Mass Spectrom. 10:889−896)。サンプルは、金でコーティングしたホウケイ酸ガラス毛細管(Protana A/S、デンマーク)から噴霧される。衝突性解離(CID)のために、適切な衝突ガス、例えば、アルゴン(AGA、スウェーデン)が用いられる。Aβペプチドの各種形態の、溶液から消滅する(例えば、α−ヘリックス形の消滅)相対速度を定量するために、好適な獲得範囲、例えば、m/z 135−4000が選択される。5秒間持続の300走査を各時点で記録し、合わせて一つのスペクトラムとする。単一プロトン化、および複数プロトン化、例えば二重および三重プロトン化分子の合計量に対応するイオン電流が、最大エントロピーソフトウェア(Micromass)によって求められる。ポリペプチドの構造を確かめるためにCIDスペクトラムが記録される。H/D交換では、ES質量分析スペクトラムを連続記録することによってH/D交換反応の速度が監視される。特定の時点で、対象とするタンパクのH/D交換形に対応するイオンに対しCIDを実施する。交換されたプロトンの場所は、得られた断片化スペクトラムによって示される。
MALDI質量分析スペクトラムは、例えば、陽イオンモードで操作されるVoyager−DE.TM.PRO Biospectrometry Workstation (PerSeptive Biosystems Inc.)で記録することが可能である。H/D交換スペクトラムの記録のために、CH3CN/D2Oの溶液から調製し、あらかじめ乾燥させたα−シアノ−4−ヒドロキシケイ皮酸(20μg)を含む表面(例えば、100ウェルプレート)において重水化または非重水化ポリペプチドを乾燥させる。装置は335nmレーザーを備え、遅延抽出採用の直線モードで操作する。適切な獲得範囲(例えば、m/z 3500−4000)を使用し、外部較正(例えば、m/z 3660.19と5734.58の間)を用いる。スペクトラムは、獲得数(例えば、400回の獲得数)の平均として計算され、一般に、各時点について3重サンプルが記録される。各時点において重水によって交換されたプロトンの数は、重水化溶媒におけるペプチドの質量から、プロトン化溶媒におけるペプチドの質量を差し引くことによって計算される。これは、例えば、現在進行中の分子内ジスルフィド形成を補正するために行われる。このような分子内ジスルフィド形成は、ES CIDによって観察が可能である。ESと組み合わせたMALDIによって測定されるH/D交換速度を比較する実験では、同一の溶液をMALDIプレートに滴下し、ES毛細管から噴霧する。
質量分析データから速度定数の推定が、例えば、Matlab.RTMバージョン5.3(Mathworks, Natick、マサチューセッツ州、米国)の非直線的最小二乗回帰を、Mathworks Optimization Toolboxのルーチンに従って用いることによって実行される。絶対濃度は、データからは得られないが、ヘリックス展開は一次反応なので、これは要求されない(例えば、Szyperski, 1998, Protein Sci. 7:2533−2540)。その代わりに、非直線的回帰では、イオンカウントが、この計測値は各ペプチドについて濃度に比例すると仮定して、濃度単位として用いられる。速度定数の、僅かな標準偏差が、目的関数の標準的直線化工程によって計算される(Seber et al., 1988, 「非直線回帰(“Non−linear Regression”)」、John Wiley & Sons, New York)。
本明細書に記載される化合物は、不一致ヘリックスを含むAβペプチドのα−ヘリックス形を安定化する能力についてアッセイされたことがある、あるいは、これからアッセイすることが可能である。本明細書に記載された方法が、そのようなアッセイに使用することができる。典型的には、試験化合物は、ポリペプチドを含む溶液に加えられ、ヘリックス構造の含量、または、ペプチドのα−ヘリックス形が溶液から消失する速度が、例えば、CD、FTIR、NMR分光光度計測、ES−質量分析、またはMALDI質量分析によって定量される。原線維形成に及ぼす試験化合物の作用も後述のようにして定量される。試験化合物の存在下および不在下における、ヘリックス含量およびα−ヘリックスの消失速度が定量される。試験化合物の存在下における、ヘリックス含量の増大、および/または溶液からα−ヘリックスが消失する速度の低下は、該試験化合物が、ADまたは関連障害を治療するのに有用な候補化合物であることを示す。
