本願明細書の文脈において、用語「含む」は、「主として含むが、必ずしも単独ではない」ことを意味する。さらに、「含有する」や「含んでなる」のような「含む」の変形は、相応に変化した意味を有する。
本願明細書に記載のように、本発明には典型的には、対応する野生型Cpn10ポリペプチドと比較して1つ以上のアミノ酸欠失、付加又は置換を含んでなり、免疫調節活性を有するCpn10ポリペプチドが包含される。典型的には前記野生型Cpn10は真核生物由来の任意のCpn10ポリペプチドである。例えば、Cpn10は、酵母(Saccharomyces cerevisiae等)、線虫(Caenorhabditis elegans等)、カエル(Xenopus tropicalis等)、ニワトリ(Gallus gallus等)、ゼブラフィッシュ(Danio terio等)、ハエ(Drosphila melanogaster等のコバエ等)、植物(Arabidopsis thaliana等)又は哺乳類由来であってもよい。哺乳類Cpn10は霊長類、ネズミ、ヒツジ、ウシ、イヌ、ネコ、ブタ又はウマの由来であってもよい。あるいは、Cpn10は古細菌由来であってもよい。特定の実施形態において、当該Cpn10はヒトCpn10である。野生型のヒトCpn10は配列番号1又は21で示されうる。野生型ヒトCpn10をコードするヌクレオチド配列は、配列番号2又は22で示されるか、又は配列番号2又は22とハイブリダイズするための十分な同一性を示すものでもよい。
本発明によれば、Cpn10ポリペプチドは、当技術分野に周知の組み換えDNA及び分子生物学における標準的な技術を用いて調製してもよい。例えば、Sambrookら、”Molecular Cloning:A Laboratory Manual”、Cold Spring Harbor、New York、1989年、及び、Ausubelら、”Current Protocols in Molecular Biology”、Greene Publ. Assoc. and Wiley−Intersciences、1992年、等の標準テキストからガイダンスを入手してもよい。Mortonら、2000年(Immunol.Cell Biol.、78巻、603−607ページ)、Ryanら、1995年(J.Biol.Chem.、270巻、22037−22043ページ)、及びJohnsonら、2005年(J.Biol.Chem.、280巻、4037−4047ページ)に記載の方法はCpn10ポリペプチド精製の適切な例であるが、当業者であれば、本発明は用いた精製法若しくは調製法、あるいは本発明の方法及び組成物に係るCpn10調製に利用しうる他のいかなる方法であってもよく、限定されないことを理解するであろう。Cpn10ペプチドは、endoLys−C、endoArg−C、endoGlu−C、及びstaphylococcus V8−protease等の1つ以上のプロテイナーゼによるタンパク分解により調製してもよい。消化されたペプチド断片は、例えば高速液体クロマトグラフィー(HPLC)法により精製してもよい。
本発明の実施形態は、上述のCpn10をコードする単離ポリヌクレオチド並びに当該ポリヌクレオチドの変異体及び断片を提供する。野生型Cpn10をコードするヌクレオチド配列は配列番号2又は22で示されるか、又は配列番号2又は22の配列とハイブリダイズするための十分な配列同一性を有する。
本発明は、本発明のCpn10ポリペプチドに選択的に結合する抗体、並びにその断片及びアナログを提供する。好適な抗体としてはポリクローナル、モノクローナル、キメラ、ヒト化、単鎖、Fab断片、及びFab発現ライブラリが挙げられるが、限定されない。本発明の抗体は、Cpn10ポリペプチド、その断片若しくはアナログのアゴニスト若しくはアンタゴニストとして作用してもよい。
抗Cpn10抗体そのもの以外にも、本発明のポリペプチド並びにその断片及び変異体は、Cpn10と相互作用する化合物又は薬剤のスクリーニング及び同定に特に有用である。とりわけ、望ましい化合物はCpn10の免疫調節活性を調節する化合物である。かかる化合物は、Cpn10の免疫調節活性を、活性化、増加、阻害又は抑制することを通じて調節する。適切な化合物は、Cpn10と直接的(例えば結合)又は間接的に相互作用することによって影響を及ぼす。
本発明のCpn10ポリペプチド及びポリヌクレオチドは、治療薬としても有用である。例えば、これらの分子の用途としては、被検者に対して治療的有効量の当該分子を投与して被検者の疾病又は症状の治療又は予防を行うことが挙げられる。典型的には、当該疾病及び症状は、被検者の免疫応答を調節することにより治療できるものである。当該疾病及び症状の例としては、急性又は慢性的炎症疾患、喘息、過敏症、多発性硬化症、GVHD及び感染症が挙げられるがこれらに限定されない。当該感染症は細菌又はウィルス感染により生じうる。従って、Cpn10ポリペプチド及びポリヌクレオチドを含んでなる、疾病又は症状の治療又は予防用の薬理学的に有用な化合物が考慮される。
