成効率が高く、単分散性の粒径分布を示した。図8Cに示されるように、抗体封入ミセルでも同様の傾向が確認された。
Claims (16)
- 薬物送達用のキャリアを含む、投与計画に従って対象に投与するための組成物であって、
投与計画は、絶食させるか、または低血糖を誘発させた対象に該組成物を投与することおよび該対象において血糖値の上昇を誘発させることを含み、
該キャリアは、その外表面が第1の高分子と第2の高分子により修飾されており、第1の高分子は、数平均分子量4000Da以上のポリエチレングリコールであり、第2の高分子は、数平均分子量3000Da以下のポリエチレングリコールであり、
第1の高分子は、GLUT1リガンドにより修飾され、これにより該キャリアの外表面がGLUT1リガンドにより修飾されている、組成物。 - 第1の高分子の分子数と第2の高分子の分子数との比が50:50〜30:70の範囲である、請求項1に記載の組成物。
- 第1の高分子の分子数と第2の高分子の分子数との比が45:55〜35:65の範囲である、請求項1または2に記載の組成物。
- 薬剤を脳に送達するための、請求項1〜3のいずれか一項に記載の組成物。
- 薬剤に血液脳関門を通過させるための、請求項1〜3のいずれか一項に記載の組成物。
- 薬剤を脳血管内皮細胞に送達するための、請求項1〜3のいずれか一項に記載の組成物。
- 血糖値の上昇がグルコース投与により誘発される、請求項1〜6のいずれか一項に記載の組成物。
- 組成物がグルコースの投与と同時にまたはその前に投与される、請求項7に記載の組成物。
- 組成物が、静脈内に輸液投与され、輸液投与が10分以上継続される、請求項1〜8のいずれか一項に記載の組成物。
- キャリアが送達する薬剤を内包している、請求項1〜9のいずれか一項に記載の組成物。
- GLUT1リガンドがグルコースである、請求項1〜10のいずれか一項に記載の組成物。
- 薬剤が、生理活性物質、抗体、核酸、生体適合性の蛍光色素、および造影剤から選択される少なくとも1つの薬剤である、請求項1〜11のいずれか一項に記載の組成物。
- 薬物送達用のキャリアを含む、組成物であって、
キャリアは、その外表面が第1の高分子と第2の高分子により修飾されており、第1の高分子は、数平均分子量4000Da以上のポリエチレングリコールであり、第2の高分子は、数平均分子量3000Da以下のポリエチレングリコールであり、
