Description
本発明は、4℃から8℃までの温度で実質的に安定な液体ワクチン製剤を提供する。本発明の具体的な実施形態である前記およびその他の液体製剤は、本明細書において、例えば、「本発明のワクチン製剤」、「冷蔵庫安定型ワクチン製剤」、「本発明の液体製剤」、「本発明の製剤」、「本発明の冷蔵庫安定型(RTS)製剤」、あるいは単に「本発明の組成物」または「本発明のウイルス組成物」と呼ばれる。
本発明は、4℃〜8℃の温度で実質的に安定な液体ワクチン製剤を提供する。本発明の具体的な一実施形態では、本発明の液体ワクチン製剤は、少なくとも1ヶ月、または少なくとも2ヶ月、または少なくとも3ヶ月、または少なくとも4ヶ月、または少なくとも5ヶ月、または少なくとも6ヶ月、または少なくとも9ヶ月、または少なくとも12ヶ月、または少なくとも18ヶ月、または少なくとも24ヶ月、または少なくとも36ヶ月、もしくは少なくとも48ヶ月の期間、2℃〜8℃の温度または4℃で実質的に安定であり、その際、該期間の終了時点で、効力の許容可能な損失(例えば、インフルエンザウイルス効力損失)、例えば、0.5〜1.0 logの効力損失(例えば、TCID50または蛍光フォーカスアッセイ(FFA)により測定)が存在する。
本発明の安定化剤は、例えば、下記のうち1以上を含む:アルギニン(例えば、0.5〜1%、1〜2%;1%;1.2%;1.5%、0.75%〜2%);ポロキサマー;ショ糖(例えば、2〜8%;2%;6〜8%;3%;4%;5%;6%;7%;もしくは8%);加水分解ゼラチン(例えば、1%;0.5〜2%;1.5%;0.5%;0.75%);ならびにグルタミン酸塩(例えば、0.05〜0.1%、0.02〜0.15%、0.03%、0.04%、0.06%、0.02〜0.3%、もしくは0.094%)。
一実施形態では、本発明の液体製剤を製造する方法により、10%未満、16%未満、20%未満、30%未満、40%未満、50%未満、60%未満、70%未満、80%未満、90%未満、または93%未満、もしくは95%未満の実際のプロセス収率または平均プロセス収率(ウイルス収集物(VH)から最終製剤までの)が得られる。
本発明者らによる以前の研究により、鼻内噴霧により投与される三価の低温適合性インフルエンザ生ワクチン(CAIV-T、FluMist(登録商標)、一般にFluMistと称されるが、FluMist(登録商標)と考えるべきである)が開発されている。本発明は、CAIV-Tの製剤の開発を含み、該製剤は、冷蔵温度での安定性プロフィールが改善されている。このような改善された製剤を製造する方法を本明細書に記載するが、該方法は、下記ステップのうち1以上を含みうる:汚染の危険性を低減するための無菌濾過ステップ、超遠心分離(例えば、速度ゾーン遠心分離)、ならびに透析濾過。上記以外にも、本発明の液体FluMistおよびその他の冷蔵温度安定型(RTS)製剤を製造する多数の方法も本明細書に含まれている。
低温適合性インフルエンザウイルス(またはその他の類似ウイルス)の精製、および組成物の実際の製剤化はRTS、または液体ウイルス組成物の特徴である。アラントイン液からのインフルエンザウイルスの分離は、不活性化ワクチンを製造する商業的方法の一環として実施されていた。このような不活性化ワクチンの選択方法は、超遠心分離であった。商業規模での連続フロー超遠心機は1969年に利用可能になり、その後すぐに不活性化インフルエンザワクチンの調製に適用された。生きたインフルエンザウイルスのクロマトグラフィー精製は、別の魅力的な手段ではあるが、ウイルス活性を保持する頑健なラージスケールプロセスはまだ利用できる段階ではない。これは、インフルエンザウイルス粒子の膜コートおよび多態性によるものと考えられる。
最初の製剤スクリーニングにより、冷蔵温度での精製CAIVの液相安定性がアニュアルワクチンに適していることを決定した。凍結FluMist安定化剤SPGに添加した加水分解動物ゼラチンが最良の安定性結果をもたらし、CTM-1計画(campaign)からの最も安定性の低い株およびロットが示す損失は6.9ヶ月で1logであった。別の製剤開発試験により、液体FluMistの安定性は、製造直後に凍結保存し、最終的販売の前に解凍することによりさらに改善できることがわかった。最も悪い結果の株の安定性も、アルギニンの添加により改善された。動物ゼラチンは、伝染性海綿状脳症(TSE)に関する懸念を高めうるが、ゼラチンを含まない入手可能な製剤では、すべての実施形態で、要求される安定性に達しなかった。従って、安定化特性を有することと、ブタにはTSEの発生が報告されていないことから、いくつかの液体FluMistの実施形態ではブタゼラチンを選択した。また、コラーゲン加水分解に用いる過酷な化学処理工程も、プリオンサイズのタンパク質の不活性化を起こすと考えられる。