表2に、遺伝子的パラメータ、具体的には下記のCpn10ポリペプチドの調製に使用される発現系(すなわちプラスミドの名称、抗生物質による選抜、及び宿主細胞)を記載する。
Cpn10の調製プロセスをさらに詳細に説明するため、Ala−Cpn10の調製プロセスを例示する。
Cpn10ポリペプチドの免疫調節活性を、基本的に国際特許出願PCT/AU2005/000041号に記載の方法に従い、HAW264−HIV−LTRルシフェラーゼバイオアッセイを用いて試験した。
分子シャペロンとして作用し、GroELと共にタンパク質をフォールディングするCpn10ポリペプチドの能力を、Weber F.及びHayer−Hartl M.K.(Chaperon in Protocols、Ed Schneider C.、Humana Press Inc.、2000年、117−126ページ)に記載の適切な方法を用い、インビトロでのロダネーゼのリフォールディングをアッセイすることによって測定した。未変性のウシのロダネーゼ(30μM、SIGMA)を、5M 塩酸グアニジン及び8mM DTTを含む20mM MOPS−KOH(pH7.5)、100mM KCl、及び20mM MgCl2溶液(バッファA)中で変性させ、次いでロダネーゼの最終濃度が400nMとなるように、変性剤からGroEL(400nM)を含むバッファA中に希釈した(75倍)。GroELは、迅速かつ安定に変性ロダネーゼ(D−Rho)と結合し、一方、バッファのみではD−Rhoはミスフォールディングし、凝集した(すなわち非効率的な自発的リフォールディング)。予め形成させた安定GroEL結合ロダネーゼ複合体に、Cpn10(下記参照)及びATP(20.6mM)を添加することにより、効率的にリフォールディングが進行した。Cpn10の非存在下でATPを添加すると、D−Rhoはフォールディング能力が喪失された状態でGroELへの着脱サイクルを生じさせ、結局はミスフォールディング及び凝集を生じさせた(この反応は適切なアッセイブランクとして有用である)。各々のフォールディング反応液は全体で290μLであり、所定の時点(すなわち0、15、30、45、60、75、90分)でアリコートを30μL採取し、70μLのロダネーゼ活性のアッセイ混合物(57.1mM KH2PO4(pH7.5)、71.4mM EDTA、71.4mM チオ硫酸ナトリウム、及び71.4mM KCN)と6分間混合した。ATPを用いたリフォールディング反応の開始前に採取したアリコート30μLを、T=0分のリフォールディング時点とした。ロダネーゼ活性アッセイ混合物中のEDTAはMg2+イオンをキレートするため、GroELのATP結合を妨げ、結果としてフォールディング反応が直ちに停止した。次いで、15体積%のホルムアルデヒドを50μL添加し(最終濃度5体積%)、6分後にロダネーゼ活性を停止させた。
組換え型大腸菌GroESを精製し、実質的にエンドトキシン汚染を含まない(0.14EU/mg)ことが示された(図3K参照)。精製GroESを、上記のように、Ala−Cpn10との比較で、RAW264.7−HIV−LTR−LUC阻害アッセイにおいて試験した。図2に示すように、試験した濃度(25−100μg/ml)のいずれにおいても、GroESは、HIV−LTRのLPS誘発活性を阻害しなかった。これらの結果は、Cpn10について観察された免疫調節活性が、実際の有意な生物学的効果であることを裏付ける。
(IMU23−25の部位特異的変異体)
可動ループ領域の疎水性IML部分(残基23−25)を変異導入し、Cpn10とCpn60との間の相互作用の強度(表1参照)を変化させた。IMLを、Cpn60との相互作用を撹乱させると予測される荷電トリペプチドEEEで置換し、また、IMLを、疎水性を高めることによってCpn10のCpn60との相互作用を潜在的に強化するものと予想されるIII又はIFI部分として変異させた。図3C、E、及びFに、Ala−Cpn10−EEE−cHis、Ala−Cpn10−IFI、Ala−Cpn10−IIIの精製を示すSDS−PAGEゲルを示す。
Ala−Cpn10を構成すると予測されるアミノ酸配列を、配列番号21に示す。Ala−Cpn10をコードする合成DNA配列(配列番号22)を、pPL550プラスミドのNcoI及びEcoRl部位に挿入した(Somodevilla−Torresら、2003年、Prot.Exp.Purif.、32巻、276−287ページ)。バッチCH001とCH003におけるAla−Cpn10の精製を示すSDS−PAGEゲルを、図3のAとBにそれぞれ示す。
