Abstract
Description
本出願は、2004年7月30日に提出された米国仮特許出願No. 10/903,191号に基づく優先権を主張するものである。前記の出願の内容全体を、引用をもってここに援用することとする。
免疫応答は、身体全体の組織に常在する樹状細胞(DC)及びマクロファージ(Mg)を含む専門的な抗原提示細胞(APC)のレベルで惹起される。DCは高レベルの細胞表面分子や、Tリンパ球と相互作用する補完的な受容体を発現するため、効力ある免疫応答を誘導する。DCはまた、免疫応答を惹起すると共に、細胞免疫及び体液性免疫の両方の増幅で蓄積するサイトカイン、ケモカイン及びプロテアーゼを分泌する。
クラス II 分子との結合に向けてプロセッシングされる。対照的に、MHC クラス I分子の細胞内位置のために、内因的にAPCに送達された抗原は、主にwith
MHC クラス I 分子との結合に向けてプロセッシングされる。これはAPCにとってのみ真実ではない。なぜなら全ての有核細胞がMHCクラスI分子を発現し、それらの表面上に内因的に産生された抗原を、MHCクラスI分子と結合させて継続的に提示しているからである。
本発明は、数多くの疾患の有効な治療に必要な、抗体ベースのワクチンと、抗原特異的T細胞媒介性免疫応答を生ずる方法を、提供するものである。具体的には、効力ある抗原特異的細胞障害性Tリンパ球(CTL)応答が、APC上に発現する特定の受容体に結合する抗体を用いて一つ以上のたんぱく質抗原を抗原提示細胞(APC)に標的決定することにより、誘導される。好適な受容体には、樹状細胞(DC)及びマクロファージの両方の上に発現する、C-レクチン、特にヒトマンノース受容体、がある。本発明の例で実証するように、抗体−抗原複合体を用いてマンノース受容体を標的に決定すると、抗原プロセッシングがMHCクラスI及びクラスII経路の両方を通じて起きる。このように、抗原特異的CTL(例えばCD8+T細胞)や、ヘルパT細胞(例えばCD4+T細胞)を含む他の重要なエフェクタT細胞が誘導される。従って、ある局面では、本発明は、抗原と、例えばヒトAPC上に発現するヒトマンノース受容体に結合するモノクローナル抗体など、ヒトAPCに結合するモノクローナル抗体との複合体を形成することにより、この抗原に対するCTL応答を誘導又は亢進する方法を提供するものである。次にこの複合体を、in vivo又はex vivoでAPCに接触させて、この抗原に対するCTL応答(例えば、CD8+細胞障害性T細胞により媒介される応答など)を誘導又は亢進する態様で、この抗原を内部移行させ、プロセッシングさせ、そしてT細胞に提示させる。ある好適な実施態様では、これは、当該抗原に対するヘルパT細胞応答(例えばCD4+ヘルパT細胞により媒介される応答など)を誘導するためにも役立つ。従って、この免疫応答はMHCクラスI及びMHCクラスII経路の両方を通じて誘導される。更にAPCを、樹状細胞の増殖を刺激して免疫応答を更に亢進する、アジュバント又はサイトカインなどの免疫刺激性物質に接触させることもできる。このような物質を、当該分子複合体とは別に接触させることも、あるいは、当該分子複合体に連結することもできる。

本発明は、特定の細胞受容体を狙った抗体を用いて抗原を抗原提示細胞(APC)の標的に決定することにより、重要なT細胞媒介性免疫応答を生じさせることができるという発見に基づく。具体的には、癌及び感染性疾患などの数多くの疾患の有効な治療のためには、ワクチンは、MHCクラスIの流れで抗原を認識するCD8+T細胞に主に媒介される、効力ある抗原特異的細胞障害性Tリンパ球(CTL)応答を惹起せねばならない。最適な免疫処理のためには、MHCクラスII経路の流れで抗原を認識するCD4+T細胞などの抗原特異的ヘルパT細胞の誘導を含め、他の重要なエフェクタT細胞機能がこれに伴っていることが好ましい。このように、有効なワクチンは、好ましくは他のT細胞媒介性免疫応答との組み合わせで、複数のMHC経路を通じて、抗原特異的CTLを誘導しなければならない。

