Abstract
Description
(a)ヒトの生物学的サンプルを得る工程と、
(b)細胞表面のCD127、CD4及びCD25に対する抗体と霊長類の生物学的サンプル中の少なくとも1つの細胞を接触させる工程と、
(c)少なくとも1つの細胞をフローサイトメトリーにかける工程と、
(d)CD127lowCD4+CD25+の発現のフローサイトメトリーのシグナルを分析する工程と
を含み、CD127lowCD4+CD25+の発現が調節性T細胞又は調節性T細胞の集団を示す。

本明細書の文脈において、用語「含むこと」は、「主に包含するが、必ずしも単独では含まないこと」を意味する。さらに、「含む」及び「含む(三人称単数)」のような、単語「含むこと」の変形は、対応して意味を変える。
サンプル
末梢血は、健康な成人のドナー及び炎症性腸疾患を有する患者から得られた(Centenary Institute)及びRoyal Prince Alfred Hospital, Camperdown, NSW, Australia)。軟膜は、オーストラリアの赤十字血液サービス(Australian Red Cross Blood Service)(Sydney, NSW, Australia)から得られた。Nepean Hospital, Penrith, NSW, Australiaからの臍帯血サンプルは胎盤娩出直後に臍帯静脈から得られた。新生児は、満期であり、血液学的異常や感染合併症を有していなかった。正常な胸腺標本は、Children's Hospital, Westmead, NSW, Australiaで矯正心臓外科手術を受ける1〜7ヶ月齢の子供由来のものであった。研究は、児童倫理委員会(Children Ethics Committee)のためのシドニー中部及び西部領域の保健サービス(Central and Western Sydney Area Health Services)及びRoyal Alexandra Hospitalの承認により行なわれた。
末梢血、軟膜及び臍帯血単核細胞はフィコールパック勾配(Lymphoprep, Nycomed, Oslo, Norway)における遠心分離により調製された。
抗CD4モノクローナル抗体、抗CD25モノクローナル抗体及び抗CD45ROモノクローナル抗体(mAb)(それぞれ、クローンOKT4、7GB6及びUCHTL−1)は、標準的なプロトコルによるAlexa488(Molecular Probes, Oregon, USA)及びFITC(Sigma)で標識された。ヒト分子に対する特異性を有する以下のマウスmAbが、この研究で使用された:ビオチン−、Alexa488−、FITC−、PE−、PerCp又はPECy5結合した抗CD3、抗CD4、抗CD5、抗CD8、抗CD38、抗CD40L、抗CD44、抗CD45RA、抗CD45RO、抗CD54、抗CD56、抗CD57、抗CD58、抗CD62L、抗CD69、抗CD71、抗CD74、抗CD84、抗CD95、抗CD95L、抗CD103、抗CD122、抗CD134、抗CD152、抗HLA−DR、抗CXCR4、抗CXCR5(PharMingen, SanDiego, CA)、抗CD21、抗CD23、抗CD25、抗GITR(BD Biosciences, San Jose, CA)、抗CD27(Ancell, Bayport, MN)、抗CD28、抗CD30(Miltenyi Biotec GmbH, Gladbach, Germany)、抗CD70、抗CD127、抗CD244(Immunotech, Marseille, France)、抗CD148、抗CD150(DNAX Research Institute, Palo Alto, CA)、抗CD11a(Caltag, Burlingame, CA)及びFox P3(eBioscience又はBiolegend)。ビオチン複合体は、Alexa594(Molecular Probes)又はPerCp(PharMingen)と結合したストレプトアビジンにより生じた。PE−及びFITC結合した抗マウスmAb(Southern Biotechnology, Birmingham, AL)を使用して、非結合抗CCR7(PharMingen)を検出した。