Ala−Cpn10−cHisを構成すると予測されるアミノ酸配列を、配列番号41に示す。Ala−Cpn10−cHisをコードする合成DNA配列(配列番号42)を、終止コドン以外のAla−Cpn10のDNA配列(配列番号22)を、pET23aプラスミド(Novagen)のNdeI及びXhol部位に挿入して調製した。当該クローニングにより、Ala−Cpn10のC末端にヘキサヒスチジンのタグが付与される。図3Dに、Ala−Cpn10−cHisの精製を示すSDS−PAGEゲルを示す。
Cpn10の可動ループ領域(配列番号12)から16のアミノ酸を除去し、Ala−Cpn10−Δml(配列番号24)と称する86アミノ酸の変異体を作製した。Ala−Cpn10−Δmlをコードする合成DNA配列(配列番号25)を、pPL550プラスミドのNcoI及びEcoRI部位に挿入した(Somodevilla−Torresら、2003、Prot.Exp.Purif.32巻、276−287ページ)。可動ループはCpn10ポリペプチド鎖の中央に位置するため、可動ループの2つの残基(両端の1つの残基)を残してN末端フラグメントとC末端フラグメントを連結させた状態にすることで、適切なフォールディングと7量体の形成がなされた。

Cpn10−NtermESを構成すると予測されるアミノ酸配列を、配列番号14に示す。ヒトX−Cpn10の残基1−AGQAFRKFL−9(配列番号23)を、大腸菌GroESの残基1−MNIR4(配列番号11)で置換し、Cpn10−NtermESタンパク質を作製した。Cpn10−NtermESをコードする合成DNA配列(配列番号43)を、pET23aプラスミドに挿入した。図3Lに、Cpn10−NtermESの精製を示すSDS−PAGEゲルを示す。
可動ループのIMLトリペプチド(Cpn60との相互作用、さらにはタンパク質のフォールディングに極めて重要)の変異体を作製し、可動ループのCpn60との相互作用を撹乱又は強化した(実施例6参照)。
可動ループ領域(及び従ってCpn60)が免疫調節活性に必要でないことを確認するため、16アミノ酸を可動ループ領域(配列番号12)から欠失させ、Ala−Cpn10−Δmlと称する86アミノ酸の変異体を作製した(実施例6参照)。Ala−Cpn10−Δmlのアミノ酸配列を、配列番号24に示す。
Ala−Cpn10−Δmlを、Ala−Cpn10との比較において、RAW264.7−HIV−LTR−LUC阻害アッセイにおいて試験した。このアッセイにおいて、ルシフェラーゼレポーターはNFKBシグナル伝達と間接的に連結しており、NFKBはLPSによって誘導される一次転写因子である。ルシフェラーゼ活性は、使用した測定方法に応じて、相対光単位(RLU)又はカウント/秒(CPS)として測定した。図5A及び5Bに示すように、Ala−Cpn10−Δmlは、1−100μg/mlの濃度範囲で、LPS誘発によるHIV−LTRの活性化を阻害した。これは単独のアッセイであるが、2つの反復した実験を別なマイクロタイタープレートで行い、やはり同様の活性が示された。このデータセットにおいて、Ala−Cpn10−Δmlは、Ala−Cpn10と比較して一貫して大きい阻害活性を有することが観察された。
Cpn10のβ−ヘアピンルーフループ領域(図1参照)は、哺乳類において正味の正電荷を有するが、多くの細菌においては正味の負電荷を有する(大腸菌GroESが代表例)。興味深いことに、バクテリオファージT4もまた、大腸菌GroELと共にT4主要カプシドタンパク質Gp23をフォールディングする機能を有する固有のCpn10(Gp31)を持つ。なお、GroES及びCpn10はいずれもこの機能を有さない。Gp31とCpn10/GroESの間の主な相違点は、Gp31がルーフβ−ヘアピンループを完全に欠くことであり、Gp31の異常な機能及び性能の理由ともなりうる(Huntら、1997年、Cell、90巻、361−371ページ)。
上記実施例9及び10に記載のデータの裏付けとして、本発明者らは、可動ループとβ−ヘアピンルーフループの双方を欠くヒトAla−Cpn10変異体を作製した(「Ala−Cpn10−β−バレル」、配列番号28、実施例6参照)。興味深いことに、Ala−Cpn10−β−バレル変異体は、Ala−Cpn10とAla−Cpn10−Δml変異体に比較し、ゲル濾過クロマトグラフィーにおいて若干大きい分子として溶出された。このことから、当該ルーフがサブユニットを強固に結合した高次構造中に保持することを助長すると示唆される。一方、可動ループは7量体を不安定にしてその分解を促進するが、それにもかかわらず当該7量体は分解された個々のモノマーよりもエネルギー的に安定である。