本発明のワクチン複合体を用いて、多種の疾患及び状態を治療及び/又は防止(例えば免疫化)することができる。
方法及び材料
全血又はロイコパックからのDCの作製: ヒト末梢血単核細胞(PBMC)を、ヘパリン処理済全血又はアフェレーシス製剤のフィッコール-パックによる密度勾配遠心分離で得た。次に、プラスチック製培養皿への接着又は風ひにより単離し、サイトカイン(10 ng/ml GM-CSF 及び2 ng/ml IL-4) を培養基に加えることで未熟DCに分化させた。5日目と7日目との間にDCを採集し、フローサイトメトリで分析した。この方法で調製されたDCは、CD14-、HLA-DR+、CD11c+ マンノース受容体+ であり、高レベルのMHC クラス I 及びII、CD80 及びCD86を発現していた。
と一緒に、96ウェル平底マイクロプレートで0.2ml最終容量にして同時培養した。この混合物は37℃で同時培養された。4日目に培養物に3H-チミジン (1μCi/ウェル)をパルス印加し、18時間後に細胞をフィルタ(Millipore社)上に直接、採取した。フィルタを水で3回、洗浄した後、エタノールで1回、洗浄して、5乃至10分間、フード下で乾燥させた。その後、シンチレーション液(パッカード社、20μl/ウェル)をこのフィルタに加えた。フィルタに結合した放射活性をWallacベータ・カウンタで計数して判定した。その結果を、抗原で刺激したCTL、対、抗原又はコントロール抗原なしで刺激を与えたもの、のcpmによる刺激指数(S.I.)で表す。MHC遮断分析については、標識済みのターゲットをすべてのクラスI HLA分子遮断するためにHLA特異mAb W6/32 と一緒に、そして全てのクラス II HLA 分子を遮断するためにL243(20μg/ml)と一緒に30分間、室温でプレインキュベートした。結合しなかったmAbは遠心分離で取り除いた。
で60分間、37℃で標識した。標識後の標的をRPMI培地で3回、洗浄した後、ペレットを再懸濁させて、細胞濃度を3 x 104 細胞/mlにした。抗原特異的CTLを96ウェルV底プレートで力価測定して100:1 (エフェクタT細胞、E: 標的、T)乃至12:5:1又はそれ以下の比とした。一定数の標識済み標的を加え(100μl/ウェル又は3,000 標的細胞/ウェル)、プレートを低速(180 x g)で遠心し、37℃でインキュベートした。4時間後、100-120μlの上清を採取し、放出された放射活性をγカウンタ計数法(Wallac Instruments、Perkin-Elmer社)で判定した。 CTL活性を計算し、以下の等式を用いて%特異的溶解(致死)で表した:

但し式中、実験的 (cpm) とは、CTL (E) 及び標的 (T)を含有するウェルからの放射活性(放出されたクロム)を言い;自発 (cpm)とは、標的を0.1 ml の培地に単独で入れた(即ちCTLを添加していない)ウェルからの放射活性を言い、最大放出とは、0.1 ml 界面活性剤溶液(Igepal CA 630; syn. NP-40; RPMI 培養基に入れた5%溶液)を加えた、標的を入れたウェルからの放射活性を言う。よくコントロールされた実験条件下では、自発的な放出の数値は、最大訪中の10%以下のはずである。MHC 遮断分析のために、標識済みの標的を、全てのクラス Iを遮断するためにHLA特異的mAbであるW6/32と一緒に、そして全てのクラス II HLA 分子(20μg/ml)を遮断するためにL243と一緒に30分間、室温でプレインキュベートした。未結合のmAb を遠心分離で取り除き、mAbで被膜された標的をCTLに加えた。アイソタイプの一致するmAbをコントロールとして用いた。
ワクチン複合体のデザイン: このコンストラクトは、樹状細胞上のヒトマクロファージマンノース受容体に結合する完全ヒト抗体であるB11にβhCG抗原を連結することにより、作製された。 連結は、図3に示すように遺伝子融合により抗原を抗体重鎖に共有結合させることにより、行われた。