一実施形態では、本発明のワクチン製剤は、最終製剤中に、以下に示すもののうち1以上を含む:ショ糖:6〜8重量%/容量(w/v);アルギニン一塩酸塩1〜2%w/v;グルタミン酸一水和一ナトリウム0.05〜0.1%w/v;ゼラチン水解物、ブタA型(またはその他の供給源)0.5〜2%w/v;二塩基性リン酸カリウム1〜2%;および一塩基性リン酸カリウム0.25〜1%w/v。
別の具体的な実施形態では、本発明のワクチン製剤を鼻内投与する方法が含まれる。例えば、0.5 mL/用量;0.75 mL/用量;0.1 mL/用量;0.15 mL/用量;0.2 mL/用量;0.25 mL/用量;0.5〜0.1 mL/用量;0.5〜2mL/用量;0.5〜2.5 mL/用量の最終用量で本発明のワクチン製剤を鼻内投与することができる。
論考および説明を容易にするために、ワクチン組成物製造の様々なステップはおおまかに4つの大きなグループ(上記で概要を記載したものと大体類似している)からなる、またはこれらに属するものとして考えることができる。第1グループは、共感染、再構築、再構築体の選択、ならびに再構築体のクローン化などの態様を含む。第2グループは、再構築体の精製および増殖などの態様を含む。第3グループは、卵における再構築体のさらなる増殖、ならびにこうして収集したウイルス溶液の収集および精製を含む。第4グループは、収集したウイルス溶液の安定化、およびウイルス溶液の効力/無菌性アッセイを含む。しかし、前記4つのおおまかなカテゴリーに本発明の態様を分類するのは、説明/系統化の目的のためにすぎないこと、ステップ同士の相互依存などを推断すべきではないことに留意されたい。また、本発明の方法および組成物と一緒に、その他のステップ(類似または異なる)を随意に用いてもよい(例えば、RTSワクチン組成物のための方法および組成物)ことは理解されよう。
第1グループに属するものとしておおまかに分類されるワクチン組成物製造の形態は、具体的に所望する再構築ウイルスの生産を目的とする、例えば、マスタードナーウイルスおよび1以上の野生型ウイルスを用いた細胞培養系の共感染の最適化;適切な再構築ウイルスの選択;および選択した再構築ウイルスのクローン化に関する方法および組成物を含む。インフルエンザウイルス株の再構築は、当業者には周知である。細胞培養および卵の両方においてA型インフルエンザウイルスおよびB型インフルエンザウイルス両者の再構築により、再構築ウイルスが製造されている。例えば、Tannockら、Preparation and characterisation of attenuated cold-adapted influenza A reassortants derived from the A/Leningrad/’134/17/’57 donor strain, Vaccine (2002) 20:2082-2090を参照。また、プラスミド構築物を用いたインフルエンザ株の再構築も記載されている。例えば、下記の文献を参照されたい:2002年4月26日出願の米国特許出願第60/375,675号、2003年4月25日出願のPCT/US03/12728 および2003年4月25日出願の米国特許出願第10/423,828号、2005年5月20日出願のPCT/US05/017734;ならびにUS200501 86563。
第1グループのステップを用いた様々な実施形態は、再構築工程を最適化して、必要な再構築の数を減らす(そして、ワクチン製造工程のスループット/安定性を高める)などすることができる。このような最適化技術を用いたステップは、B型インフルエンザ株の再構築により実施し、典型的には、細胞培養、例えば、CEK細胞において実施する。例えば、いずれも2004年2月25日出願の米国特許出願第10/788,236号およびPCT/US04/05697(本節および本明細書全体を通じ、あらゆる目的のために全文を参照として組み込む)を参照されたい。