試験化合物とは、Aβペプチドを含む溶液のヘリックス含量を増大させる能力、および/または、Aβペプチドのα−ヘリックス形をβ−ストランド構造に変換する速度を遅くさせる能力についてスクリーニングされる化合物である。候補化合物とは、Aβペプチドのα−ヘリックス形を安定化することが判明した化合物である。候補化合物は、β−ストランド構造およびアミロイドの形成を阻止する、または遅らせるのに有用である。
試験化合物をアッセイするさらに別の方法
ある場合には、α−ヘリックス立体配座の安定化を定量するものとは別の方法が、本発明にとって有用なことがある。例えば、原線維形成の直接検出は、α−ヘリックス立体配座を測定するアッセイの代わりに、またはアッセイと組み合わせて用いることが可能である。試験化合物の存在下または不在下における原線維形成の程度と速度は、形成される原線維の電子顕微鏡観察(EM)によって直接測定することが可能である。例えば、原線維を含むペレットが、低エネルギー超音波処理を5秒間用いて少量の水に縣濁される。次に、原線維縣濁液の分液が、炭素安定化Formvar(商標)フィルムによって被われた格子の上に載せられる。30秒後、余分な液体がグリッドから吸引除去される。次に、格子を空気乾燥し、水に溶解した3%酢酸ウラニルで陰性に染色する。次に、染色された格子を、電子顕微鏡、例えば、80kVで動作するPhillips CM120TWINで調べ、撮影する。原線維形成の速度または量の低下をもたらした化合物は、本発明にとって有用であり、例えば、ADまたは関連障害を治療するための候補化合物となる。
原線維形成もまた、α−ヘリックス形の消失を測定することによって間接的に評価することが可能である。α−ヘリックス形の消失は、α−ヘリックスからβ−ストランドへの変換および凝集後、原線維を生じるからである。このためには、ES質量分析が使用可能な方法である。なぜなら、この方法は、不一致ヘリックスを含むAβペプチドのモノマー形と凝集形を区別するために使用が可能だからである。別態様として、凝集体は、例えば、遠心によって除去し、次に、溶液の中に残存するペプチド(主に、その不一致ヘリックス領域におけるα−ヘリックスである)を、ゲル電気泳動、アミノ酸分析、または逆相HPLCのような技術を用いて定量する。
実験データおよび理論モデルから、Aβの16−23領域のα−ヘリックスからβ−ストランドへの転換がアミロイド原線維形成にとって決定的に重要であることが示されている。Aβの16−23位置は、膜様溶媒または界面活性剤の存在下ではヘリックス構造を示すので、この領域は、膜連結APPにおいても螺旋形であると考えられる。しかしながら、水溶液における遊離Aβでは、螺旋形立体配座はほんの一時しか安定ではなく、これは、分光光度法で検出すると水中のAβ立体配座は無秩序であるという所見(Serpell, 2000, Biochim. Biophys. Acta 1502:16−30)と一致する。β−シートの蓄積によって原線維が形成されるためには、16−23領域が、延長型/β−ストランド立体配座へと変換されることが必要とされる。

アルツハイマー病または他のアミロイドーシスの動物モデルを用い、本明細書に開示される化合物についてさらに試験することが可能である。使用される動物は、天然発生モデル(例えば、アミロイドを天然に蓄積する動物、例えば、非ヒト霊長類、例えば、非限定的例として、アカゲザルを含む霊長類)、アミロイドペプチドを発現するように遺伝子工学的に加工された動物か、または、アミロイド生産を誘発するように処理された動物のいずれかによって表される。一般に、化合物の、Aβペプチドの蓄積抑制効力を定めるには、遺伝子工学的に加工された動物モデルが使用される。Aβペプチド蓄積の抑制としては、動物モデルにおいて検出されるAβ含有構造体(例えば、原線維(可溶性または不溶性)、またはアミロイドプラーク)の量の低下;化合物の存在下においてAβ−含有原線維の蓄積が、該化合物によって処理されない動物に比べると、より遅いこと;アミロイド蓄積と関連する症状の低下;または、アミロイド蓄積と関連する症状獲得の遅延、が挙げられるが、ただしこれらに限定されない。このようなモデル、およびAβおよびアミロイド構造を検出する方法は従来技術で既知である。
本明細書に記載される化合物は、様々な方法を用いて投与することが可能である。ある場合には、化合物の効力について、および/または、動物における、例えば、抹消血または脳脊髄液(CSF)における化合物の半減期について、様々な投与法が評価される。