Cpn10のN末端は、(サイトゾル中での合成後の)ミトコンドリアマトリクスのターゲティングを補助することが知られている。しかしながら、殆どのミトコンドリアマトリクスのタンパク質は、切断可能なN末端のターゲティング配列を有するものの、Cpn10のN末端は切断されず、さらなる機能を有することを示唆する。
X−Cpn10を、Ala−Cpn10との比較において、RAW264.7−HIV−LTR−LUC阻害アッセイを用いて試験した。実施例6で議論したように、X−Cpn10は、追加的なN末端アラニン残基及び天然ヒトCpn10に存在するアセチル基を欠く。このアッセイにおいて、ルシフェラーゼレポーターを、NFkBシグナル伝達と間接的にリンクさせた。NFkBは、LPSによって誘発される一次転写因子である。ルシフェラーゼ活性は、使用した測定方法に応じて、相対光単位(RLU)又はカウント/秒(CPS)として測定した。図9A及び9Bに示すように、X−Cpn10は、HIV−LTRのLPS誘発活性を部分的に阻害した(Ala−又はGly−Cpn10活性の約50%)。このデータセットにおいて、アラニン等のCpn10のN末端上の付加的な残基は、TLR4シグナリングの免疫調節活性に関与しうることが観察された。
Gly−Cpn10を、Ala−Cpn10との比較において、RAW264.7−HIV−LTR−LUC阻害アッセイを用いて試験した。実施例6で議論したように、Gly−Cpn10は、Ala−Cpn10の追加的なN末端アラニン残基をグリシン残基で置換している。このアッセイにおいて、ルシフェラーゼレポーターを、NFkBシグナル伝達と間接的にリンクさせた。NFkBは、LPSによって誘発される一次転写因子である。ルシフェラーゼ活性は、使用した測定方法に応じて、相対光単位(RLU)又はカウント/秒(CPS)として測定した。図10A及び10Bに示すように、Gly−Cpn10は、HIV−LTRのLPS誘発による活性化を、Ala−Cpn10よりも大きい規模で阻害した。
図5−7及び10に示すデータは、上記のRAW264.7−HIV−LTR−LUC阻害アッセイにおいて、粗LPSを用いることによって得た。図11−14では、TLR4に特異的な超高純度LPSを使用した結果を示す。図11(Ala−Cpn10Δml)、図12(Ala−Cpn10Δroof)、図13(Ala−Cpn10−β−バレル)、及び図14(Gly−Cpn10)の結果は、図5−7、及び10において示した対応するデータと非常に類似するものである。このことは、Cpn10による免疫調節活性を解析するために本願明細書において使用され開示されたアッセイデータが、TLR4に特異的に由来するデータであることを示す。
種々のCpn10ポリペプチド(すなわち、Ala−Cpn10−Δml、Ala−Cpn10−Δroof、及びX−Cpn10)のインビトロ免疫調節活性が、インビボ活性を反映するか否かを解析するため、敗血症モデルマウスを用い、内毒血症の試験を行った。
以下のマウス内毒血症の試験に使用した材料及び方法を、下記の実施例17a(1)〜17(a)2に記載する。
本試験は、84匹のメスBalb/cマウスを用いて行った。いずれも成体であり(>9週齢、平均体重約20g(0.02kg))、1つの群あたり7匹のマウスとして、12群に分けた(表5参照)。マウスを12/12の明/暗サイクル条件下において収容し、標準的な実験動物用飼料(Specialty Feeds、Glen Forrest、オーストラリア)及び水を自由に摂取させた。各マウスの体重を注射の前に測定した。下記に示す要領(表5参照)で、尾静脈からの静脈(IV)ルートを経て注射を行った。
実施例16a(1)マウス内毒血症の試験に使用した薬剤/溶液を以下((A)から(H)に掲げる)に示す。(A)タンパク質製剤バッファ(FB):このバッファは、本試験のネガティブコントロールであり、50mMのトリスHCl(pH7.6)と150mM NaClを含む(<0.02EU/ml)。このバッファは、試験物質として、またポジティブコントロール及び試験サンプルの希釈用液として使用する。(B)Ala−Cpn10:このCpn10ポリペプチドは、本試験のポジティブコントロールであり、5mg/mlのストック濃度を有する(<0.01EU/mg)。400μlのタンパク質溶液を1.6mlの製剤バッファ中に希釈することによって、1mg/mlの試験用溶液を調製した。(C)Ala−Cpn10−Δml:このCpn10ポリペプチドは、3.5mg/ml(<0.03EU/mg)のストック濃度を有する。571μlのタンパク質溶液を1.429mlの製剤バッファ中に希釈することによって、1mg/mlの試験用溶液を調製した。