軟膜単核細胞(5×108)は、ストレプトアビジン−Alexa594又はストレプトアビジン−PerCPで生じたCD4−FITCモノクローナル抗体、CD25−APCモノクローナル抗体、CD127−PEモノクローナル抗体及びCD45RA−ビオチンモノクローナル抗体の組み合わせによって染色された。選別前にAutoMacs(商標)(Miltenyi Biotech)を使用して、抗FITCビーズ(Miltenyi Biotech GmbH)による正の選択が行われた。胸腺細胞(5×108)は、CD4−PE−Cy5.5、CD3−PE、CD8−APC CD25−Alexa488、及びCD45RA−ビオチンmAbの組み合わせで染色された。FACSVantage(商標)又はFACSAria(商標)細胞分別装置を使用して、分別を行った。
RNA抽出のために、全RNAは、トリゾール(Invitrogen, Life Technologies)での溶解後に1〜5×105個の分別細胞から抽出された。Superscript IIリバーストランスクリプターゼ及びオリゴ(dT)2−18プライマー(Invitrogen)を最終容量20μlで使用して、サンプル全体を逆転写させた。リアルタイム定量PCR(RT qPCR)のために、反応混合物(18μl)は、2μlのcDNAと、10μlのPlatinum SYBR Green SuperMix UDG(Invitrogen)と、0.25pmolのフォワードプライマー及びリバースプライマーとを含んでいた。qPCRは、Rotor−Gene3000システム(Corbett research)で行った。アンプリコンがイントロン/エキソン境界にかかり、ゲノムDNAの増幅が最小限になるように、プライマーを設計した。プライマー配列は以下のようなものであった:Foxp3センス、GGCAAATGGTGTCTGCAAGTG(配列番号1)及びアンチセンス、GGATGATGCCACAGATGAAGC(配列番号2)。他の遺伝子に対するプライマー配列は、GATA3センス AACTGTCAGACCACCACAACCACAC(配列番号3)、GATA3アンチセンス GGATGCCTTCCTTCTTCATAGTCAGG(配列番号4)、T−betセンス CACTACAGGATGTTTGTGGACGTG(配列番号5)、T−betアンチセンス CCCCTTGTTGTTTGTGAGCTTTAG(配列番号6)であった。プライマーは、Invitrogenにより供給された。β−アクチンに対するプライマーは、以下のようなものであった:センス TCGACAACGGCTCCGGCATGTGCAAG(配列番号7)及びアンチセンス AGCCACACGCAGCTCATTGTAGAAG(配列番号8)(Sigma Genosys, Australia)。
in vitroの抑制分析は、96ウェルの丸底ウェルプレート中の、5%熱不活性化FCS、2mMのL−グルタミン、100U/mlのペニシリン及び100μg/mlのストレプトマイシンを追加したRPMI 1640から成る培地中で実行された。すべてのウェルは、5×104個のAPC、2×104個のレスポンダー細胞(responder cells:キラー細胞)(選別されたCD4+CD25−CD45RA+細胞又はCD4+CD25−細胞)及び0.25μg/mlの抗CD3(UCHT−1又はHit3a、PharMingen)を含んでいた。各々のウェルに加えられたサプレッサー細胞の推測上の数は、2×104、0.5×104、2×103又は0のいずれかであり、それぞれ1:1、0.25:1、0.1:1又は0:1のサプレッサー細胞対レスポンダー細胞の最終的な比率を与えた。対照培養では、レスポンダー細胞がサプレッサー細胞の代わりに加えられた。CFSE分析については、レスポンダー細胞はCFSEにより標識され、細胞分裂はフローサイトメトリーを使用して72時間後に測定された。増殖のパーセントは、レスポンダー細胞のみを含む対照ウェル中で分裂した細胞の平均の数に対して計算された。チミジン分析については、3日間、4日間又は5日間の培養後に培養物がパルスされた予備実験は、抑制の程度がその期間にわたって安定していることを示し、したがって、続く培養物はすべて72時間での3HTdRによりパルスされ、16時間後に採取されて、CFSE及びチミジンの分析の直接的な比較を可能にした。
