Abstract
Description
第1の観点では、骨量減少の予防又は治療のための医薬品の製造における、BiP、又はその変異体、相同体、誘導体、若しくは断片の使用を提供している。
好ましくは、本発明は骨量減少に対する医薬品の製造におけるBiPの使用に関する。

骨量減少に関連する又は関係する疾患としてとしては特に制限されないが、パジェット病、原発性骨粗しょう症及び続発性骨粗しょう症、閉経後骨粗しょう症、老人性骨粗しょう症、グルココルチコイド誘発性骨粗しょう症、歯周病、歯槽骨の喪失、骨切り術後の骨量減少及び小児突発性骨量減少(childhood idiopathic bone loss)、骨粗しょう症の長期合併症(例えば、脊椎の湾曲及び身長の低下)、補綴手術、並びに股関節、膝関節等の人工関節の弛緩を挙げることができる。
別の観点では、本発明は、骨量減少又は骨吸収に起因する又は関連する疾患又は症候群の診断方法に関する。
本明細書において「サンプル」という用語は、その自然な意味を有する。
別の観点では、本発明は骨破壊又は骨量減少を調節する薬剤を同定するアッセイを提供する。
BiP並びに骨吸収の調節及び破骨細胞成熟の調節との関係において、「調節」という用語は、好ましくは、BiPを用いた破骨細胞の成熟及び骨吸収の予防、抑制、緩和、減少、阻害、又は予防を意味する。

本発明はBiPの断片、変異体、相同体、及び誘導体の使用を包含する。
さらに、本発明はBiPをコードするヌクレオチド配列をインビボで調節することを特徴とする遺伝子治療に関する。例えば、コーディングヌクレオチド配列、又はBiPのコーディングヌクレオチド配列に関連する調節領域、又はその対応するRNA転写産物に結合する薬剤を投与して、転写速度又は翻訳速度を調節(好ましくは促進)することで、発現を調節することができる。

[実施例]
BiP、すなわちグルコース制御性タンパク質78(grp78)は、タンパク質の正確なフォールディングを担う機能のある小胞体(ER)シャペロンである。グルコース飢餓、低酸素、又は活性酸素種によってERの機能が妨害され、折り畳まれていないタンパク質が蓄積されると、BiPのアップレギュレーションが誘導される。これらは炎症関節においてよく見られ、関節リウマチ(RA)の発症に影響する因子でもある。
免疫磁気ビーズを用いたネガティブアイソレーション(negative isolation)によって単球を精製した。BiP(20μg/ml)で24時間刺激した又は刺激しなかったこれらの細胞からトータルRNAを単離した。次いで、RNAをアフィメトリクス遺伝子アレイ(U95 バージョン2)又はR&Dサイトカイン遺伝子アレイ、にメーカーの説明書に従い用いた。1つの対象由来のサンプル2連(duplicate)をアフィメトリクスアレイに用い、2つの正常コントロール対象由来サンプル2連をR&D遺伝子アレイに用いた。
結果を表1及び図6に示す。
BiPは、炎症反応中に単球において活性化される多くの遺伝子のダウンレギュレショーンを誘導した。これらの遺伝子には、抗原提示、接着及び細胞遊走、細胞シグナル伝達、並びに破骨細胞分化に関するものが含まれる。R&Dサイトカイン遺伝子活性化アレイを用いて得られた結果とアフィメトリクスアレイを用いて得られた結果は一致していた。
BiPは、受容体様分子を介してヒトの単球に結合することが示され、この受容体様分子は、HSP60又は70及びgrp94が利用しているものとは異なるようである。単球のBiP活性化の結果を調べるために2種類の遺伝子アレイを用いた。
全てのインビトロ実験はヒト細胞及びマウス細胞を用いて行った。単核球を単離するために、ヒト細胞を末梢血から密度遠心(Corrigall VM, Solau-Gervais E, Panayi GS. Lack of CD80 expression by fibroblast-like synoviocytes leading to anergy in T lymphocytes. Arthritis Rheum 2000; 43(7):1606-1615)で分離する。マウス細胞は、以前に記載の通りに(Li X, Udagawa N, Takami M, Sato N, Kobayashi Y, Takahashi N. p38 Mitogen-activated protein kinase is crucially involved in osteoclast differentiation but not in cytokine production, phagocytosis, or dendritic cell differentiation of bone marrow macrophages. Endocrinology 2003; 144(11):4999-5005)、5〜8週齢成獣マウスの大腿骨及び脛骨から単離した。TRAP陽性の細胞数(マウス)、又はビトロネクチン受容体(VnR)及び/若しくはFアクチンリングの発現陽性の細胞数(ヒト)によって、両方の系における破骨細胞分化を評価した。象牙質切片上で細胞を培養した後の吸収窩形成を定量することで、両方の系における吸収機能を評価する。実験条件の下、4〜5日後(マウス)又は10〜12日後(ヒト)に破骨細胞培養を回収し、マウス破骨細胞及びヒト破骨細胞に対してそれぞれ7日後及び14日後に吸収アッセイを行う。
これは、細胞表面から転写因子(例えば、RANK及びc−Fms、TRAFs、c−Fos、NFATc1、MAPK[p38、jun、及びERK1/2]、IKK及びNF−κβ、並びにAkt)までの間の分子のリン酸化及び活性化を阻害することで直接調べるか、又はIL−10などの抑制性サイトカインの放出によって間接的に調べる。
卵巣摘出CD1マウスは自発的に破骨細胞を過剰産生し、骨粗しょう症様疾患を発症する。これを、疾患の異なる段階でBiPで治療する。
BiPは、破骨細胞活性化が見られるリウマチ性疾患の新規な生物学的治療法である。ヒト及びマウスのインビトロ破骨細胞形成系における破骨細胞形成をBiPが阻害するという考えを支持するインビボ及びインビトロ実験の証拠が提供されている。