(D)Ala−Cpn10−Δroof:このCpn10ポリペプチドは、4.2mg/mlのストック濃度を有する(<0.1EU/mg)。タンパク質溶液477μlを1.523mlの製剤バッファ中に希釈することによって、1mg/mlの試験用溶液を調製した。(F)X−Cpn10:このCpn10ポリペプチドのストック濃度は5mg/mlである(<0.04EU/mg)。400μlのタンパク質溶液を1.6mlの製剤バッファ中に希釈することにより、1mg/mlの試験用溶液を調製した。(G)エンドトキシン:リポ多糖(LPS)をSigma Chemical Company(Cat.No.L6529)から入手した。使用の直前に、バイアルの内容物(1mg)を1mlの無菌の生理食塩水中で再調製した。各群への注射の前に、当該内容物を無菌の生理食塩水中に100μg/mlまでさらに希釈(1/10)した。(H)エンドトキシンコントロール:注射用の無菌生理食塩水を、900mg/ml(0.9%)の濃度で、Pfizer、オーストラリア(Cat. No. DW−SC0010)から入手した(<0.01EU/ml)。
マウスの異なる群における薬剤投与の手順は、表5に示す通りである(下記参照)。投与はいずれも、ある拘束したマウスに対する意識下での100μlの尾静脈注射により行った。LPS用量はいずれも10μg/マウスとした。Cpn10変異体はいずれも100μg/マウス(体積100μL)で注射した。
炎症関連のサイトカインのレベルにおける変化を評価するため、血清サンプルについてマウスの炎症CBA分析(Cat#552364、BD Biosdences)を行った。すなわち、LPS投与マウス(表5、群1、3、5、7、9、及び11)又は生理食塩水対照マウス(表5、群2、4、6、8、10、及び12)から調製したTNFα、IL6、IL−10、MCP−1、IL12p70、IFN−γ血清を、必要により、分析前にアッセイ希釈剤に希釈した(LPS処理群について5倍、生理食塩水対照について2倍)。各サンプルを、CBAソフトウェアを備えたBD FACS−Array装置を用い、メーカーの使用説明書に従って、2回試験した。
下記の式を使用し、LPS感作マウスに及ぼすCpn10処理の効果を測定した。Cpn10変異体で予備処理したマウス(すなわち試験群)の予備処理していないマウス(すなわち対照群)に対するLPS誘発サイトカインの低下率を、次の式に従って計算した。
低下%=100−[(試験群の平均サイトカインレベル/対照群の平均サイトカインレベル)×100]
CBA分析を裏付けるため、メーカーの使用説明書に従ってR&D Systems Duoset ELISAキット(Cat#DY410)を用い、マウスTNF−α ELISAを行った。希釈剤としてPBS+10%FCSを用い、サンプルとスタンダードを希釈(3倍希釈)した。サンプルを2回試験した。
表5に記載の種々の群のマウスの挙動を、LPS又は生理食塩水の注射後90分間以内又は心臓穿刺による出血の前に解析した。表6に全ての観察を記録してまとめた。観察は、LPS/生理食塩水の注射の直後と心臓穿刺(C.P.)によるマウスの出血の前に行った。臨床的観察の比較は、群1、2、又は3に関して行った。通常、LPSのみで処理したマウス(群1)は、注射後15分間以内にLPS誘発による敗血症を示した。LPS処理のマウスでは移動性の低下を示し、刺激(例えば騒音又は接触)に対する応答が少なかった。下痢や毛の乱れのようなLPS感作によるいくつかの典型的な悪影響は、本試験においてはいずれのマウスにも観察されなかった。このことは、この試験に使用したLPSのロットが相対的に高い効果を有することを反映していると考えられる。生理食塩水処理の対照マウス(群2)は、刺激に対する応答が大きく、通常の応答性と移動性を示した。
LPS感作マウス(図15参照)におけるTNF−α、IL−6、及びIL−10サイトカインの平均レベルを示す。Cpn10で処理しなかった動物に対して炎症促進性サイトカインの低下において統計的有意性を示すCpn10処理群を星印で表示する。予備処理しなかったマウス(表5に示す)と比較した、種々のCpn10で予備処理したマウスにおいて分析した種々のサイトカインの低下率を括弧内に表示する。
各マウスからの血清サンプルを、循環炎症促進性サイトカインの検出用のBD Mouse炎症CBAアッセイを用いて分析した。このアッセイは、試験サンプル中のTNFα、IL−6、IL−10、IL−12p70、MCP−1、及びIFN−γサイトカインを検出する。分析は、アッセイの限界内での最適な検出のために、連続的に希釈したサンプル(実施例16a参照)について行った。全てのサンプルを2回試験した。