[実施例1]
図1A及び図1Bは、マウス及びヒトのCD4+T細胞によるCD25の発現における差を示す。マウス末梢血において、6%のCD25を発現する細胞の別個の集団は、陰性の集団とは明らかに識別可能である(図1A)。10%のCD4+T細胞による発現の同じようなプロファイルは、ヒト臍帯血中に見られる。しかしながら、成人血のCD4+T細胞についてのプロファイルは、分離した亜集団として識別可能なCD25を最も高いレベルで発現する細胞はわずか1〜2%であるが、98%の他の細胞における発現レベルは中間から真の陰性に及んでいるというように、全く異なる。臍帯血プロファイル(点線、図1B)により示されたレベルでのゲートの設定は、成人の末梢血中でCD4+T細胞の大多数の集団を横断する。同じようなゲートジレンマはヒトのリンパ節により提起されるが、マウスのリンパ節では提起されない。種間差に対する主要原因は、ヒト末梢血CD4+T細胞の大部分における中間レベルの発現であり、それは抗原による感作を受けた「従来の」CD4+細胞に対応する。CD4+T細胞の多量の画分が活発に分裂している(図1A「刺激された」、これはRAG−/−動物を再構成する恒常性を保って分裂する集団を示す)マウスにおいてでさえ、従来の活性化T細胞におけるCD25の発現の平均のレベルは、CD25+Tregの同定を著しく妨害しない。ヒト抗原による感作を受けた「従来の」CD4+T細胞の大多数に対する高親和性IL−2受容体の発現の生理的な意義は、不明瞭なままである。
[実施例2]
様々なリンパ組織におけるCD25+CD4+T細胞の2つの集団を区別するCD127の発現の能力は、CD4、CD25及びCD127に対するmAbによって、正常成人血、リンパ節、臍帯血及び胸腺の染色サンプルで検査された。
[実施例3]
CD25+CD127lo集団中のFoxp3タンパク質の発現を測定するために、成人血及び臍帯血、リンパ節及び胸腺由来の細胞は、Foxp3及びCD127に対するmAbにより共染色された(図2b)。血液及びリンパ節では、FoxP3+細胞の集団はCD127loであり、図2aにおけるCD25+CD127lo細胞の集団の大きさに類似していた。これとは対照的に、胸腺のFoxP3+集団は、CD25+CD127lo集団よりもかなり多かった。末梢血では、CD4+CD127lo細胞(図2c、左上パネル中のようにゲートされた)の87%は、CD25+Foxp3+ゲート内にあり(図2c、左下パネル)、反対に、CD25+Foxp3+細胞の84%は、CD4+CD127loゲート内に検出された(図2c、右側のパネル)。しかしながら、胸腺のCD4+CD8−T細胞において、Foxp3+細胞の45%はCD25−であり(図2d、右下パネル)、それゆえにCD25+CD127lo細胞はFoxp3+細胞よりも有意に小さな集団から成っていた。それにもかかわらず、胸腺のCD4+CD8−Foxp3+細胞はすべてCD127loであった。したがって、CD25、CD127及びFoxP3の発現は、胸腺と末梢血との間で異なっていた。
[実施例4]
制御活性を有するナイーブCD4+CD45RA+CD25+細胞のサブセットがFoxP3+CD127lo表現型も有するか否かを検査するために、本発明者らは、CD3、CD4、CD45RA、CD25、CD127及びFoxP3に対するmAbにより成人血、リンパ節及び臍帯血細胞を染色した(図3a)。CD3+CD4+細胞はCD45RA−サブセット及びCD45RA+サブセットへ分離され、Foxp3+ゲート内のCD25+CD127lo細胞のパーセンテージが計算された。すべての組織において、全Foxp3+細胞の90%超はCD25+CD127loであったが、残りの細胞はCD25intCD127hiであった。さらに、CD127hi細胞の比率はCD45RA−Foxp3+サブセット及びCD45RA+Foxp3+サブセット内で同様であった。
[実施例5]