第1グループのステップは、再構築したインフルエンザウイルスの選択も含む。再構築A型インフルエンザ株は、細胞培養(例えば、CEK細胞)または卵のいずれかのものを選択することができる。しかし、再構築B型インフルエンザ株は、細胞培養(例えば、CEK細胞を選択した場合)で再構築したとき、問題を生じる。CEK細胞は、B型インフルエンザ株におけるM遺伝子を妨害するため、生産全体を低下させると考えられている。ワクチン組成物を製造する多様な方法(例えば、いずれも2004年2月25日出願の米国特許出願第10/788,236号およびPCT/US04/05697を参照)では、ウイルス再構築のための様々なステップ(例えば、選択)を使用しており、このようなステップを随意に用いて、ワクチン組成物のためのウイルスを作製することができる。
ウイルス/ワクチン製造のさらに別の方法では、高スループット一本鎖コンホメーション多型/キャピラリー電気泳動(SSCP/CE)アッセイを実施することにより、本発明で用いるインフルエンザウイルスの遺伝子配置を決定する。インフルエンザウイルスは、8つの遺伝子セグメントを含み、前述したように、2つの異なるインフルエンザ株を用いた単一細胞の共感染により、それぞれの親ウイルスとは異なる新しい遺伝子配置を有する再構築ウイルスを作製することができる。例えば、いくつかの方法では、SSCP/CEアッセイを用いて、多数のインフルエンザウイルスサンプルの遺伝子セグメント配置を速やかに決定することができる。インフルエンザウイルス遺伝子セグメントは、8セグメントの各々について特異的な蛍光標識プライマーを用いたRT-PCRにより随意に増幅する。Arvinら(2000)Clin. Micro. J38(2):839-845(あらゆる目的のために参照として本明細書に組み込む)も参照されたい。
ウイルス/ワクチン製造のいくつかの方法は、インフルエンザウイルスが生産される卵の微生物汚染を検出および/または予防/検出するステップを含んでもよい。本発明の微生物検出戦略は、高速/高スループット微生物検出に有用であり、従って、他の多くのステップと同様に、スループットを高めると同時に、場合によっては、ウイルス/ワクチン製造における安定性を向上させる上で有用である。
第2グループに分類されるウイルス/ワクチン製造の形態はさらに、精製およびウイルス増殖などを含む。適正な再構築および再構築体(すなわち、6:2ウイルス)のクローン化工程後、このような再構築ウイルス粒子を有精鶏卵においてさらに精製し、適正なクローンを多数増殖する(やはり、鶏卵での増殖により)ことにより、マスターウイルス株(MVS)またはマスターウイルスシードを作製し、次に、これをさらに増殖してマスターワーキングウイルス株(MWVS)または製造者のワーキングウイルスシードを作製する。卵からのウイルス粒子の精製、ならびに、ウイルス粒子の量を増大するために、これらの精製ウイルスを用いてさらに多くの卵に接種する多数の態様は、当業者には周知である。このような多くの技術は、現行のウイルス粒子生産には一般的であり、40年以上用いられている。例えば、Reimerら、Influenza virus purification with the zonal ultracentrifuge, Science 1966, 152:1379-81を参照されたい。精製プロトコルは、例えば、ショ糖勾配(例えば、10〜40%ショ糖)超遠心分離などを含んでもよい。また、本明細書に記載するように、他のグループに記載される他の手順なども、随意に第2グループに存在してもよく、例として、微生物汚染の予防などが挙げられる。
第3グループの項目に分類されるウイルス/ワクチン製造の態様は、例えば、有精卵のコンディショニング(例えば、ウイルス感染卵のインキュベーションに関与する取扱いおよび環境条件)、ならびに卵のアラントイン液からのインフルエンザウイルスの収集および分類を含む。
本発明は、本グループに分類される超遠心分離(前記参照)の態様を含む。加えて、いずれも2004年2月25日出願の米国特許出願第10/788,236号およびPCT/US04/05697には、本発明の方法および組成物に随意に用いることができるその他の濾過および加温ステップも提供されている。記載されているように、FluMistTM製造方法は、有精鶏卵を用いて、マスターウイルスシード(MVS)、製造者のマスターワーキングウイルスシード(MWVS)およびウイルス収集物(VH)を作製する。上記シードおよびウイルス収集物は、バイオバーデン(典型的には細菌汚染)を含んでいる可能性があり、そのような場合、前記シードまたはバルクウイルス製品ロットはワクチン製造工程で不合格になってしまう。言うまでもなく、用いる特定の製品タイプ、サイズの具体的一覧または記載は、そのように明記していない限り、本発明に関して限定的と考えるべきではない。