本明細書に記載される化合物は、製薬組成物の中に取り込むことも可能である。このような組成物は、通常、該化合物、および製薬学的に受容可能な担体を含む。本明細書で用いる「製薬学的に受容可能な担体」という用語は、製薬学的投与と適合する、溶媒、分散媒体、コーティング、抗菌剤および抗真菌剤、等調剤および吸収遅延剤等を含む。補足的活性化合物を組成物の中に取り込んでもよい。
本発明は、Aβペプチドに関連する障害、例えば、アルツハイマー病、ボクサー認知症、重度の頭部外傷、および、ダウン症候群のいくつかの病理および症状、を患う危険性のある(または、感受性を持つ)被験者、または、現に患っている被験者に対処する、予防法および治療法の両方を供給する。本明細書で用いる「治療」という用語は、病気、病気の症状、または病気に対する素因を、治す、治癒する、寛解する、解除する、変える、救済する、緩和する、改善する、または作用する目的で行われる、患者に対する治療薬剤の塗布または投与、または、病気を抱える、病気の症状を持つ、または病気に対する素因を持つ患者から分離された組織または細胞系統に対する治療薬剤の塗布または投与と定義される。治療薬剤は、本明細書に記載される化合物を含む。
実施例
残基16および23の間に不一致領域を含む、残基12から28までの、ヒトAβアミノ酸配列(Aβ12−28)を、分子グラフィックス(Insight II, Accelrys Inc.、サンディエゴ、カリフォルニア州)を用いてα−ヘリックス立体配座の中に組み込んだ。Aβ不一致ヘリックスの構造に基づき、Aβヘリックスの表面に対して相補的な化学的構造を持つ化合物を、Insight IIシステムを用いて設計した。一般に、Aβのα−ヘリックス立体配座の消失を抑えるのに有用な化合物は、下記の部位の内少なくとも二つと相互作用を持つ(Aβペプチドを指す)ことの可能な一つ以上の部位を持つ化合物であることが判明した。その部位とは、Lys16の側鎖、His13の側鎖、Glu23の側鎖、Phe19の側鎖、Phe20の側鎖、およびAsp22の側鎖である。
リガンド3
式1(リガンド1、図1)の特定例であるリガンド3(図7に示す)は、活性化ペンタフルオロフェニル・エステル使用の溶液合成によって合成した。合成時、既知の保護基、および、結合および脱保護プロトコールを用いた。保護アミノ酸は全てBachem(King of Prussia、ペンシルバニア州)から購入した。他の試薬は全て市販級であり、溶媒は全て分析級であり、Fluka(セントルイス、ミズーリ州)、Merck(Whitehouse Station、ニュージャージー州)、またはAldrich Chemical Co.(セントルイス、ミズーリ州)から購入した。
Z−Dab(Fmoc−D−Trp)−OH:2.52g(10.00mmol)のZ−Dab−OHを、50mM Na2B4O7(pH8.5)に溶解した。5.93g(10.00mmol)のFmoc−D−Trp−OPfpを、50mlの冷却DMF(0℃)に溶解し、Z−Dab−OH液にゆっくりと加えた。この混合液を0℃で1時間、次に室温で10時間攪拌した。反応を薄層クロマトグラフィー(TLC)で監視した。反応の完了後、溶媒(H2O/DMF)を減圧留去した。油状産物Z−Dab(Fmoc−D−Trp)−COOHは、酢酸エチルから結晶化し、収集し、減圧下に乾燥した。この過程による収量は5.94g(90%)であった。
前述の分子モデル法を用いてさらに別のリガンドを設計し、それらのモデルに対し前述の分子運動力学分析を行った。このようにして作製された追加リガンドは全てAβ螺旋構造の消失を抑制した。合成され、試験された追加リガンドは、リガンド3(図7)の立体化学変異種、構造的異性体リガンド2a(図5および図8)、近縁リガンド2b−2e(図5に示す)、およびその異性体から生じた化合物を含む。