正常なレベルの循環しているCD4+CD25+CD127lo細胞を定義するために、43人の健康なボランティアの集団由来の末梢血サンプルを調べた(図4)。CD4+T細胞のパーセンテージとしてのCD45RA−CD25+CD127lo細胞の平均数(±SEM)は、4.29±0.24%であったが、CD45RA+CD25+CD127lo細胞のパーセンテージは2.05±0.14%であり、CD4+T細胞は合計で6.35±0.26%であった(図4b)。これは、本発明者らによりこれまでに観察されたマウス血液におけるCD4+T細胞の6.42±0.50%の図と一致しており(Tan and Fazekas de St Groth、未発表データ)、ヒト末梢血における1〜2%の従来の推測とは対照的であった(19)。さらに、エフェクター/メモリーTreg対ナイーブTregの比率は(図4a)、マウスについてこれまでに本発明者らが求めたエフェクターTreg対ナイーブTregの2:1比率の類似していた(Higgins and Fazekas de St Groth、未発表データ)。
[実施例6]
本発明者らは、末梢血CD4+T細胞の選別されたサブセット内のFoxp3 mRNAのレベルを測定した(図5a)。CD25+CD45RA−CD127lo細胞(集団1)はCD25−CD45RA−CD127hi細胞(集団4、図5b)よりも100倍多くのFoxp3 mRNAを発現した。中間レベルのFoxp3 mRNAは、CD25+CD45RA−CD127hi細胞(集団3)及びCD25−CD45RA−CD127lo細胞(集団2)で存在した。他方では、集団2はT−bet(Th1エフェクター機能のマスター調節因子)について最も高いレベルでmRNAを発現したが、GATA3(Th2機能のマスター調節因子)は等しくすべての集団で発現された(図5c)。これらの結果は、集団2がCD127loエフェクター細胞を含むことを示す。CD45RA+画分内で、CD25+CD127lo細胞は、ナイーブCD25−CD127hi細胞よりも100倍多いFoxp3を発現した(図5b)。
[実施例7]
CD45RA+サブセット及びCD45RA−サブセットへ分割された成人血のCD4+T細胞は、図6a(左パネル)において図示されたゲートに従って選別された。選別されたCD45RA+CD25−自己細胞(集団5)を、抑制活性を測定するために、共培養においてレスポンダー細胞として使用した。細胞増殖の指標としてチミジン(図6b)又はCFSE(図示せず)のいずれかを使用する分析は、各々のCD45サブセット(集団1及び集団3、図6b)内のCD25+CD127loT細胞のみがin vitroの抑制を媒介したことを示した。CD45RA+Tregは、それらのCD45RA−相当物と同じくらい有力であった。臍帯血分析については、臍帯血細胞の大部分がある程度までCD45RAを発現するので、CD45アイソフォーム発現は細胞をさらに分けるためには使用されなかった。図6a(右側パネル)におけるゲートに従って選別されたCD25+CD127loサブセット及びCD25+CD127hiサブセットは、CD4+CD25−CD127hi自己レスポンダー細胞(集団8)と共培養された。前と同じように、チミジン(図6b)及びCFSE分析(図示せず)の両方は、CD4+CD25+細胞の抑制活性が、CD127loサブセット(集団6、図6b)に限定されることを示した。
[実施例8]
活性化T細胞からのTregの区別において、CD4/CD25/CD127/CD45RA染色戦略がさらなる明瞭さを与えたことを示したので、本発明者らは、43人の対照及び38人のIBD患者の集団由来の末梢血サンプルにこの方法を適用した(表1)。
[実施例9]
末梢血においてナイーブTregの数が年齢に応じて減少すると主張するこれまでの報告とは対照的に、本発明者らは、ナイーブTregのパーセンテージがIBD患者にとって本質的に一定であったことを示し、それゆえに若年患者でのナイーブTreg欠損は、高齢患者においては明白ではなかった(図9a)。より若い患者における欠損の一部は、対照と比較して、患者におけるCD4+CD45RA+細胞のパーセンテージの減少のためであったが(図9b)、CD4+細胞のパーセンテージ全体では差はなかった(図9e)。
[実施例10]
血液の研究に加えて、本発明者らは、IBDの患者、又は憩室症若しくは失調症のために結腸を切除した患者から切除された新鮮な結腸の標本由来の粘膜及びリンパ節を得た。
[実施例11]