ワクチン製剤/組成物製造の態様からなる第4グループは、例えば、第1に安定化(例えば、成分の添加、バッファー/NAF比の改変などにより)に関するステップ、およびウイルス含有溶液の効力/無菌性についてのアッセイを含む。前述した本発明の説明から、本カテゴリーに随意に分類することができる様々な形態がわかるだろう。上記を参照されたい。
いくつかのワクチン組成物製造方法では、好適なカラムを用いて、場合によってはウイルス収集物を濃縮する。例えば、いずれも2004年2月25日出願の米国特許出願第10/788,236号およびPCT/US04/05697を参照されたい。
ワクチン組成物製造はまた、場合によっては、目的のウイルスと、例えば、ショ糖、アルギニン、ゼラチン、EDTAなどの組合せを含むNAF(典型的には非分画NAF)の様々な希釈物を含んでもよい。例えば、各種のワクチン組成物に関して考えられる様々な組合せについては、米国特許出願第10/788,236号およびPCT/US04/05697を参照されたい。好ましくは、このような方法および組成物は安定である:例えば、所望の温度(例えば、典型的には、4℃、5℃、8℃、約2℃〜約8℃、または2℃より高い)で、選択した期間(典型的には、少なくとも6ヶ月、少なくとも9ヶ月、少なくとも12ヶ月、少なくとも15ヶ月、少なくとも18ヶ月、少なくとも24ヶ月など)、効力の許容不可能な損失を示さない(例えば0.5〜1.0 log、または0.5 log未満、もしくは1.0 log未満の効力損失)。
特に記載のない限り、科学用語および技術用語はすべて、それらが関連する分野で一般に用いられるのと同じ意味を有するものとする。本発明の目的のために、下記の用語について以下のように定義する。
本明細書に記載する組成物および方法は、主として、ワクチンのためのインフルエンザウイルスの製造に関する。インフルエンザウイルスは、セグメント化した一本鎖RNAゲノムを含む内部リボヌクレオチドタンパク質コアと、マトリックスタンパク質で覆われた外部リポタンパク質エンベロープとから構成される。A型インフルエンザゲノムは11のポリペプチドをコードする。セグメント1〜3は、RNA依存性RNAポリメラーゼを形成する3つのポリペプチドをコードする。セグメント1は、ポリメラーゼ複合体タンパク質PB2をコードする。残るポリメラーゼタンパク質PB1およびPAは、セグメント2およびセグメント3によりそれぞれコードされる。加えて、特定のインフルエンザ株のセグメント1は、PB1コード領域内の選択的リーディングフレームから生産される小さなタンパク質であるPB1-F2をコードする。セグメント4は、感染時の細胞接着および侵入に関与する赤血球凝集素(HA)表面糖タンパク質をコードする。セグメント5は、ウイルスRNAに関連する主要な構造成分であるヌクレオキャプシド核タンパク質(NP)ポリペプチドをコードする。セグメント6は、ノイラミニダーゼ(NA)エンベロープ糖タンパク質をコードする。セグメント7は、別々にスプライシングされたmRNAから翻訳される2つのマトリックスタンパク質(M1およびM2と称する)をコードする。セグメント8は、選択的にスプライシングされたmRNA変異体から翻訳される2つの非構造タンパク質、NS1およびNS2をコードする。
歴史的に、インフルエンザウイルスワクチンは主として、問題となる株についての経験による推定に基づき選択したウイルスの株を用いて、有精鶏卵において生産されてきた。さらに近年、認可された弱毒化温度感受性マスター株に関して、選択した赤血球凝集素およびノイラミニダーゼ抗原を組み込んだ再構築ウイルスが生産されている。鶏卵中での複数回の継代によるウイルスの培養後、インフルエンザウイルスを回収した後、場合によっては、例えば、ホルムアルデヒドおよび/またはβプロピオラクトーンを用いて不活性化する(あるいはまた、弱毒化生ワクチンに用いる)。
既述したように、インフルエンザワクチンには多数の例および種類がある。インフルエンザワクチンの一例としてFulMistが挙げられるが、これは、インフルエンザから小児および成人を防御する弱毒化生ワクチンである(Belsheら、(1998) The efficacy of live attenuated, cold-adapted, trivalent, intranasal influenza virus vaccine in children N. Engl. J. Med. 338:1405-12;Nicholら、(1999) Effectiveness of live, attenuated intranasal influenza virus vaccine in healthy, working adults: a randomized controlled trial JAMA 282:137-44)。典型的な実施形態では、好ましくは本発明の方法および組成物をFulMistワクチンの製造に合わせて改変するか、またはこれと一緒に用いる。しかし、当業者は、本明細書に記載するステップ/組成物が、同様の、あるいは別のウイルスワクチンおよびその組成物の製造に合わせて改変できることを理解されよう。