Aβペプチドのα−ヘリックス形を安定化するのに有用なもう一つのリガンド(リガンド7a、図9)を下記のように合成した。1.05当量(2.625mol)のデカン酸を、20mlのジクロロメタンに溶解し、2.0当量のトリエチルアミンを、この溶液に滴下し、これを、攪拌しながら0℃に維持した。次に、1当量(2.5mmol)のクロロギ酸イソブチルを1分間に渡って加え、反応混合物を0℃で30分攪拌した。0℃に冷却した、3当量のNH(C2H5NH2)2、6当量のトリフルオロ酢酸(TFA)、および100mlのメタノールから成る混合物に、活性化脂肪酸を5分間に渡って滴下した。この反応混合物を0℃で1時間攪拌し、次に室温で一晩攪拌した。溶媒を減圧留去し、油状物を100mlのCH2Cl2に溶解し、1M NaOHで洗浄した。溶液を水で洗浄し、分子ふるいを用いて乾燥し、溶媒を減圧留去した。遊離アミンの精製後、この遊離アミンに対するTFA添加(遊離アミンに対し2当量のTFAを加える)後、TFA塩が得られた。次にTFAを蒸発させ、減圧下乾燥した。油状残留物を酢酸エチル(10ml)に溶解し、2−3滴の乾燥ジエチルエーテルを加えた。混合物を室温で保存したところ、結晶化が起こった。結晶をろ過し、減圧下乾燥した。この過程の収率は98%であった。1H−NMRデータは下記の通り。(D3COD+1%TFA) δ3.50(2H t J=5.67Hz CH2), 3.39(2H t J=6.55Hz CH2); 3.37−3.27(2H m CH2); 3.22(2H t J=5.67Hz CH2); 2.24(2H t J=7.70 Hz デカノイル−α−CH2); 1.61(2H m 7.1Hz デカノイル−β−CH2); 1.38−1.20(12H m 6X CH2); 0.90(3H t J=6.76Hz CH3))。
本明細書に記載される処理過程を用いて作製および特定されるリガンドについてさらに説明するために、もう一つのリガンド(リガンド5、図6および16)を下記のように合成した。
さらに別の、Aβペプチドのα−ヘリックス立体配座の消失を阻止することが可能なリガンド、リガンド17(図16)の合成例は下記の通り。
Aβペプチドのα−ヘリックス立体配座の消失を阻止するためのさらに別のリガンドも、リガンド5、7a、および17に関して記載したものと同様のやり方で合成される。これら補足的リガンドは、本明細書に記載される分子モデル法によって設計された。分子運動力学分析を課したリガンドは全てAβ螺旋構造の消失を抑制した。これらの化合物は、リガンド4、5、7、8、9、および17の性質を組み合わせるものを含む。このようなリガンドの例が図10Aに示される(すなわち、リガンド10、10a、11、12、12a、13、14、および14a)。一般に、このクラスのリガンドは、Lys16およびHis13の相互作用とAsp22とGlu23の相互作用とを、これもPhe20を含む相互接続疎水面と新たな相互作用を実現する共有結合リンカーを介して結合することによって分子当たりの相互作用の合計数を増すように設計される。図10Bに、追加化合物が依拠する一般構造が示される(リガンド18およびリガンド19)。上記化合物のいずれについても、n=3−17である。これらのリガンドは1個または2個のR基を持ってもよい。一般に、リガンドは、荷電性の基であって、一方または両方の基が同じ電荷を持つ(例えば、リガンド10−14)。同じ分子の中に二つのR基がある場合(すなわち、リガンド19)、それらは、ある場合には、異なる電荷を持ってもよい。電荷は、一般に、生理的pHにおいて決められることに注意されたい。
本明細書に記載される化合物は、Aβペプチドの二次構造に影響をおよぼす、それらの能力について試験することが可能である。一例として、溶液としたAβ(1−40)またはAβ(1−42)に対してリガンドを添加した場合、円偏光二色(CD)分光光度計測で測定すると、Aβペプチドの螺旋含量は増加した。これは、208および222nmにおける分子楕円率の低下、および、190−195nmにおける分子楕円率の上昇によって裏付けられる(一例が図11に示される)。これらの実験では、Aβペプチド(濃度は約20μM 100μM)を、リガンドと、リガンド/Aβモル比が1から10の範囲となるように混合した。測定は、リン酸バッファー(チオフラビンT測定で後述するように)、または、30%トリフルオロエタノール含有リン酸バッファーにて行った。サンプルは、測定前、振とうしながら、または振とうせずに、インキュベートした。CD測定は、1mmキュベット(サンプル容量200−300μl)によるJasco 810装置を用いて室温で行った。各サンプルについて、3通りのスペクトラムを収集した。スペクトラムは260と185nmの間で測定し、バッファーおよびリガンド単独のスペクトラムを、混合物のスペクトラムから差し引いた。記録されたスペクトラムおよびペプチド濃度を用いて分子楕円率を計算し、kdeg x cm2/dmolで表した。α−ヘリックスAβペプチドの集団を増すことが知られる、10−30%(v/v)のトリフルオロエタノールのAβペプチド溶液に対する添加後、リガンドによる螺旋含量の増加はさらに著明になる。これらのデータは、試験されたリガンドが、その設計から予測されたように、ヘリックス形Aβに結合することを示す。