この研究は、乳児期から後期成人期への調節性T細胞の数の変化を評価するために、細胞内のFoxP3染色対CD4/CD25/CD127の組み合わせを比較した。このデータは、図13〜図17において示されるように、CD45RAコンパートメントとCD45ROコンパートメントとの間の分布は著しく移行するが、Treg細胞のプールが一生を通じて狭い範囲内で維持されることを示す。この移行は、従来のT細胞の移行に匹敵する。
[実施例12]
75歳を超えた4人の健常な個体由来の末梢血Tregは、アルツハイマー病を患う年齢の一致した5人の個体と比較された。この研究は、ラッシュ施設内倫理委員会(Rush Institutional Review Board)の承認によりラッシュ大学メディカルスクール(Rush University Medical School)で実行された。この研究の被験体の詳細は表2に示される。
[実施例13]
Westmead hospitalに通院する重症のアトピー性湿疹の患者(20,000I.U.の平均のIgEレベル)由来の末梢血は、CD4/CD25/CD127/FoxP3で染色された。患者のうちの1人についてのCD127対FoxP3による染色の比較は、図19に示される。健康な対照に関しては、FoxP3の発現とCD127loCD25+表現型との間で明確な相関性があった。図20は、CD4+CD25+細胞の従来のゲート対CD4+CD25+CD127loゲートに由来したTreg数の比較を示す。前者は健康な被験体と湿疹患者との間で差を示さないが、新しいゲートは湿疹患者におけるTreg細胞の有意に高い増加を示す。
[実施例14]
末梢血サンプルは、表3に示されるような個体から得られた。
[実施例15]
18人の原発性のシェーグレン症候群患者(すべて女性)及び17人の年齢の一致した健康な女性の対照由来の末梢血を、CD4/CD25/CD127/CD45RA mAbの組み合わせで染色した。原発性シェーグレン症候群はヨーロッパの基準に従って診断された(29)。図23に示されるように、ナイーブTreg細胞と全(しかし活性化されていない)Treg細胞とのパーセンテージは、シェーグレン症候群患者においてより高かった。この増加は年齢分布全体にわたって見られた。
[実施例16]
末梢血は、28人の進行性疾患のHIV+患者(Rush University Medical Center, Chicago, IL)、10人の一次感染をともなう患者(セロコンバータ、St Vincents Hospital, Darlinghurst, NSW, Australia)、及び7人の免疫再構築症候群(IRD)を発症したHIV+患者(St Vincent Hospital)から得られた。一次感染をともなう患者及び28人中の3人の後期疾患の患者以外のすべて患者は、抗レトロウイルスの薬剤による治療を受けていた。この研究は、中央領域保健サービス(Central Area Health Services)及びラッシュ施設内倫理委員会の承認で行なわれた。
[実施例17]
メルボルンのオースティン病院のルードウィヒ癌研究所(Ludwig Institute For Cancer Research at the Austin Hospital)で行なわれた研究において、黒色腫の患者由来の末梢血においてTregが列挙された。表5に示される6人の患者は、ワクチン治験に登録されなかったが、個体中のTreg数の日々の変動を評価するために多くの週にわたって1週当たり3回採血された。

60人の慢性腎不全(CRF)の患者群がPrince of Wales Hospital(Sydney)で登録された。患者の詳細は表7において示される。
[実施例19]
末梢血は、メルボルン大学及びCSIRO畜産業動物実験及び倫理委員会の承認により行なわれたワクチン接種研究の一部として32匹のマカクから得られた。対照(11匹の被験体)、Gagタンパク質により治療された群(10匹の被験体)、並びにGag、Env、Pol及びRTNVVVにより治療された群(10匹の被験体)の異なる3群があった。

Claims (48)