既述したように、場合によってはインフルエンザウイルスを細胞培養で増殖することができる。典型的には、ウイルスの増殖は、宿主細胞を通常に培養する培地組成物中で達成される。インフルエンザウイルスの複製に適した宿主細胞として、例えば、Vero細胞、BHK細胞、MDCK細胞、293細胞およびCOS細胞(例えば、293T細胞、COS7細胞)が挙げられる。一般に、上記細胞株のうち2つ(例えば、MDCK細胞と293T細胞または COS細胞のいずれか)を含む共培養を1:1の比で用いることにより、複製効率を高める。典型的には、標準的な市販の培地、例えば、血清(例えば10%ウシ胎仔血清)を添加したダルベッコ改変イーグル培地、または無血清培地において、中性緩衝pH(例:pH7.0〜7.2)を維持するのに適した調節湿度およびCO2濃度下で細胞を培養する。培地は、細菌増殖を阻止する抗生物質、例えば、ペニシリン、ストレプトマイシンなど、および/または追加栄養素、例えば、L-グルタミン、ピルビン酸ナトリウム、非必須アミノ酸、好ましい増殖特性を促進する別の補充物質、例えば、トリプシン、βメルカプトエタノールなどを随意に含む。
本発明の方法および組成物の使用を容易にするために、ワクチン成分および/または組成物、例えば、各種製剤におけるウイルスなど、ならびに、実験または治療ワクチンの目的で、インフルエンザウイルスのパッケージングおよび感染に有用な追加成分、例えば、バッファー、細胞、培地をキットの形態でパッケージングすることができる。典型的には、このキットは、前記成分以外に、例えば、本発明の方法の実施に関する説明書、パッケージング材料、および容器などの追加要素を含む。
液体FluMistの開発
4つの開発ロット(CB0006H、CB0008H、CG0017H、CB0018HおよびCB0019H)を含む計19のウイルスの一価バルクロットをプロトコルに従い開始した。ロットCB0018HおよびCB0019Hを実施することにより、各種試験のための材料を供給したが、本明細書ではこれ以上説明しない。超遠心分離工程の後、2つのロット(CB0018HとCB0012H)を一緒にして、A/Sydney一価バルクCB0020Hを生産した。約2,000個の卵を用いて、各ロットを開始した。
液体組成物の製法を開発したが、これには一価バルクロットCAZOO1〜024およびCAZ035〜043を含むものであった。臨床試験材料(CTM)には、一価バルクロットCAZ025〜CAZ034が含まれた。
段階1:SPF卵の受取りおよび衛生化;段階2:ウイルス収集物の生産;段階3:ゾーン遠心分離によるウイルスの濃縮;段階4:ピーク画分のプールおよび希釈;段階5:無菌濾過および一価バルク保存;ならびに段階6:混合と充填。
SPAFAS, Inc(病気のない卵を確実に提供するために設計された群れ管理および監視プログラムを用いている)から、特定の病原体を含まない(SPF)有精卵を購入した。卵は米国産で、ClorwashおよびQuat 800スプレーを用いて衛生化した。次に、空路および陸路で卵を輸送したが、その際、清潔さを維持し、凍結または過熱を防止するのに十分な梱包を施した。受取り後、冷却した受精卵を検査し、14℃±2℃および相対湿度(RH)が60〜80%のSPFインキュベーションユニットで最大7日保存した。販売者が供給したボール箱から卵をトレーに移し、バッチ番号を割り当てた。卵を36卵ジェームズウェイ(Jamesway)トレーに配置し、自動卵衛生化装置において卵殻の表面をChlorwashおよびQuat 800で衛生化することにより、インキュベーターに配置する前のバイオバーデンを最小限に抑えた。Chlorwashは43〜44℃および48〜49℃の範囲で、またQuat 800は48〜49℃の範囲で、それぞれ調製した。衛生化の後、卵をトロリーの上に載せ、室温で2時間以上空気乾燥させた。次に、卵をBuckeyeインキュベーターに配置し、37.5℃±1℃および60〜80%RHで264時間±12時間インキュベートした。インキュベーション後、SPF卵ユニットからSPF卵をキャンドリングエリアに移した。光ファイバーランプを用いて、各卵について気嚢の位置を決定した。死んだか、または無精卵は廃棄し、接種のために受精卵に印をつけた。
