Abstract
【解決手段】本発明は、エタノールを製造する方法を提供し、この方法は、2個以上のグルコースからなる糖およびキシロースを含む培地において、該糖を分解する酵素の存在下で、グルコースおよびキシロースを資化できる微生物により発酵させる工程を含む。本発明はまた、上記方法において有用に用いられる酵母も提供する。

Description
ともにピチア・スチピチス由来のキシロースレダクターゼ(XR)遺伝子およびキシリトールデヒドロゲナーゼ(XDH)遺伝子であるXYL1(INSDアクセッション番号X59465)およびXYL2(INSDアクセッション番号X55392)、ならびにサッカロマイセス・セレビシエ由来キシルロキナーゼ(XK)遺伝子であるXKS1(INSDアクセッション番号X82408)遺伝子の細胞内発現のためのプラスミドpIUX1X2XK(図1a)を、非特許文献8に記載の手順に基づいて構築した。
本実施例では、実施例1で調製した形質転換サッカロマイセス・セレビシエがキシロース資化性であること、および実施に際してはグルコースおよびキシロースの共存下での使用が考えられるため、グルコースおよびキシロースが混在する培地におけるエタノールの生産について調べた。
実施例1で調製した形質転換サッカロマイセス・セレビシエMT8−1/pIUX1X2XK/pIBG13を、発酵培地に炭素源として50g/l セロビオースおよび50g/l キシロースを含有すること以外は、実施例2と同様にして、エタノール生産について調べた。
Claims (6)
- エタノールを製造する方法であって、2個以上のグルコースからなる糖およびキシロースを含む培地において、該糖を分解する酵素の存在下で、グルコースおよびキシロースを資化できる微生物により発酵させる工程を含む、方法。
- 前記微生物が、酵母である、請求項1に記載の方法。
- 前記酵母が、キシロース資化性遺伝子を有するサッカロマイセス・セレビシエである、請求項2に記載の方法。
- 前記酵素が前記微生物の細胞表層に提示されている、請求項1から3のいずれかに記載の方法。
- 前記培地が、キシロース量(重量基準)に対してグルコースを40%未満の量で含む、請求項1から4のいずれかに記載の方法。
- キシロース資化性遺伝子を有し、かつデンプン分解酵素および/またはセルロース分解酵素が細胞表層に提示されている、サッカロマイセス・セレビシエ酵母。