- 調節性T細胞又は調節性T細胞の集団を同定する方法であって、霊長類の生物学的サンプル中の少なくとも1つの細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析することを含み、CD127lowCD4+CD25+の発現が調節性T細胞又は調節性T細胞の集団を示す方法。
- 前記分析する工程が、少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドについて細胞の発現レベルを測定することをさらに含む、請求項1に記載の方法。
- 前記少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドが、細胞表面に結合又は細胞内に存在する、請求項2に記載の方法。
- 前記少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドが、CD45RA、CD45RO、Foxp3、CTLA−4又はCD95を含む群から選択される、請求項2又は3に記載の方法。
- 前記霊長類の生物学的サンプルが、細胞株、体液又は組織を含む群から選択される、請求項1〜4のいずれか一項に記載の方法。
- 前記体液が血液又はリンパ液を含む、請求項5に記載の方法。
- 前記サンプルが、胸腺、リンパ節、脾臓、扁桃腺、少なくとも1つの単離リンパ球又は少なくとも1つのT細胞を含む、請求項5に記載の方法。
- (a)霊長類の生物学的サンプルを得る工程と、
(b)細胞表面のCD127、CD4及びCD25に対する抗体と前記霊長類の生物学的サンプル中の少なくとも1つの細胞を接触させる工程と、
(c)前記少なくとも1つの細胞をフローサイトメトリーにかける工程と、
(d)CD127lowCD4+CD25+の発現の前記フローサイトメトリーのシグナルを分析する工程と
を含み、前記CD127lowCD4+CD25+の発現が調節性T細胞又は調節性T細胞の集団を示す、請求項1〜7のいずれか一項に記載の方法。 - 調節性T細胞又は調節性T細胞の集団を同定する方法であって、霊長類の生物学的サンプル中の少なくとも1つの細胞を分析し、細胞のCD127の発現レベルを測定する、分析することを含み、該サンプル中の該少なくとも1つの細胞が、あらかじめCD4及び/又はCD25の発現について分析されて、CD4+及び/又はCD25+であることが明らかにされており、CD127lowCD4+CD25+の発現が、調節性T細胞又は調節性T細胞の集団を示す方法。
- 前記少なくとも1つの細胞が、あらかじめCD4の発現について分析されており、前記分析する工程が、細胞のCD25の発現レベルを測定することをさらに含む、請求項9に記載の方法。
- 前記少なくとも1つの細胞が、あらかじめCD25の発現について分析されており、前記分析する工程が、細胞のCD4の発現レベルを測定することをさらに含む、請求項9に記載の方法。
- 前記サンプルが、胸腺、リンパ節、脾臓、扁桃腺、少なくとも1つの単離リンパ球、少なくとも1つのT細胞、少なくとも1つのCD4+T細胞、少なくとも1つのCD25+T細胞又は少なくとも1つのCD4+CD25+T細胞を含む、請求項9〜11のいずれか一項に記載の方法。
- 霊長類の被験体において、調節性T細胞又は調節性T細胞の集団を同定するためのキットであって、少なくともCD127の細胞の発現レベルの測定ための少なくとも1つの薬剤を含むキット。
- 前記少なくとも1つの薬剤が、少なくとも1つの抗CD127抗体又はCD127に対して特異的な少なくとも1つのオリゴヌクレオチドを含む群から選択される、請求項13に記載のキット。
- 霊長類の被験体において、少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドの細胞の発現レベルを測定するための少なくとも1つの薬剤をさらに含む、請求項14に記載のキット。
- 前記少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドが、CD4、CD25、CD45RA、CD45RO、Foxp3、CTLA−4又はCD95を含む群から選択される、請求項15に記載のキット。
- 調節性T細胞の集団を定量するためのキットであって、少なくともCD127の細胞の発現レベルの測定ための少なくとも1つの薬剤を含むキット。