ワクチン複合体のデザイン: βhCG抗原をB11一本鎖融合体(ScFv)に連結することにより、第二のコンストラクトを作製した。前記B11一本鎖融合体(ScFv)は、樹状細胞上のヒトマクロファージマンノース受容体に結合する一本鎖抗体であり、完全ヒトB11抗体のVL及びVHフラグメントを含有する。連結は、図1に示すように(B11sfv-βhCGコンストラクトと言及)遺伝子融合によりB11 ScFvにカルボキシ末端に抗原を共有結合させることにより、行われた。
抗体で標識決定されたワクチンが、APC表面上でその認識受容体に認識されることは、この送達段階での最初のステップである。フローサイトメトリ研究が、βhCG-B11及びB11sfv-βhCGコンストラクトが、MRを発現している培養ヒトDCに特異的結合することを実証するために用いられてきた(図4)。
DC上のMHC クラス I及びクラス II 分子を通じたT細胞へのβhCG提示(交差提示)に向けた、βhCG-B11コンストラクトのDCによる被プロセッシング能を評価した。具体的には、正常T細胞のプールを、ワクチンに暴露したDCと一緒に培養することにより、βhCG-B11コンストラクトを用いて抗原特異的T細胞を惹起した。次に、できた「感作した」T細胞をそれらの活性(増殖及び致死)及び特異性について分析した。T細胞の特異性は、βhCG抗原を有する標的細胞に応答したT細胞活性を、抗原陰性コントロールに比較することにより、実証することができる。存在する場合の細胞障害性T細胞(CTL)は、βhCG関連抗原を提示する標的のみを致死させ、抗原を欠いているか、あるいは無関係の抗原を提示しているようなコントロール標的には影響しないはずである。CTL媒介性抗原認識は通常、当該ペプチドを担持しているMHC分子の流れで起きるため、MHC特異的mAbでMHC:ペプチド-CTL 相互作用が遮断されると、クラス I又はクラス II 提示が裏付けられる。
未熟DCは、ピノサイトーシス又は受容体媒介性エンドサイトーシス機序によって可溶性抗原を取り込むことができる (55)。この抗原内部移行機序はその細胞内での運命を決定し、それに対する免疫応答の質に影響を与えていると思われる (54, 55, 56)。MRを通じた内部移行は、急速なクラスリン媒介性内部移行事象として解説されてきた (57, 58)。MR自体はその細胞質側の尾に2つの推定クラスリン標的決定配列を有しており、マンノシル化した金粒子の内部移行が、EMによりクラスリンで被膜された膜穴に位置特定されている (58, 59)。クラスリン依存性エンドサイトーシスは、急速な高浸透圧ショック又はK+枯渇により特異的に破壊することができる (61)。マンノース受容体に結合したマンノシル化抗原又はマンノースがクラスリン被覆ピットを通じて内部移行するのかどうかを判定するために、未熟DCを氷上でAIM5培地と一緒に、400mM ショ糖を加えて、又は加えずに30分間、B11 mAb 又はマンノシル化BSAのいずれかの存在下でインキュベートした。その後細胞を37℃まで暖め、20分間、内部移行させた。洗浄及び固定後の細胞を共焦点顕微鏡法で分析した(データは図示せず)。B11をMRに結合させた場合には、その取り込みは、高浸透圧ショックで阻害されたことから、その内部移行機序がクラスリン被覆ピットを通じてであることが示された。対照的にマンノシル化BSAの取り込みは高浸透圧ショックでは阻害されなかったことから、その内部移行機序はクラスリン被覆ピットの形成に依存するものではないことが示された。B11よりも20倍高い濃度でも、マンノシル化BSA FITCによる表面染色は比較的に弱かった。その後の研究では、内部移行後のマンノシル化BSA FITCは、非特異的な流体相トレーサと同時局在したが、内部移行後のB11を含有するベシクルは、該非特異的トレーサを排除したことが明らかになった(データは図示せず)。B11-FITCとは対照的に、マンノシル化BSA-FITC及び流体相トレーサの両者の取り込みとも、PI3K 阻害剤ワートマニンによる前処理により、大きく遮断された(データは図示せず)。これらの結果は、マンノシル化BSAの大半が未熟樹状細胞により取り込まれ、それは非特異的なマクロピノサイトーシス機序を通じたものであることを示しており、マンノシル化抗原に対する免疫応答の質は、MRに特異的に標的決定された抗原とは大きく異なる可能性を示唆している。