本試験は、種々のCpn10ポリペプチド(すなわち、Ala−Cpn10、Ala−Cpn10−Δml、Ala−Cpn10−Δroof、及びX−Cpn10)によるマウスの予備処理が、LPS誘発による内毒血症の臨床的効果を低下することを示した。LPSのみを投与されたマウスでは、内毒血症の典型的な症状(すなわち活動、応答、及び俊敏さの低下)を示した。他方で、いずれかのCpn10タンパク質で予備処理したマウスではLPS感作による影響が少ない(すなわち刺激に対する高い応答性、及び機動性)ことが観察された(表6)。
Cpn10ポリペプチド調製用のバイオプロセスを開発した。下記に示すように、このプロセスを用いて、100Lの大腸菌培養液から約20gの組換え型ヒトCpn10(>99%純度、≦0.03EU/mgのエンドトキシン、及び≦155.3pg/mgのDNA)を調製した。このプロセスを以下に概説する。
AlaCpn10_pPL550プラスミドを有する大腸菌XL1−Blue株を含むバイアルを、マスターセルバンク(上記の「一般的方法」参照)から取り出し、30℃にて抗生物質を添加せずにSoya Brothで一晩「前培養」した。次に、上記の培地と成長温度のパラメータに設定した100Lのバイオリアクター用の種培養液を調製した。この種アリコートを、動物由来の生成物を含まず、かつ抗生物質を添加しない、大豆ベースのペプトンリッチな最少培地(BresaGen、SA、オーストラリア)100Lを仕込んだバイオリアクター中に添加した。バイオリアクターの培養ではバッチのフィードを必要とせず、培養温度を増殖期全体にわたり30±1℃に維持した。アンモニアを添加してpHを7.0±0.2に維持した。Cpn10の誘導は、温度を42℃にシフトさせてOD600値=10の時点で行い、42±0.1℃でさらに3時間インキュベートし、OD600値が20〜25に到達した時に終了させた。
培養終了後3時間以内に、細菌細胞(約3.5kgの湿重量)を遠心分離(5,000×g)してペレットを調製し、25mM トリスHCl(pH8.0)中に再懸濁し、APV Gaulin Model 30CD圧力ホモジナイザー(APV、USA)を48.2MPa(7000psi)で3回通すことによって溶解させた。細胞残渣を沈殿させた後、細菌可溶化液に含まれる可溶性Cpn10を、フロースルーWestfalia MSB−7遠心分離機を用いて回収した。約20Lの透明な可溶化液を4℃で一晩保存した。
Big Bead Sulfopropyl−Sepharose(BBSP)カチオン交換、Q−Sepharose Fast Flow(QFF)アニオン交換、及びHigh Performance Sepharose SP(HPSP)カチオン交換クロマトグラフィーを含んでなる3つの後続プロセスを行い、Cpn10を精製した。K−prime 40−II Bioprocess Unit (Millipore)を用い、クロマトグラフィーを行った。
全てのイオン交換クロマトグラフィーのカラムを1M NaOHで洗浄して脱パイロジェンし、溶出液が所定のバッファのpHと伝導率となるまでバッファで平衡化した。バッファの保存、及び種々の精製工程におけるカラム溶出液の貯留に使用した全ての容器はパイロジェンを含有しなかった。精製に使用した全てのバッファは、注射用蒸留水(WFI)を用いて調製した。
25mM トリスHCl(pH8.0)(バッファA)を用い、毎時75cmの一定流速、樹脂1Lあたり10g以下のCpn10のローディング速度で、予め平衡化したBBSPカチオン交換カラム(BPG300/500、8.6LBBSP SP−Sepharose resin、GE Biosciences)に20〜40分間かけて可溶化液を添加した。バッファAで洗浄した後、捕捉されたCpn10を150mM NaClを含むバッファAを用いて溶出し、1Lの画分を回収した。SDS−PAGE分析は、BBSPプールが95%超の純度であることを示した。
BBSP溶出液を、室温にて15倍容のバッファAで2回バッファ交換して脱塩した。第1の透析工程を2〜3時間実施した後、新たなバッファAのタンクに移し、一晩透析を行った。透析バッグからタンクバッファへのNaClの再分配は、較正済のスタンダードに対する透析液の導電率を測定することによってモニターした(Cyberscan 100、Eutech Instruments、シンガポール)。透析したBBSPプールは、毎時75cmの線流速、1Lの樹脂あたり15g以下のCpn10の負荷割合で、バッファAの中で予め平衡化した4.7LのQFFアニオン交換樹脂(GE Biosciences)を充填したBPG 200/500カラムに添加した。Cpn10を含有するQFFアニオン交換のフロースルー画分を回収した。ローディング条件下では、大腸菌細胞タンパク質、核酸、及びエンドトキシンの大部分はマトリクスに依然結合していた。