- 霊長類の生物学的サンプル中の調節性T細胞の量を定量化する方法であって、細胞のCD127、CD4及びCD25の発現レベルを測定するために該サンプル中に分析細胞を含み、該サンプル中のCD127lowCD4+CD25+細胞の量が該サンプル中の調節性T細胞の量を示す方法。
- 霊長類の生物学的サンプル中の調節性T細胞の量を定量化する方法であって、細胞のCD127の発現レベルを測定するために該サンプル中に分析細胞を含み、該細胞があらかじめCD4及び/又はCD25の発現について分析されて、CD4+及び/又はCD25+であることが明らかにされており、該サンプル中のCD127lowCD4+CD25+細胞の量が該サンプル中の調節性T細胞の量を示す方法。
- 霊長類の被験体において、調節性T細胞の過剰生産又は低生産を診断する方法であって、
(a)前記被験体から生物学的サンプルを得ること、及び
(b)前記サンプル中の細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析すること
を含み、前記サンプル中の少量のCD127lowCD4+CD25+細胞が前記被験体の調節性T細胞の低生産を示し、該サンプル中の多量のCD127lowCD4+CD25+細胞が該被験体の調節性T細胞の過剰生産を示す方法。 - 調節性T細胞の低生産が、炎症性腸疾患若しくはその素因、又は臓器移植に関連する、請求項20に記載の方法。
- 調節性T細胞の過剰産生が、原発性シェーグレン症候群、湿疹、喘息、肝細胞癌又はHIV感染に関連する、請求項20に記載の方法。
- 少なくとも1つの対照サンプル中のCD127lowCD4+CD25+細胞の量と、前記霊長類の被験体から得られたサンプル中のCD127lowCD4+CD25+細胞の量を比較することを含む、請求項20〜22のいずれか一項に記載の方法。
- 前記少なくとも1つの対照サンプルが、自己免疫性疾患、免疫炎症性疾患、若しくはアレルギー性疾患、肝細胞癌、臓器移植、HIV感染、それらの素因、又は調節性T細胞の量の変化に関連する他の疾患をともなわない霊長類の被験体由来である、請求項20〜22のいずれか一項に記載の方法。
- 霊長類の被験体において疾患を診断する方法であって、
(a)前記被験体から生物学的サンプルを得ること、及び
(b)前記サンプル中の細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析すること
を含み、前記サンプル中の少量のCD127lowCD4+CD25+細胞が前記被験体の疾患を示す方法。 - 前記疾患が、炎症性腸疾患、全身性紅斑性狼瘡、乾癬性関節炎、若年性特発性関節炎、若年性糖尿病、川崎病、肝細胞癌、HIV感染又は調節性T細胞の量の変化に関連する他の疾患を含む群から選択される、請求項25に記載の方法。
- 病状、感染又は治療の経過の間に霊長類の被験体の調節性T細胞の量をモニタリングする方法であって、
(a)前記被験体から生物学的サンプルを得ること、及び
(b)前記サンプル中の細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析すること
を含み、前記サンプル中のCD127lowCD4+CD25+細胞の量が該サンプル中の調節性T細胞の量を示す方法。 - 前記病状又は前記感染が、自己免疫性疾患、免疫炎症性疾患若しくはアレルギー性疾患、又はそれらの素因、感染症又は癌を含む群から選択される、請求項27に記載の方法。
- 前記治療が化学療法である、請求項27に記載の方法。
- 霊長類の被験体の病状又は感染のための治療に対する反応を、該被験体の調節性T細胞の量に基づいて予測する方法であって、
(a)前記被験体から生物学的サンプルを得ること、及び
(b)前記サンプル中の細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析すること
を含み、前記サンプル中のCD127lowCD4+CD25+細胞の量が該サンプル中の調節性T細胞の量を示す方法。 - (a)霊長類の被験体における調節性T細胞の前記過剰生産又は前記低生産、
(b)霊長類の被験体における自己免疫性疾患、免疫炎症性疾患若しくはアレルギー性疾患、又はそれらの素因、及び/又は
(c)霊長類の被験体における調節性T細胞の量の変化に関連する疾患