単球由来DCを、B11sfv-βhCG 又はβhCG-B11 のPBS-BSA 緩衝液溶液に45分間、37℃で暴露して、CD40Lの存在下で一晩、成熟させた。次に、採取したDCを洗浄し、マウス抗βhCG で、続いてヤギ抗ヒトIgG(Fc)-PE 複合体で染色した。染色した細胞をフローサイトメータ (BD-LSR)で分析した。ほぼ10,000 回の事象を各試料で採集した。バックグラウンドの自己蛍光及びアイソタイプを適合させた抗体染色をコントロールとして役立てた。平均蛍光強度 (MFI) (データは図示せず)に基づくと、B11sfv-βhCG の、DC上に発現したMRへの結合は、βhCG-B11に対するそれと同様である。
DCにより提示されたβhCG-B11に対して生じたCTLを、βhCG-B11 及びB11sfv-βhCGに暴露した自己由来DC標的に対して検査し、他方、未処理のDCか、又はB11に暴露したDCをコントロールとして役立てた。抗原暴露後、上清中の放射活性放出を測定する4時間検定において標的に51クロムで標識し、CTLと混合した。この実験では、βhCG-B11特異的T細胞は、抗原をMHCクラスI分子上に提示する四つの標的のうちの二つを認識する。DCが抗原を欠く場合は、標的の致死は認められない(図11)。このように、DCによるβhCG-B11 の取り込みの結果、βhCG由来T細胞エピトープがCTLに認識されるのだと考えられる。
ヒトマンノース受容体(MR)を狙う抗体は、サルを含め、研究室での実験に通常用いられている他の種を由来とするMRとは交差反応しない。従って、ヒトMRを発現するトランスジェニックマウスを開発した。ヒトMRを発現するトランスジェニックマウスは、標準的なマイクロ注射技術を用い、全30エキソン遺伝子や5'側及び3'側末端からの付加的な配列を用いて、作製された。ヒトMRトランスジェニックマウス(huMR-tg)のバックグランドに株C57Bl/6 が選択されたが、それはなぜなら、この株については免疫処理及び腫瘍刺激実験がよく確立されていたからである。多様な方法を用いて、該ヒトMR導入遺伝子の発現及び調節が、ヒト組織におけるヒトMRの発現及び調節と同様であることを実証した(データは図示せず)。
B11-βhCGのin vivo ターゲティング能を、ヒトマンノース受容体(hMR)を発現するトランスジェニックマウス由来の組織をB11-βhCGの投与から24時間に分析することで調査した。注射部位近くのリンパ節及び周囲組織中のβhCG抗原の免疫組織化学染色では、リンパ節組織や周囲のマクロファージ及び間質性樹状細胞においてこの抗原の顕著な蓄積が実証された(図16)。hMRを発現しないコントロールマウスでは何らの染色も観察されなかったことから、hMRのターゲティングにより、抗原提示細胞及びリンパ節で抗原が効果的に負荷されたことが示された。
MRに特異的な抗体で標的決定されたワクチンの可能性を、huMR-tg マウス及びそれらのWt同腹仔をB11-βhCGで免疫することにより調査した。B11-βhCGに対する抗原特異的体液性免疫性が、非トランスジェニックの同腹仔に比べてトランスジェニックマウスでより急速かつおり大きく生じた(図17を参照されたい)。具体的には、図17に示すように、マウスに10μgのB11-βhCG、又は等量の遊離βhCGで3回、(矢印で示す通りに1日目、15日目及び29日目 )で免疫した。各免疫から1週間後にマウスを交配させた。
抗βhCG抗体価を標準的ELISAで判定した。

- 免疫刺激性物質が分子複合体に連結されている、請求項39に記載の方法。
- T細胞媒介性免疫応答がβhCGに対して生じるように、請求項31に記載の組成物を投与するステップを含む、対象を免疫する方法。
- 免疫刺激性物質が分子複合体に連結されている、請求項41に記載の方法。
- アジュバントを投与するステップを更に含む、請求項41に記載の方法。