リガンドがAβに結合することをさらに実証するために実験を行った。これらの実験では、結合を示すのに蛍光発射におけるシフトを用いた。これらの実験では、リガンド2aまたは3(約10μM)を、トリフルオロエタノール添加または無添加において、リン酸ナトリウムpH7に溶解した。蛍光は、Hitachi F−4000分光光度計にて測定し、発射は、280nmで励起した後、300と430nmの間で測定した。
リガンド添加および無添加における、Aβ(1−40)またはAβ(1−42)の原線維形成を、原線維結合性染料チオフラビンT(ThT)を用いて評価した。リガンド無添加でAβペプチドを約20μMから200μMの濃度で含む溶液、あるいは、様々なペプチド/リガンド比でペプチドとリガンドを含む溶液を、10mMリン酸バッファーpH7を溶媒として調製した。サンプルを、絶えず穏やかに振とうしながら37℃でインキュベートした。様々な時点で分液を取り出し、150mM NaClを含む10mMリン酸バッファーpH6に溶解した10μM ThTを用いてThT測定を行った。
デカノイルグルタミン酸およびパルミトイルグルタミン酸(図16)は、リガンド5に関連し、および/または、リン酸塩がカルボン酸塩によって置換されたリガンドであるという点で、リガンド8および9の変異種である。実験は、実施例5に記載したようにThT法を用いて、デカノイルグルタミン酸およびパルミトイルグルタミン酸の、原線維形成を抑制する能力を試験するために行われた。簡単に言うと、デカノイルグルタミン酸またはパルミトイルグルタミン酸の添加または無添加における、Aβ(1−40)またはAβ(12−28)の原線維形成を、原線維結合性染料チオフラビンT(ThT)を用いて評価した。溶液は、リガンド無添加で、または、各種リガンド/ペプチド比において、Aβペプチドを約20μMから200μMの濃度で含んでいた。これらの実験におけるAβペプチドの濃度は25μMであった。図17Aおよび17Bに示す比は、リガンド/Aβのモル比である。ペプチド/リガンドサンプルは、10mMリン酸ナトリウムバッファーpH7で調製した。サンプルは、絶えず穏やかに振とうしながら37℃でインキュベートした。様々の時点で、分液を取り出し、ThT測定を、150mM NaClを含む10mMリン酸ナトリウムバッファー、pH6に溶解した10μM ThTを用いて実行した。
リガンド3は、アルツハイマー病ショウジョウ蝿(Drosophila melanogaster)モデルの寿命を延ばす。
リガンド7aは、Aβの二次構造、およびインビトロ原線維形成に影響を及ぼす
もう一つの候補化合物が、アルツハイマー病または関連障害を治療するのに有用な化合物を示す作用を提示するかどうかを調べるために実験を行った。
リガンド7aは、アルツハイマー病のショウジョウ蝿モデルにおいて寿命を延ばし、運動活性を増す。

- Aβペプチド関連障害を治療するための候補化合物となる化合物を特定する方法であって、
a.請求項1、請求項2、請求項3、請求項6、または請求項7の化合物を準備すること;
b.Aβペプチドを該化合物に接触させ、それによってサンプルを準備すること;
c.サンプルにおける、αヘリックス形またはβ形のAβペプチドの量を定量すること;
αヘリックス形またはβ形のAβペプチドの量を基準と比較することを含み、基準に比べ、サンプル中のαヘリックス形のAβペプチドの量を増しβ形のAβペプチドの量を減らす化合物が候補化合物であるとすることを特徴とする、前記方法。 - Aβペプチドがインビトロで調製されることを特徴とする、請求項14の方法。
- β形のAβペプチドの量はチオフラビンTを用いて定量されることを特徴とする、請求項14の方法。
- Aβペプチドは動物において準備されることを特徴とする、請求項14の方法。
- Aβペプチド関連障害を患う危険性のある、または現に患う被験者を処置する方法であって、
a)Aβペプチド関連障害を患う危険性のある、または現に患う被験者を特定すること;および、
b)請求項1、請求項2、請求項3、請求項6、または請求項7の化合物の治療有効量を、該被験者に投与することを含む前記方法。 - Aβペプチド関連障害がアルツハイマー病であることを特徴とする、請求項18の方法。
- 被験者が哺乳動物であることを特徴とする、請求項18の方法。
- 被験者がヒトであることを特徴とする、請求項18の方法。