QFFフロースルーを、1.67LのSPHP樹脂(GE Biosciences)を充填したBPG100/500カラム(50mMトリスHCl(pH7.6)(バッファB)で予め平衡化)を用い、樹脂1Lあたり15〜20gのCpn10を添加した。結合したCpn10を、15Lの0〜120mMのNaCl(直線勾配)で溶出し、0.5Lの画分を回収した。この画分を、限外濾過クロマトグラフィー(SEC)、HPLC、及びSDS−PAGE分析に従ってプールした。Cpn10タンパク質とNaClイオンの濃度を、それぞれ、280nmでのUV吸光度とイオン選択電極法(IDEXX、オーストラリア)によって測定した。
プールしたSPHP画分の伝導率測定とNa+及びCl−イオン測定の組み合わせに基づいて、バッファを150mM NaClを含む50mMのトリスHCl(pH7.6)となるように最終調整した。調製したCpn10を、無菌条件下で0.2μmのフィルタを通して濾過した。濾過した溶液を、パイロジェンを含まない500mlプラスチックボトル中に、各ボトルあたり500mlずつ、合計2.5gのCpn10となるように分配した。
大腸菌細胞の可溶化液とクロマトグラフィーの画分のSDS PAGE分析を、メーカーの手順書に従いNuPAGE 4−12% Bis−Tris勾配ゲル(Invitrogen、Carlsbad、米国カリフォルニア州)を用いて行った。ゲルをクーマシーブリリアントブルー染色し、各ゲル中にCpn10タンパク質と分子量マーカーが含まれるようにした。
精製Cpn10の濃度を、0.353mg−1mL−1cm−1の吸光係数を用い、280nmにおいてUV吸光度(BioRad SmartSpec−3000分光光度計)により測定した。BioRad SmartSpec−3000は通常、ウシ血清アルブミン(Pierce)に基づいた場合に0.59のA280nm値を示すが、文献値は0.67である(Pierce technical resource No.TR0006.0)ことに留意すべきである。
本発明に係る分子及び薬剤、並びに本発明の方法により同定される物質を用いて、種々の疾病の状態及び症状の治療又は予防を行ってもよい。かかる分子及び薬剤は、単独で投与してもよいが、典型的にはそれらは医薬組成物として投与する。
無菌緩衝水1mL及び適切な薬剤又は分子1mgを含む態様で筋肉内注射用の組成物を調製した。
10体積%のポリエチレングリコール及び水に、1重量%の適切な薬剤又は分子を混合することにより、注射投与に適する組成物を調製した。この溶液は、濾過滅菌することができる。
カプセル形態に適切な薬剤又は分子の組成物を、標準的な2片の硬ゼラチンカプセル内に、粉末の形態の薬剤又は分子50mg、ラクトース100mg、タルク35mg、及びステアリン酸マグネシウム10mgを充填して調製した。

- 単離されたシャペロニン10ポリペプチドであって、対応する野生型シャペロニン10ポリペプチドと比較してルーフβ−ヘアピン領域に又は複数のアミノ酸置換、欠失及び/又は付加を含んでなる、免疫調節活性を有する、ポリペプチド。
- 単離されたシャペロニン10ポリペプチドであって、対応する野生型シャペロニン10ポリペプチドと比較して、可動ループ領域及びルーフβ−ヘアピン領域を両方とも実質的に欠いている、ポリペプチド。
- 配列番号9又は28で示されるアミノ酸配列を含んでなる、請求項21に記載のポリペプチド。
- 配列番号10又は29で示されるヌクレオチド配列でコードされる、請求項21に記載のポリペプチド。
- 少なくとも対応する野生型シャペロニン10ポリペプチドの免疫調節活性のレベルと同等の免疫調節活性を示す、請求項21又は請求項22に記載のポリペプチド。
- 前記ポリペプチドは、対応する野生型シャペロニン10ポリペプチドと比較して追加的なN末端アラニン残基の欠失を含んでなる、単離されたシャペロニン10ポリペプチド。
- 配列番号23で示されるアミノ酸を含んでなる、請求項25に記載のポリペプチド。
- 配列番号44で示されるヌクレオチド配列でコードされる、請求項25に記載のポリペプチド。
- 対応する野生型シャペロニン10ポリペプチドの免疫調節活性のレベルと比較して減少したレベルの免疫調節活性を示す、請求項25又は請求項26に記載のポリペプチド。
- 前記ポリペプチドのN終端は実質的にバックテリア性シャペロニン10N終端で置換される、単離されたシャペロニン10ポリペプチド。
- 配列番号14で示されるアミノ酸を含んでなる、請求項29に記載のポリペプチド。
- 配列番号43で示されるヌクレオチド配列でコードされる、請求項29に記載のポリペプチド。
- 対応する野生型シャペロニン10ポリペプチドの免疫調節活性のレベルと比較して減少したレベルの免疫調節活性を示す、請求項29又は請求項30に記載のポリペプチド。