の診断で用いられるキットであって、
少なくともCD127の細胞の発現レベルの分析のために少なくとも1つの薬剤を含むキット。 - 前記少なくとも1つの薬剤が、少なくとも1つの抗CD127抗体及び/又はCD127に対して特異的な少なくとも1つのオリゴヌクレオチドから選択される、請求項31に記載のキット。
- 少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドの細胞の発現レベルを測定するための少なくとも1つの薬剤をさらに含む、請求項31又は32に記載のキット。
- 病状、感染又は治療の経過の間に、霊長類の被験体における調節性T細胞の量をモニタリングするのに用いられるキットであって、少なくともCD127の細胞の発現レベルを分析するための少なくとも1つの薬剤を含むキット。
- 前記少なくとも1つの薬剤が、少なくとも1つの抗CD127抗体及び/又はCD127に対して特異的な少なくとも1つのオリゴヌクレオチドから選択される、請求項34に記載のキット。
- 前記病状又は前記感染が、自己免疫性疾患、免疫炎症性疾患若しくはアレルギー性疾患、又はそれらの素因、感染症又は癌を含む群から選択されてもよい、請求項34又は35に記載のキット。
- 前記治療が化学療法である、請求項34に記載のキット。
- 少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドの細胞の発現レベルを測定するための少なくとも1つの薬剤をさらに含む、請求項34〜37のいずれか一項に記載のキット。
- 霊長類の被験体における病状又は感染のための治療に対する反応を、該被験体における調節性T細胞の量に基づいて予測するのに用いられるキットであって、少なくともCD127の細胞の発現レベルの分析のための少なくとも1つの薬剤を含むキット。
- 調節性T細胞又は調節性T細胞の集団を単離するための方法であって、
(a)霊長類の生物学的サンプル中の少なくとも1つの細胞を分析し、細胞のCD127、CD4及びCD25の発現レベルを測定する、分析することであって、CD127lowCD4+CD25+の発現が調節性T細胞又は調節性T細胞の集団を示す、分析すること、及び
(b)前記少なくとも1つの細胞を単離することであって、該少なくとも1つの細胞がCD127lowCD4+CD25+である、単離すること
を含む方法。 - 前記分析する工程が、少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドについての細胞の発現レベルを測定することをさらに含む、請求項40に記載の方法。
- 調節性T細胞又は調節性T細胞の集団を単離するための方法であって、
(a)霊長類の生物学的サンプル中の少なくとも1つの細胞を分析し、細胞のCD127の発現レベルを測定する、分析することであって、該サンプル中の該少なくとも1つの細胞があらかじめCD4及び/又はCD25の発現について分析されて、CD4+及び/又はCD25+であることが明らかにされており、CD127lowCD4+CD25+の発現が調節性T細胞又は調節性T細胞の集団を示す、分析すること、及び
(b)前記少なくとも1つの該細胞を単離することであって、該少なくとも1つの細胞がCD127lowCD4+CD25+である、単離すること
を含む方法。 - 前記少なくとも1つの細胞があらかじめCD4の発現について分析され、前記分析する工程が細胞のCD25の発現レベルを測定することをさらに含む、請求項42に記載の方法。
- 前記少なくとも1つの細胞があらかじめCD25の発現について分析され、前記分析する工程が細胞のCD4の発現レベルを測定することをさらに含む、請求項42に記載の方法。
- 前記分析する工程が、少なくとも1つの追加の細胞のポリペプチド又はポリヌクレオチドについての細胞の発現レベルを測定することをさらに含む、請求項42〜44のいずれか一項に記載の方法。

- 請求項40〜45のいずれか一項に記載の方法によって単離した場合の少なくとも1つの調節性T細胞を使用する、調節性T細胞療法についての方法。
- 前記T細胞療法が、自己免疫性疾患、アレルギー性疾患、免疫炎症性疾患若しくは感染症、癌のための治療法又は臓器移植を含む群から選択される、請求項47に記載の方法。