摂取前に受精卵の表面を70%工業メチル化酒精(IMS)で衛生化してから、大量接種のために移動させた。希釈した製造者のワーキングウイルスシード(MWVS)の調製に際しては、フェーズ1臨床試験のためのMWVSを製造するのに用いるマスターウイルスシードを、古典的再構築方法を用いて作製した。MWVSをAVUに移し、移動のたびに滅菌使い捨てピペットを用いて、微生物防御キャビネット内で、解凍したMWVSの連続的希釈物を無菌0.01Mリン酸緩衝食塩水(pH 7.7)において調製した。1トレー当たり36個の卵に貫入および接種する半自動Bibby接種機を用いて、クラス10,000室内の層流下で接種を実施した。接種材料の目標力価は、0.1 mL当たりlog10 2.1 TCID50で、希釈はMWVSの予め定めた力価から計算した。各ウイルス接種材料の調製は、冷凍庫からバイアルを取り出して2時間以内に完了した。接種材料のアリコートを使用まで5±3℃で保存した。接種材料を調製から8時間以内に用いた。
卵の各トレーを手で自動Bibby接種機に供給した。パンチを用いて、卵殻に孔をあけ、予定した調節可能な深さまで卵に貫入した。接種針を卵内に延ばし、投薬ポンプで0.1 mLのMWVS接種材料を各卵に送達し、その後、パンチを引き抜いた。接種後、卵トレーを操作者が取り出し、新しい卵トレーで同じ工程を繰り返した。ウイルス株の増殖曲線により決定した時間、33±1℃でCAP/接種卵をインキュベートした。接種後、SPF有精卵を28℃まで18±6時間冷却した。冷却終了後、収集ステップのために卵を移動させた。
ウイルス収集物(VH)
自動収集機(Bibby)で卵を収集した。切断ステーションにトレーを手で運び、そこで、卵殻の上部を除去することにより、収集針を入れるための貫入孔を形成した。収集前に卵を肉眼で検査し、不良の卵(例えば、変色したもの)を排除した。合格卵は収集ステーションまで進むが、そこでアラントイン液を針で抜き取り、真空装置により吸い上げる。ジャケット温度が2〜8℃の100 Lジャケット付きステンレス容器に収集物を回収し、混合した。ウイルス収集物のサンプルをVHプールから回収し、以下のことについて試験した:効力(TCID50)、安全性、トリ白血病、結核菌、マイコプラズマ、外来ウイルス、同一性、逆転写酵素アッセイ、ウイルス遺伝子型、ウイルス表現型、および弱毒化。
プールしたウイルス収集物(VH)を直ちに、2〜8℃で、10×SPG(9部のVHと1部の10×SPG)を用いて、0.2Mショ糖、0.01Mリン酸、0.005Mグルタミン酸(SPG)の最終濃度まで安定化した。清澄化前の効力およびバイオバーデン試験のため、サンプルを回収した。5μmデプスろ過により、安定化ウイルス収集物を清澄化して、粒状物質を除去した後、高速連続フロー遠心分離による精製に付した。清澄化ウイルス収集物を効力について試験した。
使用前に、遠心分離ローターを濃縮ホルムアルデヒド溶液の1:20希釈液で衛生化した。清澄化VHをショ糖勾配(pH7.2のリン酸バッファー中10〜60%ショ糖)に重層し、Hitachi CP40Yゾーン遠心機を用いた高速遠心分離に付す。勾配は、リン酸バッファーの60%ショ糖溶液、次に、10%ショ糖溶液を遠心機に添加することにより形成した。ショ糖溶液が密度勾配を形成できるように、遠心速度を20分間4,000 rpmに設定した。勾配形成中、遠心機温度は2〜8℃に設定した。勾配形成後、バッファーフローを開始し、ローター速度を40,000 rpmまで上昇させた後、40,000 rpmで毎時20Lの速度で清澄化VHを勾配に重層した。収集した10,000個の卵を含み、6mL/卵の典型的CTM-Iバッチの場合、重層ステップの時間を約3時間にした。清澄化VHの重層後、遠心分離を40,000 rpmでさらに1時間続けることにより、ウイルスをバンド形成させる。ウイルス粒子は、勾配および濃縮物の38〜45%ショ糖部分に移動し、「バンド」に濃縮した。このステップの終了時に、遠心分離の速度を徐々に下げてから停止し、ウイルスピークを回収した。層流下で125 mL滅菌ポリカーボネート瓶に100 mL画分を回収した。画分を2〜8℃で約1時間保持しながら、HA活性についてアッセイした。一般にピーク画分は38〜45%のショ糖勾配に存在した。