Claims (31)
- 骨量減少を予防又は治療するための医薬品の製造における、BiP、又はその変異体、相同体、誘導体、若しくは断片の使用。
- 骨吸収を予防又は治療するための医薬品の製造における、BiP、又はその変異体、相同体、誘導体、若しくは断片の使用。
- 骨量減少又は骨吸収が、筋骨格系疾患に関連していることを特徴とする、請求項1又は2に記載の使用。
- 筋骨格系疾患が骨粗しょう症であることを特徴とする、請求項3に記載の使用。
- 破骨細胞成熟の調節におけるBiPの使用。
- 調節が、破骨細胞成熟の阻害、減少、又は予防であることを特徴とする、請求項5に記載の使用。
- 破骨細胞成熟の調節がインビボ又はインビトロで行われることを特徴とする、請求項5又は6に記載の使用。
- 破骨細胞が破骨細胞前駆体、多核前駆体、又は成熟破骨細胞であることを特徴とする、請求項5〜7のいずれかに記載の使用。
- 有益な予防効果又は治療効果を生じさせるために、BiP、又はその変異体、相同体、誘導体、若しくは断片を投与することを含む、骨量減少の予防方法又は治療方法。
- 有益な予防効果又は治療効果を生じさせるために、BiP、又はその変異体、相同体、誘導体、若しくは断片を投与することを含む、骨吸収の予防方法又は治療方法。
- 破骨細胞を、BiP、又はその変異体、相同体、誘導体、若しくは断片と接触させることを含む、破骨細胞発達の調節方法。
- 調節が、破骨細胞発達の阻害、減少、又は予防であることを特徴とする、請求項11に記載の方法。
- 方法がインビボ又はインビトロで行われることを特徴とする、請求項11又は12に記載の方法。
- (a)対象中のBiP活性を検出するステップ、
(b)BiP活性を、罹患していないコントロールのBiP活性と比較するステップ、及び
(c)コントロールから得られた値と対象から得られた値とを比較するステップ、
を含む、骨量減少に起因若しくは関連する疾患又は症候群の診断方法であって、コントロールから得られた値と対象から得られた値との差が、対象が疾患又は症候群に罹患していることの指標となることを特徴とする、前記方法。 - BiP活性が、コントロールから得られた値と比較して低下していることを特徴とする、請求項14に記載の方法。
- 破骨細胞発達又は骨吸収を測定する任意のステップを含む、請求項14又は15に記載の方法。
- (i)BiP活性を調節する薬剤を選択するステップ、及び
(ii)前記薬剤存在下における破骨細胞成熟を測定するステップ、
を含む、骨破壊又は骨量減少を調節する薬剤を同定するためのアッセイ方法であって、薬剤非存在下における破骨細胞成熟と薬剤存在下における破骨細胞成熟との差が、骨破壊又は骨量減少を調節する薬剤の指標となることを特徴とする、前記方法。 - 薬剤がBiP活性を増大又はアップレギュレートすることを特徴とする、請求項17に記載のアッセイ方法。
- (a)請求項17又は18に記載のアッセイ方法を行うステップ、
(b)骨破壊又は骨量減少を調節する1又は複数の薬剤を同定するステップ、及び
(c)これらの同定された1又は複数の薬剤を大量に調製するステップ、
を含むプロセス。 - 請求項17又は18に記載のアッセイ方法により得られる薬剤。
- (a)破骨細胞分化の1又は複数の異なる段階にある、1又は複数の破骨細胞に、BiPを添加するステップ、及び
(b)破骨細胞形成に対するBiPの効果を決定するステップ、
を含む、破骨細胞分化に対するBiP、又はその変異体、相同体、誘導体、若しくは断片の効果を決定する方法。 - BiPが0.02〜20μg/mlの濃度で添加されることを特徴とする、請求項21に記載の方法。
- TRAP活性、VnR、及び/又はFアクチンリングを測定することによって破骨細胞数を定量することを特徴とする、請求項21又は22に記載の方法。
- 骨吸収を定量する任意のステップを含む、請求項21〜23のいずれかに記載の方法。
- 1又は複数の破骨細胞特異的マーカーの発現をPCRを用いて測定することで、破骨細胞分化を確認することを特徴とする、請求項21〜24のいずれかに記載の方法。
- 破骨細胞特異的マーカーがカルシトニン受容体遺伝子又はカテプシンK遺伝子であることを特徴とする、請求項25に記載の方法。
- (a)BiPの存在下及び非存在下において破骨細胞を分化させるステップ、及び
(b)BiP非存在下において分化させた破骨細胞と比べて、BiP存在下で分化させた破骨細胞中で異なる発現をしている1又は複数のタンパク質を同定するステップ、
を含む、破骨細胞分化中にBiPによって調節されている1又は複数のタンパク質を同定する方法。 - タンパク質がシグナル伝達分子又は転写因子であることを特徴とする、請求項27に記載の方法。
- 培養の始めの24〜72時間、又は多核破骨細胞が存在する、培養の終わりの方の24〜72時間、BiPを添加することを特徴とする、請求項27又は28に記載の方法。
- タンパク質又はタンパク質をコードする核酸を、qt−RT−PCR及び/又はウェスタンブロットで検出することを特徴とする、請求項27〜29のいずれかに記載の方法。
- 骨粗しょう症の予防又は治療のための医薬品の製造における、BiP、又はその変異体、相同体、誘導体、若しくは断片の使用。