- 対応する野生型シャペロニン10ポリペプチドと比較して、グリシン残基が前記ポリペプチドの追加的なN末端アラニン残基を置換する、単離されたシャペロニン10ポリペプチド。
- 配列番号30で示されるアミノ酸配列を含んでなる、請求項33に記載のポリペプチド。
- 配列番号31で示されるヌクレオチド配列でコードされる、請求項33に記載のポリペプチド。
- 少なくとも対応する野生型シャペロニン10ポリペプチドの免疫調節活性のレベルと同等の免疫調節活性を示す、請求項33又は請求項34に記載のポリペプチド。
- 請求項1から請求項36のいずれかに係るシャペロニン10ポリペプチドをコードする、単離された核酸。
- 適宜、又は複数の調節配列に操作可能にリンクした請求項37に係る核酸を含んでなる、発現コンストラクト。
- 請求項1から請求項36のいずれかのポリペプチドを発現するか、又は請求項37の核酸又は請求項38の発現コンストラクトを含んでなる、ホスト細胞。
- 請求項1から請求項36のいずれかのポリペプチドと選択的に結合する抗体。
- 請求項1から請求項36のいずれかのポリペプチド、請求項37の核酸、請求項38の発現コンストラクト又は請求項40の抗体を含んでなる、医薬品組成物。
- 又は複数の追加薬剤を含んでなる、請求項41に記載の医薬品組成物。
- 請求項1から請求項36のいずれかのシャペロニン10ポリペプチド又は請求項37の核酸の有効量を被検者に投与するステップを含んでなる、被検者の治療方法。
- 前記治療は前記被検者の免疫応答を調節する、請求項43に記載の方法。
- 前記免疫応答はトール様受容体シグナリングの調節により調節される、請求項44に記載の方法。
- 前記方法は、請求項1から請求項36のいずれかのシャペロニン10ポリペプチド又は請求項37に記載の核酸の有効量を被検者に投与することを含んでなる、被検者の疾患又は症状の治療又は予防方法。
- 前記疾患、障害又は症状は、急性又は慢性炎症性疾患、喘息、アレルギー、多発性硬化症、GVHD、他の自己免疫疾患又は感染症から選択される、請求項45に記載の方法。
- 前記感染症は細菌又はウィルス感染である、請求項47に記載の方法。
- 前記細菌感染症はグラム陰性細菌により生じる、請求項48に記載の方法。
- 患者又は少なくとも1つの細胞、組織又はこれらの臓器内でTLR4シグナリングを調節する方法であって、請求項1から請求項36のいずれかのシャペロニン10ポリペプチド又は請求項37に記載の核酸の有効量を被検者に投与することを含んでなる、方法。
- 前記シャペロニン10ポリペプチドはアゴニスト誘導TLRシグナリングを調節する、請求項50に記載の方法。
- 患者又は少なくとも1つの細胞、組織又はこれらの臓器内で又は複数の免疫調節剤の生産及び/又は分泌を調節する方法であって、請求項1から請求項36のいずれかのシャペロニン10ポリペプチド又は請求項37に記載の核酸の有効量を被検者に投与することを含んでなる、方法。
- 前記ポリペプチドはTLR4シグナリングを調節する、請求項52に記載の方法。
- 免疫調節剤は炎症誘発性サイトカイン又はケモカイン、若しくは抗炎症性サイトカイン又はケモカインである、請求項52又は請求項53に記載の方法。
- 前記ケモカイン又はサイトカインがTNF−アルファ、IL−6、RANTES、TGF−β又は1型インターフェロンから選択される、請求項54に記載の方法。
- 請求項1から請求項36のいずれかのポリペプチドに結合する化合物を同定する方法であって、
(a)候補化合物を前記ポリペプチドに接触させるステップ、及び
(b)前記候補化合物及び前記ポリペプチドの錯体生成を試験するステップ、を含んでなる方法。 - 前記錯体生成を試験するステップが、競争結合試験又はツーハイブリッド試験である、請求項56に記載の方法。
- 請求項1から請求項36のいずれかのポリペプチドの活性を調節する化合物をスクリーニングする方法であって、
(a)前記候補化合物の前記ポリペプチドに対する好適な相互作用を可能にする条件下において、前記ポリペプチドを候補化合物に接触させるステップ、及び
(b)前記ポリペプチドの活性を試験するステップ、を含んでなる方法。 - 前記ポリペプチドの活性を試験するステップが、標識した基質を添加するステップ及び前記標識した基質における変化を計測するステップを含んでなる、請求項58に記載の方法。
- 前記ポリペプチドがヒトシャペロニン10ポリペプチドであり、並びに配列番号1又は21で示されるアミノ酸配列を含んでなる、請求項1から請求項59のいずれかに記載の、対応する野生型シャペロニン10ポリペプチド。
- 前記ポリペプチドが配列番号2又は22で示されるヌクレオチド配列でコードされる、請求項60に記載の、対応する野生型シャペロニン10ポリペプチド。