HAアッセイにより特定した遠心ピーク画分は、5L無菌ガラス瓶に該画分を無菌状態で注ぎ込むことにより層流フード下でプールし、攪拌により混合した。屈折率(RI)を読み取って、ショ糖濃度を決定した後、効力について該プールを試験した。計算量の無菌冷却(2〜8℃)リン酸−グルタミン酸バッファー、pH7.2(PBGバッファー)を最終濃度:0.2Mショ糖、0.1Mリン酸および0.005Mグルタミン酸まで、無菌状態で添加することにより、遠心ピーク画分プール(CP)を希釈した。これは典型的に1:6希釈であった。希釈遠心ピーク画分プール(DCP)を効力およびバイオバーデンについて試験した。
無菌濾過のために、無菌チューブを通じてDCPをクラス100充填室にポンプ輸送した。0.22μmフィルターを用いて、DCPを層流フード下で無菌5Lガラス瓶中に濾過した。濾過した一価バルクを混合し、効力、同一性および無菌性について試験した。濾過工程の前および後に0.22μmフィルターを完全性について試験した。次に、MBを無菌1リットルポリカーボネート瓶に取り分けた。MBを総量3〜4リットルの500 mLアリコートとして集めた。無菌1リットルPC瓶を−60℃以下で保存した。バルクウイルスの力価をTCID50アッセイにより決定した。一価バルクを混合および試験のために<−60℃で輸送した。
バルク三価混合調製物の混合および充填工程のための主なステップは、解凍、希釈剤の調製、混合、充填およびパッケージングである。
好適な瓶に入った一価バルクを凍結保存(MBの場合は、<−60℃)から取り出し、解凍室に移した。3つの個別の一価バルクとSPG希釈剤の必要量をバルク感染力価および要求される製剤力価に基づき計算した。MBの瓶を33±3℃の水浴に入れ、5分毎に手で攪拌した。解凍を肉眼でモニターすることにより、水浴から取り出す前にすべての瓶が解凍したことを確認し、解凍したすべての瓶を15分毎に取り出した。いったん解凍したら、MBの瓶を5±3℃の冷蔵庫に移し、必要な瓶がすべて解凍されるまで保持した。
pH調節後、バルク三価混合容器を充填室に移し、INOVA充填機に接続した。INOVA充填機は、予定量の製品を一列に並ぶ8個のBD HYPAK使い捨て噴霧器に充填した後、これら噴霧器を密栓した。噴霧器充填および密栓作業ならびに重量確認は指定通りに継続した。各充填サイクルの完了時に、噴霧器の新しいタブ(tub)を供給(in-feed)ステーションに手で配置し、充填した噴霧器のタブを排出ステーションに運んだ。充填工程中、冷却パックを用いて、混合容器を5±3℃で維持した。
濾過(5μm)による収集物の分類
濾過と低速遠心分離:凍結FluMistについて標準的低速遠心分離を用いた場合の清澄化効力損失は一般に0.2〜0.3 log10 TCID50/mLと見積もられる。2つの株についての推定遠心分離損失は次の通りであった:3,400gで20分間の標準的条件を用いた場合の力価損失は、一方の株については無視できる程度で、第2の株については0.3 log(ステップ収率59%)であった。プレCTM開発ランにおいて、遠心分離に代わり、5ミクロンデプスフィルターを用いたところ、平均清澄化ステップ収率は41%と見積もられた。TCID50アッセイ生存能を考慮に入れ、いずれの清澄化方法も、清澄化後に加える場合より、優れた結果をもたらすと考えられた。操作しやすさおよびスケール変更の容易さの点から、清澄化として濾過を選択した。
遠心スケール
ラージスケールCP40Y遠心機とRP40CT D型連続フローローターを用いて、遠心重層および温度試験を実施した;これらの比較した各種卵バッチサイズおよび遠心温度設定点を示す(図3参照)。CAZ015(B/Harbin)、CAZ018(A/Sydney)、CAZ020(A/Beijing)およびCAZ022(A/Sydney)は、バッチサイズが10K卵で、遠心機設定温度が4℃であった。CAZ016(B/Harbin)、CAZ019(A/Sydney)、およびCAZ021(A/Beijing)は、バッチサイズが20K卵で、遠心機設定温度が14℃であった。すべての試験について20L/時の重層流量を用いた。希釈したピーク画分プールにおける回収率(ステップ収率)は、図に示す清澄化収集物力価に基づいている。図からわかるように、10K卵バッチのロットは40〜184%の回収率であったのに対し、20K卵バッチは40〜55%の範囲であった。10Kバッチサイズの平均回収率は113%であり、20K卵バッチの平均回収率は46%であった。CTM製造については、開発工程の本ステップにおける少なめの限界として、1ラン当たり卵10,000個のバッチサイズを選択した。
4℃または14℃いずれかの超遠心ローター設定温度を用いて、開発ランCAZ015〜CAZ022を実施した。1つの例外はあるが、14℃での開発ランもまた、典型的スケール(1バッチ当たり卵20,000個)より大きなスケールで実施した。一般に、これらのバッチは、生きたウイルスの回収率が低いが、これは、温度の影響よりもむしろ遠心重層によるものと考えられる。4℃での結果が優れていることと、温度が上昇すれば微生物増殖速度が高くなる可能性を考慮し、開発工程でのこの時点で設定する超遠心温度として、温度4℃を選択した。
CTM-1の一価ウイルスロットを製造したが、これは、一価バルクロットランCAZ025〜CAZ034を含んでいた。CTM-1の一価バルクロットについての工程内QCアッセイ結果を図3および本節に示す。1株当たりの平均ステップ収率を図3および本節に示す。以下に結果をまとめる。
本明細書に記載する様々な実施形態および実施例では、収集液の卵黄汚染が起こらないことが好ましく、このような汚染は、例えば、不適切な収集機設定で生じうる。
2つの臨床試験製造計画(CTM-1およびCTM-2)に基づき、液体FluMist製法開発および製造作業から、該製法が流通規格に合格する臨床供給品を生産するのに適していることがわかる。両CTMランおよび各種開発研究からの総計データから、下記のようなプロセス収率中央値の推定値が得られる:
清澄化:収率50%
超遠心精製:収率50%
ピーク希釈および無菌濾過:収率20〜50%。
3つの異なる再構築インフルエンザウイルス(7.0±0.5 log10 FFU/用量[約7.0±0.5 log10 TCID50/用量])を含み、さらに100 mMリン酸カリウムバッファー(pH 7.2)中に200 mMショ糖;1%(w/v)ブタゼラチン水解物;1.21%(w/v)アルギニン一塩酸塩[1%(w/v)アルギニン塩基と同等];および5mMグルタミン酸一ナトリウムを含む三価ワクチン製剤を調製した。製剤の安定性を−25.0℃±5.0℃で24時間以上、しかし2週間以下の期間保存した後、様々な期間2〜8℃で保存することにより調べた。この製剤(例えば、ロット0141500003)は、少なくとも12週間4〜8℃で安定していると決定された。特に、各ウイルス株の効力は、4〜8℃での保存前の初期効力の0.5 log10以内に留まった。
Claims (21)
- 冷蔵庫安定型インフルエンザウイルス組成物の製造方法であって、以下の各ステップ:宿主卵の集団または宿主細胞の集団をインフルエンザウイルスに感染させるステップ;適切な温度で上記宿主卵の集団または宿主細胞の集団を培養するステップ;ウイルス収集物としてインフルエンザウイルスを回収するステップ;濾過によりウイルス収集物を清澄化し、これによって清澄化ウイルス収集物を取得するステップ;該清澄化ウイルス収集物を連続フロー遠心分離に付すことにより、さらに清澄化したウイルス収集物を取得するステップ;該さらに清澄化したウイルス収集物を濾過により滅菌するステップを含む、上記方法。
- 前記インフルエンザウイルスが、下記:弱毒化インフルエンザウイルス、低温適合性インフルエンザウイルス、温度感受性インフルエンザウイルス、もしくは弱毒化低温適合性温度感受性インフルエンザウイルスのうちの1以上を含む、請求項1に記載の方法。
- 前記インフルエンザウイルスが、1以上のA型インフルエンザウイルス株および/または1以上のB型インフルエンザウイルス株を含む、請求項1に記載の方法。
- 請求項1、2または3に記載の方法により製造された冷蔵庫安定型インフルエンザウイルス組成物。
- 少なくとも1種のインフルエンザウイルス株と、下記のもの:
a)6〜8%ショ糖;
b)1〜2%アルギニン一塩酸塩;
c)0.05〜0.1%グルタミン酸一ナトリウム一水和物;および
d)0.5〜2%ゼラチン水解物
からなる群より選択される1以上の構成要素とを含む、冷蔵庫安定型インフルエンザウイルス組成物。 - 6.84%ショ糖、1.21%アルギニン一塩酸塩、0.094%グルタミン酸一ナトリウム一水和物;および1%ゼラチン水解物を含む、請求項5に記載の冷蔵庫安定型インフルエンザウイルス組成物。
- 前記組成物が、少なくとも3ヶ月間4℃で保存した場合、1.0 log未満の効力損失を示す、請求項4に記載の冷蔵庫安定型インフルエンザウイルス組成物。
- 前記組成物が、少なくとも3ヶ月間4℃で保存した場合、1.0 log未満の効力損失を示す、請求項5、6または7に記載の冷蔵庫安定型インフルエンザウイルス組成物。
- 前記組成物が、低温適合性温度感受性弱毒化インフルエンザウイルスをさらに含む、請求項8に記載の冷蔵庫安定型インフルエンザウイルス組成物。
- 請求項7に記載の冷蔵庫安定型インフルエンザウイルス組成物を含む、免疫原性組成物。
